A total of 52 faba-bean (Vicia faba L.) fields, located in the main growing areas in Morocco were surveyed for viruses. From 240 samples with symptoms suggestive of virus infection, the following viruses were detected using electron microscopy, serology, and biological indexing: Alfalfa mosaic virus (AMV), bean yellow mosaic virus (BYMV), broad bean mottle virus (BBMV), broad bean stain virus (BBSV), broad bean true mosaic virus (BBTMV), pea earlybrowning virus (PEBV), pea enation mosaic virus (PEMV), pea seed-borne mosaic virus (PSbMV), and a complex of luteoviruses including bean leafroll virus (BLRV). This is the first report of the occurrence of BBTMV, PEMV, PSbMV, and the luteoviruses (including BLRV) of faba bean in Morcco. The luteoviruses and BBMV were found to be the most prevalent. They were detected in 56 and 50%, respectively, of the surveyed fields; while AMV, BBSV, and PEBV were found in single fields only. The remaining viruses were less prevalent, and were detected in a range of 4 to 15% of the fields surveyed. The incidences per field of the prevalent viruses varied and ranged from 1 to 33% for BBMV and up to 20% in the case of luteoviruses. BBMV was found confined to the central and northern parts of the country, BBTMV and PEMV mainly occurred in the central area, while the luteoviruses and BYMV were spread over the faba-bean growing regions of the country.  相似文献

Commercial capsicum fields in Lusaka Province were surveyed for the presence of viruses during 1996-01/06. The survey demonstrated the occurrence of five viruses in 13 fields. Virus identification was based on field disease syndrome, host range studies, DAS-ELISA and electron microscopy. The identified viruses were alfalfa mosaic alfamovirus (AMV), potato Y potyvirus (PVY), tobacco mosaic tobamovirus (TMV), cucumber mosaic cucumovirus (CMV) and pepper mild mottle tobamovirus (PMMV). The most prevalent and widespread virus, recovered from 70% of the surveyed fields, was AMV, and this was followed by PVY and TMV, which were detected in 50% and 40% of the fields respectively. CMV and PMMV were less common and occurred in 30% of fields. The study also showed that PVY could infect capsicum plants alone or in combination with CMV or TMV. The average plant infections caused by AMV was 80–100% and by TMV and PVY 50% and 60% respectively. The disease incidence in the case of CMV was 15–60% and for PMMV 30–40%. This is a first report on the identification of viruses that infect capsicum in Zambia.  相似文献

Serological tests have been used to detect viruses associated with tomato in 257 samples collected in different regions in Tunisia, Cap-Bon, Sahel and South during successive seasons. The viruses detected were cucumber mosaic cucumovirus (CMV) and tomato aspermy cucumovirus (TAV), potato Y potyvirus (PVY), tobacco etch potyvirus (TEV) and pepper veinal mottle potyvirus (PVMV), tomato mosaic tobamovirus (ToMV) and tobacco mosaic tobamovirus (TMV), tobacco rattle tobravirus (TRV), alfalfa mosaic alfamovirus (AMV), tomato spotted wilt tospovirus (TSWV), tomato ringspot nepovirus (TomRSV) and potato X potexvirus (PVX). Some were detected in all three regions surveyed, at variable frequencies: TMV, CMV, TEV, PVY, ToMV, AMV, TAV, TSWV and TRV. Others were only detected in two regions (PVMV in Cap-Bon and Sahel and PVX in Sahel and in the south) or one region (TomRSV in Cap-Bon). Movement of individual viruses from one region to another may be due to movement of specific vectors, as in the case of the thrips-transmitted TSWV moving from the south to the north. Some of these viruses were found for the first time in Tunisia.  相似文献

 1986年至1990年,从豌豆田中采集了150余份病毒病样本,鉴定出蚕豆萎蔫病毒(BB-WV)、芜菁花叶病毒(TuMV)、马铃薯Y病毒组分离物、黄瓜花叶病毒(CMV)、莴苣花叶病毒(LMV)、大豆花叶病毒(SMV)、豌豆花叶病毒(PMV)、菜豆黄花叶病毒(BYMV)和苜蓿花叶病毒(AMV)等9种病毒。样本中,BBWV所占的比例最高,达59.2%,其次为CMV,占15.5%。BBWV常与CMV复合侵染豌豆,LMV发生也较普遍。田间调查表明,豌豆病毒病发病率因种植地区及品种不同而有差异,平均发病率为12.4%。  相似文献

The incidence, severity and distribution of six viruses infecting capsicum were determined in the main growing areas of Turkey during the 2004 growing season. The surveys covered 50 randomly selected capsicum fields from four different areas in south-east Anatolia and the eastern Mediterranean region. 515 samples were individually collected and tested by DAS-ELISA for Cucumber mosaic cucumovirus (CMV), Alfalfa mosaic alfamovirus (AMV), Potato X potexvirus (PVX), Potato Y potyvirus (PVY), Pepper mild mottle tobamovirus (PMMoV) and Tobacco etch potyvirus (TEV). 64.8% of ELISA-tested capsicum samples (334 out of 515) were infected by one (41.7%) or more (23.1%) viruses. PVY was the most widespread (26.4%), followed by PVX (25.8%), AMV (25.2%), TEV (23%), PMMoV (9.1%) and CMV (8.3%). Surprisingly high AMV infection was found in three areas (Kahramanmaraş, Şanlıurfa and Gaziantep) where AMV is reported for the first time in this study. However, AMV was not detected in Hatay. Field surveys were carried out in the main vegetable-growing areas of Western and Central Albania to evaluate the sanitary status of open-field and protected cultivations of capsicum, tomato, potato, watermelon, cucumber, courgette, aubergine, lettuce, cabbage, chicory, leek and celery. The following viruses were detected: Alfalfa mosaic alfamovirus (AMV), Cucumber mosaic cucumovirus (CMV), Potato Y potyvirus (PVY), Tomato spotted wilt tospovirus (TSWV) and Watermelon mosaic potyvirus 2 (WMV-2). The virus found most frequently was CMV and all the isolates identified were of subgroup IA. AMV was also detected in several areas and all isolates were of subgroup II, suggesting a French origin. Finally, at the time of this survey, TSWV infections appeared to be moderately relevant and absent in protected crops. This is the first documented record of AMV, CMV, PVY, TSWV and WMV-2 in Albania. No infection by Tomato yellow leaf curl begomovirus (TYLCV) or Zucchini yellow mosaic potyvirus (ZYMV) was detected during this survey.  相似文献

A potyvirus that induced stunting and a characteristic bushy appearance at the apical region, due to proliferation of terminal branches with narrowed, reduced and deformed leaflets, was isolated from chickpea in India. The virus was sap-transmissible to 14 species of Chenopodiaceae, Leguminosae, Solanaceae and Malvaceae; Chenopodium amaranticolor was a good local lesion host. Virus particles, trapped by immunosorbent electron microscopy and stained with uranyl acetate, were 710 ×10 nm long. Purified virus preparations contained a single polypeptide species of 32,500 Da and one nucleic acid species of 3.1 · 106 Da. The virus was serologically related to soybean mosaic, azuki bean mosaic and peanut mottle viruses but not to clover yellow vein, pea seed-borne mosaic and bean yellow mosaic viruses.
On the basis of these properties, the virus was identified as a previously undescribed potyvirus in chickpea, for which the name chickpea bushy dwarf virus is proposed.  相似文献

Nine tobacco fields of small- and large-scale farmers in Central, Lusaka and Southern provinces of Zambia with an experimental area in the range of 4–52 ha were surveyed for the incidence, prevalence and identification of virus diseases during the growing season of 1997. Samples were collected from three tobacco fields in each of the three provinces, and a total of 72 samples was analysed. Virus identification was based on field disease syndrome, host range studies, DAS-ELISA and electron microscopy of virus particles in some cases. The study demonstrated the occurrence of Tobacco mosaic virus (TMV), Potato virus Y (PVY), Alfalfa mosaic virus (AMV) and Tobacco ringspot virus (TRSV). TMV and PVY occurred widely and were common in all three provinces, while AMV and TRSV were relatively less common. The prevalence of the four viruses was TMV 78%, PVY 67%, AMV 33% and TRSV 22%. Serological tests for Tomato spotted wilt virus (TSWV) and Cucumber mosaic virus (CMV) showed that these viruses were not present in the tobacco samples analysed.  相似文献

In a survey for viruses of cultivated legumes in West Asia and North Africa, pea seed-borne mosaic virus (PSbMV) was found in faba bean, lentil and pea. Using ELISA, it was detected in 107 out of 1554 faba bean samples and 40 out of 496 lentil samples with virus-like symptoms collected in Algeria, Egypt, Ethiopia, Jordan, Lebanon., Libya, Morocco, Sudan, Tunisia and Turkey.A pea isolate (SP9-88) from Syria was further characterized. Out of 57 plant species tested, 35 were found susceptible, 19 of which are newly reported hosts of the virus. The virus was transmitted efficiently in the non-persistent manner by five aphid species, especiallyMyzus persicae. Purification from systemically infected faba bean plants yielded 10–15 mg of purified virus per kg of infected tissue. Sap-inoculation of the food and forage legume species chickpea, faba bean, lentil, pea,Vicia narbonensis, V. sativa, Lathyrus ochrus andL. sativus at flowering stage led to 66.0, 40.5, 44.6, 49.2, 31.7, 7.5, 35.7 and 12.0% yield loss, respectively, and to seed-transmission, rates of 0.7, 6.0, 10.8, 1.1, 0.3, 0.2 and 0.4%, respectively. No transmission was detected in chickpea seed embryo axes. However, the virus was detected in the seed coat of SPbMV-infected chickpea at an estimated rate of 1.81%.  相似文献

Biological indexing of faba-bean samples collected during an earlier virus survey in Morocco revealed variation in symptom severity among isolates of broad bean mottle virus (BBMV). When seven selected isolates from Morocco and three from Algeria, Sudan, and Tunisia were further compared, they could be divided into mild, severe, and intermediate isolates, according to their pathogenicity on a number of food-legume genotypes tested. The Moroccan isolate SN1 and the Sudanese SuV256 were very mild, and deviant also in their effect onGomphrena globosa, whereas the Tunisian TV75-85 and the Moroccan VN5 were virulent. Representative isolates were indistinguishable, however, in coat-protein molecular weights, and they reacted similarly to the antisera to a Moroccan and a Syrian isolate in electro-blot immunoassay.Promising ICARDA breeding lines and accessions—ten each of pea and lentil, nine of chickpea, and twelve of faba bean-were all found vulnerable (susceptible and sensitive) to all isolates. Within each food-legume species, vulnerability varied from high to moderate, and no immunity was detected. Virus concentrations in faba-bean lines suggest that isolates differ in virulence rather than in aggressiveness, and that the differences in vulnerability among the lines are due to differences in sensitivity rather than in susceptibility.When pooled seed samples were germinated and seedlings were tested for BBMV in DASELISA, the virus was found seed-transmitted in faba bean, chickpea, and pea at transmission rates of ca 1.2, 0.9, and 0.1%, respectively. This is the first report on seed transmission of BBMV in faba bean, when occurring on its own, and the first record of such seed transmission in chickpea and pea.  相似文献

2012年6月至2013年9月对甘肃省宕昌县、漳县、岷县、渭源县、陇西县、临洮县等地区种植的具有疑似病毒病症状的半夏等18种药用植物进行调查及采样。采用黄瓜花叶病毒(CMV)、苜蓿花叶病毒(AMV)、烟草花叶病毒(TMV)、番茄花叶病毒(ToMV)、马铃薯Y病毒(PVY)、马铃薯X病毒(PVX)、芜菁花叶病毒(TuMV)等7种双抗体夹心免疫酶联检测(DAS-ELISA)试剂盒初检, 并对CMV和ToMV采用反转录PCR(RT-PCR)方法复检, 结果表明, 半夏、掌叶大黄和红花3种药用植物受到黄瓜花叶病毒(CMV)侵染; 马兜铃、土贝母及当归3种植物受到番茄花叶病毒(ToMV)侵染; 样品中未检测到上述其他病毒种类, 其余12种病毒样品的病原尚未确定。本研究为国内首次报道CMV病毒侵染掌叶大黄、红花, ToMV病毒侵染马兜铃、当归和土贝母。此研究为防治上述病毒病提供了依据。  相似文献

A luteovirus isolate infecting chickpea in Morocco was experimentally transmitted by Myzus persicae to Physalis floridana, on which it produced mild symptoms. When tested in western blots against antisera to known legume luteoviruses, this isolate reacted strongly to beet western yellows virus (BWYV) antiserum, moderately to bean leafroll virus antiserum, while no reaction was recorded with the antiserum against subterranean clover red leaf virus. In PCR, a fragment of ca. 950 bp was amplified, comprising the 3' end of the open reading frame (ORF) 3, the complete coat protein gene (ORF 4), and the non-translated region in between these ORFs. The nucleotide sequence of the amplified fragment showed high similarity with BWYV (approximately 96%), and lower (50–60%) with other luteoviruses reported to infect legumes. On the basis of these data, the Moroccan isolate was identified as BWYV. This is the first molecular evidence for the occurrence of BWYV on chickpea in Morocco, and on food legumes in general in North Africa.  相似文献

The presence of viruses in the weedLeonotis nepetaefolia in central México is reported from two field surveys.L. nepetaefolia, with viral-like symptoms such as mosaic, leaf deformation and calico, was observed growing next to cultivated fields in the Valley of Atlixco, Puebla, an important agricultural region in Mexico. The viruses harbored by this plant were characterized biologically, serologically and by molecular methods. The viruses detected wereAlfalfa mosaic virus (AMV),Cucumber mosaic virus (CMV), a satellite RNA of CMV (CMV satRNA) andTobacco mosaic virus (TMV). This last one was detected only during the first survey. CMV was the predominant virus found in both surveys, and was associated mostly to mosaic symptom. Phylogenetic analysis based on the coat protein gene sequence of CMV indicated that this isolate belongs to subgroup IA and confirmed that it is a mosaic-inducing isolate, whereas AMV belongs to subgroup II. Finally, CMV satRNA was found to be a non-necrogenic ameliorative variant, both by symptomatology and by phylogenetic analysis. Agar double diffusion tests and, later, ELISAs were used to detect viruses associated with potato in 242 samples collected in 16 trips to Tabuk and Hail, northern regions of Saudi Arabia, in four consecutive growing seasons (autumn 1989, spring and autumn 1990 and spring 1991). Eleven different viruses were detected in Tabuk and 12 in Hail. The viruses detected in Tabuk were alfalfa mosaic (AMV), cucumber mosaic (CMV), tobacco mosaic (TMV), potato leaf roll (PLRV), tomato spotted wilt (TSWV), tobacco ringspot (TRSV) and potato A, M, S, X and Y. The same viruses, plus potato yellow dwarf (PYDV), were detected in Hail. AMV was most frequently and CMV least frequently detected in Tabuk, whereas in Hail the most and least common were PVA and PLRV respectively.  相似文献

Pepper production is affected by several viral diseases in Samsun, Turkey. To determine the identity of these viruses, a total of 313 samples from field-grown peppers were collected during surveys in 1998 and 1999, and tested by enzyme linked immunosorbent assay (ELISA). Six viruses,Alfalfa mosaic virus (AMV),Cucumber mosaic virus (CMV),Potato virus Y (PVY),Tomato mosaic virus (ToMV),Tobacco mosaic virus (TMV) andTomato spotted wilt virus (TSWV) were detected in the samples. Of 313 plants tested, 42 were doubly infected, and TMV+PVY (15.4%) was the most common double infection. This is also the first report of AMV in pepper fields in Turkey. The effect of some weed species that may act as reservoir of these viruses was also investigated in the region and of 24 weed species belonging to 15 families tested, 16 were found to be infected with at least one virus.Amaranthus retroflexus (Redroot pigweed) appeared to be a common host of CMV, PVY, ToMV, TMV and TSWV, whereasHibiscus trionum (Venice mallow) was recorded as a new weed host of PVY and TSWV. The majority of weed species found to be virus infected were very common in the pepper growing areas of the region. An account is given of strain‐specific diagnostic methods currently adopted in Italy to detect alfalfa mosaic alfamovirus (AMV), cucumber mosaic cucumovirus (CMV) and tomato spotted wilt tospovirus (TSWV). These viruses are highly detrimental to tomato crops as the introduction of new virus strains has deeply changed the population structure of resident viruses. RT‐PCR coupled with restriction analysis of amplicons proved suitable for differentiating CMV and AMV strains whereas single‐stranded conformational polymorphism (SSCP) was used for the identification of TSWV isolates. Application of strain‐specific diagnostic methods to phytosanitary inspection of plant propagules is suggested as a powerful tool for timely identification of new virus strains  相似文献

Alstroemeria plants were surveyed for viruses in Japan from 2002 to 2004. Seventy-two Alstroemeria plants were collected from Aichi, Nagano, and Hokkaido prefectures and 54.2% were infected with some species of virus. The predominant virus was Alstroemeria mosaic virus, followed by Tomato spotted wilt virus, Youcai mosaic virus (YoMV), Cucumber mosaic virus (CMV), Alstroemeria virus X and Broad bean wilt virus-2 (BBWV-2). On the basis of nucleotide sequence of the coat protein genes, all four CMV isolates belong to subgroup IA. CMV isolates induced mosaic and/or necrosis on Alstroemeria. YoMV and BBWV-2 were newly identified by traits such as host range, particle morphology, and nucleotide sequence as viruses infecting Alstroemeria. A BBWV-2 isolate also induced mosaic symptoms on Alstroemeria seedlings.  相似文献

Chickpea is one of the major legume crops in the northern parts of Algeria, with 60 000 ha sown. The different viral diseases observed on this crop are partially responsible for the low yields obtained. Of the viruses involved, bean yellow mosaic potyvirus (BYMV), associated with 'narrow-leaf disease, has been identified in the central parts of the country since 1994. The host range was screened and found to include species of Fabaceae, Solanaceae and Cucurbitaceae. Three aphid species ( Acyrthosiphon pisum, Aphis craccivora and Aulacorthum solani ) have been shown to transmit the virus. Two chickpea cultivars (ILC 3279 and ILC 482) carry BYMV in the seed cotyledons. The virus has been purified, antiserum has been produced and electrophoretic analysis has been conducted.  相似文献

In surveys conducted in 2011 and 2012 to identify the viruses causing diseases on pepper and tomato in the department of Alibori in northern Benin, 451 samples of pepper and tomato were analyzed by ELISA using 11 specific antibodies. The highest virus incidence among the surveyed districts was recorded on pepper in Malanville (56.18%), followed by Karimama (39.32%). The most frequently found viruses were Pepper veinal mottle virus (PVMV), Cucumber mosaic virus (CMV), and Potato virus Y-necrotic (PVY-n), accounting respectively for 22.39%, 21.73% and 15.96% of the collected samples. Tomato yellow leaf curl virus (TYLCV) was detected in only 2.43% of the samples, whereas Alfalfa mosaic virus (AMV), Chilli veinal mottle virus (ChiVMV), Tomato mosaic virus (ToMV) and Tomato spotted wilt virus (TSWV) were not detected in any of the samples tested. Double and triple infections involving different virus combinations were found, respectively, in 14.86% and 4% of the samples. Five plant species (Euphorbia hirta Linnaeus, Moringa oleifera Lam, Leucas martinicencis (Jacquin) R. Brown, Combretum micranthum G. Don, Abelmoschus esculentus L. Moench.) out of 30 samples belonging to 13 botanical families, collected within or nearby tomato and pepper fields, were found infected with PVMV, PVY-n, and CMV. Control measures to reduce the impact of viruses on pepper and tomato production are discussed. This is the first report of viruses infecting pepper and tomato in Benin.  相似文献

Mosaic on leaves, necrosis, stunting and flower symptoms were observed in lisianthus ( Eustoma russellianum ) raised from seed. Three viruses, bean yellow mosaic (BYMV), cucumber mosaic (CMV) and tobacco mosaic (TMV), were isolated.
