An experiment was performed to study some serum enzyme changes taking place in artificially fed lambs made dystrophic on a skim milk ration supplemented with α-tocopherolextracted cod liver oil. In the course of two to three weeks the lambs showed highly increased serum values of aspartate- and alanine aminotransferase (AspAT = GOT and A1AT = GPT), total lactate dehydrogenase (LDH), and creatine phosphokinase (GPK). The latter enzyme proved to be the most sensitive test of myopathies, with serum values above 1000 times the normal level. A high-grade hyaline skeletal muscle degeneration was confirmed by histology. Myocardial changes were also present.Contrary to expectations, electrophoretic separation of LDH isoenzymes in the serum of the dystrophic lambs did not reflect any increase of the prevalent muscular cathodic thermolabile fraction LDH5. The use of a so-called LDH heat stability test as an aid in clinical diagnostic work, as previously suggested, therefore does not seem to be valid in this disease. Trace elements in liver were examined in vitamin B12 deficient lambs which developed ovine white-liver disease (OWLD), in cobalt/vitamin B12 supplemented lambs on the same pastures as well as clinically healthy, but sometimes subclinical B12 deficient lambs on other pastures (H). Liver Co was marginal to deficient in both OWLD lambs (S lambs) and H lambs. Supplementation with B12 or Co elevated liver Co. Liver copper was significantly lower in OWLD lambs than in the H lambs, and Co/B12 supplementation on pasture generally had no significant effect on the contents. Dosing lambs on OWLD pastures with copper oxide needles (SCuO), however, resulted in high/toxic liver Cu. Dosing with Co, Se and Cu glass boluses resulted in adequate liver Cu, except for 1 lamb with toxic amounts indicating dissolution and absorption of the bolus. OWLD lambs had significantly lower liver molybdenum than H lambs, and Co/B12 supplementation elevated values, while CuO treatment depressed them. AIM: To investigate growth responses of cobalt-deficient lambs to increasing doses of microencapsulated vitamin B12, and to measure associated changes in serum and liver vitamin B12 concentrations over 243 days.

METHODS: From a flock grazing pastures that had low cobalt (Co) levels (about 0.06 mg Co/kg dry matter), 4-6-week-old lambs (n=137) were assigned to four groups and received either no treatment or a subcutaneous injection of 3.0, 4.5 or 6.0 mg of microencapsulated vitamin B12 on Day 1. At approximately monthly intervals, all lambs were weighed and blood samples were collected from a selection (n=10) of monitor animals, up to Day 243. Liver biopsies were also carried out on the monitor lambs (n=8) on Days 1, 124 and 215.

RESULTS: The vitamin B12-treated lambs grew significantly faster (p<0.001) than untreated animals. Liveweights after 243 days were 28, 45, 45 and 47 kg for the untreated, 3.0, 4.5 and 6.0 mg vitamin B12-treated lambs, respectively. Of the initial group of untreated lambs, 68% had to be removed before the end of the trial because of substantial weight loss, but none of the treated animals were similarly afflicted. Serum vitamin B12 concentrations increased in all vitamin B12-treated lambs, reaching a peak at Day 25, and those of the 4.5 and 6.0 mg vitamin B12-treated lambs remained significantly higher (except at Day 124) than the untreated lambs to Day 187. However, at Day 124, but not Day 215, the liver vitamin B12 concentrations of treated lambs were two to three times higher than those of controls.

CONCLUSIONS: The growth rates of Co-deficient lambs were markedly improved by injection of 3.0, 4.5 or 6.0 mg of microencapsulated vitamin B12, and liveweights were maintained for at least 243 days. Serum vitamin B12 concentrations were related to this growth response; concentrations of <220 pmol vitamin B12/l were associated with a 95% probability that lambs were Co-deficient and would thus respond to Co/vitamin B12 supplementation. Based on these data, the current New Zealand reference criteria for Co deficiency should be reviewed.

CLINICAL SIGNIFICANCE: An injection of 3 mg microencapsulated vitamin B122 given to lambs at tailing will treat Co deficiency and will increase and maintain liveweights in a flock for up to 8 months.  相似文献

Hereditary Chondrodysplasia or Spider Lamb Syndrome (SLS) is an inherited, semi-lethal, musculo-skeletal disease affecting lambs primarily of Suffolk or Hampshire breeding. Deformities of the limbs and spinal column along with multiple sites of ossification at the anconeal process are diagnostic for the disease. Muscle atrophy is also predominant. We have investigated the relationship between SLS and circulating levels of IGF-I and the IGF-BPs in older (50-80 d of age) animals. Serum IGF-I levels were lower (P less than 0.01) in SLS affected lambs (117 ng/ml) than in phenotypically normal lambs (188 ng/ml) while serum levels of the 32 kDa BP increased (P less than 0.01) 77% in SLS affected lambs as compared to contemporary controls. All other IGF-BPs appeared to be unaffected in this group. Gene expression of IGF-I and -II in the liver and muscle of younger (16-22 d of age) lambs was also measured. There were no differences in IGF-II expression in either muscle or liver between SLS affected and phenotypically normal control lambs. Muscle IGF-I expression also did not differ. However, liver IGF-I expression in SLS affected lambs was nearly double that of control lambs (P less than 0.01). These data suggest that the regulation of IGF-I and the IGF-BPs may be involved in the physical manifestations of this disorder.  相似文献

Alveld is a hepatogenous photosensitization disease seen in lambs grazing Narthecium ossifragum pastures in Norway. Mycotoxins, possibly sporidesmin, have been suspected to cause the liver damage in alveld as in facial eczema. The histological changes in the liver of alveld cases and in lambs photosensitized after experimental sporidesmin intoxication were compared. The liver damage characterized by necrosis in single centrilobular hepatocytes, was of the same type in both conditions. Minor to moderate portal fibroplasia and bile duct proliferation were almost always present. Accumulated glycogen was seen in hepatocytes in the centrilobular areas. This was significantly correlated to the enzymatically measured glycogen content and there was good correlation between parenchymal damage and glycogen accumulation. The glucose-6-phosphatase and glycogen phosphorylase activities were normal. These findings indicate that parenchymal damage, rather than obstruction of the bile ducts, caused the retention of phylloerythrin both in alveld cases and in experimentally sporidesmin-intoxicated lambs. Vitamin E deficiency myopathy (white muscle disease) was induced in 14 suckling lambs (2 experiments; 7 lambs/experiment) by addition of cod liver oil to the diet. Disulfiram, an antioxidant, was administered orally once each day to 8 of the 14 lambs at 2 different doses. Serum creatine kinase (CK) activity was measured weekly for 5 weeks. Increased CK activity was evident in some lambs beginning at week 3. By week 4, serum CK was abnormally increased in 5 of the 6 nontreated lambs (ie, disulfiram not given) and in 4 of the 8 treated lambs. The combined disulfiram groups had significantly lower serum CK values during the study (P less than 0.05). Serum alpha-tocopherol, measured on samples from week 5 for lambs of experiment 1, was significantly higher in treated lambs (P less than 0.01). Microscopic examination of the vastus lateralis muscle indicated that the most severe lesions, consistent with nutritional myopathy, were seen in nontreated lambs. Therefore, disulfiram may have an antioxidant effect in lambs with vitamin E deficiency.  相似文献

For the investigation of the cause of white muscle disease (WMD), tocopherol (Toc) and selenium (Se) levels and blood glutathione peroxidase (GSH-Px) activities were examined using lambs with WMD and their ewes. Serum Se levels of 4 lambs with WMD were low under 30 ppb, lambs showing very low levels below 15 ppb. The serum Se level was correlated with blood GSH-Px activity showing remarkably low activities in the lambs with WMD. Se contents in the organs of lambs with WMD were lower than those of control lambs, and particularly liver Se contents were deficient levels below 50 ppb. Serum Toc levels were normal, but alpha-Toc contents in organs showed very low levels, especially in the liver. The serum Toc and Se levels and blood GSH-Px activities of their ewes and other sheep kept in the same farm revealed similar results to those of lambs with WMD. Feedstuffs supplied on the farm showed the deficient level of the Se content below 50 ppb and a very low level of alpha-Toc. It was concluded that WMD of lambs in Hokkaido was nutritional muscular dystrophy resulted from deficiencies of Toc and Se to their ewes.  相似文献

Abstract

Lambs grazing cobalt-deficient pastures and injected with hydroxocobalamine gained significantly more weight and excreted significantly less methylmalonic acid in the urine than untreated controls. Lambs with liver vitamin B12 levels in the range 0.1–0.2?g/g excreted less than 25 ?g of methylmalonic acid per ml of urine, whereas lambs with liver vitamin B12 concentrations of less than 0.1 ?g/g excreted greater amounts. Lambs in both groups had serum vitamin B12 concentrations less than 500 pg/ml.

No consistent diurnal variation in urinary methylmalonic acid concentrations was found for four lambs studied.

There was a decrease in the methylmalonic acid levels of urine after storage for more than 24 hours which could be prevented by acidification of the urine.

A mean urinary methylmalonic acid concentration greater than 30 ?g/ml for 10 animals randomly selected from a flock would indicate a cobalt deficiency in the flock as a whole.  相似文献

Sixteen lambs exhibiting hepatogenous photosensitization (alveld) after grazing pasture containing Narthecium ossifragum and seven nonphotosensitized lambs grazing the same pastures were studied. All the alveld-affected lambs revealed liver damage dominated by single cell necrosis, portal fibroplasia and bile duct proliferation. Crystalloid clefts were demonstrated in the bile ducts of two and in the hepatocytes and Kupffer cells of nine photosensitized lambs. Plasma bilirubin concentration was severely increased in ten of the cases of alveld whereas the activity of aspartate aminotransferase was moderately to severely increased in seven cases. The activity of glutamate dehydrogenase was moderately elevated in one of the photosensitized lambs. The main histopathological findings in the kidneys from the alveld-affected lambs were dilated tubules, often with eosinophilic material in the tubular lumina. Regenerative changes were seen in a large proportion of the renal sections. Elevated plasma concentrations of urea and creatinine, and the renal histopathological changes, suggested that the photosensitized lambs had been through a phase of renal injury. Analysis of the free and conjugated sapogenin content in liver tissue and bile was performed by gas chromatography–mass spectrometry. There were significantly higher concentrations of conjugated episapogenins in both the liver and bile in the alveld-affected lambs than in the nonphotosensitized lambs.  相似文献

Alveld is a disease in lambs of domestic sheep (Ovis aries L.), characterized by a combination of photosensitivity and liver damage. Generation of singlet oxygen play a major role in phototoxicity reactions. The compound phylloerythrin (phytoporphyrin) is so far assumed to be the main photodynamic agent in hepatogenous photosensitivity diseases in sheep. Phylloerythrin is a potent photosensitizer and an efficient source of singlet oxygen. The compound accumulates in the peripheral circualtion upon liver damage. Liver dysfunction is also likely to cause an increase in the blood level of bilirubin. Formation of singlet oxygen by bilirubin is reported. In the present work the photosensitizing potential of serum has been measured and related to the bilirubin- and phylloerythrin levels in lambs suffering from alveld and in clinically healthy controls. The singlet oxygen level of the serum was taken as a measure of the photosensitizing potential. The observed singlet oxygen values in serum from alveld lambs were significantly higher than the corresponding values observed in clinically healthy control lambs. This indicates that the serum of the alveld lambs contains an elevated concentration of photosensitizer. The singlet oxygen level was not correlated to the concentration of bilirubin or phylloerythrin. The results indicate that the photosensitizing mechanism is quite complex and may involve other sensitizer(s) than phylloerythrin.  相似文献

Mycoplasmas were isolated from yak calves in Mongolia, the 1st experience worldwide with this species. They were also isolated from pathologically affected sheep and goat lambs. The disease was experimentally reproduced in yak calves as well as in sheep and goat lambs. Clinical manifestations were identical with those recordable from animals with spontaneous outbreak of the disease. This had been for the 1 time ever that a therapy was tested on animals with mycoplasmosis, using water with addition of oligodynamic silver. Something between 95 and 100% of all patients were clinically cured. The experimental vaccine involved was found to offer protection against mycoplasmosis to sheep and goat lambs. Damage association with mycoplasmosis was found to occur 1st in liver, spleen, and lymph nodes and to be subsequently proliferated to lung and other organs. Respiratory mycoplasmosis is the common definition used at present but is not in conformity with the pathological processes involved, as the major role is played by liver damage. Mycoplasmosis in these 3 species, therefore, should better be named mycoplasmosis bovi hepatica.  相似文献

The objective of the experiment was to determine the optimal dose of copper oxide wire particles (COWPs) to reduce infection of Haemonchus contortus in male lambs. Five to six-month-old hair breed lambs were housed on concrete and fed 450 (L; n = 25) or 675 g (H; n = 25) corn/soybean meal supplement and bermudagrass hay. In July, lambs were inoculated with 10,000 L(3) larvae (97% H. contortus; Day 0). Lambs were administered 0, 2, 4, or 6 g COWP on Day 28. Concentrations of copper in the liver were determined. There were no effects of supplement level on concentrations of copper in the liver and a linear relationship existed between COWP treatment and concentrations of copper in liver (P < 0.001). Least squares means of the 0, 2, 4, 6 g COWP treatments were 62.2, 135.7, 161.1, and 208.4 ppm (P < 0.001). Between Days 0 and 28, PCV declined and by Day 42, PCV of all COWP-treated lambs was markedly higher than control lambs and remained higher (COWP x day, P < 0.05). By Day 21, PCV was greater in the H compared with the L group of lambs (P < 0.001). Within 14 days of COWP treatment FEC declined from more than 8000 eggs/g (epg) to less than 250 epg in all COWP-treated lambs (COWP x day, P < 0.001). The numbers of H. contortus in the abomasum were greatly reduced in all COWP-treated groups of lambs and remaining nematodes were predominantly males. FEC were greater in L versus H supplemented lambs and values decreased to a greater extent in H lambs when treated with COWP (supplement x COWP; P < 0.02). The 2 g COWP was effective in alleviating H. contortus infection and reducing number of egg-laying nematodes in the abomasum with the lowest concentration of copper in the liver of the COWP treatment groups. PCV values were more favorable for lambs fed the higher level of supplement, especially when FEC were greater than 8000 epg.  相似文献

The effect of age, sex and growth rate on total serum lactate dehydrogenase (LDH) and its isoenzymes was assessed in Suffolk X Halfbred lambs from 1.5 days to 10 weeks old. The total LDH activity was higher at 1.5 days than in older lambs and the percentage of LDH1 tended to increase while LDH4 and LDH5 decreased with age from 2 weeks, indicating that the isoenzyme distribution was changing towards the adult pattern. In female lambs, the percentage of LDH1 was higher than in male lambs at 1.5 days, but little importance was attached to this finding since the difference was not significant at 10 weeks. No clear relationships existed between the level of total LDH or its isoenzymes and daily liveweight gain.The age-related changes were considered to be of greater significance than those related to sex and growth rate when interpreting serum LDH levels in lambs.  相似文献

Fat‐tailed sheep breeds can tolerate periods of negative energy balance without suffering from elevated concentration of plasma non‐esterified fatty acid (NEFA). This ability was attributed to unique metabolism of fat‐tailed adipose depot, whereas role of liver as an influential organ in fatty acid metabolism was not evaluated yet. Hence, current study was conducted to evaluate the effects of negative and positive energy balances on liver expression of genes related to fatty acid metabolism in fat‐tailed and thin‐tailed lambs. Lambs experienced negative (21 days) and positive (21 days) energy balances and were slaughtered at the beginning and end of negative energy balance and at the end of positive energy balance. Real‐time quantitative polymerase chain reaction (RT‐Q‐PCR) was conducted to evaluate changes in gene expression. Expression of diglyceride acyltransferase 1 (DGAT1), 3‐hydroxy‐3‐methylglutaryl‐CoA synthase 2 (HMGCS2) and apolipoprotein B (APOB) was not affected by genotype, energy balance and their interaction. Expression of carnitine palmitoyltransferase 1 (CPT1) was significantly higher in liver of fat‐tailed comparing to thin‐tailed lambs regardless of energy balance (p < 0.02). Catalase mRNA abundance was increased in response to negative energy balance (p < 0.02), and severity of this enhancement was higher in fat‐tailed lambs (p < 0.06). Expression of CPT1 was positively correlated with expression of HMGCS2 in both fat‐tailed (p < 0.05) and thin‐tailed lambs (p < 0.002); however, the correlation was weaker in fat‐tailed lambs (0.72 vs. 0.57, respectively, for thin‐tailed and fat‐tailed lambs). There was a positive correlation between DGAT1 and APOB genes expression in fat‐tailed lambs (0.94; p < 0.001), whereas this correlation was not observed in thin‐tailed lambs. Results demonstrate that liver of fat‐tailed lambs has higher capacity for metabolism of mobilized NEFA exposed to liver during negative energy balance.  相似文献

SUMMARY Outbreaks of ovine white liver disease (WLD) on 7 farms in eastern Victoria were investigated. Most occurred in late spring and mainly affected lambs 3 to 6 months old, with a morbidity of 20 to 100% and mortality of 8 to 15%. Clinically affected lambs showed illthrift, emaciation and bilateral, serous, ocular discharge. Clinical pathology showed mild anaemia, elevated serum liver enzymes (GGT, OCT, AST) and low levels of serum vitamin B12. Grossly, the livers were pale, fatty and friable; microscopically there was parenchymal fatty change, bile duct proliferation and ceroid pigmentation. Liver cobalt values were consistently low (mean 0.4 ± 0.4 μmol/kg D.W.). Levels of cobalt in pasture from 2 properties were very low (0.34 μmol/kg D.W.). The diagnosis of white liver disease was made on the basis of clinical features, specific liver pathology and low cobalt status. Treatment trials established that cobalt injections or oral bullet administration resulted in clinical improvement, significant weight gains, and improved serum vitamin B12 levels. WLD did not recur in previously affected sheep using these treatments. However, when blocks containing cobalt were available continuously, WLD recurred 2 years after the initial outbreak.  相似文献

The influence of plane of nutrition, growth hormone (GH) treatment and dietary polyunsaturated fat on serum concentrations of GH and insulin (INS) and binding capacities of GH, INS and prolactin (PRL) in liver, mammary parenchyma and adipose tissue was assessed in prepubertal ewe lambs. Ten lambs were assigned to each of four treatment groups. Treatments included: (A) lambs with ad libitum access to a high-energy ration; (G) lambs fed as group A and treated with bovine GH (.08 mg/kg/d); (R) lambs with feed intake restricted to limit ADG to about 120 g; and (S) lambs with ad libitum access to a ration including formaldehyde-protected sunflower seed. Diets, all approximately isonitrogenous and isocaloric, were fed from about 7 to 22 wk of age. Weekly blood samples were collected during wk 6 to 14 of the trial. Averaged across sampling dates, mean serum concentrations of GH were elevated in G lambs (P less than .05) and INS concentrations differed in the order G greater than A greater than R = S (P less than .05). Crude membranes for binding assays were prepared from liver, mammary parenchyma and adipose tissue. Mean concentrations of GH receptors in liver and PRL receptors in mammary parenchyma were elevated in group S lambs (P less than .01). Dietary polyunsaturated fat increased the number of GH receptors in liver and PRL receptors in mammary parenchyma. Increased availability of receptors may mediate the stimulation of mammary growth observed in lambs fed polyunsaturated fat.  相似文献

Splanchnic metabolite flux was measured in young lambs given access to a high-concentrate diet either ad libitum (ADLIB) or at a maintenance level (MAINT) for 21 d. Net fluxes of urea N (UN), ammonia N (NH3 N), alpha-amino N (AAN), amino acids, glucose (G), and lactate (L) across the liver and portal-drained viscera (PDV) were measured in 11 crossbred ram lambs (35 kg) surgically fitted with indwelling catheters in the portal, hepatic, and mesenteric veins and mesenteric artery. During the 21-d period, daily N and ME intakes were 24.6 and 10.7 g N/d and 3.02 and 1.28 Mcal/d, respectively, for ADLIB and MAINT lambs. Intakes, thus, were 42% lower for MAINT than for ADLIB lambs. Net portal fluxes of UN, NH3 N, AAN, and L in MAINT lambs were 46%, 84%, 50%, and 74%, respectively, of that in ADLIB lambs. Expressed as a percentage of N intake, the proportion of AAN absorbed by the PDV was higher in MAINT lambs (P less than .05) than in ADLIB lambs. There was no net portal glucose absorption in either group of lambs; however, net hepatic glucose production in MAINT lambs was 48% of that in ADLIB lambs. There was net utilization of glutamine by the PDV; net glutamine flux in MAINT lambs was 49% of that in ADLIB lambs. The liver utilized AAN and NH3 N and produced UN. Splanchnic tissues modulate metabolite flux following changes in feed intake in young ruminants.  相似文献

A study was designed to characterize potential relationships between liver concentration of nine specific elements and age in preweaned Michigan lambs. All lambs that died before weaning in 1986 from 33 Michigan sheep producers were submitted to the Michigan State University Animal Health Diagnostic Laboratory for necropsy. Inductively coupled argon plasma emission spectroscopy was used to measure liver concentrations of Ca, Cu, Fe, K, Mg, Na, Na, P, and Zn in 888 lambs. Linear and non-linear regression techniques were employed to evaluate the relationship between liver element concentration and age. The potentially confounding variables farm of origin and cause of death were significant (P<0.01) in all models. No linear relationship was found between any liver element concentration and lamb age. Significant (P<0.05) non-linear relationships were found between liver concentrations of Cu, Fe, K, Na, Mg, and Zn and lamb age. Because of the large sample size of lambs selected from non-experimental populations, these results should provide diagnosticians with a useful guide for the interpretation of similar data when investigating perinatal lamb mortality.  相似文献

The difference in susceptibility to alveld between lambs and adult sheep may be caused by differences in the microsomal enzyme activities in their livers. There was no difference in NADPH-cytochrome c reductase or 7-ethoxyresorufin-O-deethylase (EROD) activity between ewes, control lambs and phenobarbitone-dosed lambs 3 weeks after dosing ceased. However, aldrin epoxidase activity was at that time significantly highest in the phenobarbitone-dosed lambs and significantly lowest in the ewes. The liver cytosolic glutathione transferase activity was significantly highest in the ewes and significantly lowest in the control lambs at the same time.  相似文献

One hundred and eight Texel sired lambs were weaned at 37 days old and allocated three days later to one of two treatments, using two pens per treatment, in order to evaluate lamb hepatic copper (Cu) values. In Treatment 1 (T1) the lambs were confined to pens having recycled plastic slatted flooring, while in Treatment 2 (T2) the lambs were confined to pens having straw bedded floors, with fresh straw added on alternate days. The lambs had ad libitum access to a coarse lamb concentrate. The crude protein (g/kg DM), crude fibre (g/kg DM), total zinc (mg/kg) and background levels of copper (Cu) (mg/kg) and molybdenum (Mo) (mg/kg) of the diet were 17.2, 10.9, 152, 9.7 and 0.92, respectively. The lambs were slaughtered when they reached about 39 kg live weight. Following slaughter, the liver was removed and weighed and the caudal lobe frozen for subsequent Cu analysis. The livers of lambs on the plastic floor treatment had higher liver weights (843 vs. 804 g, S.E.M. 11.0; P < 0.05) and higher hepatic copper (396 vs. 315 mg/kg DM, S.E.M. 18.2; P < 0.05) than those from lambs that were bedded with straw. The Cu accumulation per kg of liver DM per week was estimated to be 8.4 and 13.7 mg for lambs on the straw bedding and plastic slated treatments, respectively. It is concluded that when lambs are housed on plastic slatted flooring, relative to straw bedding, there is a higher accumulation of hepatic copper levels likely to result in a greater risk of Cu toxicity when lambs are reared on all concentrate diets.  相似文献
