Tritrichomonas mobilensis is an intestinal parasite of squirrel monkeys. There are few reports concerning the morphological aspects of this parasite. In addition, the taxonomic relationship between T. mobilensis and Tritrichomonas foetus, a serious pathogen that causes bovine and feline trichomonosis, has been questioned. For this reason, in the present study, we examined and compared both tritrichomonads with regard to their morphology, ultrastructure, endocytic activity and cytotoxicity when in the presence of host cells. Electron microscopy demonstrated consistent morphological differences between the hydrogenosomes of both parasites. Moreover, T. mobilensis and T. foetus had striking differences in their endocytic behavior. Thus, this work provides additional data that support the hypothesis that T. mobilensis is a distinct species from T. foetus.  相似文献

OBJECTIVE: To determine whether infection with Tritrichomonas foetus causes diarrhea in specific-pathogen-free or Cryptosporidium coinfected cats. ANIMALS: 4 cats with subclinical cryptosporidiosis (group 1) and 4 specific-pathogen-free cats (group 2). PROCEDURE: Cats were infected orogastrically with an axenic culture of T. foetus isolated from a kitten with diarrhea. Direct microscopy and protozoal culture of feces, fecal character, serial colonic mucosal biopsy specimens, and response to treatment with nitazoxanide (NTZ; group 1) or prednisolone (groups 1 and 2) were assessed. RESULTS: Infection with T. foetus persisted in all cats for the entire 203-day study and resulted in diarrhea that resolved after 7 weeks. Group-1 cats had an earlier onset, more severe diarrhea, and increased number of trichomonads on direct fecal examination, compared with group-2 cats. Use of NTZ eliminated shedding of T. foetus and Cryptosporidium oocysts, but diarrhea consisting of trichomonad-containing feces recurred when treatment was discontinued. Prednisolone did not have an effect on infection with T. foetus but resulted in reappearance of Cryptosporidium oocysts in the feces of 2 of 4 cats. During necropsy, T. foetus was isolated from contents of the ileum, cecum, and colon. Tritrichomonas foetus organisms and antigen were detected on surface epithelia and within superficial detritus of the cecal and colonic mucosa. CONCLUSIONS AND CLINICAL RELEVANCE: After experimental inoculation in cats, T. foetus organisms colonize the ileum, cecum, and colon, reside in close contact with the epithelium, and are associated with transient diarrhea that is exacerbated by coexisting cryptosporidiosis but not treatment with prednisolone.  相似文献

OBJECTIVE: To determine the long-term outcome of cats infected with Tritrichomonas foetus and identify treatment and management strategies influencing resolution of infection or associated diarrhea. DESIGN: Prospective study. SAMPLE POPULATION: 26 cats with T. foetus-associated diarrhea at least 22 months prior to the study. PROCEDURE: A standardized survey regarding clinical course and management was administered to owners of cats with T. foetus infection and associated diarrhea. Fecal samples were obtained from each cat; the presence of T. foetus was assessed via microscopic examination of smears, culture in commercial media, and polymerase chain reaction amplification of T. foetus rDNA involving species-specific primers. RESULTS: Survey responses were obtained from owners of all 26 cats. Twenty-three cats had complete resolution of diarrhea a median of 9 months after onset. Analysis of fecal samples obtained from 22 cats revealed persistent T. foetus infection in 12, with a median of 39 months after resolution of diarrhea. History of implementation of a dietary change, treatment with paromomycin, or higher numbers of cats in the household was associated with significantly longer duration of time to resolution of diarrhea. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested chronic T. foetus-associated diarrhea in most cats is likely to resolve spontaneously within 2 years of onset. Chronic infection with T. foetus (without clinical signs) after resolution of diarrhea appears to be common. Although often temporarily effective in decreasing severity of diarrhea, attempts to treat cats with T. foetus infection may result in prolongation of time to resolution of diarrhea.  相似文献

A 10-week-old female Ocicat was presented at a primary care feline veterinary practice for failure to thrive and diarrhoea. Numerous trophozoites, atypical for Giardia sp., were detected on a direct faecal examination, in addition to Giardia cysts. Although the failure to thrive and diarrhoea resolved following treatment for giardiasis, further diagnostic tests performed on faecal specimens from the kitten and 15 other Ocicats from the same cattery, including culture of trophozoites in In Pouch medium, PCR testing and molecular sequencing of PCR amplicons, confirmed infection with Tritrichomonas cf. foetus. This is the first report in Australia of feline trichomoniasis, which appears to be an emerging infectious disease of cats. Pertinent information regarding the clinical features, diagnosis, therapy, and potential source of feline trichomoniasis within Australia are discussed.  相似文献

Scanning electron microscopy was used to examine canine tracheal cartilage and to determine the relationship between the fibrous and amorphous matrix in this tissue. Collapsed tracheal (CT) cartilage was hypocellular, compared with normal tracheal cartilage. The amorphous matrix of CT cartilage had a porous, fissured texture and did not have the homogeneous appearance of normal tracheal cartilage. Capillaries were seen to pass through CT cartilage. Randomly distributed connective tissue fibers, evident in CT cartilage matrix, were frequently attached to irregular shapes and sizes of amorphous matrix.  相似文献

Faecal samples from 74 cats with chronic large bowel diarrhoea that were living in a rescue colony in Italy, were submitted for assessment of Tritrichomonas foetus by direct microscopy of faecal smears (n=20) and T foetus-specific culture (n=74), with confirmation by polymerase chain reaction (n=1). Twenty-four of the cats (32%) were found to be infected with T foetus. The infected cats were predominantly over a year of age (67%) and were all neutered non-pedigree domestic cats.  相似文献

Immunosorption has become a very important biochemical and serological tool and it is the purpose of this paper to visualize this process qualitatively using the scanning electron microscope. Different carriers (i.e. CNBr activated cellulose and Sepharose, glutaraldehyde treated acrylamide-agarose beads Magnogel, and polystyrene cover slips) were coated with different antibodies and incubated with their homologous antigens such as pneumococci, ferritin, polymeric Salmonella flagellin and mechanically detached flagella, as well as tetanus toxoid, E. coli bacteriophage T4 and Rota virus particles. OBJECTIVE: To determine the efficacy of tinidazole for treatment of cats with experimentally induced Tritrichomonas foetus infection. ANIMALS: 8 specific-pathogen-free kittens. PROCEDURES: Tinidazole was tested for activity against a feline isolate of T foetus in vitro. Kittens were infected orogastrically with the same isolate and treated or not with tinidazole (30 mg/kg, PO, q 24 h for 14 days). Amoxicillin was administered 28 weeks after completion of tinidazole administration to induce diarrhea. Feces were repeatedly tested for T foetus by use of PCR assay and microbial culture for 33 weeks. RESULTS: Tinidazole killed T foetus at concentrations >or= 10 microg/mL in vitro. In experimentally induced infection, tinidazole administered at 30 mg/kg decreased T foetus below the limit of molecular detection in 2 of 4 cats. Recrudescent shedding of T foetus, as elicited by amoxicillin-induced diarrhea, was diminished in cats that received prior treatment with tinidazole. CONCLUSIONS AND CLINICAL RELEVANCE: Although tinidazole decreased the detection of T foetus and treated cats were resistant to later efforts to incite the infection, inability of tinidazole to eradicate infection in many cats poses a serious impediment to the drug's effectiveness in practice.  相似文献

Almost 20 years has passed since trichomonosis was first recognized as a potential cause of diarrhea in domestic cats. Despite progress in confirming disease causation, developing means for diagnosis, and identifying approaches to treatment of the infection, we still know very little about how this parasite causes diarrhea. With increasing recognition of resistance of trichomonosis to treatment with 5‐nitroimidazole drugs, new treatment strategies based on an understanding of disease pathogenesis are needed. In this review, lessons learned from the pathogenesis of venereal trichomonosis in people and cattle are applied to clinical observations of trichomonosis in cats in effort to generate insight into areas where further research may be beneficial.  相似文献

Background

In recent years, the protozoan Tritrichomonas foetus has been recognised as an important cause of chronic large-bowel diarrhoea in purebred cats in many countries, including Norway. The aim of this cross-sectional study was to determine the proportion of animals with T. foetus infection among clinically healthy cats in Norway and to assess different risk factors for T. foetus infection, such as age, sex, former history of gastrointestinal symptoms and concurrent infections with Giardia duodenalis and Cryptosporidium sp.

Methods

The sample population consisted of 52 cats participating in three cat shows in Norway in 2009. Samples were examined for motile T. foetus by microscopy, after culturing and for T. foetus-DNA by species-specific nested PCR, as well as for Giardia cysts and Cryptosporidium oocysts by immunofluorescent antibody test (IFAT).

Results

By PCR, T. foetus-DNA was demonstrated in the faeces of 11 (21%) of the 52 cats tested. DNA-sequencing of five positive samples yielded 100% identity with previous isolates of T. foetus from cats. Only one sample was positive for T. foetus by microscopy. By IFAT, four samples were positive for Giardia cysts and one for Cryptosporidium oocysts, none of which was co-infected with T. foetus. No significant associations were found between the presence of T. foetus and the various risk factors examined.

Conclusions

T. foetus was found to be a common parasite in clinically healthy cats in Norway.  相似文献

Tritrichomonas foetus is the causative agent of sexually transmitted trichomoniasis in cattle. In females, the infection can be associated with infertility, vaginitis, endometritis, abortion or pyometra, leading to significant economic losses in cattle raising. T. foetus is devoid of the ability to synthesize purine nucleotides de novo, depending instead on salvaging purines from the host environment. Ecto-5'-nucleotidase catalyzes the final step of extracellular nucleotide degradation, the hydrolysis of nucleoside 5'-monophosphates to the corresponding nucleosides and Pi. In this work we show that living, intact cells of T. foetus were able to hydrolyze 5'AMP at a rate of 12.57 ± 1.23 nmol Pi × h(-1) × 10(-7) cells at pH 7.2 and the 5'AMP hydrolysis is due to a plasma membrane-bound ecto-enzyme activity. The apparent K(m) for 5'AMP was 0.49 ± 0.06 mM. In addition to 5'AMP, the enzyme hydrolyzed all substrate monophosphates tested except 3'AMP. No divalent metals or metal chelators were able to modulate enzyme activity. Phosphatase inhibitors did not have an effect on ecto-5'-nucleotidase activity while ammonium molybdate did inhibit the activity in a dose dependent manner. The presence of adenosine in the culture medium negatively modulated the enzyme. These results indicate the existence of an ecto-5'-nucleotidase that may play a role in the salvage of purines.  相似文献

To test the efficacy of a polyvalent Tritrichomonas foetus vaccine, 130 nulliparous heifers were randomly assigned to either receive the test T foetus vaccine or to serve as nonvaccinated controls. The polyvalent test vaccine consisted of a Campylobacter fetus/Leptospira canicola-grippotyphosa-hardjo-icterohaemorrhagiae-pamona bacterine containing 5 x 10(7) killed T foetus/dose. The polyvalent control vaccine consisted of the aforementioned formulation without T foetus. Heifers were administered 2 doses of control or experimental vaccine at 3-week intervals. Heifers were bred to T foetus-infected bulls and their conception and pregnancy rates were determined throughout gestation. In addition, serum samples were analyzed to determine induced concentrations of antitrichomonal antibodies and vaginal secretions were sampled to determine T foetus infection rates in control and vaccinated animals. One week after each of the 15-day breeding periods, 60% (6 of 10) of tested vaccinates and 80% (8 of 10) of tested control animals were T foetus culture-positive. The mean duration of infection of vaccinates was 3.8 weeks (+/- 7.5 days), compared with 5.4 weeks (+/- 7.5 days) of infection for control heifers. All vaccinates developed increased immunofluorescence and serum neutralizing antibody titers following the first immunization, and had additional increases of at least fourfold in response to the second injection. In contrast, no consistent increase in immunofluorescence or serum neutralizing antibodies was observed in control animals. Faecal samples from 111 cats with diarrhoea that were living in the UK were submitted for the assessment of Tritrichomonas foetus infection by polymerase chain reaction (PCR). Sixteen (14.4%) samples were found to be positive. In agreement with studies from the USA, infected cats were predominantly of a year of age or less and of a pedigree breed, with Siamese and Bengal cats specifically over-represented in this population.  相似文献

Morphologic features of the canine atrioventricular (AV) node were evaluated, using histochemical cholinesterase reactions and scanning electron microscopy. Three distinct regions of the AV node were observed: the transitional zone, superficial AV node, and deep AV node. The transitional zone lacked distinct cellular arrangement, and the cells were large and round with extensive branching on the ends. Superficial AV nodal cells were elongated, tightly packed, and smaller than were transitional cells. The superficial AV node was the densest zone of the AV node. Cell-to-cell contact was end-to-end. Deep AV nodal cells were long, formed laminated fascicles, were larger than the superficial AV nodal cells, and were continuous with the AV bundle.  相似文献

A photometrically-measured indirect fluorescent antibody (IFA) test was developed to measure antibody levels in three heifers vaccinated intramuscularly with two injections (two-week interval) of a vaccine containing formalin-killed Tritrichomonas foetus in oil adjuvant; a separate animal served as a nonvaccinated control. Sera were collected weekly and tested for specific IgG against T. foetus. Antibodies were detectable within two weeks of the initial injection and reached reciprocal titers as high as 7700 as estimated from previously tested reference sera. Titers peaked at six to eight weeks and remained at relatively high levels for the eleven-week study period. The fluorometric assay was easily developed and economical to perform; in addition, it more accurately estimated IgG levels than standard slide IFA tests.  相似文献

Tritrichomonas foetus is a very intriguing trichomonad protozoan with respect to its varied choice of residence in the different host species. It is an obligate parasite of the reproductive and the gastrointestinal tract of bovine and feline host respectively, leading to trichomonosis. Bovine trichomonosis is a sexually transmitted disease whereas feline trichomonosis is a disease with a purported fecal-oral route of spread. Further, the trichomonad is a commensal in the nasal passages, stomach, cecum and colon of swine host. Advances have been exponential in understanding the trichomonad biology and specifically feline trichomonosis since late 1990s and early 2000s when T. foetus was soundly determined to be a causative agent of chronic diarrhea in the domestic cat. It is a challenging task, even for a skilled investigator not to mention the busy clinical veterinarian, to keep up with the vast volume of information. Here we comprehensively reviewed the trichomonad biology, clinical manifestations, pathogenesis, host immunity, world map of distribution, risk factors, diagnosis and treatment. Risk factors associated with T. foetus-positive status in the domestic cat include young age, purebred, history of diarrhea, co-infections with other enteral pathogens. In addition, molecular similarity of bovine and feline isolates of T. foetus in DNA sequence was concisely discussed. The data presented serve as an information source for veterinarians, and investigators who are interested in biology of T. foetus and feline trichomonosis.  相似文献

The aim of the present study was to evaluate possible injury provoked by an intensive reproductive programme based on rectal palpation in the cow. Alterations to the mesothelial surface of the peritoneum and to the oviductal mucosa were explored by scanning electron microscopy (SEM). Nine multiparous Friesian cows were selected from each of two commercial dairy herds. Cows in herd 1 were maintained on a weekly reproductive health programme, while those in herd 2 were not subjected to gynaecological exploration. Surface features of the mesothelial cells of the reproductive tract and adjacent ligaments, and the oviductal mucosa were explored by SEM observation of tissue specimens from each animal. Six cows in the intensive reproductive programme showed microscopic alterations of the peritoneal surface. Strands were observed in the oviducts and adjacent ligaments in five of these cows. These strands, composed of filaments covered with mesothelial cells, were probably adhesions. Cells with abundant vacuoles on their surface were detected in the mesothelium corresponding to three animals, two of which also showed microscopic strands. No peritoneal surface alterations were observed in the cows from herd 2. No oviductal mucosal modifications were detected in either group. These findings suggest that intensive reproductive programmes based on rectal palpation may increase the risk of peritoneal surface alterations in the cow, but effects on reproductive physiology are unlikely.  相似文献
