The progress of gametogenesis was studied in oysters Crassostrea gigas having the same origin (Tremblade), but cultured during 1 year in two distinctive French marine areas, the Baie des Veys and Marennes-Oléron. We assessed seasonal changes in the reproduction cycle on the basis of stereological techniques to estimate reproductive investment and measurement of gonad evolution area by quantitative histology. From a qualitative point of view, both oyster groups presented typical reproductive stages, but showed differential timing, in particular during the sequence of spawning and duration of the re-absorption stage. Oysters in Baie des Veys had a single partial spawning in August and a re-absorption stage that extended until winter. Oysters in Merennes-Oléron had a partial spawning in July and massive release of gametes during August. Spawnings in both the areas were related to maximum temperature (19°C). The quantitative analysis showed, on an annual basis, a higher reproductive investment by oysters from Baie des Veys, 86% against 53% in the other group. Larger gonads, higher gamete production, and more intensive spawning were the characteristics of oysters in Baie des Veys. Recently, the reproduction pattern and investment has been related with summer mortalities; therefore, a quantitative understanding of reproductive processes becomes necessary for C. gigas. Environmental conditions at each site may explain differences in the progress and intensity of gametogenesis. While temperature regulated the time and speed of gametogenesis, results suggest that the intensity was influenced by the quantity of available food but may need further research. However, nutrient recycling from unreleased gametes in the gonads of oysters from Baie des Vey is a factor to be considered in the results of this study.  相似文献

This work aimed to evaluate the whole weight (g), shell height (mm), mortality (%) and reproductive aspects (condition index and histology) of triploid (3N) Pacific oysters (Crassostrea gigas) cultivated in southern Brazil. To accomplish this, a comparative experiment was performed with a diploid (2N) control group in two cultivation sites on Santa Catarina Island: North Bay (NB) and South Bay (SB). Whole weight, height and cumulative mortality were evaluated monthly for eight months and condition index (CI) for six months. Histological analysis was performed every two weeks for four months to investigate reproductive aspects. At the end of the experiment, whole weight and height were similar for 3N and 2N oysters. However, while more growth in whole weight and height was observed in the initial phase of cultivation in NB, such growth was not detected in SB until intermediate and final grow‐out phase. At harvest, cumulative mortality in 3N oysters was similar that in 2N oysters. The results suggest that diploid and triploid oysters grown in southern Brazil have similar developmental patterns based on whole weight and height, as well as reproductive cycle. In addition, cultivation sites have more influence on growth and mortality than ploidy levels.  相似文献

Triploid Crassostrea gigas were cultured during 13 months in Nestier‐type oyster trays. The impact of environmental parameters on the physiological and immunological parameters was evaluated. Temperature, salinity and seston were recorded monthly. Seventeen oysters were sampled monthly for immunological and condition index (CI) analyses. Samples were obtained as a haemolymph lysate supernatant (HLS). Protein content was determined using the Bradford method. The activity of hydrolytic enzymes was determined using the API ZYM kit and the lysoplate assay. Seston showed different patterns throughout the cycle. Condition index showed a positive correlation with the protein content of HLS. Protein showed a negative correlation with temperature. Eleven hydrolytic enzymes were detected in samples and higher enzymatic activity corresponded to leucine arylamidase and esterase. Leucine arylamidase and lysozyme activity showed a positive correlation with temperature. Oyster mortality was 28% in our modules and 70% in the oyster farm. Oysters showed low values of CI and haemolymph protein content in summer–autumn when mortalities were observed in the culture system. This finding suggests that these stressed oysters may have insufficient energy to invest in their immune system. It appears that oyster mortality in the culture system resulted from a combination of animal overcrowding, high temperature and low salinity.  相似文献

The Pacific oyster, Crassostrea gigas, is cultivated intertidally in Kinmen Island by both a traditional way of growing oysters on the surface of stone blocks, nicknamed “rock oysters”, and a recently introduced and more efficient way of hanging them in clusters on horizontal nylon lines, i.e. “hanging oysters”. We investigated the growth and mortality of both types of oysters from late July to early December 2003, and measured condition index (CI) and rates of oxygen consumption and ammonium excretion for the hanging oyster. Growth of oyster shells stopped in early October for the hanging oysters and early December for the rock oysters. Mortality rates were higher for hanging oysters than rock oysters. Hanging oyster's O/N ratio and CI, determined by a home-made CI meter that measured the volume of oyster's inner shell cavity with high precision, decreased significantly in October. Hanging oysters were apparently in poor physiological condition during the winter Monsoon, which is characterized by cold and persistently strong wind. On Kinmen Island the monsoon season begins in late September and is accompanied by declining seawater temperature and high seston loads in oyster farms. Chlorophyll a concentrations and seston food quality decreased significantly in early October after the winter Monsoon began. The hanging oyster's poor acclimation to the local climate was probably caused by the import of its spat from southwest coast of Taiwan where it is warmer than Kinmen Island in fall and winter. We recommend that oyster growers collect spat locally or from areas of similar climate.  相似文献

Growth is an important aquaculture trait, which has been one of the most important traits for genetic improvement in aquatic species. Numerous fast‐growing domesticated aquaculture organisms are being cultured, but the physiological processes underlying growth remain largely unexplored. The Pacific oyster (Crassostrea gigas) is one of the most widely farmed bivalves. In order to improve the productivity traits, a fast‐growing selective strain of C. gigas was established. In this study, we compared physiological energetics of the fast‐growing selective strain and an unselected hatchery population of C. gigas under different conditions of temperature. Components of energy gain (clearance rates, ingestion rates, absorption efficiency and absorption rates), energy loss (metabolic rates) and resulting energy budget (scope for growth) were analysed at four temperatures (15, 20, 25 and 30°C). The results indicated that selective breeding oysters had a higher energy gain compared to unselected oysters, but the selected strain and hatchery population of oysters did not significantly differ in metabolic rates at all tested temperatures (> 0.05). The enhanced energy gain resulted in almost two times higher scope for growth in selective breeding oysters compared with unselected oysters. Physiological characteristics of C. gigas were greatly affected by temperature and showed a fluctuation with the increase in temperatures. The selective breeding oysters outperformed those of hatchery population in growth performance by an increased energy gain along with a similar energy loss. This study provided insight into physiological energetics accounting for growth improvements in fast‐growing selective strain of C. gigas.  相似文献

The benefits of exposing eastern oysters to air during commercial culture have not been well‐characterized. An adjustable longline system (ALS) designed in Australia and recently adopted by the nascent aquaculture industry in the northern Gulf of Mexico, allows growing oysters at any position in the water column and is perfectly suited to study the benefits of air exposure. Four‐month old diploid oysters were deployed in an ALS and divided into three groups: 1) oysters exposed to air daily for 8–12 hr during low tide, 2) oysters exposed to air weekly (~24 hr once a week), and 3) oysters kept subtidally. Oyster mortality and growth rates, Perkinsus marinus load and condition index were then determined every 3 months over 2 years, while refrigerated shelf life and Vibrio vulnificus load were determined in summer and early fall of the second year. Summer mortalities were delayed, P. marinus infection intensities tended to be lower and condition index was significantly higher in oysters exposed to air daily compared with oysters exposed to air weekly or held subtidally. Shell heights of oysters exposed to air daily were lower for most of the study due to a lower growth rate during the initial sampling interval following deployment. No consistent differences were found in V. vulnificus loads or refrigerated shelf lives between the groups. It is recommended that ALS be set so that oysters are kept subtidally fall through spring to promote growth, and exposed to air daily during summer to delay P. marinus proliferation.  相似文献

Cytogenetic abnormalities associated with viral infections, including from viruses of the Herpesvirales order, have been reported in vertebrate species. Ostreid herpesvirus 1 (OsHV‐1) has been detected worldwide during mortality outbreaks of the Pacific oyster Crassostrea gigas. On the other hand, a high proportion of aneuploid cells in somatic tissues have been observed in C. gigas. In this study, we analysed the putative association between aneuploidy levels and the detection of OsHV‐1 in gills of C. gigas, the Portuguese oyster C. angulata and their F1 hybrids cultured in Ria Formosa (Portugal). OsHV‐1 was detected by PCR in 5.4% of the total of oysters analysed (n = 111) namely in 11.1%, 8.0% and 1.7% of C. gigas, C. angulata and F1 hybrid respectively. Sequencing analysis of a viral fragment amplified with the C2/C6 primer pair revealed a high similarity with the OsHV‐1 reference type. Moreover, in situ hybridization confirmed the presence of OsHV‐1 in gill tissue. Oysters where OsHV‐1 was detected had a significantly higher mean percentage of aneuploid cells (25%) than the ones where the virus was not detected (18%). However, the overall low percentage of positive samples contrasted with the high mean percentage of aneuploidy observed, with 50% of the oysters analysed showing a percentage of aneuploid cells between 20% and 30%. We hypothesize that somatic aneuploidy may adversely affect oysters making them more prone to OsHV‐1 infection, but the virus is unlikely to be the cause of somatic aneuploidy.  相似文献

This study examined the seasonal variation in the condition index (CI) of Crassostrea gigas postlarvae (<5 mm) that were cultivated at a commercial hatchery. Oysters were sampled weekly at the nursery using seawater from a lagoon for the grow‐out that precedes commercialization. Temperature, salinity, seston, chlorophyll a, oxygen and pH were recorded at each sampling and water samples were taken to identify phytoplankton groups and their abundance. High levels of primary productivity, chlorophyll a and seston were detected during summer, but the highest CI occurred in winter. During winter, elevated phytoplankton biomass was composed by diatoms and phytoflagellates, which served as the main food source and promoted weight gain in this season. Variations in salinity, oxygen and pH were not related to differences in the CI. However, it appears that the wide temperature variation affected functions, such as feeding activity, apparently enhancing ingestion during winter (mean 16.5±1.4 °C) and reducing ingestion during summer (mean 31±1.5 °C). Winter production resulted in postlarvae with a homogeneous size range and a high CI, indicating that winter is more favourable to start cultivation. The CI represents a practical means to determine the physiological state of postlarvae before transfer to cultivation sites.  相似文献

To investigate the possibility of establishing oyster culture in Malta, growth trials with C. gigas and O. edulis were undertaken. The oysters were placed in floating cages at depths of 3 and 8 m at Marsaxlokk bay, Mistra bay and Rinella creek. These oysters were sampled at two-month intervals for their growth, meat-shell index and survival. The temperature, salinity, oxygen tension, chlorophyll-a levels and the levels of particulate matter in suspension in the water at the test sites were monitored concurrently. A programme of qualitative and quantitative analyses of the natural phytoplankton population was also carried out.O. edulis suffered catastrophic summer mortalities at all locations when the ambient temperature of the water reached about 25°C; this virtually rules out any possibility of starting any flat oyster industry using this system. As for C. gigas, Marsaxlokk bay 3 m appears to hold some promise for its culture. At this location, this species showed reasonable growth, relatively low mortality and normal shell growth. Although at Rinella overall growth of C. gigas was comparable to that at Marsaxlokk at 3 m, the Rinella oysters accumulated copper and exhibited abnormal and thickened shell growth with the result that the oysters were of a much inferior condition. Growth at Mistra (both depths) and Marsaxlokk 8 m was stunted.No clear correlation could be established between the performance of the oysters and the environmental parameters monitored. Several factors that may have been responsible for the observed differences in oyster performance are discussed. In particular, the nature of the water movements is thought to have played a key role in dictating the growth of the oysters.  相似文献

In an investigation of the potential for pearl production in New South Wales (NSW), Australia, changes in the physical and reproductive condition of the pearl oyster, Pinctada imbricata, were monitored for over 2 years. Using wild oysters gathered from close to the southern extent of the species' range in Port Stephens, NSW, a series of macroscopic and histological observations were made. Reproductive activity in P. imbricata was greatest from late spring to early autumn with oysters in poor reproductive condition during winter. Peaks in reproductive indices occurred in November 1998, March 1999, December 1999 and April 2000. Four indices of physiological condition were used: shell growth, byssal attachment, mantle thickness and mucoprotien layer. With the exception of the thickness and extent of the mucoprotein layer, these indices either showed little variation or the variation that occurred was not related to seasonal or reproductive changes. Changes in the mucoprotein layer were correlated with water temperature and suggest that this layer is metabolized during periods of high demand such as during gonadogenesis. Collectors deployed at two sites in Port Stephens demonstrated that spatfall occurs largely in the months of December and January following the spring and early summer peaks in reproductive activity (November 1998 and December 1999). Spatfall was not observed following the autumnal peaks (March 1999 and April 2000) in reproductive activity. Overall, reproductive patterns in P. imbricata are poorly suited to spat supply in Port Stephens. Farmers require spat in early spring (September) to allow maximum use of the ‘growing’ season (September–May). Reproductively capable oysters are not available from the wild until September and quantity of natural spatfall is too variable and occurs too late in the season (December–January). As a result, oysters are being conditioned in the hatchery in July, spawned in August and spat are supplied to farmers in mid September.  相似文献

Winter mortality in hatchery reared oyster spat (Ostrea edulis) that received three different diets during the summer period was investigated. Oysters fed a natural type diet had a winter mortality of 18.3 ± 6.3% while oysters fed cultivated algae (a mixture of Tetraselmis suecica, Isochrysis galbana and Chaetoceros muelleri) had a mortality of 73.0 ± 9.7%. A group of oysters fed a mix between the two diets had a mortality of 54.7 ± 10.6%. Tissue samples were taken at the start of the experiment, after the summer period and after the winter period in order to determine growth and the content of glycogen and fatty acids. The glycogen content decreased for all groups during the winter but the decrease was highest in oysters fed the natural diet. This group also contained the largest variety of fatty acids, but there was no difference in the content of the essential fatty acids EPA, DPA and DHA between the groups. It is concluded that transplantation of spat to the sea in spring and early summer may reduce winter mortality since the feeding period on a more varied natural algal diet is prolonged compared to transplantation of spat later in the season.  相似文献

European oysters, Ostrea edulis, L. were kept in plastic mesh cages at three depths at two commercial oyster farm sites within Mali Ston Bay, Croatia, between April 2004 and August 2005. Oysters from each cultivation depth were sampled at both sites at approximately 3‐month intervals for growth parameters, mortality rate control and condition index calculation. The growth parameters of experimental groups from both sites only revealed a considerable distinction for shell length growth (P=0.011). Total weight gain and soft tissue weight gain were the highest for groups cultivated in the middle of the water column as compared with the top and bottom positions for both studied sites. Survival rates were very similar in all experimental groups, situated at different depths of both experimental sites, and differences in the mortality peak during the summer months were not significant. Condition indices fluctuated throughout the trial period and were the highest in spring and summer, with a reduction during the winter months and improvement again in the spring.  相似文献

Temperature and quality of the available food are important factors that influence the physiology of oysters; however, the combined effects have not been well studied. We evaluated the impacts of the temperature and diet on the growth, survival and biochemical composition in the Pacific oyster Crassostrea gigas spat, cultured in the laboratory for 8 weeks at 23, 26, 29 and 32°C and fed Isochrysis sp.‐Pavlova lutheri (IP) and Dunaliella tertiolecta (Dt). The growth and biochemical composition showed a pattern, which changed in response to rising temperature. The shell length was significantly longer, in spat fed the IP diet, except at 32°C, where both diets produced poor growth results. The survival was <50% after 5 weeks at 32°C, whereas at all other temperatures it was >88%. High temperatures directly increased lipids and saturated fatty acids, while the proteins, carbohydrates and unsaturated fatty acids decreased. High temperatures achieved in the environment, as those reached on clear summer days during low tides, are an important stressor in oyster spat, especially when the quality of the available food is poor.  相似文献

In order to study the possibility of gaining commercial benefit from culturing an excess of one sex of the Pacific oyster (Crassostrea gigas), comparative data on the growth rate and condition of male and female oysters are reported. Historically, measurement of sex‐specific growth rate in oysters has been overlooked or confounded by protandric sex. The recent conclusion that the sex ratio of Pacific oysters is predominantly under genetic rather than environmental control introduces the possibility of manipulating sex ratio for commercial gain if they exhibit asynchronous sex‐specific growth rates. Pacific oysters were cultured intertidally in Smoky Bay, south Australia. The observations, made over the 7‐month gametogenic cycle from August to February to ensure no sex reversal, were of growth rates of male and female oysters and ambient chlorophyll a concentrations. Mean shell growth of female oysters was significantly faster than that of males (4.5 ± 3.3 compared with 3.8 ± 3.2 μm day?1 mm?1 total length). Sex‐specific asymmetries in length and weight were generally significant and increased in magnitude during the 7‐month study period, suggesting potential commercial benefits from increasing the proportion of cultured female oysters. The fastest increase in the sex‐specific disparity in growth and condition came after the October chlorophyll a peak, suggesting that females utilize blooms more efficiently than males. Our results compare favourably with methods currently used to increase oyster growth (e.g. triploidy can provide growth gains of 13–51%).  相似文献

The Portuguese oyster Crassostrea angulata shows great potential in oyster farming. The conservation of pure populations of this species is important for production diversification and biodiversity preservation. In this way, the zootechnological development for seed hatchery production is extremely important. Broodstock conditioning is a key step in the process of rearing bivalves in a hatchery. Many factors regulate the reproductive cycle, being food one of the most important ones. To evaluate the effect of different diets on C. angulata reproductive performance, broodstock were conditioned with different food regimes formulated fundamentally by flagellates (Diet 1 – Pavlova lutheri and Isochrysis galbana clone T‐ISO; Diet 2 – P. lutheri, T‐ISO and Skeletonema costatum) and constituted fundamentally by diatoms (Diet 3 – S. costatum and Chaetoceros calcitrans; Diet 4 – P. lutheri, S. costatum and C. calcitrans). During conditioning, samples of oysters were collected to evaluate condition index, gonadal development and biochemical composition. At the end of the conditioning period, oysters were induced to spawn to evaluate reproductive output (fecundity, fertilization rate and D‐larvae development). The diets had an impact on the gametogenesis process, energy storage and reproductive output performance, being the best results those obtained in broodstock fed with the diatoms‐predominant diets. However, those fed with diets majority flagellates had an unsuccessful performance. Holistic approaches incorporating all results in this study reveal and reinforce the idea that the diatom species used presented the nutritional requirements to C. angulata broodstock, being essential in the conditioning phase.  相似文献

In the present study, we investigate the short‐term adaptive physiological strategies to polycyclic aromatic hydrocarbon (PAH) of Pacific oysters, Crassostrea gigas, by exposing the oysters to varying levels of PAH (0, 50, 100 and 200 μg L−1) for 7 days with a 3‐day acclimation period under laboratory conditions. The filtration rate (FR) and respiration rate (R) increased significantly at 50 μg L−1 PAH and decreased at 100 and 200 μg L−1 compared with the control. The absorption efficiency (Abs. eff.) was significantly impaired at 200 μg L−1 PAH. Ammonia excretion (E) increased with increasing PAH levels, with a significant elevation at 200 μg L−1. Although a significantly elevated FR was observed, oysters exposed to 50 μg L−1 PAH showed scope for growth (SFG) similar to the control. This indicates that even at nominal levels, PAH contamination is a possible cause of reduced oyster production because of increased food demand. The oysters exposed to 200 μg L−1 PAH showed negative SFG values, which could be a possible cause of growth stagnation or even mortality when the exposure is chronic.  相似文献

This study investigates the vulnerable period in postspawning Pacific oysters (Crassostrea gigas) through physiological and immunological assessments. After spawning, the oyster condition index reduced by 50% and required 70 days to recover to the prespawning level. The mantle glycogen reduced quickly while the reduction in tissue protein occurred slowly. The mantle tissue also lost more protein than gills. The analysis of adenylate energy charge indicated that oysters were stressed in the first 8 days after spawning. As a result of spawning, haemocyte phagocytosis was reduced and remained at a low level for 3 days. In contrast, the reduction of haemolymph antimicrobial activity did not occur until 3 days after spawning and continued to decline until day 8. This immunesuppression was not directly correlated to the changes in haemocyte density. Our study suggests that the first 8 days after spawning are a critical period for oyster survival due to the loss of energy and low immunity. This study further improves our understanding of the coincidence between spawning and summer mortality in oyster aquaculture.  相似文献

The effect of ploidy on the mortality of Crassostrea gigas spat caused by the ostreid herpesvirus (OsHV‐1) genotype μVar was investigated at five sites along the Atlantic coast in France in 2011. Sibling diploids and triploids were produced using either unselected or selected OsHV‐1‐resistant oysters. No significant interactions were found between the factors of environment, genotype and ploidy at the endpoint dates. The mean mortality rates at the sites were 62% and 59% for diploids and triploids, respectively, and the two rates were not significantly different. The mean mortality rates were 33% and 32% for sibling diploids and triploids, respectively, when OsHV‐1‐resistant parents were used and 91% and 85%, respectively, when unselected parents were used. The results were confirmed through other broodstocks tested in 2013. Our study is the first to clearly show that mortality related to OsHV‐1 is similar between diploids and triploids in C. gigas when the same germplasm is used for both ploidy. Furthermore, OsHV‐1 resistance was not substantially altered by triploidization, indicating that the achieved selective breeding of diploid oysters for OsHV‐1 resistance can be translated into improved survival in triploids.  相似文献

现生头足类的性腺发育滞后于个体生长,研究其生殖投入方式对于掌握它们的生长发育过程有着重要的意义。为此,本实验利用形态学和残差指标分析方法,对东太平洋赤道海域茎柔鱼的体征生长及其生殖投入进行了分析。结果显示,雌、雄个体均为匀速生长,雌性个体性腺成熟的最小胴长和最小体质量分别为270 mm和766 g,雄性个体性腺成熟的最小胴长和最小体质量分别为270 mm和479 g。雌、雄个体的空胃率低,摄食等级以1~2级为主;随着月份推移,个体的体征质量逐步趋好,在性腺发育阶段最差。雌性成熟个体的性腺指数平均值为2.26%±1.33%,缠卵腺指数平均值为34.34%±18.62%;雄性成熟个体的性腺指数平均值为1.31%±0.35%。雌、雄个体二者的体征指标与生殖投入指标均呈显著的正相关关系;随着性腺发育,体征指标和生殖投入指标呈下降趋势;随着胃饱满度等级增加,体征指标呈下降趋势,生殖投入指标则呈上升趋势。研究表明,茎柔鱼在性腺发育过程中持续摄食生长,生殖投入倾向于外源性,肌肉组织存储能量部分转化以满足生殖发育所需。  相似文献

This study examines the relationship of the reproductive and reserve storage cycles and spat settlement of the Queen scallop (Aequipecten opercularis) to environmental factors. The possibility of farming this species from spat settled on collectors is considered. The gonad condition index shows maximum values between mid-May and early August. Another peak occurs in mid-November. In spring and summer, the energy needed for gametogenesis is derived from food, although it is possible that direct transfer may occur from the digestive gland to the gonad. The glycogen and lipid content of the reserve organs reaches maximum values between late summer and early autumn. The energy requirements related to gametogenesis, which begins in autumn, involve the consumption of accumulated reserves with the resulting reduction in the condition indices. There is a positive correlation between temperature and the condition indices of the digestive gland, the adductor muscle and their lipid and glycogen content, respectively. Two settlement periods were recorded, one in winter, which could not be associated with the spawning of the experimental animals, and another in summer, which coincided with the spawning that took place in spring. The reduced gonad indices recorded at the end of the summer and in winter did not lead to settlement.  相似文献
