One known and two novel antioxidant compounds have been isolated from bamboo (Phyllostachys edulis). The butanol-soluble extract of the bamboo leaves was found to have a significant antioxidant activity, as measured by scavenging the stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical and the superoxide anion radical (O(2)(-)) in the xanthine/xanthine oxidase assay system. Antioxidant activity-directed fractionation of the extract led to the isolation and characterization of three structural isomeric chlorogenic acid derivatives: 3-O-(3'-methylcaffeoyl)quinic acid (1), 5-O-caffeoyl-4-methylquinic acid (2), and 3-O-caffeoyl-1-methylquinic acid (3). Compounds 2 and 3 were isolated and characterized for the first time from the natural products. In the DPPH scavenging assay as well as in the iron-induced rat microsomal lipid peroxidation system, compounds 2 (IC(50) = 8.8 and 19.2 microM) and 3 (IC(50) = 6.9 and 14.6 microM) showed approximately 2-4 times higher antioxidant activity than did chlorogenic acid (IC(50) = 12.3 and 28.3 microM) and other related hydroxycinnamates such as caffeic acid (IC(50) =13.7 and 25.5 microM) and ferulic acid (IC(50) = 36.5 and 56.9 microM). Among the three compounds, compound 1 yielded the weakest antioxidant activity, and the DPPH scavenging and lipid peroxidation inhibitory activity (IC(50) = 16.0 and 29.8 microM) was lower than those of chlorogenic and caffeic acids. All three compounds exhibited both superoxide scavenging activities and inhibitory effects on xanthine oxidase. Their superoxide anion (O(2)(-)) scavenging activities (IC(50) = 1, 4.3 microM; 2, 2.8 microM; and 3, 1.2 microM) were markedly stronger than those of ascorbic acid (IC(50) = 56.0 microM), alpha-tocopherol (IC(50) > 100 microM), and other test compounds, although their inhibition effects on xanthine oxidase may contribute to the potent scavenging activity. alpha-Tocopherol exerted a significant inhibitory effect (65.5% of the control) on superoxide generation in 12-O-tetradecanoylphorbol-13-acetate-induced human promyelocytic leukemia HL-60 cells, and compound 3 showed moderate activity (36.0%). On the other hand, other compounds including 1, 2, chlorogenic acid, and other antioxidants were weakly active (24.8-10.1%) in the suppression of superoxide generation.  相似文献

Purpose

Moso bamboo (Phyllostachys edulis), an important economic crop, is distributed from low- to medium-elevation mountains in Taiwan. Bamboo is a fast-growing herbaceous species with an extensive rhizome structure. With the hypothesis that the characteristics of soil organic matter and microbes might change after long-term bamboo plantation, we investigated different fractions of organic C and N as well as soil microbial biomass and activities in five moso bamboo plantations along an elevation gradient in Central Taiwan.

Materials and methods

Five soil samples (top 10 cm of soil) were collected from each bamboo plantation (600, 800, 1,000, 1,200, and 1,400 m above sea level (asl)) in January 2011. Soil was processed and analyzed for soil total C and N contents, biologically available C, potentially mineralizable N, soil microbial biomass and soil respiration (CO2). Two extraction methods (2 M KCl and hot-water extraction) were used to estimate soil soluble organic C and N (SbOC and SbON) and soil inorganic N (NH4 + and NO3 ?) concentrations to evaluate the relationship with soil organic matter and microbe characteristics in bamboo plantations.

Results and discussion

Soil total C and N contents as well as soil microbial biomass and soil respiration (CO2) of the bamboo plantations increased along the elevation gradient. Temperature changes along elevation contributed to such variations observed among the selected bamboo plantations. The SbON in hot-water extracts was highest in the 1,200-m plantation, then in the 1,400-m plantation, and lowest in the low-elevation plantations (600, 800, and 1,000 m). However, SbON in 2 M KCl extracts did not differ by elevation. The SbON was strongly correlated with soil total N in both 2 M KCl and hot-water extracts, but only SbON in hot-water extracts was strongly correlated with microbial biomass N and potentially mineralizable N. SbOC was strongly correlated with soil total C content, microbial biomass C, and biologically available C in both 2 M KCl and hot-water extracts.

Conclusions

Soil total C and N, SbOC and SbON, and microbial biomass characteristics increased in the moso bamboo plantations with increasing elevation. No altitudinal difference in specific soil respiration (CO2) rate suggested that the enhanced potentially mineralizable N and soil respiration (CO2) in the high-elevation plantations were associated with increased microbial biomass rather than microbial activities.  相似文献

The composition of microbial communities and the level of enzymatic activity in the soil are both important indicators of soil quality, but the mechanisms by which a soil bacterial community is generated and maintained are not yet fully understood. Two soil samples were collected from the same location, but each had been subjected to a different long-term fertilization treatment and was characterized by different microbial diversity, biomass and physicochemical properties. These samples were γ-sterilized and swap inoculated. Non-sterilized soil samples along with sterilized and inoculated soil samples were incubated for eight months before their nutrient content, microbial biomass, enzymatic activity and bacterial composition were analyzed. Total phosphorus, and potassium concentrations along with the overall organic matter content of the non-sterilized soil were all equal to those of the same soil that had been sterilized and self/swap inoculated. Additionally, the microbial biomass carbon concentration was not affected by the specific inoculum and varied only by soil type. The activities of catalase, invertase, urease, protease, acid phosphatase and phytase were smaller in the sterilized soils that had been inoculated with organisms from chemical fertilizer amended soil (NPK) when compared to sterilized soil inoculated with organisms from manure and chemical fertilizer amended soil (NPKM) and non-sterilized soil samples. Bacterial 16S rRNA examined by 454-pyrosequencing revealed that the composition of bacterial community reconstructed by immigrant microbial inoculum in the soil was mainly influenced by its physicochemical properties, although the microbial inoculum contained different abundances of bacterial taxa. For example, the pH of the soil was the dominant factor in reconstructing a new bacterial community. Taken together, these results demonstrated that both soil microbial composition and functionality were primarily determined by soil properties rather than the microbial inoculum, which contributed to our understanding of how soil microbial communities are generated and maintained.  相似文献

A short-term mesocosm experiment was conducted to ascertain the impact of tebuconazole on soil microbial communities. Tebuconazole was applied to soil samples with no previous pesticide history at three rates: 5, 50 and 500 mg kg−1 DW soil. Soil sampling was carried out after 0, 7, 30, 60 and 90 days of incubation to determine tebuconazole concentration and microbial properties with potential as bioindicators of soil health [i.e., basal respiration, substrate-induced respiration, microbial biomass C, enzyme activities (urease, arylsulfatase, β-glucosidase, alkaline phosphatase, dehydrogenase), nitrification rate, and functional community profiling]. Tebuconazole degradation was accurately described by a bi-exponential model (degradation half-lives varied from 9 to 263 days depending on the concentration tested). Basal respiration, substrate-induced respiration, microbial biomass C and enzyme activities were inhibited by tebuconazole. Nitrification rate was also inhibited but only during the first 30 days. Different functional community profiles were observed depending on the tebuconazole concentration used. It was concluded that tebuconazole application decreases soil microbial biomass and activity.  相似文献

Organic complexed super-phosphates (CSPs) are formed by the complexation of humic acid (HA) with calcium monophosphate. The aim of this study was to determine whether two CSPs, characterized by different HA concentrations, added to a calcareous soil at an agronomic dose, were able to maintain the phosphorus (P) in a soluble form longer than the superphosphate fertilizer. Another important goal was to verify if CSP could positively influence soil microbial biomass and soil microbiological activities. Organic complexed super-phosphates were capable of keeping a large portion of P in a soluble form under different soil water conditions. In particular, the CSP with the highest organic C content was the most effective product, capable of maintaining, in an available form, the 73 % of the initially added P at the end of the experiment. In addition, it was the most effective in increasing C–CO2 soil emission, microbial biomass carbon (C) and nitrogen (N), fluoresceine diacetate hydrolysis and activities of alkaline phosphomonoesterase, β-glucosidase and urease. The addition of CSPs to soil probably produced a priming effect, increasing several times C–CO2 release by the treated soil. The significant correlation (p?<?0.05) between C–CO2 emission and the amount of C added to soil by CSP suggests that the added HA acted as trigger molecules.  相似文献

The relationships between arylsulfatase and microbial activity were investigated in regional and microenvironmental scales, at three study sites in Israel, that represent different climatic regions—Mediterranean (sub-humid), mildly arid and arid.Total arylsulfatase activity was divided into extracellular and intracellular (microbial biomass enzyme) activities according to the chloroform-fumigation method. The results show that with increasing aridity, Corg (soil organic carbon), Cmic (soil microbial biomass carbon), Nmic (soil microbial biomass nitrogen) and respiration rate decreased, while Cmic/Corg and metabolic quotient (qCO2) increased. Total, extracellular and microbial biomass arylsulfatase activities decreased with aridity. Expressed as percentage of total activity, the arylsulfatase activity of microbial biomass in the soil, at 0-2 cm and 5-10 cm depths, accounted for more than 50% of the total, in most measurements. This activity was significantly higher in the arid sites than that found in the Mediterranean one for the 0-2 cm soil. The results indicate the importance of the microflora as an enzyme source in soils, especially in arid climate conditions.Enzyme activity in the different study sites was found to be influenced by microenvironmental conditions. The Mediterranean site showed a much higher enzyme activity under shrubs than that under rock fragments and in bare soil. In the arid site rock fragments created a favorable microenvironment for microbial activity on soil surface, which resulted in a much higher microbial biomass and arylsulfatase activity than that in bare soil.The total, extracellular and intracellular arylsulfatase activities, were significantly correlated with Corg, Cmic, Nmic and respiration rate (p<0.05) at all study sites. The correlation coefficients between microbial biomass and arylsulfatase activity were usually higher than those between organic carbon and enzyme activity, especially in the arid sites. Close relationships between microbial biomass and arylsulfatase activities in all the studied sites supported the hypothesis that Corg content and enzyme activities should be related to each other via microbial biomass. Arylsulfatase activity was found to be a good indicator of microbial one. The regression equations between these factors can be incorporated into models of biogeochemical cycling for their easy method of analysis.  相似文献

The study dealt with the assessment of the impact of deforestation on tropical soil through a comparative analysis of physicochemical and microbiological parameters of natural forest and a deforested barren site. With significant decline in clay, texturally the soil of the deforested barren site was observed to be different from that of natural forest. Bulk density and porosity data revealed structural deterioration of deforested barren soil. The soil hydrological regime was also adversely affected by the deforestation. Levels of soil organic carbon, total nitrogen, microbial biomass C, N and microfungal biomass also exhibited significant decline in deforested site. Estimation of microbial biomass and activity in soil using microcalorimetry   总被引:2,自引:0,他引:2  
Relationships between the rate of heat output from soil, the rate of respiration and the soil microbial biomass were investigated for 25 soils from northern Britain. The rate of heat output, measured in a Calvet microcalorimeter at 22°C, correlated well with the rate of carbon dioxide respiration. The average amount of heat evolved per cm3 of gas respired. 21.1 J cm?3, suggests that the biomass metabolism was largely aerobic. The rate of heat output per unit of total microbial biomass was remarkably uniform over a wide range of soils, but showed differences depending upon whether the soil had been stored or amended. Mineral soils that had been stored at 4°C had the lowest heat output, 12.0 mW g?1 biomass C, compared with a mean of 20.4 mW g?1 biomass C for freshly-collected soils. Amendment with glucose (0.5% w/w) caused an immediate increase in respiration and heat output, up to 59.4 mW g?1 biomass C for stored soils and 188.2 mW g?1 biomass C for freshly collected soils. There was a consistent relationship between the biomass and the rate of heat output from freshly collected and amended mineral and organic soils which gave a linear fit using log transformed data: y= 0.6970+ 1.025x (r= 0.98, P < 0.001) (y=log10 biomass C, μgC g?1; x=log10 rate of heat output at 22°C, μW g?1). The overall relationship between biomass and the rate of heat output for all the amended samples was: 1 g biomass C= 180.05 ± 34.61 mW.  相似文献

以燕麦田土壤为研究对象,探讨了聚丙烯酸盐类土壤改良剂及其复配(聚丙烯酸钾、聚丙烯酰胺、腐植酸钾、聚丙烯酸钾+腐植酸钾、聚丙烯酰胺+腐植酸钾)对燕麦田土壤微生物量氮及土壤酶活性的影响。结果表明,不同土壤改良剂均能提高土壤有机质、碱解氮、速效磷和速效钾的含量,各指标分别比对照增加了8.24%~30.22%、7.60%~19.29%、5.15%~29.45%和27.86%~68.86%;土壤改良剂能促使燕麦全生育期内0~10、10~20和20~40 cm各土层的土壤微生物量氮含量显著提高,聚丙烯酸钾+腐植酸钾和聚丙烯酰胺+腐植酸钾复配处理较其各单施效果显著,随土壤深度的增加土壤微生物量氮逐层递减;与对照相比,土壤改良剂能显著提高燕麦全生育期各土层过氧化氢酶活性,在抽穗期活性最高,且以聚丙烯酸钾+腐植酸钾较高;但对于脲酶,聚丙烯酸钾+腐植酸钾、聚丙烯酰胺+腐植酸钾和腐植酸钾3个处理在苗期显著低于对照,在抽穗期和成熟期高于对照,两种酶活性均随土壤深度的增加逐渐降低。  相似文献

Purpose

Intensive long-term management practices in forest ecosystems can markedly influence soils’ physicochemical and microbial properties. However, their effects on the magnitude of nutrient pools and activities of enzymes regarding nutrient cycling in subtropical forest soils remain unclear. This study aimed to examine effects of long-term intensive management (organic mulching and chemical fertilization) on concentrations of different C, N, and P fractions and activities of enzymes involved with nutrient cycling in a subtropical Lei bamboo (Phyllostachys violascens) forest soil.

Materials and methods

Soil samples were taken from a chronosequence of Lei bamboo forests with intensive management spanning 0, 5, 10, and 15 years. Concentrations of various forms of C, N, and P, as well as activities of β-glucosidase, cellobiohydrolase, urease, protease, and acid phosphatase were measured.

Results and discussion

The results revealed that the concentrations of different classes of C (water-soluble organic C, hot-water-soluble organic C, and readily oxidizable C), N (NH4+-N, NO3?-N, and water-soluble organic N), and P [resin-inorganic P (Pi), NaHCO3-Pi, NaHCO3-organic P (Po), NaOH-Pi, NaOH-Po, HCl-Pi, and residual-P] were enhanced markedly with prolonged duration of intensive management. Furthermore, activities of β-glucosidase, cellobiohydrolase, urease, protease, and acid phosphatase were increased following a 5-year treatment, while they were markedly reduced from 5- to the 15-year treatments. The 15 years of intensive management significantly reduced microbial biomass C and N concentrations by 8.2% and 31.9%, respectively, compared to the control.

Conclusions

We concluded that long-term intensive management led to the accumulation of C, N, and P, while it negatively impacted microbial biomass and activities of enzymes involved in nutrient cycling in subtropical Lei bamboo forest soils. Consequently, a reduction in chemical fertilizers should be considered toward the long-term sustainable development of subtropical Lei bamboo forests.

Management of soil ecosystems requires assessment of key soil physicochemical and microbial properties and the spatial scale over which they operate. The objectives were to determine the spatial structure of microbial biomass and activity and related soil properties, and to identify spatial relationships of these properties in prairie soils under different management histories. Soil were sampled along a transect at 0.2 m intervals in each of five long-term treatments, namely, undisturbed, cattle grazed at two intensities, and cultivated with either wheat (Triticum aestivum L.) or cotton (Gossypium hirsutum L.). Contents of organic carbon (Corg), dissolved organic C (DOC), soluble nitrogen (Nsol), and microbial biomass C (Cmic) and N (Nmic) as well as dehydrogenase activity (DH) in 70 samples were evaluated. Results showed that long-term soil management altered the spatial structure and dependence of Corg and microbial biomass and activity. Cultivation has contributed to high nugget variance for Corg, Cmic, Nmic and DH which interfered with detection of spatial structure at the sampling scale used. Contents of Corg were spatially connected to microbial biomass and activity and to DOC in the uncultivated but not in the cultivated soils, indicating that various factors affected by management may operate at different spatial scales.  相似文献

Accumulation of inorganic and labile organic phosphorus (P) in intensive agricultural systems leads to P loss from soil which can cause serious environmental problems. Soil microbes are important in mobilizing soil non-available P, however, little is known about the role of soil microbes in immobilizing P to reduce P loss. Here, we test whether stimulating microbial biomass to immobilize P could reduce the amount of labile P available for leaching. The distribution characteristics of Olsen P, organic P and microbial biomass P were determined in three intensive agricultural systems. In addition, we conducted a pot experiment with three P and four carbon (C) levels. CaCl2 extractable P was measured and used to indicate the risk of P leaching. We found that there was a positive relationship between soil organic C and microbial biomass P. Carbon addition drove the process of P immobilization and reduced CaCl2 extractable P. Microbial biomass P increased by 64% (< .05) with the addition of C, and Olsen P and CaCl2 extractable P decreased by 28% and 17%, respectively. Our results show that C addition increased microbial immobilization of P and reduced forms of labile P susceptible to leaching. Stimulating microbes to immobilize P by adding C to soils may have the potential to reduce P loss from intensive agricultural systems, reducing their environmental impact.  相似文献

We performed an assay of nutrient limitations to soil microbial biomass in forest floor material and intact cores of mineral soil collected from three North Carolina loblolly pine (Pinus taeda) forests. We added solutions containing C, N or P alone and in all possible combinations, and we measured the effects of these treatments on microbial biomass and on microbial respiration, which served as a proxy for microbial activity, during a 7-day laboratory incubation at 22 °C. The C solution used was intended to simulate the initial products of fine root decay. Additions of C dramatically increased respiration in both mineral soil and forest floor material, and C addition increased microbial biomass C in the mineral soil. Additions of N increased respiration in forest floor material and increased microbial biomass N in the mineral soil. Addition of P caused a small increase in forest floor respiration, but had no effect on microbial biomass.  相似文献

以黄淮海平原潮土为研究对象,通过室内恒温恒湿培养方法,比较研究了土壤中纤维素是否存在时,外源无机氮和葡萄糖添加对土壤微生物生物量及其活性的影响变化。实验设8个处理,包括不加任何物质的对照(CK)、添加无机氮(N)、葡萄糖(G)、纤维素(C)处理及葡萄糖和无机氮同时添加(G+N)处理,以及在纤维素存在基础上添加无机氮(C+N)、葡萄糖(C+G)、葡萄糖和无机氮同时添加(C+G+N)处理。在33天培养时间内,分别在不同的时间间隔内测定了土壤CO2累积释放量、微生物生物量碳(Cmic)、及脱氢酶(DHD)、β-葡萄糖苷酶(GLU)、过氧化氢酶(CAT)、碱性磷酸酶(APH)活性。结果表明,所有测定的微生物性质在CK与C处理间均没有显著性差异。与CK和C处理相比,其他所有处理的土壤CO2累积释放量均显著增加,其中C+G+N处理达最大值;G、G+N、C+G、C+G+N处理的土壤Cmic含量及DHD和APH活性显著提升,尤其在培养的前14天,而N和C+N处理则与CK处理相似,表示添加葡萄糖可显著增加上述处理生物活性水平,而添加无机氮则不能。添加无机氮和葡萄糖对GLU和CAT的影响不明显,大部分情况下它们在处理间没有表现出显著性差异。相关性分析表明,CO2释放速率始终与APH活性成显著正相关,但与Cmic和其他酶活性之间的相关关系则随着培养时间的不同而发生变化,这可能与不同培养时间的微生物组成或微生物利用底物的模式发生改变有关。聚类分析结果进一步表明,8个处理的土壤微生物活性水平可明显分成3组,其中活性水平最高的组只包含C+G+N处理,该结果提示在难分解纤维素存在时,无机氮和易利用有机碳的同时添加对提升土壤微生物活性的重要性。  相似文献

我国喀斯特区域面积分布较广,而喀斯特生态系统的退化已成为当前西南地区面临的严重的生态问题。本研究选取贵州中部两种不同植被类型的生态系统—乔木林和灌木林,以乔木林中的白栎、园果化香和灌木林中的火棘、竹叶椒等主要优势树种为对象,研究不同的植物树种对根际土壤微生物生物量及其细菌群落结构的影响。结果显示:乔木林系统中根际土壤微生物生物量碳、氮显著性高于灌木林,植物的根际效应在乔木林中表现更为显著;同时乔木林中的优势树种通过根系分泌物的作用显著提高根际土壤细菌多样性指数,而灌木林中优势树种的根际土壤微生物量及多样性均未表现出明显的根际效应。因此,植被的演替通过改变土壤微生物的特性影响植物-微生物-土壤之间的物质和能量循环,进一步影响喀斯特生态系统的稳定和健康功能。  相似文献

The effect of harvesting bamboo savanna on the dynamics of soil nutrient pools, N mineralization, and microbial biomass was examined. In the unharvested bamboo site NO inf3 sup- -N in soil ranged from 0.37 to 3.11 mg kg-1 soil and in the harvested site from 0.43 to 3.67 mg kg-1. NaHCO3-extractable inorganic P ranged from 0.55 to 3.58 mg kg-1 in the unharvested site and from 1.01 to 4.22 mg kg-1 in the harvested site. Over two annual cycles, the N mineralization range in the unharvested and harvested sites was 0–19.28 and 0–24.0 mg kg-1 soil month-1, respectively. The microbial C, N, and P ranges were 278–587, 28–64, and 12–26 mg kg-1 soil, respectively, with the harvested site exhibiting higher values. Bamboo harvesting depleted soil organic C by 13% and total N by 20%. Harvesting increased N mineralization, resulting in 10 kg ha-1 additional mineral N in the first 1st year and 5 kg ha-1 in the 2nd year following the harvest. Microbial biomass C, N and P increased respectively by 10, 18, and 5% as a result of bamboo harvesting.  相似文献

Land degradation causes great changes in the soil biological properties.The process of degradation may decrease soil microbial biomass and consequently decrease soil microbial activity.The study was conducted out during 2009 and 2010 at the four sites of land under native vegetation(NV),moderately degraded land(LDL),highly degraded land(HDL) and land under restoration for four years(RL) to evaluate changes in soil microbial biomass and activity in lands with different degradation levels in comparison with both land under native vegetation and land under restoration in Northeast Brazil.Soil samples were collected at 0-10 cm depth.Soil organic carbon(SOC),soil microbial biomass C(MBC) and N(MBN),soil respiration(SR),and hydrolysis of fluorescein diacetate(FDA) and dehydrogenase(DHA) activities were analyzed.After two years of evaluation,soil MBC,MBN,FDA and DHA had higher values in the NV,followed by the RL.The decreases of soil microbial biomass and enzyme activities in the degraded lands were approximately 8-10 times as large as those found in the NV.However,after land restoration,the MBC and MBN increased approximately 5-fold and 2-fold,respectively,compared with the HDL.The results showed that land degradation produced a strong decrease in soil microbial biomass.However,land restoration may promote short-and long-term increases in soil microbial biomass.  相似文献
