The use of a concentrate emulsion of Melia azedarach green fruits and a suspension of the fungus Beauveria bassiana was evaluated in the control of Rhipicephalus microplus on artificially infested cattle. The evaluation was conducted following the protocol established by the Brazilian Agriculture Ministry. Five groups of 4 or 5 animals were allocated to one of the following treatments: emulsion concentrate of M. azedarach at 0.25% (T AZED 0.25%), emulsion concentrate of M. azedarach at 0.5% (T AZED 0.5%), B. bassiana at 2.4 × 10(8) conidia (T BASS), association of the concentrate of M. azedarach at 0.25% with B. bassiana at 2.4 × 10(8) conidia (T AZED 0.25%+BASS), and control (untreated). The association of the two compounds provided better results than any one isolated treatment, indicating compatibility or perhaps a synergy between M. azedarach and B. bassiana. This treatment resulted in fewer engorged females (129 ± 70) than in the control group (233 ± 82), showing high performance against all developmental stages of the tick. Results revealed an apparent synergistic effect of M. azedarach and B. bassiana in the control of R. microplus that should be further investigated.  相似文献

OBJECTIVE: To evaluate the pathogenicity of 4 strains of the entomopathogenic fungi Metarhizium flavoviride var flavoviride and Metarhizium anisopliae var anisopliae in vitro against the bovine tick Boophilus microplus. SAMPLE POPULATION: 300 B. microplus females were used for each series of 5 conidial suspensions of the 4 fungal strains; a control group of 60 females was treated with sterilized water. PROCEDURE: For each fungal strain, sporulating cultures were used to produce conidial suspensions containing 10(4), 10(5), 10(6), 10(7), and 10(8) conidia/ml. A bioassay was performed by immersing B. microplus females in conidial suspensions; the procedure was repeated with a new group of 60 females for each dilution (a total of 300). A control group was immersed in the same volume of sterilized distilled water. Ticks were maintained at 27 C and 80% relative humidity for 7 days, and the following variables were evaluated: weight before egging, egging weight, percentage eclosion, reproductive efficiency, and percentage control. RESULTS: A low index of eclosion was found in the treated groups, with a progressive decrease in percentage eclosion (control = 100%) with increased conidial concentration. The highest level of effectiveness of biocontrol was detected in groups treated with concentrations of 10(7) and 10(8) conidia/ml. CONCLUSIONS AND CLINICAL RELEVANCE: Strains of the entomopathogenic fungi M. anisopliae var anisopliae and M. flavoviride var flavoviride were effective as biological control agents against B. microplus, with M. flavoviride strain CG-291 being the most effective. The pathogenicity of these strains and the development of an application method for this fungus for control of ticks may result in improved animal productivity and a reduction in environmental pollution through the use of fewer chemical insecticides.  相似文献

The efficacy of Metarhizium anisopliae on the control of Boophilus microplus in cattle infested naturally in the Mexican tropics was evaluated. The study was carried out on a ranch in Veracruz, Mexico. Twenty steers were randomly allocated into two groups of 10 cattle. Animals were naturally infested with B. microplus. Animals in the treated group were sprayed with M. anisopliae (strain Ma34) at a concentration of 1x10(8)conidia/ml every 15 days (four treatments). The other group remained as untreated control. Standard engorged female ticks were recorded on days 0, 1, 3, 5, 7 and 14 post-treatment. From the second application treatment (day 7) to the end of the experiment, animals in the treated group had lower tick infestation (P<0.05) with an efficacy of 40.0-91.2%. The results demonstrate the efficacy of repeated treatment with M. anisopiae (Ma34 strain) to control natural infestation of engorged female B. microplus on cattle in the Mexican tropics.  相似文献

This work aimed to assess the effect of the fungus Metarhizium anisopliae on off-host Rhipicephalus (Boophilus) microplus from tick-infested Brachiaria decumbens pasture undergoing cattle grazing. For this purpose a naturally tick-infested Brachiaria decumbens pasture of 60 m × 100 m with twelve grazing Holstein Friesian-Nelore (Bos indicus) cross breed bovines was sprayed 12 times, 21 days apart with an aqueous conidial suspension of the E9 isolate of M. anisopliae fungus. Control pasture was treated with conidial suspension vehicle only. Efficacy of treatment was evaluated by tick larvae counts on the pasture and that of engorged female ticks on grazing cattle. Fungus persistence on grass stems as well as soil pasture was assessed after each spray. Efficacy of fungus against ticks was also measured by an in vitro immersion test. Whereas in vitro test showed a clear pathogenic effect of the fungus on ticks, no pathogenic effect over R. (B.) microplus ticks was detected in the field trial by spraying pasture with fungal suspension. Fungus from the suspension could be recovered from both the grass stem as well as the soil of sprayed pasture, even though the numbers obtained varied distinctly and could only be recovered shortly after spray. Pasture environments with exposure to sun and rain, seem to be very detrimental to the fungus, thus suspensions which provide fungus with protection or more resistant isolates to these should be looked for. In order to achieve high fungal efficacy against tick under field conditions, accurate laboratory assays, optimization of application strategy and knowledge of interactions between fungal strains and environmental factors are warranted.  相似文献

The tick is a common ectoparasite of livestock and humans, and is responsible for the transmission of pathogens among hosts. Direct and indirect impacts of ticks include limiting the sustainable development of the animal husbandry industry and detrimental effects on human health. Despite these negative effects, the main method of controlling ticks remains the application of chemical acaricides, which can lead to ambient pollution and the development of tick resistance to them. The biocontrol of ticks is one of the alternative control methods that has received recent research attention. The present study used Tenebrio moliter bait methods to collect 13 species of entomopathogenic fungi from different areas in China that were then tested to observe their effects on engorged female Hyalomma anatolicum anatolicum ticks. The results showed that more than half of the isolates had some pathogenic effects on the ticks; in particular, two Beauveria bassiana strains (B.bAT01, B.bAT17) and one Metarhizium anisopliae strain (M.aAT26) were highly virulent, causing up to 90% mortality. In addition, H. anatolicum anatolicum females were treated with B. bassiana B.bAT17 using different concentrations of the fungus. Results revealed that B. bassiana B.bAT17 is highly pathogenic against engorged H. anatolicum anatolicum females. This is the first report of the pathogenic effect of entomopathogenic fungi on engorged H. anatolicum anatolicum females. However, studies of the efficiency of this fungus against ticks in the field are required before it can be used for tick management in practice.  相似文献

Previous studies have shown that one Brazilian Anaplasma marginale isolate presents an inclusion appendage (tail), while other isolates do not present such inclusion. Studies on tick transmission have been carried out with tailless isolates but little is known about transmission of tailed isolates by Boophilus microplus. Two splenectomized calves were experimentally inoculated with the tailed A. marginale isolate. During ascending rickettsemia, B. microplus larvae, free from hemoparasites, were fed on the calves and the resulting nymphs, adult males and engorged females were examined by optic and electronic microscopy. No A. marginale colonies were observed in the gut cells of engorged females and the larvae originated from them did not transmit A. marginale to susceptible calves. In addition, no colonies of A. marginale were seen in the gut cells or in salivary glands of adult males and nymphs. These results suggest that B. microplus is not the biological vector for this tailed isolate.  相似文献

The tick Boophilus microplus causes economic impact to cattle producers and has a great capacity to develop resistance to chemical acaricides. It is very important the development of new techniques to complement the control of this parasitosis. Biological control is a promising option to maintain acceptable levels of tick populations in cattle. Therefore, the present study evaluated the association of deltamethrin and the entomopathogenic fungus Metarhizium anisopliae against B. microplus larvae resistant to pyrethroid. The synthetic pyrethroid, deltamethrin, was used at concentrations of 0.39, 0.78, 1.56, 3.12 and 6.12 ppm, M. anisopliae was used at concentrations of 10(5), 10(6), 10(7) and 10(8) conidia ml(-1). Their associations were also evaluated in in vitro tests. The assays showed that the tick strain is resistant to deltamethrin, but high mortality rates were observed when deltamethrin was associated with the entomopathogen. The larvae's mortality rates ranged from 7 to 36.5% for the different concentrations of deltamethrin, however, for the different concentrations of M. anisopliae, the mortality rates ranged from 10 to 96.9%. Mortality rates were proportional to the concentrations used for both deltamethrin and the fungus. Predominantly, the association of pyrethroid and fungus resulted in higher larvae mortalility rates than those obtained with the respective non-associated concentrations. Thus, the present study indicates that this association can be used as a tool for integrated control of the tick B. microplus.  相似文献

The aim of this study was to assess the influence of different exposure times of engorged female the Rhipicephalus microplus to infective juveniles of Heterorhabditis bacteriophora isolate HP88. The engorged females were divided into seven groups (six treatments and one control) of 20 ticks each with statistically similar average weights (p>0.05) and exposed to concentrations of 300 nematodes/tick for periods of 3, 6, 12, 24, 48 and 72 h. The following biological parameters were monitored: pre-oviposition period; egg mass weight; hatching percentage; and efficacy of treatment. There was no influence of the exposure time on the pre-oviposition period (p>0.05), while there were significant differences (p<0.05) between the groups exposed for 24, 48 and 72 h and the control group with respect to the egg mass weight, and 24h for hatching percentage. Treatment efficacy reached 100% after exposure for 48 and 72 h. These results demonstrate that infective juveniles of H. bacteriophora HP88, under laboratory conditions, interfere in the majority of the biological parameters of the non-parasitic phase of engorged R. microplus females when the exposure time is greater than or equal to 24h.  相似文献

Babesia spp. infections were investigated in Bos taurus x Bos indicus dairy cows and calves and in Boophilus microplus engorged female ticks and eggs. Blood samples and engorged female ticks were collected from 25 cows and 27 calves. Babesia spp. was detected in ticks by microscopic examination of hemolymph of engorged female and by squashes of egg samples. Cattle infection was investigated in blood thin smears and by DNA amplification methods (PCR and nested PCR), using specific primers for Babesia bovis and Babesia bigemina. Merozoites of B. bovis (3 animals) and B. bigemina (12 animals) were detected exclusively in blood smears of calves. DNA amplification methods revealed that the frequency of B. bigemina infection in calves (92.6%) and in cows (84%) and of B. bovis in calves (85.2%) and in cows (100%) did not differ significantly (P > 0.05). Babesia spp. infection was more frequent in female ticks and eggs collected from calves (P < 0.01) than from cows, especially in those which had patent parasitemia. Hatching rates of B. microplus larvae were assessed according to the origin of engorged females, parasitemia of the vertebrate host, frequency and intensity of infection in engorged female tick, and frequency of egg infection. Hatching rate was lower in samples collected from calves (P < 0.01) than from cows, and in those in which Babesia spp. was detected in egg samples (P < 0.01).  相似文献

As part of a research programme designed to identify biological agents for the control of sheep scab, the pathogenicity of the fungus Metarhizium anisopliae to Psoroptes mites in the presence of sheepskin and wool was examined in the laboratory. No inhibitory effects of skin and wool were observed and high levels of infection were recorded. Subsequently the pathogenicity of formulations of both M. anisopliae and Beauveria bassiana to Psoroptes ovis was studied in vivo. For this, 36 batches of 20 adult female Psoroptes mites were confined in 25 mm diameter chambers which were attached to the backs of 6 scab-naive sheep. In some treatments, mites were exposed to the fungal pathogens for 48 h in vitro prior to being placed on the host, while other treatments involved mites with no prior exposure placed directly onto the skin of a host treated with a fungal pathogen. After 48 h on the host, mites were removed, incubated individually and all fungal infections were recorded. Fungal infection was observed in all treatments, except untreated controls. However, B. bassiana infected a significantly greater number of mites than M. anisopliae with all the formulations examined. Infection rates were highest when mites were exposed to dry conidia (>90%) and lowest with M. anisopliae in diatomaceous earth. Overall, the infection rate was not affected by whether or not the mites were given prior exposure to the conidia, before being placed on the sheep. The results demonstrate that Psoroptes mites can become infected by entomopathogenic fungi on the skin of sheep and provides a first demonstration of the potential of this technology for the control of sheep scab.  相似文献

The effect of four extracts from neem seeds (Azadirachta indica) containing 2000, 5000, 9000 and 10,000 ppm of azadirachtin A (AZA), quantified by high-performance liquid chromatography (HPLC) and diluted to 1.25%; 2.5%; 5.0%; 10.0% and 12.8% was verified by in vitro tests with engorged females and larvae of the cattle tick Rhipicephalus microplus. The results from the bioassays with the engorged females showed that the main toxic effect of the extracts was reduction of the reproductive parameters, with a sharp drop in the number of eggs laid and the hatching rate, mainly when the extracts were diluted to 10.0% and 12.8%. The product effectiveness (PE) calculations for all the solutions tested showed that the AZA solution at 10,000 ppm (N10) was the most effective. However, statistical analysis of the PE data obtained for the proportional AZA concentrations in the different diluted extracts showed significance (P<0.05) of the effects included in the model (extract dilution, principle effect (classificatory) of the assay (extract) and the interaction between the two), indicating significant variations due to the dilution, the test and the interaction between the two factors in the tests with engorged females. For solutions N2, N5, and N9, it was not possible to estimate LC(90) values in the dilution range tested. The lowest LC(50) was observed for extract N5, and although extract N10 was the only extract for which the LC(90) could be estimated within the range tested, the LC(50) was higher than for N5 and N9. These results suggest that substances other than AZA present in the extracts influenced the efficacy, especially up to a certain LC range. In the tests with larvae, no mortality was observed, indicating zero effectiveness of all the extracts tested. The results of the tests with engorged females showed that the neem extracts had acaricide activity, inhibiting egg laying and the larval hatching rate. Complementary studies are necessary to develop new methods to isolate and/or identify other substances besides AZA contained in this plant, to enable using products made from it as acaricides.  相似文献

OBJECTIVE: To assess the efficacy of ivermectin and moxidectin to prevent transmission of Babesia bovis and Babesia bigemina by Boophilus microplus to cattle under conditions of relatively intense experimental challenge. DESIGN: Naive Bos taurus calves were treated with either pour-on or injectable formulations of either ivermectin or moxidectin and then exposed to larvae of B microplus infected with B bovis or larvae or adults of B microplus infected with B bigemina. One calf was used for each combination of haemoparasite, B microplus life stage, drug and application route. PROCEDURE: Groups of calves were treated with the test drugs in either pour-on or injectable formulation and then infested with B microplus larvae infected with B bovis or B bigemina. B bigemina infected adult male ticks grown on an untreated calf were later transferred to a fourth group of animals. Infections were monitored via peripheral blood smears to determine haemoparasite transmission. RESULTS: Cattle treated with either pour-on or injectable formulations of ivermectin and moxidectin became infected with B bovis after infestation with infected larvae. Similarly, larvae infected with B bigemina survived to the nymphal stage to transmit the haemoparasite to animals treated with each drug preparation. Cattle treated with pour-on formulations of ivermectin and moxidectin then infested with adult male ticks infected with B bigemina did not become infected with B bigemina whereas those treated with the injectable formulations of ivermectin and moxidectin did show a parasitaemia. CONCLUSIONS: Injectable or pour-on formulations of ivermectin and moxidectin do not prevent transmission of Babesia to cattle by B microplus. Use of these drugs can therefore not be recommended as a primary means of protecting susceptible cattle from the risk of Babesia infection.  相似文献

Virgin females of both Boophilus decoloratus and Boophilus microplus, when mated with males of the other species, subsequently produced sterile eggs. Counts of spermiophore capsules in female seminal receptacles showed that the males of both species will mate with the females of both species and that B. microplus males show a slightly greater, but statistically insignificant, mating capacity than B. decoloratus males. South African B. microplus females, when mated with an Australian strain of B. microplus males, produced a 62% yield of viable hybrid progeny while the reciprocal cross produced only a 1,82% hatch of non-viable larvae. The hybrids were sterile when interbred and no hatch resulted when the Fl males were backcrossed with parent females. The reciprocal backcross of hybrid Fl females to parent males resulted in a low percentage hatch of non-viable larvae.  相似文献

Two isolates of Beauveria bassiana and one of Purpureocillium lilacinum (=Paecilomyces lilacinus) were found infecting Amblyomma cajennense engorged females collected on horses (0.15% infection rate from a total of 1982 specimens) and another two isolates of P. lilacinum and one Metarhizium anisopliae detected in soils (2.1% from 144 samples) collected in typical pasture habitats of this tick in Central Brazil from October 2009 to March 2011. Fungi were isolated from soils with Rhipicephalus sanguineus as surrogate baits. No fungi were found in ticks or soils during the driest months (May to August). Testing pathogenicity of fungi all R. sanguineus females were killed regardless of the isolate and fungi sporulated abundantly on the cadavers. A. cajennense was less susceptible to infection with P. lilacinum within 20 days than R. sanguineus. All three fungal species probably act as natural antagonists of A. cajennense particularly in the rainy season and have interest for integrate control of vectors of Rocky Mountain spotted fever.  相似文献

Experiments were conducted to determine if Metarhizium anisopliae isolates which are capable of growth at cattle surface temperatures could produce pathogenicity to Boophilus microplus in laboratory and field studies. The diurnal temperature fluctuation on the surface of cattle was monitored. The temperature tolerance of M. anisopliae isolates (ARSEF3297 and IMI386697) was determined and their pathogenicity to B. microplus compared at a standard bioassay temperature (28 degrees C) and at a temperature similar to the cattle surface (31-35 degrees C). The effect of the two isolates on the B. microplus population on cattle under field conditions was determined. The temperature of the fore udder, rear udder, ribs and neck regions of the mixed Holstein cattle fluctuated between 30 and 35 degrees C, in a similar pattern to the prevailing environmental temperature. However, wider fluctuations were obtained on the ears (28-35 degrees C) and spine (30-41 degrees C). The colony radius of both isolates declined as temperature increased, however, the growth of IMI386697 was five times greater than ARSEF3297 at 34 degrees C. At 28 degrees C, the pathogenicity of both isolates to B. microplus was similar, however, at 31-35 degrees C, IMI386697 was more pathogenic than ARSEF3297. Both isolates reduced the B. microplus population on cattle in comparison to the control formulation. However, IMI386697 (8.5+/-0.64 ticks/animal) produced a greater reduction in tick numbers than ARSEF3297 (19.1+/-0.64 ticks/animal). M. anisopliae was re-isolated from 8.9% of the ticks collected from IMI386697 treated cattle as compared to 2.8% of ticks from ARSEF3297 treated cattle.  相似文献

The effects of the entomopathogenic fungus Metarhizium anisopliae (Metschnikoff) were evaluated against the common louse parasite of cattle, Bovicola bovis (Piaget) (Trichodectidae: Ischnocera). Two different concentrations and formulations of conidial suspensions were applied to contained populations of adult female lice. In vitro, lice immersed in suspensions of M. anisopliae formulated in 0.03% Tween 80 developed infections; at the highest concentration (1x10(8)conidia ml-1) a mean of 71% (+/-11.52%, 95% C.I.) of lice became infected. Lice exposed to the Tween 80 only in vitro, showed high levels of survival and zero infection. In vivo, fungal conidia were applied to louse populations contained in 7 cm diameter circular arenas glued to the backs of Holstein cattle, maintained in controlled climate conditions. Conidia were formulated in either Tween 80 or silicone oil. The treatment with M. anisopliae resulted in high levels of infection and there was no overall difference between the two formulations in the number of infections observed. At the highest concentration (1x10(8)conidia ml-1) a mean of 73% (+/-15.57%, 95% C.I.) lice became infected. It is concluded that the strategic seasonal use of a fungal pathogen on cattle, applied in early winter, may be of value in suppressing the winter increase in abundance, preventing the population increasing to clinically significant levels.  相似文献

以诱集自土壤中的对桑天牛(Apriona germari)幼虫具有较高致病性的球孢白僵菌(Beauveria bassiana)Bb00菌株为原发菌株R0,通过反复接种桑天牛幼虫分别获得菌株R1、R2、R3和R4,并反复在普通查氏培养基上传代分别获得菌株M1、M2、M3和M4。将R0、R2、R4和M2、M4菌株感染桑天牛幼虫进行生物测定,发现在普通培养基上传代,会导致菌株致病力的降低,而通过桑天牛幼虫传代培养可以提高菌株的致病力。R4、R2、R0、M2、M4菌株的LT50分别为3.23、4.01、4.13、4.78和6.34d,LC50分别为0.686×106、1.470×106、3.050×106、7.940×106和9.580×106mL-1。表明通过菌株在桑天牛幼虫虫体上反复接种可以提高白僵菌对桑天牛幼虫的侵染力。  相似文献

As in most parts of the world where the cattle tick Boophilus microplus is established, resistance of ticks to acaricides occurs in New Caledonia. In order to implement laboratory resistance tests on larvae, engorged females collected in suspected farms are necessary. Investigations on the detachment schedule of the engorged females were conducted to explain certain field situations such as the lack or scarcity of engorged females on highly infested cattle driven from the pasture to the pen in the morning. Three experiments on Charolais steers naturally infested on pastures showed that: (1) engorged female burdens at sunrise are similar whether the steers spend the night in pasture or in a pen; (2) compared with steers maintained in a pen, morning detachment of females increases when the steers stay on the pasture or move from the pasture to the pen; (3) detachment rhythm of engorged females on steers staying the morning in a pen, is not influenced by feeding activity or exposition of steers to sun; (4) detachment occurs earlier for females attached on anatomical sites exposed to sun, and earlier from these sites for the steers in pasture or walking than for steers in a pen.  相似文献

The present work aimed to evaluate the endectocide activity of a new injectable long-action formulation, containing ivermectin (IVM) and abamectin (ABA). In each one of the four experiments performed, the following groups were formed: group I: 2.25% IVM (450 microg/kg)+1.25% ABA (250 microg/kg), group II: 3.15% IVM (630 microg/kg) and group III: control. Eighteen bovine naturally infected by gastrointestinal nematoda were selected for anthelmintic evaluation and necropsied on posttreatment day (PTD) 14 to estimate the total parasitic burden. For the Rhipicephalus (Boophilus) microplus field trial, 30 bovine were selected by means of counts of semi-engorged R. (B.) microplus and the therapeutic and residual efficacy evaluated by tick counts on PTDs 1, 3, 7, 14, 21, 28, 35, 42, 49, 56, 63, 70, 77, 84 and 91. In the stall test, 15 calves were artificially infested with 5000 R. (B.) microplus (Mozzo strain) larvae three times a week and daily collections of all the engorged female ticks detached from each calf were performed until the PTD 80. Forty bovine naturally infected with Dermatobia hominis larvae were selected and the number of larvae was counted by visual and tactile inspection on PTDs 3, 7, 14, 28, 35, 49, 63, 77, 91 and 105. In this trial, a formulation containing 1% doramectin (200 microg/kg) was also used. IVM+ABA formulation and 3.15% IVM eliminated four of the eight species of nematode identified. The anthelmintic efficacy of the avermectins association against Haemonchus placei, Cooperia spatulata and C. punctata was 89.64%, 98.84% and 97.69%, while 3.15% IVM achieved 30.98%, 84.79% and 75.56%, respectively. The two formulations evaluated showed reduced acaricide action on the PTD 1 and 3, reaching high efficacy percentages from PTD 14 onward. The IVM+ABA showed efficacy above 95% in the period between PTDs 21 and 49. In the stall test, it observed no difference (P>0.05) between the two formulations regarding the R. (B.) microplus counts during the entire evaluation period. IVM+ABA reduced the number of ticks from the PTD 1 to 77 (P<0.05) and 3.15% IVM reduced (P<0.05) the tick number from PTD 4 up to PTD 80. The three endectocides showed no difference (P>0.05) regarding the number of D. hominis larvae and prevented this parasite reestablishment until PTD 105. These results indicate that the IVM+ABA association showed higher anthelmintic activity and similar efficacy against arthropods to the formulation containing 3.15% IVM.  相似文献
