It is surmised that domestication has reduced geneticvariability in the common bean (Phaseolusvulgaris L.). However, little is known about leaf anatomy responses to water stress in wild or domesticated forms ofP. vulgaris. In this study,anatomical variation was evaluated in watered and water stressed plants of wild and domesticated genotypes of P.vulgaris from Mexico, growing under greenhouse conditions. Five terminal leaflets of completely expanded leaves wereused to evaluate leaf area (LA) and theirmiddle-regions to evaluate leaf thickness (LT), and various anatomical features. Domesticated genotypes showed bigger LAthan that of the wild ones; there was an effect due to the origin of materials, LA being bigger in genotypes from warm and humid lands that those from dry and temperate ones. No differences were detected in LT among materials; however, in domesticated plants the palisadeparenchyma predominated over the spongy one, contrasting with thewild genotypes. Under drought conditions, stomatal abortion was observed in wild and domesticated genotypes, especially on theabaxial surface. We conclude that, in fact, there are contrasting differences in leaf anatomy among wild and domesticated forms of Mexican Phaseolus vulgaris. Results suggest that domestication not only favours the enlargement of the LA, but also an increase in palisade parenchyma, suggesting a divergence toward assimilation in domesticated forms, in contrast with a better internal gas diffusion in wild forms. Domestication seems to have accentuated stomatal abortion as means of a transpirational control mechanism to counteract the effects of drought.  相似文献

Common bean (Phaseolus vulgarisL.) is a traditional crop in Portugal, where farmers growvarieties selected and maintained by themselves. A collection of 88landraces of common bean was evaluated for 17 quantitative andqualitative traits and the biochemical marker phaseolin to displaythe degree of variation of this germplasm. Agronomic data weresubjected to cluster analysis and several groups were identified,with three groups clustering most of the landraces. Regardingphaseolin variation the C and T banding patterns are the mostfrequent ones, so the origin of the Portuguese beans is thus probablythe Andean region of South America. These results give informationabout the origin, diversity and breeding value of the Portuguesegermplasm, that could be useful to widen the genetic base ofcurrently cultivated bean varieties in Europe.  相似文献

Summary Plants grown from seed with high (1.5–7.3 g Mo seed-1) and low (0.07–1.4 g Mo seed-1) Mo contents were grown in the presence and absence of Mo in growth media (perlite) or in a flowing-solution culture, in a controlled environment. Neither the high (1.5 g Mo seed-1) nor the low (0.1 g Mo seed-1) Mo content in seed from a small-seeded genotype (BAT 1297) was able to prevent Mo deficiency (reduced shoot, root and nodule dry weight, N2 fixation and seed production) in growth media without an external supply of Mo, whereas both the high (7.3 g Mo seed-1) and the low (0.07 g Mo seed-1) contents in seed were able to prevent Mo deficiency in a large-seeded genotype (Canadian Wonder). Responses to Mo treatment by the Two genotypes were inconsistent between the growth media and solution culture experiments. Seed with a large Mo content (3.5 g Mo seed-1) from the Canadian Wonder genotype was unable to prevent Mo deficiency (reduced shoot and nodule dry weight and N2-fixation) in a solution culture without an external source of Mo, whereas both the large (1.7 g Mo seed-1) and the small (0.13 g Mo seed-1) contents in seed prevented a deficiency in BAT 1297. Growing plants from seed with a small Mo content, without additional Mo, reduced the seed Mo content by 83–85% and seed production by up to 38% in both genotypes. Changes in seed size and increases in shoot, root and nodule dry weight occurred, but varied with the genotype and growth conditions. These effects were also observed in some cases where plants were grown with additional Mo, demonstrating that the amount of Mo in the seed sown can influence plant nutrition irrespective of the external Mo supply. Nodule dry weight, total N content of shoots and seed production were improved by using seed with a small Mo content (1.64–3.57 g Mo seed-1) on acid tropical soils in Northern Zambia. Plants of both the large- and small-seeded genotypes grown from seed with a small Mo content (<1.41 g Mo seed-1) had a smaller nodule weight, accumulated less N and produced less seed. The viability of seed with a small Mo content was lower (germination up to 50% less) than that of seed with a large Mo content.  相似文献

Ninety-five common bean (Phaseolus vulgaris L.) landraces from Spain were evaluated in three different environments in northern Spain for their agronomic performance and seed quality. Significant differences among landraces were found for 14 quantitative traits related to phenology, yield and its components, and seed quality traits. Environmental effects were significant for all traits evaluated except for seeds pod?1, seed width/thickness, seed weight, and seed water absorption. Landrace by environment interactions were significant for all traits except for seeds pod?1 and seed water absorption. Selection of new breeding lines for agronomic performance and seed quality within landraces should be reliable because many of them are mixture of lines. Some heirloom varieties belonging to the types faba, caparrón,riñón,ganxet and tolosana had the best performances regarding to seed quality and yield. Principal component analysis revealed differences among environments affecting the performance of the bean landraces. Variation in the landraces seems to be organized in a different way in each one of the test environments, therefore, low plasticity and specific adaptation of Spanish bean landraces to different environments is derived from this study. Some landraces, especially those large and white seeded should be an useful resource for sustainable farming systems in different biogeographical areas and a worthy germplasm for the genetic improvement of agronomic value and seed quality.  相似文献

The genetic diversity of 10 commercial cultivars of common beans, developed in Northern Argentina was analyzed based on RAPD markers. Sixteen primers were assayed and among them only 4 showed polymorphisms. A similarity matrix was generated by applying three different association coefficients, Simple Matching, Jaccard and Dice. By the UPGMA method dendrograms were generated and also the principal coordinate analysis was performed. The similarity values found were higher than 40% suggesting that genetic diversity is low. Both cluster analysis and principal coordinates analysis associated commercial cultivars either to the Andean or the Mesoamerican gene pool.  相似文献

We examined the competitiveness of five effective Rhizobium leguminosarum biovar phaseoli strains in the nodulation of kidney beans (Phaseolus vulgaris L.), either alone or in pairwise combination, against the indigenous strains. The results showed that the introduced Rhizobium sp. strains (B2, B17, B36, T2, or CIAT 652) occupying 64–79% of the total nodules (as single inocula) were more competitive in nodulation than the native rhizobia. However, the competitiveness of the individual Rhizobium sp. strain either increased or decreased when used in a pairwise combination of double-strain inocula. For example, strain B17, although quite competitive against the indigenous population (68% nodule occupancy), became poorly competitive in the presence of strain B2 (reduced from 68 to 2.5%). A similar reduction in nodule occupancy by strain B17 was observed in the presence of B36 or CIAT 652, indicating that two competitive strains may not always be compatible. These results suggest that it is important to co-select competitive as well as compatible rhizobia for multistrain inoculant formulation.  相似文献

The phenotypic variation found in four common bean (Phaseolus vulgaris L.) complex primitive landraces, among a group of accessions collected in Northwestern Argentina in several missions is described, with particular attention to the wide diversity found in some small areas. It is presented a hypothesis about the maintenance of such diversity in bean mixtures or complex primitive landraces that grow close to their wild relative. Wide diversity regarding to seed type and plant characteristics was displayed by the landraces MCM-SV (composed of 11 lines), MCM-292 (14 lines), MCM-298 (5 lines) and VAV-3716 (14 lines). Food uses of dry seed and fresh pod seemed to be more relevant than the aesthetic use although all of them were presumably considered by humans for centuries resulting in the current phenotypes of these complex primitive landraces. Additionally, some weedy types (intermediate between wild and domesticated types) were detected in the landracesMCM-292 and MCM-298. The four complex landraces described consisted of highly diverse mixtures and they could play a role in breeding to enlarge the genetic basis of domesticated bean varieties belonging to the Andean gene pool.  相似文献

The diversity, genetic structure, and genetic flow of wild populations of Phaseolus vulgaris L. within its Mesoamerican area of domestication, were analyzed by means of morphological and inter-simple sequence repeat molecular markers. Overall, 89% of the loci studied were polymorphic, 35% in the least diverse population and 65% in the most diverse. Genetic diversity in the populations was high, between h = 0.14 and 0.29, as was the maximum distance between populations (D = 0.3). Between 40% and 45% of the diversity was explained by the differences among populations, indicating that a large number of populations is necessary to represent the wild gene pool in the germplasm collections. We found uniformity in allele frequencies among the populations, suggesting presence of outcrossing. We did not find correlation between genetic and geographic distances, but the dendrogram topology suggests geographical isolation due to the mountainous topography. Negative correlations were observed between the coefficient of variation of seed size and the distance between wild populations and fields . We obtained a highly negative correlation between percentage of polymorphic loci and distance to the nearest crop field, which also suggests gene flow from the domesticated populations. These observations suggest that genetic flow is taking place from domesticated toward wild populations and that the farmer, through his agricultural activities, could be influencing the magnitude and the characteristics of the gene flow, and along with this, the differentiation of wild populations. New approaches should be established for conservation in situ and maintaining bio-safety, given the risk of introducing genotypes from the Andes and transgenic varieties and causing genetic assimilation.  相似文献

Strains of Rhizobium tropici IIB, CIAT899 and F98.5, both showing good N2 fixation, and a R. etli strain W16.3SB were introduced into a field which had no history of bean culture. Plant dilution estimates showed that in the presence of its host (Phaseolus vulgaris cv. Carioca) during the cropping seasons and the subsequent fallow summer periods, the bean rhizobial populations increased from less than 30 to 103 g–1 dry soil after 1 year and to 104 g–1 dry soil after 2 years. In the 1st year crop, the inoculated strains occupied most of the nodules, which resulted in a higher nodulation and C2H2 reduction activity. Without reinoculation for the second and third crops, however, little R. tropici IIB was recovered from the nodules and the bean population consisted mainly of R. etli, R. leguminosarum bv. phaseoli, and R. tropici IIA. Reinoculation with our superior R. tropici IIB strains before the second crop resulted in R. tropici IIB occupying the main part of the nodules and a positive effect on nodulation and C2H2 reduction activity, but reintroduction of the inoculant strain in the third season did not have any effect.  相似文献

Summary In an experiment performed under greenhouse conditions, four cultivars of Phaseolus vulgaris L. (Venezuela-350; Aroana; Moruna; Carioca) were inoculated with three Rhizobium leguminosarum biovar phaseoli strains (C-05; C-40 = CIAT 255; C-89 = CIAT 55) and were fertilized with an N-free mineral nutrient solution. The plants were harvested 25, 40, and 55 days after emergence and the following paramenters were evaluated: Nitrogenase activity of nodulated roots, H2 evolution by the nodules; relative efficiency of nitrogenase; respiration rates of nodulated roots and detached nodules; dry weight and total N of stems, leaves, pods, roots, and nodules. Generally the bean cultivar, Rhizobium strain, had an effect and there was an interaction effect with both symbiotic partners, on all parameters. On average, nodules represented 23% of total root respiration but the best symbiotic combinations showed lower ratios of C respired to N fixed. The maximum N-assimilation rate (between 40 and 55 days after emergence) of 11.93 mg N plant–1 day–1 occurred with the symbiotic combination of Carioca × C-05, while the poorest rate of 0.55 mg N plant–1 day–1 was recorded with Venezuela-350 × C-89. The best symbiotic combinations always showed the highest relative nitrogenase efficiency, but the differences in N2-fixation rates cannot be explained solely in terms of conservation of energy by recycling of H2. Summary The common bean (Phaseolus vulgaris L.) is generally regarded as a poor N2 fixer. This study assessed the sources of N (fertilizer, soil, and fixed N), N partitioning and mobilization, and soil N balance under field conditions in an indeterminate-type climbing bean (P. vulgaris L. cv. Cipro) at the vegetative, early pod-filling, and physiological maturity stages, using the A-value approach. This involved the application of 10 and 100 kg N ha-1 of 15N-labelled ammonium sulphate to the climbing bean and a reference crop, maize (Zea mays L.). At the late pod-filling stage (75 days after planting) the climbing bean had accumulated 119 kg N ha-1, 84% being derived from fixation, 16% from soil, and only 0.2% from the 15N fertilizer. N2 fixation was generally high at all stages of plant growth, but the maximum fixation (74% of the total N2 fixed) occurred during the interval between early (55 days after planting) and late podfilling. The N2 fixed between 55 and 75 days after planting bas a major source (88%) of the N demand of the developing pod, and only about 11% was contributed from the soil. There was essentially no mobilization of N from the shoots or roots for pod development. The cultivation of common bean cultivars that maintain a high N2-fixing capacity especially during pod filling, satisfying almost all the N needs of the developing pod and thus requiring little or no mobilization of N from the shoots for pod development, may lead to a net positive soil N balance.  相似文献

The relationship between soil acidity and polyamine (putrescine, spermidine, spermine) concentrations in roots was evaluated and compared to (1) nodule numbers in common beans and (2) tannin concentrations in roots. Six Phaseolus vulgaris cultivars were grown in pots in a greenhouse in soil at pH 4.5 or 5.5 and inoculated with Rhizobium leguminosarum bv. phaseoli strains. At pH 4.5 nodule numbers were strongly reduced but polyamine concentrations were 50% higher than at pH 5.5. At both pH levels putrescine and spermidine were the dominant polyamines, while the concentration of spermine was less than half of the other two. There was also a cultivar effect on the polyamine concentration, but this was much less pronounced than the pH effect. By 22 days after planting the uninoculated control plants had about 13% lower levels of polyamines than the inoculated plants. The concentrations of putrescine, spermidine, and spermine were negatively correlated with nodule numbers, but positively correlated with the root tannin content. There were no significant correlations with either root or plant dry weight.  相似文献

A common bean genomic library was constructed using the ‘IAC-UNA’ variety enriched for (CT) and (GT) for microsatellite motifs. From 1,209 sequenced clones, 714 showed microsatellites distributed over 471 simple and 243 compound motifs. GA/CT and GT/CA were the most frequent motifs found among these sequences. A total of 123 microsatellites has been characterized. Out of these, 87 were polymorphic (73.7%), 33 monomorphic (26.8%), and 3 (2.4%) did not amplify at all. In a sample of 20 common bean materials selected from the Agronomic Institute Germplasm Bank, the number of alleles per locus varied 2–9, with an average of 2.82. The polymorphic information content (PIC) of each marker varied from 0.05 to 0.83, with a 0.45 average value. Cluster and principal coordinate analysis of the microsatellite data were consistent with the original assignment of the germplasm accessions into the Andean and Mesoamerican gene pools of common bean. Low polymorphism levels detected could be associated with the domestication process. These microsatellites could be a valuable resource for the bean community because of their use as new markers for genetic studies. Extensive germplasm resources are present in collections of Lotus corniculatus and L. uliginosus, two forage species that are valuable in situations where other legumes are not persistent. Naturalised populations in many regions are the primary gene pool for genetic improvement of these species and interspecific hybrids are an important source of variation for agronomic and forage quality traits. Although previous research has provided evidence that selection for optimal levels of condensed tannins and herbage productivity are feasible breeding objectives, less is known about correlations of condensed tannin with other parameters of nutritive value. A germplasm collection of 38 accessions in both species from 9 geographic regions was used as a data base to determine these correlations. Forage of each accession was sampled in two successive years from a field site in south-eastern Australia and analysed for condensed tannin, in vitro digestible dry matter, nitrogen, acid detergent fibre and neutral detergent fibre. In both species condensed tannin was negatively correlated with in vitro digestible dry matter and nitrogen. This result helped to identify potentially valuable accessions with low condensed tannin (<4% of dry weight) and high in vitro digestible dry matter (>70%). One of these was an interspecific hybrid of L. uliginosus and L. corniculatus, confirming the view that interspecific hybridization would be a valuable avenue for improvement of agronomic and forage quality characters in Lotus species.  相似文献

选用对低磷胁迫抗性各异的3个甜菜品种:品20、品17和品14,在人工培养室内采用沙培试验法,研究了甜菜抗耐低磷胁迫的生理机制。结果表明,低磷胁迫限制了甜菜对磷的吸收,导致植株含磷量和生物产量显著下降;不同品种间差异显著,品14降幅最大,品17次之,品20最小。与足磷处理(P 100μmol/L)相比,低磷胁迫后甜菜品种间体内抗性生理特征差异显著。其中,品14和品17的丙二醛含量、脯氨酸含量、过氧化物酶活性均显著增加;品20的丙二醛含量、脯氨酸含量极显著下降,而过氧化物酶活性变化不显著。叶片中Mg2+-ATPase活性降低,不同品种降幅各异,从小到大依次为品20品17品14,其中品20与后两者间的差异均达显著。品20和品17体内的钙调素(CaM)含量显著增加,而品14变化不明显,其相对值从大到小依次为品20品17品14,差异显著,与品种自身抗磷胁迫能力顺序一致。不同磷素营养条件对甜菜抵御外界不良环境有较大影响,叶片在受到热伤害时,抗磷胁迫能力较弱的品14和品17在低磷胁迫时质膜损伤率显著增加;而抗磷胁迫能力较强的品20叶片质膜的损伤率显著下降,抗热能力得到改善。  相似文献

Heat susceptibility is a cause of yield loss in common bean (Phaseolus vulgaris L.) in temperate and tropical lowland growing regions. Interspecific hybridization with related species that include tepary bean (Phaseolus acutifolius A. Gray) can potentially be used to transfer heat tolerance traits to common bean. This study evaluated 25 tepary bean plant introductions (PIs) following exposure to high-temperature (35 °C day/32 °C night) and control temperature (27 °C day/24 °C night) during reproductive development. These 25 PIs were selected for high yield potential in a 14 h photoperiod greenhouse environment. High temperature when compared to non-stress treatment resulted in a mean reduction in seed yield of 88.8% among tepary beans and caused 100% yield loss in common beans. Twelve P. acutifolius PIs exhibited negligible yield under heat-stress when compared to control treatment. Although yields of these accessions were low under very high-temperature conditions, PI 200902, PI 312637, PI 440785, PI 440788, and PI 440789 exhibited the highest yield component stability under the high temperature treatment.  相似文献

A collection formed from 53 accessions of white clover stolonswas carried out in naturalized populations from Argentina betweenparallels 26° and 37° South latitudes and 58° to 62°West longitude. Almost half of them (43%) wereobtained from grazed sites. Collected samples were grown in the fieldat Pergamino Experimental Station and compared with a local cultivar.Attributes scored or measured were petiole length, width and lengthof central leaflet, relation width/length, height, growing habit,leaf mark, stolon thickness and density, days to flowering,cyanogenesis, seed yield and winter herbage yield. There weresignificant correlations among seven pairs of characters. Allpopulations were cyanogenic within a range from 40 to 100%.Populations from grazed sites had prostrate habit and lower canopyheight than those from non grazed sites. The collection was clusteredin seven groups. Differences among groups were attributed toagroecological differences in sites of collection. Results obtainedencouraged in Argentina further collection missions for naturalizedpopulations of white clover.  相似文献

In order to evaluate the genetic diversity in Lathyrus genus, the Inter Simple Sequence Repeats method (ISSR) was exploited in five populations. These consisted of two cultivated species belonging to section Lathyrus (L. sativus L. and L. cicera L.) and a wild one belonging to the section Clymenum (L. ochrus DC.). Two 3′anchored ISSR primers and two unanchored ones, generated a total of 60 useful polymorphic DNA bands. Our data provide evidence of high molecular polymorphism at the intra- and the inter-specific levels showing that both wild and cultivated forms constitute an important pool of diversity. Moreover, among the generated DNA bands, a 500 bp band, totally absent in the banding patterns of the section Clymenum, appears to be a molecular marker of section Lathyrus. Results provided for lineage and suggest recent origin of these species that might have evolved from a common ancestor producing both L. ochrus species and the two other species L. sativus and L. cicera. These relationships support previous studies based on morphological variation and molecular analysis.  相似文献

The effect of dual inoculation on three local cultivars (Miss Kelly, Portland Red, Round Red) of red kidney beans (Phaseolus vulgaris, L.) with four strains of Rhizobium leguminosarum bv. phaseoli and three species of vesicular-arbuscular mycorrhizal (VAM) fungi was examined in a clay loam soil. Rhizobial strains B 17 and B 36, each paired with Glomus pallidum or G. aggregatum, were the most effective pairings for cv. Miss Kelly. Inoculation of Miss Kelly with any of these pairings significantly (P=0.05) increased growth, number of nodules, nodule dry weight, mycorrhizal colonization, and shoot N and P content than other pairings. The growth response by cv. Portland Red was significantly improved by pairings of B 36 or B 17 with any of the three VAM fungi. For both cultivars (Miss Kelly and Portland Red), CIAT 652 or T 2 paired with VAM fungi did not give a positive growth response. In contrast, for cv Round Red the T 2 rhizobial strain in combination with any of the three VAM fungi showed a significant (P=0.05) growth improvement in all parameters. Our results suggest that while dual inoculation of VAM fungi and rhizobia significantly improves the growth response by red kidney beans, the best pairings of VAM fungus and rhizobia for each cultivar need to be carefully selected.  相似文献
