Studies to determine baseline levels of microbial contaminants and foodborne bacterial pathogens are needed to evaluate the effectiveness of Hazard Analysis Critical Control Point (HACCP) programs, Good Manufacturing/Production Practices, and various interventions. In 2004 and 2005 poultry carcass rinses from provincially inspected abattoirs in Alberta, Canada, were tested to determine the levels of aerobic plate count bacteria, coliform bacteria, and generic Escherichia coli, the prevalence and levels of Campylobacter spp., and the prevalence of Salmonella spp. and Shiga toxin-producing E. coli (STEC). Samples were collected from 3 high volume and 62 low volume abbatoirs. All samples (1296) were positive for aerobic plate count bacteria, with 98.8% of samples having counts of 100 000 or less colony forming units (CFU)/cm2. Coliform bacteria were isolated from 99.7% of the 1296 carcasses and were recovered at levels of ≤ 1000 CFU/cm2 for 98.3% of the samples. Generic E. coli were recovered from 99.1% of the 1296 carcasses at levels of ≤ 1000 CFU/cm2 for 98.6% of the samples. Seventy five percent of 1234 samples that were tested for Campylobacter were positive; 37.5% of 1295 samples that were tested for Salmonella were positive; and only 2 of 1296 samples tested for STEC were positive (0.15%). 相似文献

Postmortem proteolysis in skeletal muscle and factors affecting this process were examined in pork, lamb and beef longissimus muscles (LM) to determine the cause of differences in meat tenderness among these species. Fat thickness differed among species in the following order: pork greater than beef greater than lamb. The following patterns were observed for rate of temperature and pH decline: lamb greater than pork greater than beef and pork greater than beef greater than lamb, respectively. At 1 d postmortem, pork was the most tender, followed by beef and lamb, respectively. Between 1 and 14 d of postmortem storage, lamb LM was the most improved in tenderness, followed by beef and pork, respectively. Species did not differ (P greater than .05) in LM collagen solubility. Pork LM from fed pigs had the highest (P less than .05) level of cathepsins B + L and cystatin(s) activities, whereas no differences (P greater than .05) were observed among the species for cathepsin B activity. The lowest (P less than .01) Ca2(+)-dependent protease (CDP)-II and CDP inhibitor activities were observed in pork LM. Beef LM had the highest CDP inhibitor activity (P less than .05) but was intermediate in CDP-II activity. No differences were observed among species for CDP-I activity. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of myofibrils isolated at 0, 1 and 14 d postmortem indicated that by d 1, desmin hydrolysis was most extensive in pork muscle, followed by lamb and beef.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Giardia infections in domestic cattle has come under increasing scrutiny owing to the potential contamination of surface and ground waters through manure distribution on fields and pasture runoff. The objective of the study was to determine the prevalence and genotypes of Giardia duodenalis in beef calves in major beef cow calf farms in Alberta, Canada. Fecal samples were collected from beef calves aged 2-10 weeks at nine farms in Alberta. Samples were examined for the presence of G. duodenalis cysts by immunofluorescent staining. Giardia cysts were found in 168 of the 495 fecal samples examined, with prevalence ranging from 7 to 60% among farms. Genotypic analysis of positive isolates utilizing PCR and sequencing of a 292 bp fragment of the 16S-rRNA locus, revealed the hoofed livestock genotype in 41 of the 42 isolates. One isolate was identical to the Assemblage A genotype. The results of this study demonstrate that beef calves in this area are primarily infected with the livestock genotype which is thought to be specific to artiodactyl hosts and non-infective to humans. Numbers of mesophilic bacteria were estimated on carcasses of 25 heifers and 25 steers of beef breeds in a modern, high-line-speed abattoir. One side of each carcass from each sex was sampled at the end of the kill-floor, before the carcass wash, on each of 25 visits. Two adjacent excision samples (5 x 5 x 0.5 cm) were taken from each of ten sites and processed for automatic enumeration of aerobic bacteria on hydrophobic grid membrane filters. The effects of sex and carcass weight on bacterial counts were examined. Groups of carcasses were examined to determine the sample size required for future assessments of kill-floor hygiene. The log10 of the most probable number of growth units (MPNGU)/cm2 did not differ significantly between heifers and steers (average over the ten sites of 2.2) and there was no effect of carcass weight on bacterial counts for nine of the ten sites. There were, however, highly significant (p < 0.001) differences in the counts between sites and the counts from the ten sites clustered into five homogenous groups. The between-carcass component of variation at a site was generally larger than the within-carcass component. We conclude that, to estimate the mean log10 MPNGU/cm2 at a site to within +/- 0.5 units, future group-carcass evaluations require about 200 samples from 10 (two adjacent samples/site) or 20 carcasses (one sample/site). 相似文献

The bacteriological quality of beef and pig carcasses was assessed at 9 Norwegian abattoirs by sampling 10 carcasses at multiple sites on each of several visits. On beef carcasses the following sites consistently carried higher numbers of bacteria: the Brisket, the Fore-ribs, the Flank groin, and the Round medial. There was no evidence that beef slaughter and dressing in the hanging position was superior to methods where the carcass was lying until the hide puller. On pork carcasses the Cheek, and the Abdomen lateral surface (belly) were most heavily contaminated. The hygienic quality of pork carcasses in abattoirs where singeing was a separate step tended to be better than where a combined singeing and dehairing machine was used.This survey suggests that bacteriological monitoring of slaughter at this level of sampling and visiting is able to detect consistently poor hygienic practices.Where direct comparisons with data from other countries could be made, the present investigation indicates that the bacterial counts on Norwegian beef and pork carcasses are of the same order or better.Key words: abattoir, slaughter, beef, pork, hygiene 相似文献

A survey to estimate the prevalence of antimicrobial resistance in Escherichia coli was conducted in 7 Canadian federally inspected processing plants during 2001. Escherichia coli isolates were recovered during routine hazard analysis critical control point sampling from beef carcasses and trim and subsequently tested for their antimicrobial susceptibility by using susceptibility panels. Of the 2653 isolates analyzed, 68% were sensitive to all 18 antimicrobials tested. For 14 of the 18 antimicrobials evaluated, the percentage of resistant isolates was < or = 1. Twenty-five percent of the isolates were resistant to tetracycline, 9% to sulfamethoxazole, 7% to streptomycin, and 3% to ampicillin. Multiple resistance was found in 12% of the isolates, with 7% showing resistance to 2 antimicrobials, 2% to 3 antimicrobials, 2% to 4 antimicrobials, and 1% to 5 or more antimicrobials. Forty-five different antimicrobial resistance patterns were observed. The reasons for the development of the antimicrobial resistance were not investigated in this study. This study was useful as a pilot to help to develop a national antimicrobial resistance surveillance program in Canada. This study indicates that laboratory standardization is possible for consistent results across the country and that the indicator organism, E. coli, is fairly easy to obtain for surveillance but Salmonella are not, due to their low prevalence in beef. 相似文献

This fecal prevalence study targeted cattle from 7 large (10 000 to > 40 000 head) commercial feedlots in Alberta as a means of establishing Campylobacter levels in cattle just prior to animals entering the food chain. Overall, 87% [95% confidence interval (CI) = 86–88] of 2776 fresh pen-floor fecal samples were culture positive for Campylobacter species, with prevalences ranging from 76% to 95% among the 7 feedlots. Campylobacter spp. prevalence was 88% (95% CI = 86–90) in the summer (n = 1376) and 86% (95% CI = 85–88) in the winter (n = 1400). In addition, 69% (95% CI = 66–71) of 1486 Campylobacter spp. positive samples were identified as Campylobacter jejuni using hippurate hydrolysis testing. Of those, 64% (95% CI = 58–70) of 277 and 70% (95% CI = 67–72) of 1209 Campylobacter isolates were identified as C. jejuni in winter and summer, respectively. After accounting for clustering within pen and feedlot, feedlot size and the number of days on feed were associated with Campylobacter spp. isolation rates. The high isolation rates of Campylobacter spp. and C. jejuni in feedlot cattle feces in this study suggest a potential role for feedlot cattle in the complex epidemiology of campylobacters in Alberta. 相似文献

This paper describes a large-scale investigation into the effects of licensed air emissions from sour-gas processing plants on the health and productivity of beef cow-calf herds in the province of Alberta, Canada. In conjunction with a geographical information system, two atmospheric-dispersion models were used to assess historical exposures at 5726 beef cow-calf farm-sites from 1987 to 1990. We did secondary analyses of health, productivity, and management data, from a government-extension survey previously administered to beef cow-calf producers across the province. Statistical models (adjusted for potential confounding and clustering within herd and over time) were used to determine associations with estimates of sour-gas emission exposure. All analyses were conducted at the herd-level. There were no significant (P>0.05) detrimental associations of exposure and the annual herd risk for culling, calf-crop delivered, calf-crop season profile, stillbirth and twinning, calfhood mortality, or calf-crop weaned. 相似文献

The study objectives were to determine the prevalence and serotypes of non-O157 Shiga toxin-producing Escherichia coli (STEC) in pens of feedlot cattle and on corresponding beef carcasses. We collected 25 fecal samples from 84 pens in 21 Alberta feedlots and 40 carcass swabs from each preslaughter pen for analysis by culture and polymerase chain reaction (PCR). Non-O157 STEC were recovered from feces from 12 (14%) of the 84 pens and 12 (57%) of the 21 feedlots by examination of 1 E. coli isolate positive for 4-methylumbelliferyl-beta-beta-glucuronide per sample. Twelve non-O157 serotypes were detected, but 7 of the 15 STEC isolates lacked the accessory virulence genes eae and hlyA. Although 115 (7%) of the carcass broths were PCR-positive, no STEC isolates were recovered from the 1650 carcasses sampled. Our data indicate that multiple non-O157 STEC serotypes may be present in cattle feces, yet are unlikely to be recovered from the corresponding beef carcasses when 20 colonies per sample from PCR-positive broth cultures are analyzed. 相似文献

The aim of this study was to compare the prevalence of Salmonella serotypes at two different sites on pig carcasses from herds classified as high-risk or low-risk and to elucidate the relationship between carcass contamination levels and serological status. Caecal samples and carcass surface swabs were cultured for Salmonella from a total of 210 pigs from low risk herds (< 19% of pigs in herd Salmonella seropositive) and 209 pigs from high risk herds (> 32% of pigs in herd Salmonella seropositive) in three abattoirs. Meat juice samples were collected for analysis by ELISA. The prevalence of Salmonella in the caecal contents of "low-risk" pigs was 10%, which was significantly lower than the 19% prevalence in "high-risk" pigs (p < 0.01). The corresponding figures for skin samples collected immediately post-evisceration were 2% and 12%. The predominant Salmonella serotype in the caecal contents of both the low-risk and high-risk pigs was Salmonella Typhimurium. Salmonella Kentucky and Salmonella Derby were the most frequent isolates from the carcass surface swabs of low- and high-risk pigs respectively. There was a positive association between seropositivity of pigs from high-risk herds and caecal carriage (p < 0.05). Results showed that herd categorisation based on serological results was useful in predicting Salmonella isolation rates from caecal samples and surface swabs of slaughtered pigs. 相似文献

The primary objective of this 7-month study was to determine the prevalence of porcine pathogens of the tonsil of the soft palate of swine at slaughter. Additional objectives were to determine if sampling the carcasses of normal or abnormal hogs provided different microbiological profiles and if the slaughter plant provides a feasible sampling frame and environment for detecting and monitoring important pathogens in tonsils that have health implications for both swine and humans. A total of 395 samples were collected from 264 farms. Of these, 180 tonsils were collected from normal carcasses and 215 tonsils were collected from carcasses that were diverted to the hold rail. Laboratory testing included bacteriological culture and identification as well as real time-polymerase chain reaction (PCR) testing for porcine reproductive and respiratory syndrome virus (PPRSV) and immunohistochemistry (IHC) for porcine circovirus-2 (PCV-2). The most commonly isolated bacteria included: Streptococcus suis (53.7%), Arcanobacterium pyogenes (29.9%), Pasteurella multocida (27.3%), and Streptococcus porcinus (19.5%). Virus screening revealed evidence of PRRSV and PCV-2 in 22.0% and 11.9% of the samples, respectively. Salmonella Typhimurium and Yersinia enterocolitica were isolated in 0.5% and 1.8% of the samples, respectively. Tonsils collected from the hold rail were more likely to be positive for Staphylococcus hyicus [odds ratio (OR) = 7.51, confidence interval (CI) = 2.89 to 19.54], Streptococcus porcinus (OR = 9.93, CI = 4.27 to 23.10), and Streptococcus suis (OR = 2.16, CI = 1.45 to 3.24). The objectives were to describe the pattern of losses through culling, sales of breeding stock, mortality, and disappearance, and to characterize the causes of mortality of cows and replacement heifers of breeding age from Western Canadian beef herds. Cows and replacement heifers from 203 herds were observed for a 1-year period starting June 1, 2001. Veterinarians examined dead animals on-farm using a standard postmortem protocol. The incidence of culling in cows and replacements heifers was 14.3 per 100 cow-years at risk, and the frequencies of sales for breeding stock, mortality, and cows reported missing per cow-years at risk were 4.0, 1.1, and 0.4, respectively. During the study, 355 animals died or were euthanized, 209 were examined postmortem, and the requested tissues were submitted for histopathologic examination from 184. A cause of death was determined for 70% (128/184) of the cows with complete gross postmortem and histopathologic examinations. Hardware disease (traumatic reticuloperitonitis), malignant neoplasia (cancer), calving-associated injury, rumen tympany (bloat), myopathy, and pneumonia accounted for 56% (72/128) of the animals where a cause of death was determined. Twenty-three other causes of death accounted for the remaining 44% (56/128). Factors relating to cow nutrition accounted for 25% of the deaths, emphasizing the importance of feeding management as a determinant of cow health in western Canada. 相似文献

Non-suppurative encephalitis occurs sporadically in beef cattle in western Canada, leading to loss of animals. This retrospective study investigated the presence of viral, bacterial, and protozoal antigens or DNA in 37 western Canadian feedlot cattle with non-suppurative encephalitis for which a cause had not been identified. Cases were selected based on the age of the animal (> 7 months), and clinical history of recumbency and depression. The identification of rabies in 1 case stresses the importance of including this viral disease in the list of differential diagnoses. Because there was variation in the severity, distribution, and type of lesions, it is possible that there may be more than 1 cause, but failure to identify an infectious agent might also suggest that non-infectious agents could play a role. 相似文献

The dispersion of air pollutants from all 231 licensed sour-gas processing plants in Alberta, Canada, was modeled on a monthly basis over a 10-year period (1985-1994). Exposure estimates for sulfur dioxide (SO(2)-used as a surrogate for exposure to combusted emissions) then were assigned to 1382 provincial dairy farms using a geographical-information system. Individual average and peak exposure for periods prior to each of 15 months of age and conception (four exposure-averaging periods for each of two dispersion models) were estimated for 163,988 primiparous female dairy-cattle between 1986 and 1994. Monthly or annual average farm-site exposure estimates likewise were assigned to associated herd-level data sets for the biologically relevant period of interest for each of three additional reproductive outcomes: monthly herd-average calving interval, stillbirth risk, and twinning risk. In one of the main-effects models, the maximum (i.e., peak) monthly sour-gas exposure experienced by individual-animals from birth to conception was associated with an increased time to first-calving in the very-highest exposure category (hazard ratio=0.86, 95% CI=0.80, 0.92). This equates to a decreased hazard (lambda) of calving (in each month subsequent to 22 months of age) for the highest-exposure animals (lambda=0.170) versus the zero-exposure animals (lambda=0.198) in a model with referent values for agro-ecological region and season of birth. The dose-response was not consistent across the full range of exposure categories. There was significant (P=0.003) interaction of emissions with agro-ecological region. After accounting for the interaction, a more-consistent dose-response was evident for some (but not all) agro-ecological regions. This suggests that any effect of emissions on dairy-heifer reproduction is subject to modification by features of soil type, vegetative cover, and/or climate.The increase in monthly herd-average calving interval on farms exposed to the very-highest levels of emissions appeared quite small and of limited practical importance within the range of expected exposures. Determination of percentage of protein in a pork carcass is a very costly and lengthy process. Therefore, we attempted to identify an easy and economical means to predict protein content of pork carcasses. Forty-seven market-weight pigs were slaughtered. The right side of each carcass was skinned, deboned and ground for composition analysis. Carcass data were collected on the left side and specific gravity analysis was performed on the side, ham and 8-9-10 rib loin section of each carcass. The hams and loin sections were dissected into skin, bone, subcutaneous fat, seam fat and individual muscles. After these components were weighted, they were analyzed for chemical composition. Percentage of chemical protein of each animal was determined and correlated with each of the weights and measurements taken on the carcass, ham and loin sections. Stepwise regression was used to predict decimal fraction of protein using either the carcass data, specific gravity ham measurements, loin section measurements or various combinations of these. In prediction equation, specific gravity of the carcass accounted for 73.4% of the variation found in percentage of protein. Prediction equations using indicators from the ham and loin section resulted in equations with R-square values as high as .783. These results suggest that specific gravity or indicators from the carcass, ham or loin section can be employed to predict percentage of protein in pork carcasses. 相似文献

Forty-seven market-weight pigs were slaughtered in order to determine percentage of chemical fat and in an attempt to determine an easily obtainable and inexpensive method to predict this value. The hams and 8-9-10 rib loin sections were removed from the left side of each carcass and dissected into subcutaneous and seam fat, individual muscles, skin and bone. Weights and chemical analysis were determined for each component. Numerous weights, measurements and specific gravity were determined on the carcass, ham and loin section of each pig. Percentage of chemical fat of each ham, loin section and carcasses was determined and correlated with the various weights and measurements taken. Stepwise regression was used to develop prediction equations using carcass data, specific gravity, ham or loin measurements or various combinations of these as dependent variables. The single best indicator of the decimal fraction of chemical fat in the pork carcass was determined to be specific gravity of the carcass half, the prediction equation using this variable had an R-square of .64. By adding 10th rib fat thickness to this equation, the R-square increased to .72. The best equation using carcass variables included 10th rib fat and marbling (R-square = .67). The loin section proved to be an accurate indicator of composition; ham measures were not as accurate as specific gravity and carcass measurements for predicting percentage of carcass fat. This research suggests that the percentage of chemical fat in the pork carcass can be predicted by an easy and inexpensive means. 相似文献

Carcasses from five trim cows and five choice steers were used to study the effects of spray-chilling on cooler shrink, chill rate, purge loss from vacuum-packaged cuts, cook loss, shear values and bacterial growth. Spray-chilling reduced cooler shrink but had no effect on chill rate, purge loss from vacuum-packaged cuts, cook loss or shear values. Aerobes, facultative anaerobes, aerobic psychrotrophs, facultative anaerobic psychrotrophs and lactic acid bacteria all tended to be higher on rounds from spray-chilled sides. Leaner (and lighter) cow carcasses chilled faster and had lost a higher percentage of their weight at 24 h than fatter and heavier steer carcasses. The leaner carcasses had higher bacterial counts initially and throughout storage. This difference may have been due to differences in the level of initial contamination during dressing and not due to the carcasses' leanness. Purge-weight loss for each carcass increased and cooking weight loss decreased with increased storage times, making the total weight loss from meat aged 5 vs 10 wk similar. 相似文献
