While embryo transfer (ET) is widely practiced, many of the transferred embryos fail to develop in cattle. To establish a more effective method for selecting bovine embryos for ET, here we quantified morphological parameters of living embryos using three-dimensional (3D) images non-invasively captured by optical coherence tomography (OCT). Seven Japanese Black embryos produced by in vitro fertilization that had reached the expanded blastocyst stage after 7 days of culture were transferred after imaged by OCT. Twenty-two parameters, including thickness and volumes of the inner cell mass, trophectoderm, and zona pellucida, and volumes of blastocoel and whole embryo, were quantified from 3D images. Four of the seven recipients became pregnant. We suggest that these 22 parameters can be potentially employed to evaluate the quality of bovine embryos before ET.  相似文献

Optical coherence tomography (OCT) is a rapid non‐invasive imaging technique that has shown high sensitivity for intra‐operative surgical margin assessment in human breast cancer clinical trials. This promising technology has not been evaluated in veterinary medicine. The objective of this study was to correlate normal and abnormal histological features with OCT images for surgical margins from excised canine soft tissue sarcoma (STS) and to establish image evaluation criteria for identifying positive surgical margins. Fourteen client‐owned dogs underwent surgical resection of a STS and OCT imaging of 2 to 4 areas of interest on the resected specimen were performed. Following imaging these areas were marked with surgical ink and trimmed for histopathology evaluation. Results showed that different tissue types had distinct characteristic appearances on OCT imaging. Adipose tissue exhibited a relatively low scattering and a honey‐comb texture pattern. Skeletal muscle and sarcoma tissue were both dense and highly scattering. While sarcoma tissue was highly scattering, it did not have organized recognizable structure in contrast to muscle which showed clear fibre alignment patterns. In this investigation, we showed different tissue types had different and characteristic scattering and image texture appearances on OCT, which closely correlate with low‐power histology images. Given the differentiation between tissue types the results support that OCT could be used to identify positive surgical margins immediately following resection of STS. Further research is needed to assess the diagnostic accuracy of this method for surgical margin assessment.  相似文献

Objectives  To document the clinical manifestations, development, progression, and mode of inheritance of the retinopathy of Coton de Tulear dogs.
Materials and methods  Multiple Coton de Tulear dogs were examined with biomicroscopy, indirect ophthalmoscopy, photopic and scotopic electroretinography, fluorescein and indocyanine green angiography, optical coherence tomography, ultrasonography, and fundic photography for 3 years.
Results  The retinopathy of Coton de Tulear dogs is inherited as an autosomal recessive condition and manifests as multifocal serous retinal detachments in homozygous puppies, between 3 and 4 months of age. Optic coherence tomography and ultrasonography confirm multiple focal serous retinal detachments. Serial fundic photographs confirmed minimal progression of lesions beyond 1 year of age. Electroretinography identified diminished scotopic and photopic amplitudes; however, the only significant differences between affected and age-matched control Coton de Tulear dogs were noted during photopic flicker electroretinograms. Leakage of fluorescein or indocyanine green through the blood ocular barriers was not detected during repeated angiograms prior to, during, and after development of the retinopathy. There was no focal pooling of fluorescein in any of the dogs examined. Focal retinal thinning was detected with optical coherence tomography over each detachment and the serous content of some of the bullae diminish after several years leaving focal areas of hyper-reflectivity.
Conclusions  Multifocal retinopathy of Coton de Tulear dogs is inherited as an autosomal recessive condition. The retinopathy manifests early in life as nonprogressive multifocal bullous retinal detachments. The blood ocular barrier remains intact. Photopic and scotopic electroretinography are minimally diminished compared to age-matched dogs.  相似文献

采用气相色谱法对牦牛酥油和普通奶油进行测试,分析其脂肪酸含量和组成,尤其是共轭亚油酸(CLA).结果表明:酥油中硬脂酸含量(83.23 g/kg)高于普通奶油(57.15 g/kg),棕榈酸含量(155.73 g/kg)比普通奶油(178.26g/kg)低;酥油中反式油酸(C18:1,t11)的含量是普通奶油的近8倍,酥油中总CLA含量是普通奶油的2倍多,尤其是C18:2,c9t11CLA异构体,在普通奶油中含量为3.38 g/kg,在酥油中为9.86 g/kg,是普通奶油的近3倍.相对于普通牛乳来说,牦牛乳具有更高的营养价值.  相似文献

Surgical excision is the foundation of treatment for early-stage solid tumors in man and companion animals. Complete excision with appropriate margins of surrounding tumor-free tissue is crucial to survival. Intraoperative imaging allows real-time visualization of tumors, assessment of surgical margins, and, potentially, lymph nodes and satellite metastatic lesions, allowing surgeons to perform complete tumor resections while sparing surrounding vital anatomic structures. Clinicians are frequently presented with laboratory test results that are not consistent with preconceived expectations for a given case. One important reason for such results is the occurrence of preanalytical errors. In this article preanalytical errors are discussed in 2 parts. The first part covers the steps of sample collection, preparation, and transportation, in which preanalytical errors often occur. This part would be most useful if read in full before collecting a sample. The second part of this article includes a systematic review of preanalytical errors divided according to individual analytes or parameters or, when appropriate, groups of analytes and parameters that represent the same biological system. This part will hopefully serve the clinician as a quick and user friendly guide for identification of possible pitfalls when presented with unexpected laboratory test results for a given case. This article is limited to errors that can affect the complete blood count, chemistry, and coagulation panels.  相似文献

OBJECTIVES: To assess the feasibility of optical coherence tomography (OCT) for examining the cat ocular fundus, to provide normative data on retinal thickness in different fundus regions, and to demonstrate selected surgically induced vitreoretinal pathologies in the cat. ANIMAL STUDIED: Forty-five eyes of 28 healthy domestic cats and two eyes of domestic cats that had undergone subretinal implantation surgery for a visual prosthesis were examined. PROCEDURES: An optical coherence tomograph (Zeiss-Humphrey) was used to examine the anesthetized animals. At least five vertical and five horizontal scans in regular distribution were recorded for each cat including (1) the peripapillary region, (2) the area centralis, and (3) the peripheral retina. Thickness was measured manually at five locations in each scan. Retinal thickness was compared in the three above-mentioned fundus regions, between eyes and between vertical and horizontal scans. OCT was additionally performed in animals with retinal detachment and a subretinal visual prosthesis. RESULTS: OCT measurements required only minimal adjustments of human settings and yielded high quality images. In comparison to humans intraretinal layers were more difficult to differentiate. Retinal thickness was highest in the peripapillary region (245 +/- 21 microm), followed by the peripheral retina (204 +/- 11 microm) and the area centralis (182 +/- 11 microm; all P < 0.0001). There was no statistically significant difference between right and left eye or between vertical and horizontal scans. OCT demonstrated retinal detachment, an iatrogenic break and a subretinal prosthetic device in high detail. CONCLUSIONS: Retinal thickness was measurable with high precision; values compare well to older histologic studies. OCT bears significant advantages over histology in enabling one to repeat measurements in living animals and thus allowing longitudinal studies. Various vitreoretinal pathologies common in feline eyes are detectable and quantifiable by OCT.  相似文献

To compare the pathogenesis of calf and rabbit strains of E. coli, gnotobiotic pigs were infected with 10(10) colony forming units (cfu) of verotoxigenic strain RDEC-1 or S102-9, or a non-verotoxigenic E. coli (X114/83). Pigs were killed 4 days later, and intestinal tissue was fixed and examined by light, scanning, and transmission electron microscopy. Strains S102-9 and RDEC-1 caused diarrhea, attached to enterocytes, and effaced microvilli, confirming that the calf and rabbit strains possessed similar mechanisms of pathogenicity. Non-verotoxigenic strain X114/83 did not cause diarrhea, but in 5/5 piglets it was detected in histological sections adherent to enterocyte surfaces. Exfoliated enterocytes were seen in 4/5. Bacteria attached to enterocytes by "cups" and "pedestals," with effacement of microvilli, were seen by electron microscopy in 1/5 piglets. It was concluded that strain S102-9 appears to be an animal equivalent of human enterohemorrhagic E. coli, that verotoxin is not essential in the pathogenesis of attaching and effacing lesions, and that the lesions induced by S102-9 are more severe in gnotobiotic pigs than in gnotobiotic or conventional calves.  相似文献

In several veterinary institutions, adjustments of CT machines have been made that allow for imaging of the standing horse. The risk of general anesthesia is eliminated and the shorter scan completion time reduces cost to clients. The objective of this retrospective, analytical study was to evaluate the technique, imaging diagnoses, feasibility, and image artifacts of multi-slice helical CT of horses’ distal limbs acquired under standing sedation. The CT images of 250 horses of various breeds, aged 3–23 years, that underwent standing distal limb CT were evaluated. Three observers assessed the CT images for artifacts and inter-observer agreement was calculated. Eighty-six percent (95% confidence interval (CI), 81–90) of the scans were carried out on the forelimbs, while 14% (95% CI, 10–19) were of the hindlimbs. A total of 65% (95% CI, 59–71) of horses that underwent standing sedated CT had single imaging diagnoses. Seventy-one percent (95% CI, 65–77) of the cases had unilateral lesions, 27% (95% CI, 22–33) had bilateral lesions and 2% (95% CI, 1–4) had no diagnosed lesions. The average CT acquisition time was 17.5 minutes (range = 15–20). The average number of acquisitions per horse was 1.7 (median = 1; range = 1–4). There was good to excellent agreement between all three observers for the presence of motion artifact in the metacarpo/metatarsophalangeal joints, identification of marked beam hardening artifact, mild solar/ skin dirt, and photon starvation artifact (kappa 0.61-0.80). No complications were encountered. Standing examination of the distal limb achieved diagnostic image quality that was obtained with minimal acquisition attempts and in a timely manner.  相似文献
