AIM: To conduct a longitudinal serological survey for evidence of Brucella spp and Leptospira spp infection of pre-weaned New Zealand fur seals in a colony on the Otago Peninsula.

METHODS: Seal pups were repeatedly captured on a monthly basis from February through to July 2001. Pups were tagged at first capture and a blood sample was taken at each capture event. A total of 163 sera were collected from 118 seal pups. Where sufficient volume was collected, the sera were tested for leptospirosis using the microscopic agglutination test (MAT), and for brucellosis using the competitive enzyme-linked immunosorbent assay (ELISA) for Brucella abortus.

RESULTS: None of 128 sera from 101 seals tested positive to the ELISA for B. abortus. All tests for Leptospira interrogans serovars Grippotyphosa, Copenhageni, Bratislava and Leptospira borgpetersenii serovar Ballum were negative at a cut-off of <1/100 dilution. Positive or suspicious titres were found to L. interrogans serovars Canicola and Pomona and L. borgpetersenii serovar Hardjo. The highest titres (12,800) were found to serovar Pomona. The titre to serovar Pomona in one seal rose from <1/50 in March to 12,800 in April and was <1/50 when re-sampled in July. The titre to serovar Pomona in another seal dropped from 12,800 in May to <1/50 in June. These seals also had titres to serovar Hardjo, which rose or fell in the same manner. All suspicious or positive titres occurred in late April and early May, when the pups were approximately 4–5 months old. In June and July, all seals tested were negative.

CONCLUSIONS: There was no serological evidence of Brucella infection in the pre-weaned fur seals at the colony. Positive titres to serovars Pomona, Hardjo, or Canicola suggest that a Leptospira species was present at the colony, however isolation or visualisation of the organism is required to confirm this. Care should be exercised when handling New Zealand fur seals to prevent human infection or inadvertent transfer of leptospirosis to another marine mammal species.  相似文献

Ophthalmic examination under anesthesia of a 5-yr-old, captive, male New Zealand fur seal (Arctocephalus forsteri) with reduced visual capacity revealed bilateral cataracts. Surgical extraction of the lenses was performed by phacofragmentation. Preoperative mydriasis was unsuccessful using common topical mydriatic agents. Intraoperative intracameral epinephrine 1:10,000 produced minimal pupillary dilation, but excellent dilation was achieved with intracameral epinephrine at 1:1,000. Postoperative uveitis was controlled with oral prednisolone. Postoperative complications included generalized corneal edema persisting for approximately 1 wk and corneal ulceration caused by the lateral canthotomy sutures. Mortalities and abortions associated with starvation occurred at Cape Cross, Namibia, in Cape fur seals (Arctocephalus pusillus pusillus). Affected seals showed lethargy and emaciation, and the most common pathological signs were those of a respiratory infection, both in adults and offspring. Streptococcus phocae was isolated from adult seals, a cub and aborted foetuses.  相似文献

Abstract

AIM: To provide baseline data on the levels and patterns of antibacterial drug resistance expressed by Gram-negative bacteria isolated from poultry carcasses in New Zealand.

METHODS: Between July and December 2006, isolates of Escherichia coli (n=407) and Salmonella spp. (n=3) originating from carcass-rinse samples were submitted by testing laboratories affiliated to five major poultry processing plants. Isolates of Campylobacter jejuni (n=193) originating from retail poultry carcasses in 2005–2006 were retrieved from the Massey University archives. All isolates underwent disc diffusion susceptibility testing against panels of 12 (Enterobacteriaceae) and six (Campylobacter spp.) antibacterial drugs. Cephalothin-resistance in isolates of E. coli was confirmed using ETest strips, and confirmation of the resistance phenotypes for a subset of C. jejuni isolates used microbroth dilution assays. Patterns within the resistance phenotypes of the isolates were investigated using hierarchical clustering, and logistic regression modelling.

RESULTS: The majority of isolates (71.5% E. coli, 99% C. jejuni, and all three Salmonella spp. isolates) were fully susceptible to the drugs that were tested. Four (1%) E. coli isolates showed resistance to three or more drugs. The proportions of susceptible E. coli differed between the five processing plants. Resistances were detected in E. coli isolates, using disc diffusion to cephalothin (18.2%), ampicillin (4.4%), tetracycline (4.4%) and gentamicin (1.5%). There was an association between cephalothin-resistant isolates of E. coli and decreased susceptibility to gentamicin. Using ETests to ascertain the minimum inhibitory concentrations (MIC) of E. coli for cephalothin gave inconsistent results. One of 193 C. jejuni isolates was resistant to erythromycin, and microbroth dilution assays confirmed that this panel of C. jejuni was generally susceptible to antibacterial drugs.

CONCLUSIONS: The levels of resistance shown by Gram-negative bacteria isolated from chicken carcasses in New Zealand are among the lowest reported around the world. No resistance to extended-spectrum cephalosporin drugs was detected in E. coli, suggesting that CTX-M and AmpC beta-lactamases are rare or absent. Salmonella spp. are rarely isolated from poultry carcasses during routine testing in New Zealand, and the isolates identified during this study were fully susceptible to the drugs tested. A panel of C. jejuni isolates originating from retail poultry carcasses were susceptible to first-line and second-line antibacterial drugs. The use of cephalothin as a marker of resistance to first-generation cephalosporins may not be appropriate for non-type-specific E. coli of animal origin.  相似文献

Intermediate hosts of Fasciola hepatica in New Zealand are the native species Lymnaea tomentosa, and introduced species, L. columella and L. truncatula. The latter is of little significance in New Zealand. Fascioliasis has become more widespread since 1950 following colonisation of much of the North Island by L. columella. L. tomentosa can produce eggs at 5°C and L. columella at 2°C. L. columella produces about the three times as many eggs as L. tomentosa and the eggs of the former develop more rapidly and over a wider range of temperatures. Both species inhabit ponds and marshes, although L. columella is better suited to pond habitats. Neither species will survive in marsh habitats that dry out in summer. Both species tolerate wide ranges of Ca and Mg concentrations and pH. Marsh habitats of the two species are indistinguishable though there is evidence of subtle differences in habitat preferences. Snail populations are minimal in winter and fluctuate irregularly throughout the rest of the year. In general, L. tomentosa populations contain proportionally more mature snails than those of L. columella. The epidemiology of fascioliasis in New Zealand is discussed. The absence of outbreaks of acute disease, the multiplicity of snail hosts and variations in climate pose problems for investigators. Overwintering infections of snails have been observed in L. tomentosa, but not in L. columella. With the latter host, transmission of Fasciola occurred over 6–7 months from mid-summer to mid-winter.The rate of infection of tracer sheep over 5 years varied but was not related to temperature or increased summer rainfall. Infection rates tended to increase when summers were dry and animals forced to graze the snails habitats. Rates in infection were not related to snail densities but to the numbers of mature snails on habitats. The infection rate of L. columella was extremely low (1790 snails examined). Sheep appear to be more important than cattle in maintaining Fasciolainfections from year to year. The control of fascioliasis is discussed briefly. It is argued that prevention of infection is preferable to removing infections already acquired, and low this might be achieved by a strategic anthelmintic treatment when L. columella is involved is described. Further studies on the prevalence and epidemiology of fascioliasis in New Zealand are required.  相似文献

Anthelmintic resistance was first confirmed in New Zealand in 1979 and since then has become common-place; more than 50 % of sheep farms now have detectable levels of resistance to one or more chemical classes of anthelmintic. Farmer drenching practices have changed little over the last 15–20 years and are clearly exerting a significant level of selection for resistance. In the absence of new chemical classes of anthelmintics, current parasite control practices will be unsustainable in the long-term. Once substantial resistance has developed, significant reversion to susceptibility is unlikely and re-introduction of failed drugs is likely to result in the rapid re-emergence of control problems. The number of anthelmintic treatments applied is not necessarily a reliable indicator of selection pressure and should not be the only factor considered in strategies for minimising the development of resistance. The relative potential of the different anthelmintics now available, particularly the long- acting products, to select for resistance varies with the way they are used and with other epidemiological and management factors; generalisations about their respective roles in the development of resistance are often unreliable. In many cases, literal extrapolation of recommendations for the management of resistance from Australia to New Zealand is unsupportable, given the differences in climate, parasite ecology and farming practices between the 2 countries. In the absence of a refuge for susceptible genotypes, as occurs when anthelmintic treatments are used as a means of generating low-contamination ‘safe’ pasture for young stock, the rapid development of resistance is likely. Anthelmintic treatments applied to animals with a high level of immunity, or which become immune while the anthelmintic is active, are likely to select for resistance faster than treatments applied to non-immune stock.  相似文献

In grasses and clovers the highest concentrations of selenium were found in the leaves. The selenium content of different species in spring pastures on the same soil type did not vary widely although the slower growing and more deeply rooting species contained higher levels. The concentration of selenium in a pasture varied with time of the year, the lower levels being usually associated with conditions that stimulated rapid growth of the herbage, although factors other than growth rate were also involved. Lucerne appeared to be unusual in that in some areas it accumulated much higher levels of selenium than was found in other associated species. A very wide variation in levels of selenium in pastures growing on different soils has been observed (3-1500 ng/g).

Selenium-responsive unthriftiness in grazing lambs occurred where the selenium content of spring pastures was <20 ng/g. Pastures containing >30 ng/g were apparently adequate while intermediate levels were probably marginally deficient.  相似文献
