Blood samples were taken thrice a week for 2 weeks from 33 pregnant cows in 9 groups, 1 month apart. In addition, blood samples were collected twice daily 1 week from 8 cows before parturition. Plasma progesterone and total oestrogens were quantified by validated radioimmunoassays and total corticosteroids by a competitive protein binding technique. During the 1st 3 month of pregnancy, no remarkable changes were observed from the 3 hormones studied. From the 4th month on, concentrations of progesterone and total corticosteroids decreased significantly (P less than 0.05) and remained fairly constant until the 9th month. Values of total oestrogens increased significantly (P less than 0.05) from the 4th to 6th months and stayed unchanged afterwards. Progesterone levels in periparturient cows decreased linearly (P less than 0.01) during 3 days preceding delivery. Concentrations of total oestrogens rose sharply from day -5 and peaked at calving. Values of total corticosteroids increased dramatically on the 6th day and 24 hours before parturition. The results obtained suggest that in contrast to studies cited earlier plasma corticosteroids could play a key role for triggering parturition in cows.  相似文献

The effects of gonadotrophin releasing hormone injection into pregnant mare serum gonadotrophin treated cows on serum levels of luteinizing hormone, estradiol-17 beta and progesterone and ovulation rates was evaluated in 20 cows. Pregnant mare serum gonadotrophin treatment does not always induce superovulation as was shown in this study. Luteinizing hormone levels reached peak concentrations at 1.0 h and remained significantly (P less than or equal to 0.05) elevated at 2.0 h, 3.0 h, 4.0 h, and 7.0 h after the first gonadotrophin releasing hormone injection. Progesterone levels in the gonadotrophin releasing hormone treated group were significantly (P less than or equal to 0.05) higher than in the controls on the third and fourth days following injection. A significant correlation (r = 0.58-0.72) was noted between the number of corpus luteum and progesterone levels on each day between day 2 and day 7. More embryos were recovered (46 vs 26) from the ten treated than from the ten control cows.  相似文献

Radioimmunological investigation (RIA) of the level of pituitary FSH in the peripheral blood of cows after parturition demonstrated that this level increased successively. The FSH value of 32.93 +/- 3.71 ng per 1 ml, recorded the fourth to sixth day post partum, increased to as much as 57.78 +/- 40.98 ng per 1 ml 40 days after parturition. The LH level changed only slightly over the same period (from 1.12 +/- 0.21 ng per ml to 1.72 +/- 1.15 ng per ml). The concentration of progesterone in the blood of the cows was about 0.40 ng per ml during the first 15 days after parturition, but from the 25th day post partum it trebled, on an average (1.53 +/- 1.19 - 1.59 +/- 1.25 ng per ml). The response of the adenohypophysis of the cows to the administration of 200 micrograms of synthetic gonadoreline (spec. Dirigestran inj. Spofa) increased with increasing length of time from parturition. FSH concentration increased less markedly during the first 28 days p.p. and this increase was not uniform in time (the average increase was 1.5 to three times); in the later period the highest increase 300-500% was recorded, as a rule, 120 minutes after GnRH administration. The situation was similar in the increase in LH concentration in peripheral blood, but after the 26th day post partum the average increase in LH accounted for 500 to 600%. The concentration of progesterone in peripheral blood increased by more than 300%, on an average, from the 25th day after parturition. Three components of Ponderosa pine trees were administered via gavage to pregnant cows to determine their ability to induce premature parturition. When tips of branches, bark or needles were administered (2.7 kg/day) starting at 250 d of pregnancy, early parturition was induced in an average of 3.4, 5.0 and 9.1 d, respectively, compared to 31.8 d for water gavaged controls (P less than 0.01). Less pine material was required to induce parturition with tips and bark (9.4 and 11.2 kg/cow, respectively) than with needles (25.0 kg/cow P less than 0.05). Cows fed pine material retained fetal membranes longer after parturition; 13.0 d for needles, 10.5 d for bark and greater than 3.0 d for tips versus less than 0.5 d for controls (P less than 0.01). However, 2 of the cows fed tips died within 48 hours after parturition with fetal membranes intact and the other 2 cows fed tips became depressed, pyrexic and emaciated. They survived after extended veterinary care with antibiotic and fluid therapy. Administration of pine materials modified serum progesterone concentration profiles; there was significant elevation in progesterone 1-3 days after pine material was administered. Subsequently, pine material induced a premature decline in progesterone levels prior to parturition, which occurred more rapidly in cows fed bark and tips suggesting a dose response relationship. Pine material also modified serum cortisol concentration profiles; the main difference was a prolonged elevation in cows receiving tips. Branch tips and bark from Ponderosa pine are more potent in inducing parturition than needles.  相似文献

The influence of exposure to exogenous estradiol on the interval from parturition to first ovulation, luteinizing hormone (LH) secretion and luteal function was examined in cows. Cows were assigned at parturition to one of three treatments. Cows received either a 3.0 (1-E; n = 30) or .75 cm (1/4-E; n = 28) implant containing 17 beta-estradiol or served as untreated control animals (C; n = 33). Implants were administered within 2 days following parturition and removed on day 40 postpartum (day 0 = day of parturition). Single blood samples were collected twice weekly and analyzed for progesterone to determine length of postpartum anestrus and duration of the initial increase in progesterone. Sequential blood samples were collected on day 35 +/- .1 postpartum (15 min intervals for 18 hrs) from 5 cows in each treatment and analyzed for LH. Concentrations of estradiol were higher (P less than .01) in the 1-E (5.3 +/- .24) than in C (3.9 +/- .23) or 1/4 E (3.9 +/- .25) cows on day 35 postpartum. The interval from parturition to the first estrous cycle of normal duration was similar for cows in the C and 1-E treatment (53 +/- 2.4 and 56 +/- 2.4 days, respectively). Cows in the 1/4-E treatment had a longer (P less than .05) interval (68 +/- 2.5 days). Secretion of LH was similar among treatments on day 35 postpartum. The first normal luteal phase after parturition was preceded by a transient rise in progesterone in 81, 64 and 85% of the cows in the C, 1-E and 1/4-E treatments, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Udder oedema was studied in eight Jersey cows with rectovaginal constriction (RVC) and three normal Jersey cows. A complete physical examination was performed at least two weeks before parturition, at parturition and two weeks after parturition. Jersey cows with RVC developed udder oedema as early as 14 days before parturition, and condition persisted for three to 10 days following parturition. Mammary blood flow and cranial superficial epigastric venous blood pressure were measured in three Jersey cows with RVC with udder oedema and three normal Jersey cows. The former with udder oedema had a significant decrease in mammary blood flow at parturition compared to the normal Jersey cows (P less than 0.05); there was no significant difference in mammary blood flow between Jersey cows with RVC and normal Jersey cows two weeks before parturition (P greater than 0.05). A significant mean increase in cranial superficial epigastric venous blood pressure occurred at parturition when compared to two weeks before parturition in Jersey cows with RVC (P less than 0.05), but normal Jersey cows had no significant increase in cranial superficial epigastric vein pressure during these two periods of measurement (P greater than 0.05). There was no significant increase in jugular vein blood pressure during the two periods of measurement in Jersey cows with RVC or normal Jersey cows (P greater than 0.05). There was a significant negative correlation between mammary blood flow and cranial superficial epigastric pressure at parturition (r = 0.733, P less than 0.05).  相似文献

Peripartal serum alkaline phosphatase activity and lactate dehydrogenase activity were measured in 30 dairy cows in order to examine the association between retained fetal membranes and enzyme activity. Daily blood samples were obtained from pregnant cows, starting 15 days before the expected day of calving until eight days after parturition. Sera from 15 cows which retained fetal membranes longer than 24 hours and 15 cows which shed fetal membranes within six hours after parturition were analyzed for alkaline phosphatase and lactate dehydrogenase enzyme activities. Mean alkaline phosphatase enzyme activities ranged from 15.93 to 32.6 U/L in retained and nonretained placenta cows. There was a trend towards higher serum alkaline phosphatase activities in retained placenta cows but the differences were not significant among the groups (P greater than 0.05). Mean lactate dehydrogenase activities ranged from 307.2 to 438.86 U/L in nonretained and retained placenta cows. Lactate dehydrogenase enzyme activities in nonretained and retained placenta cows were similar (P greater than 0.05). The alkaline phosphatase and lactate dehydrogenase enzyme activities peaked at the time of parturition in both groups. However, the differences in alkaline phosphatase and lactate dehydrogenase activities on different days within non-retained and retained placenta cows were significant (P less than 0.05). Results indicate that prepartal changes in alkaline phosphatase and lactate dehydrogenase enzyme activities are not predictive of placental retention postpartum.  相似文献

Three cross-bred cows calved in March and April and were followed until day 62 after parturition. Each animal was suckled by 2 calves ad libitum. All calves were removed from the cows on day 55 after parturition. Blood was collected 3 times per day from the jugular vein by venipuncture. On 4 occasions after parturition--i.e. days 7-8, 21-22, 35-36 and 49-50, the cows were bled through a jugular venous catheter every 30 min during the 24 h. The plasma samples were analyzed for the content of 15-keto-13,14-dihydro-PGF2 alpha (main PGF2 alpha metabolite), LH, prolactin, cortisol and progesterone by radioimmunoassay methods. The concentration of PGF2 alpha increased from 280 to 730 pmol/l within the last 4 days before parturition. The highest geometric mean was 3106 pmol/l on the day of parturition. Thereafter a steady decrease of PGF2 alpha metabolite concentration was seen until day 21 when it reached plateau at 148 pmol/l. In all cows plasma LH concentrations increased significantly (P < 0.05) from about 1.6 micrograms/l on days 7-8 to 2.4 micrograms/l on days 21-22 post partum. The frequency of LH pulses showed no tendency to increase as the postpartum period progressed and averaged 6.5 pulses/24 h. Mean plasma LH concentrations increased from 2.1 micrograms/l 2 days before weaning to 3.2 micrograms/l 2 days after weaning (P < 0.05). LH peaks occurred less frequently in association with prolactin and cortisol peaks than in their absence. A partial positive correlation between PGF2 alpha metabolite and cortisol (r = 0.30) was found on days 7-8 post partum. Correlation between prolactin and cortisol on days 7-8 and 21-22 post partum was also positive (r = 0.20 and r = 0.27, respectively). There was a negative correlation between LH and cortisol on days 7-8 (r = -0.27) and days 49-50 (r = -0.21) post partum. The first and short progesterone increase observed after weaning was terminated in conjunction with PGF2 alpha metabolite peaks.  相似文献

Changes in follicular and luteal structures were assessed and concentrations of estradiol and progesterone were measured in 13 Hereford X Angus suckled beef cows during resumption of estrous cycles. Transrectal ultrasonography was used to monitor follicular size, ovulation, and formation and regression of the corpus luteum (CL). The interval from parturition to first postpartum ovulation (FO) was 82 +/- 4.7 d. Serum progesterone remained low before FO. One cow exhibited standing estrus, two cows showed other signs of estrus, and 10 displayed no signs of behavioral estrus preceding FO. All cows exhibited standing estrus before the second postpartum ovulation (SO). All cows had a short luteal phase after FO, with an average interval of 8.5 +/- .2 d between FO and SO. Concentrations of estradiol in serum during the 8 d preceding ovulation were similar before FO and SO. Maximal diameter of the preovulatory follicle was similar before FO and SO. However, the ovulatory follicle was larger in diameter at 2 d (P = .02) and 3 to 8 d (P less than .005) before FO than before SO. The time from detection until ovulation was less (P = .005) for the ovulatory follicle preceding SO than for the follicle associated with FO (8.5 vs 10.2 d, respectively, SE = .4). The second-largest follicle was larger (P less than .005) in diameter during the 8 d preceding the FO than before the SO. We studied the relationship of serum prolactin, estradiol-17 beta, and progesterone concentrations to plasma calcium, phosphorus, and free hydroxyproline concentrations, as well as to dry matter intake, in 14 aged dairy cows (mean of 4.5 parities), 7 of which became paretic, from 28 days before to 4 days after calving. Plasma calcium and phosphorus concentrations and dry matter intake decreased more at parturition in paretic cows than in nonparetic cows. Prolactin concentrations were not different between paretic and nonparetic cows, but were variable. Concentrations of estradiol were higher in paretic cows from 15 to 5 days before parturition, whereas hydroxyproline concentration was lower in paretic cows on days 10 through 3 before parturition. Progesterone concentration was lower in paretic cows and decreased earlier at parturition, compared with that in nonparetic cows. The findings suggested that high estradiol concentrations in late pregnancy inhibit bone resorption and predispose aged cows to parturient paresis. The earlier decrease in progesterone concentration at parturition and lower concentrations throughout late pregnancy might have contributed to the greater inappetence in paretic cows at parturition. The importance of prolactin in the pathogenesis of parturient paresis is not clear.  相似文献

One hundred and eighty-five Holstein-Friesian dairy cows received either sterile water or 250 micrograms of gonadotrophin releasing hormone intramuscularly on the fourth day after the first service postpartum. Heparinized blood samples were taken immediately prior to treatment (day 4) and on day 8 postinsemination for analysis of plasma progesterone concentration. Pregnancy diagnosis was carried out by rectal palpation at 42 days postinsemination and reconfirmed after 60 days postbreeding. The pregnancy rates after first, second or third service were not significantly different between gonadotrophin releasing hormone-treated and control cows. Plasma progesterone concentrations on day 4 and day 8 postinsemination, as well as the change in plasma progesterone concentration from day 4 to day 8, were similar for gonadotrophin releasing hormone-treated and control cows. The plasma progesterone concentrations on day 8 postbreeding were significantly higher (p less than 0.005) and the change in progesterone concentrations between days 4 and 8 were significantly greater (p less than 0.002) in pregnant cows compared to nonpregnant cows.  相似文献

将14头荷斯坦奶牛完全随机分为2组,每组7头。试验组从围产前期开始至产后1个月内每天每头饲喂10 g过瘤胃氯化胆碱,测定所有供试牛产后第1个泌乳月内日均产奶量;同时分别在产前1周、产后24 h、产后1、2、3、4周共6个时间点采血,测定血清中血糖、D3 羟丁酸、血脂(甘油三酯、总胆固醇, 包括高密、低密、极低密)、谷丙转氨酶、谷草转氨酶的含量。结果表明,与对照组相比,饲喂过瘤胃氯化胆碱的奶牛产后第1个月内日均产奶量比对照组提高1.7%(P>0.05);围产期及泌乳初期血糖能维持在相对稳定的水平;D3 羟丁酸含量降低(P>0.05),血脂、谷丙转氨酶、谷草转氨酶均无显著差异(P>0.05)。因此,围产期饲喂过瘤胃氯化胆碱能缓解奶牛产后能量负平衡状态,预防酮病,帮助奶牛快速恢复健康,提前进入产奶高峰期,提高牛场经济效益。  相似文献

The acyclic postpartum period in cows is prolonged due to frequent suckling when compared with twice daily milking. In conventional milking systems twice daily milking is common, while in automatic milking systems (AMS) three or more milkings are performed in early lactation. In this study, the hypothesis was tested that an increased milking frequency in AMS causes a delay in resumption of ovarian cyclicity postpartum and increases days open in German Fleckvieh cows (n = 124). Calvings were equally distributed throughout the 1-year experimental period. All cows were housed in one barn. Sixty-three cows were milked in an AMS (AC) and 61 cows were twice daily milked in a conventional milking parlour (PC). Forty-two AC and 36 PC remained in the herd until conception. After parturition, milk samples were analysed for progesterone twice weekly to record the ovarian activity until pregnancy was diagnosed. The first and the second ovulation were determined by the progesterone profile of each individual cow. The mean milking frequency during the first 3 weeks after parturition was higher in AC (3.17 +/- 0.09 milkings/day) than in PC (milked twice daily). The individual mean milking frequency of AC for the first 3 weeks of lactation ranged from 2.0 to 5.9 daily milkings. The time span from parturition until the first and second ovulation did not differ between AC and PC. However, first ovulation was observed earlier in primiparous (23 +/- 1 days) than in multiparous cows (29 +/- 1 days, P < 0.05). Within AC no relationship between milking frequency and duration of the acyclic period was observed. Days open did not significantly differ between AC and PC and between primiparous and multiparous cows. In conclusion, the increased milking frequency in AMS did neither cause a delay in resumption of ovarian cyclicity postpartum nor did it increase days open.  相似文献

We investigated the changes in the plasma concentrations of oxidative stress markers in periparturient dairy cows in hot conditions. This study was performed on 10 cows from day ?5 to day 10 after parturition. The experimental duration was divided into three periods: period 1 (5 days before parturition); period 2 (5 days after parturition); and period 3 (from day 6 to day 10 after parturition). The average rectal temperature in the cows during the experimental period was more than 39°C. Plasma aspartate aminotransferase activities in periods 2 and 3 were higher than that in period 1 (P < 0.05). Blood urea nitrogen concentration in period 3 was lower than those in periods 1 and 2 (P < 0.05). The total ascorbic acid concentration in the plasma in period 2 was greater than those in periods 1 and 3 (P < 0.05). Thiobarbituric acid reactive substances (TBARS) concentrations in periods 2 and 3 were greater than that in period 1 (P < 0.05). TBARS concentration was positively correlated with aspartate aminotransferase activity (P < 0.05). These results suggested that oxidative stress increased in cows after parturition under hot climatic conditions.  相似文献

采用1∶2.5比例将雌二醇和孕酮制成"诱乳素"处理空怀和泌乳量低母牛.107头供试母牛每日2次注射诱乳素,连续7~8 d.结果表明,处理后2~3 d内出现乳汁,第4 d开始出现正常乳汁,第10~15 d产奶量最高可达10~15 kg.处理后60 d,母牛平均产奶量以对干奶期6个月的母牛处理后为最高(P<0.05).此外,经诱导泌乳处理后,显著提高了空怀母牛的发情率和受胎率.  相似文献

The present study examined the Serum 3-methylhistidine concentrations and energy-associated variables of 5 healthy Holstein cows and 5 Holstein cows with ketosis. The serum total cholesterol and apolipoprotein B-100 concentrations and lecithin-cholesterol acyltransferase (LCAT) activity of the ketotic cows were lower than those of the healthy cows 14 days before parturition. The serum non-esterified fatty acid (NEFA) concentration on the day of parturition and 3-methylhistidine concentration 14 days after parturition were higher in the ketotic cows. The serum 3-methylhistidine concentration 14 days after parturition was negatively correlated with the serum LCAT activity 14 days before parturition and was positively correlated with the serum NEFA concentration on the day of parturition. Insufficiency of cholesterol metabolism and acceleration of body fat degradation occur before parturition in cows with ketosis, and these characteristics are correlated with acceleration of protein degradation after parturition.  相似文献

Udder edema was studied in 9 affected and 3 control Holstein cows ranging in age from 2.5 to 8 years. A complete physical examination was done in conjunction with serum analysis for total protein, albumin, globulin, sodium, potassium, chloride, calcium, and inorganic phosphorus 2 weeks before parturition, at parturition and 2 weeks following parturition. Mammary interstitial fluid samples also were obtained from cows with udder edema and analyzed for total protein, albumin, globulin, sodium, potassium, chloride, calcium, and inorganic phosphorus. Clinical signs of udder edema appeared as early as 7 days prepartum and persisted for as long as 9 days postpartum. There were no significant differences in serum constituents between affected and control cows at the same stage of pregnancy or lactation. There was a significant difference (P less than or equal to 0.05) between serum and mammary interstitial fluid in the total protein, albumin, globulin and calcium concentrations.  相似文献

The calves of the Red Spotted breed and their first-calver mothers were subject to the study of total proteinemia and of the changes in protein fractions before parturition and in a ten-day period of post-partal development. The relations between these parameters were also studied. Calves up to the 10th day of life show a rise in total proteinemia whereas cows show insignificant changes. The level of albumins in calves exhibits a slight fluctuation, whereas in mothers it increased before parturition, followed by a drop which lasted until the end of the test period. alpha-globulins show a decreasing tendency in calves from the fourth day of age and have higher values than in cows. beta-globulins in cow blood serum before parturition increase. The level of gamma-globulins in calves rises until the end of the first week of life. A linear correlation dependence was found between the values of beta-globulins in cows and calves in the first two post-partal days and between the value of beta-globulin level of mothers prior to parturition and of calves at birth. In gamma-globulins the correlation dependence is always significant, starting the first day after birth.  相似文献

Neutrophil responses were evaluated in dairy cows at parturition and on 1, 2, 3, 5 and 7 days after they were given dihydroheptaprenol (DHP) (0.5 mg/kg body weight) at parturition. The chemiluminescent index increased significantly (P less than 0.05) from parturition to 1 day after parturition in cows injected with DHP. No significant differences were observed in the number of neutrophils between untreated and DHP-treated groups of dairy cows during the immediate postparturient period.  相似文献

Data from 65 cows were collected to clarify the relationship between parity and mineral status in Holstein cows during the periparturient period. Milk fever occurred in a second‐ and a fourth‐lactation cow at parturition. Bodyweight of cows decreased immediately after parturition, and the decreased weights of cows from 1 to 6 days after parturition ranged from 11 to 27 kg. Milk yield of multiparous cows was higher than that of primiparous cows, but Ca and P concentrations in colostrum were not affected by the parity. Plasma glucose of primiparous cows was higher than that of multiparous cows, and plasma nonesterified fatty acids of fourth‐and‐more‐lactation cows at parturition was highest. Plasma Ca concentration of cows at parturition decreased with advancing parity, and plasma inorganic P concentration of third‐and‐more‐lactation cows was lowest at parturition. Plasma parathyroid hormone of fourth‐and‐more‐lactation cows at parturition was highest, but plasma hydroxyproline and alkaline phosphatase was lowest. These results suggest that the large transfer of Ca and P to colostrum is a factor in the development of milk fever in third‐and‐more‐lactation cows.  相似文献
