The plasma inhibitor of carbonic anhydrase (CA) from rainbow trout was purified using ion exchange chromatography. The inhibitor has a high isoelectric point value (pI>10). SDS-PAGE electrophoresis and gel filtration demonstrated that the inhibitor is a low molecular weight compound of about 6,000 daltons. The plasma inhibitor was more effective against gill CA than against blood CA in vitro, probably reflecting the presence of various CA-isoenzymes in red blood cells and gill tissue. The apparent Root effect, i.e., the impairment of the oxygen binding capacity of hemoglobin in red blood associated with increased blood PCO 2was counteracted by the plasma inhibitor, probably by acting on membrane-bound and/or cytosolic blood CA. This interaction may be of importance in adaptive mechanisms, e.g., during the acidemic phase, when the fish is being acclimated to hypercapnic conditions.  相似文献

Twenty-three smooth dogfish, Mustelus canis L., and 20 spiny dogfish, Squalus acanthias (Mitchill), were examined for the presence of diseases. Sharks were collected from the north-western Atlantic between April and September 2000. Major organs were sampled for histopathology and bacterial cultures were taken from kidneys. Macroscopic lesions were infrequent and included intestinal cestodiasis and gastric and dermal erosions. Microscopic lesions were common in both shark species. Mustelus canis had numerous degenerative lesions involving the pancreatic ducts, seminiferous tubules and coronary vessels. The most frequent lesions in S. acanthias were parasitic and included pancreatic nematodiasis and biliary myxosporeosis. Additionally, both species had focal meningitis, encephalitis, dermatitis, gastritis, enteritis and glomerulomesangial thickening. Microscopic parasites included intestinal coccidiosis, unclassified gastric and testicular protozoa, skeletal muscle and renal tubular myxosporeans, a branchial trichodinid ciliate, olfactory and branchial trematodes, gastrointestinal cestodes and larval nematodes. Shewanella putrefaciens , Photobacterium sp., Vibrio sp., Pseudomonas sp. and Alteromonas sp. were isolated from kidneys of nine sharks. The role of the above lesions in the natural mortality and morbidity of the two shark species is unknown. The finding that apparently healthy sharks can harbour potentially debilitating lesions warrants the inclusion of histopathological studies in the management and conservation of sharks.  相似文献

The associations between spiny dogfish ( Squalus acanthias ) and hydrographic variables (temperature, salinity and depth) were examined in the Bay of Fundy and Scotian Shelf from 1970 to 1998. Data were obtained from standard groundfish bottom trawl surveys. Dogfish sex affected habitat associations. Males were found to occupy bottom water of significantly higher salinities and depths than that occupied by females. Length also significantly affected habitat associations. Smaller dogfish occupied relatively deep, high salinity bottom water compared with larger dogfish. Overall, the occupied temperatures, salinities and depths were significantly different from those which were available. Dogfish occupied warmer temperatures along a narrow range (6.62–9.19°C) compared with those which were available (1.57–9.35°C). Occupied salinity (32.70–34.43 ppt) and occupied depth (88.62–184.66 m) were also distributed along a narrower range than available salinity (32.16–34.79 ppt) and available depth (55.00–218.10 m). Sex-specific, length-specific and overall environmental preference by dogfish may bias traditional `offshore' groundfish surveys while large scale changes in hydrographic parameters may alter dogfish distribution and their interactions with other marine fauna.  相似文献

The North Pacific spiny dogfish (SPD), Squalus suckleyi, is a commercially exploited shark species that plays an important role in the ecosystem. To elucidate the distribution of the SPD in the North Pacific and to evaluate the effects of sea surface temperature (SST) and prey availability on its distribution, we estimated the probability of SPD presence using a generalized additive model with a binomial error distribution from SPD presence/absence data on 14,824 operations in fishery‐independent gillnet surveys between 1972 and 2011. The habitat model was structured in the east and west to reflect differences in the North Pacific oceanic environments. In the east, a higher probability of SPD presence was identified along the coast from the eastern Gulf of Alaska to Queen Charlotte Sound. In the west, it was identified around northern Japan. The estimated distribution was continuous between the two areas, whereas the probability of SPD presence was relatively low. Although the probability of SPD presence was higher at SSTs between 6°C and 12°C, the SST at the peak probability of SPD presence differed between the west and east. The prey species, Japanese sardine, Sardinops melanostictus, and walleye pollock, Gadus chalcogrammus, in the west and boreal clubhook squid, Onychoteuthis borealijaponica, in the east significantly affected the probability of SPD presence, which was higher if the prey species co‐existed with SPD. Therefore, SPD might adapt their distribution to that of available prey species. SPD stock assessment and management in these two important areas are required for its sustainable utilization.  相似文献

Gill Na+-K+ ATPase and carbonic anhydrase activities were measured, on a fortnightly basis, from February to July, in 0+ age Atlantic salmon (Salmo solar), hatched and reared in a freshwater experimental station in Covas, northern Portugal. Plasma osmolarity and ionic composition were also measured. Gill Na+-K+ ATPase activity increased slowly until April (15–19 moles Pi mg prot–1 h–1). From April to late May there was a great increase in activity (19–32 moles Pi mg prot–1 h–1) followed by a sharp decline in June (15 moles Pi mg prot–1 h–1). In contrast, carbonic anhydrase activity decreased significantly from early April to early June (170-70 moles p-nitrophenol mg prot–1 h–1) and increased in late June, suggesting the existence of a compensatory mechanism for the changes in Na+-K+ ATPase activity. Plasma osmolarity and sodium concentration showed lower levels during the period of high ATPase activity. On the other hand, plasma calcium concentrations showed an increase during the same period (3.47–5.98 mm1–1 of plasma). A transitory decrease in osmolarity and plasma sodium and chlorine concentrations occurred in March, prior to the surge in Na+-K+ ATPase activity, suggesting that the physiological changes, characteristic of parr-smolt transformation can be a consequence of this loss of freshwater osmoregulatory capacity.  相似文献

The pseudobranch in rainbow trout was investigated in order to clarify its metabolic and physiological function. Contents of lactate, glycogen, and glycerides and activities of carbonic anhydrase (CA), lactate dehydrogenase (LDH), and 3-hydroxy-acyl CoA dehydrogenase (HAD) were measured and compared with those in the gills and liver. Contents of lactate and glycerides, and activities of CA and HAD were higher in the pseudobranch than in the gills and liver. The results show that the pseudobranch is well equipped with enzymes (HAD and CA) suggested to be coupled to fatty acids breakdown and synthesis respectively. Lactic acid, possibly formed from glycogen and the glycerol component in glycerides, may be involved in the increase in blood P0 2 (Root effect) that is necessary to meet the oxygen needs of the eye. Pseudobranch carbonic anhydrase is probably involved in this process.  相似文献
