Summary The relationship between Folsomia candida and chitin-degrading microorganisms was studied. On chitin agar, 1010 bacteria were isolated per g faeces, and 3.8×1011 bacteria per g gut contents, 1/3 of them showing a clear (chitin-free) zone around the colony. The most abundant chitin-degrading bacteria were Xanthomonas maltophilia and Curtobacterium sp. To determine the bacterial contribution in the use of chitin by F. candida, a feeding experiment was carried out. F. candida were fed with chitin, either amended with or without tetracycline as an inhibitor of bacteria. When tetracycline was omitted the biomass of F. candida was increased compared to those fed chitin with tetracycline. However, this result was observed only when the food replacement intervals were long enough to allow bacterial colonization before ingestion of the food. In a food-selection experiment, a preference for chitin colonized with microorganisms as opposed to sterile chitin was found. The results indicate that a mutualistic symbiosis of F. candida with chitinolytic microorganisms is likely to enhance chitin degradation. The effect of sonication during chitin extraction from freshwater prawn shells on yield, purity, and crystallinity of chitin was investigated. Dry prawn shells were suspended for 4 h in 0.25 M HCl at 40 degrees C while they were sonicated for 0, 1, and 4 h. Demineralized shells were lyophilized, resuspended in 0.25 M NaOH, and sonicated again for 0, 1, and 4 h for protein removal. The yield of chitin decreased from 8.28 to 5.02% for nonsonicated and sonicated samples, respectively, which was attributed to losses of depolymerized materials in the wash water. The application of ultrasound enhanced the removal of proteins. In nontreated shells, the amount of protein was 44.01% and was reduced to 12.55, 10.59, and 7.45% after 0, 1, and 4 h of sonication treatments. The glucosamine content slightly decreased with sonication probably because of losses due to depolymerization. The crystallinity indices of chitins decreased as the time of sonication increased. The degree of acetylation of chitins was unaffected by sonication, but the degree of acetylation of chitosans produced from sonicated chitin decreased from 70.0 to 68.7 and 61.4% for 1 and 4 h sonicated samples, respectively.  相似文献

Degradation of chitin, which is an aminopolysaccharide used as a soil amendment, has been often monitored in soil via its degradation products such as carbon dioxide and ammonium. We report here the applicability of thermogravimetry to measure the amount of chitin added to soil. The maximum pyrolysis rate of the upland surface soil of Brown Forest soil supplemented with chitin was strongly correlated with added chitin content (r = 0.999) when the content exceeded 6.0 g kg?1. The maximum pyrolysis rates of chitin-added soil (around 385°C) was distinctive from those of soil supplemented with cellulose, chitosan, N-acetylglucosamine, and N,N’-diacetylchitobiose (around 340°C, 300°C, 200°C, and 240°C, respectively), indicating the specific detection of chitin. Soil incubation study demonstrated that 60 g kg?1 chitin added to the soil declined exponentially (r = 0.993) within days and could not be detected at 90 days after the addition of chitin. Total carbon (C) content also decreased within days whereas total nitrogen (N) remained almost constant over the 90 days. The amount of ammonium-N increased in the initial 30 days after the addition of chitin and reached about 3.6 g kg?1, which corresponded to the amount of N in the added chitin (4.1 g kg?1) while the amount of nitrite-N and nitrate-N were below 2.0 and 15 mg kg?1, respectively. Comparison of the measured ammonium-N and total-C contents with those calculated from the measured chitin-content implied that addition of chitin enhanced degradation of native organic compounds in soil.  相似文献

Antioxidant thio-conjugates were obtained from white grape pomace by a clean and efficient one pot extraction and depolymerization method using water and cysteamine hydrochloride. To evaluate the potential of grape pomaces of different origins as sources of proanthocyanidins and conjugates, we conducted varietal comparative studies of polyphenol content, antioxidant power, and procyanidin composition. Xarel.lo proved to be the richest source of polyphenols. The total conversion into the conjugates was as high as 8 g/kg of Xarel.lo grape pomace, with a 50-fold excess of cysteamine, and 3 g/kg, with a 5-fold excess of cysteamine. After purification by preparative cation exchange and reversed phase high-performance liquid chromatography, 17 g of 63% pure 4beta-(2-aminoethylthio)epicatechin (acetate salt) was obtained from 35 kg of moist pomace. The procedure described here will make the antioxidant thio-derivatives efficiently available directly from raw plant byproducts such as grape pomace.  相似文献

Starch and Sago production from cassava (Manihot esculenta Crantz) roots is an increasingly important agro industry. Casssava is one of many cyanogen containing plants. In the present study the amount of cyanogens present in the waste waters and ground waters collected from the starch/sago processing industries in Salem, the main centre of cassava starch and sago production in India was assessed. Concentration of total cyanogens ranged between 12.9 to 66.6 mg l-1 (as HCN) in the initial processing stages whereas the final discharge contained 10.4 to 27.4 mg l-1. Linamarase activity of the waste water samples ranged between 3350 to 3370.6 µmoles l-1 15 min-1. The ground water sources near cassava starch/sago factories showed much higher cyanogen concentration than the acceptable level. The level of total cyanogen in the samples of ground water ranged between 1.2 mg l-1 to 1.6 mg l-1.  相似文献

The process for extracting sugarcane juice (Saccharum officinarum) represents the point of greatest contamination in sugarcane mills. Sodium dithiocarbamate also known as metam-sodium or MS is added to inhibit the growth of microorganisms especially Leuconostoc mesenteroides which is responsible for forming polysaccharides. Metam-sodium, upon decomposition, produces highly toxic byproducts. According to literature, under acidic conditions, MS is hydrolyzed resulting in methylamine (MA), CH3NH2, and carbon disulfide (CS2), and in dilute alkaline solutions, MS produces an oxidation reaction characterized by the formation of elemental sulfur (S) and methyl isothiocyanate (MITC). In this paper, it was studied how MS decomposes to MITC and/or MA considering the effects of the matrix (methanol and water); of temperature (4 and 25 °C); of processing time (0, 1, 2, 3, 4 days); and of pH (4.0, 4.5, 7.0). A second experimental design considering the effects of the matrix (water and sugarcane juice); of temperature (4, 25, 35, 45 °C); of processing time (30, 300 min); and of pH (4.0, 4.5, 7.0) was derived from the results obtained considering MITC and/or MA formation. According to the statistical analysis of these results (p < 0.05), the order of the influential factors was as follows: time > matrix > pH > temperature. Results also indicated that the water matrix at pH = 4.5 and 45 °C had the lowest degradation rate (k), with a value of 8.82 day?1, while for the sugarcane juice matrix at the same pH and temperature conditions was larger, with a k value of 30.07 day?1. These results show that the matrix is also important for the degradation of dithiocarbamate to MITC and to MA.  相似文献

采用改良硫酸铵盐析法从鸡胆汁中分离鸡分泌型免疫球蛋白A(S IgA),M acro-prep H igh Q阴离子交换柱层析提纯,经二次过柱后获得的鸡S IgA浓度达22 m g/mL。SDS聚-丙烯酰胺凝胶电泳检测表明,重链分子质量为63~64 ku,轻链为26~28 ku,J链约16 ku。用纯化的鸡S IgA免疫兔,所得的抗血清经双向琼脂扩散,证明兔抗鸡S IgA血清效价达1¨128。  相似文献

Cellulose acetate butyrate (CAB) and cellulose acetate propionate (CAP) were prepared homogeneously in a 1-allyl-3-methylimidazolium chloride (AmimCl) ionic liquid system from sugarcane bagasse (SB). The reaction temperature, reaction time, and molar ratio of butyric (propionic) anhydride/anhydroglucose units in the cellulose affect the butyryl (B) or propionyl (P) content of CAB or CAP samples. The (13)C NMR data revealed the distribution of the substituents of CAB and CAP. The thermal stability of sugar cane bagasse cellulose was found by thermogravimetric analysis to have decreased after chemical modification. After reaction, the ionic liquid was effectively recycled and reused. This study provides a new way for high-value-added utilization of SB and realizing the objective of turning waste into wealth.  相似文献

The preemergence chloroacetamide herbicide metazachlor was encapsulated in biodegradable low molecular weight poly(lactic acid) micro- and submicroparticles, and its release to the water environment was investigated. Three series of particles, S, M, and L, varying in their size (from 0.6 to 8 μm) and with various initial amounts of the active agent (5%, 10%, 20%, 30% w/w) were prepared by the oil-in-water solvent evaporation technique with gelatin as biodegradable surfactant. The encapsulation efficiencies reached were about 60% and appeared to be lower for smaller particles. Generally, it was found that the rate of herbicide release decreased with increasing size of particles. After 30 days the portions of the herbicide released for its highest loading (30% w/w) were 92%, 56%, and 34% for about 0.6, 0.8, and 8 μm particles, respectively. The release rates were also lower for lower herbicide loadings. Metazachlor release from larger particles tended to be a diffusion-controlled process, while for smaller particles the kinetics was strongly influenced by an initial burst release.  相似文献

A new rapid and low-cost preparation of the (3E,5Z)-3,5-alkadienyl system, encountered in several insect pheromone constituents, was developed. Knoevenagel condensation of (E)-2-alkenals with ethyl hydrogen malonate in dimethyl sulfoxide, in the presence of a catalytic amount of piperidinium acetate, led to a mixture of geometrical isomers of ethyl 3,5-alkadienoates and ethyl 2,4-alkadienoates, from which the (3E,5Z)-3,5-alkadienoate was conveniently separated, by the use of urea inclusion complex formation. The importance of this procedure has been illustrated by the preparation of the (3E,5Z)-3,5-tetradecadienoic acid (megatomoic acid) 1, the (3E,5Z)-3,5-dodecadienyl acetate 2, and the (3E,5Z)-3,5-tetradecadienyl acetate 3. These compounds are the main components of insect sex pheromones and constitute synthetic targets of considerable interest for the semiochemical community.  相似文献

Salt solubility of pH-shift isolated herring ( Clupea harengus ) muscle proteins was studied in relation to pH exposure and microstructure using transmission electron microscopy (TEM). Using protein solubilization at pH 11.2 with subsequent precipitation at pH 5.5, salt solubility of the proteins decreased from 78 to 17%. By precipitating the alkali-solubilized proteins at the pH of native herring muscle, 6.5, salt solubility only decreased to 59%, proving that pH values between 6.5 and 5.5 affected protein salt solubility more than the pH cycle 6.5 → 11.2 → 6.5. Precipitation at pH 5.5 resulted in hydrogen bonds, hydrophobic interactions, and S-S bridges, whereas precipitation at pH 6.5 resulted only in the formation of hydrophobic interactions. The alkaline pH-shift isolation process severely rearranged the protein microstructure, with precipitation at pH 6.5 forming a finer, more homogeneous network than precipitation at pH 5.5. The former protein isolate also contained less lipid oxidation products and formed more deformable gels, without affecting protein yield.  相似文献

Lysis of chlamydospores of Fusarium solani f. sp. cucurbitae is enhanced both in chitin and in laminarin amended soils. In soil amended with both chitin and laminarin, lysis of chlamydospores resembles that in control soil. Addition of chitin and laminarin to soil stimulates the growth of both bacteria and actinomycetes. In soils amended with chitin and with both chitin and laminarin, a chitinolytic microflora is stimulated. Penetration of chlamydospore cells by soil microorganisms has not been observed using the electron microscope. The mechanism of lysis of chlamydospores in soil is discussed.  相似文献

The main objective of the present study was to compare the composition and functional and nutritional properties of whole yeast cells (WY) from an ethanol distillery with those of a phosphorylated protein concentrate (PPC) prepared from the same cells. Comparisons were also made of PPC with texturized soy protein (TSP) and soy protein isolate (SPI), both acquired in the local market. Yeast (Saccharomyces cerevisiae) is a rich source of protein, soluble fiber, and some minerals. Saturated fatty acids predominated over monounsaturated and polyunsaturated in both WY and PPC. The functional properties of PPC were similar to those of SPI and TSP. Both soy products and PPC replaced 20 or 40% chuck roll protein without affecting the emulsion properties of the meat products. Amino acid scoring was high for both WY and PPC; digestibility was higher (90%) for PPC and lower (68%) for WY. The protein nutritive value of PPC did not differ from that of casein and was significantly higher than that for WY.  相似文献

 The effect of the interaction between a vesicular-arbuscular (VA)-mycorrhiza (Glomus intraradices no. LAP8) and Streptomyces coelicolor strain no. 2389 on the growth response, nutrition and metabolic activities of sorghum (Sorghum bicolor) plants grown in non-sterilized soil amended with chitin waste was studied in a greenhouse over 8 weeks. Chitin amendment resulted in an increase in the microbial population and chitinase activity in soils. Growth of mycorrhizal G. intraradices no. LAP8 and non-mycorrhizal sorghum plants increased as compared with other treatments either in the presence or absence of S. coelicolor strain 2389. VA-mycorrhizal inoculation significantly increased the growth, photosynthetic pigments, total soluble protein and nutrient contents of sorghum compared to non-mycorrhizal sorghum. Such increases were related to increased mycorrhizal colonization. Inoculation with S. coelicolor 2389 significantly increased the intensity of mycorrhizal root colonization and arbuscular formation, but the levels of mycorrhizal infection and their beneficial effects were significantly reduced with the addition of chitin waste to the soil. Analysis of the content of total amino acids and ammonia in leaves on the basis of dry matter production showed that, in most instances, total amino acids of mycorrhizal plants were significantly higher than those of non-inoculated plants. The microflora of the rhizosphere was highly affected by mycorrhizal inoculation. Quantitative changes in acid and alkaline phosphatase activities of the roots in response to the mycorrhizal inoculation are discussed. Received: 11 August 1999  相似文献

Establishment of vegetation on residues produced from the bauxite refining process is a beneficial part of their environmental management. Of the two fractions produced in the refining, the coarse fraction has greater efficiency in the leaching of excess salts and alkalinity. However, these same properties can result in increased loss of nutrients and low water-holding capacity. The current study investigated the use of mixing coarse fraction residue with fine fraction residue, at two different application rates (10% and 25%), with and without the use of gypsum as an ameliorant, for re-vegetation of the residue with Trifolium pratense. Optimum plant growth was observed in treatments that had also received gypsum amendment, with higher plant biomass, Mn nutrition and lower Al and Fe concentration. However, use of process sand at the higher application rate (25%) promoted lower levels of soluble Al and Fe and exchangeable Na in the substrate and, consequently, lower plant uptake of Na. Results indicate that co-disposal of the coarse fraction sand at 25% w/w with fine fraction residue can improve the substrate and, therefore, plant uptake and growth. Further monitoring is recommended to determine the effect of the absence of gypsum and other nutrient sources on plant growth.  相似文献

A novel method for the discrimination of bluefin tuna (Thunnus thynnus) from Atlantic bonito (Sarda sarda) was developed, based on species-specific amplification of a region of the mitochondrial cytochrome b gene by Polymerase Chain Reaction (PCR). The method, which uses a one-step amplification reaction, is more rapid to perform than any of the currently described techniques for species determination in fish. The species of origin of the DNA is indicated by the distinctive size of the PCR product on electrophoresis, but the test could readily be adapted to other forms of electrophoresis or fluorescence-based systems for quantification. Given the possibility of intraspecific variability in mitochondrial DNA and the consequent desirability of performing two independent tests, the new method constitutes a valuable addition to the range of tuna speciation methodologies currently available.  相似文献
