It is well known that after a stressor, levels of plasma cortisol rise, inducing physiological changes within the animal that are directed toward maintaining homeostasis. Less well understood is the role of cortisol in regulating food intake in teleosts. This study investigated the effect of cortisol on food intake and regulation of the neuroendocrine appetite-stimulating hormones, neuropeptide Y (NPY) and ghrelin, in tilapia (Oreochromis mossambicus). Male and female tilapia were randomly assigned to one of the following treatments: unhandled control, vehicle-injected control, or cortisol (2 μg/g BW). Food intake was determined 24 h after injection during a 1-h feeding trial. Cortisol reduced food intake (P < 0.001). An identical study was conducted to measure the effects of 24-h cortisol treatment on the endocrine regulators of food intake. Cortisol reduced stomach expression of ghrelin mRNA (P < 0.05) and plasma concentrations of ghrelin (P < 0.05). In the hypothalamus/optic tectum cortisol reduced levels of GHSR1a-LR (biologically active ghrelin receptor) mRNA. In the telencephalon/preoptic area cortisol significantly reduced levels of NPY and GHSR1b-LR (biologically inactive ghrelin receptor) mRNA. These findings suggest that anorexigenic actions of cortisol may be mediated via two separate pathways: (1) reducing circulating ghrelin levels as well as GHSR1a-LR expression in the hypothalamus/optic tectum and/or (2) suppressing NPY expression in the telencephalon/preoptic area.  相似文献

OBJECTIVE: To investigate the physiologic endocrine effects of food intake and food withholding via measurement of the circulating concentrations of acylated ghrelin, growth hormone (GH), insulin-like growth factor-I (IGF-I), glucose, and insulin when food was administered at the usual time, after 1 day's withholding, after 3 days' withholding and after refeeding the next day in healthy Beagles. ANIMALS: 9 healthy Beagles. PROCEDURES: Blood samples were collected from 8:30 AM to 5 PM from Beagles when food was administered as usual at 10 AM, after 1 day's withholding, after 3 days' withholding, and after refeeding at 10 AM the next day. RESULTS: Overall mean plasma ghrelin concentrations were significantly lower when food was administered than after food withholding. Overall mean plasma GH and IGF-I concentrations did not differ significantly among the 4 periods. Circulating overall mean glucose and insulin concentrations were significantly higher after refeeding, compared with the 3 other periods. CONCLUSIONS AND CLINICAL RELEVANCE: In dogs, food withholding and food intake were associated with higher and lower circulating ghrelin concentrations, respectively, suggesting that, in dogs, ghrelin participates in the control of feeding behavior and energy homeostasis. Changes in plasma ghrelin concentrations were not associated with similar changes in plasma GH concentrations, whereas insulin and glucose concentrations appeared to change reciprocally with the ghrelin concentrations.  相似文献

Hyperglycemia was produced in female Brown Leghorn fowls by oral or intravenous injection of glucose solutions, at high and low levels of feeding motivation. A significant effect on food intake during a subsequent 15‐min test period was found only for the oral/low‐motivation condition. This may have been due to the osmotic effect of the solution on the gut rather than a true lowering of feeding motivation. It is unlikely that feeding motivation is directly related to blood glucose levels.  相似文献

  1. The aim of the current study was to investigate the interaction of the nitric oxide and cannabinoidergic systems on feeding behaviour in neonatal chicken.

  2. A total of 6 experiments were designed to evaluate the interaction between cannabinoidergic and nitrergic systems on food intake in 3-h food-deprived (FD3) neonatal chickens. In Experiment 1, chickens received intracerebroventricular (ICV) injections of saline, 2-arachidonoylglycerol (2-AG) (a CB1 receptor agonist, 2 µg), l-arginine (nitric oxide precursor, 200 nmol) and co-administration of 2-AG + l-arginine. In Experiment 2, ICV injection of saline, 2-AG (2 µg), l-NAME (a nitric oxide synthesis inhibitor, 100 nmol) and their combination (2-AG + l-NAME) were applied to the birds. In Experiment 3, injections were saline, CB65 (a CB2 receptor agonist, 1.25 µg), l-arginine (200 nmol) and CB65 + l-arginine. In Experiment 4, birds received ICV injection of saline, CB65 (1.25 µg), l-NAME (100 nmol) and CB65 + l-NAME. In Experiment 5, chickens were ICV injected with saline, l-arginine (800 nmol), SR141716A (a selective CB1 receptor antagonist, 6.25 µg) and l-arginine + SR141716A. In Experiment 6, birds were injected with saline, l-arginine (800 nmol), AM630 (a selective CB2 receptor antagonist, 5 µg) and l-arginine + AM630. Cumulative food intake was recorded until 2-h post injection.

  3. ICV injection of CB1 and CB2 receptor agonists increased food intake. Co-injection of 2-AG + l-NAME increased the hyperphagic effects of CB1 receptors. CB2 receptor-induced food intake was not affected by co-administration of CB65 + l-NAME. l-Arginine decreased food intake and this effect was amplified by co-injection of l-arginine + SR141716A. However; CB2 receptor antagonists had no effect on l-arginine-induced hypophagia.

  4. The results suggest that there is an interaction between endogenous nitric oxide and the cannabinoidergic system on feeding behaviour which is mediated via CB1 receptors in the neonatal chicken.

These studies were conducted to determine whether ghrelin, a 28-amino acid peptide produced mainly by the stomach, was involved in tryptophan-mediated appetite stimulation in swine. In experiment 1, 36 crossbred (Long WhitexLarge White) barrows were used in a 2x3 factorial design to determine the effects of food intake (ad libitum versus limit fed) and tryptophan level (0.12%, 0.19% and 0.26%) on growth performance as well as ghrelin expression, plasma insulin, ghrelin and leptin levels. Ad libitum fed pigs gained more weight, but had poorer feed conversion than limit fed pigs. Weight gain, food intake and feed conversion all improved with increased ingestion of dietary tryptophan. Ad libitum feeding increased plasma insulin. Plasma insulin was unaffected by the level of dietary tryptophan. However, plasma leptin was significantly lower in pigs fed 0.19% tryptophan compared to those fed 0.12% tryptophan. Plasma ghrelin levels and ghrelin mRNA level in gastric fundus and duodenun was significantly higher in pigs fed 0.19% and 0.26% tryptophan diet compared with pigs fed 0.12%. In the second experiment, 18 crossbred barrows were divided into three treatments involving oral infusion of saline, tryptophan (40mg/kg BW) or 5-hydroxytryptophan (40mg/kg BW). Plasma ghrelin levels at 20, 40 and 60min after infusion of tryptophan were higher than after saline and 5-hydroxytryptophan infusion, 5-hydroxytryptophan infusion induced lower food intake than saline infusion, and tryptophan infusion increased food intake 2, 8 and 24h after infusion. In conclusion, oral tryptophan ingestion increased ghrelin expression in gastric fundus and plasma ghrelin level.  相似文献

1. Male broiler chickens were exposed in separate experiments to: (a) constant ambient temperatures (Ta) ranging between 10 and 35°C; (b) 12 h: 12 h diurnal high‐low temperature of 10:30°C, 15:35°G and 15:30°C in different trials; and (c) 12 h of 30 or 35°G and 12 h of variable lower temperature.

2. A progressive decline in weight gain and food intake was obtained as Ta increased from 18 to 35°C. Under diurnally cycling temperature, weight gain and food intake were lower than in the average corresponding constant temperature, with the exception of chickens exposed to 15:30°C, where weight gain and food intake were not significandy different from those of the constant average temperature.

3. As the diurnal cold period was made colder, chickens exposed during half of the diurnal cycle to high temperature (30, 35°C) demonstrated a significant (P ≤ 0.05) increase in weight gain and food intake.

4. There was a good correlation between plasma triiodothyronine (T3) and Ta. The correlation between T3 and food intake was highly significant, but that between T3 and weight gain was poor.

5. The results indicate the difficulties in predicting the performance of broilers exposed to diurnally cycling temperatures from knowledge of the average temperature.  相似文献

Respiratory quotients (RQ) smaller than 0.70 must be associated with partial oxidation of the biological substrates and storage of oxygen in the carcass or the excreta. An equation for the calculation of the amount of oxygen stored in relation to the RQ is described. The fluxes of N, C, H and O in the carcass of the fasting fowl were negative and compatible with the complete oxidation of the biological substrates. The amounts of these elements in the excreta corresponded with the nitrogenous excretion products, carbohydrates and minerals. No substantial oxygen storage could be demonstrated. The very low RQ values reported in the past would require a magnitude of oxygen storage which is incompatible with life.  相似文献

The acute effect of leptin on the regulation of food intake was investigated in layer and broiler chickens. In an initial study, we observed that a single intraperitoneal injection of recombinant chicken leptin (1 mg/kg BW) dramatically reduced (38%) food intake in 56-day-old layer chickens, more moderately reduced (15%) food intake in 9-day-old layer chicks, and had no significant effect in 9-day-old broiler chicks. In a subsequent study, body weight and plasma concentrations of leptin were measured weekly in layer and broiler chicks from day 1 to 35 of age and brain leptin receptor and neuropeptide Y (NPY) mRNA expression were analyzed at 1, 9, and 35 days of age. At day 1 of age, peripheral concentrations of leptin were significantly greater in layer than broiler chicks. Subsequently, despite increases in body weight and differences in growth rates between layer and broiler chicks from day 8 to day 35 of age, peripheral concentrations of leptin were constant and similar in both genotypes. Leptin receptor and NPY mRNA were expressed in brain from day 1 in chicks of both genotypes and increased significantly to day 35 of age. These observations provide evidence that the inhibitory effect of leptin on the regulation of food intake in growing chicks is an age dependent process. Furthermore, acquisition of the anorectic effect of leptin is likely to be associated with greater expression of the leptin receptor and NPY mRNAs than to changes in blood levels of leptin. Finally, this study provides evidence that chickens selected for high growth rates may be less sensitive or responsive to peripheral concentrations of leptin than chickens with low growth rates (layers), suggesting that the faster growth of broiler chicks may be related to a lessened responsiveness to anorexigenic factors.  相似文献

Circadian variations in heat production (HP) rate and respiratory quotient (RQ) were measured in growing broilers maintained at 5 ambient temperatures (14 degrees, 17 degrees, 22 degrees, 27 degrees and 32 degrees C) and at 5 rates of feeding [ad libitum intake and 75%, 50%, 25% and 0% (fasting) of ad libitum intake]. 2. In most cases, the HP rate decreased from 10.30 h just after food was given) until 00.30 h (the 1-h dark period), showed an overshoot just after the 1-h dark period and then changed little. 3. Circadian variation in RQ, except in the fasted group, showed a similar pattern, which consisted of increase, decrease and constant phases. 4. Food intake affected the pattern of circadian variation in RQ, although ambient temperature had little effect. Possible effects of food intake on the pattern of circadian variation in HP rate were discussed.  相似文献

Selection for increased leanness and improved food efficiency in pigs has resulted in a decreased voluntary food intake. It has been argued that voluntary food intake during lactation should be considered in sow breeding programmes. The aim of the present study was to investigate the phenotypic correlation of food intake during growth and food intake at maturity with maximum lactation food intake in a mouse model. A total of 179 records were available on female mice selected for litter size at birth (S-line) and non-selected control females (C-line) from 3 weeks of age to 21 days in lactation. Half of the litters were standardized to eight pups per litter (s) and half were not standardized (ns). Growth intake was measured as the average intake between 21 and 42 days of age, mature intake was measured from a linear regression of food intake against age between 42 and 69 days of age, and lactation intake was measured as a linear regression of food intake against days in lactation between 5 and 14 days in lactation. In both lines, females with a higher growth intake also had a higher mature intake (r = 0.63 to 0.75, P < 0.0001). Lactation intake was related with growth intake and mature intake in Sns females (r = 0.50 and 0.46, P < 0.01) and with growth intake in Ss females (r = 0.32, P < 0.05). In both lines, lactation intake was highly correlated with litter weight (r = 0.48 to 0.94, P < 0.001). Sns and Ss females with higher growth intake supported larger litter weights during lactation (r = 0.32 and 0.30, respectively, P < 0.05) and Sns females with higher mature intakes tended to support larger litter weights (r = 0.28, P = 0.060). It is suggested that lactating C-line females eat to support a given litter size, while S-line females support the maximum litter weight that is allowed for by their intake capacity, and still produce at high levels when litters are standardized. Since lactating sows mobilise body reserves, the relationship of food intake during growth with that during lactation may be reflected in the relationship between growth intake and body condition.  相似文献

  1. The aim of the current study was to investigate the interaction of the lipopolysaccharide (LPS) and histaminergic systems on appetite regulation in broilers. Effects of intracerebroventricular (ICV) injection of α-fluoromethylhistidine (α-FMH, histidine decarboxylase inhibitor), chlorpheniramine (histamine H1 receptor antagonist), famotidine (histamine H2 receptor antagonist) and thioperamide (histamine H3 receptor antagonist) on LPS-induced hypophagia in broilers were studied.

  2. A total of 128 broilers were randomly allocated into 4 experiments (4 groups and 8 replications in each experiment). A cannula was surgically implanted into the lateral ventricle. In Experiment 1, broilers were ICV injected with LPS (20 ng) prior to α-FMH (250 nmol). In Experiment 2, chickens were ICV injected with LPS followed by chlorpheniramine (300 nmol). In Experiment 3, broilers were ICV injected with famotidine (82 nmol) after LPS (20 ng). In Experiment 4, ICV injection of LPS was followed by thioperamide (300 nmol). Then, cumulative food intake was recorded until 4 h post-injection.

  3. According to the results, LPS significantly decreased food intake. Chlorpheniramine significantly amplified food intake, and LPS-induced hypophagia was lessened by injection of chlorpheniramine. α-FMH, famotidine and thioperamide had no effect on LPS-induced hypophagia.

  4. These results suggest that there is an interaction between central LPS and the histaminergic system where LPS-induced hypophagia is mediated by H1 histamine receptors in 3 h food-deprived broilers.

1. This study was conducted to examine whether oral administration of lysine solution affect food intake and the ventromedial hypothalamic (VMH) monoamines in chickens fed on a lysine-free diet.

2. Chickens were assigned to four treatment groups. Two groups of chickens were given two different doses of lysine solution (0·1?g and 0·07?g in 1?ml of saline) exogenously (orally) while being fed on a lysine-free diet, and these results were compared with a control diet plus saline group. Another group of chickens was fed on a lysine-free diet without lysine supplementation, and their results were compared with the lysine treated groups. The extracellular dopamine (DA), norepinephrine (NE) and serotonin (5-HT) in the VMH of freely moving chicken were measured by in vivo microdialysis.

3. There was no significant difference in food intake between the control diet and the lysine supplemented groups during the time-course of the experiments. Food intake significantly decreased at 4, 5 and 6?h in the lysine-free diet plus saline group compared with the lysine supplemented groups. Of the VMH monoamines, the DA concentration remained close to the baseline in the lysine supplemented groups. This DA concentration was significantly lower than the baseline in the lysine-free diet plus saline group at 3·5?h and thereafter.

4. No significant difference from the baseline was observed for NE in the lysine-free diet plus saline group. The 5-HT concentrations were close to the baseline for all groups throughout the experiments.

5. The findings suggest that oral administration of lysine solution to chickens fed on a lysine-free diet restored food intake which was associated with the variations of VMH DA concentration.  相似文献

To evaluate the effect of corticosterone on nutrient transport, 10-week-old male chickens were grouped in 4 categories and treated as follows: sham-operated, adrenalectomised, corticosterone (4 mg/kg injected subcutaneously for 5 d) in both sham-operated and adrenalectomised. The food intake, food passage time and uptake of calcium, phosphorus and glucose were determined by standard procedures. Corticosterone administration to both sham-operated and adrenalectomised groups stimulated significantly higher food intake, delayed food passage time and increased uptake of calcium, phosphorus and glucose, as compared to sham-operated control and adrenalectomised groups. Corticosterone administration increased absorption of these nutrients significantly more in the adrenalectomised group than in the sham-operated controls. Corticosterone also significantly elevated the plasma concentrations of these nutrients. The responses to the hormone were significantly greater in adrenalectomised birds. It is concluded that corticosterone increases food intake and retention and the absorption of calcium, phosphorus and glucose in the alimentary tract.  相似文献

The aim of these in vitro experiments was (1) to examine the effects of ghrelin on the basic functions of ovarian cells (proliferation, apoptosis, secretory activity); (2) to determine the possible involvement of the GHS-R1a receptor and PKA- and MAPK-dependent post-receptor intracellular signalling cascades; (3) to identify the active part of the 28-amino acid molecule responsible for the effects of ghrelin on ovarian cells. We compared the effect of full-length ghrelin 1-28, a synthetic activator of GHS-R1a, GHRP6, and ghrelin molecular fragments 1-18 and 1-5 on cultured chicken ovarian cells. Indices of cell apoptosis (expression of the apoptotic peptide bax and the anti-apoptotic peptide bcl-2), proliferation (expression of proliferation-associated peptide PCNA), and expression of protein kinases (PKA and MAPK) within ovarian granulosa cells were analysed by immunocytochemistry. The secretion of progesterone (P(4)), testosterone (T), estradiol (E(2)) and arginine-vasotocin (AVT) by isolated ovarian follicular fragments was evaluated by RIA/EIA. It was observed that accumulation of bax was increased by ghrelin 1-28, GHRP6 and ghrelin 1-18, but not by ghrelin 1-5. Expression of bcl-2 was suppressed by addition of ghrelin 1-28, GHRP6 and ghrelin 1-5, but promoted by ghrelin 1-18. The occurrence of PCNA was reduced by ghrelin 1-28, GHRP6, ghrelin 1-18 and ghrelin 1-5. An increase in the expression of MAPK/ERK1, 2 was observed after addition of ghrelin 1-28, GHRP6 and ghrelin 1-18, but not ghrelin 1-5. The accumulation of PKA decreased after treatment with ghrelin 1-28 and increased after treatment with GHRP6 and ghrelin 1-18 but not ghrelin 1-5. Secretion of P(4) by ovarian follicular fragments was decreased after addition of ghrelin 1-28 or ghrelin 1-5 but stimulated by GHRP6 and ghrelin 1-18. Testosterone secretion was inhibited by ghrelins 1-28 and 1-18, but not by GHRP6 or ghrelin 1-5. Estradiol secretion was reduced after treatment with ghrelin 1-28 but stimulated by ghrelins 1-18 and 1-5; GHRP6 had no effect. AVT secretion was stimulated by ghrelin 1-28, GHRP6 and ghrelin 1-18, but inhibited by ghrelin 1-5. The comparison of the effects of the four ghrelin analogues on nine parameters of ovarian cells suggest (1) a direct effect of ghrelin on basic ovarian functions-apoptosis, proliferation, steroid and peptide hormone secretion; (2) that the majority of these effects can be mediated through GHS-R1a receptors; (3) an effect of ghrelin on MAPK- and PKA-dependent intracellular mechanisms, which can potentially mediate the action of ghrelin at the post-receptor level; (4) that ghrelin residues 5-18 may be responsible for the major effects of ghrelin on the avian ovary.  相似文献

Glucagon‐related peptides, such as glucagon‐like peptide (GLP)‐1, GLP‐2 and oxyntomodulin (OXM), are processed from an identical precursor proglucagon. In mammals, all of these peptides are suggested to be involved in the central regulation of food intake. We previously showed that intracerebroventricular administration of chicken OXM and GLP‐1 significantly suppressed food intake in chicks. Here, we show that central administration of chicken GLP‐2 potently suppresses food intake in chicks. Male 8‐day‐old chicks (Gallus gallus domesticus) were used in all experiments. Intracerebroventricular administration of chicken GLP‐2 significantly suppressed food intake in chicks. Plasma glucose concentration was significantly decreased by chicken GLP‐2, whereas plasma nonesterified fatty acid concentration was significantly increased. Intracerebroventricular administration of chicken GLP‐2 did not affect plasma corticosterone concentration. In addition, the anorexigenic effect of GLP‐2 was not reversed by the corticotropin‐releasing factor (CRF) receptor antagonist α‐helical CRF, suggesting that CRF is not a downstream mediator of the anorexigenic pathway of GLP‐2 in chicks. Intracerebroventricular administration of an equimolar amount of GLP‐1 and GLP‐2, but not OXM, significantly suppressed food intake in both broiler and layer chicks. All our findings suggest that GLP‐2 functions as a potent anorexigenic peptide in the brain, as well as GLP‐1, in chicks.  相似文献
