反相高效液相色谱法测定牛奶中土霉素残留   总被引:4,自引:0,他引:4

建立了测定牛奶中土霉素残留的RP-HPLC方法.采用草酸甲醇液提取牛奶中的土霉素,离心,用C18色谱柱(5 μm,250 mm×4.6 mm),甲醇 乙腈 0.01 mol/L草酸溶液(20∶ 12∶ 68)为流动相,流速1 mL/min,检测波长360 nm,以HPLC外标法测定牛奶中土霉素的残留量.结果:土霉素峰面积与浓度呈良好的线性关系,其线性范围为50~1 500 μg/L(r=0.999 9),平均回收率为88%.本法简便、准确、快速,是一种较好的测定牛奶中土霉素残留的方法.  相似文献

摘要试验选用;35头健康无病、处于泌乳中期、胎次相同、体重相近(体重约450kg)、产奶量相近(P〉0.05)的荷斯坦奶牛,随机分为2组:对照组10头,用产奶期正常饲料饲喂;试验组25头,每头牛在对照组日粮基础上投5kg新鲜红薯渣替代5kg苜蓿干草。结果表明:在产奶期日粮中添加新鲜红薯渣对奶牛的产奶量没有显著影响(P〉0.05);从乳成分试验统计数据看,其对奶牛的乳成分影响不明显,乳脂、乳蛋白、乳糖、乳固形物和非脂固形物均无显著差异(P〉0.05)。  相似文献

对五种土霉素注射液产品(A、B、C、D和得米先20%)进行了猪体内的药代动力学比较研究。五组猪分别按20 mg/kg的剂量耳后肌肉注射,HPLC测定血药浓度,3P97软件拟合血药浓度-时间数据,计算药代动力学参数。五种长效土霉素注射液均符合二室开放模型,峰浓度值分别为1.511、.772、.132、.022、.72μg/mL,AUC分别为48.456、0.22、52.41、66.045、9.8(μg.h)/mL。比较而言,得米先20%药动学最好,其次为产品C,产品B最差。  相似文献

Plasma concentrations of doramectin in 40 cattle dosed by subcutaneous (sc) or intramuscular (i.m.) injection (200 μg/kg) were compared to assess the bioequivalence of the two routes of administration. Peak concentration ( C max), and areas under the concentration curve ( AUC0– ) were determined from plasma concentrations. Animals treated by the sc route showed a mean AUC0– of 457 ± 66 ng±day/mL (± SD) and a mean C max of 27.8 ± 7.9 ng/mL. Results from the i.m. treatment group showed a mean AUC 0– of 475 ± 82 ng-day/mL and a mean C max of 33.1 ± 9.0 ng/mL Absorption constants ( k a) determined by modelling were 0.542 ± 0.336 day-1after sc administration and 0.710 ± 0.357 day-1after i.m. administration. The 90% confidence limits on the difference between mean AUC 0– values for the sc and i.m. groups fell within 20% of the mean value for the subcutaneous group. C max was somewhat greater for the i.m. route. The 90% confidence limits on the difference in mean In ( T max+1) also fell within 20% of the mean sc value. Based on this analysis, bioequivalence of the sc and i.m. formulation has been established.  相似文献

Tulathromycin is a triamilide antibiotic that maintains therapeutic concentrations for an extended period of time. The drug is approved for the treatment of respiratory disease in cattle and swine and is occasionally used in goats. To investigate the pharmacokinetics of tulathromycin in meat goats, 10 healthy Boer goats were administered a single 2.5 mg/kg subcutaneous dose of tulathromycin. Plasma concentrations were measured by ultra-high pressure liquid chromatography tandem mass spectrometry (UPLC–MS/MS) detection. Plasma maximal drug concentration (Cmax) was 633 ± 300 ng/ml (0.40 ± 0.26 h post-subcutaneous injection). The half-life of tulathromycin in goats was 110 ± 19.9 h. Tulathromycin was rapidly absorbed and distributed widely after subcutaneous injection 33 ± 6 L/kg. The mean AUC of the group was 12,500 ± 2020 h ng/mL for plasma. In this study, it was determined that the pharmacokinetics of tulathromycin after a single 2.5 mg/kg SC injection in goats were very similar to what has been previously reported in cattle.  相似文献

The aim of this work was to determine the pharmacokinetics of intravenous (i.v.) and intramuscular (i.m.) ceftazidime administered to lactating (LTG; n=6) and non-lactating (NLTG; n=6) healthy Creole goats in 2 trials (T1 and T2). During T1 and T2, goats randomly received a single dose of i.m. or i.v. ceftazidime (10 mg/kg). Serum concentration of iv ceftazidime in NLTG and LTG goats is best described by 2 and 3 compartment models, respectively. The pharmacokinetic parameters of iv and im ceftazidime administered to LTG and NLTG showed statistically significant differences (P < 0.05) in the constants (lamda(z), T1 vs. T2 [i.v.] 0.5 +/- 0.1 vs. 0.3 +/- 0.1/h; T1 vs. T2 [i.m.] 0.5 +/- 0.2 vs. 0.3 +/- 0.1/h) and in the mean times (t(1/2), T1 vs. T2 [i.v.] 1.6 +/- 0.3 vs. 2.3 +/- 0.6 h; T1 vs. T2 [i.m.] 1.6 +/- 0.7 vs. 2.6 +/- 0.9 h) of elimination. The bioavailability of ceftazidime in LTG and NLTG was 113.0 +/- 17.8 and 96.0 +/- 18.0%, respectively. Ceftazidime concentration in milk at 2 h was: i.v. = 1.9 +/- 0.2 and i.m. = 2.4 +/- 0.5 microg/ml; the penetration in milk was i.v. = 18.3 +/- 13.5 and im = 14.3 +/- 10.6%. Ninety-six hours after i.v. and i.m. administration, residues of the drug were not found in milk. In conclusion, ceftazidime, when administered to goats, showed high concentration times in serum, good penetration into milk and a bioavailability that makes it suitable to be used by the i.m. route.  相似文献

The pharmacokinetics of butorphanol tartrate were investigated following intravenous administration of 0.25 mg/kg of body weight to six healthy non-lactating Jersey cows. Three lactating Holstein cows also received 0.045 mg of butorphanol/kg of body weight intravenously to determine the extent and duration of drug transfer into milk. A radioimmunoassay technique was used to measure butorphanol concentrations in plasma and milk. The disposition of butorphanol following intravenous administration was characterized by rapid and extensive distribution followed by a slower elimination phase. Apparent volume of distribution was 4.178 ± 1.145 (mean ± SD) I/kg, mean elimination half-life was 82 min, and clearance was 34.6 ± 7.7 ml/min/kg. Trace quantities of butorphanol were detected in the cow's milk for up to 36 h following administration. These pharmacokinetic data were compared with pharmaco-kinetic and pharmacodynamic data for butorphanol in other species and for three other potent opioids in related ruminant species.  相似文献

建立了一种测定鸡蛋及牛奶中四环素类药物残留 液相色谱-串联质谱的方法。样品经Mcllvaine-Na2EDTA缓冲液提取,经快速滤纸过滤,并经加脱脂棉的HLB固相萃取柱净化,洗脱液氮气吹至小于1 mL,加30%甲醇至1mL并混匀,液相色谱-串联质谱法检测,基质匹配标液外标法定量。结果表明,四环素类药物在5 μg/kg~100 μg/kg的浓度范围内呈现良好的线性关系。在鸡蛋及牛奶中四环素类药物的检测限均为2 μg/kg,定量限均为5 μg/kg。在LOQ~2MRL(多西环素为LOQ-10LOQ)添加浓度范围内,四环素类药物的回收率均在81.2~110.0%之间,批内批间变异系数均小于11.4%。该方法各项技术指标均能满足残留检测要求,且方法的重现性良好,满足国内外兽药残留检测的相关规定。  相似文献

本试验研究单剂量静脉注射、肌肉注射和乳房灌注恩诺沙星(2.5mg/kg)在健康奶山羊的药动学及乳中药物浓度。采用HPLC法测定血浆和乳中恩诺沙星及其代谢产物环丙沙星的浓度,用统计矩原理处理血浆中药物浓度-时间数据,计算非房室模型的药动学参数。静脉注射、肌肉注射和乳房灌注恩诺沙星的t1/2β分别为1.32、1.55、0.99h;AUC为1.06、3.04、2.66mghL^-1;恩诺沙星的代谢分数为35.01%、44.06%、45.73%;环丙沙星的t1/2β为1.81、2.94、2.32h。乳中的药物浓度高于同期血中药物浓度,且乳中环丙沙星浓度高于恩诺沙星浓度并维持更长的时间。  相似文献

建立了链霉素在牛奶中残留的微生物学检测方法.样品用10%(W/V)三氯乙酸提取,通过冷冻离心使牛奶中残留的链霉素得到分离,再用枯草芽孢杆菌作为检定菌进行含量测定.结果显示,链霉素在0.10~3. 20 μg/mL的浓度范围内具有良好线性关系(r=0.998 1),牛奶中3种添加浓度0.20、0.80、1.60 μg/g的平均回收率为97.6%(n=9),RSD分别为1.6 %、3.7 %、1.2 %(n=3).  相似文献

建立了牛奶中苯并咪唑类药物及代谢物残留的UPLC-MS/MS检测方法。牛奶样品在弱碱性条件下经乙腈沉淀蛋白后,用乙酸乙酯提取,再经正己烷脱脂,MCX固相萃取柱净化,采用UPLC-MS/MS多反应监测(MRM)模式检测,外标法定量。本方法对苯并咪唑类药物及代谢物的检测限均为5μg/g,定量限均为10μg/g,在10~200μg/L添加浓度范围内回收率为69.9%~113.1%之间,批内变异系数在1.36%~11.48%之间,批间变异系数在1.09%~9.87%之间。本方法分析速度快,灵敏度高,重现性好,各项技术指标均满足国内外相关法规要求,可用于牛奶中苯并咪唑类药物及代谢物残留的检测。  相似文献

试验建立了反相高效液相色谱(RT-HPLC)法测定盐酸多西环素的浓度,探讨了盐酸多西环素在猪体内的药物动力学和残留特征。结果表明,盐酸多西环素以2.5mg/kg单剂量肌内注射给猪(n=6),药物动力学模型符合有吸收一室模型,药物动力学参数:吸收半衰期(t1/2ka)、消除半衰期(t1/2ke)为(0.400±0.312)h、(9.530±0.956)h,药时曲线下面积(AUC)为(44.414±4.123)mg·h·L-1,最大血药浓度(Cmax)为(2.811±0.136)mg/L,达峰时间(Tp)为(1.910±0.213)h。另外,以相同剂量肌内注射给猪(n=6),每天1次,连续给药4d后,在不同时间测定盐酸多西环素在猪的肌肉、肝脏、肾脏、皮肤和脂肪中的残留量。在给药后16d,盐酸多西环素在各组织均能检测到,且残留均低于残留限量。盐酸多西环素注射液在猪体内消除缓慢,残留期较长,建议休药期不低于16d。  相似文献

AIMS: To determine the plasma disposition of meloxicam in goats following S/C, oral or I/V administration at a single dose of 0.5?mg/kg bodyweight.

METHODS: Five healthy Saanen goats, aged 12–14 months and weighing 35–40?kg, were used for a three phase cross-over design with a 10-day washout period, with meloxicam administered I/V, then orally and S/C. Heparinised blood samples (5?mL) were collected from all animals prior to drug administration (0 hours) and subsequently up to 96 hours. Concentrations of meloxicam in plasma were measured using high performance liquid chromatography. Concentration-time curves were fitted and pharmacokinetic parameters were estimated for each administration group.

RESULTS: Subcutaneous administration of meloxicam exhibited unique plasma distribution characteristics that differed from oral and I/V administration. Mean peak plasma concentrations were greater (1.91 (SD 0.39) vs. 0.71 (SD 0.17) µg/mL) and the time to reach them shorter (3.20 (SD 1.64) vs. 14.33 (SD 2.19) hours) following S/C compared with oral administration (p<0.05). The terminal half-life was longer (15.16 (SD 4.74) vs. 10.69 (SD 1.49) hours) and the MRT was shorter (15.67 (SD 2.37) vs. 24.33 (SD 3.12) hours) following S/C than oral administration (p<0.05), but bioavailability was similar (98.24 (SD 9.62) vs. 96.49 (SD 10.71)%).

CONCLUSION AND CLINICAL RELEVANCE: Subcutaneous administration of meloxicam resulted in long-term presence of drug at high concentration in goat plasma. This unique plasma disposition characteristic may offer an advantage in some clinical cases towards potentially improving the treatment efficacy in goats.  相似文献

Background

Toxoplasma gondii is a major problem for the sheep industry as it may cause reproduction problems. The importance of T. gondii in Norwegian goat herds is uncertain, but outbreaks of toxoplasmosis in dairy goat farms have been recorded. The aim of this study was to describe the prevalence of T. gondii infection in Norwegian dairy goats by using serology.

Findings

Goat serum originally collected as part of two nationwide surveillance and control programmes between 2002 and 2008 were examined for T. gondii antibodies by using direct agglutination test. In total, 55 of 73 herds (75%) had one or more serologically positive animals, while 377 of 2188 (17%) of the individual samples tested positive for T. gondii antibodies.

Conclusions

This is the first prevalence study of T. gondii infection in Norwegian goats. The results show that Norwegian goat herds are commonly exposed to T. gondii. Nevertheless, the majority of goat herds have a low prevalence of antibody positive animals, which make them vulnerable to infections with T. gondii during the gestation period.  相似文献

Closantel (CLS) is currently used in programs for the strategic control of gastrointestinal nematodes. CLS is extralabel used in different dairy goat production systems. From available data in dairy cows, it can be concluded that residues of CLS persist in milk. The current work evaluated the concentration profiles of CLS in plasma and milk from lactating orally treated dairy goats to assess the residues pattern in dairy products such as cheese and ricotta. Six (6) female Saanen dairy goats were treated orally with CLS administered at 10 mg/kg. Blood and milk samples were collected between 0 and 36 days post‐treatment. The whole milk production was collected at 1, 4, 7, and 10 days post‐treatment to produce soft cheese and ricotta. CLS concentrations in plasma, milk, cheese, whey, and ricotta were determined by HPLC. The concentrations of CLS measured in plasma were higher than those measured in milk at all sampling times. However, the calculated withdrawal time for CLS in milk was between 39 and 43 days postadministration to dairy goats. CLS residual concentrations in cheese (between 0.93 and 1.8 μg/g) were higher than those measured in the milk used for its production. CLS concentrations in ricotta were sixfold higher than those in the milk and 20‐fold higher than those in the whey used for its production. The persistent and high residual concentrations of CLS in the milk and in the cheese and ricotta should be seriously considered before issuing any recommendation on the extralabel use of CLS in dairy goat farms.  相似文献

This study was conducted to determine the passage ratio of amoxicillin into milk and its pharmacokinetics in milk and plasma after intramuscular administration. Five healthy dairy cows (Holstein, weighing 450–500 kg, aged 2–4 years) were used in this study. They received single intramuscular amoxicillin at a dose of 14 mg/kg body weight. Blood and milk samples were collected prior to drug administration (0); after 15, 30, 45, 60, and 90 min; and 2, 3, 4, 6, 8, 10, and 12 hr after administration. The plasma and milk concentrations of amoxicillin were determined using high‐performance liquid chromatography with ultraviolet detection. The passage ratio of amoxicillin into milk and plasma was determined using both AUC‐based calculation and milk and plasma concentrations at sampling times; it was calculated 0.46 and 0.52, respectively. The terminal half‐life and mean residence time of amoxicillin were 6.05 and 8.60 hr in plasma and 2.62 and 5.35 hr in milk, respectively. The Cmax2 levels of amoxicillin in plasma and milk were measured as 1,096 and 457 ng/ml, respectively. It was observed that amoxicillin exhibited a secondary peak in plasma and milk. This study was the first to report on the passage ratio of amoxicillin into milk in lactating cows.  相似文献

Buprenorphine is absorbed following sublingual administration, which would be a low‐stress delivery route in foals. However, the pharmacokinetics/pharmacodynamics are not described in foals. Six healthy foals <21 days of age participated in a blinded, randomized, 3‐period, 5‐sequence, 3‐treatment crossover prospective study. Foals received 0.01–0.02 mg/kg buprenorphine administered SL or IV with an equivalent volume of saline administered by the opposite route. Blood was collected from the cephalic vein for pharmacokinetic analysis. Physiologic parameters (HR, RR, body temperature, GI sounds), locomotion (pedometer), and behavioral data (activity level, nursing time, response to humans) were recorded. Plasma concentration of buprenorphine exceeded a presumed analgesic level (0.6 ng/ml) in five foals in the IV group and one in the SL group but only for a very brief time. Pharmacokinetic analysis following IV administration demonstrated a short elimination half‐life (t1/2β 1.95 ± 0.7 hr), large volume of distribution (6.46 ± 1.54 L/kg), and a high total clearance (55.83 ± 23.75 ml/kg/min), which differs from adult horses. Following SL administration, maximum concentrations reached were 0.61 ± 0.11 ng/ml and bioavailability was 25.1% ± 10.9%. In both groups, there were minor statistical differences in HR, RR, body temperature, locomotion, and time spent nursing. However, these differences were clinically insignificant in this single dose study, and excitement, sedation, or colic did not occur.  相似文献
