Seedlings of maritime pine (Pinus pinaster Ait.) were inoculated with different dikaryons of Pisolithus sp. from South Africa to determine the influence of extension of the extramatrical phase and diameter of the mycelial strands on water relations parameters including xylem water potential (Psi(x)), soil water potential at the soil-root interface (Psi(s)) and hydraulic conductance (L(p)) during and after a period of water stress. Seedlings inoculated with dikaryons having an extensive extramatrical phase and large diameter mycelial strands showed higher Psi(s) (-2 MPa) during severe water stress than seedlings inoculated with dikaryons producing fine hyphae and sparse extramatrical phases (-3.8 MPa). Seedlings inoculated with strand-forming dikaryons recovered faster from water stress than did non-inoculated seedlings or seedlings inoculated with non-strand-forming dikaryons. Architectural aspects of the extramatrical phase, including the presence of large diameter mycelial strands or fine hyphae, influenced the soil-root contact and the water relations of an inoculated host plant. When water stress was not limiting, the architecture of the extramatrical phase did not have a large effect on Psi(s). It is suggested that the architecture of the extramatrical phase influences the resistance to water flow through the soil-root interface and that large mycelial strands increase the water flow by bridging the gap between the soil and the root. These changes in physiology indicate that dikaryons can improve the survival of Pinus pinaster under dry conditions.  相似文献

We conducted a study to find out if arbuscular mycorrhizal (AM) fungi (Acaulospora scrobiculata, Scutellospora calospora) and phosphate solubilizing bacteria (PSB, Paenibacillus polymyxa) inoculation either individually or in combinations can improve Acacia auriculiformis seedling growth, uptake of nutrients and quality in a phosphorus deficient tropical Alfisol. The seedlings were assessed for various growth and nutrient uptake parameters after 60 days of treatment. Inoculation with P. polymyxa stimulated mycorrhizal formation. Seedling height, stem girth, taproot length, number of leaves and leaf area, plant dry matter production, nodulation, and nodular dry weight were significantly higher for seedlings that were either dual inoculated or triple inoculated compared to individual inoculation of AM fungi or PSB, and uninoculated seedlings. Dual and triple application of AM fungi and PSB also significantly improved the nutrient contents of shoots and roots and nutrient uptake efficiencies. The calculated seedling quality indexes of the AM fungi and PSB inoculated seedling were 25–208% higher than uninoculated seedlings. These findings show that A. auriculiformis seedlings when dual inoculated or triple inoculated performed better than seedlings inoculated with the microbes individually and compared with uninoculated control seedlings. We conclude that bioinoculation is important for the production of high-quality A. auriculiformis seedlings in tree nurseries for planting in nutrient deficient soils.  相似文献

以接种褐环乳牛肝菌和未接种的马尾松幼苗为试验材料,采用水培法研究不同浓度铝(Al^3+)处理(0、0.2、0.4、0.8和1.2 mmol·L^-1)对菌根化和非菌根化马尾松幼苗生长、营养元素吸收的影响,揭示菌根化马尾松苗木对Al^3+的适应机制,为减轻马尾松苗木铝毒害现象提供理论和技术依据。结果表明,在相同浓度Al^3+胁迫下,接种褐环乳牛肝菌的马尾松幼苗苗高、生物量、根系形态以及对氮(N)、磷(P)、钾(K)的吸收能力均优于不接种的马尾松苗。马尾松幼苗苗高、干物质量积累以及根系生长随着Al^3+浓度的增大均呈现先升后降再升的趋势,随着Al^3+浓度的逐渐增大,马尾松幼苗根、茎、叶中的N、P、K含量呈现出低促高抑的趋势,根、茎、叶中Al^3+含量的分配为根>茎>叶。Al^3+胁迫下,接种褐环乳牛肝菌的马尾松幼苗,通过改变马尾松幼苗根系结构,促进了幼苗的生长,特别是根系的生长,以及对N、P、K等营养元素的吸收,抑制了根系吸收Al^3+,提高了马尾松幼苗的耐铝能力。  相似文献

试验表明接种固氮菌能显著提高桉树苗木的生长及苗木各器官的生物量和N含量,接种苗木高生长比对照(不接种)增加29.81%、叶片数增加37.31%、叶片干重增加35.01%、茎干重增加55.59%、根干重增加78.93%、总生物量干重增加55.89%。不同菌株的增产效果存在差异,接种催娩克氏菌的效果较接种阴沟肠杆菌的好。接种+施N80mg/株处理的效果优于接种+施N30mg/株处理的效果。接种与施N肥的分析结果表明接种催娩克氏菌的效果等于施N80mg/株的效果。  相似文献

This study was conducted to compare the growth and mineral nutrient composition of Alnus hirsuta seedlings when treated with different aluminum concentrations and acidic nutrient solutions with and without Frankia inoculation and to examine microscopic changes in the roots treated with different aluminum levels with and without Frankia inoculation. When inoculated with Frankin, A. hirsura seedlings without A1 treatment showed the greatest growth rate in height; root length and fresh weight. The seedlings treated with Al grew better in height, root length and fresh weight in the presence of Frankia than those uninoculated with Frankia. As the concentration of Al treatments increased, the concentration of Al in both shoot and root increased regardless of Frankia inoculation. As the concentration of treated Al increased, the concentration of cations (Ca, Mg, K) in the tissue decreased. The root tissues treated with A1 showed the following microscopic characteristics as compared to those without Al treatment; presence of electron dense bodies in vacuoles, and between plasmalemma and cell wall, accumulation of phenolic materials in vacuoles, disrupted tonoplast, and increase in vacuolation. These alterations in root anatomy were observed at 50 mg/L Al treatment. The degree of alteration was more severe in the root tissues uninoculated with Frankia than those inoculated with Frankza.  相似文献

We tested the effects of ectomycorrhizal (ECM) inoculation on greenhouse-grown white spruce (Picea glauca (Moench) Voss) and jack pine (Pinus banksiana L.) seedlings to be used for revegetation of salt-affected tailing sands resulting from the exploitation of oil sand in northeastern Alberta, Canada. White spruce and jack pine seedlings were inoculated with three ECM fungi selected for their in vitro tolerance to excess Na+ and Cl-: Hebeloma crustuliniforme (Bull) Quel. UAMH 5247, Laccaria bicolor Maire (Orton) UAMH 8232 and a Suillus tomentosus (Kauff.) Sing., Snell and Dick isolate from a salt-affected site. The physiological responses of the seedlings to a gradient of NaCl concentration (0, 50, 100 and 200 mM) were assessed over four weeks by: (1) Na+ accumulation and allocation; (2) chlorophyll a fluorescence; (3) growth, (4) water content; and (5) organic osmolyte accumulation. Jack pine seedlings were more sensitive than white spruce seedlings to increasing Na+ and Cl- concentrations. Both species showed decreasing biomass accumulation, and increasing concentrations of organic osmotica and Na with increasing NaCl concentration. White spruce seedlings inoculated with the S. tomentosus isolate had the best growth response at all NaCl concentrations tested. Although jack pine seedlings inoculated with the L. bicolor or S. tomentosus isolate exhibited the highest growth in the 50 and 100 mM NaCl treatments, both fungi increased the photochemical stress and dehydration of their hosts in the 200 mM NaCl treatment. At the latter concentration, jack pine seedlings inoculated with H. crustuliniforme showed the greatest tolerance to salt stress. Although the different fungi altered the physiological response of the host in different ways, inoculation with salt-stress-tolerant ECM fungi increased growth and reduced the negative effects of excess NaCl. Use of controlled mycorrhization may increase survival of coniferous seedlings used for revegetation of salt-affected sites.  相似文献

9 mycorrhizal fungi, including 4 ectomycorrhizal fungi (ECMF) and 5 arbuscular mycorrhizal fungi (AMF), were used to explore their effect on seedling growth of Mytilaria laosensis. The study shows that M. laosensis is one of the species with both VAM and ECM. Mycorrhizal infection rate of all ECM inoculation methods reached level 3, and that of AMF infection rate was 88%-93%, of which AM91 became the highest. In different periods, different inoculation treatment expressed different effects on seedling growth. At the end of the experiment, the height, ground diameter, underground dry mass and upper ground dry mass of seedling inoculated with PX0801 and 9006 respectively increased by 71%, 45%, 128%, 184%, and 65%, 54%,150%, 208%, which exhibited the best overall effect. Coinoculated seedlings with AM90036, AM3008 and AM91 had advantages over uninoculated ones in all the tested growth indicators, which suggest to significantly promote the growth of M.laosensis. The results obtained provide reference for mycorrhizal fungi application on M. laosensis.  相似文献

用Frankia纯培养接种木麻黄苗木试验,获得了明显效果,苗高、地径、根瘤数、根瘤鲜重及生物量均比对照高。在4个供试菌株中,接种Br的效果最好,苗高、地径分别为对照的3.5倍和2.7倍,其次是P1。接种效果随着苗龄增加越来越明显。苗木的高度、地径、生物量与根瘤数量、根瘤重呈极显著的直线正相关。  相似文献

Sphagnum peat (peat), spruce bark compost (compost), peat +perlite (1 : 1, v : v) and compost + perlite (1 : 1, v : v)substrates were inoculated with vegetative alginate-bead inoculumof Hebeloma crustuliniforme, Hygrophorus agathosmus or Paxillusinvolutus or left uninoculated prior to the addition of Norwayspruce seed. Growth and percentage of mycorrhization of barerootseedlings cultivated in a greenhouse were evaluated after thefirst growing season. Seedlings grown in peat-based substrateshad significantly larger aboveground and total dry weight, butsignificantly lower mycorrhization percentage than those grownin compost-based substrates. There were no significant differencesbetween fungal treatments (including control) for both the percentageof mycorrhization and growth of seedlings. The artificiallyintroduced fungi were not efficient in mycorrhizal formation– naturally occurring fungi were common in all treatments.However, a significant interaction between substrate and fungustreatments in root dry weight was detected. Hebeloma-inoculatedseedlings grown in peat showed the highest value of root dryweight. This fact indicates other possible effects of the fungion root growth independent of mycorrhization. Growth parameterswere negatively correlated with the extent of mycorrhization,indicating allocation of host photosynthates to the fungi. Tothe best of our knowledge, operational inoculation of Norwayspruce seedlings with the test fungi has not been reported previously.  相似文献

The aim of this study was to test the pathogenicity of two Spanish isolates of Heterobasidion annosum sensu stricto in 2‐year‐old Pinus pinaster seedlings. Two types of inocula (woodchips and sawdust) were used to infect the seedlings by two different routes (stem inoculation and soil infestation). The mortality rates of the stem‐inoculated seedlings differed significantly from controls, but those of the seedlings infected via soil infestation did not differ. For both types of inoculation, the lesions were longer, and wilting symptoms were more severe in the seedlings inoculated with H. annosum than in control seedlings. For stem inoculation, biomass allocation did not differ significantly between the infected and control seedlings. However, the percentage of fine roots was lower in seedlings infected via soil infestation than in the control seedlings. To our knowledge, this is the first pathogenicity test with H. annosum isolates and P. pinaster.  相似文献

An investigation was carried out to screen and select efficient vesicular arbuscular mycorrhizal (VAM) fungi for inoculating the forest tree species, Casuarina equisetifolia. The seedlings were inoculated with 10 different VAM fungi, obtained from various sources. Inoculated seedlings generally had greater plant height, stem girth, biomass and P content than uninoculated plants. They also had more mycorrhizal root colonization and spore numbers in root zone soil. C. equisetifolia seedlings responded best (in biomass) to inoculation with Glomus mosseae (Nicolson and Gerdemann) Gerdemann and Trappe, closely followed by Acaulospora laevis Gerdemann and Trappe and G. fasciculatum (Thaxter Sensu Gerdemann) Gerdemann and Trappe; all the three being statistically on par with each other.  相似文献

At germination, container-grown shortleaf pine seedlings were inoculated with Pisolithus tinctorius (Pers.) Coker & Couch or left uninoculated, and both groups were fertilized semiweekly with a modified Hoagland's solution supplemented with 0 or 0.4 mM boric acid. After 12, 16 and 24 weeks, seedling root tissue was analyzed for ectomycorrhizal colonization, phenolic concentration and phenoloxidase activity. In addition, phenoloxidase activity was assayed in P. tinctorius that had been cultured in a liquid medium containing boric acid. Inoculation with P. tinctorius increased the root phenolic concentration of 16- and 24-week-old seedlings, and increased root phenoloxidase activity in 12-, 16- and 24-week-old seedlings. Fertilization with boric acid reduced the phenolic concentration of P. tinctorius ectomycorrhizae after 24 weeks. Although boric acid fertilization did not affect the phenoloxidase activity of 12-, 16- and 24-week-old inoculated roots, or that of 16- and 24-week-old uninoculated roots, it increased the phenoloxidase activity of P. tinctorius grown in vitro and 12-week-old uninoculated roots. We conclude that boric acid fertilization influences the phenolic relations of the shortleaf pine-P. tinctorius ectomycorrhizal association, possibly through a boric acid-induced increase in phenoloxidase activity.  相似文献

The effects of two fungicides (benomyl and captan ‐ at recommended doses and up to three soil drenches) on root development and mycorrhizal colonization of Sitka‐spruce (Picea sitchensis) and ash (Fraxinus excelsior) were examined after 20 weeks growth, under environmentally controlled conditions, in soil from an Irish tree nursery. Although four mycorrhizal types have been found on Sitka‐spruce at the nursery, only one ectomycorrhizal type (Piceirhiza horti‐inflata) was identified on the short roots in this study. An inoculant (Vaminoc: MicroBio Ltd, Hemel Hempstead, UK) was used to inoculate ash and 20‐week‐old seedlings had a higher arbuscular mycorrhizal (AM) colonization in comparison with uninoculated controls. Multiple applications (2–3) of benomyl reduced the length of root and shoot and shoot dry mass of Sitka‐spruce, whereas in ash, it only depressed root length. Benomyl decreased the numbers of ectomycorrhizas of Sitka‐spruce and arbuscular mycorrhizal colonization of Vaminoc‐inoculated ash. A single application of captan stimulated root length and ectomycorrhizal colonization of Sitka‐spruce and root dry mass in ash compared with uninoculated controls. Applications of captan reduced arbuscular mycorrhizal colonization of Vaminoc‐inoculated ash to levels near to those of uninoculated controls. Of the two fungicides used, benomyl had the most deleterious effect on root length and mycorrhizal colonization of Sitka‐spruce and ash.  相似文献

The effect of ectomycorrhizae on damage caused by Otiorhyncus larvae and on plant growth was evaluated in a 3-yr field experiment. Russian larch seedlings, inoculated with ectomycorrhizal fungi, were compared with uninoculated plants. Assessment of larval damage at the end of the first growing season showed a significant effect of treatment on damage intensity, but not on damage incidence: 11.2% of uninoculated seedlings were severely damaged, but only 3.5% of inoculated plants. There was a significant effect of treatment on plant mortality. The accumulated mortality at the end of the third growing season was 34.5% for uninoculated plants, but 17.6% for inoculated plants. The height increment of inoculated plants (4.5 cm) was significantly greater than that of uninoculated plants (2.7 cm) in the first growing season. No difference in height increment between treatments was observed during the second and third growing seasons.  相似文献

We investigated the effects and interaction of four irradiances, 37, 53, 70 and 100% full sunlight, and the endomycorrhizae Glomus macrocarpum and Glomus fasciculatum on the development of seedlings of Fraxinus pennsylvanica Marsh. At all irradiances, mycorrhizal seedlings were taller and larger in diameter than nonmycorrhizal seedlings. Seedlings inoculated with G. macrocarpum showed the best height growth at 70% or less of full sunlight, whereas seedlings inoculated with G. fasciculatum grew best at 37 and 53% full sunlight. Leaf area was greatest in shaded seedlings and was enhanced by mycorrhizal inoculation. Shoot/root ratios were greater for mycorrhizal than for nonmycorrhizal seedlings. At intermediate light intensities, percent root length colonized was greatest, whereas the concentrations of soluble sugar and starch in roots were at intermediate levels.  相似文献

Abstract

Individual and interactive effects of simulated acidic rainfall and mycorrhizal inoculation on growth and nutrient and water relations of loblolly pine (Pinus taedaL.) and white oak (Quercus albaL.) grown in a loam soil were examined. Seedlings of each species inoculated with basidiospores of the ectomycorrhizal fungus Pisolithus tinctorius(Pers.) Coker and Couch, a known my-cobiont of both loblolly pine and white oak, and uninoculated control seedlings received two simulated rains per week of either pH 3.6, 4.2, or 4.8 for 26 weeks. Higher acidity rainfall reduced the growth but increased mycorrhizal colonization of loblolly pine, while both loblolly pine and white oak exposed to these rains exhibited greater foliar injury. Inoculation with P. tinctoriusincreased growth and reduced foliar injury of both species. Foliar concentrations of P, S, and Cu in loblolly pine and white oak, Ca in loblolly pine, and Fe and Zn in white oak decreased with increasing rain acidity while the Al concentration of both species increased. Higher rainfall acidity also reduced soil pH and Ca and Mg concentrations while increasing soil AI. Foliage of inoculated seedlings of both species had higher N and P concentrations and lower Al concentrations than control seedlings. Following the final rain applications, a drought cycle was simulated by withholding irrigation for two weeks during which seedling xylem pressure potential and soil water potential were measured. One day after cessation of irrigation, xylem pressure potential of loblolly pine that had received pH 3.6 rains was lower than that of other treatments. Thereafter, xylem pressure potential and soil water potential of the inoculated treatment decreased below those of the control treatment in both species. These results suggest that acid deposition is detrimental to juvenile loblolly pine and white oak, but the magnitude of this effect is less than the positive response to ectomycorrhizal inoculation.  相似文献

接种苗木在生长量和生物量上均表现出极其显著的生长优势,ECM真菌蜡蘑菌单接种及其与4种VA菌根菌剂混合接种,对苗木生长的促进作用尤为显著。在接种后16周时,与对照苗相比,蓝桉和尾叶桉接种苗木的高生长量最大增幅分别为28.86%(LS)和86.65%(LG);两种桉树地上部分平均干质量最大增幅分别为129.93%(LS)和133.34%(L),地下部分分别为119.93%(LF)和174.83%(LG)。桉树对不同菌根菌及其组合的菌根依赖性表现出很大的差异,对混合接种的真菌组合有相对高的菌根依赖性,而对土壤菌剂依赖性很小。菌根感染与苗木生物量间有显著的相关性。试验结果还证实了P素对桉苗生长的促进作用,P素水平对菌根的接种效应有一定的影响。  相似文献

Simulated acid rain (SAR) at three pH levels (pH 4, 3, 2) was applied to only the top or both the top and roots of 4-month-old Japanese black pine seedlings repeatedly for two months. Then the seedlings were inoculated with a virulent isolate (S10) of pinewood nematode,Bursaphelenchus xylophilus. The exposure to SAR at any level did not kill the seedlings. When inoculated with nematodes, however, the nematode population build-up was significantly higher in the seedlings pretreated with SAR at any level than in the control at the 7th and 17th day after inoculation, and subsequent disease symptom development was also significantly accelerated by the exposure to SAR at pH 2 and 3. This result indicates that even acid rain at pH 4 has the potential for promoting population growth of pinewood nematodes in 4-month-old Japanese black pine seedlings.  相似文献
