In artificially inseminated cows (AI on day 1) peak concentrations of oestradiol-17 beta in defatted milk occurred at median times of day 0 during the pre-ovulatory period, days -5 to 2, and day 6 during the post-ovulatory period, days 2-15. Median peak concentrations during these periods were approximately 5 pg/ml and 3 pg/ml respectively. There were no significant differences in the timing or magnitude of oestradiol-17 beta concentrations between cows that became pregnant to the AI and those that entered normal length oestrus cycles immediately after AI. 相似文献

为了研究同期发情定时输精技术对牛群体繁殖力的影响,试验选取同一牛场内不同单元的经产奶牛,随机分为2个组,分别采用自然发情输精和同期发情定时输精两种方法对奶牛进行人工输精,比较两种方法对奶牛群体繁殖力的影响。研究结果显示:同期发情组和自然发情组的参配率分别为,产后54 d之前为96.3%VS 12.1%,55~90 d为64.6%VS 44.6%,91~130 d为48.1%VS 50.1%,131~170 d为39.9%VS 38.7%,180 d内的总参配率分别为249.1%VS 145.5%;相应时段的同期发情组的受胎率为39.2%、36.7%、36.5%、33.9%、92.6%,自然发情组的受胎率为44.8%、47.3%、46.2%、46.1%、 67.5%;2个组的年繁殖率分别为93.8%VS 84.6%、空怀天数为(91.6±18.3)d VS (131.7±26.8)d、配种次数分别为2.5 VS 1.8、淘汰率分别为29.1%VS 13.4%。以上结果表明,在奶牛生产中,使用同期发情定时输精技术,能显著提高牛群的繁殖力,缩短空怀时间。 相似文献

The effect of two insemination policies after synchronization of oestrus on reproductive performance in two groups of cows was investigated. Oestrus was synchronized by two treatments with prostaglandin F2 alpha (PGF2 alpha) at a 14-day interval. Cows in Group 1 (n = 234) were inseminated twice on appointment 66 and 90 h after the second treatment. Cows in Group 2 (n = 222) were intensively watched for signs of oestrus after the synchronization protocol and inseminated on observed oestrus. Cows with abnormal discharge after synchronization were excluded from breeding and inseminated later during the study period. Service rate within 1 week after synchronization was higher in Group 1 than in Group 2 (89.3 versus 67.1%). Conception rates on first service did not differ between groups (33.2 versus 30.0%). Days to first service and days open were shorter in Group 1 (P < 0.05). The number of cows culled for infertility did not differ between the groups. Endometritis 14-20 days post-partum decreased the percentage of cows pregnant at the end of the study period in both groups but did not have a significant effect on conception rates and days open. It is concluded that additional inseminations required in the timed artificial insemination protocol were economically justified by the reduction in days open in comparison with insemination on observed oestrus after synchronization. 相似文献

We determined whether an ovulatory estrus could be resynchronized in previously synchronized, AI nonpregnant cows without compromising pregnancy from the previous synchronized ovulation or to those inseminated at the resynchronized estrus. Ovulation was synchronized in 937 suckled beef cows at 6 locations using a CO-Synch + progesterone insert (controlled internal drug release; CIDR) protocol [a 100-microg injection of GnRH at the time of progesterone insert, followed in 7 d by a 25-mg injection of PGF(2alpha) at insert removal; at 60 h after PGF(2alpha), cows received a fixed-time AI (TAI) plus a second injection of GnRH]. After initial TAI, the cows were assigned randomly to 1 of 4 treatments: 1) untreated (control; n = 237); 2) progesterone insert at 5 d after TAI and removed 14 d after TAI (CIDR5-14; n = 234); 3) progesterone insert placed at 14 d after TAI and removed 21 d after TAI (CIDR14-21; n = 232); or 4) progesterone insert at 5 d after TAI and removed 14 d after TAI and then a new CIDR inserted at 14 d and removed 21 d after TAI (CIDR5-21; n = 234). After TAI, cows were observed twice daily until 25 d after TAI for estrus and inseminated according to the AM-PM rule. Pregnancy was determined at 30 and 60 d after TAI to determine conception to the first and second AI. Pregnancy rates to TAI were similar for control (55%), CIDR5-14 (53%), CIDR14-21 (48%), and CIDR5-21 (53%). A greater (P < 0.05) proportion of nonpregnant cows was detected in estrus in the CIDR5-21 (76/110, 69%) and CIDR14-21 (77/120, 64%) treatments than in controls (44/106, 42%) and CIDR5-14 (39/109, 36%) cows. Although overall pregnancy rates after second AI service were similar, combined conception rates of treatments without a CIDR from d 14 to 21 [68.7% (57/83); control and CIDR5-14 treatments] were greater (P = 0.03) than those with a CIDR during that same interval [53.5% (82/153); CIDR5-21 and CIDR14-21 treatments]. We conclude that placement of a progesterone insert 5 d after a TAI did not compromise or enhance pregnancy rates to TAI; however, conception rates of nonpregnant cows inseminated after a detected estrus were compromised when resynchronized with a CIDR from d 5 or 14 until 21 d after TAI. 相似文献

Most fixed-time insemination protocols utilize an injection of GnRH at the beginning of the protocol to initiate a new follicular wave. However, the ability of GnRH to initiate a new follicular wave is dependent on the stage of the estrous cycle. We hypothesized that administering PGF(2α) 3 d before initiating a fixed-time AI protocol would improve synchrony of follicular waves and result in greater pregnancy success. Therefore, our objective was to determine whether inducing luteal regression 3 d before a fixed-time AI protocol would improve control of follicular turnover and pregnancy success to fixed-time AI. Multiparous crossbred cows at 3 locations (n = 108, 296, and 97) were randomly assigned to 1 of 2 treatments: 1) PGF(2α) [25 mg; intramuscularly (i.m.)] on d -9, GnRH (100 μg; i.m.) and insertion of a controlled internal drug-releasing device (CIDR) on d -6, PGF(2α) (25 mg; i.m.) and CIDR removal with PGF(2α) (25 mg; i.m.) at CIDR removal on d 0 (PG-CIDR) or 2) GnRH (100 μg; i.m.) and insertion of a CIDR on d -5 and CIDR removal with PGF(2α) (25 mg; i.m.) at CIDR removal and 4 to 6 h after CIDR removal (5-d CIDR). Cows were time-inseminated between 66 and 72 h (PG-CIDR) or 70 to 74 h (5-d CIDR) after CIDR removal, and GnRH was administered at the time of fixed-time AI. At location 1, ovulatory response to the first injection of GnRH was determined by ultrasonography at the time of GnRH and 48 h after GnRH administration. Among cows with follicles ≥10 mm in diameter, more (P = 0.03) PG-CIDR-treated cows ovulated after the initial GnRH injection (88%, 43/49) compared with the 5-d CIDR-treated cows (68%, 34/50). Pregnancy outcome was not influenced by location (P = 0.96), age of the animal (P = 1.0), cycling status (P = 0.99), BCS (P = 1.0), or any 2-way interactions (P ≥ 0.13). However, pregnancy success was influenced by synchronization protocol (P = 0.04). Pregnancy outcome was greater (P = 0.04) for the PG-CIDR protocol (64%) compared with the 5-d CIDR protocol (55%). In summary, control of follicular turnover was improved by inducing luteal regression 3 d before initiation of a fixed-time AI protocol, and pregnancy success was improved with the PG-CIDR protocol compared with the 5-d protocol. 相似文献

Mating and calving records for 47,533 first-calf heifers in Australian Angus herds were used to examine the relationship between days to calving (DC) and two measures of fertility in AI data: 1) calving to first insemination (CFI) and 2) calving success (CS). Calving to first insemination and calving success were defined as binary traits. A threshold-linear Bayesian model was employed for both analyses: 1) DC and CFI and 2) DC and CS. Posterior means (SD) of additive covariance and corresponding genetic correlation between the DC and CFI were -0.62 d (0.19 d) and -0.66 (0.12), respectively. The corresponding point estimates between the DC and CS were -0.70 d (0.14 d) and -0.73 (0.06), respectively. These genetic correlations indicate a strong, negative relationship between DC and both measures of fertility in AI data. Selecting for animals with shorter DC intervals genetically will lead to correlated increases in both CS and CFI. Posterior means (SD) for additive and residual variance and heritability for DC for the DC-CFI analysis were 23.5 d2 (4.1 d2), 363.2 d2 (4.8 d2), and 0.06 (0.01), respectively. The corresponding parameter estimates for the DC-CS analysis were very similar. Posterior means (SD) for additive, herd-year and service sire variance and heritability for CFI were 0.04 (0.01), 0.06 (0.06), 0.14 (0.16), and 0.03 (0.01), respectively. Posterior means (SD) for additive, herd-year, and service sire variance and heritability for CS were 0.04 (0.01), 0.07 (0.07), 0.14 (0.16), and 0.03 (0.01), respectively. The similarity of the parameter estimates for CFI and CS suggest that either trait could be used as a measure of fertility in AI data. However, the definition of CFI allows the identification of animals that not only record a calving event, but calve to their first insemination, and the value of this trait would be even greater in a more complete dataset than that used in this study. The magnitude of the correlations between DC and CS-CFI suggest that it may be possible to use a multitrait approach in the evaluation of AI and natural service data, and to report one genetic value that could be used for selection purposes. 相似文献

1. Guinea fowl hens were inseminated weekly once with two doses of spermatozoa (75 million and 100 million) in two different diluents, Beltsville poultry semen extender (BPSE), and Instruments for Veterinary Medicine (IMV), each with and without pre-insemination vaginal douching. Per cent fertility, hatchability, dead germ, dead in shells along with data on sperm egg interaction and vaginal microbial counts were recorded. 2. Artificial insemination had significantly improved the per cent fertility and hatchability compared to natural mating. Dose dependent improvement in fertility was noticed with both diluents. 3. There was a beneficial effect of vaginal douching, which was more pronounced at lower insemination doses. 4. For optimum fertility and hatchability in guinea fowl, insemination of 75 million spermatozoa diluted in BPSE once in 4 d and 100 million spermatozoa diluted in BPSE or IMV once in 5 d coupled with vaginal douching is recommended. 相似文献

The objective of this study was to examine the effects of increasing circulating progesterone in multiparous Holstein cows, using an intravaginal progesterone implant between days 3 and 7 following fixed-time artificial insemination (FTAI). The animals were randomly assigned into two groups: (1) treatment group and (2) control (no treatment). The experiment took place during periods of high and mild temperatures. Pregnancy rates were evaluated by ultrasound at 30 and 60 days after FTAI; embryonic and fetal mortality rates were also evaluated on these dates. The temperature and humidity index on the day of FTAI were different ( P?<?0.05) during the times of interest, but it did not affect conception rates. Neither the conception rates at days 30 and 60 nor the embryonic and fetal mortality rates differed between the groups during either the high- or mild-temperature periods ( P?>?0.05). Throughout the experimental period, the serum progesterone concentration was higher in the treatment group than in the control ( P?<?0.05). We conclude that the intravaginal progesterone implant inserted from days 3 to 7 following FTAI had no effect on conception rates at days 30 and 60 or on embryonic or fetal mortality rates but enhances the serum progesterone concentrations following treatment in multiparous Holstein cows during high- and mild-temperature periods. 相似文献

The purpose of this study was conducted to evaluate the use of a system of timed artificial insemination (TAI) and early diagnosis
of pregnancy in Nelore beef cows in order to reduce the breeding season (BS) from 150 to 90 days and analyze the factors affecting
the conception rate of cows selected for TAI. The study included 110 Nelore cows belonging to the Experimental Farm Capim
Branco, Brazil. The body condition scores (BCS) were evaluated, and those above 2.50 were included in the TAI protocol. Five
groups of cows were formed at intervals of 21 days postpartum and then commenced a 9-day progesterone protocol. Progesterone
devices were used three times. Statistical analysis of binary variables was performed by logistic regression in the program
Proc Logistic of Sas. A 5% of significance ( P < 0.05) was used to determine the presence of statistical differences and the effect of independent variables on conception
rate. The TAI and early diagnosis of pregnancy reduces the BS from 150 to 90 days without changing the pregnancy rate. The
conception rate was not affected by the presence of the calf ( P > 0.05), the corpus luteum ( P > 0.05), the BCS ( P > 0.05), or the number of reuses of the intravaginal progesterone device ( P > 0.05). Therefore, this technique is beneficial for breeding Nelore beef cattle and can be used in anestrous cows with a
BCS above 2.50. Additionally, the progesterone devices can be reused to reduce the cost of the protocol without compromising

95 cows with large cystic degeneration of the ovaries were treated with luteinizing hormone-follicle stimulating hormone releasing hormone (dosages of .25-1.0 mg). 12 of the cows were treated further with repeated Lutal doses. 70 cows became pregnant, a success rate of 73.6%, which corresponds to the experiences of Bentele and Humke and Christl. The 1st venereal ardor occurred 22.8 days after Lutal injection with a standard deviation of 15.4 days. The unpredictability of the onset is a disadvantage of the therapy and use of prostaglandins should be researched in this context. Otherwise this therapy is practical and effective. No clear correlation was found with respect to effectiveness of the medication and the dosage level. 相似文献

Three experiments were designed to evaluate methods to optimize the use of sex-sorted sperm in timed AI (TAI) programs for suckled beef cows. In all 3 experiments, suckled Bos indicus cows were synchronized using an intravaginal progesterone (P4) device during 8 d and a 2.0-mg injection of intramuscular estradiol benzoate (EB) at device insertion. The females received PG and eCG (300 IU) at P4 device removal and 1.0 mg of EB 24 h later. The cows were inseminated 60 to 64 h after P4 device withdrawal. All cows had their ovaries scanned by transrectal ultrasound at TAI to indentify and to measure the largest follicle (LF) present. In Exp. 1, a total of 853 cows had their LF classified as <9 mm or ≥9 mm at the time of TAI; these cows were then randomly assigned to 4 groups according to their LF diameter (<9 mm or ≥9 mm) and the type of sperm used (sex-sorted or non-sex-sorted). There was an interaction (P = 0.02) between the type of sperm and LF diameter beginning at TAI[non-sex-sorted ≥9 mm = 58.9%a (126/214); non-sex-sorted <9 mm = 49.5%b (106/214);sex-sorted ≥9 mm = 56.8%ab (134/236); and sex-sorted <9 mm = 31.2%c (59/189), a≠b≠c = P < 0.05]. In Exp. 2, suckled cows (n = 491) were classified immediately before TAI as having displayed estrus or not (estrus or no estrus) between P4 device removal and TAI. These cows were randomly assigned to 4 groups according to the occurrence of estrus and the type of sperm (sex-sorted or non-sex-sorted). There were effects of the occurrence of estrus (P = 0.0003) and the type of sperm (P = 0.05) on pregnancy per AI [P/AI; no estrus, non-sex-sorted = 43.6% (27/62); estrus, non-sex-sorted = 58.5%; (107/183); no estrus, sex-sorted = 33.9% (21/62), and estrus, sex-sorted = 50.0% (92/184)]; however, no interaction between the occurrence of estrus and type of sperm was observed (P = 0.87). In Exp. 3, a total of 200 suckled cows presenting LF ≥9 mm at TAI were randomly assigned to receive sex-sorted sperm deposited into the uterine body (n = 100) or into the uterine horn ipsilateral to the recorded LF (n = 100). No effect of deeper AI on P/AI was found (P = 0.57). Therefore, the LF diameter at TAI and the occurrence of estrus can be used as selection criteria to identify cows with greater odds of pregnancy to receive sex-sorted sperm in TAI programs. Thirty nonlactating Holstein cows were superovulated to determine the effect of artificial insemination time on fertilization status and embryo quality. During the luteal phase of the estrous cycle, cows were administered 38 mg FSH-P in a 4-d descending dose regimen. Luteolysis was induced with two injections of prostaglandin on the last day of FSH-P treatment. All cows were continuously monitored for behavioral estrus using the HeatWatch estrus detection system. All cows were inseminated once with one .5-mL straw (50 x 10(6) sperm) at either 0 (n = 10), 12 (n = 10), or 24 h (n = 10) after the first standing event. The elapsed time (mean +/- SD) from the first prostaglandin dose to the first standing event was 39.4 h +/- 7.7 h. The (mean +/- SD) duration of behavioral estrus was 13.2 h +/- 4.1 h. The (mean +/- SD) number of standing events was 27 +/- 17. Five hundred twenty-nine embryos and ova were recovered nonsurgically 6 d after insemination. Fertilization rates were 29 (0 h), 60 (12 h), and 81% (24 h) (P < .01). Percentages of excellent and good, fair and poor, and degenerate embryos were not different (P > .05). Percentages of embryos with accessory sperm were 5 (0 h), 8 (12 h), and 41 (24 h) and differed between the 0 and 24 h and the 12 and 24 h inseminations (P < .01). Artificial insemination of superovulated, nonlactating Holstein cattle 24 h after onset of estrus increased fertilization rate and percentage of embryos with accessory sperm compared with insemination at 0 or 12 h after onset of estrus. The experimental objective was to compare pregnancy rates after fixed-time AI in postpartum suckled beef cows following administration of two progestin-based protocols to synchronize ovulation. Cows (n = 424) at three locations (n = 208, 122, and 92 per location) were stratified by age, BCS, and days postpartum (DPP) and assigned randomly to one of the two treatment protocols. The MGA Select-treated cows (MGA Select; n = 213) were fed melengestrol acetate (MGA, 0.5 mg x cow(-1) x d(-1)) for 14 d and carrier for 8 d, and then GnRH (100 microg i.m. Cystorelin; d 26) was injected 12 d after MGA withdrawal, and PG (25 mg i.m. Lutalyse) was administered 7 d after GnRH. Cows assigned to the 7-11 Synch protocol (7-11 Synch; n = 209) were fed carrier for 15 d and MGA for 7 d, and then injected with PG on d 22 (d 7 of MGA), GnRH on d 26, and PG again on d 33. Artificial insemination was performed at fixed times for cows in both treatments at 60 or 72 h after d 33 PG for 7-11 Synch and MGA Select groups, respectively. All cows were injected with GnRH (100 microg of i.m. Cystorelin) at AI. There was no treatment x location interaction for age (P = 0.90), BCS (P = 0.64), or DPP (P = 0.93), and the results were therefore pooled for the respective treatments (age [7-11 Synch, 5.5 +/- 0.2; MGA Select, 5.5 +/- 0.2], BCS [7-11 Synch, 5.7 +/- 0.1; MGA Select, 5.6 +/- 0.1], and DPP [7-11 Synch, 41.1 +/- 1.1; MGA Select, 42.1 +/- 1.1]). Blood samples were collected 8 and 1 d before MGA or carrier to determine pretreatment estrous cyclicity (progesterone >or=1 ng/mL; 7-11 Synch, 59/209 [28%]; MGA Select, 54/213 [25%]; P = 0.50) and again on d 33 PG to evaluate treatment response as a percentage of cows with progesterone concentrations in serum >or=1ng/mL (7-11 Synch, 184/209 [88%]; MGA Select, 177/213 [83%]; P = 0.15). Pregnancy rates resulting from fixed-time AI did not differ (P = 0.25) between treatments (7-11 Synch, 128/209 [61%]; MGA Select, 142/213 [67%]), nor did pregnancy rates (P = 0.77) at the end of the breeding season (7-11 Synch, 198/208 [95%]; MGA Select, 204/213 [96%]). These data indicate that pregnancy rates were comparable after fixed-time AI, following administration of the 7-11 Synch and MGA Select protocols. Both protocols provide opportunities for beef producers to use AI and eliminate the need to detect estrus. 相似文献

This experiment was designed to compare pregnancy rates in postpartum beef cows resulting from fixed-time AI (FTAI) after treatment with 1 of 2 protocols to synchronize estrus and ovulation. Cross-bred, suckled beef cows (n = 650) at 4 locations (n = 210; n = 158; n = 88; and n = 194) were assigned within a location to 1 of 2 protocols within age group by days postpartum and BCS. Cows assigned to the melengestrol acetate (MGA) Select treatment (MGA Select; n = 327) were fed MGA (0.5 mg x head(-1) x d(-1)) for 14 d, GnRH (100 microg of Cystorelin i.m.) was injected on d 26, and prostaglandin F2alpha (PG; 25 mg of Lutalyse i.m.) was injected on d 33. Cows assigned to the CO-Synch + controlled internal drug release (CIDR) protocol (CO-Synch + CIDR; n = 323) were fed a carrier for 14 d, were injected with GnRH and equipped with an EAZI-BREED CIDR insert (1.38 g of progesterone, Pfizer Animal Health, New York, NY) 12 d after carrier removal, and PG (25 mg of Lutalyse i.m.) was injected and the CIDR were removed on d 33. Fixed-time AI was performed at 72 or 66 h after PG for the MGA Select or CO-Synch + CIDR groups, respectively. All cows were injected with GnRH (100 microg of Cystorelin i.m.) at the time of insemination. Blood samples were collected 8 and 1 d before the beginning of MGA or carrier to determine estrous cyclicity status of the cows (estrous cycling vs. anestrus) before treatment [progesterone > or = 0.5 ng/mL (MGA Select, 185/327, 57%; CO-Synch + CIDR, 177/323, 55%); P = 0.65]. There was no difference (P = 0.20) in pregnancy rate to FTAI between treatments (MGA Select, 201/327, 61%; CO-Synch + CIDR, 214/323, 66%). There was also no difference (P = 0.25) between treatments in final pregnancy rate at the end of the breeding period (MGA Select, 305/327, 93%; CO-Synch + CIDR, 308/323, 95%). These data indicate that pregnancy rates to FTAI were comparable after administration of the MGA Select or CO-Synch + CIDR protocols. Both protocols provide opportunities for beef producers to utilize AI and potentially eliminate the need to detect estrus. 相似文献

The records kept by 4 artificial breeding centres in south western Victoria were examined and analysed by multiple regression analysis to determine whether daily fluctuations in the number of dairy cows presented for insemination, and daily, monthly and annual fluctuations in the result of first service, were associated with either climatic conditions or nutritional factors assessed as the availability of conserved hay and oats as feed supplements in the district. Over a 122 day observation period from July to October 1974, the number of cows presented daily for insemination was negatively correlated with daily rainfall total (p less than 0.01), wind speed (p less than 0.001) and barometric pressure (p less than 0.05). When the mean value for barometric pressure was used in the regression equation, rainfall totals and wind speeds above zero were associated with a mean reduction of 13.8% in the number of cows presented daily for insemination. The daily percentages of cows not resubmitted for service within 60 days of first service (non-return rates) were unrelated to climatic conditions and to the number of cows presented for insemination. The monthly non-return rate had a negative correlation (p less than 0.001) with the cost of oats but was not related to corresponding climatic changes. Annual non-return rate fluctuations were negatively correlated with the cost of oats (p less than 0.001) and with the July to October rainfall total (p less than 0.01). It is suggested that the correlation of non-return rate with the July to October total rainfall was due to an effect of rainfall total on the nutritional value of the pasture. 相似文献
