The production of viable oospores of Peronospora parasitica under laboratory conditions and the recovery of isolates (referred to as sexual progeny) from these oospore populations are described. Oospores were produced when isolates of opposite sexual compatibility type, specialized to the same or different Brassica species, were grown together in seedling cotyledons of a host line capable of supporting growth of both isolates. Recovery of sexual progeny from oospore populations produced from two out of four pairings between isolates specialized to the same host species (homologous pairings) proved relatively easy. On the basis of their characterization with respect to virulence, response to phenylamide fungicides, sexual compatibility type and isoenzyme polymorphisms, there was evidence that the sexual progeny from these homologous pairings could be of hybrid origin. For the first time in a member of the Peronosporaceae, it proved possible to recover and successfully characterize a few sexual progeny from pairings between isolates specialized to different host species (heterologous pairings). However, the majority of such isolates sporulated weakly and as a consequence proved difficult to maintain and were lost. The response of cotyledons of oilseed rape ( Brassica napus ssp. oleifera ) accessions to infection by isolates of Peronospora parasitica under controlled conditions was assessed on a 0–7 scale (disease reaction). In interactions scored 0–3, 4–5 and 6–7, the host was considered resistant, partially resistant and susceptible, respectively. Accession RES-26, selected from the spring oilseed rape cultivar Janetzkis, was partially resistant to isolate R1 and resistant to isolate P003 of P. parasitica , which distinguishes it from three previously described differential response groups ('A', 'B' and 'C') of accessions in B. napus . The resistance of RES-26 to isolate P003 seemed to be conditioned by a single, partially dominant gene and the resistance of RES-02, which belongs to group 'A' (resistant to R1 and P003), by two independent partially dominant genes. The gene for resistance to P003 in RES-26 is either closely linked, allelic or identical to one of the two genes for resistance in RES-02. Resistance of RES-02 to R1 is conditioned by a single, incompletely dominant gene. The genes for resistance to isolates R1 and P003 in RES-02 are either closely linked, allelic or identical. The growth of two isolates of the downy mildew fungus Peronospora parasitica , one obtained from cauliflower ( Brassica oleracea ) and the other from oilseed rape ( B. napus ) was assessed in their respective hosts of origin, and also in the alternative combination. Both isolates were capable of infecting either host, but there were marked contrasts in the time course and extent of mycelial development, the amounts of associated host-cell necrosis, and eventual intensity of sporulation. Oilseed rape, which was partially resistant to the isolate from cauliflower, exhibited extensive necrosis of mesophyll cells, in conjunction with reduced mycelial development, and delayed and reduced sporulation by the pathogen. The isolate from oilseed rape was virulent on both host species. Pathogenesis in the susceptible combinations was accompanied by large increases in electrolyte leakage, and increased activity of the enzymes β-glucosidase, ribonuclease, and peroxidase. Effects on chlorophyll content were variable and activities of acid phosphatase and acid phosphodiesterase were unaffected. Electrophoretic analyses of extracts from fungal sporangia and infected seedlings indicated that the large increases in β-glucosidase were of pathogen origin, while evidence from inhibitor studies suggested that enhanced ribonuclease activity was due to a new post-infectional form of the enzyme. The significance of these findings is discussed in relation to pathogenesis and host resistance mechanisms. 相似文献

The oilseed rape cultivar Cresor was resistant to 14 isolates of Peronospora parasitica derived from crops of Brassica napus in the UK. Segregation for resistance to one isolate among F2 plants and F3 progeny of crosses between Cresor and the susceptible cultivars Victor and Jet Neuf indicated that resistance was controlled by a single gene. There was evidence that genetic background and environment could influence the phenotypic expression of this resistance. Two sexual progeny isolates derived from a homothallic isolate of P. parasitica avirulent on Cresor were completely virulent on this cultivar. This suggested that the parental isolate was heterozygous at a matching locus or loci for avirulence and demonstrated the race-specific nature of the resistance. 相似文献

Seedlings of six cauliflower cultivars ( Brassica oleracea convar. botrytis var. botrytis ) were assessed for resistance to a Danish isolate of Peronospora parasitica , under controlled conditions. Resistance, characterized by restricted sporulation and necrotic dark flecks at the inoculation site on the cotyledons, was expressed in the hybrids 9306 F1, 9311 F1, and the open pollinated cultivar Perfection. Testing of the parent lines and F2 generations of the two resistant hybrids suggested that resistance was a dominantly inherited trait controlled by a single gene. Inoculation of the cultivars with seven isolates, from different geographical origins, showed that the resistance was isolate specific. The two hybrid cultivars expressing cotyledon resistance and two hybrids expressing susceptibility were assessed for adult plant resistance under field conditions. The AUDPC (Area Under the Disease Progress Curve), based on disease incidence and severity, revealed significant differences between the cultivars. At harvest, the cultivars exhibited significantly different levels of defoliation and curd attack. The cultivars 9306 F1 and 9311 F1 showed high levels of resistance in all assessments, whereas the two cultivars exhibiting susceptibility at the seedling stage, 9304 F1 and 9305 F1, also exhibited susceptibility through the adult plant stage. Thus, the resistance exhibited under field conditions resembled that identified at the seedling stage under controlled conditions. Downy mildew disease caused by Peronospora sparsa, also known as ‘dryberry’ disease, is a serious threat to the cultivation of arctic bramble (Rubus arcticus) and boysenberry (Rubus spp. hybrid). A quantitative and sensitive screening method is necessary for the breeding of downy mildew resistant cultivars and for determining efficient disease control methods. A quantitative real-time PCR method using SYBR® Green I fluorescent dye was developed for the analysis of P. sparsa in arctic bramble, other Rubus species and roses. Primers were designed to amplify a P. sparsa specific 94-bp fragment from the internal transcribed spacer region (ITS1) and a 140-bp fragment from a conserved region of plant 5.8S ribosomal DNA, which served as an internal control in the samples. Linear amplification from genomic DNA and control plasmids was achieved with both primers, and even 37 fg of P. sparsa conidial DNA was detected. In the samples collected from the field, quantities as low as 0.2 ppm of P. sparsa DNA in plant DNA were detected, thus enabling the diagnosis of weak and latent infections. Arctic bramble cvs Pima, Mespi and Mesma, all showing distinct foliar symptoms, were tested to assess the relative amount of downy mildew DNA present. The symptoms and the amount of P. sparsa DNA detected correlated only in cv. Pima, indicating that visual inspection of symptoms is not a reliable method for assessing the extent of tissue infection. The number of conidiophores of P. sparsa on in vitro inoculated leaves of two arctic bramble cultivars correlated with the results obtained by real-time PCR. 相似文献

Under controlled inoculation, a set of 56 accessions belonging to 19 wild species of the genusCucumis was studied for resistance to seven isolates of cucurbit downy mildew (Pseudoperonospora cubensis (Berk. et Curt.) Rostow.) from cucumber. No resistance toP. cubensis was detected in theseCucumis accessions. In three host accession/pathogen isolate combinations, limited sporulation was observed. Nine newCucumis species are described as hosts forP. cubensis: C. africanus, C. ficifolius, C. figarei, C. meeusii, C. metuliferus, C. myriocarpus, C. leptodermis, C. sagittatus andC. zeyheri. Of the three isomers of aminobutyric acid, only the β isomer (BABA) was effective in inducing resistance against Peronospora parasitica , the causal agent of downy mildew, in cauliflower ( Brassica oleracea var. botrytis ). A single foliar spray applied to 7-day-old seedlings protected the plants against Peronospora parasitica for at least 15 days. Of the enantiomers (R and S), only the R was effective. Resistance was accompanied by a hypersensitive-like reaction (necrotic spots) which was evident before inoculation. BABA was systemically effective when applied to the roots, but failed to protect cotyledons adjacent to treated ones. Unlike other chemical inducers, BABA was effective when applied several hours postinoculation. It had no effect on P. parasitica spore germination. In cauliflower seedlings, BABA did not induce the accumulation of the pathogenesis-related protein PR-1, PR-2, PR-3, PR-5 and PR-9. Only treated and challenged seedlings accumulated PR-2. 相似文献

A total of 101 Brassica napus ssp. oleifera accessions with seed differing in glucosinolate and erucic acid contents were screened for resistance to four isolates of Peronospora parasitica at the cotyledon stage. Two groups of accessions with different resistance factors were identified. Lines that were homogeneous for resistance were selected from seedling populations of accessions that exhibited a heterogeneous reaction to some isolates. The resistance of one group differs from that of cv, Cresor, the only oilseed rape cultivar reported to have an isolate-specific gene for resistance to P. parasitica. The isolate specificity of the second group was identical to that of cv, Cresor, A comparison of the response of host accessions which expressed moderate to full susceptibility at the cotyledon stage, with no clear differential response to any of the four P. parasitica isolates, indicated that those with high glucosinolate and high erucic acid contents (12 accessions) were slightly but significantly less susceptible than those with high glucosinolate and low erucic acid (19 accessions), or low glucosinolate and low erucic acid contents (28 accessions). The mean differences between accessions with low erucic acid but differing in glucosinolate content were inconsistent. The last result was further confirmed by investigating the expression of resistance to three isolates of P. parasitica at three different seedling growth stages among 11 accessions of oilseed rape with seeds low in erucic acid but differing in glucosinolate content. 相似文献

Yellowish lesions with downy, gray growth developed on the leaves of carnations (Dianthus caryophyllus) in a greenhouse in Hokkaido Prefecture, Japan, in March 2011. We identified the fungus on diseased plants as Peronospora dianthicola on the basis of the morphologies of the conidia, conidiophores, and oospores. To confirm pathogenicity of the fungus, we inoculated healthy plants with a conidial suspension of the fungus, and the plants developed the same signs and symptoms as the naturally infected plants. We then reidentified P. dianthicola from the diseased lesions. This is the first report of downy mildew of carnation in Japan. 相似文献

The development of downy mildew on spinach (Peronospora farinosa f. sp.spinaciae) was stratified according to leaf layers and represented in three-dimensional computer graphics, in which percentage diseased leaf area was plotted against time for each leaf layer. Distinction was made between a point source and an area source. Variation for virulence among Scandinavian isolates of Peronospora viciae f.sp. pisi (downy mildew) on different pea genotypes was investigated. Variation for virulence was found within and between mass-conidial isolates which were originally obtained from oospore populations. Virulence towards pea cultivars that have not been commerically cultivated in Scandinavia was observed. The specific resistance of the pea cultivars Puget, Cobri, Gastro, Starcovert and Starnain was confirmed. One pea breeding line showed a stable partial resistance to different isolates of the fungus and it was also highly resistant to race 8'from The Netherlands which is considered to be virulent on most pea genotypes carrying known specific resistance factors. 相似文献

The anamorphic state of a powdery mildew, which first appeared on tomato in the UK in 1987, currently listed as Erysiphe orontii (anamorph Oidium violae ), was examined microscopically on both tomato and alternative hosts. Host range studies were also carried out. In 1993–95, species and cultivars of plants from 26 of the 30 families listed as hosts of E. orontii were inoculated with tomato powdery mildew in the glasshouse and assessed for susceptibility on a 0–3 scale based on degree of sporulation and growth. Thirteen families contained species or varieties that were susceptible and a total of 80 varieties, including many economically important Cucurbitaceae and Solanaceae, supported sporulation to some extent (susceptibility score 1–3). Necrosis was not a constant feature of plants exhibiting resistance, as it could accompany sporulation on some susceptible varieties. Microscopic measurements showed that conidial size, conidiophore length, conidiophore foot-cell length and width, and the number of cells distal to the foot-cell were remarkably constant between samples of powdery mildew taken from tomato each year and from the various alternative hosts. Based on the production of conidia singly, the presence of nipple-shaped to moderately lobed appressoria, and occurrence of straight conidiophores, it is concluded that this fungus on tomato should more correctly be named Oidium lycopersicum until the sexual stage is discovered. The highly polyphagous nature of this powdery mildew would suggest that it represents a potential risk to UK agriculture and horticulture, but the failure to recover any powdery mildew from 120 samples of potato, including varieties Cara and Maris Piper, on which the tomato powdery mildew could sporulate in the glasshouse, suggests that environmental factors may play a key role in preventing establishment of this powdery mildew on alternative hosts on a commercial scale in the field. 相似文献
