The semivolatile cyclic alcohols 2-methylisoborneol (MIB) and geosmin (GSM) impart muddy or musty flavors to water and food products. A rapid quantitative analytical technique has been developed whereby microwave distillation is used to remove the volatile organic compounds from a lipophilic matrix into an aqueous matrix. Solid-phase microextraction (MD-SPME) is then used to extract and concentrate the analytes, which are then desorbed in the injection port of a gas chromatograph/mass spectrometer (GC/MS) for analysis. Limits of detection are 0.01 microg/kg and limits of quantification are 0.1 microg/kg. MD-SPME is comparable in precision, requires no solvents, and is faster than current methods of analysis. This methodology allows detection of MIB and GSM at concentrations below human sensory thresholds in fish tissue.  相似文献

The role of 2-methylisoborneol (MIB) in coffee aroma is controversially discussed in the literature. MIB is known as an off-flavor compound in drinking water and food, but it has also been suggested as a key flavor component of Robusta coffee, discriminating Robusta from Arabica coffee. To check this hypothesis the role of MIB in coffee brews was studied. Two reference samples containing pure Arabica and Robusta coffee brews were compared with five samples of Arabica coffee brews containing increasing amounts of MIB. The sensory panel consisting of 12 assessors perceived a distinct difference in the Arabica coffee odor and flavor in the presence of 10-25 ng/kg MIB, which is close to its threshold value in water. The sensory impression was described as musty, mold-like, and earthy. The intensity increased with increasing concentration of MIB. The panelists agreed that there was no similarity with the Robusta reference sample. The Arabica coffee brew spiked with MIB was no longer palatable due to the odor and flavor defect formed.  相似文献

The production of 2-methylisoborneol (MIB) by certain fungi and algae can contribute musty off-flavors to foods and water supplies if uncontrolled. The goal of this research was to develop a nonsensory simple method for the detection of MIB. Anti-MIB monoclonal antibodies were produced by immunizing mice with borneol-conjugated protein and selecting positive clones with an MIB-protein conjugate. An indirect competitive immunoassay developed using this antibody had a detection limit of 0.6 microg L(-)(1) and an I(50) value of 5 microg L(-)(1). Detection was relatively specific for MIB and showed 20% cross-reactivity with borneol or isoborneol and 4-5% cross-reactivity with camphor. No cross-reactivity to geosmin was observed.  相似文献

The perceived intensities of savory flavors in hydrocolloid-thickened solutions were investigated using sensory paired comparison tests between two distinct thickener concentrations (high and low viscosities). The perceived saltiness of 3.5 g/L NaCl was found to be significantly reduced (P < 0.01) at the higher thickener concentration of both hydroxypropylmethyl cellulose (HPMC) and lambda-carrageenan, relative to the lower concentration. Mushroom flavor (8 ppm of 1-octen-3-ol with 3 g/L NaCl) was perceived as significantly more intense (P < 0.05) in 1.7 g/L lambda-carrageenan as compared with the same concentration of flavoring in 10.2 g/L lambda-carrageenan. Garlic flavor (2.5 ppm of diallyl disulfide with 2 g/L NaCl) was perceived to be significantly more intense in 2 g/L HPMC (P < 0.01) than in 10 g/L HPMC. However, when the NaCl concentration in the more viscous sample was increased to 3 g/L, the garlic flavor intensities of the two systems were not significantly different, suggesting a perceptual interaction (enhancement) between salt taste and garlic flavor. In vivo aroma release measurements from the same samples, using API-MS, showed that hydrocolloid concentration did not significantly alter the amount of mushroom or garlic aromas released when solutions were consumed. It was concluded that changes in perceived saltiness were driving the reduction in savory flavor perception even though the aroma stimulus was unchanged (a taste-aroma interaction). These findings parallel previous results in sweet hydrocolloid-thickened solutions.  相似文献

The effects of 2-methylisoborneol (MIB) and geosmin, the main sources of soil odor produced by microorganisms, on plant seed germination were examined for 15 kinds of Brassicaceae seeds including radish (Raphanus sativus var. radicula). Authentic MIB or geosmin was used at the concentration of 100 mg L-1 in the assay, the effects were quantitatively indexed based on four parameters of cumulative seed germination: the final germination percentage, germination onset, weighted mean rate, germination index. MIB and geosmin inhibited germination of all kinds of seeds tested for these parameters. Radish variety “Comet” was the most affected of all of the tested plant seeds. The estimated 50% inhibitory concentration values on germination of “Comet” were 70.5 and 7.5 mg L-1 for MIB and geosmin, respectively. We also conducted the primary characterization of the inhibition by MIB and geosmin on “Comet” seeds. Our findings were as follows: (1) onset of seed imbibition was not affected; (2) germination of seeds pre-imbibed in the control solution until approx. 2 h before germination onset was inhibited by subsequent MIB or geosmin treatment; (3) growth of seedlings was not significantly affected; (4) the germination was restored by stratification, gibberellin A3 treatment, or seed coat removal; and (5) light irradiation affected the degree of inhibition. These results suggest that inhibition by MIB and geosmin is restricted to the germination process, at least in the early stage of growth, that they act as growth -regulating substances.  相似文献

Atlantic salmon (Salmo salar L.) juveniles were fed either 100% fish oil (FO), 75% vegetable oil (VO), or 100% VO throughout their life cycle to harvest weight followed by a finishing diet period when all groups were fed 100% FO. The two experimental VO diets were tested at two different locations (Scotland and Norway) against the same control diet (100% FO). The VO blend was composed of rapeseed oil, palm oil, and linseed oil using capelin oil as a control for fatty acid class compositions. Flesh fatty acid profiles were measured regularly throughout the experiment, with the times of sampling determined by changes in pellet size/lipid content and fish life stage. Growth and mortality rates were not significantly affected by dietary fatty acid compositions throughout the life cycle, except during the seawater winter period in Norway when both growth and protein utilization were increased in salmon fed 100% VO compared to 100% FO. Flesh fatty acid composition was highly influenced by that of the diet, and after the finishing diet period the weekly intake recommendations of very long chain n-3 polyunsaturated fatty acid (VLCn-3 PUFA) for human health were 80 and 56% satisfied by a 200 g meal of 75% VO and 100% VO flesh, respectively. No effect on flesh astaxanthin levels was observed in relation to changing dietary oil sources. Sensory evaluation showed only minor differences between salmon flesh from the dietary groups, although prior to the finishing diet period, flesh from 100% VO had less rancid and marine characteristics and was preferred over flesh from the other dietary groups by a trained taste panel. After the finishing diet period, the levels of typical vegetable oil fatty acids in flesh were reduced, whereas those of VLCn-3 PUFA increased to levels comparable with a 100% FO fed salmon. No differences in any of the sensory characteristics were observed between dietary groups. By blending VOs to provide balanced levels of dietary fatty acids, up to 100% of the fish oil can be replaced by the VO blend without compromising growth or flesh quality. At the same time, 75% of the dietary fish oil can be replaced without compromising flesh VLCn-3 PUFA content, thereby providing a beneficial nutritional profile for human consumption.  相似文献

The effect of the addition of marine algae in fish feed on the levels of bromophenols in fish flesh was studied. These bromophenols include 2-bromophenol, 4-bromophenol, 2,4-dibromophenol, 2,6-dibromophenol, and 2,4,6-tribromophenol. Two types of algae-containing fish feeds with 30% Padina arborescens and 30% Sargassum siliquastrum were developed. The total bromophenol contents of these feeds were 132 and 340 ng/g respectively, which were significantly higher than that of the control feed (8.9 ng/g) (ANOVA, p < 0.05). Silver seabream was used as the model fish for the feeding experiment. Bromophenol contents of both fish gut and flesh were monitored at 2-week intervals throughout the 8-week period. Two-way ANOVA showed that only the 30% S. siliquastrum-containing feed significantly (p < 0.05) increased the total bromophenol content in the fish flesh with time. 为实现鱼粉加工副产物的高值化利用,本研究对鱼粉加工压榨液经酶解发酵后的挥发性风味及游离氨基酸含量进行了分析。结果表明,鱼粉加工压榨液经酶解发酵后,其滋味成分、游离氨基酸含量、呈味强度和呈鲜能力明显增加,且其牛磺酸含量高达0.263 g·100mL-1,远高于市售鱼露。挥发性风味分析表明,酶解和发酵显著改善了鱼粉加工压榨液的风味,其醛醇类化合物、吡嗪类以及含硫类化合物含量均有所增加,酮类物质含量下降,其风味与市售鱼露相当。本研究为鱼粉加工压榨液或其他水产品下脚料的综合利用提供了有效手段。  相似文献

Determination of the microbial origin of geosmin in Chinese liquor   总被引:1,自引:0,他引:1  
Geosmin is the major cause of the common earthy off-flavor in light-aroma type Chinese liquor and, thus, highly detrimental to the aromatic quality. To find out its origin, the evolving process of geosmin in light-aroma type liquor making was monitored, and microbial analysis of Daqu containing geosmin was carried out. The results showed that geosmin appeared in all the fermented sorghums at different fermentation periods. About 57% geosmin in the fermented sorghums was distilled into liquor. During the distillation process, the peak of geosmin concentration appeared when alcohol content was 50-60% vol. More importantly, high geosmin content was observed during the Daqu-making process. Furthermore, five Streptomyces strains were isolated from different types of Daqu used for the fermentation of light-aroma type liquor. All of them produced only geosmin as the main volatile metabolite but no 2-methylisoborneol (2-MIB). It appears that microorganisms developing in Daqu are responsible for the presence of geosmin in liquor. Because of the relatively low detection threshold estimated at 110 ng/L in 46 vol % hydroalcoholic solution, the presence of geosmin in Daqu may pose a risk for Chinese liquor producers.  相似文献

对茶叶感官审评分属性与其化学组分的相关性进行探讨,该文以不同等级、产地红茶和绿茶为代表,利用传统感官审评方法鉴定茶叶品质(外形、香气、汤色、滋味和叶底),并对各品质指标进行相关性分析,筛选出对茶叶品质影响最重要的品质指标,即滋味。在利用尺度评价法对茶叶滋味品质的10个分属性(浓度、厚度、甘度、鲜度、醇度、涩度、嫩度、陈味、纯正度、火工度)进行更为细致的评定和对茶叶中主要呈味物质—茶多酚、咖啡碱、茶氨酸进行定量分析的基础上,对不同等级、不同产地茶叶的滋味分属性进行差异分析,并对化学组分与滋味分属性进行相关性分析,结果表明,各种茶样之间的产地和等级间差异主要是鲜度和涩度,而与之相关的组分主要是茶氨酸和咖啡碱,茶多酚的影响还有待进一步考察。该研究实现了茶叶感官品质的定量分析,提高了感官审评结果的客观性和确定性,为茶叶原产地保护和分等分级提供核心技术和标准支撑。  相似文献

为了满足受控生态生保系统对鱼类生长和水资源循环利用的需求,该文围绕密闭循环水产养殖系统(recirculating aquaculture system,RAS)平台构建以曝气生物滤池(biological aerated filter,BAF)工艺为核心,辅以蔬菜栽培系统的水质净化工艺,探讨其对RAS持续运行的作用及原理。试验初始,向平台养殖池内输入300 L市政原水和60尾罗非鱼鱼苗,随后系统连续循环运行200 d,通过分析系统水质及鱼类生长状况,考察BAF耦合蔬菜栽培工艺在RAS中的适用性。结果表明,1~60 d单独BAF工艺对密闭RAS内水质污染物控制效果较差,运行60 d后水中TOC、N和P类污染物均出现大量累积。针对该问题,构建2级BAF和蔬菜栽培耦合工艺,在后续140 d内,TOC和NH4+-N累积浓度分别由60.3和2.9 mg/L降低至11.2和1.2 mg/L;NO2--N持续稳定在0.1 mg/L以下;NO3--N和PO43--P分别由累积浓度114.2和43.1 mg/L降低并稳定至54.7和15.6 mg/L左右。在BAF耦合蔬菜栽培工艺保障下,密闭RAS内95%以上水资源持续再生;罗非鱼生长状态良好,零死亡率,最大养殖密度达41.9 kg/m3;同步收获4批次蔬菜,总质量达23 420 g,平均株质量达195.17 g/株。试验结果说明,BAF耦合蔬菜栽培可有效保障RAS持续稳定运行,可为密闭受控生态生保系统的水生动物养殖提供技术借鉴。  相似文献

通过试验研究了芜湖市景观水体多环芳烃(PAHs)的污染状况。采集芜湖市主城区29个景观水体沉积相、水相和悬浮颗粒相样品,运用气相色谱—质谱联用仪(GC—MS)测定样品中16种PAHs的浓度,分析PAHs的含量分布特征及其与有机碳、黑碳的相关性,利用特征比值法、正定矩阵因子分解法(PMF)对样品中PAHs进行来源解析,并通过效应区间值法和风险熵值法进行生态风险评价。结果表明:PAHs的浓度范围在沉积相中为43.48~6 388.40ng/g,水相中为0.08~52.23ng/L,悬浮颗粒相中为2.46~60.46ng/L,平均含量分别为1 307.47ng/g,8.60,13.71ng/L。样品中PAHs与有机碳(TOC)、黑碳(BC)的相关性明显,尤其是中高环PAHs,且与湿化学氧化法(BC_(Cr))的相关性最为显著。来源解析表明芜湖市景观水体PAHs主要来源于木材等生物质燃烧和煤炭燃烧,部分来源于石油类物质燃烧,其中PMF表明生物质燃烧贡献率为50.66%(沉积相)和52.51%(悬浮颗粒相加水相)。生态风险评价结果显示芜湖市景观水体整体处于中等污染水平,研究结果可为城市水体污染的治理提供科学依据。  相似文献

Sixteen species (44 samples) of marine polychaetes and 10 species (14 samples) of bryozoans from eastern Australia were analyzed by GC/MS for the key seafood flavor components 2- and 4-bromophenol, 2, 4- and 2,6-dibromophenol, and 2,4,6-tribromophenol. All five bromophenols were found in 91% of polychaetes and 64% of bryozoans. The remaining samples all contained at least three bromophenols. 2,4, 6-Tribromophenol was found in all polychaetes and bryozoans and, with few exceptions, was present in the highest concentrations. The total bromophenol content determined on a wet-weight basis varied widely between species: for polychaetes, from 58 ng/g for Australonuphis teres to 8.3 million ng/g for Barantolla lepte, and for bryozoans, from 36 ng/g for Cladostephus spongiosus to 1668 ng/g for Amathia wilsoni. Species of polychaetes with the highest concentrations of bromophenols were all collected from muddy environments. The possible effects that dietary polychaetes and bryozoans have on the ocean-, brine-, or iodine-like flavors of certain seafoods are discussed.  相似文献

2-Methylisoborneol (2-MIB) and trans-1,10-dimethyl-trans-9-decalol (geosmin) are two commonly observed taste and odor compounds present in drinking water sources. The effects of pH on the analysis of the two chemicals are investigated using a gas chromatograph and mass spectrometric detector (GC/MSD) coupled with three pre-concentration methods, namely solid-phase micro-extraction (SPME), purge-and-trap concentration (PTC), and liquid–liquid extraction (LLE). At neutral and alkaline pH conditions, the concentrations detected for both compounds remain constant. However, a substantial reduction of concentration for both chemicals is observed when the water solution pH is less than 5. Under acidic conditions (pH ≅ 2.5), the 2-MIB concentrations detected by GC/MSD coupled with SPME, PTC, and LLE are 87%, 16%, and 37% lower than those measured at pH 6–7, respectively. For geosmin, a decrease in concentration is only observed when using GC/MSD-SPME, presumably due to the higher extraction temperature compared to the other two techniques. The pH-dependent behavior was attributed to dehydration of the tertiary alcohols of 2-MIB and geosmin under acidic conditions. The dehydration for 2-MIB and geosmin is reversible, and the analysis can be mitigated by adjusting the water solution pH back to a neutral condition.  相似文献

Whereas freshly pressed linseed oil provides a delicate nutty flavor, a lingering bitter off-taste is developing upon storage at room temperature. Using a sensory guided fractionation approach, the key bitter compound was identified in stored linseed oil, and its structure was determined as the methionine sulfoxide-containing, cyclic octapeptide cyclo(PLFIM OLVF) by means of FTIR, LC-MS, NMR spectroscopy, and amino acid analysis. Although this peptide is known in the literature as cyclolinopeptide E, the bitter taste activity of this compound has not previously been described. Sensory evaluation revealed a recognition threshold concentration of 12.3 micromol/L water.  相似文献

水产种业的高质量发展是推动养殖业发展的基础,现阶段中国鱼类人工繁育生产方式设施装备化程度低,产业大而不强,转型升级的需求迫在眉睫。该研究在文献调研整理的基础上,首先就循环水人工繁育设施装备在亲鱼产卵、鱼卵孵化、鱼苗培育等方面的研究和应用情况展开论述。相对于常规培育方式,循环水系统能够最大程度上构建出符合亲鱼交配和产卵的环境条件,养殖密度0.01~4.5 kg/m3,系统循环率9~76%/h,换水率0.7~3%/d;针对不同性质的鱼卵,介绍了国内外目前常用的孵化器主要有瓶式孵化罐、平列式孵化槽、漏斗式孵化器等,阐明了其适用对象、工作原理和主要性能表现;针对育苗和养殖系统构建需求的差异,综述了目前在循环水育苗设施装备应用研究中关注的重点和难点。其次,概述了中国鱼类人工繁育发展现状和问题,分析了循环水人工繁育技术的优势和面临的挑战。最后提出,鱼类工厂化循环水人工繁育具有较高的可行性和引领性,但是要实现产业化应用仍需要进一步开展基于品种对象的人工繁育环境构建及循环水处理、繁育过程鱼类对环境应激源的生物学响应、智能繁育装备等方面技术攻关和装备研发。  相似文献

为改善单一菌株发酵制备青稞甜醅的风味与口感,提高其质量品质,本试验采用米根霉和酵母菌为发酵菌株,以氨基酸态氮含量及感官评分为指标,确定混菌发酵黑青稞甜醅的最佳工艺条件,并比较单一菌株发酵黑青稞制品与混菌发酵黑青稞制品中酚类物质含量、抗氧化活性及风味物质组成的差异。结果表明,混菌发酵黑青稞制品最佳发酵条件为:发酵温度33℃,发酵时间48 h,菌种比例(酵母菌J7∶米根霉)1∶1.20,接种量6.81%,在此条件下混菌发酵黑青稞制品的氨基酸态氮含量为9.32 mg·100 g-1,感官评分为95.48分。与单一菌株发酵黑青稞制品相比,混菌发酵黑青稞的黄酮含量(32.22 mg·100 g-1)、 多酚含量(230.68 mg·100 g-1)及DPPH自由基清除能力(95.03 μmol·L-1)显著提高。气相色谱-质谱(GC-MS)分析结果表明,酵母菌单独发酵的黑青稞中共检出33种挥发性风味物质,米根霉单独发酵的黑青稞中共检出41种挥发性风味物质,混菌发酵的黑青稞中共检出46种挥发性风味物质,其中酯类和醇类是3种发酵方式黑青稞制品的主要风味组分。混菌发酵黑青稞的醇类、酯类和酸类种类及含量均显著高于其余两种发酵方式,其相对含量分别达到59.09%、29.44%和6.46%,风味更丰富。综上分析,混菌发酵使黑青稞制品在功能及风味方面具有一定的优势。本研究结果为混菌发酵黑青稞制品的开发和应用奠定了基础。  相似文献

为降低烟熏即食鱼片中的苯并芘(BaP)含量,同时保留其感官特征和风味品质,开发熏烟在线筛滤技术,优化烟熏即食鱼片的加工工艺。本试验通过比较不同固相吸附材料(棉花、硅胶、分子筛、活性氧化铝)对烟熏即食鱼片品质的影响,采用高效液相色谱(HPLC)和气相色谱/质谱联用(GC/MS)对熏鱼的BaP和风味物质进行检测。比较不同固相吸附材料发现,当吸附剂为硅胶时,BaP含量下降至4.88 μg·kg-1,产品质地、色泽、熏香等感官评分较高,酚类等友好型挥发性物质得到保留。响应面优化后的烟熏鲟鱼片最佳工艺条件为硅胶25 g,烟熏时间30 min,烟熏温度85℃。本研究结果对健康烟熏食品的开发具有重要的参考意义。  相似文献

Methods for determination of triaryl/alkylphosphates (TAPs) in water, fish, and sediment have been extended to determination of the diarylphosphate (DAP) degradation products. DAPs were extracted from water (adjusted to pH 0.5) by use of XAD-2 resin and determined by gas-liquid chromatography as butyl esters. Recovery of diphenylphosphate (DPP) and o-, m-, p-dicresylphosphates (DoCP, DmCP, DpCP) were greater than 95% in water samples fortified at 1, 10, and 50 micrograms/L. DAPs were extracted from fish with methanol and the extracts were cleaned up on reverse phase (C18) silica cartridges. Recoveries were greater than 87% for DPP, DoCP, DmCP, and DpCP in fish muscle fortified at 50, 100, and 500 ng/g. Sediments were refluxed with aqueous methanol and DAPs were recovered by use of XAD-2 resin. Recoveries of DAPs from sediments fortified at 50 and 100 ng/g were greater than 76%. Interferences (1-10 ng/g) from phosphorus or nitrogen-containing GLC peaks prevented sub- ng/g level analysis for DAPs in sediment and fish extracts.  相似文献

A new sample preparation and enrichment technique, headspace liquid-phase microextraction (HS-LPME) linked to gas chromatography-mass spectrometry (GC-MS), was developed for the determination of the off-flavor odorants, 2-methylisoborneol and geosmin, produced by Streptomyces sp. and Anabaena PCC7120. Some of the factors that influence the extraction efficiency of HS-LPME, such as the type of extraction solvent, ionic strength of sample solution, and sample agitation rate, were studied and optimized by a single factor test. Other factors, including extraction temperature, extraction time, microdrop volume, and headspace volume were optimized by orthogonal array design. Extraction of 2-methylisoborneol and geosmin was conducted by exposing 2.5 microL of 1-hexanol for 9 min at 50 degrees C in the headspace of a 20 mL vial with a 10 mL of sample solution saturated by NaCl and stirred at 800 rpm. The developed protocol demonstrated good repeatability (relative standard deviations (RSDs) < 5%), wide linear ranges (10-5000 ng/L, r2 > 0.999), and low limits of detection (LODs) for 2-methylisoborneol and geosmin (0.05 ng/L for both analytes). Subsequently, the method was successfully applied to extract the analytes in bacterial cultures with high recoveries (from 94% to 98%). Compared with headspace solid-phase microextraction (HS-SPME), HS-LPME demonstrates better linearity, precision, and recovery. Importantly, the sensitivity is about 1 order of magnitude higher than that of most HS-SPME. The results showed that HS-LPME coupled with GC-MS is a simple, convenient, rapid, sensitive, and effective method for the qualitative and quantitative analysis of 2-methylisoborneol and geosmin.  相似文献
