This study examined the effects of exposure to p-Nonylphenol (NP) on the motility of the spermatozoa of Japanese medaka (Oryzias latipes). Spermatozoa collected from adult males reared for 2 weeks in aqueous solutions of 20 and 100 μg/L NP showed similar swimming speeds but significantly lower percentages of motile spermatozoa than those of untreated controls. These results suggest that even a short exposure to NP might reduce significantly spermatozoan viability in fish.  相似文献

Glycoconjugates are known to be involved in many physiological events in vertebrates. Sialidase is one of the glycosidases, which removes sialic acid from glycoconjugates. In mammals, the properties and physiological functions of sialidases have been investigated, while there is little understanding of fish sialidase. Here, to investigate the significance of fish neu4 sialidase, neu4 gene was cloned from medaka brain mRNA and identified. Sialidase-specific motifs (GPG, YRVP and Asp-Box) were well conserved in the medaka neu4 polypeptide. Optimal pH of medaka neu4 sialidase was 4.6, but its activity was sustained even at neutral and weak alkaline pH. The neu4 considerably cleaved sialic acid from 4-methylumbelliferyl-N-acetyl-α-d-neuraminic acid and sialyllactose, but not from ganglioside and fetuin, which are good substrates for human NEU4. neu4 activity was mostly detected in mitochondria/lysosome fraction after biochemical fractionation, and indirect immunofluorescence assays revealed neu4 localization in lysosome in neu4 overexpressed cells. Next, developmental change in medaka neu4 and other sialidase mRNA levels were estimated by real-time PCR. Each sialidases showed different expression patterns in embryonic development: neu4 was up-regulated at late developmental stage in embryo, and neu3a mRNA level was quite high in 0.5 dpf. On the other hand, neu3b expression was drastically increased after hatching, suggesting that each sialidase may play a different role in embryonic development.  相似文献

Arachidonic acid (AA; 20:4n-6) is the precursor of a range of highly biologically active derivatives, collectively termed eicosanoids, including prostaglandins, thromboxanes, leukotrienes and lipoxins, that act as autocrine hormones regulating many physiological processes including haemostasis, reproduction, immune and inflammatory responses. Eicosapentaenoic (EPA; 20:5n-3) and dihomo-γ-linolenic (20:3n-6) acids modulate eicosanoid metabolism by both inhibiting the conversion of AA to eicosanoids whilst simultaneously being converted to eicosanoids with different, often attenuated, properties compared to their AA homologues. Eicosatetraenoic acid (20:4n-3) is a naturally occurring C20 polyunsaturated fatty acid (PUFA), present in fish oil at levels of around 1–2%, that has been suggested to be the active metabolite responsible for the anti-inflammatory effects of plant oils containing stearidonic acid (18:4n-3). However, the biochemical properties of 20:4n-3 in terms of cellular biology have rarely been investigated, partly due to difficulties in obtaining the fatty acid in high purity. In this paper, we describe methods for the medium scale laboratory preparation of high purity 20:4n-3, and investigate its metabolism in fish cell culture systems which normally contain significant amounts of n-3 PUFA. Thus the incorporation and metabolism of 18:4n-3 and 20:4n-3, and their distribution in phospholipid classes was studied in an established cell line from Atlantic salmon (Salmo salar) (AS), and the effects of 20:4n-3 on eicosanoid production studied in freshly isolated macrophages from rainbow trout (Oncorhynchus mykiss). Both 18:4n-3 and 20:4n-3 were preferentially esterified into phosphatidylcholine, phosphatidylethanolamine and phosphatidylserine in contrast with the accumulation of AA in phosphatidylinositol. Incorporated 18:4n-3 was readily converted to 20:4n-3, and both fatty acids were further desaturated and elongated to EPA and 22:5n-3 but not 22:6n-3. Supplementation with 20:4n-3 decreased the conversion of AA into prostaglandins, as demonstrated by the decreased levels of PGF produced in trout macrophages supplemented with 20:4n-3 and AA compared to cells supplemented with AA alone. In addition, 20:4n-3 was converted into eicosanoids in fish cells as indicated by the presence of Δ17,18 12-HETE, Δ17,18 PGE1 and Δ17,18 PGF in extracts from rainbow trout macrophages incubated with 20:4n-3.  相似文献

Abstract –  Temperature fluctuation has been shown to affect somatic growth rates and metabolic functions of fish, but little is known about the effects of fluctuating temperatures on life-history traits. We examined these effects on female Japanese medaka ( Oryzias latipes ) reared from hatch to maturity at two mean temperatures (27 and 30 °C) with diel temperatures either held constant or allowed to fluctuate ±5 °C. Feeding regimes were manipulated to equilibrate medaka growth rates in all treatments. Age and length at maturity decreased as a result of thermal fluctuation, whereas there was no effect of temperature fluctuation on the proportion of fish attaining maturity or on the number of eggs produced by a mature female. We hypothesise that short-term high temperatures associated with temperature fluctuation decreases development time, whereas reduced energy conversion efficiency at higher temperatures does not allow for increased egg production.  相似文献

给大菱鲆亲鱼投喂高蛋白和高水平 n 3HUFA的饲料。结果表明,与投喂低蛋白和低水平 n 3HUFA饲料组相比,大菱鲆亲鱼繁殖力明显提高。高水平试验组,亲鱼体重增重大,产卵量高、所产卵子卵径大,受精后上浮率、孵化率高,仔稚鱼的存活力强。其中卵子脂肪酸含量受亲鱼所摄饲料中脂肪酸含量的影响较大。当亲鱼组投喂试验组饲料的蛋白质为 49 3%、脂类含量为14 6%,其中∑n 3HUFA的比例为 29 3%时,平均每尾亲鱼的产卵量为 1 45×106,所产卵子的卵径为1 053 mm,卵子脂类∑n 3HUFA的比例为20 38%,浮性卵率为95%。经过30 d的饲育,大菱鲆苗种的存活率为 23%,明显高于低蛋白和低脂肪酸试验组的数据。本研究认为,大菱鲆亲鱼培育的饲料中适宜蛋白质含量应≥45%;脂类的适宜含量为≥10%,其中∑n 3HUFA的适宜比例≥20%。  相似文献

The objectives of this study were to determine the effects of the dietary docosahexaenoic acid (DHA) to arachidonic acid (ARA) ratio on the survival, growth, hypersaline stress resistance and tissue composition of black sea bass larvae raised from first feeding to metamorphic stages. Larvae were fed enriched rotifers Brachionus rotundiformis and Artemia nauplii containing two levels of DHA (0% and 10% total fatty acids=TFA) in conjunction with three levels of ARA (0%, 3% and 6% TFA). On d24ph, larvae fed the 10:6 (DHA:ARA) treatment showed significantly (P<0.05) higher survival (62.3%) than larvae fed 0:0 (DHA:ARA) (27.4%). Notochord length and dry weight were also significantly (P<0.05) greater in the 10:6 (DHA:ARA) treatment (8.65 mm, 2.14 mg) than in the 0:0 (DHA:ARA) (7.7 mm, 1.65 mg) treatment. During hypersaline (65 g L−1) challenge, no significant differences (P>0.05) were observed in the median survival time (ST50) between larvae fed 10% DHA (ST50=25.6 min) and larvae fed 0% DHA (ST50=18.2 min). The results suggested that black sea bass larvae fed prey containing 10% DHA with increasing ARA within the range of 0–6% showed improved growth and survival from first feeding through metamorphic stages.  相似文献

The effect of dietary 22:6n-3 (docosahexaenoic acid, DHA) on growth and survival was determined in striped trumpeter during metamorphosis and the Artemia-feeding period (16–36 days posthatch, dph). Artemia were enriched on one of five experimental emulsions that contained graduated concentrations of DHA and constant 20:4n-6 (arachidonic acid, ARA). We also compared larval performance using a commercial enrichment product high in n-3 PUFA. Final DHA concentrations in Artemia enriched on the experimental emulsions ranged from 0.1–20.8 mg/g DM, while Artemia fed the commercial product had 18.2 mg DHA/g DM. Each of the six diets was fed to larvae in four replicate 300-l tanks. Standard length (range 10.0–11.2 mm) and dry weight (range 1.6–2.5 mg) of larvae at the end of the experiment were directly related to dietary DHA, with the highest growth recorded in the experimental diet with the greatest concentration of DHA (20.8 mg/g DM). Survival at 36 dph was not influenced by dietary DHA and ranged from 20–44%. Mortality increased noticeably, regardless of dietary treatment, when larvae attained a standard length of approximately 9.5 mm. Mortality was related to a nocturnal behaviour where larvae would migrate to the tank bottom during the dark phase. Fatty acid profiles of the larvae were generally correlated to dietary fatty acids. Dietary DHA was found to be important in larval striped trumpeter growth, where enhanced growth probably shortened the critical period of metamorphosis and the window where nocturnal downward migration and mortality occurred.  相似文献

The incorporation and metabolism of (n-3) and (n-6) polyunsaturated fatty acids (PUFA) supplemented to growing cultures were studied in Atlantic salmon (AS) cells. A fatty acid concentration of 25 μM considerably altered the fatty acid composition of AS cells without increasing the neutral lipid content of the cells or inducing the production of cytoplasmic lipid droplets. Whereas Δ6 and Δ5 desaturase activities were significantly expressed in AS cells, Δ4 desaturase activity was very low. Both the Δ6 desaturase activity and the Δ5 desaturase activity showed some preference for (n-3) PUFA.  相似文献

为研究饲料中n-3长链多不饱和脂肪酸(n-3LC-PUFA)水平对花鲈幼鱼血清生化指标的影响,以初始体重(29.2±1.34)g的花鲈幼鱼为研究对象,分别投喂n-3LC-PUFA含量分别为0.13、0.30、0.56、1.03、1.45、2.41(%饲料干物质)的饲料,实验周期63d。结果表明,饲料中n-3LC-PUFA水平对花鲈幼鱼血清碱性磷酸酶(ALP)、乳酸脱氢酶(LDH)活性、总甘油三酯(TG)、总胆固醇(TC)含量无显著性影响(P0.05),而血清中肌酐(CREA)水平在饲料中n-3含量为1.45%时显著高于其余各组(P0.05)。结果表明,以血清CREA含量为标准,花鲈幼鱼饲料中n-3LC-PUFA含量应低于1.45%。  相似文献

Together with docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), arachidonic acid (ARA) is being considered to be an essential fatty acid in marine fish larval diets. The objective of the present study was to determine the importance of dietary ARA levels for larval European sea bass performance, when EPA and DHA are also present in the diet. Eighteen‐day‐old larvae were fed, for 14 days, gelatine‐based microdiets containing the following ARA levels: 0.3%, 0.6% or 1.2%. Elevation of dietary ARA up to 1.2% showed a positive correlation with larval survival and a significant improvement in the specific growth rates, body weight and total length. Arachidonic acid was efficiently incorporated into larval lipids, even at a higher proportion than that in the diets. Increased accumulation of ARA did not affect the incorporation of DHA or EPA from the diet into larval total lipids. A significant positive correlation was found between dietary ARA levels and survival after handling stress, indicating the importance of this fatty acid in sea bass larvae response to acute stressors. The results show the importance of ARA for sea bass larvae, but higher dietary levels should be tested to determine whether there is a negative effect of ARA in sea bass as reported for other species.  相似文献

Monounsaturated fatty acids (MUFA)-rich and n-6 polyunsaturated fatty acid (n-6 PUFA)-rich vegetable oils are increasingly used as fish oil replacers for aquafeed formulation. The present study investigated the fatty acid metabolism in juvenile European sea bass (Dicentrarchus labrax, 38.4 g) fed diets containing fish oil (FO, as the control treatment) or two different vegetable oils (the MUFA-rich canola/rapeseed oil, CO; and the n-6 PUFA-rich cottonseed oil, CSO) tested individually or as a 50/50 blend (CO/CSO). The whole-body fatty acid balance method was used to deduce the apparent in vivo fatty acid metabolism. No effect on growth performance and feed utilization was recorded. However, it should be noted that the fish meal content of the experimental diets was relatively high, and thus the requirement for n-3 long-chain polyunsaturated fatty acid (n-3 LC-PUFA) may have likely been fulfilled even if dietary fish oil was fully replaced by vegetable oils. Overall, relatively little apparent in vivo fatty acid bioconversion was recorded, whilst the apparent in vivo β-oxidation of dietary fatty acid was largely affected by the dietary lipid source, with higher rate of β-oxidation for those fatty acids which were provided in dietary surplus. The deposition of 20:5n-3 and 22:6n-3, as % of the dietary intake, was greatest for the fish fed on the CSO diet. It has been shown that European sea bass seems to be able to efficiently use n-6 PUFA for energy substrate, and this may help in minimizing the β-oxidation of the health benefiting n-3 LC-PUFA and thus increase their deposition into fish tissues.  相似文献

Copepod oil (CO) from the marine zooplankton, Calanus finmarchicus, is a potential alternative to fish oils (FOs) for inclusion in aquafeeds. The oil is composed mainly of wax esters (WE) containing high levels of saturated fatty acids (SFAs) and monounsaturated fatty alcohols that are poorly digested by fish at low temperatures. Consequently, tissue lipid compositions may be adversely affected in salmon‐fed CO at low temperatures. This study examined the lipid and FA compositions of muscle and liver of Atlantic salmon reared at two temperatures (3 and 12 °C) and fed diets containing either FO or CO, supplying 50% of dietary lipid as WE, at two fat levels (~330 g kg?1, high; ~180 g kg?1, low). Fish were acclimatized to rearing temperature for 1 month and then fed one of four diets: high‐fat fish oil (HFFO), high‐fat Calanus oil (HFCO), low‐fat fish oil (LFFO) and low‐fat Calanus oil (LFCO). The fish were grown to produce an approximate doubling of initial weight at harvest (220 days at 3 °C and 67 days at 12 °C), and lipid content, lipid class composition and FA composition of liver and muscle were determined. The differences in tissue lipid composition between dietary groups were relatively small. The majority of FA in triacylglycerols (TAG) in both tissues were monounsaturated, and their levels were generally higher at 3 °C than 12 °C. Polyunsaturated fatty acids (PUFA), particularly (n‐3) PUFA, predominated in the polar lipids, and their level was not significantly affected by temperature. The PUFA content of TAG was highest (~26%) in the muscle of fish fed the HFCO diet at both temperatures. Tissue levels of SFAs were lower in fish‐fed diets containing HFCO than those fed HFFO, LFFO or LFCO, particularly at 3 °C. The results are consistent with Atlantic salmon being able to incorporate both the FA and fatty alcohol components of WE into tissue lipids but, overall, the effects of environmental temperature on tissue lipids were more pronounced in fish fed the CO diets than FO diets.  相似文献

Herbivorous grass carp (Ctenopharyngodon idella) has been reported to exhibit low capacity to utilize high dietary lipid, but different lipid sources might affect this limited capacity. In order to compare the effects of different lipid sources with different lipid levels, juvenile grass carp were fed one of nine diets containing three oils [lard, plant oil mixed by maize and linseed oil, and n‐3 high unsaturated fatty acid‐enriched (HUFA‐enriched) fish oil] at three lipid levels (20, 60 and 100 g kg?1 dry diet) for 8 weeks. Decreased feed intake, poor growth performance, hepatic pathology and higher blood lipid peroxidation were found in 60 and 100 g kg?1 fish oil groups. Conversely, in lard and plant oil groups, even at 100 g kg?1 dietary lipid level, feed intake and growth performance did not decrease, despite histological observation revealed hepatic pathology in these groups. Plasma triglyceride and cholesterol contents increased significantly in all 100 g kg?1 dietary lipid groups. In the comparison of hepatic FA β‐oxidation among three oil groups at 60 g kg?1 dietary lipid level, impaired mitochondrial and peroxisomal FA oxidation capacity was observed in fish oil group. The results confirmed the relatively low capacity of grass carp to utilize high dietary lipid, and furthermore excess HUFA intake will result in more serious adverse effects than other FA.  相似文献

A growth trial was conducted to examine the effect of dietary digestible energy (DE) content on methionine (Met) utilization and requirement in juvenile Nile tilapia (Oreochromis niloticus). Ten iso‐nitrogenous (288 g kg?1 protein) practical diets, with two DE levels (10.9 MJ kg?1; 12.4 MJ kg?1) and five methionine supplementation levels (0, 1, 2, 4 and 6 g kg?1), were hand‐fed twice daily to triplicate groups of Nile tilapia (initial body weight 8.95 ± 0.06 g) for 8 weeks. Weight gain (WG) and specific growth rate (SGR) increased significantly with increasing dietary methionine concentration at the same DE content (< 0.001). At the same dietary methionine level, WG and SGR of fish fed high‐DE diets were significantly higher than that of fish fed low‐DE diets (= 0.0001), although no interaction was found between dietary DE and methionine supplementation. Based on quadratic regression analysis between dietary methionine concentration and weight gain, optimal methionine requirement for maximum growth, expressed as g Met required kg?1 diet (low‐ versus high‐DE diets), increased as diet DE concentration increased (7.34 versus 9.90 g kg?1 diet, respectively; with cysteine 4.70 g kg?1 diet). The results indicated that diet DE content affects methionine utilization and requirement in juvenile Nile tilapia, fish fed high‐DE diets required more methionine for maximum growth.  相似文献

The effects of dietary folic acid on biochemical parameters and gene expression of three heat shock proteins (HSPs) of blunt snout bream (Megalobrama amblycephala) fingerling under acute high temperature stress. Six dietary folic acid groups (0.0, 0.5, 1.0, 2.0, 5.0, and 10.0) mg/kg diets were designed and assigned into 18 tanks in three replicates each (300 l/tank) and were administered for 10 weeks in a re-circulated water system. The fingerlings with an initial weight of 27.0 ± 0.03 g were fed with their respective diets four times daily. At the end of the experiment, samples were collected before challenge, 0, 24, 72 h, and 7 days. Serum total protein (TP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), cortisol, glucose, complement C3 (C3), complement C4 (C4, immunoglobulin M (IgM) hepatic superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), and the expression of heat shock protein 60 (HSP60), 70 (HSP70), and 90 (HSP90) were studied. The results showed that fish fed with dietary folic acid between 1.0, 2.0, and 5.0 mg/kg significantly (P < 0.05) increased serum TP, C3, C4 hepatic SOD, CAT, and the expression of HSP60, HSP70, and HSP90 before and after temperature challenge of 32 °C. Also, serum ALP, cortisol, glucose, and hepatic MDA were significantly (P < 0.05) reduced by supplementation of dietary folic acid level 1.0, 2.0, and 5.0 mg/kg before and after the same temperature challenge of 32 °C. Before stress, 0, 24, 72 h, and 7 days significantly (P < 0.05) affects serum biochemical parameters, immune and antioxidant capacities, and expression level of three HSPs. Furthermore, there was no statistical evidence to show that dietary folic acid inclusion level and temperature duration have significant interactive effect on serum biochemical parameters, antioxidant parameters, and gene expression level (P > 0.05) of the three HSPs. However, there were statistical significant interactive effect between dietary folic acid inclusion level and temperature duration on serum C3 and C4 (P < 0.05) except IgM (P > 0.05). The present results indicate that supplementation of basal diet from 1.0 mg/kg; 2.0 and 5.0 mg/kg can enhance acute high temperature resistance ability in M. amblycephala fingerling to some degree and improve physiological response, immune and antioxidant capacities, and expression level of three HSPs.  相似文献

This study investigated in rainbow trout (Oncorhynchus mykiss) if diet composition and feeding level affect faecal bile acid loss, and whether this reflects on the apparent digestibility coefficient (ADC) of fat. Six diets were formulated with either fishmeal or plant protein as main protein source. This created a contrast in the supply of bile acids, the bile acid precursor cholesterol, taurine and the taurine precursors (methionine + cysteine) involved in bile acid conjugation. For both protein sources, three diets were formulated with increasing inclusion of a non‐starch polysaccharide (NSP)‐rich ingredient mixture (0.0, 82.0 and 164.2 g/kg diet). This aimed at enhancing faecal bile acid loss. Fish were fed both restrictively (1.2% BW/day) and to satiation. A similar fat ADC was found when substituting fishmeal with a plant protein mixture, suggesting that the lower content of bile acids, cholesterol, taurine, methionine and cysteine in the plant‐based diets did not limit fat digestion. Faecal bile acid loss increased alongside dietary NSP level, however, only during satiation feeding and most strongly for fish fed the fishmeal‐based diets. Enhanced faecal bile acid loss was not caused by NSP‐bile acid binding/entrapment, but by an increase in faeces production. During satiation feeding, fat ADC negatively correlated with faecal bile acid loss. From this it is concluded that bile acid availability/synthesis can become limiting for fat digestion in rainbow trout under conditions that enhance faecal bile acid loss (i.e. dietary NSP level and feeding level).  相似文献

Ovulation was stimulated in carp females of Polish and Hungarian provenance using injections of ( d ‐Ala6) GnRH ProNHEt (Kobarelin) (20 μg kg?1) with metoclopramide (10 mg kg?1) or carp pituitary (3 mg kg?1). For estimating the effects of the substance stimulating ovulation and of the female provenance, analysis of variance was carried out using the least squares method. It was found that the stimulating substance, provenance and interaction of these factors did not significantly affect the weight of eggs obtained. On the other hand, the stimulating substance, provenance and interaction had a highly significant (P ⩽ 0.0001) effect on the percentages of fertilization and live embryos. Females of Hungarian provenance treated with Kobarelin yielded eggs of much better quality but a smaller weight than after the pituitary treatment. In the case of carp females of Polish provenance, a yield of eggs of smaller weight and very poor quality was noted compared with the weight and quality of eggs obtained after hypophysation.  相似文献

We wanted to optimize the procedure for the selenium enrichment of farmed African catfish, using garlic as dietary selenium source. In the first experiment we established the relation between the length of the selenium enrichment period and the resulting total selenium level in the fillet of the fish. It was found that at a dietary level of 11.7 mg kg?1 Se, a total selenium level in the fillet of 0.7 mg kg?1 was reached in a relatively short enrichment period of 10 days before harvest. In the second experiment we studied the effect of depuration on the selenium level in the fillet and the sensory properties of selenium‐enriched African catfish. It was found that total selenium levels in the fillet were not affected during a 7‐day depuration period, while garlic odours and flavours in the raw and cooked fillets were significantly reduced after 2 days of depuration. We concluded that selenium enrichment of farmed African catfish can be obtained by selenium‐enriched finishing diets, while garlic odours and flavours resulting from dietary garlic can be effectively reduced in the fillet during a short depuration period without negatively affecting fillet levels of total selenium.  相似文献

This study investigated the effect of the replacement of fish oil (FO) with DHA‐Gold (DHA‐G)‐supplemented plant oils (PO) in rainbow trout fed plant‐protein‐based diets. Five diets (450 mg g?1 digestible protein and 150 mg g?1 crude lipid) were fed to rainbow trout (initial weight 37 ± 0.5 g) for 12 weeks in a 15 °C recirculating water system. The lipid inclusion types and levels were FO, PO and PO with DHA‐G supplemented at 30 mg g?1, 60 mg g?1 or 90 mg g?1 of the diet replacement for corn oil. Fish fed 90 mg g?1 DHA‐G were significantly larger and consumed more feed than fish‐fed PO or FO (218 g and 2.6% bwd?1 versus 181 g and 2.4% and 190 g and 2.3%, respectively). Feed conversion ratio was significantly increased in fish fed 90 mg g?1 DHA‐G (0.99) as compared to fish‐fed FO (0.90) and 30 mg g?1 DHA‐G (0.91). Panellists found trout fillets from fish fed the 90 mg g?1 DHA‐G diet to have significantly fishier aroma and flavour than fish fed the FO diet. Fatty acid analysis demonstrated that 60 mg g?1 or 90 mg g?1 DHA‐G supplementation increased PO fed fish fillet DHA to fatty acid levels equivalent or higher than those fish fed a FO diet.  相似文献
