The contents of volatile fatty acids in the rumen fluid were studied in calves at an early (seven weeks of age) and traditional weaning term (nine weeks of age). Weaning at the age of seven weeks enabled to save 16.2 kg of milk replacer per calf; there was also a lower content of concentrates in the starter feed with a supplement of Amylastim. The health condition of the calves was good in both groups. The average daily weight gains for the period from the second to the ninth week of age were 0.550 kg in the early weaned calves and 0.690 kg in the calves weaned at a normal time. Early weaning had a positive influence on the development of rumen metabolism. Calves weaned at the age of seven weeks, compared with those weaned at the age of nine weeks, had much higher concentrations of VFA (p less than 0.05) in rumen fluid (at the age of seven weeks: 130.49 mmol per litre vs. 111.53 mmol per litre; at the age of eight weeks: 119.74 mmol per litre vs. 96.98 mmol per litre). Early weaned calves had the statistically significantly (p less than 0.05) higher proportions of propionic acid, butyric acid, i-butyric acid, and valeric acid. Later-weaned calves had the significantly higher (p less than 0.05) contents of acetic acid and i-valeric acid.  相似文献

In this experiment, effect of wheat processing on rumen conditions and development were investigated. Fifty‐six neonatal Holstein‐Friesian calves (22 male and 34 female) were fed calf starters and post‐weaning diets containing 35 (pre‐weaning) and 21.90% (post‐weaning) popped wheat (PW), steam‐flaked wheat (SFW), dry‐rolled wheat (DRW) or ground wheat (GW) till 12 weeks of age. Calves were weaned at the end of 9th week, and a post‐weaning‐specific starter diets were fed for 1 month. Rumen liquor was analysed in days 30, 60 and 90 of the experiment to determine volatile fatty acids (VFA), pH and ammonia nitrogen concentrations. Twelve male calves (three calves/treatment) were slaughtered, and digestive tract was emptied. Forestomach empty weight and rumen parameters were assessed. Results indicated that calves received PW had the highest total VFA, acetate, propionate, butyrate, ammonia nitrogen, rumen wall thickness, papilla width and density. Calves fed DRW experienced the lowest rumen pH throughout the experiment probably because high proportion of fine particles in GW. Calves consuming PW apparently had more functional rumen in comparison with other groups.  相似文献

Effects of the presence or absence of ciliate protozoa on methanogenesis in the rumen and hindgut were investigated in young calves during a 7-week period. Ten Holstein calves, aged 7 days, were divided in two groups (n = 5) and fed an increasing amount of a commercial milk replacer and small amounts of a calves starter. One group was inoculated with ciliate fauna on two occasions, week 5 and 6, while the second remained ciliate-free. The absence of protozoa in the rumen decreased rumen empty weight (-23%, P < 0.01), and rumen pool size of N (-36%, P < 0.01) and crude fat (-37%, P < 0.05). Rumen bacteria of non-faunated calves contained a higher proportion of total amino acid-N per 16 g N (+3%, P < 0.01) and D-alanine-N per 16 g N (+13%, P < 0.05) compared to faunated calves. Further results contain a reference for a higher bacterial mass in the ciliate-free rumen with an increased number of bacteria adherent to rumen mucosa. The CH4 production in the rumen increased exponentially with the increase in protozoa population size (R2 = 0.68). In presence of 46 x 10(4) protozoa per ml rumen fluid, the in vitro CH4 production of rumen fluid per mol total VFA was about 34% higher in faunated than in non-faunated calves (P < 0.001). Hydrogen (2H) recovery of rumen fermentation was positively correlated (R2 = 0.55) to the CH4 production rate. Methanogens were attached on rumen mucosa. Methanogenesis, induced by rumen mucosa attached bacteria, was stimulated by ruminal protozoa. In the absence of protozoa in the rumen, the acetate-propionate ratio and butyrate proportion of VFA were reduced. In vivo, in the absence of protozoa not only the whole animal CH4 production (-30%, P < 0.05) but also the digestibility of carbohydrates (-4%, P < 0.05) was reduced. Thereby no difference was observed in the intake of ME per kg DM between the groups. In conclusion, the methanogenesis in the rumen, but not in hindgut, is associated with the development of the ruminal protozoa population. The level of methanogenesis (mol/mol VFA) in the hindgut amounts to 20% of the ruminal methanogenesis.  相似文献

Digestive activities were studied in test calves (n = 12) in relation to age and feed intake. The calves were isolated after birth from adult cows. Since the 14th day of age, milk replacers were fed with concentrate feed mixture TG and alfalfa hay, i.e. the calves were in the period of plant feeding. The investigation lasted from the end of milk feeding (50th day) to the age of six months. Overall health condition was studied clinically. The characteristics of digestive activities studied in rumen fluid at weekly--monthly intervals were pH, ammonia content, total content of volatile fatty acids, content of particular volatile fatty acids, incidence and number of infusoria. At the age of 50 days, actual pH of rumen fluid was on the average 6.07 and it ranged from 5.84 to 6.76 in the next period. Ammonia content was the highest at the age of 50 days (21.49 mmol/l rumen fluid), then it dropped to 9.83 mmol/l rumen fluid at the age of 180 days. The average total concentration of volatile fatty acids (C2-C5) made 117.00 mmol/l during the first examination, it increased to 132.98 mmol/l at the age of 84 days, and it dropped to 94.16 mmol/l rumen fluid at the age of six months. A similar tendency was found in the volatile fatty acids. The infusoria were found out only at the age of 120 days--their numbers were 155 000 per ml; at the age of 180 days their numbers rose to 368 000 per ml rumen fluid. A trial was conducted with wethers to study the effect of the administration of a synthetic diet (composition: 30.125% starch, 30.125% sucrose, 25% cellulose, 5.25% urea, 8.125% mineral supplement, 1.25% maize oil and 0.125% cholinechloride) upon rumen fermentation. The adaptation to the synthetic diet lasted three months, the proportion of the synthetic diet increasing every week (by 10%) to the detriment of a traditional diet (composition: 0.5 kg meadow hay, 0.3 kg barley, 0.2 kg wheat bran, salt and straw ad libitum). In the 10th week the animals consumed 0.5 kg granular synthetic diet, 0.2 kg cellulose flakes and 0.01 kg polystyrene. After three weeks of the administration of the fully synthetic diet, the rumen fluid was sampled after morning feeding in intervals of 0, 1, 3, 5 and 7 hours. In the dynamics of fermentation, statistically significant differences were found only in isobutyric and isovaleric acid between the 0th and 1st and between the 5th and 7th hours (P less than 0.05--P less than 0.001). The data for all the time intervals were recalculated to average values. These were as follows: total volatile fatty acids 63.03 mmol/l, acetic acid 51.00 mol%, propionic acid 26.75 mol%, butyric acid 19.43 mol%, isobutyric acid 0.91 mol%, isovaleric 1.27 mol%, valeric acid 0.62 mol%, energy efficiency of VFA production 78.23%. The obtained data are confronted with literary data on synthetic diets which contained urea and various energy sources.  相似文献

Three sheep fitted with a ruminal cannula and an abomasal catheter were used to study water kinetics and absorption of VFA infused continuously into the rumen. The effects of changing VFA concentrations in the rumen by shifting VFA infusion rates were investigated in an experiment with a 3 x 3 Latin square design. On experimental days, the animals received the basal infusion rate of VFA (271 mmol/h) during the first 2 h. Each animal then received VFA at a different rate (135, 394, or 511 mmol/h) for the next 7.5 h. Using soluble markers (polyethylene glycol and Cr-EDTA), ruminal volume, liquid outflow, apparent water absorption, and VFA absorption rates were estimated. There were no significant effects of VFA infusion rate on ruminal volume and water kinetics. As the VFA infusion rate was increased, VFA concentration and osmolality in the rumen were increased and pH was decreased. There was a biphasic response of liquid outflow to changes in the total VFA concentration in the rumen, as both variables increased together up to a total VFA concentration of 80.1 mM, whereas, beyond that concentration, liquid outflow remained stable at an average rate of 407 mL/h. There were significant linear (P = 0.003) and quadratic (P = 0.001) effects of VFA infusion rate on the VFA absorption rate, confirming that VFA absorption in the rumen is mainly a concentration-dependent process. The proportion of total VFA supplied that was absorbed in the rumen was 0.845 (0.822, 0.877, and 0.910 for acetate, propionate, and butyrate, respectively). The molar proportions of acetate, propionate, and butyrate absorbed were affected by the level of VFA infusion in the rumen, indicating that this level affected to a different extent the absorption of the different acids.  相似文献

The pharmacokinetics of chloramphenicol, either administered as the monosuccinate ester or as a veterinary formulation, were studied in calves from the first day of life to the age of 10–12 weeks and compared with the results obtained in adult cattle. (1) After intravenous injection of 0.15 mmol/kg chloramphenicol monosuccinate, the plasma elimination half life of intact ester fell from a value of 33 min on the first day of life to 15 min at the age of 10–12 weeks (value in cows = 14 min). Free chloramphenicol reached maximal plasma concentrations after 2–3 h on the first day of life, but in less than 15 min in cows. The elimination half-life fell from about 15 h on day 1 to 4.8 h at the age of 10–12 weeks (4.2 h in cows). The bioavailability of the ester was more than 90% on Day 1, but declined to 50–60% from Day 7 on account of rapid renal excretion: 21–28% of the total dose was excreted as intact ester in a 2 h period following injection in calves aged 10–12 weeks. (2) The veterinary formulation of chloramphenicol proved toxic when administered intravenously at a dose of 0.15 mmol/kg, and even a dose of 0.093 mmol/kg was less well tolerated than 0.15 mmol/kg of the monosuccinate ester. (3) The pharmacokinetics of chloramphenicol fitted an open two-compartment model, the half-life of the elimination phase corresponded well to the values determined in the experiments with the monosuccinate ester. (4) The intramuscular injection of 0.15 mmol/kg of the ester or 0.093 mmol/kg of chloramphenicol provided ‘therapeutic’ plasma concentrations (≥ 5 μg/ml) within 15–30 min and for about 24 h in calves aged 7 days. (5) Chloramphenicol crossed the placenta when given to cows shortly before a Caesarian section, but equilibrium was not reached within 50–100 min. (6) The binding of chloramphenicol to serum proteins was dependent both on total protein and drug concentrations. It rose from less than 30% on day 1 to about 40% in adult cattle. (7) Recommendations for a dosage regime for chloramphenicol in calves are made on the basis of the pharmacokinetic data.  相似文献

Three hundred bucket-fed diarrhoeic calves up to the age of 21 days were used to investigate the degree in which D-lactic acid contributes to metabolic acidosis in bucket-fed calves with naturally acquired neonatal diarrhoea. Fifty-five percent of all diarrhoeic calves had serum D-lactate concentrations higher than 3 mmol/l. Mean (+/-SD) D-lactate values were 5.7 mmol/l (+/-5.3, median: 4.1 mmol/l). D-lactate values were distributed over the entire range of detected values from 0 to 17.8 mmol/l in calves with base excess of -10 to -25 mmol/l. Serum D-lactate concentration was higher in patients with ruminal acidosis (6.6 +/- 5.2 mmol/l; median: 5.9 mmol/l) than in those with physiological rumen pH (5.3 +/- 5.4 mmol/l; median: 3.7 mmol/l). There was no evidence of a correlation (r = 0.051) between the serum levels of D-lactate and creatinine (as an indicator of dehydration). D-lactate was correlated significantly with both base excess (r = -0.685) and anion gap (r = 0.647). The proportion of cured patients was not significantly different between the groups with elevated (>3 mmol/l) and normal serum D-lactate concentrations. This study shows that hyper-D-lactataemia occurs frequently in diarrhoeic calves, has no impact on prognosis but may contribute to the clinical picture associated with metabolic acidosis in these animals.  相似文献

Two sheep with a ruminal fistula and an isolated small rumen were studied for the secretion of ammonia nitrogen, urea nitrogen, and amino nitrogen into the isolated rumen at different levels of volatile fatty acids (VFA) (50, 133-97, and 97-66 M Mol 1(-1)) in the rumen. The VFA level in the rumen was found to exert a great influence on the quantitative secretion of endogenous nitrogen from the blood through the rumen wall into rumen content. When the VFA level in the rumen was increased by administration of a single dose of acetic, propionic, and butyric acid, the secretion of ammonia nitrogen and amino nitrogen abruptly dropped and the secretion of urea into the isolated rumen slightly increased. The over-all amount of nitrogen (NH3-N + urea-N + amino-N) that had passed into the isolated rumen in the course of an hour showed a highly significant correlation with the passage of nitrogen in the form of ammonia and amino nitrogen and was greatest before the application of VFA to the rumen, i.e. at the level of 50 m mol 1-1. Of the metabolites under study, which were passing to the isolated rumen, amino nitrogen shared the greatest proportion (45.38-46.54%). When the VFA level in the rumen was raised, the proportion of ammonia secreted to the isolated rumen decreased and the proportion of urea in the total amount of nitrogen increased.  相似文献

Four steers fitted with a ruminal cannula and chronic indwelling catheters in the mesenteric artery, mesenteric vein, hepatic portal vein, hepatic vein, and the right ruminal vein were used to study the absorption and metabolism of VFA from bicarbonate buffers incubated in the temporarily emptied and washed reticulorumen. Portal and hepatic vein blood flows were determined by infusion of p-aminohippurate into the mesenteric vein, and portal VFA fluxes were calibrated by infusion of isovalerate into the ruminal vein. The steers were subjected to four experimental treatments in a Latin square design with four periods within 1 d. The treatments were Control (bicarbonate buffer) and VFA buffers containing 4, 12, or 36 mmol butyrate/kg of buffer, respectively. The acetate content of the buffers was decreased with increasing butyrate to balance the acidity. The butyrate absorption from the rumen was 39, 111, and 300 +/- 4 mmol/h for the three VFA buffers, respectively. The ruminal absorption rates of propionate (260 +/- 12 mmol/h), isobutyrate (11.4 +/- 0.7 mmol/h), and valerate (17.3 +/- 0.7 mmol/h) were not affected by VFA buffers. The portal recovery of butyrate and valerate absorbed from the rumen increased (P < 0.01) with increasing butyrate absorption and reached 52 to 54 +/- 4% with the greatest butyrate absorption. The liver responded to the increased butyrate absorption with a decreasing fractional extraction of propionate and butyrate, and with the greatest butyrate absorption, the splanchnic flux was 22 +/- 1% and 18 +/- 1% of the absorbed propionate and butyrate, respectively. The increased propionate and butyrate release to peripheral tissues was followed by increased (P < 0.05) arterial concentrations of propionate (0.08 +/- 0.01 mmol/kg) and butyrate (0.07 +/- 0.01 mmol/kg). Arterial insulin concentration increased (P = 0.01) with incubation of VFA buffers compared with Control and was numerically greatest with the greatest level of butyrate absorption. We conclude that the capacity to metabolize butyrate by the ruminal epithelium and liver is limited. If butyrate absorption exceeds the metabolic capacity, it affects rumen epithelial and hepatic nutrient metabolism and affects the nutrient supply of peripheral tissues.  相似文献

本试验旨在探讨不同精粗比日粮对奶山羊瘤胃液pH值、VFA以及血液中VFA含量的影响。选择8只安装永久性瘤胃瘘管的奶山羊作为试验动物,采用完全随机分组试验设计随机分为2组,分别饲喂精粗比为6∶4和4∶6的日粮,预饲期15 d,采样期3 d。结果表明,高精料组(HC组)瘤胃液pH值显著低于低精料组(HR组)(P<0.05);在采食后3 h,HC组与HR组瘤胃液pH值均下降至最低值,分别为5.71和6.08。除了乙酸含量外,HC组瘤胃液丙酸、异丁酸、丁酸、异戊酸、戊酸以及总挥发性脂肪酸(TVFA)含量分别比HR组提高4.99%、5.58%、21.81%、17.95%、18.27%、1.66%。HC组血浆中各种VFA的含量均高于HR组,其中丙酸、丁酸含量两组间差异达到显著水平(P<0.05)。HC组瘤胃液以及血浆中乙酸与丙酸比值均低于HR组,但两组间差异均不显著(P>0.05)。HC组瘤胃液乙酸、丙酸、TVFA浓度在采食后2 h达到最大值,HR组在采食后3 h达到最大值,两组日粮血浆中VFA浓度均在采食后2 h达到最大值,然后逐渐恢复到采食前水平。结论:高精料日粮导致瘤胃液pH值显著降低,瘤胃液和血浆中VFA含量增加;瘤胃液VFA生成速率HC组高于HR组。  相似文献

The objective of this study was to determine whether the nutritional regimen of rearing calves would influence the morphometric and histological development of rumen mucosa. Twelve male Holstein calves 7 d of age were assigned to three groups of 4 animals each: milk group (I), early weaned (6 weeks) group (II) and late weaned (9 weeks) group (III). All animals received additional solid feed. Animals of group I were slaughtered after 6 weeks of age, whereas those in groups II and III were slaughtered after 9 weeks of age. At slaughter, the ruminal digesta amounted to 2035 g (milk group), 3092 g (late weaned group) and 5374 g (early weaned group). The differences in the ruminal molar percentage of SCFA were not significant. There was a trend for lower pH and higher SCFA concentrations in the order late weaned, early weaned and milk fed animals (pH: 6.4, 6.6 and 6.7, respectively; SCFA: 96, 87 and 77 mmol/l, respectively). The mean length (1.07 mm in milk group, 1.45 mm in late weaned group and 1.87 mm in early weaned group), width (0.43, 0.58 and 0.71 mm, respectively) and surface of papillae (190, 232 and 241 mm2/cm2 mucosa, respectively) increased with both the age of the animals and the elevated intake of solid feed, whereas the number of papillae (210, 140 and 92 per cm2 mucosa, respectively) decreased. In both milk-fed groups type A and B corneal cells were present in the Stratum corneum, whereas in the earlier weaned calves type C-cells could be also seen. These findings indicate a more advanced stage of development of the rumen epithelium in the earlier weaned calves fed higher amounts of concentrate and hay.  相似文献

The effect of feeding level and interval between feedings on the fermentation pattern in the bovine rumen have been investigated in experiments with hay. The animals were completely adapted heifers fitted with rumen cannulas, and the parameters measured were ammonia, pH, volatile fatty acids (VFA) and non-glucogenic/glucogenic ratio (NGGR) in the VFA mixture.Increasing feeding levels, ranging from 2/3 to 4/3 of maintenance level, resulted in higher average concentrations of VFA and lower pH in the rumen fluid. Further the highest level of intake caused a considerable diurnal variation in the pH and the concentration of total VFA, and increased the variation in the molar composition of the VFA mixture.Three feeding intervals ranging from 8 to 16 hrs., with hay administered at maintenance level, caused no changes in the fermentation pattern.Typical variations in the concentrations of ammonia and valeric acid as related to time after feeding were demonstrated, but the concentrations of the branched-chain fatty acids in the rumen fluid were found to be quite constant.It may be concluded that a representative mean value of the parameters measured, except for ammonia and valeric acid, may be based on relatively few samples when feed intake does not exceed maintenance level, whereas sampling every hour is required at higher feeding levels.  相似文献

Acid-base balance and electrolyte concentrations, including ionised calcium, were monitored during intravenous fluid therapy of 11 collapsed diarrhoeic suckler calves aged five to 10 days. Six healthy calves of similar age and type were used to provide control data. All the diarrhoeic calves were severely acidotic (TCO2<12 mmol/litre). Isotonic sodium bicarbonate (1-3 per cent) was administered until the metabolic acidosis was half corrected, as indicated by the TCO2 increasing to 17 to 24 mmol/litre when the infusion was changed to an extracellular volume replacement fluid containing 144 mmol/litre Na+, 35 mmol/litre HCO3-, 4 mmol/litre K+ and 113 mmol/litre Cl- which was administered until the calf was discharged. Milk feeding was started as soon as the calf had a suck reflex. The treatment was successful in 10 calves. At admission the diarrhoeic calves were hypocalcaemic compared with the control calves, but their ionised calcium was significantly higher, with significantly less calcium being protein bound. Treatment with isotonic sodium bicarbonate resulted in a significant improvement in acid-base balance, but both total and ionised calcium decreased significantly, the decrease in ionised calcium being proportionately greater owing partly to a significant increase in the protein binding of calcium. The mean total, bound and ionised calcium concentrations were all significantly lower in the treated calves after they had received isotonic sodium bicarbonate than in the control calves. Further treatment with replacement fluid had no significant effect on any of the parameters apart from pCO2 which increased significantly. Milk feeding had no significant effect on plasma calcium concentrations. The calves' mean ionised calcium concentration was significantly lower at the end of the treatment than before it, but there was no difference in the mean total and bound calcium concentrations. The calves' mean plasma potassium and magnesium concentrations decreased significantly during the course of the treatment.  相似文献

The origin of D-lactate, the most important acid contributing to metabolic acidosis in the diarrheic calf, is unknown. We hypothesized that because D-lactate is produced only by microbes, gastrointestinal fermentation is the source. The objective of this study was to determine whether D-lactate production occurs in the rumen, colon, or both, and to measure D- and L-lactate concentrations in urine. Fecal, rumen, blood, and urine samples were obtained from 16 diarrheic and 11 healthy calves. Serum electrolyte concentrations were measured in both groups, and blood gas analyses were performed for diarrheic calves. All samples were analyzed for D- and L-lactate by high performance liquid chromatography (HPLC). Diarrheic calves were generally hyperkalemic with high serum anion gap, depressed serum bicarbonate, and low blood pH. L-lactate was markedly higher in rumen contents (22.7 mmol/ L [median]) and feces (8.6 mmol/L) of diarrheic calves than healthy calves (0.5 mmol/L and 5.1 mmol/L, respectively), but not different in serum or urine. Rumen, fecal, serum, and urine D-lactate concentrations were all significantly higher (P < .05) in diarrheic calves (17.0, 25.4, 13.9, and 19.2 mmol/L, respectively) than in healthy calves (0.5, 9.1, 1.4, and 0.5 mmol/L, respectively). Higher D-lactate concentrations in the rumen and feces of diarrheic calves suggests these sites as the source of D-lactate in blood and urine.  相似文献

本试验旨在研究断奶前后犊牛在不同固液比例饲喂模式下营养物质代谢及其瘤胃发酵的异同,从而探索基于不同饲喂模式下的犊牛断奶方式。试验选用7日龄荷斯坦公犊牛36头,随机分成3组,每组12头,各组采用相同原料组成、相同营养成分的代乳粉和颗粒料。在总干物质饲喂量保持一致的情况下,改变固液饲料饲喂比例,形成3种饲喂模式组:高液体饲料比例(HL)组,28~56日龄内,颗粒料∶代乳粉保持在1∶2,56日龄断奶;对照(LS)组28~56日龄内,颗粒料∶代乳粉从1∶2逐步降低到1∶1,56日龄断奶;高固体饲料比例(HS)组,28~42日龄内,颗粒料∶代乳粉从1∶2逐步降低至1∶0,并于42日龄断奶。试验期77 d。分别于犊牛28、42、56和84日龄采集瘤胃液,35和63日龄进行断奶前后消化代谢试验。结果表明:断奶前,与HS组相比,HL、LS组犊牛总能代谢率相对较高,但差异不显著(P0.05);断奶后,HS组消化能代谢率、氮利用率及氮的生物学价值较HL组显著提高(P0.05)。与HL组相比,HS组84日龄瘤胃液微生物蛋白含量显著提高(P0.05)。3组间瘤胃液氨态氮浓度无显著差异(P0.05)。84日龄时,HS组犊牛瘤胃液总挥发性脂肪酸浓度和丁酸比例显著高于HL组(P0.05),各组间丙酸、戊酸比例无显著差异(P0.05)。综上可得,适量增加固体饲料饲喂比例有助于改善断奶前后犊牛瘤胃发酵环境,促进瘤胃微生物蛋白的合成,提高断奶后犊牛饲粮能量代谢率、氮的生物学价值及氮利用率;采用高固体饲料饲喂模式,犊牛在42日龄固体饲料采食量达到1.0 kg/d时实施断奶具有一定优势。  相似文献

Fifty-six calves in the period of a milk nutrition, randomly assigned to two groups (control and alpha amylase fed) were used to monitor the changes in the rumen microbial populations and total volatile fatty acids (VFA) concentrations associated with feeding Amylosubtilin G10X (0.6 g.day-1). Statistically significant increase was observed for amylolytic counts in experimental calves on 20th or 30th days. The counts of rumen cellulolytic bacteria and selenomonas tended to have increase in the value, but were not significantly different from those in the control valves. For evaluation of physiologically significant organic anions in the colonic environment, 87 samples of normal feces were collected from the rectum of 15 calves less than 60 days old. The calves were fed milk replacer with free access to starter diet and hay. After fecal extraction with water, pH, D- and L-lactate, succinate, and volatile fatty acid (VFA) concentrations were determined. There was wide variation in fecal pH (4.3 to 7.7). Higher lactate concentrations were observed in feces samples with lower pH, and most of these samples were collected during the first 4 weeks of life. Elevated lactate concentrations included both the D- and L-isomers, and the D-isomer comprised approximately 30-50% of total lactate. Elevated succinate concentrations were observed in only 8 fecal samples, while other samples had lower or trace amounts of succinate. Elevated fecal succinate showed no relationship with fecal pH or VFA concentrations. Fecal VFA concentrations were lower in samples collected in the early postnatal stage, but fecal VFA concentrations were not necessarily related to age. We confirmed that fecal D- and L-lactate concentrations increased with a concomitant decrease of VFA in the acidic lumen of the colon, although acidic feces were not necessarily accompanied by elevated concentrations of lactate. In contrast, succinate production was not related to fecal pH or VFA concentrations.  相似文献

Age-related changes in the absorption and distribution patterns of trimethoprim/sulfadiazine were studied following oral or subcutaneous administration of 15 mg/kg of the drug combination in calves. Following oral administration, the time course of trimethoprim/sulfadiazine appearance and dissipation in serum, synovial fluid and urine was followed for periods up to 48 hours in calves one day, one week and six weeks of age. The profiles of drug appearance-disappearance in these body fluids were also determined after subcutaneous administration in seven week old calves. The peak serum and synovial fluid levels of trimethoprim/sulfadiazine achieved following oral administration were substantially lower with increasing maturation. In ruminating (six and seven week old) calves, subcutaneous or oral administration of the combination led to high serum levels of sulfadiazine but little or no serum trimethoprim was detected in animals at this age. The data indicate that, while therapeutic concentrations and optimum ratios of the drugs may be achieved for extended time periods in neonatal life, this dosage is unable to produce optimum serum and synovial fluid concentrations as the calves mature.  相似文献
