The effects ofSclerotinia sclerotiorum live and autoclaved sclerotia, and sclerotial exudates, and commercial oxalic acid were testedin vitro on sevenConiothyrium minitans strains differing in aggressiveness towardsS. sclerotiorum. Only sclerotial exudates and autoclaved sclerotia affected the mycelial growth rate of almost all the strains tested, whereas a change in theC. minitans mycelial growth pattern was observed in the presence of autoclaved sclerotia and live sclerotia germinating by the myceliogenic eruptive germination. In addition, sclerotial exudates had a stimulatory effect on spore germination. These findings indicate that the various treatments could influence theC. minitans strains regardless of their aggressiveness.  相似文献

The effect of the main construction factors and design features of greenhouses on the incidence of white rot disease, caused by Sclerotinia sclerotiorum (Lib.) de Bary, was studied in peppers grown under plastic-roofed greenhouses typical of Almería (SE Spain). At the height of the pepper-growing season in the province, fifty greenhouses were repeatedly sampled. Incidence of the disease was measured and the different construction features of the greenhouses were evaluated: types of roof cover, colour and age of the plastic cover, dimensions (ground plan, surface area, average height), estimated ventilation capacity and the presence of netting on the sides of the greenhouses (laterals). Statistical analyses were undertaken to see if the choice of various design options for each factor had any significant influence on the incidence of the disease. To achieve this single factor ANOVA was undertaken. The incidence of this disease was significantly affected (p<0.05) by the following greenhouse characteristics: ground plan, surface area, mean height, colour of the plastic cover, estimated ventilation and use of lateral netting.  相似文献

Studies were made of the variability ofSclerotinia sclerotiorum (Lib.) de Bary: (i) growth rate when cultivated on potato-dextrose agar (PDA); (ii) thein vitro resistance of this organism to the fungicides benomyl, vinclozolin and procymidone; and (iii) the size and number of sclerotia formed on PDA. The isolates used were collected from eggplants, French beans and tomatoes cultivated in the same locality, but although marked differences were observed among them, especially in growth rate and formation of sclerotia, it was not possible to establish groups or categories among the isolates studied, or to relate the observed variability to their origins. Temperature was a decisive factor for formation of sclerotia on PDA. No correlation was found between size and number of sclerotia, and growth rate. The ED50 level for both vinclozolin and procymidonein vitro was 2 mg l−1, whereas that for benomyl was below the minimum level used, and therefore could not be established.  相似文献

The effects of the mycoparasites Coniothyrium minitans and Trichoderma atroviride on the suppression of alfalfa blossom blight caused by Sclerotinia sclerotiorum were evaluated under indoor and field conditions. When T. atroviride (9·0 × 104 conidia/floret) + S. sclerotiorum (6·0 × 103 ascospores/floret) or C. minitans (9·0 × 104 conidia/floret) + S. sclerotiorum (6·0 × 103 ascospores/floret) were applied to detached young alfalfa florets, T. atroviride effectively inhibited saprophytic growth of S. sclerotiorum, whereas C. minitans showed no inhibition under the same conditions. When T. atroviride (6·9 × 104 conidia/floret) + S. sclerotiorum (6·0 × 103 ascospores/floret) or C. minitans (6·9 × 104 conidia/floret) + S. sclerotiorum (6·0 × 103 ascospores/floret) was applied to young alfalfa petals in vivo just after pollination, the percentage of pod formation was higher for T. atroviride+S. sclerotiorum than that for C. minitans+S. sclerotiorum, and the percentage of pod rot was lower for T. atroviride+S. sclerotiorum than that for C. minitans+S. sclerotiorum. However, when they were applied to senescent petals attached to developing pods of alfalfa at 9·2 × 104 conidia/floret together with S. sclerotiorum at 4·5 × 103 ascospores/floret at 14 days after pollination, C. minitans was more effective than T. atroviride in suppressing sclerotinia pod rot and seed rot of alfalfa. Field experiments showed that three applications of C. minitans (5·4 × 106 conidia mL−1) or T. atroviride (5·4 × 106 conidia mL−1) at a 7-day interval to blossoms of alfalfa effectively suppressed sclerotinia pod rot in two out of three annual trials. Coniothyrium minitans effectively suppressed sclerotinia seed rot in all three years, whereas T. atroviride was not effective against seed rot in any of the trial years. The efficacy of C. minitans was not significantly different (P > 0·05) from benomyl (250 µg ai mL−1). This study suggests that C. minitans has potential as a biocontrol agent to control blossom blight of alfalfa caused by S. sclerotiorum.  相似文献

In 2002, rotted flower clusters and blighted shoot tips and leaves were observed on highbush blueberry (Vaccinium corymbosum L.) and rabbiteye blueberry (V. ashei Reade) plants in Chiba, Japan. The causal fungus isolated from the diseased plants was morphologically identified as Sclerotinia sclerotiorum (Libert) de Bary. The fungus reproduced natural symptoms after inoculation, then reisolated from the symptomatic parts. This is the first report of blueberry sclerotinia rot caused by S. sclerotiorum.  本试验建立一种可同时检测黄瓜炭疽病(Colletotrichum orbiculare)、黄瓜菌核病(Sclerotinia sclerotiorum(Lib.)de Bary)和黄瓜细菌性萎蔫病(Erwinia tracheiphila)等黄瓜主要病害病原菌的三重PCR检测体系。采用正交试验设计方法, 对三重PCR的影响因素分析研究, 进行退火温度优化, 并以3个引物组、Taq DNA聚合酶、dNTP和Mg2+ 共6因素3水平进行多重PCR体系优化, 成功建立了适合黄瓜主要病害的三重PCR最佳检测体系, 即25 μL的反应体系中含有0.24 μmol·L-1 CY1/CY2;0.72 μmol·L-1 SSFWD/SSREV;0.336 μmol·L-1 ET-P1/ ET-P2;1 U Taq聚合酶;0.15 mmol·L-1 dNTP;1 mmol·L-1 MgCl2, 最适退火温度为63℃。该方法能够快速从田间黄瓜发病植株和根围土壤中将黄瓜炭疽病菌、黄瓜菌核病菌和黄瓜细菌性萎蔫病菌检测出来, 灵敏度可以达到10 pg·μL-1。  相似文献

陕西省核盘菌不同分离株对油菜的致病性   总被引:2,自引:0,他引:2  
采用带菌牙签活体茎秆穿刺接种法和离体叶片菌饼接种法,以3个品种的油菜为供试寄主,对陕西省5个地区10个县的油菜和其它5种寄主共495个核盘菌分离株进行了致病性测定。结果显示,两种接种方法接种3个不同油菜品种所反映的不同分离株的致病趋势一致。离体叶片菌饼接种蓉油12号除来自勉县和城固县的2个分离株不能致病外,其余493个分离株均能致病,但各分离株所致病斑直径差异很大,为0.5~42.6mm,其中病斑直径在10~40mm的分离株占94.7%,属于优势群体。同一地区甚至同一地块均存在不同致病力的分离株,不同寄主来源的分离株在油菜离体叶片上均能致病。说明核盘菌种内存在明显的致病性分化,这种分化与地理来源和寄主来源均无明显的相关性。  相似文献

为获得对紫花苜蓿Medicago sativa生长有益的生防菌,以尖孢镰刀菌Fusarium oxysporum为指示菌从紫花苜蓿根际土壤中分离筛选拮抗菌株,基于形态学特征、生理生化特性和16S rDNA与gyrB基因序列分析对其进行鉴定,并评价其对紫花苜蓿根腐病的生防效果以及提高寄主植物耐盐碱胁迫的能力。结果表明,从紫花苜蓿根际土壤中筛选到1株对尖孢镰刀菌有较强拮抗作用的菌株YB-2,该菌株对尖孢镰刀菌菌丝生长的抑制率为71.37%;结合形态学特征、生理生化特性和16S rDNA与gyrB基因序列分析结果将菌株YB-2鉴定为枯草芽胞杆菌Bacillus subtilis。菌株YB-2具有溶解无机磷的能力,且具有产氰化氢、羧甲基纤维素酶、1-氨基环丙烷-1-羧酸脱氨酶、吲哚乙酸和嗜铁素的能力,在NaCl含量为1%~9%、pH 7~9条件下均能生长。菌株YB-2对紫花苜蓿根腐病的相对防效达56.83%;在盐碱条件下,与对照相比,接种YB-2的紫花苜蓿株高极显著增加了17.82%,根长显著增加了28.22%,地上部鲜重和地上部干重分别显著增加了51.12%和48.57%。表明菌株YB-2兼...  相似文献

为明确金龟子绿僵菌Metarhizium anisopliae在促进植物生长和提高植物抗性方面的作用,以金龟子绿僵菌拌土种植紫花苜蓿Medicago sativa,于室内测定植株的生长参数、苜蓿斑蚜Therioaphis trifolii的取食选择性及其发育历期和繁殖力。结果表明,金龟子绿僵菌处理后7 d,植株株高比对照显著增加了0.87 cm;处理后14 d,植株分枝数为3.88个,根长为54.67 mm,均显著高于对照。株龄14 d的植株接入苜蓿斑蚜,至株龄28 d时,金龟子绿僵菌处理的植株株高、分枝数和根长均较对照显著增加;同时生物量、叶绿素含量和类胡萝卜素含量也有不同程度的升高。选择性试验结果显示,24 h内选择并驻留在金龟子绿僵菌处理植株叶片上的苜蓿斑蚜数量较对照显著减少28.57%。以金龟子绿僵菌处理的植株叶片饲喂苜蓿斑蚜,1~4龄若虫的发育均有所减缓,各龄若虫的发育历期均较对照叶片饲喂的各龄若虫有所延长,而成虫寿命较以对照叶片饲喂时显著缩短了30.09%,总生命历期显著缩短了18.83%。表明金龟子绿僵菌促进了紫花苜蓿植株的生长,同时增强了植株对苜蓿斑蚜的耐害性、排趋性与...  相似文献

本文评价了不同接种鉴定方法在油菜菌核病抗(耐)病性筛选上的应用。结果表明:花期离体或活体叶片菌丝体接种和限期接种法能显著区分不同品种(系)的抗感水平;苗期菌丝体悬浮液接种和试管苗接种法适于早期育种世代进行大规模筛选。对83份品种(系)鉴定结果,821、083等呈轻度感病,大部分属中感至高感范围。  相似文献

为探究自噬在核盘菌Sclerotinia sclerotiorum致病过程中的作用,利用酵母Saccharomyces自噬相关基因(autophagy-related gene,ATG)编码的蛋白序列比对核盘菌基因组,获得核盘菌假定ATG,并以核盘菌1980菌株为出发菌株,基于同源重组的原理对假定ATG进行敲除和回补,并测定不同突变体的生长表型和致病能力。结果表明,从核盘菌基因组中比对到2个ATG,分别命名为SsATG5SsATG8,两者在核盘菌致病过程中均上调表达。SsATG5SsATG8敲除突变体在菌丝生长、产草酸和侵染垫形成方面与野生型菌株无明显差异,但SsATG5敲除突变体在离体拟南芥Arabidopsis thaliana叶片上的致病力显著下降了约40%,在活体拟南芥植株上的致病力显著下降了约80%,同时SsATG5回补突变体恢复了正常的致病力。表明SsATG5参与了核盘菌的致病过程,证实自噬在核盘菌致病过程中发挥着重要作用。  相似文献

Sclerotinia stem rot (SSR) caused by Sclerotinia sclerotiorum (Lib.) De Bary is a serious fungal disease of soybean. Senescing petals provide a starting nutrient source for the invasion of healthy tissue by the advancing oxalic acid secreting fungal hyphae. Since oxalic acid is a major pathogenicity factor of SSR, transgenic soybean capable of degrading oxalic acid may be resistant to the pathogen. Transgenic soybean plants were produced byAgrobacterium -mediated transformation with the wheat germin gene (gf-2.8) encoding an oligomeric protein, oxalate oxidase (OxO), which oxidizes oxalic acid to carbon dioxide and hydrogen peroxide (H2O2). Transgenic soybean homozygous for 35S- gf-2.8 produced an approx. 130 kDa protein indistinguishable from wheat germin, and with OxO activity. OxO activity was prominent in cell walls proximal to the site of pathogen attack. The transgenics had greatly reduced disease progression and lesion length following cotyledon and stem inoculation with S. sclerotiorum indicating that the germin gene product conferred resistance to SSR. This is the first report of plant resistance to the fungal pathogen S. sclerotiorum in transgenic plants expressing OxO.  相似文献

The development of a polymerase chain reaction (PCR) assay for the detection of inoculum of the plant pathogenic fungus Sclerotinia sclerotiorum is described. The PCR primers were designed using nuclear ribosomal DNA internal transcribed spacer sequences. Specific detection of DNA from S. sclerotiorum was possible even in the presence of a 40-fold excess of DNA from the closely related fungus Botrytis cinerea. PCR products were obtained from suspensions of untreated S. sclerotiorum ascospores alone, but DNA purification was required for detection in the presence of large numbers of B. cinerea conidiospores. Specific detection of inoculum of S. sclerotiorum was possible in field-based air-samples, using a Burkard spore trap, and from inoculated oilseed rape petals. The assay has potential for incorporation into a risk management system for S. sclerotiorum in oilseed rape crops.  相似文献

Baseline sensitivity of Sclerotinia sclerotiorum, causal agent of stem rot of rapeseed, to a dicarboximide fungicide iprodione was determined using 50 isolates (historic population) collected in 2001 from the rapeseed fields without a previous history of dicarboximide usage. The 50% effective concentration (EC50) values to iprodione of these wild-type isolates ranged from 0.163 to 0.734 μg/ml with a mean of 0.428 μg/ml. In 2007 and 2008, 111 isolates (current population) were collected from rapeseed fields with 4–5 years of iprodione application. The EC50 values of these 111 isolates ranged from 0.117 to 0.634 μg/ml. The historic and current populations were not significantly (> 0.05) different in sensitivity to iprodione. The EC50 values of these 161 isolates to a carboxamide fungicide boscalid ranged from 0.002 to 0.391 μg/ml with a mean of 0.042 μg/ml. In the laboratory, three iprodione-resistant (IR) isolates HA17-IR, SZ31-IR, and SZ45-IR were induced from wild-type isolates HA17, SZ31, and SZ45, respectively. The EC50 values of the IR isolates were 200-fold higher than those of the original wild-type parents. The IR isolates showed an increase in osmotic sensitivity. The IR isolate HA17-IR lost its ability to produce sclerotia, and showed a significantly lower virulence on rapeseed leaves than its parent isolate HA17. In contrast, the IR isolate SZ31-IR had a significantly higher virulence than its wild-type parent SZ31. PCR assays showed that the partial two-component histidine kinase (os-1) gene, which is the putative target gene of iprodione, was deleted in the low virulent IR isolate HA17-IR. DNA sequence analysis showed that each of the other two IR isolates SZ31-IR and SZ45-IR had two point mutations in their partial os-1 genes. These results indicate that the mutations in os-1 gene may be associated with dicarboximide sensitivity, sclerotial development, and virulence in S. sclerotiorum.  相似文献

Ninety-six isolates of sunflower Sclerotinia sclerotiorum (Lib.) de Bary from Inner Mongolia (IM) in China, from Canada and the United Kingdom (UK) were sampled to investigate the genetic diversity and structure using Sequence-Related Amplified Polymorphism. A total of 123 polymorphic bands were obtained, ranging in size from 100 to 500 base pairs. The five populations of S. sclerotiorum isolated from the three countries showed various levels of genetic variability. The percentage of polymorphic loci varied from 30.89% in the UK population to 97.56% in the Middle IM population. The values of Shannon index (i) varied from 0.1876 in the UK population to 0.5301 in the West IM population. The heterozygosity of the five geographic populations obtained by estimating allele frequency varied from 12.91% in the UK population to 35.44% in the West IM population. The genetic identity, as indicated by the Nei unbiased identity index, ranged from 0.9744 between populations from Canada and East IM to 0.6477 between populations from West IM and UK. UPGMA cluster analysis using Nei’s genetic distance gave distances ranging from 0.0259 to 0.4343. The rates of gene flow among five geographic populations ranged from 1.5406 between West IM and UK populations to 18.4149 between West IM and Middle IM populations. The four populations from West IM, Middle IM, East IM and Canada were clustered into one subgroup in which the isolates from West and Middle IM belonged to one population, whereas those from East IM and Canada essentially were another population. 采用菌丝生长速率法测定了四霉素对采自山东省不同地区不同蔬菜作物的151株菌核病菌的毒力作用,同时比较了其对蔬菜菌核病菌不同生育阶段的抑制活性,并通过离体叶片法评价了四霉素对蔬菜菌核病的防治效果。结果表明:菌核病菌对四霉素比较敏感,敏感性频率呈单峰正态分布,151株病菌菌丝生长的平均EC50值为 (0.29 ± 0.01) μg/mL,该值可作为蔬菜菌核病菌对四霉素的敏感基线。此外,经四霉素处理后,该病菌的菌核数量以及干重明显降低,菌核明显变小;2 μg/mL的处理对菌核萌发的抑制率达到100.00%。 离体黄瓜叶片接种试验表明,四霉素对菌核病具有较好的保护和治疗效果,且保护作用较为显著。在质量浓度为20 μg/mL时,四霉素对该病的防效显著高于对照药剂多菌灵和异菌脲。因此,四霉素具有防治蔬菜菌核病的潜在价值,可进一步通过田间试验验证其应用效果。  相似文献

A study was conducted to investigate production of antifungal substances (AFS) by Coniothyrium minitans (Cm), a mycoparasite of Sclerotinia sclerotiorum (Ss), in modified Czapek-Dox (MCD) broth and potato dextrose broth (PDB), and effects of AFS of Cm on mycelial growth and germination of sclerotia and ascospores of Ss and incidence of leaf blight of oilseed rape caused by Ss. Results showed that mycelial growth of Ss was reduced by 41.6 and 84.5% on 3 day-old cultures grown on potato dextrose agar (PDA) amended with 10% (v v−1) of cultural filtrates of Cm grown in MCD (MCDcm) after incubation for 6 and 15 days, respectively, and by 2.7 and 15.7% on PDA amended with 10% (v v−1) of cultural filtrates of Cm grown in PDB for 6 and 15 days, respectively. In addition to retardation of mycelial growth, morphological abnormality of Ss such as hyphal swellings and cytoplasm granulation were also observed in colonies grown on PDA amended with cultural filtrates of MCDcm. Sclerotia of Ss soaked in the filtrates of MCDcm for 24 h remained viable, but their ability to undergo myceliogenic germination on PDA was delayed, compared to sclerotia treated with MCD. Germination of ascospores of Ss was unaffected on PDA amended with 10% of the filtrates of MCDcm. However, germ tubes of Ss were shortened and deformed by the formation of hyphal swellings in the treatment of MCDcm. Treatment of leaves of oilseed rape with cultural filtrates of MCDcm reduced incidence of leaf blight caused by Ss, compared to the controls (water or MCD). This study suggests that AFS produced by Cm plays an important role in the suppression of mycelial growth and germ-tube development of ascospores of Ss and that there is potential for using AFS of Cm to control leaf blight of oilseed rape caused by ascospores of Ss.  相似文献

采用硅胶室温干燥法对陇东紫花苜蓿(Medicago sativa L. cv. Longdong)种子进行超干处理,使其含水量由9.03%(CK)分别降至7.09%~4.59%共八个不同含水量水平,用铝箔纸密封,常温下贮藏1年后,采用沙培盆栽试验,研究种子超干贮藏对苜蓿生长和生理的影响。结果表明,超干贮藏后,紫花苜蓿种子的出苗数、植株的株高比CK显著提高,MDA含量显著降低。播种第30天时,含水量为6.36%~4.59%处理种子的出苗数较CK提高25%~50%;播种第60天时,种子含水量为6.36%~5.46%处理的植株株高较CK提高11.02%~15.68%,各超干处理种子的植株MDA含量仅为CK的44.92%~70.29%;所有处理植株的根体积、地上干重、地下干重和可溶性糖含量仅为CK的55.36%~89.29%、63.06%~88.05%、50.71%~76.69%和63.35%~97.86%;除4.59%含水量处理的叶片数和5.18%含水量处理的根瘤数与CK比差异不显著,其它各处理的单株叶片数与根瘤数均显著低于CK,其中各超干处理的根瘤数仅为CK的63.00%~92.56%。说明超干贮藏能够提高苜蓿种子的出苗数、植株的株高和耐受性,但降低了植株的生物量及根体积,并影响了叶片生长及根瘤的形成。  相似文献

A study was conducted to determine the feasibility of using sclerotia ofSclerotinia sclerotiorum for producing conidia ofConiothyrium minitans in liquid culture. The medium (SST) was made of water containing 2.0, 1.5, 1.0 or 0.5% (w/v) ground sclerotia ofS. sclerotiorum and 100 μgl −1 thiamine hydrochloride (HCl). One ml of conidial suspension (2 × 107 conidia ml−1) ofC. minitans LRC 2534 was inoculated into 100 ml of SST medium or control (thiamine HCl in water) and incubated at 20 ± 2°C on a shaker at 200 rpm. Subsamples were removed periodically and examined under a compound microscope. Conidia in the SST media germinated within 24 h, developed into branched hyphae within 48 h, produced pycnidia after 3–4 days, and the pycnidia released mature conidia after 7 days. Production of conidia varied with the concentration of sclerotia in the SST medium. It was lower (3.6 × 106 conidia ml−1) at 0.5% but higher (1.2 × 108 conidia ml−1) at 2%. The new conidia were viable and the colonies developing from them showed the original morphological characteristics. It was concluded that using SST liquid medium as a substrate for mass production of conidia ofC. minitans has potential for use in commercial development of this mycoparasite as a biocontrol product. http:www.phytoparasitica.org posting Jan. 23, 2007.  相似文献
