Abstract

Channel catfish Ictalurus punctatus injected with Aeromonas hydrophila, Pseudomonas fluorescens, Edwardsiella tarda, or E. ictraluri were frozen at ?20°C after death. Bacterial isolation at 2-d intervals after freezing indicated that A. hydrophila could be recovered for 20 d, P. fuorescens for 60 d, E. tarda for 50 d, and E. ictaluri for 30 d in frozen fish.  相似文献

Abstract

The in vitro and in vivo efficacies of two aryl-fluoroquinolones, A-56619 and A-56620, against two bacterial pathogens of channel catfish Ictalurus punctatus were determined The minimum inhibitory concentrations (MIC) of A-56619, A-56620, oxytetracycline, nalidixic acid, spectinomycin, ampicillin, doxycycline, and ormetoprim-sulfadimethoxine against 10 isolates of both Edwardsiella ictaluri and Aeromonas sobria were determined by the agar-dilution method. The in vivo efficacies of A-56619 and A-56620 were determined by bacterial challenge of channel catfish with E. ictaluri. Fish were fed a prepared diet that supplied 12.5, 25.0, or 50.0 mg of active drug per kilogram body weight per day. In vitro tests showed that all strains of E. ictaluri and A. sobria were sensitive to A-56619 and A-56620. The two aryl-fluoroquinolones demonstrated a lower mean MIC against these organisms than any of the other antibiotics tested. Significant reductions in mortality occurred within all channel catfish groups treated with these two chemicals as compared with controls. Mortality from E. ictaluri infection of fish given feed medicated with A-56619 and A-56620 ranged from 4 to 12%. Mortality among unmedicated controls ranged from 20 to 68%. No significant difference in mortality among dosage levels was demonstrated.  相似文献

Abstract

One hundred seven Aeromonas spp., 26 Edwardsiella ictaluri, 6 E. tarda, 12 Plesiomonas shigelloides, and 6 Pseudomonas spp. (131 piscine isolates and 26 reference isolates) were studied with 36 biochemical tests from the Minitek system, 20 tests from the API 20E system, and corresponding standard tube tests. Isolates were incubated at 25°C. Arginine dihydrolase, ornithine decarboxylase, mannose, and citrate showed less than 95% agreement between the Minitek system and the tube tests. Arginine dihydrolase, lysine decarboxylase, nitrite reductase, Voges-Proskauer, and citrate showed less than 95% agreement between the API 20E system and the tube tests. The 26 reference isolates were examined with the three systems and were incubated at both 25 and 37°C. There were no major differences between tests run at 25 and 37°C except with nine Aeromonas spp. that did not grow well at 37°C. Both the Minitek and API 20E systems will reproduce standard biochemical tube test results with at least 95% accuracy when used to test warmwater fish pathogens incubated at 25°C. However, the numerical identification databases for both the Minitek and API 20E systems were not usable for identifying fish pathogens.  相似文献

Abstract

To determine the scope of antibacterial resistance among Aeromonas spp., bacterial cultures were taken from a variety of tropical fish species imported from Singapore. The samples were plated on Rimler-Shotts medium for bacterial isolation and identified with both the API20E and Nonfermenter Test Strip systems. Aeromonas hydrophila, A. sobria, and A. caviae were identified in monomicrobic and concomitant infections. Following identification of bacterial isolates, 11 antimicrobials routinely used in the tropical fish industry and 1 new fluoroquinolone were tested for their effectiveness against Aeromonas spp. with the Kirby-Bauer disk-diffusion technique. Aeromonas sobria proved to be the most resistant, often showing susceptibility to only 3 of 12 test drugs. Aeromonas hydrophila was consistently the least resistant. Based on these results, antimicrobial resistance appears to be a rapidly emerging problem in the pet fish industry.  相似文献

Abstract

The objective of this study was to determine whether common bacterial catfish pathogens could attach and colonize surfaces commonly found in aquaculture facilities. In addition, we evaluated the role of calcium in biofilm formation. Attachment to polystyrene plates was used to quantify biofilm formation by five bacterial pathogens (i.e., Flavobacterium columnare, Aeromonas hydrophila, Edwardsiella ictaluri, E. tarda, and E. piscicida). Flavobacterium columnare and A. hydrophila formed thick biofilms that were enhanced by calcium supplementation. Biofilm formation was significantly lower in all Edwardsiella species tested and calcium had little to no effect on Edwardsiella biofilm formation. Attachment to natural and artificial surfaces was quantified by a standard plate count method. Scanning electron microscopy (SEM) was used to confirm biofilm formation on the substrates. Flavobacterium columnare formed biofilm on the liner, flexible PVC, and nets. Bamboo prevented F. columnare attachment and inhibited cell growth. Aeromonas hydrophila and E. ictaluri formed biofilm on all materials tested, although significant differences were found among substrates. While E. ictaluri failed to form biofilm on microtiter polystyrene plates, it was able to colonize and multiply on all aquaculture materials tested. Our results demonstrated that common bacterial pathogens had the potential of colonizing surfaces and may use biofilm as reservoirs in fish farms.

Abstract

Intestinal and rectal smears from 137 birds (4 snowy egrets Egretta thula, 22 great egrets Casmerodius albus, 30 great blue herons Ardea herodias, and 81 double-crested cormorants Phalacrocorax auritus) were examined by indirect fluorescent antibody test for the presence of Edwardsiella ictaluri. Edwardsiella ictaluri was detected in 53% of the birds sampled. Rectal samples from eight birds were placed in a special antibiotic broth for isolation of viable E. ictaluri. Two of these samples produced colonies of viable E. ictaluri, and the identity of these colonies was confirmed biochemically and serologically.  相似文献

Abstract

Edwardsiellosis is an important bacterial infection of freshwater and marine fishes. Edwardsiella ictaluri causes enteric septicemia of catfish, and E. tarda causes emphysematous putrefactive disease of catfish and fish gangrene in various species; these diseases have considerable economic effects on the aquaculture industry. In addition, E. tarda is an important zoonotic pathogen. Thus, the reduction or elimination of these pathogens from an aquarium or aquaculture facility is imperative. This study examined a variety of commercially available chemicals for their ability to reduce or eliminate E. ictaluri and E. tarda from the aquatic environment. The various concentrations of chemicals were tested in vitro in microcentrifuge tubes with a known concentration of bacteria at room temperature. In this study, ethyl alcohol (30, 50, or 70%), benzyl-4-chlorophenol/phenylphenol (1%), sodium hypochlorite (50, 100, 200, or 50,000 mg/L), n-alkyl dimethyl benzyl ammonium chloride (1:256), povidone iodine (50 or 100 mg/L), glutaraldehyde (2%), and potassium peroxymonosulfate/sodium chloride (1%) were effective disinfectants, as each reduced or eliminated the number of detectable organisms within 1 min of contact time. However, neither Chloramine-T (15 mg/L) nor formalin (250 mg/L) substantially reduced bacterial counts even after 60 min of contact time.  相似文献

Thirty-one Aeromonas hydrophila, 13 A. sobria and two A. salmonicida strains of diverse sources were tested for enterotoxigenicity, hemagglutination and cell surface hydrophobicity. Although 93% of the culture supernatant fluids of the Aeromonas strains exhibited cytotoxic effects on Y1 adrenal and Chinese hamster ovary (CHO) cells, typical rounding of Y1 adrenal cells was reproducibly observed before cytotoxicity for 80% of the isolates within 1 h of exposure.Twenty-eight strains were positive for delayed permeability factor (DPF) activity in rabbit skin. Culture filtrates of 16 of 20 strains that were positive both in the Y1 adrenal cell test and for DPF activity elicited fluid accumulation in rabbit ileal loops. The DPF and ileal loop activities were neutralizable by cholera antitoxin. All, except two strains each of A. sobria and A. hydrophila, produced a heat-stable, rapid permeability factor (RPF) detected in rabbit skin. Heat-treated culture supernatant fluids of two A. hydrophila and one A. sobria isolate gave positive responses in the infant mouse assay. Nine other strains gave borderline reactions.When A. hydrophila and A. sobria isolates were grown in broth, approximately 90% agglutinated bovine, chicken, human group A and guinea-pig erythrocytes in the presence of mannose at 4°C and/or 20°C. The two A. salmonicida isolates produced mannose resistant hemagglutination (MRHA) of these four blood types.Hydrophobic interaction chromatography indicated adhesive potential in 61% A. hydrophila and 100% A. sobria strains expressing weak to strong hydrophobic cell surface properties. The results of these investigations strongly imply that the Aeromonas strains produce a cytotonic enterotoxin immunologically related to cholera toxin. Adhesive characteristics were commonly found in both clinical and routine isolates.  相似文献

The present study was carried out to assess the prevalence of motile Aeromonas spp. in the faeces of clinically healthy sheep, cattle and horses and evaluate their susceptibility to some anti-microbial agents. Rectal swabs from 120 sheep, 85 cattle and 20 horses were examined for Aeromonas species using alkaline peptone water (pH 8.4) as the enrichment medium and Aeromonas Selective Agar containing 5 mg/l ampicillin as the isolation medium. Identification and antibiotic resistance of motile Aeromonas strains was performed using Gram Negative Enteric ID panel. Motile aeromonads were isolated from 12 (10%) sheep, 7 (8.2%) cattle and 1 (5%) horse. Of these 20 aeromonad isolates, 13 were A. caviae, 6 were A.sobria and 1 was A. hydrophila. Aeromonas species in the faeces of livestock might pose a public health problem for humans who are in direct contact with contaminated animals. However, further studies should be performed on aeromonads relating to their transmission between animals and humans.  相似文献

Abstract

Tannic acid, propyl gallate, methyl gallate, and gallic acid were tested for their inhibitory effects on selected aquatic microorganisms by the well assay technique. Tannic acid, propyl gallate, and methyl gallate in deionized water inhibited the growth of Aeromonas hydrophila, A. sobria, Edwardsiella iclaluri, E. tarda, Pseudomonas fluorescens, and Escherichia coli, but gallic acid did not. When 500-μg/mL concentrations of these four compounds were tested in sterilized fish pond water at pH 7.0 and with a low bacterial inoculum of 103–104 colony-forming units (CFU) per milliliter, they inhibited the growth of P. fluorescens and (except for tannic acid) E. coli. Pseudomonas fluorescens inoculated at 103–104 CFU/mL in pond water was inhibited by methyl gallate, propyl gallate, and tannic acid concentrations as low as 50 μg/mL, but a gallic acid contration of 100 μg/mL was required for inhibition. Escherichia coli was inhibited by methyl gallate and propyl gallate at 250 μg/mL and by gallic acid at 500 μg/mL, but it was not inhibited by tannic acid at concentrations up to 500 μg/mL in water of pH 7.0. Tannic acid (500 μg/mL) did inhibit E. coli growth at pH 4.5.  相似文献

The presence of potentially human pathogenic strains of Aeromonas was investigated in 84 samples of seafood which were purchased from retail traders in Berlin, Germany in spring 2000. A total of 134 Aeromonas strains were isolated on selective [GSP agar and Aeromonas (Ryan) agar] and unselective (standard count agar and enterohaemolysin agar) media from 27 (32.1%) of the samples and were classified as Aeromonas hydrophila (67.9%), A. caviae (26.1%) and A. sobria (6.0%) by biotyping. Thirteen (48.1%) of the 27 positive samples contained more than one species of Aeromonas. Production of haemolysins on enterohaemolysin agar was found with 132 (98.5%) of the strains at 28°C and with 130 strains (97.0%) at 37°C growth temperature. Vero cytotoxins were produced by 99 (73.9%) of the strains when grown at 28°C but only by 24 of the strains (17.9%) at 37°C. The latter strains were identified as A. hydrophila (n = 22) and A. sobria (n = 2) which came from 17 (20.2%) samples of raw seafood and from ready‐to‐eat salted herring ‘Matjes’ products. Cytotoxin‐encoding genes for aerolysin (aer) and haemolysin A (hlyA) were investigated by PCR. Aer and hlyA genes were detected in both, strains which produced toxins only at 28°C and strains which produced toxins at 37°C. Our data indicate that raw seafood and ready‐to‐eat fish products can harbour potential human pathogenic, cytotoxin producing Aeromonas strains.  相似文献

Abstract

The ability of Edwardsiella ictaluri to attract macrophages of channel catfish Ictalurus punctatus was investigated. Exoantigen from E. ictaluri was tested for macrophage chemotactic activities both in vitro and in vivo. The exoantigen was chemotactic and chemokinetic for macrophages in vitro. Peritoneal injection of 750 μg of exoantigen protein into normal (E. ictaluri-free) channel catfish induced a marked increase in macrophage accumulations at 24 and 48 h. Neutrophil accumulation did not occur at the injection sites. Edwardsiella ictaluri exoantigen attracts macrophages, and this attraction may play an important role in macrophage responses during E. ictaluri infections.  相似文献

Mycoplasma spp. are highly contagious pathogens and intramammary Mycoplasma infection is a serious issue for the dairy industry. As there is no effective vaccine for Mycoplasma infection, control depends on good husbandry and chemo‐antibiotic therapy. In this study, antimicrobial susceptibility of Mycoplasma strains recently isolated from cases of bovine mastitis in Japan was evaluated by minimum inhibitory concentration (MIC). All Mycoplasma bovis strains were sensitive to pirlimycin, danofloxacin and enrofloxacin, but not kanamycin, oxytetracycline, tilmicosin or tylosin. M. californicum and M. bovigenitalium strains were sensitive to pirlimycin, danofloxacin, enrofloxacin, oxytetracycline, tilmicosin and tylosin, but not to kanamycin. This is the first report to describe the MIC of major antimicrobial agents for Mycoplasma species isolated from bovine mastitis in Japan.  相似文献

Abstract

Enteric septicemia of catfish (ESC) was transmitted horizontally from channel catfish Icialurus punctatus that had died from Edwardsiella ictaluri infection to contact channel catfish during 2 d of habitation in a tank. The contact channel catfish became positive for E. ictaluri antibody, became infected with this bacterium, and had signs of ESC and died within 12 d postexposure. Edwardsiella ictaluri was recovered from 24 of the 30 contact channel catfish that died from ESC, as well as from 9 of the 25 tested contact survivors. The cannibalizing of E. ictaluri-infected fish, or the shedding of E. ictaluri from dead fish, or both, were shown to be mechanisms of horizontal transmission of ESC among channel catfish.  相似文献

Abstract

Wild and domestic populations of channel catfish Ictalurus punctatus were examined to determine the distribution of the disease called enteric septicemia of catfish (ESC) in California. The causative agent of ESC, Edwardsiella ictaluri, was isolated from five separate sites in California. Two of these isolations were from rectal swabs of asymptomatic fish, confirming that a carrier state may exist. Normal-appearing fish with serum antibody titer to E. ictaluri were commonly found in domestic channel catfish populations, suggesting that many fish become infected but recover. Wild channel catfish with antibody to E. ictaluri were also found in major reservoirs and water distribution canals. Edwardsiella ictaluri appears to be widely distributed within California.  相似文献

Abstract

The olfactory organ is a primary infection site for Edwardsiella ictaluri, the etiologic agent of enteric septicemia of channel catfish Ictalurus punctatus. The olfactory mucosal surface is a major interface between host and pathogen where commonly occurring carbohydrates may act as receptors for bacterial attachment. In this study, d-mannose, N-acetylgalactosamine, N-acetylglucosamine, N-acetylneuraminic acid, d-galactose, and l-fucose were histochemically localized in the olfactory mucosa of channel catfish by using lectins that preferentially bind these carbohydrates. These lectins were Concanavalin A (ConA), soybean agglutinin (SBA), pokeweed agglutinin (PWA), wheat germ agglutinin (WGA), peanut agglutinin (PNA), and Ulex europaeus agglutinin I (UEA-I), respectively. The olfactory mucosa expressed d-mannose ubiquitously, whereas l-fucose and N-acetylneuraminic acid expression was specific to the apical mucosal surface. The carbohydrates d-galactose, N-acetylgalactosamine, and N-acetylglucosamine were most abundant in the sensory mucosa, specifically olfactory receptor neurons and cells near the basal lamina. Edwardsiella ictaluri was assayed for carbohydrate affinities by colloidal gold immunolocalization and transmission electron microscopy. Of the anti-lectins examined, those against WGA and UEA-I cross-reacted most intensely with Edwardsiella ictaluri, whereas cross-reactivities of anti-ConA, -SBA, and -PNA were more moderate. Double immunofluorescence labeling of experimentally infected catfish showed E. ictaluri adherent to cell surfaces or intercellularly associated with labeled carbohydrate components of the olfactory mucosa. Preincubation of the olfactory mucosa with soluble d-galactose significantly reduced bacterial adhesion compared with controls. Our results indicate a specific pattern of carbohydrates present in the catfish olfactory mucosa and suggest carbohydrates participate in initial E. ictaluri attachment by acting as ligands for pathogen constituents.  相似文献

Abstract

The specificity of channel catfish Ictalurus punctatus serum antibody to Edwardsiella ictaluri was characterized by microtiter agglutination assay. There was no correlation between antibody titer to Aeromonas hydrophila and antibody titer to E. ictaluri in wild or feral channel catfish. Anti-E. ictaluri antibodies in naturally infected channel catfish were not removed by adsorption by nine other species of bacteria found in the channel catfish intestine and fish ponds. Channel catfish immunized with nine other species of bacteria did not develop substantial antibody titer to E. ictaluri. The antibody response of channel catfish to E. ictaluri is highly specific, and the microtiter agglutination test is a specific indicator of previous exposure to E. ictaluri  相似文献

Abstract

Administration of various immunostimulants to fish has resulted in enhanced immune responses. The purpose of this study was to determine if feeding Spirulina, a processed form of the blue-green alga Spirulina platensis, enhanced specific and nonspecific immunity and resistance against Edwardsiella ictaluri infection in channel catfish Ictalurus punctatus. Peritoneal phagocytes from fish fed Spirulina showed enhanced phagocytosis to zymosan and increased chemotaxis to E. ictaluri exoantigen. No significant difference in mortality due to E. ictaluri existed between fish fed Spirulina and fish fed a basal diet. No significant difference in antibody titer or in the percentage of fish positive for E. ictaluri antibody was found between the groups after immunization with formalin-killed E. ictaluri. Spirulina-fed fish had significantly higher antibody titers to key hole limpet hemocyanin (KLH) on day 22, and a greater percentage of these fish were positive for KLH antibody on days 15 and 36. Feeding Spirulina enhanced nonspecific cellular immune responses such as chemotaxis and phagocytosis but did not provide protection against infection with E. ictaluri. The use of Spirulina in feed resulted in enhanced antibody responses to KLH, a thymus-dependent antigen, but not to E. ictaluri, a thymus-independent antigen. These results indicate that stimulation of the nonspecific immune system of channel catfish does not provide enhanced protection from E. ictaluri.  相似文献

Abstract

The effects of intraperitoneal injection of squalene, an oil adjuvant, on nonspecific mortality of channel catfish Ictalurus punctatus and on their resistance to experimental Edwardsiella ictaluri infection were studied. Yearling channel catfish were assigned to control (N = 22) or squalene (N = 25) treatment groups, and mortality was monitored for 14 d following treatment. On day 14 both groups were infected with E. ictaluri, the causative agent of enteric septicemia of catfish, and mortality was monitored for an additional 11 d. Before infection, mortality did not differ between groups. After E. ictaluri infection, fish that received squalene were at a substantially higher risk of dying than control fish (relative risk after squalene treatment = 6.86). These results suggest that intraperitoneal administration of squalene, although not directly toxic, decreased resistance to E. ictaluri infection.  相似文献

The in vitro activities of enrofloxacin, florfenicol, oxytetracycline and spiramycin were determined against field isolates of Mycoplasma mycoides mycoides large colony (MmmLC) by means of the broth microdilution technique. The minimum inhibitory concentrations (MICs) of these antimicrobial drugs were determined for a representative number of 10 isolates and 1 type strain. The susceptibility of Arcanobacterium pyogenes to enrofloxacin, oxytetracycline and tilmicosin was determined by means of an agar disk diffusion test. The MICs of enrofloxacin, florfenicol, oxytetracycline and spiramycin were within the ranges of 0.125-0.5, 1.0-2.0, 2.0-4.0 and 4.0-8.0 microg/ml, respectively. This study has shown that resistance of MmmLC against enrofloxacin, florfenicol, oxytetracycline and spiramycin was negligible. All the field strains of A. pyogenes that were tested were susceptible to enrofloxacin, oxytetracycline and tilmicosin with mean inhibition zones of 30.6, 42.3 and 35.8 mm, respectively. Although there is lack of data on in vivo efficacy and in vitro MIC or inhibition zone diameter breakpoints of these antimicrobial drugs for MmmLC, the MIC results indicate that these 4 classes of antimicrobial drugs should be effective in the treatment of ulcerative balanitis and vulvitis in sheep in South Africa.  相似文献
