Two potato cultivars representing extremes with regard to the texture of the cooked product were divided into subcategories based on size and dry matter (DM) content. The effects of the preheating temperature and time on both the instrumentally determined firmness and the sensory-perceived firmness were measured and compared. Increasing the preheating time at 60 degrees C followed by cooking resulted in greater force required to fracture the tissue, an increase in perceived firmness, and a less mashable product. A principal component analysis showed that with higher DM contents of the potato samples, preheating resulted in a larger force required to fracture the tissue and a firmer product. The changes in fracture force were not linearly related with the changes in perceived firmness. The effects of preheating on the pectin methylesterase (PME) activity, the enzyme assumed to be responsible for the firming effect upon preheating, showed that the activity of this enzyme remained rather constant during preheating at 60 degrees C for 1 h. Preheating at 78 degrees C for 10 min abolished virtually all PME activity. To obtain insight into the consequences of preheating and preheating followed by steam cooking on the yield and composition of the cell wall material (CWM) of potatoes, a cell wall isolation followed by a pectin fractionation study was performed. Attention was also paid to the consequences of the processing conditions applied on the chemical composition of the CWM and the sequentially extracted pectic fractions. Preheating resulted in an increase in yield of the CWM of cooked potatoes and, as a consequence, all of the sequentially extracted fractions, including the residue. Preheating did not have a pronounced effect on the composition of the pectin of the sequentially extracted fractions. This altogether strongly indicates that preheating causes a PME-based firming effect, resulting in an decrease in pectin degradation and, as a consequence, a larger yield of CWM. It seems reasonable to assume that this increase in amount of CWM results in a firmer texture. The contribution of starch-based degradation products to the texture after preheating can, however, not be excluded.  相似文献

Chlorophyll and glycoalkaloid synthesis in potato (Solanum tuberosum L.) tubers occur in direct response to light. The two processes are concurrent, but independent. Color photographic indices to subjectively grade fresh market potatoes for the extent of greening were developed under lighting conditions consistent with those of retail markets. Total glycoalkaloid (TGA) and chlorophyll accumulation for four cultivars were determined over the respective greening scales, thus calibrating the scales for TGA content. On average, TGA concentrations in complete longitudinal sections of tubers (flesh samples) were highest in Dark Red Norland followed by Russet Norkotah, Yukon Gold, and White Rose. TGA concentrations of flesh samples of White Rose and Yukon Gold tubers were somewhat variable and did not increase in direct proportion to greening level and chlorophyll content, particularly at higher levels of greening. TGA concentrations in Dark Red Norland and Russet Norkotah tubers were highly correlated (P < or = 0.001) with greening level and chlorophyll concentrations. When averaged over greening levels, skin samples contained 3.4- to 6.8-fold higher concentrations of TGAs than flesh samples, depending on the cultivar. The TGA concentration in periderm samples ranged from 37 to 160 mg/100 g of dry wt. Regardless of greening level, concentrations of TGAs in the flesh samples (including attached periderm) remained within limits presumed safe for human consumption. Discrimination of greened tubers on the basis of perceived glycoalkaloid toxicity is likely unfounded for the cultivars and greening levels studied.  相似文献

A four-year study of a number of compositional parameters of potato tubers during low-temperature storage was conducted to examine the compositional differences between cold-tolerant (low sugar-accumulating) and cold-sensitive (high sugar-accumulating) tubers in relation to potato chip processing quality. Compositional parameters analyzed included sucrose, reducing sugars, nitrogen, protein, ascorbic acid, and dry matter content. Pearson correlation analysis of the data illustrated that chip color was most closely correlated with reducing sugar concentration. Multiple regression analysis revealed that the relative contribution of each parameter to chip color varied greatly among the cultivars and selections evaluated and from season to season. This analysis demonstrates that the quantitative relationships between the measured compositional parameters and chip color were not sufficient to provide a general predictive index of chip color quality for tubers processed directly from low-temperature storage.  相似文献

Steroidal glycoalkaloids occur in potatoes and are reported to impart resistance to phytopathogens including bacteria, fungi, and insects. Because glycoalkaloids can be passed to progenies during breeding programs designed to develop improved potatoes, it is of importance to determine the quality of desired characteristics and the composition of glycoalkaloids of new somatic hybrids. The objective of this study was to determine the appearance, size, and shape (morphological characters) as well as the glycoalkaloid content of potato tubers of somatic hybrids between tetraploid Solanum tuberosum cv. Dejima (2n = 4x = 48 chromosomes) and the dihaploid clone ATDH-1 (2n = 2x = 24 chromosomes) induced by anther culture from Solanum acuale-T (acl-T, 2n = 4x = 48 chromosomes). Tuber size and shape in somatic hybrids were in accord with those of cv. Dejima, whereas the tuber skin color resembled that of ATDH-1. Thin-layer chromatography, high-performance liquid chromatography, and gas-liquid chromatography/mass spectrometry studies showed that the two steroidal glycoalkaloids (alpha-chaconine and alpha-solanine) were present in the tubers of S. tuberosum, whereas acl-T and ATDH-1 tubers were found to contain alpha-tomatine and demissine. The concentrations of total glycoalkaloids in both acl-T and ATDH-1 was >100 mg/100 g of fresh weight tuber cortex, much higher than in S. tuberosum. All somatic hybrids, except one clone, contained four glycoalkaloids (alpha-chaconine, alpha-solanine, alpha-tomatine, and demissine) derived from the fusion parents. The lack of alpha-tomatine in the remaining clone may be due to somaclonal variation. The results show that character expression is influenced by ploidy level and that total glycoalkaloid levels in most somatic hybrids were intermediate between those of the fusion parents. The possible significance of these findings for plant breeding and food safety is discussed.  相似文献

We compared the antioxidant enzyme activities between Solanum tuberosum L. cultivars Kitaakari and "Danshaku" during storage at 1 degrees C and 20 degrees C. The Kitaakari and Danshaku plants contained approximately 330 microM and 120 microM ascorbic acid (AsA) immediately after the harvest, respectively. At 1 degrees C, the activity of ascorbate peroxidase (APx) in the Kitaakari plants showed the tendency to increase, while in the Danshaku its activity increased temporally by 9 weeks and thereafter returned to basal levels. Superoxide dismutase (SOD) activity increased after 12 weeks in the case of the Kitaakari at 1 degrees C. Catalase did not show any difference in both cultivars at each temperature. The contents of AsA, which was one of the substrates of APx, decreased more rapidly at 1 degrees C than at 20 degrees C in both cultivars. Particularly in the case of the Danshaku, AsA contents were already less than 30 microM at 9 weeks, which confirmed that APx was inactivated.  相似文献

Biochemical characteristics of patatin proteins purified by ion-exchange and affinity chromatography from tubers of 20 potato cultivars were studied to evaluate their genotype differences with respect to utility groups, table potato cultivars (TPCs) and processing potato cultivars (PPCs). Both groups of cultivars showed similar values of protein content in dry matter (3.98-7.39%) and of patatin relative abundance (5.40-35.40%). Three mass levels (~40.6, 41.8, and 42.9 kDa) of purified patatins were found by MALDI-TOF MS within all cultivars. Differences among mass levels corresponding with the mass of sugar antenna (~1.2 kDa) confirmed the previous concept of different glycosylation extentsin patatin proteins. It was showed that the individual types of patatin varying in their masses occur in the patatin family in a ratio specific for each of the cultivars, with the lowest mass type being the major one. Electrophoretic analyses demonstrated wide cultivar variability in number of patatin forms. Especially 2D-PAGE showed 17-23 detected protein spots independently on the utility group. Specific lipid acyl hydrolase (LAH) activity of purified patatins from the individual tested cultivars varied between 0.92 and 5.46 μmol/(min mg). Patatin samples within most of the TPCs exhibited higher values of specific LAH activity than samples of PPCs. It may be supposed that individual patatin forms do not have similar physiological roles.  相似文献

Several abiotic stresses, including ethylene, methyl jasmonate, temperature, light, and wounding, were tested for their ability to induce accumulation of phenolic compounds and antioxidant capacity in purple-flesh potatoes (cv. All Blue). Results indicated that temperature, ethylene, methyl jasmonate, and light treatments did not significantly affect the accumulation of phenolic compounds compared to control samples. Only tubers with low initial anthocyanin levels treated with methyl jasmonate showed approximately 60% anthocyanin accumulation. Wounding induced the accumulation of phenolics compounds and an increase of PAL-activity in sliced tissue compared to the control. Total phenolics increased approximately 60% with a parallel 85% increase in antioxidant capacity. These results show that selection of appropriate abiotic stresses can enhance the nutritional and functional value of potatoes.  相似文献

The carotenoid pattern of four yellow- and four white-fleshed potato cultivars (Solanum tuberosum L.), common on the German market, was investigated using HPLC and LC(APCI)-MS for identification and quantification of carotenoids. In each case, the carotenoid pattern was dominated by violaxanthin, antheraxanthin, lutein, and zeaxanthin, which were present in different ratios, whereas neoxanthin, beta-cryptoxanthin, and beta,beta-carotene generally are only minor constituents. In contrast to literature data, antheraxanthin was found to be the only carotenoid epoxide present in native extracts. The total concentration of the four main carotenoids reached 175 microg/100 g, whereas the sum of carotenoid esters accounted for 41-131 microg/100 g. Therefore, carotenoid esters are regarded as quantitatively significant compounds in potatoes. For LC(APCI)-MS analyses of carotenoid esters, a two-stage cleanup procedure was developed, involving column chromatography on silica gel and enzymatic cleavage of residual triacylglycerides by lipases. This facilitated the direct identification of several potato carotenoid esters without previous isolation of the compounds. Although the unequivocal identification of all parent carotenoids was not possible, the cleanup procedure proved to be highly efficient for LC(APCI)-MS analyses of very low amounts of carotenoid esters.  相似文献

Cooked must is the starting raw material in Aceto Balsamico Tradizionale di Modena (ABTM) production, and the cooking process is a crucial step to ensure quality and safety standards. In particular, the furfural content has to be strictly monitored. In this study, we followed, directly at the production plant, nine cooking processes, differing for grape type and process conditions in order to monitor the actual variability in cooked must reduction. 5-Hydroxymethylfurfural (5-HMF) and furfural contents were determined by high performance liquid chromatography (HPLC) technique and glucose, fructose, and tartaric and malic acids by gas chromatography (GC) technique. Furthermore, some bulk properties, such as the water content, refractive index, density, and total acidity, were also measured. The obtained results show that the product undergoes, in the worse operating conditions, remarkable degradation, leading to extremely high levels of furfurals (5-HMF and furfural), corresponding to a water content lower than 40%. On the basis of these results, a first draft of an optimal heating protocol may be suggested in order to guarantee the safety and quality of the final product.  相似文献

The influence of 2 weeks and 3 months of dark storage upon light-induced chlorogenic acid accumulation within tuber tissue of four potato cultivars and upon 5-, 4-, and 3-caffeoylquinic acid concentrations within cv. King Edward was determined. Storage period significantly affected (P < 0.05) the magnitude of the light-induced chlorogenic acid response with accumulation rates 3-4 times higher in tubers exposed to light after 2 weeks compared with those placed under light after 3 months. Comparison of chlorogenic acid concentrations in controls after 2 and 3 months of dark storage indicated that tuber chlorogenic concentrations decline during prolonged cold store at 5 degrees C. Rates of accumulation in response to light were cultivar-dependent with cv. Fianna the most light-sensitive and cv. Maris Piper relatively light-insensitive. In virtually all cases exposure to sodium and fluorescent light promoted higher rates of accumulation than did exposure to high-pressure mercury light sources. Chlorogenic acid values steadily increased over 15 days of illumination with, in the majority of cases, no indication of cessation. Light exposure increased 5-, 4-, and 3-caffeoylquinic acid accumulation rates in cv. King Edward. Irrespective of storage period and light source, ratios of 5-:4-:3-caffeoylquinic acid were ca. 85:15:0 at day 0 and 52:42:6 by day 15.  相似文献

The calystegines detected in tubers from 17 Phureja ( S. tuberosum Group Phureja) lines and five Tuberosum ( S. tuberosum Group Tuberosum) cultivars were identified as the A 3 and B 2 structural types. Their concentration in whole tubers was of a similar order of magnitude in both species, as was the variability in the ratio of B 2 to A 3. On average, calystegine concentrations in the peel were about 13 times that found in the flesh for the five Tuberosum cultivars, and 4 times higher for four Phureja lines. Removal of the peel reduced the calystegine content by an average of over 50% in Tuberosum but by only 30% in Phureja, despite the latter having the greater proportion of peel. The calystegine content of sprouts was also determined for five Tuberosum cultivars and four Phureja lines and was found to include small amounts of four additional types, B 3, B 4, N 1, and X 2, in addition to the more abundant A 3 and B 2. Concentrations in the sprouts of Tuberosum were on average 100 times higher than that in the tuber flesh and 8 times higher than in the peel, whereas for Phureja, the equivalent values were 30 and 7 times higher, respectively. No correlation was found between sprout concentration and either flesh or peel calystegine concentration.  相似文献

The potato (Solanum tuberosum L.) tuber storage protein, patatin, was purified to homogeneity with a molecular mass of 45 kDa. The purified patatin showed antioxidant or antiradical activity by a series of in vitro tests, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical (half-inhibition concentration, IC(50), was 0.582 mg/mL) scavenging activity assays, anti-human low-density lipoprotein peroxidation tests, and protections against hydroxyl radical-mediated DNA damages and peroxynitrite-mediated dihydrorhodamine 123 oxidations. Using electron paramagnetic resonance (EPR) spectrometry for hydroxyl radical detections, it was found that the intensities of the EPR signal were decreased by the increased amounts of patatin added (IC(50) was 0.775 mg/mL). Through modifications of patatin by iodoacetamide or N-bromosuccinimide, it was found that the antiradical activities of modified patatin against DPPH or hydroxyl radicals were decreased. It was suggested that cysteine and tryptophan residues in patatin might contribute to its antioxidant activities against radicals.  相似文献

Abstract

Potato (Solanum tuberosum) showed a well-established mutualistic association with arbuscular mycorrhizal fungi. In the present study, earthen pots containing autoclaved soil were taken in which surface sterilized potato seeds were sown. The seed sowed earthen pots were inoculated by sterilized spores of arbuscular mycorrhizal fungi along with sterilized inoculated maize-root fragments, while the rest half of the seed filled pots, without any inoculation depicting control but were provided with non-inoculated maize root fragments. The inoculation was performed twice; first inoculation was done 3 days prior to sowing of potato seeds and second at the time of seedling emergence. Sampling of the inoculated as well as of control plants was performed at 20-day intervals till 80?days after tuber initiation. An increment in the level of metabolites as well as mineral nutrient was found in mycorrhizal inoculated potato tubers in contrast to non-inoculated. Our work demonstrates that inoculation has a great potential in enriching storage metabolites and nutrients in potato plant in low yielding soils.  相似文献

Changes in the lipid composition of two standard infant formulas induced by 4 years of storage were determined. Lipids were thoroughly analyzed using different gas-liquid and liquid-liquid chromatographic techniques. Oleic acid and linoleic acid, which accounted for almost the total monounsaturated and polyunsaturated fatty acids, respectively, showed slight but significant decreases (P < 0.05) during the 4 years of storage (from 41.52 to 39.83% for oleic acid and from 17.35 to 15.99% for linoleic acid). Total trans fatty acid isomers showed low initial level (0.22% of total fatty acids), and such level remained unchanged during the storage period. Nonvolatile oxidation compounds including oxidized, dimeric, and polymeric triglycerides did not significantly increase during the storage period, although a significant loss of tocopherols was found in the surface oil fraction (10-15%). In general, the results obtained indicate that, although small losses of oleic and linolenic acid as well as tocopherols were found, the 4 year storage period did not lead to relevant changes in the lipid fraction of infant formulas.  相似文献

Four related phenolic amides previously undescribed from the species were revealed during metabolic profiling of potato (Solanum tuberosum) tubers. N(1),N(12)-Bis(dihydrocaffeoyl)spermine (kukoamine A) and N(1),N(8)-bis(dihydrocaffeoyl)spermidine were positively identified by comparison with authentic standards, while the structures N(1),N(4),N(12)-tris(dihydrocaffeoyl)spermine and N(1),N(4),N(8)-tris(dihydrocaffeoyl)spermidine are proposed for the other two metabolites. Each amide was present at several tens of micrograms per gram of dry matter. Several of these compounds were subsequently detected in other solanaceous species, such as tomato (Lycopersicon esculentum) and Nicotiana sylvestris. They appeared not to be present in Arabidopsis thaliana or Beta vulgaris. Bis(dihydrocaffeoyl)spermine isomers have previously been identified in only a single plant, the Chinese medicinal species Lycium chinense (Solanaceae), where they may account for some of the described biological activity. The other compounds have not until now been reported in vivo, though some of the equivalent hydroxycinnamoyl derivatives are known. The surprising discovery of kukoamine and allies in a range of solanaceous species including potato, a common food crop that has a long history of scientific investigation, provides exemplary evidence for the potential of the nontargeted techniques of metabolomics in studying plant metabolites.  相似文献

A potato field experiment was conducted for 2 consecutive years to determine the effects of nitrogen (N) and potassium (K) fertilization rates on the yield and quality of potato cv. Spunta cultivated on soil low in N and K. A 3?×?4 complete factorial experiment was used with three rates of nitrogen (330, 495, and 660 kg N ha–1) and four rates of potassium (112, 225, 450, and 675 kg K2Ο ha–1). An additional treatment without fertilization was used as the control. On soils low in N and K, potatoes showed low yield response to K fertilizer. The greatest tuber yields for both years were achieved at 495 kg N ha–1 and 112 kg K2O ha–1 (29.81 t ha–1) and 225 kg ha–1 (27.13 t ha–1), respectively. Differences in mean fresh weight due to treatment application were not significant. Application of 495 kg N ha–1 significantly reduced harvest index (the ratio of tuber dry weight to the total dry weight at harvest) compared to 330 kg N ha–1, but at 660 kg N ha–1 harvest index achieved the greatest significant value. Potassium fertilization had no significant influence on harvest index. Nitrogen rates positively influenced the number of tubers. The addition of 450 kg K2O ha–1 significantly enhanced the number of tubers compared to the lower K rates, and the number was significantly decreased by the application of 675 kg K2O ha–1. Tuber dry-matter concentration was significantly promoted by N fertilization in both cultivation years, but it was negatively affected by K fertilization in the first year of cultivation. There was no change in tuber N with N application, but N application strongly increased nitrate (NO3) concentration, which fluctuated between 360 and 1382 mg kg–1 wet mass. Tuber NO3 was negatively correlated with tuber yield, indicating that high levels of NO3 in tubers can adversely affect yield. Tuber response to K fertilization was not in accordance with the rate of applied nutrient.  相似文献

An antifungal protein, AFP-J, was purified from tubers of the potato (Solanum tuberosum cv. L Jopung) by various chromatographic columns. AFP-J strongly inhibited yeast fungal strains, including Candida albicans, Trichosporon beigelii, and Saccharomyces cerevisiae, whereas it exhibited no activity against crop fungal pathogens. Automated Edman degradation determined the partial N-terminal sequence of AFP-J to be NH2-Leu-Pro-Ser-Asp-Ala-Thr-Leu-Val-Leu-Asp-Gln-Thr-Gly-Lys-G lu-Leu-Asp-Ala-Arg-Leu-. The partially sequence had 83% homology with a serine protease inhibitor belonging to the Kunitz family, and the protein inhibited chymotrypsin, pepsin, and trypsin. Mass spectrometry showed that its molecular mass was 13 500.5 Da. This protease inhibitor suppressed over 50% the proteolytic activity at 400 microg/mL. These results suggest that AFP-J is an excellent candidate as a lead compound for the development of novel antiinfective agents.  相似文献

The multi-element (Ag, Al, Au, Ba, Bi, Ca, Cd, Co, Cr, Cs, Cu, Dy, Er, Fe, Ga, Gd, Ho, In, Ir, La, Lu, Mn, Mo, Nb, Nd, P, Pb, Pd, Pr, Pt, Rb, Re, Rh, Ru, Sb, Sc, Sm, Sn, Sr, Ta, Tb, Th, Ti, Tl, Tm, U, V, Y, Yb, and Zn) concentrations (microg/kg, fresh weight) in potatoes (Solanum tuberosum, Folva) were investigated in this study. The potatoes were grown in two fertilization practices; one with pig slurry and one with calcium ammonium nitrate at three levels of N fertilization (0, 60, and 120 kg of N/ha). The experiment field was located at the Riso National Laboratory Agronomy Farms in Roskilde, Denmark. High-resolution-inductively coupled plasma mass spectrometry (HR-ICPMS) was used for analyses of the samples. The effect of three levels of N fertilization on elemental concentrations of the crop are evaluated by use of discriminant partial least-squares regression (PLS). The results provide useful biological and nutritional information on potatoes.  相似文献
