为探讨植物源杀虫剂印楝素对马铃薯甲虫的防治作用,采用叶碟法测定印楝素乳油对其成虫的拒食作用,并通过药效试验测定印楝素乳油对马铃薯甲虫的控制效果。结果显示,印楝素对马铃薯甲虫成虫有较强的非选择性拒食作用,且拒食率随取食时间的增加而升高。在取食36 h后,浓度为6、7.5、10、15和30 mg/L印楝素的拒食率分别为55.5%、59.1%、53.4%、76.1%和63.3%,拒食率与浓度无显著相关性;但印楝素对马铃薯甲虫的选择性拒食率较低,各浓度处理间均无显著差异,取食36 h时,选择拒食率约20%;在田间10 mg/L印楝素溶液施药1 d和3 d后,虫口减退率分别为26.52%和50.76%,显著高于对照组,施药9 d后,马铃薯甲虫的校正防治效果为48.86%。表明印楝素对马铃薯甲虫有一定的控制效果。  相似文献

Green lacewings, includingChrysoperla rufilabris (Burmeister) andCeraeochrysa cubana (Hagen), are predators of small, soft-bodied insects including whiteflies. The silverleaf whitefly,Bemisia argentifolii Bellows & Perring [formerlyB. tabaci (Gennadius) strain B], is an important pest of agronomic, vegetable and ornamental crops. Practical use of these lacewings as biological control agents would be facilitated by better understanding of their responses to both biorational (selective) and broad-spectrum insecticides. The topical and residual toxicity of azadirachtin (Azatin-ECTM), insecticidal soap (M-PedeTM), paraffinic oil (Sunspray Ultra-Fine Spray OilTM) and the pyrethroid bifenthrin (BrigadeTM) to eggs, larvae and adults of the lacewings were studied in the laboratory. Larvae ofC. cubana were much more tolerant to residues of bifenthrin than wasC. rufilabris and were somewhat more tolerant to topically applied soap. At normal field concentrations, azadirachtin (0.005%, by wt a.i.), paraffinic oil (1.0% by volume) and soap (1.0% by volume) were not toxic to larvae or adults of either species either topically or residually. Oil was toxic topically to eggs but azadirachtin and soap were not. Bifenthrin was toxic topically and residually to larvae and adults but was not so toxic to eggs as was oil. Thus, selectivity of all materials tested was relative to lacewing species and lifestage. The relative tolerance to insecticide residues exhibited byC. cubana larvae may be related to its trash-carrying habit, suggesting that use of trash-carrying chrysopids in place of non-trash carriers for augmentative biological control would increase options for non-disruptive chemical intervention when necessary.  相似文献

为筛选高效、低毒的杀虫剂,测定1.5%除虫菊素、0.3%印楝素、4%阿维·啶虫脒、2.5%高效氯氟氰菊酯和70%吡虫啉5种杀虫剂对苜蓿种子田主要害虫牛角花齿蓟马Odontothrips loti和豌豆蚜Acyrthosiphon pisum的室内毒力和田间防效,并分析这5种药剂对主要传粉昆虫数量及多样性指数的影响。结果显示,0.3%印楝素对牛角花齿蓟马和豌豆蚜的毒力相对较高,LC50分别为1.093 mg/L和2.864 mg/L。70%吡虫啉以及4%阿维·啶虫脒对牛角花齿蓟马和豌豆蚜均有较高的防效,最高防效均达到85.00%以上,但这2种药剂对传粉昆虫威胁较大,而0.3%印楝素则对传粉昆虫威胁较小。推荐花期苜蓿种子田使用0.3%印楝素1 000倍稀释液防控害虫,而非开花期可交替使用70%吡虫啉1 000倍稀释液和4%阿维·啶虫脒1 000倍稀释液防控害虫。  相似文献

为有效防控黏虫Mythimna separata,选用灭多威、苦参·印楝素、苏云金芽胞杆菌Bacillus thuringiensis、甲维盐和高效氯氟氰菊酯5种药剂对其卵进行室内触杀试验,比较这5种药剂对黏虫卵和初孵幼虫的触杀效果及对卵的毒力,测定不同药剂处理后黏虫卵初孵幼虫存活率、发育历期、化蛹率、蛹重和羽化率;并通过冷冻切片技术观察不同药剂处理后黏虫卵的外部形态和胚胎发育情况。室内触杀试验结果表明,苦参·印楝素浓度大于0.1 mg/L时,对产后不同时间黏虫卵的校正死亡率介于42.89%~97.34%之间,苏云金芽胞杆菌浓度为0.01 mg/L时对产后72 h黏虫卵的校正死亡率已达71.99%;各浓度苦参·印楝素和苏云金芽胞杆菌处理产后24、48和72 h黏虫卵后,其初孵幼虫校正死亡率介于39.02%~83.59%之间。苦参·印楝素对产后24、48 h黏虫卵的毒力最大,致死中浓度LC_(50)分别为0.018、0.109 mg/L;苏云金芽胞杆菌对产后72 h黏虫卵的毒力最大,LC_(50)为0.004 mg/L。黏虫卵经5种药剂处理后,幼虫化蛹率、蛹重和羽化率分别介于91.07%~93.64%、0.272~0.284 g和86.60%~87.78%之间,与对照差异不显著,幼虫存活率较对照显著下降43.02%~60.27%,幼虫发育历期较对照延长0.23~0.88 d,其中灭多威、苦参·印楝素、苏云金芽胞杆菌处理的幼虫发育历期较对照显著延长。苦参·印楝素、苏云金芽胞杆菌使胚胎无法形成正常胚带,同时会使已发育的虫体出现蜷缩、腹部皱缩;高效氯氟氰菊酯会延缓胚胎发育时间,使已形成的虫体畸形或使其肠道断裂;甲维盐影响虫体对营养物质的吸收,造成虫体萎缩,使其不能正常孵化。在黏虫卵防治中推荐使用苏云金芽胞杆菌和苦参·印楝素。  相似文献

Biological effects of a commercial formulation of azadirachtin, AlignTM, were assessed onStreblote panda Hübner [1820] (Lasiocampidae) first instars by means of choice and nochoice bioassays. Survival, development, and food consumption were recorded. The product tested did not show clear antifeedant effects, but produced dose-dependent larval mortality. All treatments resulted in significant inhibition of growth, presumably through endocrine regulatory effects. At higher doses assayed the product appears to have direct toxic effects. 采用高效液相色谱技术研究了苦皮藤素Ⅴ在粘虫Mythimna separata和小地老虎Agrotis ypsilon幼虫中的穿透及代谢。结果表明:苦皮藤素V均不能从粘虫和小地老虎幼虫的体壁穿透到血腔或从血腔穿透到中肠,但很容易从中肠穿透到血腔,且穿透速率无差异;苦皮藤素V在小地老虎幼虫体内的代谢解毒速率远大于其在粘虫幼虫体内的代谢速率,其半衰期分别为5.5和13.1 h。本研究结果表明,苦皮藤素V对粘虫和小地老虎幼虫的选择毒杀作用与药物的穿透能力无关,其在试虫体内的解毒代谢差异才是其对昆虫具有选择作用的机理之一。  相似文献

Investigations of the antifeedant mode of action of azadirachtin and four synthetic analogues, 22,23-dihydroazadirachtin, 3-tigloylazadirachtol, 11-methoxydihydroazadirachtin and 22,23-bromoethoxydihydroazadirachtin have revealed that both polyphagous and oligophagous insects are behaviourally responsive to azadirachtin, with the most responsive species being able to differentiate extremely small changes in the parent molecule. In Lepidoptera the antifeedant response is correlated also with increased neural activity of the chemoreceptors. When locusts are treated on crop plants, the antifeedant and physiological actions of azadirachtin and analogues work in concert and result in feeding deterrence, growth and moulting aberrations and mortality with the same order of potency as for antifeedancy. Specific binding studies using [3H]dihydroazadirachtin carried out on locust testes and Spodoptera Sf9 cells have shown that the competitive binding of the different analogues of azadirachtin to these binding sites occurs in a similar order of potency to that found with antifeedant and IGR bioassays. This suggests a causal link between specific binding to membrane proteins and the ability of the molecule to exert biological effects. © 1998 Society of Chemical Industry  相似文献

为探究舞毒蛾控制和分泌滞育激素的器官,于室内对舞毒蛾幼虫的中枢神经系统进行显微解剖,对其中枢神经系统和咽下神经节进行观察,并测量舞毒蛾1~5龄幼虫咽下神经节长度、宽度和面积,运用共聚焦激光扫描显微镜对脑和咽下神经节玻片逐层扫描,获取舞毒蛾幼虫脑和咽下神经节各高度的图像,将扫描后的图片描绘成1组多边图形,将其导入软件中,软件将多边图形叠加后构建其三维模型。结果显示,舞毒蛾幼虫的中枢神经系统包括脑、咽下神经节、3个胸部神经节和7个腹部神经节;舞毒蛾不同龄期幼虫的咽下神经节在外观形态上无较大差异;随着龄期增长,舞毒蛾咽下神经节的长度、宽度以及面积也不断增加,其中4龄幼虫咽下神经节的长度、宽度以及面积的增长速度最快,而5龄幼虫咽下神经节的长度、宽度以及面积的增长速度略微下降,1~3龄幼虫咽下神经节的长度、宽度以及面积的增长速度大致相同,均小于4龄和5龄幼虫。通过测量外部形态结构重建了舞毒蛾脑和咽下神经节的三维模型。  相似文献

The structure of the columnar cells, goblet cells and peritrophic membrane was studied in the midgut of gypsy moth (Lymantria dispar L.) caterpillars fed with a suitable (Quercus cerris L.) or an unsuitable (Robinia pseudoacacia L.) host. Columnar cells in the midgut of caterpillars fed onR. pseudoacacia leaves were elongated with small nuclei, partial loss of microvilli and vacuolated cytoplasm. The number and height of goblet cells and their nuclei were greater in caterpillars fed onQ. cerris leaves in comparison with larvae fed onR. pseudoacacia leaves. The effect of low concentrations of azadirachtin applied systemically through root tissues of rape plants (Brassica napus subsp.napus) on the mortality, period of development, longevity and fecundity of the cabbage aphid (Brevicoryne brassicae L.) (Sternorrhyncha: Aphididae) was studied. The concentrations of azadirachtin A tested were: 0.25, 0.05, 0.005, 0.0005 and 0.00005 mg ml−1. The mortality of cabbage aphid nymphs, particularly during the period of ecdysis, increased significantly with increasing concentrations. Azadirachtin had no effect on the length of the development period of the respective immature stages of cabbage aphids. The average longevity of the cabbage aphids decreased with increasing azadirachtin concentration and time spent feeding. 为合理预测意大利蝗Calliptamus italicus的种群动态,分别用1%(对照)、4%和7%氮含量的食物饲喂意大利蝗5龄蝗蝻,测定不同氮含量食物处理后意大利蝗5龄蝗蝻的生长速率,羽化后7 d的体长、股节长、胫节长、翅长、翅宽、体重及羽化后1、7、13和19 d的卵巢面积。结果显示,4%氮含量食物处理试虫的生长速率最大,分别较1%和7%氮含量处理的增加了88.02%和109.33%;4%和7%氮含量食物处理后,意大利蝗成虫的后翅宽和体重均显著高于对照;羽化后7 d,不同氮含量食物处理的意大利蝗卵巢面积出现显著差异,羽化后19 d,4%氮含量食物处理的卵巢面积显著高于其他2个处理。表明中等氮含量(4%氮含量)适宜意大利蝗生长。  相似文献

利用昆虫细胞-杆状病毒表达系统(Bac-to-Bac baculovirus expression system)表达获得小菜蛾 Plutella xylostella(L.)中肠氨肽酶N2(aminopeptidase N,APN2)蛋白,成功建立了该蛋白在昆虫细胞中的表达体系。将对Cry1Ac毒素敏感和已产生抗性的小菜蛾中肠APN2基因插入表达载体pFAST Bac HTB中,并在草地夜蛾 Spodoptera frugiperda 细胞系(Sf9)中进行了重组表达。聚丙烯酰胺凝胶电泳(SDS-PAGE)及蛋白免疫印迹技术(Western-blotting)结果显示,在107 kDa处有1条特异性蛋白条带。研究表明,小菜蛾中肠APN2基因可在Sf9细胞中成功表达,这为继续研究其功能及抗性的关系奠定了基础。  相似文献

稻螟赤眼蜂Trichogramma japonicum是稻田中鳞翅目害虫重要的卵期寄生型天敌,为了规范稻螟赤眼蜂放蜂期农药的安全使用,利用卵卡浸渍法处理蛹期稻螟赤眼蜂,通过稻螟赤眼蜂室内飞行装置和单雌寄生法评估了蛹期5种非鳞翅目害虫靶标农药井冈霉素、三唑酮、吡蚜酮、印楝素、毒死蜱对稻螟赤眼蜂成蜂搜寻和寄生能力的影响。搜寻能力结果显示,在毒死蜱处理后稻螟赤眼蜂失去搜寻能力,三唑酮、吡蚜酮和印楝素处理对稻螟赤眼蜂的搜索行为无影响,但井冈霉素处理下稻螟赤眼蜂的飞行蜂比例为48.1%,显著低于对照处理的74.7%。寄生能力结果显示,毒死蜱处理对稻螟赤眼蜂高毒,能使稻螟赤眼蜂失去寄生能力;印楝素处理对稻螟赤眼蜂的寄生力和子代羽化率无显著影响;井冈霉素、三唑酮和吡蚜酮处理下稻螟赤眼蜂的寄生力分别为22.2、25.3和24.7,显著低于对照的27.4、31.1和29.6,但对子代羽化率无显著作用。表明印楝素对稻螟赤眼蜂安全,可以推广使用;井冈霉素、三唑酮和吡蚜酮对稻螟赤眼蜂的搜索能力(仅井冈霉素)和寄生能力有显著的削弱作用,应谨慎使用;毒死蜱对稻螟赤眼蜂高毒,在稻螟赤眼蜂放蜂期应该禁用。  相似文献

为获得对大黑鳃金龟甲幼虫具有较高杀虫活性的菌株,利用拌土法测定了本实验室分离的500株苏云金芽胞杆菌(Bacillus thuringiensis,Bt)对大黑鳃金龟甲的杀虫活性,建立Bt菌株比较鉴别技术来进行多样性分析,并对菌株的晶体形态及幼虫感染Bt后的中肠组织切片进行观察。结果显示,从500株Bt菌株中筛选到42株对大黑鳃金龟甲幼虫具有不同程度活性的菌株,分属于14个菌株类型;其中有10株校正死亡率大于60%,261-1菌株杀虫活性最高,7 d校正死亡率达100%。从14个菌株类型中各选取1个代表菌株进行基因鉴定,仅P65-1、1126-1、FCD114和78-2菌株分别含有cry8Ma、cry8Ca、cry8Ab、cry8Ga、cry8Ea共5个cry8类新基因,其它10个菌株均不含cry3、cry8、cry18、cry23、cry37、cry43等对鞘翅目害虫有效的杀虫基因。14个菌株中,78-3、127-2和1198-1菌株能产生双锥体型晶体,261-1、FCD114、P65-1、FTL84、78-2和1126-1菌株能产生球形晶体。幼虫感染261-1、1198-1、FCD114、1126-1、QDL40-2菌株后,中肠组织发生明显的病理变化:2 d时肠壁细胞明显出现空洞化,排列疏松,4 d时受到严重破坏并脱离底膜。表明筛选到的Bt菌株具有防治大黑鳃金龟甲幼虫的潜力。  相似文献

We studied the presence of a potential transmission barrier that blocks Tomato yellow leaf curl virus in the nonvector greenhouse whitefly, Trialeurodes vaporariorum. Because T. vaporariorum can ingest and retain the virus after acquisition feeding on an infected plant, comparable to the vector whitefly Bemisia tabaci, circumstance evidence suggested that a transmission barrier presents at location(s) where the virus moves from the digestive tract lumen to the hemolymph. To provide direct evidence for the site of a transmission barrier in the nonvector insect, we compared the accumulation levels and localization of the virus between the two species of whiteflies. Quantitative real-time and conventional PCR analysis showed that accumulation of the virus during acquisition feeding and retention after a short acquisition period were indistinguishable between the two species, but the circulation of the virus within the whiteflies differed significantly between the species. In an immunofluorescence analysis using an antibody specific to the coat protein of the virus, the virus was restricted to the luminal surface of the midgut epithelial cells and did not enter their cytoplasm or that of the salivary glands in T. vaporariorum. In contrast, the virus was localized within the cytoplasm of the midgut epithelial cells and in the paired salivary glands of B. tabaci adults. This direct evidence shows that a selective transmission barrier at the luminal membrane surface of midgut epithelial cells in the nonvector whitefly blocks entrance of the virus into the midgut epithelial cells, resulting in incompetence as a vector of the virus.  相似文献

Methanolic extracts of seeds and fruits of the chinaberry tree,Melia azedarach L. (Meliaceae), showed strong antifeedant activity against 2nd instar larvae ofSesamia nonagrioides Lefèbvre (Lepidoptera: Noctuidae), a very serious pest of maize(Zea mays L.) in Mediterranean countries. Extracts were applied in an artificial diet at concentrations of 1000 and 2000 ppm. The parameters used to evaluate the activity were larval growth rates; quantity of food ingested; phagodepression/phagostimulation index; quantity of frass produced; quantity of material ingested; duration of larval development; and cumulative mortality. Seed extract showed high bioactivity at both doses, while fruit extract proved to be less active, and only at the higher dose used (2000 ppm) did it display a slight antifeedant activity. The activity of theM. azedarach seed extract at the higher dose (2000 ppm) was comparable to that of pure azadirachtin applied at a dose of 1.25 ppm, or to ‘Mubel’, a commercial extract ofAzadirachta indica A. Juss. (Meliaceae), applied at a dose of 75 ppm.  相似文献

为探明丁布胁迫对亚洲玉米螟氧化还原系统的影响,采用酶活力测定方法和实时荧光定量PCR技术,研究了取食丁布含量差异显著的3个玉米品种登海662、浚单20和隆玉602叶片及模拟其叶片丁布含量的人工饲料后,玉米螟3龄幼虫氧化还原系统主要酶活性和谷氧还蛋白(Grx)mRNA转录表达的变化。结果显示:取食3个玉米品种叶片24 h后,亚洲玉米螟幼虫过氧化物酶和超氧化物歧化酶活性均显著高于对照,且在3个品种处理间差异显著,由大到小依次为登海662(118.30 U/mg和182.19 U/mg)、浚单20(107.13 U/mg和90.02 U/mg)和隆玉602(77.54 U/mg和93.59 U/mg)。取食浚单20可显著促进幼虫脂肪体Grx的表达,取食登海662可显著促进其中肠Grx的表达;取食模拟登海662叶片丁布含量的人工饲料12 h后,幼虫脂肪体中Grx表达量高于对应玉米叶片饲喂处理,而中肠部位则低于对应玉米叶片饲喂处理;取食模拟浚单20的结果与模拟登海662相反。表明亚洲玉米螟氧化还原系统在防御丁布胁迫过程中发挥着重要作用。  相似文献

为更好地了解苏云金芽胞杆菌Bacillus thuringiensis毒素蛋白对二点委夜蛾Athetis lepigone的毒力以及作用机理,通过饲喂含有Cry1Ac、Cry1Ab、Cry2Ab和Vip3Aa四种不同Bt毒素蛋白饲料,测定Bt毒素蛋白对二点委夜蛾幼虫的毒力,并观察取食4种毒素蛋白后幼虫中肠组织的病理学变化。结果显示,二点委夜蛾幼虫取食毒素蛋白后72 h,Cry1Ab和Cry1Ac毒素蛋白对二点委夜蛾幼虫的杀虫活性较高,校正死亡率为84.7%和76.4%;Vip3Aa和Cry2Ab毒素蛋白的毒力较弱。二点委夜蛾幼虫取食4种Bt毒素蛋白后,中肠柱状细胞微绒毛脱落,杂乱地分散在肠腔内,杯状细胞变形和腔内微绒毛脱落,线粒体和内质网等变形破裂,细胞核的核膜消失、核质凝聚和形状发生变化,经Cry1Ab和Cry1Ac毒素蛋白处理后中肠细胞的病变症状和速度明显高于Cry2Ab和Vip3Aa毒素蛋白处理。表明Cry1Ab和Cry1Ac毒素蛋白对二点委夜蛾幼虫杀虫活性较高,显著高于Cry2Ab和Vip3Aa毒素蛋白,且对其中肠细胞的破坏作用也较强。  相似文献

为明确棉铃虫Helicoverpa armigera ATP结合盒转运蛋白家族A蛋白亚家族2(ATP binding cassette transporter subfamily A member 2,ABCA2)在Cry2Ab杀虫机制中的作用,利用酵母双杂交技术研究棉铃虫ABCA2与Cry2Ab的结合特性,并利用RNA干涉(dsRNA和siRNA)降低ABCA2在细胞和幼虫中的表达,结合细胞毒理学试验分析ABCA2的功能。结果表明,棉铃虫ABCA2能与Cry2Ab特异性结合;棉铃虫的ABCA2序列与美洲棉铃虫H.zea相关序列的相似度为92.86%,利用dsRNA干涉美洲棉铃虫中肠细胞ABCA2的表达能显著降低Cry2Ab对中肠细胞的毒力;利用siRNA干涉技术降低ABCA2在棉铃虫幼虫活体中的表达也显著降低了Cry2Ab对幼虫的毒力。表明棉铃虫ABCA2不仅是Cry2Ab的特异性结合蛋白,而且参与Cry2Ab的杀虫过程,是Cry2Ab的一个重要功能受体。  相似文献

为评估转Cry1Ac/1Ab基因棉花对棉铃虫Helicoverpa armigera的抗性及对非靶标害虫甜菜夜蛾Spodoptera exigua和斜纹夜蛾S.litura生长发育的影响,采用室内生测法测定其对棉铃虫的抗性及对甜菜夜蛾和斜纹夜蛾不同龄期幼虫存活率、营养代谢及中肠酶活性的影响。结果表明,转Cry1Ac/1Ab基因棉花对棉铃虫第2代幼虫的抗性程度最高,幼虫校正死亡率达91.33%,但对甜菜夜蛾和斜纹夜蛾的抗性程度较低,幼虫校正死亡率分别为15.33%和13.33%。甜菜夜蛾和斜纹夜蛾各龄期幼虫取食转Cry1Ac/1Ab基因棉花后,其存活率与取食常规棉花对照无显著差异;甜菜夜蛾对转Cry1Ac/1Ab基因棉花叶片的相对取食量、近似消化率分别为16.68和93.12%,均高于取食常规棉花对照的10.72和92.00%,但差异不显著,而斜纹夜蛾取食转Cry1Ac/1Ab基因棉花后的各项营养指标均低于取食常规棉花对照,差异也不显著。取食转Cry1Ac/1Ab基因棉花后,甜菜夜蛾的过氧化物酶活性为0.02 U/mg prot,显著低于取食常规棉花的0.05 U/mg prot;斜纹夜蛾的酸性磷酸酶活性为0.15 U/mg prot,高于取食常规棉花的0.10 U/mg prot,但差异不显著,其它中肠酶活性均低于对照,亦无显著差异。  相似文献
