Freeze-dried ileal effluent (1% wt/vol) from cannulated pigs fed rice-based diets with the inclusion of either animal protein (CON), animal protein plus potato starch (PS), animal protein plus sugar beet pulp (SBP), or animal protein plus wheat bran (WB) was incubated anaerobically at pH 6.0 in fermenters containing 5% (wt/vol) fecal slurry comprising mineral salts medium and 50 g/L of fresh feces from pigs fed the same diets as the cannulated pigs. Samples were collected from the fermenters at 0, 2, 4, 12, 24, and 48 h during in vitro fermentation for measuring nonstarch polysaccharides (NSP), starch, and short-chain fatty acids (SCFA). Results showed that the major SCFA produced were acetate, propionate, and butyrate. The inclusion of soluble dietary fiber (diet SBP) caused the highest concentrations of acetate, propionate, butyrate, and total SCFA, whereas the increase in the production of propionate resulting from the addition of insoluble dietary fiber (diet WB) only occurred at the initial stages during 48 h in vitro fermentation. At all sampling occasions (except for 4 h), the levels of butyrate were increased (P < 0.01) by resistant starch compared with fiber sources, showing that a higher level of butyrate can be achieved through microbial fermentation by potato starch. Lowered (P < 0.05) butyrate concentrations were observed with diet WB during in vitro fermentation. With the inclusion of fiber sources, the energy originating from SCFA was similar to that from NSP disappearance, whereas the values were lower (P < 0.05) from NSP disappearance than for SCFA generated without fiber sources supplemented. We conclude that more substrate is available in ileal effluent with the addition of soluble dietary fiber, and an increased level of butyrate could be achieved through microbial fermentation by resistant starch.  相似文献

The aim of this study was to evaluate the effect of different sources of variation in gas production technique on the in vitro gas production kinetics of feedstuffs. Triplicates of commercial concentrate, grass silage, grass hay and grass pasture were incubated in three experiments: experiment 1 assessed two agitation methods; experiment 2 evaluated different rumen inocula (pooled or different donor cows for each incubation run); and experiment 3 used Goering‐Van Soest or Mould buffers for media preparation. Gas production data were fitted into the Michaelis–Menten model and then subjected to analysis of variance. Gas production (GP) at 48 h and asymptote gas production (A) were lower when bottles were continuously under horizontal movement. Time to produce half and 75% of A, and A were affected by rumen inocula, while buffer type affected time to produce half and 25% of A and GP. No interactions between substrates and sources of variation were observed, suggesting that the effects of substrates on GP parameters were not modified. It is concluded that comparison of numerical data from in vitro experiments that follow different protocols must be done carefully. However, the ranking of different substrates is more robust and less affected by the sources of variation.  相似文献

Fecal samples were collected to evaluate mathematical models to describe the kinetics of digesta passage in the segments of the equine gastrointestinal tract and to compare the passage kinetics of hay and oats. Four Norwegian Cold-blooded trotters (cecally cannulated, approximately 500 kg of BW) were fed Cr-mordanted hay and Yb-marked oats with their morning meal. The meal consisted of 2 kg of hay and 1 kg of oats processed as ground, pelleted, extruded, or micronized. Each horse was fed each type of oats on different days of collection, after a 5-d adaptation period, in a 4 x 4 Latin square design. Fecal samples were collected 5, 6, 7, 8, 9, 10, 11, 12, 14, 16, 18, 20, 22, 24, 26, 28, 30, 32, 34, 36, 38, 48, and 52 h after administration of the marker dose. The samples were analyzed for Cr and Yb, and values were plotted using 1- and 2-compartment nonlinear passage models and an algebraic model. The 1-compartment G4 model and the 2-compartment G4G1 model showed an equally good fit to the observed excretion curves, based on low mean square error and SE. The excretion curves for hay (Cr) and oats (Yb) showed a striking similarity, and there seemed to be no difference in retention time between hay and oats in the horse. The mixing compartments in the horse are believed to be the cecum or both the cecum and the right ventral and dorsal segments of the colon, but further research in this area is needed to make a final conclusion.  相似文献

目前犬瘟热发病表现腹泻症状的病犬占很高的比例,而犬瘟热疾病表现消化道症状的病理机制尚未完全阐明。5-羟色胺(5-HT)是消化系统内重要的神经递质和调节物,而且90%以上分布于肠道,在腹泻性的疾病中可能起着巨大作用。为探讨犬瘟热病犬胃肠道5-HT细胞的变化及其在病理中的作用,取5只消化道型犬瘟热病犬胃肠组织,常规切片,SP免疫组织化学技术对病犬胃肠道内5-HT胺细胞进行定位,观察其形态特征与分布密度。结果显示,胃肠道内5-HT胺细胞呈现明显多形性,大小不均一。胃部的5-HT胺细胞主要集中在胃腺上皮细胞之间;肠道内的5-HT胺细胞主要分布于肠上皮基底部、肠道黏膜上皮细胞之间。在消化道型犬瘟热病犬胃肠道中5-HT胺细胞数量明显增多,在胃中5-HT胺细胞的密度在胃幽门处显著高于胃部其他部位,在肠道内结肠处的分布数量显著低于其他肠段,在整个胃肠道内呈不规则波浪形分布,并且5-HT胺细胞密度与腹泻程度明显相关。试验结果提示5-HT胺细胞可能参与犬瘟热发病过程,其增高是由于犬瘟热病毒或者继发感染的其他病原菌破坏消化道的正常组织结构刺激胃肠道内的5-HT胺细胞代偿性增加。  相似文献

The search for alternatives to in-feed antibiotics in animal nutrition has highlighted the role dietary modulation can play in improving gut health. Current antibiotic replacement strategies have involved the use of microbes beneficial to health (probiotics) or fermentable carbohydrates (prebiotics) or both (synbiotics). The present review recognises the contribution of fermented feeds and fermentable carbohydrates in improving the gut environment in non-ruminants. It proposes the screening of probiotic bacteria for the production of fermented feeds and supplementation of these feeds with fermentable carbohydrates prior to feeding animals. It is suggested that the term ‘fermbiotics’ should be used to describe this intervention strategy.  相似文献

Previous studies have indicated that reducing dietary CP may improve N utilization and effectively diminish manure ammonia emissions; however, the response of manure odor emissions to such dietary modifications has been inconsistent. The objective of the current experiment was to induce decreased lactobacilli (DL) numbers in the distal gastrointestinal tract (dGIT; cecum + colon) of finishing pigs offered both high- and low-CP diets through consumption of chitosan, and examine the influence of this model on manure ammonia and odor emissions when compared with a positive control diet. It was hypothesized that an DL population would be accompanied by an increase in markers of protein fermentation. When compared with normal lactobacilli populations in the dGIT, generation of an DL population would result in increased manure odor emissions from pigs offered both dietary CP concentrations. A 2 × 2 factorial arrangement of treatments was conducted to investigate the effect of including chitosan [0 (positive control) vs. 20 g/kg of feed] and high or low dietary CP concentration (200 vs. 150 g/kg of feed) on nutrient digestibility, N utilization, selected bacterial populations, and metabolite composition of the dGIT and manure emissions from finisher pigs (60.3 kg). Consumption of chitosan had no influence (P > 0.05) on nutrient digestibility or N utilization. In both high- and low-CP diets, consumption of chitosan decreased the lactobacilli-to-Enterobacteriaceae ratio (P < 0.01), generating an DL population, and increased pH (P < 0.01) in the dGIT and ammonia (P = 0.02) in the cecum compared with diets that supported normal lactobacilli populations. Consumption of chitosan decreased molar proportions of butyric acid (P < 0.01) and increased valeric acid (P < 0.01) in the dGIT compared with unsupplemented diets. Furthermore, consumption of chitosan increased manure odor emissions (P = 0.05) compared with unsupplemented diets. There was no effect (P > 0.05) of chitosan consumption on manure ammonia emissions from 0 to 240 h. The current study demonstrates that dietary chitosan suppressed populations of lactobacilli in the dGIT. In response, a considerable increase in Enterobacteriaceae, markers of protein fermentation, and manure odor emissions was observed compared with the positive control diet. These effects were observed in pigs offered both high-and low-CP diets. The current study indicates a possible role for lactic-acid bacteria in modulating manure odor emissions relatively independent of the proportions of dietary CP available for fermentation in the dGIT.  相似文献

OBJECTIVE: To compare fermentation characteristics of fructooligosaccharides (FOS) and other fiber substrates that are commonly found in canine diets. SAMPLE POPULATION: Fecal samples from 3 adult dogs. PROCEDURE: The ability of fiber substrates to be used in microbial fermentation reactions was assessed by use of an in vitro fermentation system. Dogs were fed a commercially available food, and feces were collected for use as the microbial inoculum. Substrates used were beet pulp, cellulose, soy fiber, mannanoligosaccharides (MOS), FOS, and 4 inulin products (inulin 1, 2, 3, and 4). Each substrate was incubated anaerobically with fecal inoculum and growth media for 6, 12, and 24 hours, and production of short-chain fatty acids (SCFA) was measured. RESULTS: Total production of SCFA was higher for fermentation of the 4 inulin products and FOS, whereas fermentation of beet pulp, MOS, and soy fiber resulted in moderate concentrations of SCFA. Fermentation of cellulose produced the lowest concentrations of total SCFA without detection of butyrate or lactate. Butyrate production was greatest for fermentation of the 4 inulin products and FOS. Total lactate production was greatest for FOS and inulin 4. As expected, production of SCFA increased for all substrates as fermentation time increased. CONCLUSIONS AND CLINICAL RELEVANCE: Canine fecal microflora ferment FOS-containing substrates in a similar manner, with little fermentation of cellulose-based carbohydrates. Furthermore, results of an in vitro fermentation system indicate that fiber type affects the metabolic activity of microorganisms, thus influencing the amount and nature of the end products of fermentation.  相似文献

There is a discrepancy in the literature on potential digesta separation mechanisms in horses, with both a selective retention of fine and of large particles postulated in different publications. To assess the net effect of such mechanisms, we fed ponies on a hay‐only diet a pulse dose of whole (unchopped) marked hay together with a solute marker, collected faeces on a regular basis, measured marker concentrations in whole faeces and in their large (2.0–16 mm), medium (0.5–1.0 mm) and small (0.063–0.25 mm) particle fraction, and calculated the corresponding mean retention times (MRTs). For comparison, the same experiment was performed in goats. In goats, as expected, MRTsolute (35 hr) was significantly shorter than MRTparticle (51 hr); only a very small fraction of particle marker was excreted as large particles (2%); and the MRT of these large particles was significantly shorter than that of small particles (with a relevant difference of 8.6 hr), indicating that those few large particles that escape the rumen do so mostly soon after ingestion. In ponies, MRTsolute (24 hr) did not differ from MRTparticle (24 hr); a higher fraction of particle marker was excreted as large particles (5%); and the MRT of these large particles was longer than that of small particles (but with a non‐relevant difference of less than 1 hr). These results indicate that no relevant net separation of digesta phases occurs in horses and that selective particle retention mechanisms in the large intestine are unlikely to represent important characteristics of the horse's digestive physiology.  相似文献

The objective of the present study was to evaluate the effects of dietary inulin and sucrose on the fermentation profile of the gastrointestinal microflora in chicken. Day-old broilers (n = 80) were assigned to four dietary treatments, either fed a basal diet or the same diet supplemented with sucrose (4%), inulin (1%) or sucrose and inulin. At day 35, birds were killed and pH, lactate, ammonia, short chain fatty acids (SCFA) and biogenic amines were determined in different parts of the digestive tract. Final body weights and the relative weights of liver, pancreas, crop, gizzard and small intestine were not influenced by treatment. The relative weights of the empty caeca and of the caecal digesta were higher with the diets containing inulin while caecal pH and ammonia were reduced. Lactate concentration was reduced in the crop (p < or = 0.01) and gizzard (p < or = 0.001) of sucrose-fed groups, while it was increased (p < or = 0.01) in the jejunum of inulin-fed group. Ammonia in the crop (p = 0.089) and gizzard (p = 0.067) tended to be lower in the group receiving inulin plus sucrose. Amongst SCFA, only acetate was detected in the crop and gizzard contents that tended to be lower (p = 0.09) in the crop digesta of sucrose plus inulin-fed group. N-butyrate (mol %) was higher (p < or = 0.001) in the caecal digesta of inulin-supplemented groups without affecting total SCFA. Dietary inulin elevated the concentration of putrescine in the jejunal and caecal contents. In the caecal digesta, total biogenic amines were increased (p < or = 0.001) in sucrose plus inulin-fed group without affecting production of biogenic amines in the jejunum. In conclusion, inulin could reduce the pH in the lower gastrointestinal tract of broilers, while sucrose had no acidifying influence in the upper digestive tract. Inulin enhanced the concentration or metabolic activity of butyrate-producing bacteria in the caecum. Further studies are needed to investigate the potential effect of inulin on the intestinal microbial composition.  相似文献

The main objective of the present investigation was to study the impact of fermentation of cereals on the degradation of polysaccharides and other macronutrients in the small intestine and total tract of growing pigs. Eight pigs (initial BW, 34.5 ± 0.9 kg) were used in a replicated 4 × 4 Latin square design. Pigs were cannulated and housed individually in metabolism pens during sample collection. The 4 cereal-based diets were nonfermented liquid barley (NFLB), nonfermented liquid wheat (NFLW), fermented liquid barley (FLB), and fermented liquid wheat (FLW). The fermented feeds were prepared by storing the dietary cereals (barley and wheat) and water [1:2.75 (wt/wt)] in a closed tank at 25 °C for 2 d, after which 50% of the volume was removed and replaced with an equal amount of fresh cereals and water after each afternoon meal. At the time of feeding (0730 and 1430 h), the remaining dietary ingredients were added. Water was added to the dry nonfermented feeds [1:1 (wt/wt)] immediately before feeding. The fermentation process reduced the amount of DM in both cereals (P<0.001), whereas the amount of DM was similar (P=0.626) between the fermented cereals. There was an interaction of cereal and treatment for ileal flow of DM (P=0.014), OM (P=0.013), and protein (P=0.006), which were less in pigs fed the NFLB than the FLB diets, but unchanged in pigs fed the NFLW and FLW diets. Conversely, the ileal flow of protein was similar (P=0.605) in pigs fed the barley diets (average, 47.5 ± 1.7 g/kg of DMI) and increased with the FLW diet compared with the NFLW diet (43 vs. 35 g/kg of DMI, respectively). Ileal fat and CH(2)O digestibilities were 7.6 (P=0.002) and 8.9% (P<0.001) greater, respectively, when pigs were fed wheat compared with the barley-based diets, and the ileal digestibility of CH(2)O was greater when pigs were fed the fermented than nonfermented diets (86.5 vs. 84.5%, respectively; P<0.001). Fermentation reduced (P<0.0001) the fecal excretion of DM, OM, and protein in pigs fed the barley diet, but not when fed the wheat-based diet (P=0.305). Fermentation had no effect (P=0.243) on the fecal digestibility of nonstarch polysaccharides in either of the cereals but their digestibility was 10.0% greater (P<0.001) in pigs fed wheat than the barley-based diets. In conclusion, fermentation of cereal before feeding altered the dietary composition and influenced flow and composition of polysaccharides and other macronutrients at the ileum and in feces to a larger extent for barley than wheat.  相似文献

Four rumen fistulated wether and beef steers were used to evaluate differences in dry matter digestibility (DMD) between cattle and sheep. They were fed either perennial ryegrass or switchgrass hay at an ad libitum or restricted level for four experimental periods. Significant ruminant species X forage and ruminant species X level of intake (P less than .05) interactions were observed for digestible dry matter. The steers digested the switchgrass 7 percentage units greater than the wethers while ryegrass was digested equally. Digestibility differences between the steers and wethers were 6 percentage units at the ad libitum level of intake and 1 unit at the restricted level of intake. Crude protein digestibility tended to be greater (P less than .10) for sheep with a 7 unit difference for switchgrass and a 3 unit difference for ryegrass. The mean ruminal solids retention time of the digesta was approximately (P less than .01) 50% greater (26.0 vs 17.4 h) in cattle, with no difference in ruminal liquid dilution rate (LD) between animal species. Total ruminal volatile fatty acid concentration differed (P less than .01) with level of intake; however, no influence due to intake on the molar proportion of acetate (P greater than .10) or propionate (P greater than .10) was evident in spite of a difference (P less than .01) in LD. Rumen pH (P less than .05) and osmolality (P less than .01) were affected by both level of intake and forage, with the ryegrass and high level of intake decreasing pH and increasing osmolality.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Effects of sucrose (Suc) concentration on fermentation kinetics and products were evaluated using 3 concentrations of Suc, with 1 concentration of isolated NDF from Bermudagrass fermented together in batch culture in vitro with rumen inoculum. Fixed amounts of medium and inoculum were the protein sources, so protein:Suc decreased with increasing Suc. Kinetics were calculated from gas production over 48 h in a randomized complete block design (n = 28), and product yield was evaluated with sampling every 4 h for 24 h in a split-split plot in time design (n = 84). Fermentation vial was the experimental unit. Increasing Suc increased the lag time of rapidly (P < 0.01) and slowly fermented (P < 0.01) fractions and tended to decrease the rate of gas production from the rapid fraction (P = 0.07). Gas production from the slow fraction decreased linearly with increasing Suc (P = 0.02), suggesting a decrease in NDF fermentation. Sucrose was the predominant substrate at 1 indicate incorporation of C from the medium, likely from AA and peptides. The results support the premises that direct effects of Suc concentration and perhaps protein:Suc alter yields of fermentation products. That substrate concentration altered fermentation products and kinetics, possibly due to interactions with the run conditions, advises the clear definition of substrates and fermentation conditions to determine how the results integrate into our knowledge of ruminant nutrition.  相似文献

The aim of this study was to compare the in vitro antimicrobial activity of the veterinary fluoroquinolones against a panel of recently isolated porcine and bovine bacterial pathogens. The study used enrofloxacin as a benchmark against which other agents were compared, being the most common fluoroquinolone used in treatment of bovine and porcine infections. The activity of ciprofloxacin was also assessed as it is the main metabolite of enrofloxacin in cattle. Enrofloxacin and ciprofloxacin generally showed higher antibacterial activity, in terms of MIC(50) values, for most pathogen species when compared with marbofloxacin, difloxacin, danofloxacin and norfloxacin. Ciprofloxacin showed significantly greater in vitro antibacterial activity than enrofloxacin against M. haemolytica, P. multocida and E. coli, whereas enrofloxacin showed greater activity than ciprofloxacin against S. aureus. Marbofloxacin was significantly more active than enrofloxacin against M. haemolytica, E. coli and B. bronchiseptica but less active against P. multocida, S. aureus, coagulase negative Staphylococci, S. dysgalactiae, S. uberis, A. pleuropneumoniae and S. suis. Danofloxacin was significantly less active than enrofloxacin against P. multocida, E. coli, S. uberis, A. pleuropneumoniae and S. suis. Enrofloxacin and its metabolite ciprofloxacin showed the highest in vitro activities against most bovine pathogens tested and the porcine pathogens also showed a high degree of sensitivity to enrofloxacin. These data facilitate further pharmacokinetic/pharmacodynamic comparison of fluoroquinolones currently used in veterinary medicine.  相似文献

This study investigated the effects of allergic skin disease on the penetration kinetics of hydrocortisone through canine skin in vitro. Full-thickness lesional and nonlesional (normal) skin was removed from the dorsal lumbosacral and dorsocaudal thoracic regions, respectively, of five canine cadavers. The dogs were suspected of having flea allergy dermatitis based on their distribution and types of skin lesions. Nonlesional skin was confirmed to be histologically normal, and the histopathology of the lesional skin was consistent with allergic dermatitis. Excised skin was clipped, mounted in Franz-type diffusion cells, and the transdermal penetration of a saturated, radiolabelled hydrocortisone solution was measured over 30 h. When the penetration data for all five dogs were pooled, a restricted (or residual) maximal likelihood mixed model predicted that the permeability coefficient and pseudosteady-state flux of hydrocortisone was more than twice as great (95% confidence interval 1.55-2.71 times as great; P < 0.0001) through lesional compared with nonlesional skin. There was no significant difference in the lag time for hydrocortisone penetration through lesional compared with nonlesional skin of the dogs. This study has confirmed that the transdermal penetration of hydrocortisone may be altered, typically increased twofold, but could be as high as 10-fold, through lesional compared with nonlesional skin of dogs with suspected flea allergy dermatitis. This is likely to be affected by variables such as disease severity, concurrent infections and interindividual differences in skin characteristics.  相似文献
