为明确云南籼稻主栽品种对白背飞虱Sogatella furcifera(Horváth)的抗虫性及其影响产量的水稻关键生育期,为抗虫品种选育和推广提供参考依据,本文利用苗期和分蘖盛期单株接虫鉴定、成株期(移栽期—乳熟期)田间自然种群发生鉴定等方法,测定了云南籼稻区31个水稻主栽品种对白背飞虱的抗虫性及其产量。结果表明,苗期有7个品种表现高抗,23个品种表现中抗—中感,1个品种表现高感;分蘖期有20个品种表现高抗,有11个表现中抗—中感;成株期田间自然种群鉴定,31个品种成株期白背飞虱田间自然种群的累计虫量都超过了对照品种TN1,均表现为感虫。单株接虫鉴定多数品种表现抗虫,而大田期自然种群鉴定则全部表现为感虫,一定程度说明了云南籼稻区主栽品种对白背飞虱具有耐害性而无抗生性。移栽期、孕穗期、抽穗期百丛虫量与有效穗数呈显著的负相关性;移栽期、孕穗期和抽穗期的百丛虫量是造成有效穗数少和产量低的关键因子。移栽期、孕穗期和抽穗期是防治白背飞虱的关键时期。  相似文献

 西瓜花叶病毒(watermelon mosaic virus,WMV)是危害我国葫芦科作物生产的重要病原。培育和种植抗病品种是防治病毒病最经济有效的措施。本研究利用RT-PCR和血清学方法在表现环斑症状的西葫芦果实上检测到WMV,未检测到番木瓜环斑病毒(papaya ringspot virus,PRSV)。接种WMV侵染性克隆到西葫芦品种绿源冬宝,果实表面产生环斑症状,表明WMV是引起西葫芦果实表面环斑症状的重要病原。利用间接ELISA方法对山东泰安的西瓜、黄瓜、甜瓜、西葫芦和南瓜等46个样品检测发现33个样品表现WMV血清学阳性(检出率71.74%),薛庄采集的带有环斑症状的西葫芦均检测到WMV。对山东省81份葫芦科作物种质资源抗WMV鉴定发现西葫芦品种万盛丰宝、盛丰金珠表现中抗(MR);西瓜品种绿宝新秀和浪潮一号表现中抗(MR);甜瓜品种黄皮面瓜表现抗病(R);黄瓜品种星君贝贝表现中抗(MR);南瓜品种爱维80南瓜表现高抗(HR),蜜本南瓜、传统蜜本南瓜、七叶早南瓜、玲珑二号和绿贝贝迷你南瓜等5个品种表现抗病(R);瓠瓜品种均表现为感病(S)。研究结果对合理布局葫芦科作物品种防控WMV具有重要意义。  相似文献

1991 ̄1995年采用自然发病和土壤接种方法鉴定了1550份玉米品种对茎腐病的抗病性,表现高抗的397份,中抗304份,中感364份,高感485份,经重复鉴定,筛选出76份抗性稳定的自交系和杂交种。不同抗性品种用肿囊腐霉菌和串珠镰刀菌接种测定,抗性反应基本一致,人工接种比自然发病鉴定效果好,通过病圃自然发病初筛,人工土壤接处复选,可准确鉴定大量材料的抗病性。  相似文献

ABSTRACT Genetic analysis of the rice cultivar Aichi Asahi and some other Japanese cultivars for the high resistance to the blast fungus isolate CHNOS58-3-1 from China was performed. All the Japanese differential cultivars were resistant to the isolate except for 'Pi No. 4', which showed moderate resistance. Analysis of the F(2) population of a cross of the susceptible cultivar Reiho and the resistant cultivar Aichi Asahi indicated that the resistance of 'Aichi Asahi' to the isolate was conferred by one dominant gene. To identify the gene in other Japanese differential cultivars, AK lines, which were derived from a cross of 'Aichi Asahi' x 'K59' and assumed to harbor no known genes except for the new one, were used for the allelism tests. The new, completely dominant resistance gene was detected in 14 differential cultivars, but not in 'Pi No. 4', 'Yashiro-mochi', and 'K1', and was designated as Pi19(t). Pi19(t) was allelic or closely linked to Pita(2) on chromosome 12. Pi19(t) was extensively distributed among Japanese traditional local cultivars.  相似文献

Resistance of 23 important olive cultivars to Verticillium dahliae has been evaluated in four experiments under controlled conditions. Nine-month-old nursery olive plants were inoculated with a cotton non-defoliating (ND) (V4) or a cotton defoliating (D) (V117) isolate of V. dahliae. Resistance was evaluated by assessing symptom severity using a 0–4 rating scale and estimating the area under disease progress curves. The percentage of plants killed and of those which recovered from the disease were used as additional parameters for including a particular cultivar into a defined category. Most of the evaluated cultivars were susceptible, although at different levels, to both isolates of V. dahliae. All cultivars were more susceptible to the D pathotype than to the ND one. A group of 11 cultivars, including several important Spanish cultivars, were susceptible or extremely susceptible to both pathotypes of V. dahliae. A second group showed differences of resistance depending on the pathotype used. They were susceptible or extremely susceptible to the D pathotype but resistant or moderately susceptible to the ND one. Finally, 'Frantoio', 'Oblonga' and 'Empeltre' were moderately susceptible to the D isolate of V. dahliae and resistant to the ND one. The resistance of 'Empeltre' was evident by the plant ability to recover from infection with either isolates. 'Empeltre' is considered to be a valuable cultivar for inclusion in breeding programmes for resistance to Verticillium wilt.  相似文献

Head rot is a major disease of broccoli caused by the soft rot pathogens Pseudomonas fluorescens and Erwinia carotovora. Two in vitro pathogenicity tests were evaluated as a methods to identify broccoli cultivars susceptible or resistant to bacterial head rot. One test used mature heads excised from the plant and inoculated with squares of cotton lint which had been soaked in a bacterial suspension. The other test involved stab-inoculating axenically grown seedlings. With the excised head test, susceptible cultivars showed a black soft rot, whilst less susceptible or moderately resistant cultivars showed only watersoaking, or browning and slight softening of the tissue. No cultivar was completely resistant. Ten cultivars were tested, and their susceptibility ratings corresponded with previously recorded field data, with one exception. This laboratory test could be used to screen for susceptibility to head rot in broccoli breeding programmes. The seedling test distinguished differences in aggressiveness among bacterial isolates but not cultivar susceptibility. Increasing head size correlated negatively with disease resistance. Head shape, i.e. cultivars which showed a domed shape rather than a flat shape, was positively correlated with disease resistance. Thus small domed heads are more resistant to head rot than large flat heads. Other morphological characteristics, viz. floret prominence and number, and sepal stomatal number were not correlated with host resistance.  相似文献

Seedlings of tomato (19) and cucurbits (33), previously selected as rootstocks for commercial cultivars, were evaluated for their resistance to verticillium wilt under glasshouse conditions. Disease scoring was based on foliar symptoms, but in plants with no visual or ambiguous symptoms the presence of the pathogen was confirmed by isolations or PCR using specific DNA primers. Five of the cucurbit rootstocks showed resistance to infection, 11 were moderately infected, 11 were susceptible, while the remaining six were very susceptible. In general, Cucurbita pepo (courgette, pumpkin) and Lagenaria siceraria tolerated infection, Cucumis melo rootstocks were susceptible and Citrullus vulgaris was the most susceptible. When inoculated with race 1 of the pathogen, one of the tomato rootstocks was very resistant, three moderately resistant, nine tolerant, ten susceptible and one very susceptible. Pathogenicity tests with race 2 showed that none of the rootstocks exhibited high tolerance, but that there was some variation in susceptibility. Research is under way to evaluate the reaction of commercial cultivars to verticillium wilt when grafted onto resistant rootstocks.  相似文献

为明确不同百合品种对灰霉病的抗病能力,本试验采用离体叶片菌丝块接种和田间孢子液喷雾2种方法测定了百合7个不同品种对百合灰霉病的抗病性。离体测定结果表明,供试的7个品种对百合灰霉病的抗病性存在显著差异,其中‘法兰西’和‘罗蒂娜’为高感品种,‘康斯坦萨’和‘索邦’为高抗品种,‘马可波罗’、‘提拔’和‘西伯利亚’为中抗品种;田间接种发病的测定结果除‘西伯利亚’为高抗品种外其余结果与离体接种结果一致;离体接种和田间接种发病情况的系统调查分析得知,感病品种‘法兰西’和‘罗蒂娜’发病早,病情发展速度快,病情指数高;‘提拔’、‘康斯坦萨’、‘西伯利亚’和‘索邦’等抗病品种发病晚,病情发展缓慢,病情指数低;随着品种抗病性的增强,发病速度和病情指数逐渐降低。  相似文献

ABSTRACT Transformation of plants with the nucleocapsid (N) gene of Tomato spotted wilt tospovirus (TSWV) provides resistance to disease development; however, information is lacking on the response of plants to natural inoculum in the field. Three tobacco cultivars were transformed with the N gene of a dahlia isolate of TSWV (TSWV-D), and plants were evaluated over several generations in the greenhouse. The resistant phenotype was more frequently observed in 'Burley 21' than in 'KY-14' or 'K-326', but highly resistant 'Burley 21' transgenic lines were resistant to only 44% of the heterologous TSWV isolates tested. Advanced generation (R(3) and R(4)) transgenic resistant lines of 'Burley 21' and a 'K-326' F(1) hybrid containing the N genes of two TSWV isolates were evaluated in the field near Tifton, GA, where TSWV is endemic. Disease development was monitored by symptom expression and enzyme-linked immunosorbent assay (ELISA) analysis. Whereas incidence of TSWV infection in 'Burley 21' susceptible controls was 20% in 1996 and 62% in 1997, the mean incidence in transgenic lines was reduced to 4 and 31%, respectively. Three transgenic 'Burley 21' lines were identified that had significantly lower incidence of disease than susceptible controls over the two years of the study. In addition, the rate of disease increase at the onset of the 1997 epidemic was reduced for all the 'Burley 21' transgenic lines compared with the susceptible controls. The 'K-326' F(1) hybrid was as susceptible as the 'K-326' nontransformed control. ELISA analysis demonstrated that symptomless plants from the most resistant 'Burley 21' transgenic lines accumulated detectable nucleocapsid protein, whereas symptomless plants from more susceptible lines did not. We conclude that transgenic resistance to TSWV is effective in reducing incidence of the disease in the field, and that accumulation of transgene protein may be important in broad-spectrum resistance.  相似文献

Initial experiment on the reactions of five Japanese cultivars of cucumber toColletotrichum orbiculare infection in the greenhouse revealed that cv Suyo and Gibai were susceptible and moderately susceptible, respectively, while cv Shogoin fushinari and Sagami hanjiro were resistant to infection byC. orbiculare; cv Ochiai fushinari was moderately resistant. The ability of 16 plant growth promoting fungi (some isolates belonged to species ofPhoma and some non-sporulating isolates) isolated from zoysiagrass rhizospheres to induce systemic resistance in the above five cucumber cultivars was tested by growing plants in potting medium infested with barley grain inocula of PGPF in the greenhouse. The second true leaves of 21-day-old plants were challenge inoculated withC. orbiculare and disease assessed. Nine, out of 16 isolates, caused significant reduction of disease caused byC. orbiculare in at least two cultivars.Phoma isolates (GS8-1 and GS8-2) and non-sporulating isolates (GU21-2, GU23-3, and GU24-3) significantly reduced the disease in all the five cultivars. The disease suppression in cucumber was due to the induction of systemic resistance, since the inducer(s) and the pathogen were separated spatially and that the inducer did not colonize aerial portions. The resistance induced by certain isolates in a susceptible cultivar was less than that in a resistant cultivar. Disease suppression caused by isolate GU21-2 was similar to theC. orbiculare induced control in certain cultivars. The average rate of expansion of lesion diameter on leaves due toC. orbiculare was slower due to induction with the selected plant growth promoting fungi compared to the uninduced control plants. The Bayoud disease, caused by Fusarium oxysporum f. sp. albedinis (Foa), represents a major limiting factor of date palm culture in Morocco and constitutes a serious threat to the date palm plantations in Algeria and all other countries. Efficient disease prevention requires the development of resistant cultivars. In Morocco, among the cultivars listed, only six appear to be resistant to Bayoud disease, but they produce poor quality fruit. Thus, the Moroccan program of date palm genetic improvement is based on directed crossing between resistant cultivars and susceptible cultivars with good date quality traits to select resistant genotypes producing high quality fruits. In addition to the separation of the resistance to Bayoud disease and quality of the fruits characters, this breeding program is really complex due to the sex separation in the date palm, the duration of juvenile phase which is very long, and the lifespan of the date palm which requires a durable polygenic resistance. Then, the selected genotypes must be of female sex, of good date quality, and possess effective defense mechanisms against the pathogen. Moreover, the selection of the date palm resistance must necessarily take into account the mechanisms of pathogen aggressiveness. In this review, we will present and discuss studies developed on the Bayoud disease of the date palm, particularly on the disease control, the biochemical and molecular markers of resistance, the program of date palm genetic improvement of resistance, the Foa pathogenicity factors, and host defense mechanisms. It will also highlight the recent studies that showed that differential behaviour of the resistant and susceptible cultivars was not related to a difference of induction of the defense mechanisms, but to the suppression of their elicitation in the susceptible cultivars.  相似文献

Mango wilt, caused by Ceratocystis fimbriata, is one of the most important diseases affecting mango yields in Brazil. Information regarding the infection process of C. fimbriata in the stem tissues of mango from different cultivars and the basis of host resistance to the pathogen is rare in the literature. Thus, the objective of the study was to investigate how infection by two isolates of C. fimbriata can be affected by mango cultivar‐specific mechanisms of resistance. Disease progress on the inoculated stem tissues of the mango cultivars was evaluated and stem sections were obtained from the site of inoculation and prepared for histopathological observations using light microscopy. The factors mango cultivars and C. fimbriata isolates and their interaction were significant for all measures of disease development. Plants from the cultivars Espada, Haden and Palmer inoculated with isolates of C. fimbriata were more susceptible, whereas plants from the cultivars Tommy and Ubá were moderately resistant and resistant, respectively. Histopathologically, fungal isolates apparently massively colonized the stem tissues of plants from the susceptible cultivars Espada, Haden and Palmer, starting from the collenchyma and moving in the direction of the cortical parenchyma, xylem vessels and pith parenchyma. By contrast, on stem tissues of plants from the resistant cultivars Tommy Atkins and Ubá, most of the cells reacted to C. fimbriata infection by accumulating amorphous material. The results from the present study strongly indicated the importance of phenolic compounds for mango cultivar resistance against infection by Brazilian C. fimbriata isolates.  相似文献

Twenty tomato (Solanum lycopersicon) cultivars were screened for resistance against bacterial spot disease incited byXanthomonas axonopodis pv.vesicatoria under field conditions with and without pathogen infection. Screening was done by artificially inoculating aX. axonopodis pv.vesicatoria suspension to 4-week-old tomato seedlings and observing them for typical symptoms of the disease. Seedlings were categorized into highly resistant, resistant, susceptible and highly susceptible cultivars on the basis of disease incidence. Tomato cultivars were screened for defense-related enzymes, total phenols and lignin contents. The temporal patterns of all these enzymes were estimated with a moderately susceptible tomato cultivar. Native PAGE analysis of both peroxidase (POX) and polyphenol oxidase (PPO) was carried out for the time course of enzyme activities and also by selecting three different tomato cultivars, following infection with the pathogen. Based on the inducible amounts of these enzymes upon pathogen infection, the tomato cultivars were correlated with the disease incidence under field conditions. A significant (P≤0.05) correlation was observed between the degree of host resistance and the enzyme levels. In highly resistant tomato cultivars the enzyme levels, total phenols and lignin contents were increased in comparison with highly susceptible tomato cultivars. Isoform analysis of POX and PPO enzymes indicated a clear difference between resistant and susceptible tomato cultivars in the number of isoforms and also in the intensity of each isoform in the presence of pathogen infection. High populations of Pseudomonas solanacearum were detected in some, but not all stems of bacterial wilt resistant ('CRA 66', 'Hawaii 7996' and 'Caraibo') and susceptible ('Floradel') tomatoes. Latent infection, i.e. spread of P. solanacearum into xylem vessels, was confirmed in Caraibo, Hawaii 7996 and 'CRA 66' (the resistant parent of Caraibo). None of the plants within the resistant cultivars wilted and those cultivars were characterized by tolerance of the vascular tissues to high bacterial densities. In contrast, plants of cultivar Floradel showed consistent symptoms and wilted rapidly, with higher mean bacterial density than resistant cultivars. Bacterial wilt resistance was not associated with resistance to bacterial root invasion but with the capability of the plant to limit P. solanacearum colonization in the stem. The extent of bacterial colonization is proposed as a criterion to quantify tolerance, complementary to absence of external wilt symptoms used in breeding programmes for resistance.  相似文献

The development of powdery mildew on five barley varieties selected for partial resistance in the field and lacking identifiable, effective race-specific resistance factors was examined. Inoculation tests demonstrated that a reduction in the number of colonies was a major component of this resistance. Microscopic examination of segments from the fourth and fifth leaf revealed several stages at which colony development was arrested. The failure of individual penetrations to establish compatible colony growth was classified as being due either to hypersensitive or to non-hypersensitive resistance in host epidermal cells. Varying proportions of these two types of resistance mechanism were evident in all varieties examined, including the susceptible controls. In four resistant varieties, one or other mechanism predominated. The possibility of enhancing partial resistance by combining sources in which different mechanisms operate is discussed.  相似文献

通过田间定点抗病性调查,结合前人的研究结果得出:不同猕猴桃品种对细菌性溃疡病的抗性存在明显差异。金魁和两种雄株中华软、美味硬最为抗病,早鲜次之,魁蜜稍抗。华美2号、海沃德中感。秦美、金丰最感病。用最长距离法对安徽省9个猕猴桃品种进行抗细菌性溃疡病系统聚类,可以将9个品种划分为5类。在谱系图上,抗病性相同的品种大致划分在同一个类中,第1类中华软、美味硬(8、9)为免疫品种;第2类早鲜、金魁(6、7)为高抗品种;第3类魁蜜(5)为中抗品种;第4类为中感品种,华美2号(4)为偏抗病,海沃德(3)为感病品种;第5类则为感病品种,金丰(1)为高感品种,秦美(2)为感病品种。其结果与实际抗性调查情况基本相符。  相似文献

甘蔗褐锈病是目前中国蔗区发生最普遍、危害最严重的病害之一,选育和种植抗病品种是防治该病最为经济有效的措施。为明确近年国家甘蔗体系育成的50个新品种(系)对甘蔗褐锈病的抗性,确定其应用潜力,本研究通过在甘蔗褐锈病高发蔗区云南德宏、保山2个区域化试验站,采用田间自然抗性调查与分子标记辅助鉴定抗性基因的方法,于2014年和2015年对中国近年选育的50个优良新品种(系)及2个主栽品种进行抗褐锈病基因Bru1的分子检测及自然抗性评价。田间自然发病调查结果表明,50个优良新品种(系)及2个主栽品种中,高抗至中抗的有34个,占65.38%。其中15个材料表现高抗,占28.85%,16个材料表现抗病,占30.77%,3个材料表现中抗,占5.77%;分子检测结果显示,共29个抗病材料含有抗褐锈病基因Bru1,出现频率为55.77%,表明中国近年选育的优良新品种(系)中抗褐锈病性主要由Bru1控制;其余5个抗病材料均不含抗褐锈病基因Bru1,暗示除了Bru1外,可能还有其他抗褐锈病基因存在。不同系列品种田间感病品种的频率和含抗褐锈病基因Bru1的频率不同,粤糖系列品种感病品种的频率最高达到60%,含Bru1的频率最低,只有30%,抗性最弱;云蔗系列品种感锈病品种的频率最低只有12.5%,含Bru1的频率最高,达到81.25%,抗性最好。本研究结果为深入开展甘蔗抗褐锈病育种,选育和推广优良抗病品种,有效防控甘蔗褐锈病提供了科学依据和优良抗性新品种(系)。  相似文献

Ascochyta blight (AB, Ascochyta rabiei (Pass.) Lab.) is one of the most important foliar disease of chickpea (Cicer arietinum L.), globally. Chickpea is attacked by AB at any growth stage in cool and humid weather depending on the inoculum availability. However, the disease epidemics are most prominent during the flowering and podding growth stages. The main objective of this study was to determine the effect of growth stages of chickpea on the genetic resistance of AB and use this information in a resistance breeding program. Two susceptible and two moderately resistant chickpea cultivars were spray inoculated at seedling (GS1), post-seedling (GS2), vegetative (GS3), flowering (GS4) and podding (GS5) growth stages with A. rabiei conidial suspension under controlled environment conditions. Irrespective of crop cultivars the incubation period (IP) was shorter in GS1, GS4 and GS5 and was significantly extended in GS2 and GS3. Symptom development was delayed significantly in moderately resistant cultivars. The AB severity 10 days after inoculation ranged between 7 and 9 on susceptible cultivars and 3 and 5 on moderately resistant cultivars. Further the correlation coefficient of disease severity between GS1, GS4 and GS5 was highly significant (r = 0.95) indicating that, evaluation for resistance to AB can be done at GS 1 (seedling stage), and or GS4 (flowering stage) to GS5 (podding stage) growth stages of chickpea. This supports the evaluation for AB resistance using 10-day-old-seedlings in controlled environment at ICRISAT and adult plant field screening at hot-spot locations in Dhaulakuan and Ludhiana in India.  相似文献
