The aim of this experiment was to determine if the milk yield response of dairy cows to short-term treatment with bovine somatotropin (bST) was correlated with the non-esterified fatty-acid (NEFA) response to an adrenaline challenge. Twenty-six multiparous Holstein cows (58+/-5.4 days postpartum) received daily sub-cutaneous injections of saline for 7 days followed by sub-cutaneous injections of 20mg/day of bST for 14 days. On day 7 of the saline treatment and day 14 of the bST treatment the cows were given an intravenous injection of adrenaline (1.4 microg/kg body weight). Blood samples were taken before and after the adrenaline challenge. The difference in milk yield between the saline and the second week of bST treatment (MYR) varied considerably between animals (from -0.4 to +8.0 kg/day). MYR was positively correlated with the change in the basal concentration of NEFA between the saline and second week of bST treatment, as well as with the change in the area under the profile of NEFA above basal values following the adrenaline challenge. It remains to be established whether the greater lipolytic responses to adrenaline of the cows with the greater MYR reflects the deeper negative energy that these animals also experienced or a fundamental difference in the physiology of their adipose tissue.  相似文献

Effects of various doses of bovine somatotropin (bST) on plasma concentrations of nonesterified fatty acids (NEFA) were studied in a 14-d Latin square with six Holstein heifers. Animals were given daily injections of excipient or bST at 12:00 p.m. and fed twice daily at 7:00 a.m. and 7:00 p.m. On Day 14, plasma NEFA remained low through the day except around the 7:00 p.m. feeding when they were substantially elevated. The elevation was significantly greater in bST-treated animals and corresponded to the excitement of the animals in anticipation of the evening feeding. To further investigate this phenomenon, a second experiment was conducted in which nine growing Holstein steers were fed hourly and received either daily intramuscular (i.m.) injection of excipient or bST (120 mg/kg BW) for 15 d in a crossover design. Daily profiles of NEFA were obtained under undisturbed conditions or concurrently with intensive handling. Although no elevations could be detected in any case in control animals, bST caused a substantial rise in NEFA concentration only when animals were subjected to intensive handling. This suggested that NEFA peaks noted in bST-treated heifers in the first experiment resulted from increased ability of adipose tissue to respond to adrenergic stimulation associated with the anticipation of feeding. Consistent with this hypothesis, plasma NEFA concentrations in bST-treated steers were increased to a greater extent during a challenge involving i.v. injection of epinephrine. This amplification of adipose tissue response by bST must be considered when conducting intensive studies. Even the minimal excitement associated with blood sampling can confound the results regarding lipid mobilization, and this may have contributed to the notion that ST is a lipolytic hormone.  相似文献

The effect of exogenous bovine somatotropin (bST) treatment on the temporal pattern of insulin-like growth factor-I (IGF-I) in serum of four multiparous Holstein cows was examined. Cows (190 +/- 24 days postpartum) were treated with daily subcutaneous injections of recombinant bST (40 mg) or excipient for 12-day periods in a crossover experimental design. During excipient treatment, concentrations of IGF-I in serum were relatively constant throughout the day and averaged 70 ng/ml. Following the first bST injection, serum IGF-I began increasing after a lag of 5 to 7 hr and progressively increased over the first 2 days of treatment. Serum IGF-I levels were approximately 2-fold greater than control values at the end of day 1 of bST treatment, with a 3-fold elevation observed at the end of day 2. Concentrations of IGF-I in serum plateaued by day 3 of bST treatment. Serum concentrations of IGF-I did not follow the oscillating pattern of bST in serum resulting from daily bST injections. Milk yield (3.5% fat-corrected) plateaued after 6 days of bST treatment and was increased 61% (+15.3 kg). Both IGF-I and milk yield remained essentially constant across days for the remainder of treatment. Following cessation of treatment, serum IGF-I and milk yield gradually declined, returning to control values after approximately 4 days. The temporal pattern of circulating concentrations of IGF-I is consistent with a role for IGF-I in mediating a portion of the effects of exogenous bST in lactating cows.  相似文献

Milk production is increased in lactating cows treated with bovine somatotropin (bST) because a greater portion of absorbed nutrients are partitioned for milk synthesis. This homeorhetic action may be caused by alterations in response of key tissues to homeostatic signals. To examine this theory, acute metabolic challenges were administered to 8 multiparous Holstein cows (61 +/- 2 days postpartum) receiving daily subcutaneous injections of pituitary-derived bST (26.3 mg) or excipient during two 14-day treatment periods (crossover experimental design). Treatment with bST increased milk yield 12%. Feed intake did not change so that net energy balance decreased (+ .5 vs. -4.3 Mcal/day). Plasma concentrations of nonesterified fatty acids (NEFA) were chronically elevated in bST-treated cows, consistent with energy balance differences. However, baseline concentrations of glucose, insulin, and glucagon in plasma did not differ. On the last 3 days of treatment, individual metabolic challenges were administered via jugular cannulas: epinephrine (700 ng/kg BW), glucose (250 mg/kg BW), insulin (1.0 micrograms/kg BW), and glucagon (175 ng/kg BW). Plasma glucose was reduced after the insulin challenge to a lesser extent during bST treatment. In bST-treated cows, the increase in plasma NEFA in response to epinephrine was greater, and NEFA concentrations were lowered to a greater extent after insulin and glucose challenges. Glucose, insulin, and glucagon removal rates were not altered, nor was plasma glucose response to epinephrine or glucagon challenges. Treatment of lactating cows with bST primarily altered the response of adipose tissue to homeostatic signals which affect lipid metabolism.  相似文献

Basal hormone/metabolite concentrations and responses to intravenous challenges of glucose, insulin and epinephrine were examined in Friesian cows from selection lines of low or high genetic merit treated with recombinantly-derived bovine somatotropin (bST) or control formulation in a 2 x 2 factorial design. Cows from the low genetic merit (low breeding index, LBI) line had previously been shown to be more responsive to the galactopoietic effects of bST (50 mg/day) than those from the high breeding index (HBI) line. Despite this, comparisons of metabolic differences were not confounded by differences in energy balance because bST treatment had also caused an increase in voluntary intake of cut pasture. Circulating levels of somatotropin, insulin-like growth factor-I (IGF-I) and insulin were greater in bST-treated than control cows but neither bST treatment nor selection line influenced basal concentrations of glucose, non-esterified fatty acids (NEFA), beta-hydroxybutyrate, urea or creatinine. Treatment with bST produced a small increase in sensitivity of cows to the lipolytic effects of epinephrine and this effect was similar in both selection lines. HBI cows had greater circulating insulin levels following the glucose challenge than LBI cows but bST treatment did not affect the insulin response to exogenous glucose. Whereas bST treatment retarded the glycogenolytic response to epinephrine and the clearance of blood glucose in response to insulin in LBI cows, it had no effect on epinephrine-stimulated glycogenolysis, and caused enhanced glucose clearance in response to insulin, in HBI cows. Results are consistent with bST altering the homeorhetic control of metabolism but do not adequately explain the greater responsiveness of LBI cows to the galactopoietic effects of bST.  相似文献

Effects of daily injectable or sustained-release bovine somatotropin (bST) administrations on plasma and milk bST and insulin-like growth factor-I (IGF-I) concentrations were monitored in 74 lactating cows through early, mid- and late lactation. Treatments beginning at wk 4 of lactation were excipient (CO, 24 cows) at 2 wk intervals, daily injections of 10.3 mg bST (DI, 25 cows) and 350 mg sustained-release bST at 2 wk intervals (SR, 25 cows). The duration of treatments was 40 wk. Data were first analyzed for the overall mean concentrations covering the 40 wk treatment period. Overall mean plasma bST, milk bST and plasma IGF-I concentrations were significantly increased by both bST treatments (p<0.05). On the other hand, milk IGF-I concentrations were significantly increased (p<0.05) only in the DI group. Next, data were analyzed according to stage of lactation. The bST treatments resulted in significant increases (p<0.05) in plasma and milk bST concentrations for all early, mid- and late lactation periods. Even though plasma IGF-I concentrations were higher (p<0.05) in all lactation periods for bST treatment groups, higher milk IGF-I concentrations (p<0.05) occurred only in mid- and late lactation periods for the DI group. The patterns of bST and IGF-I concentrations in milk follows those of the plasma after bST treatments.  相似文献

Ninety-nine multiparous Brahman-influenced (1/4 to 3/8 Brahman) cows were managed to achieve low (BCS = 4.3 +/- 0.1; n = 50) or moderate (BCS = 6.1 +/- 0.1; n = 49) body condition (BC) to determine the influence of bovine somatotropin (bST) on estrous characteristics, reproductive performance, and concentrations of serum GH and plasma NEFA. Beginning 32 d postpartum, cows within each BC were assigned randomly to treatment with or without bST. Non-bST-treated cows received no treatment, and treated cows were administered bST (Posilac, 500 mg s.c.) on d -35, -21, and -7 before initiation of the breeding season. On d -7, all cows received an intravaginal, controlled internal drug-releasing (CIDR) device. On d 0 (initiation of the 70-d breeding season), the CIDR were removed and cows received prostaglandin F2alpha (PGF2alpha). Blood samples were collected from the median caudal vein of the cows at each bST treatment and at d -28 and 0. Estrous behavior was monitored by radiotelemetry during the first 30 d of the breeding season. Growth hormone was increased (P < 0.05) in low and moderate BC cows treated with bST. The percentage of cows detected in estrus during the first 30 d of the breeding season was decreased (P = 0.05) for low BC (64%) compared with moderate BC (82%) cows. The interval to first estrus tended (P = 0.07) to be shorter in low BC-bST-treated cows (3.7 +/- 1.9 d) than in moderate BC-bST-treated cows (9.6 +/- 1.8 d). During the first 30 d of the breeding season, cows in low BC had a decreased (P = 0.02) number of mounts received and increased (P = 0.001) quiescence between mounts compared with cows in moderate BC. The number of mounts received was reduced (P = 0.04) in bST-treated cows. More (P = 0.02) cows treated with bST became pregnant during the first 3 d of the breeding season compared with non-bST-treated cows. The cumulative first-service conception rate tended (P = 0.07) to be greater for bST-low BC cows than non-bST-treated cows in low or moderate BC. On d 0, NEFA were greater (P < 0.05) in bST-treated vs. non-bST-treated cows. Low BC and bST reduced the intensity of behavioral estrus in postpartum Brahman-influenced cows. However, bST increased the first-service conception rate during the first 30 d of breeding and pregnancy rates during the first 3 d of breeding in postpartum Brahman-influenced cows.  相似文献

The effect of chronic treatment with recombinant methionyl bovine somatotropin (USAN, sometribove) on anterior pituitary secretions and its target organs was investigated in six control and six sometribove-treated British Friesian cows. Cows averaged 112 and 119 d postpartum in their fourth lactation of treatment and, except for one control, had active corpora lutea. During each lactation, treated cows received sometribove injections (500 mg) every 2 wk (injection cycle) starting 60 ± 3 d postpartum. On Day 9 of one injection cycle, blood was sampled for 390 min, starting 30 min before an intravenous injection of thyrotropin (TRH, 0.33 μg/kg), corticotropin (100 μg), and gonadotropin (GnRH, 200 μg)-releasing hormones. Baseline somatotropin (bST) and adrenocorticotropin (ACTH) were higher in sometribove-treated cows vs. controls (3.27 vs. 1.03 ng/ml and 35.24 vs. 19.28 pg/ml, respectively). Baseline total thyroxine, free thyroxine, triiodothyronine, prolactin, follicle stimulating and luteinizing hormones, estradiol, and progesterone (P4) were similar across treatments. Circulating cortisol levels did not differ between control and sometribove cows, indicating a reduced adrenal ACTH responsiveness in the latter. Releasing factors induced similar changes across treatments in hormones studied with the following exceptions: a bST spike was seen in control cows only, cortisol response to ACTH was reduced in treated cows, and a significantly higher P4 concentration was detected in the plasma of sometribove-treated cows, suggesting increased ovarian responsiveness to GnRH-stimulated P4 output. The study demonstrated reduced bST response to TRH, consistent with physiologic feedback mechanisms, whereas the release profiles of the other pituitary hormones were unaffected. Target tissue responses affected by chronic sometribove treatment appear to be adrenal cortisol and ovarian P4 output.  相似文献

Twelve second-lactation Deutsches Fleckvieh cows, (FV, a dual-purpose breed) and 12 German Black and White (BW, a dairy breed) were used to evaluate similarities and differences in the response to a prolonged-release formulation of sometribove (N-methionyl bovine somatotropin, bST). Cows were milked twice daily and fed a basic diet (70% corn silage, 30% alfalfa hay) ad libitum plus 1 kg of concentrate. Additionally, concentrates and supplements were offered at the rate of 1 kg for each 22 kg of milk produced above 12 kg/d. Six cows of each breed were treated s.c. with 500 mg of sometribove at 14-d intervals from wk 9 to 29 postpartum (pp). Blood was collected by jugular vein puncture once weekly from wk 7 to 33 pp and daily for 14 d after the first and last (11th) injection of bST. Somatotropin (ST), insulin-like growth factor I (IGF-I), insulin, thyroid hormones, nonesterified fatty acids (NEFA), liver enzymes, and blood metabolites were evaluated. During bST treatment, plasma ST increased significantly on d 7 in both breeds and then decreased on d 14 after injection. Plasma IGF-I was positively correlated with ST. Plasma NEFA were high on d 7 and decreased on d 14. In response to bST, the average milk yield increased for FV and BW cows by 5.1 kg/d (29.7%) and 5.0 kg/d (28%), respectively, and 4% fat-corrected milk (FCM) increased by 5.0 kg/d (28%) and 5.3 kg/d (19.9%), respectively. The objectives of this study were to determine the effects of daily injection of bovine somatotropin (bST) on the metabolism of N and 1-[14C]leucine and on hormone and metabolite concentrations in growing beef steers. Injection of bST increased N retention (P less than .05) primarily through decreased (P less than .10) urinary N excretion. Plasma concentration of somatotropin, insulin and glucose increased (P less than .01) and of urea-N (P less than .01) and alpha-amino-N (P less than .10) decreased with bST compared with excipient injection. Total leucine flux was not altered by treatment; however, the partition of flux was. Leucine oxidation decreased (P less than .05) and leucine used for protein synthesis (P less than .10) increased, with bST compared with excipient injection. During excipient injection, 10.3 g protein were synthesized for each gram crude protein deposited, whereas during bST injections only 6.4 g were required. The average maximum contribution of myofibrillar protein degradation to whole body protein degradation, calculated from excretion of 3-methylhistidine, was 16%. Although the ratio of protein deposition/protein synthesis was low for both excipient- and bST-injected steers, the incremental efficiency of protein deposition was 50%, reflecting a dilution of protein synthesis required for turnover and a proportionately greater increase in protein synthesis than protein degradation with bST injection. In growing beef steers, bST stimulated whole body protein synthesis and decreased leucine oxidation. The change in partition of leucine flux, but not of total flux (irreversible loss), demonstrates a chronic redirection in metabolism consistent with homeorhetic control. These data from steers injected with bST suggest mechanisms by which bST affects metabolism during normal growth.  相似文献

This experiment was conducted to investigate the effect of naloxone (NAL), an opioid receptor antagonist, on pituitary LH secretion in anovulatory Holstein cows during the early postpartum period when cows were expected to be in negative net energy balance. Twenty-three cows (11 primiparous) received either saline (n = 12) or 1 mg/kg BW NAL i.v. (n = 11) on Day 14 or 15 postpartum. Jugular blood samples were collected at 15-min intervals for 2 hr before and 2.5 hr after NAL or saline. All cows received 3 ug gonadotropin releasing hormone (GnRH) at 2.5 hr post-NAL or -saline and blood collection was continued for 1 hr. Mean serum progesterone concentration was 0.33 ± 0.2 ng/ml. Mean net energy balance for all cows was -5.5 ± 0.6 Mcal/day. Naloxone caused a transient increase (P < 0.05) in serum LH concentrations in both primi- and multiparous cows within 45 min after administration. In contrast, serum LH concentrations remained unchanged in saline-treated cows. GnRH increased (P < 0.05) LH and there was no effect of treatment. These results suggest that modulation of LH secretion, at least in part, may be mediated via endogenous opioids in dairy cows before first postpartum ovulation.  相似文献

Multiparous beef (1/4 to 3/8 Bos indicus; n = 99) cows were managed to achieve low (BCS = 4.3 +/- 0.1; n = 50) or moderate (BCS = 6.1 +/- 0.1; n = 49) body condition (BC) to determine the influence of bovine (b) ST on the number of follicles, diameter of largest follicle, and serum concentrations of IGF-I, triiodothy-ronine (T3), thyroxine (T4), and prolactin. Beginning 32 d postpartum, cows within each BC were assigned randomly to treatment with or without bST. Non-bST-treated cows received no treatment, and treated cows were administered bST (Posilac, 500 mg, s.c.) on d 32, 46, and 60 postpartum. On d 60, all cows received a controlled internal drug-releasing (CIDR) device for 7 d and PGF(2alpha) at CIDR removal (CIDR-PGF(2alpha)). Blood samples (7 mL) were collected at each bST treatment and d 39 and 67 postpartum. Ultrasound was performed 1 d after CIDR-PGF(2alpha) to determine the number of small (2 to 9 mm) and large (>/=10 mm) follicles and the diameter of largest follicle. Cows treated with bST in low BC had increased (P < 0.05) IGF-I vs. low-BC non-bST-treated cows on d 39, 46, 60, and 67 postpartum. Prolactin and T3 were greater (P < 0.05) in moderate-BC than in low-BC cows on all sample dates. Thyroxine was greater (P < 0.001) in moderate-BC cows on d 46, 60, and 67 compared with low-BC cows. On d 67, bST-treated cows had greater (P < 0.05) T4 compared with non-bST-treated cows. Diameter of the largest follicle 1 d after CIDR-PGF(2alpha) was greater (P < 0.01) in anestrous cows treated with bST than for non-bST-treated anestrous cows. Objectives were to establish conditions for preparation, growth, and maintenance of a primary culture cell model of fetal uterine cells, and to determine whether cells maintained under those conditions would maintain their capacity to respond to estrogen stimulation in vitro. Fetal uteri (n = 19) were enzymatically dispersed and grown on Type 1 collagen in Dulbecco’s modified Eagle’s medium (DMEM) with 10% fetal bovine serum. Fetal-uterine cells appeared fibroblast-like and exhibited positive immunostaining for both vimentin and cytokeratin. Effects of gestational age (GA), passage number (p), and hormonal treatment on appearance of specific mRNAs were determined by RT-PCR; relative concentrations of products determined by densitometry were analyzed as the ratio of target cDNA to the GAPDH loading control. Cells expressed mRNAs for estrogen receptor (ER), TGF-β, EGF-R, PRL-R, IL-1 α, and IL-6. ER mRNA was greater at 185–200 than at 100–110 d GA (P < 0.01). All specific mRNAs examined were greater in p5 cells than p2 at both 100–110 (P < 0.01) and 185–200 d GA (P < 0.02). There was no effect of estradiol on these specific mRNAs in cells from 100–110 d GA; at 185–200 d GA, there was an estradiol (1.0 nm) effect both at 6 hr (P < 0.001) and 24 hr (P < 0.02). Overall, there was an effect of 8-br-cAMP (1 mm; 6 h) on specific mRNAs in cells at both 100–110 (P < 0.001) and 185–200 d GA (P < 0.001). In p5 cells from Day 185–200 GA, there was increased cell proliferation (P < 0.001) in response to estradiol (1 nm; 24 hr). These data suggest that primary fetal uterine cells retain their age-specific and hormone-responsive phenotype under these in vitro conditions.  相似文献

Luteolysis before the time of maternal recognition of pregnancy is one cause of low fertility in high‐producing dairy cows. The objective of this study was to assess whether induction of a secondary corpus luteum (CL) late in the luteal phase would delay the time of luteolysis. Twenty high‐producing Holstein cows were synchronized to ovulation (Day 0) with the Ovsynch protocol and received hCG (1500 IU im) on Day 12. Corpora lutea formation (as evaluated by ultrasonography) and plasma P4 concentrations were monitored from Days 4 to 36. hCG treatment induced the formation of one secondary CL (CL2) in 11 of 20 cows (55%) from the dominant follicle (mean diameter: 14.2 ± 0.9 mm) of two‐wave (3/11) and three‐wave (8/11) cycles. The maximal diameter of the CL2 (23.3 ± 1.9 mm) was reached approximately 6 days after hCG treatment and was correlated with its structural lifespan (p < 0.01). Cows that formed a CL2 after hCG had higher mean plasma P4 concentrations on Day 14 (+4.5 ng/ml) and Day 18 (+3.0 ng/ml) compared with cows without CL2 (p < 0.05). The structural regression of CL2 begun approximately 8 days after that of the CL1, and the median time at which the first drop in circulating P4 levels occurred was later in cows that formed a CL2 than in those that did not (Day 26 vs Day 18; p < 0.01). Thus, the induction of a CL2 by hCG on Day 12 might reduce the risk of premature luteolysis in high‐producing dairy cows after insemination.  相似文献

Strategic supplementation of P4 may be used to increase conception rates in cattle, but timing of supplementation in relation to ovulation, mass of supplementary P4 and formulation of the P4‐containing supplement has not been determined for beef cattle. Effects of supplementation of long‐acting progesterone (P4) on Days 2 or 3 post‐ovulation on development, function and regression of corpus luteum (CL) were studied in beef cattle. Cows were synchronized with an oestradiol/P4‐based protocol and treated with 150 or 300 mg of long‐acting P4 on Day 2 or 3 post‐ovulation (6–7 cows/group). Colour‐doppler ultrasound scanning and blood sample collection were performed from Day 2–21.5. Plasma P4 concentrations were greater (p < 0.05) from Day 2.5–5.5 in the Day 2‐treated groups and from Day 3.5–5.5 in the Day 3‐treated cows than in the control group. CL area and blood flow during Day 2–8.5 did not differ (p > 0.05) among groups, suggesting no effect of P4 treatment on luteal development. The frequency of cows that began luteolysis before Day 15 was greater (p < 0.04) in cows treated with 300 mg than in the controls, but there were no differences between non‐treated and 150 mg‐treated cows. The interval from pre‐treatment ovulation to functional and structural luteolysis was shorter (p < 0.01) in the combined P4‐treated groups than in the control cows. In conclusion, was showed for the first time that long‐acting P4 supplementation on Day 2 or 3 post‐ovulation increases P4 concentrations for ≥3 day, has no effect on luteal development, but anticipates the beginning of luteolysis in beef cattle.  相似文献

This study characterized the peripheral plasma placental lactogen (bPL) profile throughout gestation and examined the relationship between the stage of gestation, fetal mass, number, and postpartum lactation with circulating levels of bPL in Holstein cows after nonsurgical embryo transfer. Cows (n = 12) were divided into two groups: Group 1 = single embryo recipient cows (n = 5); Group 2 = twin embryo recipient cows (n = 7). Blood was collected about every third day from Day 0 (Day 0 was defined as the first day of standing estrus), then daily for the last 10 d of gestation, and sampling was stopped 1 d postpartum. The cows were milked twice daily at 0800 and 1800 hr. Two twin-embryo recipient cows had abnormal pregnancies; therefore, their data were excluded from that of the group and reported separately. The time trend concentrations of plasma bPL were significantly affected by the stage of gestation (P < 0.01) but not fetal number (P < 0.21). In both groups bPL levels remained low during the first two trimesters, then increased rapidly (P < 0.01) to peak concentrations between Days 200 and 220, and stabilized at this elevated level until parturition. Postpartum milk yields were indistinguishable between the singleton and twin-bearing cows. Calf birth weight and postpartum lactation were both correlated (P < 0.01) to peripheral bPL concentration in singleton cows, however, this relationship decreased with a subsequent increase in fetal number. Cows giving birth prematurely to stillborn calves or to a schistosomus reflexus calf exhibited a deviating bPL profile. These results indicate that peripheral bPL levels are positively associated with the stage of gestation but not with fetal number. Otherwise, the peripheral pattern of bPL is a valuable index for predicting feto-placental viability.  相似文献

Fifty multiparous cows, 22 Parda de Montaña (PA) and 28 Pirenaica (PI), were assigned to either suckling once‐daily for 30 min (RESTR) or ad libitum (ADLIB) from the day after calving. Blood samples were collected to analyse metabolites [non‐esterified fatty acids (NEFA), β‐hydroxybutyrate, total protein and urea)], insulin‐like growth factor‐I (IGF‐I) and progesterone (P4) at different intervals. Cows from RESTR maintained their live‐weight (LW) over the first 3 months post‐partum, whereas ADLIB cows lost nearly 4% LW. Both genotypes showed similar LW gains during this period (p > 0.10). Calf daily gains were lower in RESTR than in ADLIB treatment (p < 0.05), but similar across breeds (p > 0.10). Milk and lactose production were lower in RESTR cows than in ADLIB (p < 0.05). Milk and protein yield were greater in PA than in PI breed (p < 0.05). Serum NEFA, total protein and urea were higher in PI cows suckling ADLIB than in the rest (p < 0.05). Cows from PI breed had greater NEFA values than PA ones on the first week post‐partum (p < 0.001). Circulating IGF‐I was not affected by suckling frequency, breed nor their interaction (p > 0.10). Suckling frequency, but not breed, affected the interval from calving to first ovulation (p < 0.001), being shorter in RESTR than in ADLIB cows. In conclusion, the ad libitum suckling practice improved cow milk yield and offspring gain compared to once‐daily suckling for 30 min from the day after calving, at the expense of impairing the onset of cyclicity. The effect of calf management was confounded with breed on the studied blood biochemical constituents, but any of these metabolites influenced the role of endocrine IGF‐I in these genotypes.  相似文献

Thirty-four Holstein cows, 18 primiparous and 16 multiparous, were used to determine the effects of supplemental Cr on carbohydrate and lipid metabolism during the transition period. From approximately 3 wk prior to anticipated calving date until parturition, Cr-treated cows were given 51 mg Cr picolinate via a gelatin capsule bolus three times weekly, equivalent to an average daily intake of approximately 2 ppm Cr. Control cows were given empty gelatin capsules. All cows were fed total mixed rations, with one ration fed to dry cows, a second to lactating cows for wk 1 of postpartum (transition), and a third to lactating cows thereafter. Blood samples were collected via jugular venipuncture at 1, 2, and 3 wk prior to anticipated calving date as well as immediately following calving, 3 d post calving, and 1, 2, and 3 wk post calving. Samples were analyzed for glucose, insulin, and nonesterified fatty acids (NEFA). Within 1 wk prior to calving, a frequently sampled intravenous glucose tolerance test was performed, and blood plasma samples were analyzed for concentrations of glucose and insulin. Plasma glucose response to the frequently sampled intravenous glucose tolerance test was evaluated by calculating the half-life and fractional turnover rate for the period from 2 to 19 min after glucose infusion. Supplemental Cr did not affect circulating concentrations of glucose, insulin, or NEFA (P>0.1). Plasma NEFA concentrations were greater in primiparous cows (P<0.01) throughout the experiment, with the difference being most apparent during the 3 wk postpartum. Glucose concentrations reached a greater peak at time of calving in primiparous cows than in multiparous cows (P=0.07). There was an increase in glucose clearance rate (P=0.06) and a trend toward decreased glucose half-life (P<0.1) in primiparous cows supplemented with Cr. Insulin peak concentrations were unaffected by treatment or parity (P>0.1). Data suggest that Cr supplementation may alter carbohydrate metabolism in primiparous cows during the transition period.  相似文献

The effects of the rainy (RS) and the dry season (DS) on fertility, corpus luteum activity, and adrenal cortex response relationships were evaluated after first service (49 ± 6 d postpartum) in Carora cows, a dairy cattle of Venezuela raised in tropical conditions. Cows (n = 84 in RS and n = 98 in DS) were kept semistabled, had two or three calvings, body condition score 3.5 on a 5-point scale, and similar milk yield (2450 ± 560 kg of milk during the previous lactation). Cows were grouped retrospectively according to pregnancy status. A split-plot model with repeated measures over Days 5, 7, 10, 14, and 15 after insemination was used to analyze the effects of season, pregnancy status, and their interaction involving the day on: 1) serum concentrations of progesterone in four treatments: RS pregnant (n = 26), RS nonpregnant (n = 24), DS pregnant (n = 24), and DS nonpregnant (n = 20) cows; 2) serum concentrations of cortisol at Days 0, 10, 14, 15, and 16 postservice in the previous treatments (n = 9, 7, 6, and 8, respectively); and 3) concentrations of cortisol after 0.1 mg of adrenocorticotropin in these last four groups of cows at Day 14 postin-semination. Breeding during the DS decreased (P < 0.05) conception rate to first service and increased (P < 0.01) days in service. In addition, the DS decreased (P < 0.05) the percentages of cows with normal interestrous interval (20–22 d), expression of estrus, and (P < 0.01) luteal phase progesterone; but DS increased (P < 0.05) percentages of short and long estrous cycles, anovulatory estrus, and repeat breeding rate. Mean serum concentration of progesterone was lower (P < 0.05) at Days 10, 14, and 15 in DS nonpregnant than in DS pregnant cows, and lower during luteal phase (P < 0.05) in DS nonpregnant than RS nonpregnant cows. Serum cortisol concentration was greater (P < 0.05) at Days 10, 14, and 16 in DS nonpregnant than DS pregnant cows. A significant (P < 0.05) negative correlation (r = −0.78) between serum concentrations of progesterone and cortisol was found within DS nonpregnant cows. Concentrations of cortisol after adrenocorticotropin were greater (P < 0.05) in DS nonpregnant cows than in other groups. These results indicate that elevated concentrations of cortisol associated with the DS may decrease progesterone secreted by the corpus luteum and therefore mediate the negative effect of the DS on fertility.  相似文献

The aim of this study was to determine the effect of norgestomet treatment, in the absence or the presence of a functional corpus luteum (CL), on plasminogen activators activity (PAA) in the cervical mucus and the endometrium in dairy cows. Eleven days after oestrus (Day 0 = oestrus), 38 cows were randomly assigned to one untreated control group (n = 9) and three treatment groups (S(1), S(2) and S(3)). Animals of S(1) group (n = 9) received an implantation of norgestomet on the outer surface of the ear for 8 days, simultaneous injection of oestradiol valerate 5 mg and norgestomet 3 mg, i.m., and on Day 19 an injection of ECG 500 IU, i.m. Animals of S(2) group (n = 11) received the treatment of S(1) group, plus an administration of PGF(2)alpha on Day 10 for the regression of CL. Animals of S(3) group received the treatment of S(2) group, plus two additional norgestomet implants inserted on Day 16 for 36 h. Both types of plasminogen activators [the tissue-type (t-PA) and the urokinase-type (u-PA)] were detected in the cervical mucus and the endometrium of the cows. Plasminogen activators activity in the cervical mucus was higher in control group than in S(1), S(2) and S(3) groups (P < 0.001). In contrast, endometrial PAA did not differ among groups (P > 0.05). Oestradiol-17beta concentrations on Day 21 were higher in S(2) group than in control group (P < 0.01) and S(3) group (P < 0.05). Progesterone concentrations did not differ among groups (P > 0.05). Oestradiol-17beta concentrations could positively affect cervical mucus PAA in control group (P < 0.1), but not in other groups (P > 0.05). These findings suggest that control of estrous cycle by norgestomet administration, in dairy cows, exerts a suppressive effect on plasminogen activators synthesis and/or secretion in the cervical mucus, regardless of the absence or the presence of the CL. On the contrary, endometrial PAA is not affected by norgestomet treatment.  相似文献
