Serum samples collected from breeder chickens ranging in age from 1 day to 55 weeks were tested for CAA antibodies by enzyme-linked immunosorbent assay (ELISA) and indirect immunofluorescent antibody (IFA) test. The relationship of ELISA to IFA test was determined. The sensitivity of the ELISA relative to the IFA test was 82.64%, and the specificity of the ELISA relative to the IFA test was 56.25%. Agreement between the ELISA and the IFA test was highly significant (Kappa = 0.74, Z = 5.78). We concluded that the ELISA is as good as the IFA test for detecting CAA antibody in sera from chickens.  相似文献

Blood from 48 chicks was examined for anemia (packed cell volume), and plasma was examined for virus particles by direct transmission electron microscopy (DTEM). There was agreement between the occurrence of anemia and the presence of CAA virus particles in plasma from anemic chicks (Kappa = 0.2425, Z = 2.096, P = 0.036). Although DTEM is a method that can be used to diagnose CAA in chicks, more sensitive, economical and less laborious diagnostic assays are needed.  相似文献

Pale chicks with necrotic dermatitis, small bursas of Fabricius (BFs), small thymuses, pale bone marrow, and watery blood were suspected of having parvovirus-like virus- (PVLV) associated disease. Histologic lesions included atrophy or hypoplasia of thymuses and BFs, and septic necrotizing clostridial dermatitis and hepatitis. Clostridium perfringens was cultured from skin and liver. A PVLV was isolated in a Marek's disease tumor cell line (MDCC-MSB1) culture and was identified by physicochemical, immunofluorescent, and morphologic features. This isolate was named GA-1 PVLV. Specific-antibody-negative chicks and embryos infected with heat- or chloroform-treated GA-1 PVLV developed anemia at the same rate. Control chicks never were anemic. This is the first isolation of PVLV from clinically ill chickens in the United States and the first report of PVLV-induced anemia in chickens in the Western Hemisphere.  相似文献

The pathologic consequences of chicken anemia virus (CAV) oral inoculation in 4-wk-old broiler breeders of different major histocompatibility B complex (MHC) genotypes were evaluated. MHC B complex was determined by hemagglutination and sequence-based typing. Clinical signs, serology, gross lesions, histopathologic analysis, and CAV genome quantification were used to evaluate disease progression. Clinical disease was not apparent in the inoculated broilers throughout the experimental period. At 14 days postinoculation, antibodies against CAV were detected in 26.4% (29/110) of the inoculated birds. The distribution of percent positive was 34.6% (9/26) and 32.3% (10/31) of the chickens with B A9/A9 and B A9/A4 MHC genotypes, respectively, and seroconversion in six other genotypes was 19% (10/53). These differences among MHC genotypes for specific seroconversion rate were not statistically significant. CAV genomes were detected in the thymus of 87.7% (93/110) of the inoculated birds with no statistically significant differences between MHC genotypes. Mild thymic lymphocytolysis, lymphedema, and medullary hemorrhage were observed in the inoculated chickens. Histomorphometric analysis showed that cortical lymphocyte-to-parenchyma ratios did not differ between inoculated and uninoculated groups or among MHC genotypes. Similar findings have been reported previously in white-leghorn chickens of similar age, suggesting that broilers show a similar resistance to the effects of CAV infection at this age. The absence of significant clinical and pathological changes in the orally inoculated broilers at this age contrasts with CAV-associated thymus damage seen frequently in condemned commercial broilers at harvest.  相似文献

In the recent past, anatomic and clinical pathologic diagnoses of so-called chicken anemia agent (CAA) infections have been based on lesions such as anemia and hematopietic organ atrophy (HOA). In the present study, significant (P less than or equal to 0.05) positive and negative correlations were seen in a lesion matrix constructed from 89 cases of anemia and HOA in Georgia broilers during 1988 and 1989. Only splenic atrophy and bursal atrophy were not significantly associated. We concluded that information regarding only HOA and anemia is not sufficient to allow pathologists to diagnose CAA in broiler chickens submitted to diagnostic laboratories such as ours.  相似文献

The objective of the present study was to determine the effect of broiler breeder egg weight on egg quality, chick quality, and broiler performance using Hubbard Classic broiler breeder flock. Hatching eggs from a commercial broiler breeder flock (Hubbard classic strain) were obtained at the age of 60 wk and divided into 3 egg-weight categories, namely small, medium, and large. For this purpose, 930 eggs having weights 63.09 ± 0.21, 68.85 ± 0.23, and 74.81 ± 0.11g were divided into 3 egg categories labeled small, medium, and large respectively. We further divided these eggs into 2 categories, eggs for quality parameters and eggs for incubation and posthatch performance parameters. Eggs from the small egg category had greater proportion of the shell weight and more shell strength. Egg weight did not significantly affect the albumen and yolk weight percentage. Egg weight affected the shape index and specific gravity for which lower values were observed for large eggs. Egg weight positively affected the chick weight, chick yield, and chick length (P < 0.05) of both male and female chicks. Results of the body weight gain showed that effect of egg weight on the posthatch performance of chick diminishes with the age of broilers. Egg weight significantly affected the body weight gain of male and female chicks up to 3 wk of age. Egg weights did not significantly affect the feed intake and mortality of broiler. Different egg-weight categories significantly affected the feed conversion ratio of female chicks at wk 2 and 3 of age and did not affect the feed conversion of male and female chicks at the end of wk 5. In conclusion, egg weight positively affected the chick characteristics (e.g., chick weight, chick length) and did not affect the final live body weight, feed conversion ratio, feed intake, and mortality in broilers.  相似文献

1. To determine if selection for early response to photostimulation could be successful, 150 male broiler breeders were photostimulated at 8 weeks of age. The first 20 to produce a semen sample and have a reddened comb with an area > 10 cm2 were selected as responders (R) and 20 birds that did not show these signs of sexual development were chosen as non-responders (NR). Once sexually mature, 8 birds from each group that consistently produced a semen sample were mated with both egg-type hybrids and broiler breeder females to observe the response to 8-week photostimulation in the as-hatched offspring. 2. The AFE of the F1 females with NR or R paternity and egg-type hybrid layer maternity (F1L) were similar, but AFE was advanced in birds from R relative to NR paternity when they had broiler breeder maternity (F1B). 3. Date following a normal distribution of AFE were extracted from the overall data set. This group included offspring from both NR and R paternity, but AFE in F1L and F1B females with R paternity was advanced compared to those with NR paternity. 4. Mean testis weights, or age at most rapid testis growth predicted using parameters from Tyler and Gous (2009), of F1 males were not significantly different in birds with NR or R paternity. A strong correlation was found between predicted age at most rapid growth and AFE of full sibs and so it is likely that an advance in AFE in female offspring would also result in an advance in age of testis development of males. 5. There was no significant difference in 21-d body weight of F1B females of NR or R paternity, but the 21-d body weights of F1L females were higher from R than from NR sires, suggesting that although fertility and meat-type traits are often negatively correlated, there was no adverse effect of selection for responsiveness to early photostimulation and broiler growth rates to 21 d. 6. These findings showed that the response to early stimulation is heritable, and should be useful to the broiler breeder industry, where a reduction in photorefractoriness would improve egg production and fertility.  相似文献

To evaluate the economic effects of subclinical chicken anemia agent (CAA) infection on broiler performance, clinically normal broiler flocks were grouped into two categories: A) flocks in which none of 10 birds sampled at slaughter had antibody to CAA, and B) flocks in which six or more of 10 similarly sampled birds had CAA antibody. Production and performance parameters of 25 flocks in each category were compared. No statistically significant differences were found between category A and category B flocks in major production parameters such as sex, feed manufacturer, type of litter, stocking density, and age at slaughter. However, category A flocks achieved 13% (P less than 0.05) greater net income per 1000 birds, 2.0% better feed-conversion ratio (P less than 0.05), and 2.5% (P less than 0.05) greater average weight per bird than category B flocks. No significant differences in hockburn bonus per 1000 birds (a bonus payable to growers whose broilers have the lowest prevalence of contact dermatitis lesions on the hocks) and mortality were found between category A and category B flocks. These results show that subclinical CAA infection has a substantial, statistically significant effect on commercial broiler performance and profitability.  相似文献

Packed cell volumes (PCVs) from 3-, 7-, 14-, 21-, 28-, and 35-day-old clinically healthy chicken anemia agent (CAA)-free and specific-pathogen-free (SPF) leghorn chicks were compared with PCVs from age-matched clinically healthy CAA-free broiler chicks. The PCVs of the SPF chicks regressed significantly (F = 20.6, df = 2/3, P < 0.025) on age in a linear fashion. The PCVs of the broiler chicks regressed significantly (F = 9.56, df = 2/3, P < 0.05) on age in a cubic parabola. The mean PCVs of the broiler chicks were significantly (P < 0.05) higher than PCVs of SPF chicks at every tested time interval. Results indicate that PCV values are higher in broiler chicks than in SPF leghorn chicks, and that PCVs increase as chicks age. Clinicians, diagnosticians, and investigators who intend to work with chicken blood must be aware of these differences.  相似文献

Foot-and-mouth disease (FMD) is endemic in Kenya and has been well studied in cattle, but not in pigs, yet the role of pigs is recognised in FMD-free areas. This study investigated the presence of antibodies against FMD virus (FMDV) in pigs sampled during a countrywide random survey for FMD in cattle coinciding with SAT 1 FMDV outbreaks in cattle. A total of 191 serum samples were collected from clinically healthy pigs in 17 districts. Forty-two of the 191 sera were from pigs vaccinated against serotypes O/A/SAT 2 FMDV. Antibodies against FMDV non-structural proteins were found in sera from 30 vaccinated and 71 non-vaccinated pigs, altogether 101/191 sera (53 %), and 91 % of these (92/101) also had antibodies measurable by serotype-specific ELISAs, predominantly directed against SAT 1 with titres of 10–320. However, only five high titres against SAT 1 in vaccinated pigs were confirmed by virus neutralisation test (VNT). Due to high degree of agreement between the two ELISAs, it was concluded that positive pigs had been infected with FMDV. Implications of these results for the role of pigs in the epidemiology of FMD in Kenya are discussed, and in-depth studies are recommended.  相似文献

1. Eggs were stored for two different times at varying temperatures. The effects on hatchability, chick weight at hatch and hatching time were examined in two broiler breeder lines from 33 to 58 weeks of age. 2. Short storage (1 to 3 d). Storage at 20 degrees C compared with 16.5 degrees C reduced hatchability of all eggs set. No effect was observed on hatchability of fertile eggs, hatching time or chick weight. 3. Long storage (9 to 11 d). Storage at 16.5 degrees C compared with 10 degrees C decreased both hatchability of fertile eggs and chick weight at hatch. Incidence of early embryonic death increased and incubation time decreased at 16.5 degrees C compared with 10 degrees C. 4. Chicks from morning eggs were heavier than those from afternoon eggs irrespective of storage conditions. 5. Hatchability (all eggs set and fertile eggs) and chick weight varied with hen age irrespective of storage conditions. During long storage, hatching time varied with hen age independently of breeder line, storage temperature or egg laying time. 6. Hatchability (all eggs set and fertile eggs) was higher in line A than in line B. Line B eggs hatched later and produced heavier chicks than line A eggs irrespective of storage time.  相似文献

In 1975, a survey was conducted in East Baton Rouge Parish, Louisiana, to determine the prevalence of equine infectious anemia. Using the agar gel immunodiffusion test, 94 of 1,398 horses (6.7%) were found to be infected. Infection rates were especially high in areas where clinical cases of equine infectious anemia had been diagnosed. Clinical signs compatible with the disease were noted in 1 of the 94 seropositive horses. The sample set of 1,398 horses represented 22% of the census population obtained during the 1971 Venezuelan equine encephalomyelitis vaccination campaign.  相似文献

Chicken anemia agent (CAA) was isolated from broiler chickens in Texas with a blue wing or anemia dermatitis-like syndrome. Specific-pathogen-free chicks inoculated with field material developed anemia, and CAA was isolated in MDCC-MSB1 cells from bone marrow and lymphoid tissue from inoculated chicks. One isolate, designated EF88/78/276, was further characterized. Infectivity of EF88/78/276 was resistant to treatment with chloroform and with heat at 70 C for 5 minutes. EF88/78/276 was indistinguishable from the Cux-1 and Gifu-1 isolates of CAA by cross-neutralization tests. Almost all 1-day-old susceptible chicks inoculated intramuscularly with EF88/78/276 developed anemia, but contact-infected chicks did not. Antibody to CAA was detected in broiler breeder flocks from Texas, the Delmarva peninsula, and Alabama.  相似文献
