Cyanobacteria are producers of potent and environmentally abundant microcystins, representing an emerging global health issue. In the present study, we investigated the impact of cyanobacterial biomass on biochemical indices of common carp (Cyprinus carpio L., average weight of 246 ± 73 g) under laboratory conditions. The fish were fed a diet containing cyanobacterial biomass with microcystins in high concentration (0.4 mg/kg of fish weight and day) for 28 days. Statistical evaluation of the influence of the cyanobacterial biomass in food on the biochemical indices of the juvenile carp showed only minor differences. The activity of aspartate aminotransferase value and the urea concentration were significantly reduced compared to control group. The biochemical parameters of fish blood plasma significantly rose during the experiment in the control group as well as in the experimental group. This state was probably influenced by the environmental conditions and the fish diet. A significant rising value was established in calcium creatinine, total protein, phosphorus, lactate, urea and natrium. The present study demonstrates that the oral exposure of toxic cyanobacterial biomass has a minor influence on the biochemical indices of common carp and that the effect of other factors, e.g., nutrition is more visible.  相似文献

A non‐classical biomanipulation experiment was carried out in Gonghu Bay of Lake Taihu in 2009. Silver and bighead carp were stocked in a large fish enclosure to control cyanobacterial blooms. Water quality, plankton abundance, and the intracellular and extracellular microcystins (MCs) in lake water were investigated monthly in 2009. The concentrations of nitrogen nutrients were significantly lower in the fish enclosure than in the surrounding lake, while phosphorus (especially total phosphorus) concentration was higher in fish enclosure. During the blooming period, Cyanophyta contributed to more than 90% of the phytoplankton in the surrounding lake, whereas it represented only 40–80% in the fish enclosure. The phytoplankton and crustacean zooplankton biomasses and the zooplankton/phytoplankton ratios were all significantly lower in the fish enclosure than in the lake. This result suggested that silver and bighead carp can effectively suppress the phytoplankton biomass with the initial stocking density of 7.5 g m?3 for silver carp and 1.1 g m?3 for bighead carp, despite a simultaneous decrease in the grazing pressure of the zooplankton on the phytoplankton. During the blooming period, the intracellular and extracellular MCs in the fish enclosure were reduced by 93.8% and 69.8% compared with the surrounding lake. MCs content varied from 0.34 to 18.8 ng (mean 4.8 ng) MC‐LReqg?1 wet weight in the muscle sample of silver and bighead carp in the experimental enclosure, which suggested that these fish were safe to consume for human. However, the long‐term effects of MCs on aquatic ecosystem and on public health cannot be overlooked.  相似文献

Stress load on harvest‐size common carp (Cyprinus carpio L.) just before harvest, during harvest and during the post‐harvest storage were assessed using haematological and biochemical indices. The study was carried out at the 60 ha Dřemliny pond for 28, 7 and 1 day before harvest, during 3‐day harvest and after a 28‐day post‐harvest period. Indices monitored included the relative spleen weight (SSI), erythrocyte count (Er), haemoglobin levels (Hb), haematocrit (PCV), MCV, MCH, MCHC, leucocrit (Bc), differential leucocyte count (leucogram), blood plasma cortisol, glucose, total proteins (TP), cholesterol and chloride. During harvest, a significant decrease of SSI (P<0.05; P<0.01), increase of PCV (P<0.01), decrease of Bc (P<0.01, P<0.05), lymphopenia (P<0.05) and neutrophilia (P<0.01) were observed. A significant increase in the levels of blood plasma cortisol was found 1 day before harvest (P<0.05) due to draining off the pond before harvest itself, hyperglycaemia was observed 1 day before harvest (P<0.05) and during harvest (P<0.01); a decrease, on the other hand, was found in cholesterol and chloride levels (P<0.01). The relationships between oxygen content in water and SSI and MCHC were positive (P>0.05) and between oxygen and glucose were negative (P>0.01). A 28‐day post‐harvest storage returned the indices investigated back to physiological interval of values. Persistently lower levels of Bc, lymphopenia, neutrophilia and lower TP concentrations are indicative of deepening immunosuppression during the post‐harvest storage period. Changes of haematological and biochemical indices during harvest in relation to hypoxia, fish handling and higher stocking rates are also discussed.  相似文献

  • 1. In Australia, the carp Cyprinus carpio L. is regarded as a threat to the native fish and the aquatic environment. In recent years, Botany Wetlands, a significant coastal wetland in the Sydney region, has been invaded by the undesirable cyprinids, carp and goldfish (Carrasius auratus L.).
  • 2. In 1996 a cyprinid removal programme commenced at Botany Wetlands with the objective of managing the invasive species, increasing fish diversity, reducing cyanobacterial blooms and hence enhancing the aquatic habitat. Using electrofishing and gill netting, 4073 carp and 261 goldfish, amounting to 10 117 kg of cyprinid biomass were removed between 1996 and 2004. The captured carp ranged from 60 to 835 mm. Males matured before females. Carp between 350 and 680 mm in length grew at the rate of 1.66±0.38 g day?1.
  • 3. The success of the programme was monitored by assessing four indicators related to carp populations and two related to habitat. The former included the pattern of length–frequency distribution, mean weight per size class, condition factor (CF) and the catch per unit effort (CPUE), and the latter the Secchi disc transparency and cyanobacterial counts. After 8 yr of removal, the plots of carp length–frequency distribution flattened, CF decreased from 2.86 to 1.82 and CPUE decreased from 97 kg day?1 to 50 kg day?1. A 10‐fold decrease occurred in cyanobacterial counts and the Secchi disc transparency increased by 20%. More than 20 000 fingerlings of native Australian bass Macquaria novemaculeata (S.) have been introduced to the wetlands, increasing the potential for juvenile carp predation and biodiversity.

Healthy crucian carp (Carassius auratus) were treated by intraperitoneal (i.p.) injection of crude cyanobacterial extracts at two doses, 50 and 200 μg MC-LR equiv kg−1 BW. High mortality (100%) was observed within 60 h post injection in the high-dose group. In the treated fish, activities of four plasma enzymes, alanine aminotransferase (ALT), alkaline phosphatase (ALP), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH), all showed substantial increases, with both dose and time-dependent effects. These increases of enzyme activity indicate severe impairment occurred in the liver of crucian carp over time. Plasma concentrations of energy-related biomolecules including glucose (GLU), cholesterol (CHO), triglyceride (TG), and total protein (TP) showed marked changes in the high-dose group, possibly a nutritional imbalance correlated with the liver injury caused by intraperitoneal exposure to crude cyanobacterial extracts.  相似文献

Erythrocytes of olive flounder, Paralichthys olivaceus (Temminck et Schlegel), were treated with serial concentrations of formalin (37% formaldehyde) to investigate in vitro haemolysis and methaemoglobin formation. In addition, the short‐term toxicity of formalin concentrations of 0, 100, 212 and 300 ppm was also studied by clinical tests in which fish were subjected to 3‐h bath exposure. There was no haemolysis of fish erythrocytes exposed to formalin concentrations ranging from 31.3 to 2000 ppm. Methaemoglobin formation, however, was induced at concentrations greater than 500 ppm. Red blood cell count, haemoglobin, haematocrit, mean corpuscular haemoglobin concentration and percentage of immature erythrocytes were also markedly elevated in all formalin‐exposed groups (P<0.05). Formalin exposure also caused significant increases in alkaline phosphatase, aspartate aminotransferase, lactate dehydrogenase, potassium, chloride, magnesium and inorganic phosphorus (P<0.05). However, total protein decreased significantly in the formalin‐exposed groups (P<0.05). No significant differences in white blood cell count, mean corpuscular volume, mean corpuscular haemoglobin, albumin, glucose, total cholesterol, high‐density lipoprotein cholesterol, free cholesterol, alanine aminotransferase, calcium, creatinine and total bilirubin were observed in the formalin‐exposed groups (P>0.05).  相似文献

Silver carp Hypophthalmichthys molitrix (Valenciennes) were co‐cultured with channel catfish Ictalurus punctatus (Rafinesque) in 0.4 ha earthen ponds to determine the impacts of carp grazing on pond phytoplankton communities and cyanobacterial off‐flavours in catfish. Carp were stocked at densities of 0, 75, or 250 fish ha?1 in seven replicate ponds per treatment. The mean chlorophyll a concentrations (a measure of phytoplankton standing crop) steadily increased in all treatments from about 100 μg L?1 in April to more than 400 μg L?1 by mid‐October. Silver carp had no affect (P>0.1) on chlorophyll a concentrations across all sampling dates (April though October) or for sampling dates late in the growing season (August–October) when the prevalence of cyanobacterial off‐flavours among catfish populations is usually greatest. Silver carp did not eliminate odour‐producing cyanobacteria from pond phytoplankton communities: on sampling dates in September and October, three to six ponds in all treatments contained populations of the odour‐producing cyanobacteria Oscillatoria perornata, Anabaena spp., or both. Failure of silver carp to eliminate odour‐producing cyanobacteria resulted in a relatively high incidence in all treatments of ponds with off‐flavoured catfish. On sampling dates in September and October, catfish in three to five ponds in each treatment were tainted with either musty (2‐methylisoborneol) or earthy (geosmin) off‐flavours. The presence of silver carp had no obvious effect on off‐flavour intensity: on each sampling date, at least three ponds in each treatment contained catfish described as distinctly to extremely off‐flavored. Apparently, hypertrophic conditions in catfish ponds overwhelm the effect of silver carp grazing at the low carp densities used in this study.  相似文献

The phytoplanktivorous silver carp is an important biomanipulation fish to control cyanobacterial blooms and is also a food fish with the greatest production in China. The accumulation of the hepatotoxic microcystins (MCs) determined by LC-MS in various organs of silver carp was studied monthly in Lake Taihu dominated by toxic Microcystis aeruginosa. Average recoveries of spiked fish samples were 78% for MC-RR and 81% for MC-LR. The highest content of MCs was found in the intestine (97.48 μg g− 1 DW), followed by liver (6.84 μg g− 1 DW), kidney (4.88 μg g− 1 DW) and blood (1.54 μg g− 1 DW), and the annual mean MC content was in the order of intestine > liver > kidney > blood > muscle > spleen > gallbladder > gill. Silver carp could effectively ingest toxic Microcystis cells (up to 84.4% of total phytoplankton in gut contents), but showed fast growth (from 141 g to 1759 g in 1 year in mean weight). Silver carp accumulated less microcystins in liver than other animals in the same site or other fish from different water bodies at similar level of toxin ingestion. There was possible inhibition of the transportation of the most toxic MC-LR across the gutwall. Muscle of silver carp in Lake Taihu should not be consumed during period of dense Microcystis blooms while viscera were risky for consumption in more months.  相似文献

Dietary thiamin requirement of juvenile grass carp, Ctenopharyngodon idella, was to investigate in this experiment. Eight purified diets were formulated with graded levels of thiamin (0.1, 0.6, 1.1, 2.1, 5.5, 9.8, 21.2, and 41.8 mg/kg, respectively). Each diet was fed to triplicate groups of 40 fish (initial average weight 10.7 ± 0.2 g) for 12 wk in 400‐L aquaria (R = 1 m, h = 0.6 m). Results showed that weight gain rate, specific growth rate, feed efficiency, protein efficiency ratio, and hepatosomatic indice of fish increased before dietary thiamin increased to the optimum level, then remained similar thereafter (P > 0.05). Thiamin concentration in fish liver was positively correlated with dietary thiamin and it stayed in stable when dietary thiamin level exceed 5.0 mg/kg. The serum biochemical indices analysis showed that dietary thiamin had significant effects on serum triglycerides, total cholesterol, glucose, pyruvate contents, and lactate dehydrogenase activity. Body composition was unaffected by dietary thiamin. Broken‐line regression analysis showed that, a dietary thiamin level of 1.3 mg/kg diet was adequate for optimum growth, and 5.0 mg/kg for maximum liver thiamin accumulation.  相似文献

The haematological and biochemical characteristics of two healthy farmed cyprinids, the topmouth culter Culter alburnus and yellowcheek carp Elopichthys bambusa, were investigated in this study. Erythrocytes, thrombocytes, lymphocytes, monocytes and granulocytes (i.e. neutrophils and eosinophils) were observed in these two fish. Every type of these cells (excluding the erythrocyte and lymphocyte) showed similar sizes in the topmouth culter and yellowcheek carp. Thrombocytes and neutrophils were the two most abundant leucocytes in the topmouth culter while thrombocytes and lymphocytes were the two most frequent leucocytes observed in the yellowcheek carp. The erythrocyte counts, haemoglobin concentrations and values of serum glucose in these two fish were high. There were significant differences in the leucocyte counts, haemoglobin concentrations, mean cellular haemoglobin contents, mean cell haemoglobin concentrations and values of serum glucose, triglyceride, total bilirubin, alkaline phosphatase and chlorine between the topmouth culter and the yellowcheek carp. The information of haematology and blood biochemistry obtained here would be useful for the prevention and diagnosis of diseases of farmed topmouth culter and yellowcheek carp.  相似文献

The aim of this study was to evaluate the dynamics of the blood plasma parameters and the content of microcystins in the tissues of common carp ( Cyprinus carpio L.) in relation to the toxic cyanobacterial water bloom. Fish (average body mass 2176±697 g) in the hypertrophic pond were exposed to natural water bloom (dominated by Planktothrix agardhii, Pseudanabaena limnetica and Limnothrix redekei ), which contained microcystins (concentration in biomass 20–181 μg g−1 dry wt, concentration in water 0.3–9.5 μg L−1). Biochemical parameters in fish blood plasma were analysed in 89 fish at 14-day intervals during the whole season (nine sampling periods). Our results demonstrated high variability and fluctuations in the investigated parameters. The content of microcystins and density of cyanobacterial cells correlated with some haematological indices as lipase, alanine–aminotransferase, albumin, magnesium and chlorides. The concentrations of microcystins in the muscle and liver of the fish (determined by high-performance liquid chromatography with mass spectrometer) were below the limit of detection during the monitored period [0.31 ng g−1 fresh weight (f.w.) for the liver and 0.13 ng g−1 f.w. for muscle]. Our study demonstrates that although known cyanobacterial toxin microcystins were not detected in the fish tissues, several biochemical parameters important for the fish physiology were modulated by natural cyanobacterial bloom.  相似文献

Haematological parameters of 2‐year‐old carp (Cyprinus carpio L.) were assessed to study the protective effect of chloride on the health of fish exposed to elevated nitrite concentrations. Four groups of carp were exposed to different concentrations of nitrite and chloride for 96 h (group E1: 67 mg L?1 NO2?, 11 mg L?1 Cl?; group E2: 67 mg L?1 NO2?, 100 mg L?1 Cl?; group E3: 0 mg L?1 O2?, 100 mg L?1 Cl? and group C: 0 mg L?1 NO2?, 11 mg L?1 Cl?). The main haematological response of carp to an acute exposure to nitrite (group E1) was a significant decrease (P<0.05) in haemoglobin concentrations (53.40±6.61 g L?1), haematocrit (0.21±0.02 LL?1), erythrocyte count (1.13±0.12 TL?1), leucocyte count (7.1±4.19 GL?1) and lymphocyte count (5.28±2.51 GL?1), and a significant increase in methaemoglobin concentration (90.50±4.38%, P<0.01) and mean corpuscular haemoglobin concentration (0.27±0.2 LL?1, P<0.05). At higher chloride concentrations (group E2), a lower nitrite toxicity was observed. In group E2 carp, methaemoglobin made up 38.32±13.30%. Erythrocytes in carp exposed to nitrite showed qualitative changes. Compared with the control group C, group E1 carp showed a significantly higher number (P<0.05) of elongated erythrocytes, with the nucleus located at one cell pole (0.519±0.388 TL?1). All erythrocytes of group E1 carp had remarkably clear cytoplasms compared with the cytoplasm in the control group C. The biochemical values found were comparable with those found in controls. The main histological lesions were found in the gills of carp exposed to nitrite and consisted of hyperplasia and an elevated number of chloride cells.  相似文献

A growth trial was conducted to estimate the optimum concentration of dietary available phosphorus (P) for grass carp (Ctenopharyngodon idella). Triplicate groups of grass carp (5.59 ± 0.02 g) were fed diets containing graded levels (2.36, 4.27, 6.31, 8.36, 10.4 and 14.8 g kg?1) of available P for 8 weeks. Grass carp fed with the P‐supplemented diets had significantly higher specific growth rate, weight gain, protein efficiency ratio and feed efficiency than fish fed with the basal diet. In whole‐body composition, protein content increased, while lipid content decreased with the increase in P level in diet (P < 0.05). Fish fed with the P‐supplemented diets had significantly higher whole body, vertebrae and scales mineralization (P < 0.05), but Ca/P ratios were not influenced. The blood chemistry analysis showed that dietary available P had distinct effects on P, Ca and Mg contents, as well as on the contents of triacylglycerol and total cholesterol. Broken‐line analysis indicated that 8.49 g kg?1 dietary available P was required for maximal tissue storage and mineralization as well as optimal growth.  相似文献

The nitrite threshold concentration in rearing water of African catfish (Clarias gariepinus) was assessed. African catfish with an initial mean (SD) weight of 219.7 (57.8) g were exposed to an increasing range of water nitrite from 6 (Control) to 928 μM nitrite for 28 days. Mean (SD) plasma nitrite concentrations increased from 5.0 (3.6) to 32.5 (12.6) μM at 928 μM ambient nitrite. The increase in nitrite was accompanied by gradual increase in plasma nitrate from 41.6 (28.4) μM to 420.2 (106.4) μM. Haematocrit, haemoglobin, methemoglobin, plasma concentrations of cortisol, glucose, lactate, osmolality, gill morphology and branchial Na+/K+‐ATPase activity were not affected. Feed intake, final weight, SGR, FCR and mortality were not affected. We advise not to exceed a water nitrite concentration of 43 μM (0.6 mg L?1 NO2?‐N) to prevent the risk of reduced growth and feed intake in African catfish aquaculture.  相似文献

This study investigated the haematological and blood biochemical characteristics of Glyptosternum maculatum. The haematological and biochemical parameters were measured in 30 adult fish collected from Nyingchi Reach of Yarlung Zangbo River in Tibet. The red blood cell count (RBC), haemoglobin concentration (Hb), haematocrit (Hct), erythrocyte osmotic fragility (maxEof and minEof), the erythrocyte sedimentation rate, mean cell volume (MCV), mean cellular haemoglobin content (MCH), and mean cell haemoglobin concentration (MCHC) were determined. Compared with other Siluriformes fishes, G. maculatum showed similar mean values for Hct, Hb, MCH, and MCHC and had slightly lower RBC and higher MCV. The biochemical parameters were assayed including alanine aminotransferase, aspartate aminotransferase (AST), alkaline phosphatase, total protein, albumin, globulin, albumin/globulin ratio, total bilirubin, direct bilirubin, urea, creatinine, glucose, total cholesterol, and triglyceride. The result showed that the value of AST in G. maculatum was obviously higher than that in Rhamdia quelen as well as in Silurus merdionalis.  相似文献

The present study was conducted to investigate the effects of fenugreek seed meal (FKSM) on growth performance, blood haematological and biochemical factors, survival and stress resistance of the common carp (2.46 ± 0.06 g) fingerlings. Four practical diets containing 0 (control), 0.5%, 1% and 1.5% FKSM were used to feed fish. Fish were fed on the tested diets at a rate of 3% of body weight for 8 weeks. Results indicated that fish fed the control diet had a significantly lower weight gain, specific growth rate and condition factor compared to the other treatments (P < 0.05). Feed conversion ratio decreased significantly in fish fed diets containing FKSM. There were no significant differences in glucose levels, red blood cell count, haematocrit and haemoglobin between treatment groups and the control group (P > 0.05). Significantly lower white blood cells count was observed in fish treatment with FKSM when compared with the control group (P < 0.05). The cholesterol level in group fed supplemented diet by 1% fenugreek significantly lower than other groups (P < 0.05). Total protein levels were significantly higher in all treatments compared to the control (P < 0.05). No mortality was recorded during the feeding trial. In salinity stress experiment, highest survival rate belong to fish fed supplemented diet by 1% fenugreek. These results indicate that FKSM can be considered as a beneficial dietary supplement for improving the growth performance and blood indices of common carp fingerling.  相似文献

A 60-day feeding trial was conducted to estimate the optimum phosphorus requirement of juvenile bighead carp (Aristichthys nobilis). Fish (initial body weight: 2.42?±?0.08 g) were hand-fed with six isoproteic (437 g kg?1) and isolipidic (68 g kg?1) diets containing graded phosphorus levels (0.90, 4.40, 8.30, 11.90, 15.50, and 19.30 g kg?1) thrice daily to apparent satiation. Each diet was randomly assigned to triplicate tanks, and each tank was stocked with 30 fish. The highest weight gain rate (WGR, 288.94%) and specific growth rate (2.28% day?1) and the best feed conversion rate (FCR, 1.91) were recorded in fish fed 8.30 g kg?1 phosphorus. The body composition analysis showed that the phosphorus contents in the whole body, muscle, vertebra, and plasma of fish fed the phosphorus-supplemented diets were higher than those of fish fed the control diet, whereas the phosphorus retention rate and crude lipid contents in the whole body and muscle presented the reverse results. The highest activity of lipase (41.97 U g?1 prot) in the intestine was found in fish fed the diet with 11.90 g kg?1 phosphorus. Further, the contents of total protein, albumin, and globulin in plasma were increased as dietary phosphorus levels ranged from 0.90 to 11.90 g kg?1 and then decreased with further increased phosphorus levels. The highest contents of triglyceride (1.85 mmol L?1) and total cholesterol (2.16 mmol L?1) in plasma occurred at dietary phosphorus level of 0.90 g kg?1. Broken-line model analysis based on WGR, FCR, and the phosphorus contents of the whole body and vertebra indicated that the optimal phosphorus requirements for juvenile bighead carp were 7.16, 9.02, 10.88, and 11.04 g kg?1, respectively.

Previous research showed that stocking 1.5 rohu (Labeo rohita) and 0.5 common carp (Cyprinus carpio) m−2 yields the highest production in small holder ponds in Bangladesh. The present study looked into the effects of additional stocking of Nile tilapia (Oreochromis nilotica) in fed or non‐fed ponds on water quality and fish production. A low, additional stocking density of 0.2 Nile tilapia m−2 was tested. All treatments were executed in triplicate in 100 m2 ponds and the duration of the experiment was 4.5 months. The results showed that tilapia addition increased nutrient concentrations and reduced total suspended solid concentration and phytoplankton biomass (P<0.05). Tilapia stocking resulted in additional production without affecting the growth and production of rohu and common carp. Supplemental feeding increased the nitrogen and phosphorus concentrations, phytoplankton availability and the growth and production of rohu and common carp (P<0.01). The combination of supplemental feeding and tilapia stocking resulted in a higher net yield than the other treatments (P<0.05). Stocking 1.5 rohu, 0.5 common carp and 0.2 tilapia m−2 in fed‐ponds is a good culture combination for polyculture farmers in South Asia.  相似文献
