Jute is an important fibre crop that has dominated the packaging sector for over one and a half centuries in India. For sustenance of the trade in the face of tough competition from synthetics, there is an urgent need to redesign the ongoing breeding strategy to improve both the yield and quality of jute fibre. It is therefore, essential to understand the pattern of diversity in this important commercial crop species. In the present study, genetic diversity analysis of 20 exotic germplasm lines and 20 commercial varieties of the two cultivated species (Corchorus olitorius and C. capsularis) and two wild relatives of jute (C. aestuans and C. trilocularis) was carried out using sequence tagged microsatellite site (STMS), inter simple sequence repeat (ISSR) and random amplified polymorphic DNA (RAPD) markers. The first set of six STMS markers developed from the genomic sequence of C. olitorius was not fully transferable to the related species C. capsularis. The level of intraspecific polymorphism revealed by these markers was very low. The four ISSR and 22 RAPD primers employed in the study revealed 98.44% and 100% polymorphism, respectively, across all the species, while the level of polymorphism was significantly low within a species. The commercial varieties, particularly those of C. capsularis, had an extremely narrow genetic base that demands immediate effort for diversification. The germplasm accessions in both the cultivated species showed considerably higher levels of diversity and thus should be used in broadening the base of the varieties. All the accessions of C. olitorius together with the wild species C. aestuans clustered separately from those of C. capsularis and C. trilocularis, suggesting a polyphyletic origin of the two cultivated species.  相似文献

White jute (Corchorus capsularis) and dark jute (Corchorus olitorius) are two important cultivated crops that are used for natural fibre production. Some genetic maps have been developed for dark jute, but the genetic map information for white jute (C. capsularis) is limited. In this study, a linkage map comprising 44 sequence‐related amplified polymorphisms (SRAPs), 57 intersimple sequence repeats (ISSRs) and 18 randomly amplified polymorphic DNA (RAPD) covering 2185.7 cM with a mean density of 18.7 cM per locus was constructed in an F2 population consisting of 185 individuals derived from a cross between two diverse genotypes of ‘Xinxuan No. 1’ and ‘Qiongyueqing’ in white jute. These markers were evenly distributed in the linkage groups without any clustering. This genetic linkage map construction will facilitate the mapping of agronomic traits and marker‐assisted selection breeding in white jute.  相似文献

Jute is one of the most important fibre crops, which is second only to cotton in providing environment-friendly (biodegradable and renewable) ligno-cellulose fibre. In order to improve this largely neglected crop, we conducted a preliminary study involving the following: (i) analysis of nature and extent of the genetic variability for fibre yield and four other related traits in a set of 81 genotypes belonging to two commercially cultivated Corchorus species (45 genotypes of C. olitorius + 36 genotypes of C. capsularis), (ii) development and analysis of a set of simple sequence repeat (SSR) markers from C. olitorius, and (iii) use of a sub-set of SSRs for assessment of genetic diversity in the above set of 81 genotypes. The results suggested quantitative nature of fibre yield and other related traits, with a preponderance of dominance component in genetic variance. A sub-set of 45 SSRs derived from C. olitorius, when used for a study of DNA polymorphism and genetic diversity, showed high transferability of these C. olitorius SSRs to C. capsularis. The average number of alleles for individual SSRs was surprisingly low (3.04 for both species, 2.02 for C. capsularis and 2.51 for C. olitorius), and so was the average polymorphic information content (PIC; 0.23 and 0.24 in two species). In the dendrogram obtained using a similarity matrix, the 81 genotypes were grouped into three clusters, which largely corresponded to the two species, Cluster I belonging mainly to C. capsularis and the other two closely related clusters (clusters II and III) belonging to C. olitorius. It was also shown that a minimum of 15 SSRs could give the same information as 41 SSRs, thus making many SSRs redundant. The SSR markers developed during the present study and to be developed in future will prove useful not only for evaluation of genetic diversity, but also for molecular mapping/QTL analysis, and for comparative genome analysis of the two Corchorus species. A strong sexual incompatibility barrier that exists between the two cultivated jute species, Corchorus capsularis and Corchorus olitorius, limits the scope for improvement through genetic introgression. Protoplast fusion was carried out to generate interspecific hybrid cell lines. Cotyledonary cell protoplasts of C. capsularis and anthocyaninpigmented hypocotyl protoplasts of C. olitorius were used in the fusion experiments, which appeared to be visually useful in the early selection of the fused products. A chloroplast DNA (cpDNA) marker was developed in jute, which showed species‐specific hybridization patterns with EcoRI‐digested total genomic DNA of C. capsularis and C. olitorius. This cpDNA marker was used in the characterization of the somatic hybrid cell lines at their early stages of growth. Evidence for the presence of both types of cpDNA in the hybrid cell lines was obtained when the total genomic DNA of 4‐ to 7‐month‐old hybrid cell lines was challenged with the chloroplast DNA marker through Southern analysis. It was shown that the early segregation of the parental chloroplasts did not occur in jute, although this is common in other plant species. The hybrid nature of the fused cell lines could also be identified through peroxidase isozyme analysis. Isozyme banding patterns were complex and varied among the hybrid cell lines.  相似文献

The pollen grains of Corchorus olitorius and capsularis (jute) germinate within ten minutes in cross-pollinations as well as in self-pollinations. No perceptible difference is observed in the number of capsularis pollen grains germinating on olitorius stigmas and vice versa, though an olitorius stigma (being bigger) accommodates more pollen than capsularis.Several hundreds of flowers of different capsularis types were pollinated with those of olitorius and vice versa for three years. Very small quantities of seeds are produced in the cross-pollinations compared to self-pollinations. The percentages of empty, partially full and full seeds produced vary greatly in different crosses including their reciprocal ones. Most of the seeds obtained are shrivelled and empty. Few apparently normal looking full seeds, more or less of the same size in appearance as selfed ones that are produced in certain crosses, weigh less than the same number of selfed seeds of the respective female parents concerned; this suggests the under-development of the embryo or/and endosperm. The seeds do not germinate. Even in spite of including some types of capsularis closer to olitorius in certain characters and the wild types of both species in the crossing programme, still there was no success in getting hybrids between the two cultivated species of jute. Style reduction methods also fail. The possibilities of getting hybrids between them are indicated.

Voor de veredeling van jute (o.a. op ziekteresistentie) zou dit belangrijke perspektieven kunnen bieden.Ofschoon wel enig zaad verkregen werd, gelukte het toch niet daaruit F1 planten op te kweken. Bij de voortzetting van het onderzoek zal gezocht worden naar methoden om de beide soorten toch te overbruggen.

遴选黄麻核心种质可为黄麻种质创新及新品种选育奠定基础。本研究以300份黄麻种质资源为基础,基于SSR分子标记和农艺性状考察,结合地理来源构建核心种质。结果表明,11个农艺性状变异系数变幅在13.06%~84.87%,表现出丰富的遗传多样性。按农艺性状聚类分析可划分为8个类群,按分子标记聚类可划分为10个类群。结合2个聚类分析、地理位置并按比例取样,建立一个由108份品种(系)组成的预选核心种质。采用44对SSR引物对其进行遗传差异分析,在遗传相似系数为0.65时,可把108份品种(系)分成圆果种和长果种两大类。根据遗传差异分析,剔除遗传相似系数大于或等于0.85的遗传冗余,获得84份品种(系)的核心种质,其中圆果种60份和长果种24份。比较84份核心种质与300份种质的农艺性状变异系数及Shannon-Wiener指数发现,两者之间相差不大,表明遴选的84份核心种质可以最大限度代表300份黄麻种质资源的遗传多样性加以利用和保存。  相似文献

Genetic variability, correlation, path coefficient analysis and test of normality was conducted in an F8 recombinant inbred aerobic rice population developed by single seed descent method to evaluate its potential as a mapping population. Estimates of genotypic variance, phenotypic variance, genotypic coefficient of variance (GCV), phenotypic coefficient of variance (PCV), heritability in the broad sense (H) and expected genetic advance at 5% selection index (GA) for grain yield and other attributing characters were computed. In all the cases, PCV was higher than GCV indicating the influence of environment on the characters. High heritability coupled with high GA was observed for several plant traits; number of tillers, plant height, total number of spikelets panicle−1, biomass plant−1, straw weight, harvest index and grain yield plant−1 and hence offered good scope for selection. Grain yield plant−1 was found to be positively correlated with plant height, number of tillers, panicle length, panicle exsertion, number of panicles plant−1, single panicle weight, test weight, number of fertile spikelets panicle−1, straw weight, biomass plant−1, harvest index and grain breadth both at genotypic and phenotypic levels. Harvest index exerted maximum positive direct effect, followed by biomass plant−1 and straw weight on grain yield plant−1 at phenotypic level. Shapiro-Wilks “W test of normality” indicated that the population was skewed towards female parent IR50 for some traits and for some others towards Moroberekan, the male parent. Most of the characters that showed skewness were platykurtic with a kurtosis value of less than 3.  相似文献

Summary Sixty tetraploid wheat (Triticum turgidum L.) landrace agrotypes collected from the central highlands of Ethiopia and one commercial check cultivar (Boohai) were evaluated at Akaki experimental station for grain yield and 11 other component traits. The objectives were to estimate phenotypic (PCV) and genetic (GCV) coefficients of variation, broad-sense heritability (H) and genetic advance (GA), and to determine the interrelationships among the various traits. Genotypic differences among the agrotypes were highly significant for all the traits considered. Compared to Boohai, the landrace agrotypes were later in days-to-heading (DTH) and maturity (DTM), and had shorter grain filling period (GFP), lower fertility (KS) and lower 1000-kernel weight (TKW). By contrast, they were superior to Boohai in tiller number (TN), biological yield (BY) and grain yield plant-1 (GYP). Intermediate to high order estimate combinations of GCV, H and GA (as % of the mean) were observed for TN, GYP, number of kernels spike-1 (NKS), harvest index (HI) and TKW. GYP showed a moderate heritability which was higher than GFP, BY and Plant height (PHt). DTH and DTM were strongly correlated, but both were negatively associated with the rest of the traits except PHt. The negative correlation of DTM with GYP was largely indirect via other characters. PHt had either a weak or negative association with the other traits. TN and TKW were positively correlated with GYP, and had high and intermediate direct effects, respectively. These two traits, however, were negatively correlated and showed a substantial counter-balance effect via one another. It appears that, for the short-term, improvement of the Ethiopian wheat landraces may be possible through indirect selection for TN and TKW or direct selection for grain yield per se. In the long-run, crossing programmes between indigenous and introduced germplasm may be necessary.  相似文献

Jute (Corchorus capsularis L. and Corchorus olitorius L.) is a cash crop of India, Bangladesh and Thailand. Jute stick and caddis are by-products of its processing. These by-products are of little economic importance at the moment, and considerable quantities are wasted annually. The present paper reviews the potential of jute wastes for industrial utilization, such as in the manufacture of pulp and paper, rayon, board, cellulose derivatives and chemicals. Isatis tinctoria L. was cultivated until the 19th century to produce indigo, a natural blue pigment used principally for dyestuffs. The current search for alternative crops and interest in natural products has led to reconsidering I. tinctoria as a crop to be grown in marginal areas to produce natural indigo. To reintroduce I. tinctoria into cultivation, its behaviour under different climatic conditions as well as its morpho-physiological and genetic diversity must be assessed in order to evaluate the possibilities of future breeding work. To do this, a Eurasian collection of 15 accessions was studied in a 2-year experiment. The study was carried out in four locations in order to assess plant performance at altitudes ranging from 380 to 1,700 m a.s.l. A second experiment evaluated the morpho-physiological diversity of several traits (some related to agronomic performances) of the collection. In a third experiment the genetic traits of the collection were characterised by using eight AFLP and eight SAMPL markers. The species showed a wide adaptability to different mountainous conditions and the populations showed high morphologic and genetic variability and differed according to their origins. Both morpho-physiological and molecular characterisation allowed the accessions to be distinguished into groups of European and Asian origin. Future breeding work is recommended because some accessions have good agronomic potential.

The present investigation was carried out to estimate the interrelationships and repeatabilities of various variability parameters, i.e., genotypic (σ2g) and phenotypic (σ2p) variances, genotypic (GCV) and phenotypic (PCV) coefficients of variation, broad-sense heritability (hb2), genetic advance (GA), and genetic gain (GG), using data derived from a number of plant characters [seed yield (SY), days to heading (DH) and maturity (DM), plant height (PH), and thousand kernel weight (TKW)] of 17 spring safflower genotypes grown in 27 environments in Iran during 2003–2005. The analysis of variance for the five characters revealed significant differences among the genotypes in most of the environments, indicating a very high variability within the genotype. High genotypic and phenotypic variances in the quantitative traits, particularly for SY, were markers of increased success. A close correlation between GCV and PCV was observed for all the traits, indicating that all of the characters were less influenced by the environment and that the variability which exists in these characters is under genetic influence; however, the relatively higher values of PCV indicate the predominant role of environment. High expected genetic advances were observed for SY, but this trait showed large fluctuations in different environments (97.4%). High and low mean values of heritability were observed for DH (74.3%) and SY (59.7%), respectively, whereas high and low mean values of genetic gain were found for SY (60.3%) and DM (4.2%), respectively. High heritability coupled with low GA as a percentage of the mean was observed for DH and MD, whereas moderate heritability with moderate GA as a percentage of the mean was exhibited for SY. Analysis of correlation among the parameters showed that they were strongly and constantly correlated with each other, but none of them were consistently well correlated with the heritability parameter (hb2). Based on estimates of the genetic variability parameters within each trial, the ranking of genotypes in the low-performance subset for all traits differed from that in the high-performance subset. This result indicates poor repeatability for the genetic variability parameters. The estimates of the parameters in the multi-year trials were almost constant and repeatable, whereas the responses over years showed poor repeatability.  相似文献

Summary To find out an effective isolation distance in olitorius jute experiments were conducted for three years in two planting designs; in one the cultivar with the dominant marker allele was planted at the centre and the cultivarcarrying the recessive marker allele was planted around and in the other the planting arrangement was just the reverse of the former. A total of nearly 20 lac seedlings were counted. Irrespective of the planting designs, at the minimum distance from the pollen source outcrossing was found to be at its maximum which, however, decreased with the distance. Directions had no influence on cross pollination in this species. At a distance beyond 9.0 metres outcrossing was found to be absent.  相似文献

Garlic cultivated in rural farms of South Italy is often a heterogeneous clone population, which can comprise different cytotypes. A collection of cultivated garlic from the University of Basilicata gene-bank, was evaluated for ploidy level, and 16 different morphological traits. Out of50 accessions, 7 were hexaploids (Allium ampeloprasum L.) and43 diploids (Allium sativum L.). Significant differences in yield were observed within and between ploidy levels. The heritable traits most correlated with yield were plant height (h2 = 0.62) within diploid types and neck diameter (h2 = 0.75) within hexaploids. Discriminant analysis did show that four characters (leaf basal width, total n° of leaves, clove diameter and neck height) were able to correctly discriminate all germ plasm accessions between the two species. Zingiber zerumbet (L) Smith, a wild clonal species related to the cultivated ginger (Zingiber officinale Roscoe), is a potential resistance donor for soft rot disease in ginger caused by Pythium aphanidermatum (Edson) Fitzp. In this study we evaluated the genetic diversity and P. aphanidermatum resistance of 74 Z. zerumbet accessions belonging to 15 populations from eight districts in Kerala state, India. The disease index (DI) of the accessions varied from 0% to 72.24% and the accessions could be separated into six frequency classes according to their DI values. More than 65% of the accessions had a DI < 20%. Eight accessions were found to be immune to the infection. The relative frequency of resistant accessions was higher in the central and northern regions of Kerala. Amplified fragment length polymorphism (AFLP) analysis of Z. zerumbet accessions using five primer combinations yielded 215 bands in total, of which 175 (81.4%) were polymorphic. Nei’s genetic diversity (h) of 0.2738 and Shannon information index (I) of 0.4012 revealed a high genetic diversity in Z. zerumbet unexpected for a clonal species. In the UPGMA dendrogram, accessions were clustered mostly according to their geographical origin and no clear correspondence was observed between the clustering pattern of accessions and their responses to Pythium aphanidermatum. The study revealed high genetic diversity and variability for pathogen resistance among Z. zerumbet accessions and confirmed the value of Z. zerumbet as a potential donor for soft rot resistance for the genetic improvement of ginger.  相似文献

The diversity among 52 landraces and cultivars of tartary buckwheat (Fagopyrum tataricum Gaertn.) and one accession of its wild ancestor, F. tataricum ssp. potanini Batalin, from diverse geographic origins was examined using random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) markers. Eighteen primers produced a total of 240 fragments, of which 153 (63.75%) were monomorphic and 87 (36.25%) polymorphic bands. UPGMA-based pairwise Jaccard’s coefficient of similarity was used to deduce the relationships among 53 genetically diverse accessions. The similarity between cultivated tartary buckwheat accessions ranged from 0.61 to 1.00. Four distinct clusters were formed which corresponded well with the geographic distribution of the tartary buckwheat. Nepalese accessions showed maximum diversity followed by Chinese accessions. Tartary buckwheat accessions from the Himalayan region of northwestern India revealed a narrow gene pool. The wild buckwheat accession did not group with any of the three cultivated tartary buckwheat groups, and formed its own single-entry group. Genetic similarity (0.59) of Chinese buckwheat accessions with the wild ancestor reaffirmed that cultivated tartary buckwheat originated in the Yunnan province of northwestern China. Consistent with some earlier reports, our study demonstrated the usefulness of the RAPD technique for the characterization of plant genetic resources and assessment of diversity between species. Twenty seven accessions of Bacopa monnieri collected from semi-temperate, subtropical and tropical environments at geographically distinct locations in India were examined for genetic variability carried in them at Lucknow, India in a semi-temperate environment. The accessions were grown in earthen pans, arranged in completely random block design, replicated four times, and observed for 13 qualitative and 24 quantitative characters, including bacoside-A content of herbage. On the basis of principal component and canonical variable analyses, the accessions could be grouped into seven clusters, of 2 to 6 accession sizes. The metroglyph analysis using five qualitative traits, cluster analysis and dendrogram drawn using D2 data separated the clusters IV and V and cluster VII from all the others. The bacoside-A content of the herbage was found to be usually low in clusters of accessions possessing vigorous vegetative growth such as in cluster VII. It was also observed that some of the clusters included accessions having wide geographical origins. It was inferred that both the gross agroclimatic environment of a region and microenvironment in the vicinity of water bodies where a Bacopa monnieri genotype occurred must have interacted for the natural selection of the concerned genotype. Soil salinity, being world's gravest and increasing environmental threat, limits the growth and development in nearly all crop plants. The experiment was laid out to investigate the tomato local and exotic germplasm for morpho‐physiological and photosynthetic attributes under salinity stress (0 (control), 8 and 12 dS m?1). The phenotype data for morpho‐physiological traits were collected; meanwhile, photosynthetic pigments were analysed also at seedling stage. Analysis of variance, interaction plot, PCV and GCV revealed significantly greater diversity among all accessions for subjecting attributes. Strikingly, principle component analysis (PCA) biplot and cluster heatmap exposed potentially salt tolerant accessions (NAQEEB, BL1076, PBLA1401, 017859, TINY TIM, BL1174, BL1078, PBLA1932, TOMMY TOE, PAKIT and CLN2413) under the highest level of salinity for β‐carotene, lycopene contents, chlorophyll b, carotenoids, pheophytin (b) and total pheophytin. The overall performance of the above‐mentioned accessions was found significant and exhibits tolerant attitude under high salt levels. We suggest that these tolerant accessions might facilitate as a potential source for further breeding programme to improve the agronomically important traits by exploiting both conventional as well as molecular breeding efforts.  相似文献

A set of 292 accessions of common wheat (Triticum aestivum L.) representing 21 germplasm pools based on geographical or breeding program origins was assayed for RFLP diversity. Thirty cDNA and genomic DNA probes and the HindIII restriction enzyme were employed for RFLP analysis. About 61% of all 233 scored bands were present in 75% or more of the accessions. All but one of the 30 probes revealed polymorphism, and the average number of distinct patterns per probe over all accessions was 9.5.Polymorphic Information Content (PIC) values within a pool varied from 0 to 0.9 and depended on the identities of both the germplasm pool and the probe. Rare banding patterns with a relative frequency of ≤0.2 within a pool were detected. These rare patterns were more likely to occur in pools exhibiting high levels of heterogeneity. The highest level of polymorphism was observed in the Turkish landraces from Southwest Asia. The Eastern U.S. soft red winter wheat germplasm pool was more genetically diverse than the other advanced germplasm pools, and nearly as diverse as the Turkish landrace pool. RFLP-based genetic relationships between germplasm pools generally tracked expectations based on common geographical origin, breeding history and/or shared parentages. The Chinese wheat landraces from Sichuan, Tibet, and Yunnan provinces were distinct from other pools. Similarity matrices for among-pool genetic distance estimates based on either band frequencies or banding pattern frequencies showed good correlation with matrices derived from Nei and Li's mean genetic similarity estimates (r=−0.82** and r=−0.73**, respectively. S.288 an offspring of a putative spontaneous interspecific hybrid between tetraploid Coffea Arabica (2n = 4x = 44) and diploid C. liberica(2n = 2x = 22) and 17 arabica coffee introgression lines (representing F2 and F4) derived from the cross S.288 x Kent arabica were evaluated for introgression of C. liberica genetic material. In all, 36 AFLP primer combinations were used in the analysis. The AFLP profiles of introgressed lines were compared to five accessions each of C. arabica and C. liberica. A total of 137 polymorphic bands were scored among the 29 accessions analysed. The introgressed genotypes exhibited 102 marker bands consisting of 65 additional bands and 37missing bands associated with introgression of C. liberica genetic material. C. liberica accessions of EA group (C. liberica var liberica of Guinean origin) seemed to be the likely progenitor in the origin of natural hybrid. Analysis of genetic relationships in the introgressed lines suggested that introgression was limited to few fragments. Segregation and wide variation in number of marker fragments in the F2 and F4progenies were attributed to chromosome recombinations. The differences in the level of introgression between introgressed parent, F2 and F4 groups was not pronounced. Therefore the alien genetic material appeared to be fixed and there was no elimination or counter-selection over generations, from introgressed parent to F4.In C. arabica accessions, only 35 polymorphic bands were seen confirming the low genetic diversity. On the contrary, although representing a small amount of alien genome introgression, the Liberica-introgressed genotypes provided notable genetic diversity. Considering the fact that the diploid species of Coffea constitute a valuable source of genetic diversity, the potential implications of variability generated by Liberica-introgressed genotypes in C. arabica breeding are discussed. In order to determine genetically diverse parents for the generation of mapping populations segregating for resistance to ascochyta blight in wild Cicer species, the genetic diversity between a selection of resistant and susceptible accessions was assessed using molecular markers. Twenty Cicer accessions — comprising eight C. reticulatum accessions, six C. echinospermum accessions, five C. bijugum accessions, and one C. arietinum accession — were compared using a combination of seven RAPD primers and seven ISSR primers. A total of 231 polymorphic bands were scored and used to determine the genetic distances between accessions using Jaccard similarity coefficients. The most genetically diverse parents for the generation of intraspecific and interspecific populations segregating for resistance to ascochyta blight are reported.
