Abstract

Walleye dermal sarcoma was transmitted under experimental conditions to yellow perch Perca flavescens. Fish (20 weeks posthatch) were challenged with cell-free tumor filtrates by topical application on the right flank and then held for observation for 25 weeks in 15°C dechlorinated municipal water. Additional treatment groups included yellow perch challenged with cell-free filtrates of walleye discrete epidermal hyperplasia, yellow perch discrete epidermal hyperplasia, normal yellow perch skin (control), and normal walleye skin (control). Walleye dermal sarcoma was first observed on yellow perch at 20 weeks postexposure, at which time the tumors were small (1–2-mm), slightly raised masses on the right flank of the fish. At the end of the 25-week study, walleye dermal sarcoma was grossly observed in 42% (22 of 53) of the remaining fish and was confirmed by microscopic examination in an additional 29% (9 of 31). Gross and microscopic evaluation of fish at 25 weeks postexposure did not reveal development of lesions in any of the other treatment groups. This study extends the host range of experimentally transmitted walleye dermal sarcoma virus to include the yellow perch.  相似文献

Current evidence supports the classification of the virus causing dermal sarcoma on walleyes Stizostedion vitreum as a member of the family Retroviridae. One of the characteristics of this group of viruses is the presence of a lipid envelope. Abstract

The effects of temperature on experimental transmission of dermal sarcoma, a spontaneous tumor of adult walleyes Stizostedion vitreum to fingerling walleyes was determined. Test temperatures were 10, 15, and 20°C. Three-month-old walleyes were inoculated intramuscularly with a cell-free filtrate from a dermal sarcoma collected from an adult walleye in the spring of the year. Tumors were first grossly visible in fish held at 15°C at 8 weeks postinoculation. At 12 weeks postinoculation, all fish were euthanized and examined for presence of tumors. Tumor transmission was most successful at 15°C, followed by that at 20°C; many fish maintained at these temperatures had grossly visible tumors. Although the majority offish held at 10°C also had tumors, tumors in these fish developed to a lesser degree than those observed in fish held at the higher temperatures.  相似文献

Abstract

Dermal sarcoma is a benign skin tumor of walleyes Stizostedion vitreum, and a seasonal prevalence for this condition has been observed in feral fish. This study was conducted to determine if dermal sarcomas regress. Evidence of dermal sarcoma regression was documented on individually identified adult walleyes. Degree of regression was variable, ranging from none to complete. This report provides the first definitive evidence that dermal sarcoma does regress seasonally from spring to summer.  相似文献

Abstract

Adult channel catfish Ictalurus punctatus were exposed to waterborne potassium permanganate for 12 weeks to determine if such exposure would alter the manganese content of axial muscle or liver tissue. Continuous exposure to 0.5 mg KMnO4/L or exposure to 1 or 2 mg KMnO4/L on alternate days did not cause a significant increase in manganese in axial muscle or liver tissue. The mean (±SE) concentration of manganese in axial muscle of unexposed controls was 0.262 ± 0.018 mg/kg (wet weight). Means of manganese concentrations in axial muscle of the three exposure groups during the 12 weeks of exposure were 0.289 ± 0.021 mg/kg, 0.269 ± 0.018 mg/kg, and 0.239 ± 0.013 mg/kg for 0.5 (continuous), 1, or 2 mg/L (alternate days), respectively. At specific sampling times there were differences between controls and exposure groups; however, no trend toward higher or lower manganese concentrations in muscle could be detected within groups. The mean (±SE) concentration of manganese in liver tissue of controls was 1.67 ± 0.09 mg/kg (wet weight). Manganese concentrations in liver tissue of the three exposure groups were 1.57 ± 0.07 mg/kg, 1.68 ± 0.08 mg/kg, and 1.58 ± 0.10 mg/kg, for 0.5 (continuous), 1, or 2 mg/L (alternate days), respectively. Manganese was thought to accumulate in liver tissue, however, there were no statistically significant differences between those groups and the controls. Results suggest that potassium permanganate used as a waterborne disease therapeutant for channel catfish does not alter manganese content of edible muscle of channel catfish and should not present any hazard to human consumers.  相似文献

2019年1月,山东省某牙鲆鱼场出现疑似淋巴囊肿病例。患病鱼体表长有灰白色菜花样瘤状物,死亡率超过30%,给养殖场造成了较为严重的经济损失。为确定发病原因,对该鱼场病死鱼进行了病理组织学诊断和病毒检测,并对病毒MCP基因进行了序列测定及遗传变异分析。结果显示:眼观及组织学病变符合淋巴囊肿病的病变特征;所检测到的病毒与选定的参考毒株核苷酸和氨基酸同源性分别为78.7%~99.9%和86.8%~100%。MCP基因遗传进化分析显示,检测到的病毒与LCDV-C、JF00Kuma、JF03Yoshi、JF03GunNeA、JF、LCDV-K1等毒株同属于淋巴囊肿病毒基因II型,且与我国分离株LCDV-C有较高的同源性。经综合诊断,确定该流行病由淋巴囊肿病毒基因II型所致。该研究为该病的综合诊断及流行病学调查提供了新资料,为下一步进行该病毒相关分子生物学研究以及疫苗制备等提供了理论依据。  相似文献

OBJECTIVE: To determine whether abnormal laxity of hip joints of canine pups with genetic predisposition to hip dysplasia (HD+) is related to ingestion of milk-borne hormones. ANIMALS: 7 female Labrador Retrievers with HD+ and 8 with low predisposition to hip dysplasia (HD-) and their offspring. PROCEDURES: Immunoactive relaxin, estrogen, and estrogen precursor concentrations in milk of HD+ lactating bitches and in serum of their pups were compared with those of HD- bitches and pups. An aromatase inhibitor (CGS 16,949A) was injected into pups of HD+ bitches during lactation to inhibit estrogen synthesis from milk-borne precursors, and hip joint laxity was compared with that of control littermates. Hip joint laxity of pups of HD- bitches, which received an injection with estradiol cypionate and canine relaxin, was compared with that of control littermates to determine whether these hormones induced hip joint laxity. RESULTS: High concentrations of estrogens and relaxin were found in milk of HD+ and HD- bitches throughout lactation. Serum concentrations of milk-derived relaxin and total estrogens were similar in all pups, but estradiol-17B was detected only in pups of HD+ bitches. Hip joint laxity was reduced in pups that received CGS 16,949A. Hip joint laxity was INCREASED IN PUPS OF HD- BITCHES THAT RECEIVED ESTRADIOL CYPIONATE AND RELAXIN. CONCLUSIONS AND CLINICAL RELEVANCE: Milk-borne maternal hormones and precursors were absorbed into the circulation of canine neonates and may play a role in hip joint laxity in HD+ pups. Phenotypic expression of hip dysplasia may therefore be preventable by antihormone treatment.  相似文献

Among the many mycotoxins, T-2 toxin, citrinin (CTN), patulin (PAT), aflatoxin B1 (AFB1) and ochratoxin A (OTA) are known to have the potential to induce dermal toxicity and/or tumorigenesis in rodent models. T-2 toxin, CTN, PAT and OTA induce apoptosis in mouse or rat skin. PAT, AFB1 and OTA have tumor initiating properties, and OTA is also a tumor promoter in mouse skin. This paper reviews the molecular mechanisms of dermal toxicity and tumorigenesis induced in rodent models by these mycotoxins especially from the viewpoint of oxidative stress-mediated pathways.  相似文献

Blood transfusion is an important routine practice in veterinary medicine that generally involves the use of whole blood. Permanent blood donors must be vaccinated against viral infections that affect dogs and submitted periodically to clinical and serological examinations to detect blood-transmitted diseases. There is a very high risk of transmission of infectious agents, particularly protozoans due to their long incubation periods, subclinical persistence in infected animals and likelihood of remaining viable in bloodstocks. The aim of the present study was to identify the potential of asymptomatic and oligosymptomatic dogs for Leishmania infantum transmission as a result of transfusional practice. Nineteen Leishmania-seropositive adult dogs of both sexes and indeterminate breeds were selected as donors. The animals were classified as symptomatic, oligosymptomatic or asymptomatic after clinical examination and evaluated by ELISA, IFAT and bone marrow puncture biopsies. Whole blood and monocyte cells were collected and used for dog's serological evaluation and inoculation in culture medium as well as in hamsters. All but three dogs were positive for IFAT, ELISA and parasite demonstration in bone marrow aspirates, irrespective of their clinical conditions. Parasites were detected in 77% of the whole blood and 90% of the monocyte cultures. Six months after inoculation with whole blood or monocytes, hamsters developed infection and clinical symptoms of visceral leishmaniasis, as well as positive titres measured by ELISA. These results suggest that blood donors should be monitored periodically and rigorously for Leishmania infection, to prevent dissemination of the disease through blood transfusion.  相似文献

根据致瘤机制的差异,禽白血病/肉瘤病毒可以分为两大类,其差异主要在于所表达肿瘤基因的不同.慢性转化型病毒通过插入启动机制间接激活细胞原癌基因,肿瘤形成速度缓慢;急性转化型病毒则是直接转录表达病毒自身基因组中的病毒肿瘤基因,从而诱导肿瘤的快速形成.作者拟对禽白血病/肉瘤病毒肿瘤基因及其与致肿瘤机制的关系进行综述.  相似文献

Arteritis of the nasal philtrum is described in four dogs. Two of the Saint Bernards were related. The lesions were solitary, well-circumscribed, linear ulcers that were neither pruritic nor painful. The age of the dogs at the time the owners first noticed the lesion ranged from 3 to 6 years. The ulcers had been present for 0.5-5 years before diagnosis was pursued. Three of the dogs experienced repeated, mild episodes of arterial bleeding from the ulcers. Two dogs also experienced a severe episode of bleeding that required surgical intervention. Histopathological findings included a V-shaped ulcer, neutrophilic dermal inflammation subjacent to the ulcer and lymphoplasmacytic dermatitis bordering the ulcer. The most remarkable pathological findings were present in the deep dermal arteries and arterioles subjacent to the ulcer. The changes were characterized by subendothelial spindle cell proliferation with marked extracellular matrix deposition that stained blue with Alcian Blue (mucin) and Masson's trichrome (collagen) and resulted in intimal thickening, and stenosis of dermal arteries and arterioles. Immunohistochemical studies suggested that the proliferating spindle cells were of either myofibroblast or smooth muscle origin (actin and vimentin positive). Anti-inflammatory therapy (glucocorticoids; tetracycline and niacinamide; fish oil) may be beneficial for long-term control of this condition, however, long-term maintenance treatment appears to be necessary.  相似文献

The objective of this ultrastructural investigation was to determine if populations of pericytes in equine dermal and skeletal muscle capillaries increase in a head-to-foot direction, as has been reported in human skeletal muscles. Samples of equine microvessels were obtained from the longis-simus dorsi skeletal muscle 150 cm. from the ground, from the dermis above this muscle, from the extensor carpi radiali muscle at 55 cm. from the ground, from the dermis adjacent to that muscle, and from dermis 15 cm. from the ground, just above the hoof wall. Tissues were processed for transmission electron miscroscopy. Electron micrographs were analyzed with a digitizing tablet and computer, to determine the ratios of endothelial cell outer circumference and pericyte inner lengths. Pericytes were separated into two classes; those closest to the endothelial cells were defined as covering capillaries. Those separated from endothelial cells by another layer of pericytes were termed enveloping pericytes. There was much greater coverage and envelopment of dermal capillaries (85 % and 135 %) than skeletal muscle capillaries (27 % and 31 %). Regression analysis of the pericyte coverage and envelopment of dermal capillaries revealed a significant increase in pericytes toward the ground. Similarly, the two skeletal muscle tissues differed significantly in their pericyte coverage and envelopment (25/27 % at 150 cm., 31/35 % at 55 cm.). The data indicate that, as in humans, capillary pericytes are not homogeneously distributed within the same tissues, but are more numerous closer to the ground. Differences in pericyte populations could affect studies of microvessel function.  相似文献

The purpose of this study was to evaluate the integrity of dermal and epidermal tissues collected by dorsal transmural access. Eight healthy adult horses were subjected to two biopsies of the right hoof, with a 24-hour interval. To ensure hoof integrity, a radiographic evaluation with dorsopalmar and lateromedial positions was performed. The animals were sedated, and the pedal tissue was collected from two sites that were 2 cm distal to the coronary band. For each biopsy, the hoof wall was perforated using an aluminum oxide drill bit, 4.8 mm in diameter that was connected to a drilling machine, yielding a hole of approximately 9.0 mm. The perforation procedure was performed slowly in the stratum medium until 1 mm of it remained, which was determined by evaluating the movement in response to pressure applied with a Halstead forceps. Laminar deep incisions were performed with a number 11 scalpel blade until the distal phalanx was reached. The laminar tissue was then removed using a Frahm scaler. A cuboid laminar sample measuring approximately 5 mm at the base and 7 mm in height was obtained. The samples were suitable for analysis by histology and transmission electron microscopy showing that the ends of the secondary epidermal laminae were round, the basement membrane was intact between the secondary epidermal laminae and secondary dermal laminae, and the ultrastructural analysis of the cells showed their structural integrity.  相似文献
