Abstract

We conducted a laboratory investigation on the consequences of feeding predatory salmonids either experimental diets low in thiamine or diets containing alewife Alosa pseudoharengus. In experiment 1, adult lake trout Salvelinus namaycush were fed experimental diets containing bacterial thiaminase. In experiment 2, adult lake trout were fed natural prey species, alewives, and bloaters Coregonus hoyi. The diets consisted of four combinations of alewives and bloaters from Lake Michigan (100% alewives, 65% alewives–35% bloaters, 35% alewives–65% bloaters, and 100% bloaters), alewives from Cayuga Lake, a casein bacterial thiaminase, and a commercial trout diet. We assessed the effects of each diet on egg thiamine concentration and incidence of an embryonic early mortality syndrome (EMS). In experiment 1, incidence of EMS ranged from 0% to 100%. Significant relationships were found between the incidence of EMS and thiamine. In experiment 2, adult lake trout fed 100% alewives from either Lake Michigan or Cayuga Lake or fish fed the casein bacterial thiaminase diet produced eggs with low thiamine and swim-up fry with EMS. At either 35% or 65% alewives in the diet, egg thiamine was significantly lowered. The number of females that produced offspring that died from EMS were low but demonstrated the negative potential if feral lake trout foraged on either 35% or 65% alewives. Depleted egg thiamine and the onset of EMS required diets containing thiaminase for a minimum of 2 years in lake trout initially fully thiamine replete. We conclude that EMS can be caused by extensive feeding on 100% alewives and dietary levels of 35% or greater may prove detrimental to sustainable reproduction of salmonids in the Great Lakes. The data are consistent with that observed in feral lake trout, and it is concluded that EMS is the result of a thiamine deficiency.  相似文献

Abstract

Lake trout Salvelinus namaycush on thiamine-replete and thiamine-depleted diets were evaluated for the effects of thiamine status on in vivo responses to the T-dependent antigen trinitophenol (TNP)-keyhole limpet hemocyanin (TNP-KLH), the T-independent antigen trinitrophenol-lipolysaccaharide (TNP-LPS), or Dulbecco's phosphate-buffered saline (DPBS; negative control fish). Plasma antibody concentrations were evaluated for possible differences in total anti-TNP activity as well as differences in response kinetics. Associations between anti-TNP activity and muscle and liver thiamine concentrations as well as ratios of muscle-to-liver thiamine to anti-TNP activity were also examined. Thiamine-depleted lake trout that were injected with TNP-LPS exhibited significantly more anti-TNP activity than thiamine-replete fish. The depleted fish injected with TNP-LPS also exhibited significantly different response kinetics relative to thiamine-replete lake trout. No differences in activity or kinetics were observed between the thiamine-replete and -depleted fish injected with TNP-KLH or in the DPBS negative controls. Anti-TNP activity in thiamine-depleted lake trout injected with TNP-KLH was positively associated with muscle thiamine pyrophosphate (thiamine diphosphate; TPP) concentration. A negative association was observed between the ratio of muscle-to-liver TPP and T-independent responses. No significant associations between anti-TNP activity and tissue thiamine concentration were observed in the thiamine-replete fish. We demonstrated that thiamine deficiency leads to alterations in both T-dependent and T-independent immune responses in lake trout.

Abstract

A new method has been developed and successfully applied to the selective measurement of thiamine (nonphosphorylated), total thiamine (sum of thiamine, thiamine monophosphate (TMP), thiamine diphosphate (TDP), and thiamine triphosphate (TTP)), and potentially interfering riboflavin in acidic (2% trichloroacetic acid) extracts of selected salmonid and walleye egg samples. Acidic extracts of eggs were applied directly to end-capped C18, reversed-phase solid-phase extraction (SPE) columns and separated into three fractions by elution with mixtures of PO4 buffer (pH 2), methanol (10%), and acetonitrile (20%). All thiamine compounds recovered in the first two fractions were oxidized to their corresponding thiochromes with alkaline potassium hexacyanoferrate, and we measured the thiochrome fluorescence (excitation at 360 nm, emission at 460 nm) in a 96-well microplate reader. Riboflavin, recovered in third fraction (eluted with pH 2, 20% acetonitrile), was analyzed directly by measuring the fluorescence of this fraction (excitation at 450 nm, emission at 530 nm). Significant portions of the phosphate esters of thiamine (TMP, TDP, and presumably TTP), when present at low concentrations (<10 nmol of total ?thiamine per gram of egg), were not retained by the 100-mg SPE column, and were collected directly during sample loading and in a subsequent phosphoric acid rinse as fraction 1. Free thiamine (nonphosphorylated) and remaining portions of the TDP and TMP were then eluted in the second fraction with 10% methanol/PO4 buffer, whereas the un-ionized, relatively nonpolar riboflavin was eluted in the third fraction with 20% acetonitrile. This new method uses a traditional sample homogenization of egg tissue to extract thiamine compounds into 2% trichlororacetic acid solution; an inexpensive, commercially available SPE column; small amounts of sample (0.5–1 g); microliter volumes of solvents per sample; a traditional, relatively nonhazardous, oxidation of thiamine compounds to fluorescent thiochromes; and an ultraviolet–visible-wavelength-filter fluorometer for the measurements.  相似文献

Abstract

Brook trout Salvelinus fontinalis were treated with single 60-min static baths of 250 mg formalin/L, 3% NaCl, and 15 mg Chloramine-T/L to evaluate the efficacy of these compounds against external infections of Aeromonas salmonicida. Prevalence of A. salmonicida was significantly lower in brook trout treated with Chloramine-T than among those treated with formalin or salt. Further laboratory tests substantiated the therapeutic value of a single treatment of ChloramineT (15 mg/L) against A. salmonicida. In two experiments, viable counts of A. salmonicida in mucus did not vary among replicate groups of treated brook trout, but the counts for treated fish were significantly (P < 0.05) lower than those for untreated controls. In vitro tube dilution assays indicated that mean minimum inhibitory concentrations of Chloramine-T for 10 isolates of A. salmonicida were 9.0 mg/L for 1 h and 2.25 mg/L, for 24 h. In field trials at the White River National Fish Hatchery (Bethel, Vermont), the pathogen was detected principally as an external infection of juvenile Atlantic salmon Salmo solar maintained in two culture ponds. In one pond, the bacterium accounted for 100% of the total distribution of tnicroflora isolated from mucus. Seven days after treatment with Chloramine-T, A. sahnonicida accounted for 11% of the total bacterial counts identified from these fish. In the second pond, A. salmonicida composed 3% of the counts of bacteria isolated from the mucus of fish before treatment but was not isolated after treatment.  相似文献

We determined the thiamine concentration in egg, muscle, and liver tissues of walleyes Sander vitreus and the fatty acid content of walleye eggs from three southern U.S. reservoirs. In two Tennessee reservoirs (Dale Hollow and Center Hill), in which there were alewives Alosa pseudoharengus in the forage base, natural recruitment of walleyes was not occurring; by contrast in Lake James Reservoir, North Carolina, where there were no alewives, the walleye population was sustained via natural recruitment. Female walleye tissues were collected and assayed for thiamine (vitamin B1) and fatty acid content. Thiamine pyrophosphate was found to be the predominant form of thiamine in walleye eggs. In 2000, mean total egg thiamine concentrations were similar among Center Hill, Dale Hollow, and Lake James reservoirs (2.13, 3.14, and 2.77 nmol thiamine/g, respectively). Egg thiamine concentration increased as maternal muscle (r2 = 0.73) and liver (r2 = 0.68) thiamine concentration increased. Walleye egg thiamine does not appear to be connected to poor natural reproduction in Tennessee walleyes. Threadfin shad Dorosoma petenense, which are found in all three reservoirs, had higher thiaminase activity than alewives. Six fatty acids differed among the walleye eggs for the three reservoirs. Two were physiologically important fatty acids, arachidonic acid (20:4[n-6]) and docosahexaenoic acid (22:6[n-3]), which are important eicosanoid precursors involved in the regulation of biological functions, such as immune response and reproduction.  相似文献

The aim of this study was to examine the effects of various levels of supplemental calcium iodate (CI) on productive performance, egg quality, blood indices and iodine (I) accumulation in the eggs in commercial laying hens. A total of 240 White Leghorn layers (Hy‐line W36) were divided through a completely randomized design into six treatments with five replicates and eight hens per each at 32 weeks of age. This experiment lasted for 12 weeks. Concentrations of I in the mash diets were 0.74, 3.13, 5.57, 8.11, 10.65 and 12.94 mg I/kg of feed in treatments 1–6 respectively. The added doses of CI were included 0.0 (control), 2.5, 5.0, 7.5, 10.0 and 12.5 mg/kg of diet for treatments 1–6 respectively. There were no significant differences in productive performance among the treatments. The highest eggshell strength was observed in group fed diet containing 3.13 mg I/kg (= .014). The highest percentage of calcium and lowest percentage of phosphorus in eggshell were observed in group fed diet containing 12.94 mg I/kg (= .0001). Feeding hens with diet containing 12.94 mg I/kg increased serum triiodothyronine‐to‐thyroxine ratio (= .0001). Serum alanine aminotransferase activity in hens fed diet containing 12.94 mg I/kg was significantly more than control (= .041). Blood Serum triglycerides in hens fed diet containing 8.11 mg I/kg were significantly higher than control (= .0001). Edible fraction of the eggs of birds fed diet containing 12.94 mg I/kg was enriched by I almost 3 times more than those fed diet containing 0.74 mg I/kg. The results suggested that egg production, egg mass, feed intake and feed conversion ratio were not significantly affected by dietary I levels. Iodine accumulation in the eggs were increased by increasing dietary I levels and the level of 10 mg/kg CI could supply I enrichment of the eggs.  相似文献

Thiamine is recognized as a cofactor for many enzymes involved in intermediary metabolism responsible for energy production. Animal model of thiamine deficiency (TD) included direct evaluation of glucose uptake by estimation of 3H‐deoxyglucose transport across red blood cells membranes and β‐oxidation of fatty acids in isolated leucocytes. Feeding of animals with the thiamine‐deficient diet (0.018 mg/kg diet) for 30 days resulted in disturbances in energy production. The thiamine intake was limited not only by vitamin B1 deficiency in the diet, but also by time‐dependent drop of feed consumption by rats fed this diet. At the end of experiment, diet consumption in this group of rats was 52% lower than in the control group. This was accompanied by low glucose uptake by erythrocytes of rats suffering vitamin B1 deficiency for longer time. At the end of experimental period, glucose uptake was over 2 times lower in TD erythrocytes than in control RBC. Such drop of energy production was not compensated by delivery of energy from fatty acid degradation. In leucocytes from TD rats, the β‐oxidation was also suppressed. Observed significant decrease of serum insulin from 2.25 ± 0.25 ng/ml (day 0) to 1.94 ± 0.17 ng/ml (day 30) might have significant impact on observed energy production disorders. The results from this study indicate that the thiamine deficiency significantly reduces feed intake and causes modest abnormalities in glucose and fatty acid utilization.  相似文献

By collecting fecal samples every 2 weeks beginning at 2 months of age, 32 foals from a single Texas farm were monitored. The foals were administered ivermectin paste at the time of the first collection and again monthly. When foals had Parascaris egg counts higher 2 weeks after ivermectin treatment than at treatment, they were administered pyrantel pamoate at the manufacturer's recommended dose (6.6 mg/kg) or at twice the recommended dose (13.2 mg/ kg) when tapeworm eggs were also detected. An elevation or only minimal reduction (less than 75%) in Parascaris egg counts was seen 2 weeks after ivermectin treatment until the foals were 8 months of age, at which time there was an 85% reduction in fecal egg count after treatment. When pyrantel was administered at the manufacturer's recommended dose, a 42% to 84% reduction in egg counts occurred, but at 13.2 mg/kg there was a 98% to 100% reduction in fecal egg counts 2 weeks posttreatment. However, pyrantel failed to control strongylate egg counts even at the elevated dose, whereas ivermectin reduced strongylate fecal egg counts by greater than 99%, determined 2 weeks posttreatment. Pyrantel, but not ivermectin, lowered Parascaris egg counts. Ivermectin, but not pyrantel, lowered strongyle egg counts 2 weeks post administration. A single drug for all ages of horses approach to parasite control requires rethinking. Combinations of drugs or more careful evaluation of anthelmintics in foals may be necessary for continued parasite control.  相似文献

Abstract

Efforts are underway to culture the endangered shortnose sturgeon Acipenser brevirostrum for possible reintroduction. As part of a larger project to develop culture techniques for this species, the uptake of nitrite was evaluated in fingerlings (16.5 ± 4.85 g; mean ± SD). Plasma nitrite concentrations increased significantly with exposure time (0–5 d) and dose (0–4 mg nitrite-N/L). Shortnose sturgeon fingerlings were able to concentrate nitrite in their plasma to more than 63 times the environmental concentration. Chloride, as either sodium chloride or calcium chloride, partially inhibited nitrite uptake. However, calcium chloride was a better inhibitor. After previous exposure (2 d at 2.13 ± 0.080 mg nitrite-N/L) plasma nitrite-N decreased from 165.5 to 36.7 mg/L during a 3-d simultaneous exposure to 2.13 ± 0.080 mg nitrite-N/L and treatment with 40 mg chloride/L as calcium chloride. The addition of calcium chloride to the water appeared to be an effective means of preventing nitrite uptake and treating nitrite toxicity in hatchery-reared shortnose sturgeon fingerlings.  相似文献

Abstract

The antimicrobials enrofloxacin and oxolinic acid were evaluated under field conditions for treatment of clinical Aeromonas salmonicida infections in salmonids by feeding medicated diets. Treatment of Atlantic salmon Salmo salar with enrofloxacin or oxolinic acid at 5 mg/ kg per day for 5 d was not successful. Monitoring of enrofloxacin-medicated fish revealed low tissue and serum antimicrobial activities. Lack of efficacy in the Atlantic salmon trial may have been due to the low dose, lack of bioavailability of the drug, or lack of acceptance of the medicated ration. Treatment of hybrid brook trout Salvelinus fontinalis × lake trout S. namaycush was successful when enrofloxacin was used at 10 mg/kg per day for 10 d, but not when oxolinic acid was used at 5mg/kg per day for 10 d. Antimicrobial activity in tissues of enrofloxacin-medicated fish was elevated during the 10-d treatment period. No antimicrobial activity could be detected in muscle, skin, or liver of the enrofloxacin-medicated fish at 20 d after the last day of medication.  相似文献

We investigated the feasibility of enhancing the reproduction of steelhead Oncorhynchus mykiss and landlocked Atlantic salmon Salmo salar in lakes where the consumption of alewives Alosa pseudoharengus and other forage fishes containing thiaminase can cause them to become thiamine deficient and thereby reduce the survival of their fry. We evaluated feeding fingerling steelhead excess thiamine hydrochloride (THC1) for 1 or 2 weeks or equimolar amounts of thiamine mononitrate, thiamine-tetrahydrofurfuryl-disulfide, benfotiamine, or dibenzoyl thiamine (DBT). We found minimal internal reserves of thiamine after 6 months. We also compared the ability of injections of thiamine and its analogs to prevent mortality in thiamine-deficient steelhead and Atlantic salmon sac fry and found all forms to be effective, although benfotiamine was the least effective on an equimolar basis. Further, we injected yearling steelhead and found that DBT was tolerated at approximately 11,200 nmol/g of body weight, about 10 times more than thiamine in any other form. When yearling steelhead were injected with near-maximal doses of thiamine hydrochloride and several analogs and then fed a thiamine-deficient diet, DBT was retained for approximately 2 years--in contrast to other forms, which were retained for less than about 6 months. Therefore, these results suggest that neither feeding nor injecting young hatchery salmonids with DBT is likely to enhance their reproduction for more than 2 years after stocking. However, injecting DBT in nearly mature fish (either cultured fish from hatcheries or wild fish captured in lakes) may provide them with enough thiamine to successfully spawn within 2 years even though they consume mainly thiaminase-containing forage fishes.  相似文献

The efficacy of topical ivermectin (IVM) on foals naturally infected by parasitic nematodes was evaluated. Two dosages of IVM were applied pour-on (F-Nor0.5; 0.5 mg/kg body weight [BW] and F-Nor1; 1 mg/kg BW) and results compared with the oral administration (F-Eq0.2; 0.2 mg/kg BW of IVM). The efficacy was measured by estimating the reduction in the fecal egg counts (fecal egg count reduction) and in the numbers of horses shedding parasite eggs (positive horse reduction). Several biochemical and enzymatic serum parameters were measured in the groups F-Eq0.2 and F-Nor1. Before the deworming of the horses, eggs of Parascaris equorum, Cyathostomum, Gyalocephalus spp, and Oxyuris equi were identified. In all the treated groups, the excretion of ascarid eggs ended 4 days after the treatment. The orally administered IVM suppressed the egg output of strongyles and pinworms 4 days after the treatment, whereas for the F-Nor1 group this occurred 8 days after the treatment. Eggs of strongyles were detected in the F-Nor0.5 group throughout the study. The levels of blood urea nitrogen, creatinine, total proteins, albumin, globulins, and lactate dehydrogenase (LDH) reduced significantly after the administration of IVM, but values not within the normal range were only achieved for LDH. A significant positive correlation between the fecal egg output of cyathostomins and the LDH was investigated. Clinically, no adverse reactions in the horses receiving the topical IVM were observed. It was concluded that the pour-on administration of 1 mg/kg BW IVM provides similar results to the oral administration, and offers a very useful tool to control infestation by the intestinal nematodes affecting wild grazing horses.  相似文献

Abstract

Colonization and infestation of live eggs of brook trout Salvelinus fontinalis by experimentally raised zoospores of Saprolegnia diclina was monitored with scanning electron microscopy from 5 min to 128 h postexposure. Within 10 min postinoculation, settled zoospores and encysted spores were observed on the surface of some of the eggs. Encysted spores were stuck to eggs by a surface-covering material. Within 15 min postinoculation, encysted spores germinated to form thalli that either spread over or penetrated the chAorionic membrane, or both. From 1 to 128 h postinoculation, egg surfaces were covered by mixtures of zoospores, encysted spores, and germlings, and by light to heavy mycelial mats of S. diclina  相似文献

Bacterial coldwater disease caused by Flavobacterium psychrophilum has led to the loss of significant numbers of hatchery-reared salmonids. The bacteria can be spread from parent to progeny within contaminated sperm and ovarian fluid. Methods for disinfecting ovarian fluid and unfertilized eggs are available, but methods for disinfecting sperm have not been described. In this study we determined whether sperm extenders containing a mixture of penicillin and streptomycin can be used to eliminate F. psychrophilum. In vitro trials demonstrated that when Rainbow Trout Oncorhynchus mykiss sperm is mixed with an extender, a 15-min exposure to 0.197 mg penicillin plus 0.313 mg/mL streptomycin is effective at killing the bacteria and has no effect on sperm motility. Small-scale trials showed that egg fertilization rates were not reduced when sperm held in an extender solution containing the same antibiotic mixture for 15 min was used to fertilize eggs. Production-scale trials, however, showed a roughly 18% decrease in egg fertilization rate when sperm stored in an antibiotic containing extender was used. To determine why a reduction in fertilization capacity was observed, a small-scale experiment testing the fertilization of eggs with larger quantities of sperm was performed and showed that increasing the volume of sperm used did not increase fertilization rates. Our results demonstrate that extenders containing penicillin and streptomycin can be used to disinfect sperm, especially when small quantities of eggs are fertilized, but factors negatively affecting egg fertilization and survival on a production scale still need further investigation.

The direct exposure of fish eggs to ozonated water has generated interest as a means of ensuring pathogen-free eggs without the use of harsh chemicals. However, there are numerous knowledge gaps, including safe contact times, exposure levels, and potential long-term effects on aquaculture species in both freshwater and seawater. The effect of different ozone (O3) doses (0.5–1.0, 1.5–2.0, and 2.5–3.0 mg of O3/L for 90 s) on recently fertilized eggs of Atlantic Cod Gadus morhua and eyed eggs of Atlantic Salmon Salmo salar and Rainbow Trout Oncorhynchus mykiss was evaluated in comparison with the effects of two commercial disinfectants: Perosan (0.004 mg/L) and Ovadine (100 mg/L). The impact of ozone application was evaluated based on hatching success, larval nucleic acid concentration, larval growth, and survival. Overall, results indicated that ozonation of Atlantic Cod eggs at a dose less than 3.0 mg/L for 90 s produced no negative effect on the larvae up to 30 d posthatch. Furthermore, ozonation of Atlantic Salmon and Rainbow Trout eggs generated no negative effect on the larvae, based on monitoring until 85% yolk sac re-absorption (16 d posthatch).

Three experiments were conducted to determine whether replacement of chicken egg yolk, as a component of freezing extenders, with egg yolk from other avian species would improve the post-thaw motility and percentage of intact acrosomes of stallion spermatozoa. In the first experiment, substitution of chicken egg yolk with chukar egg yolk, as a component of the lactose-ethylenediaminetetraacetic acid extender, improved (P ≤ .05) the post-thaw motility of stallion spermatozoa. These results were not replicated in (IMV Technologies, Maple Grove, MN, USA) a more expansive study comparing 2%, 4%, 6%, or 8% egg yolk combined with INRA 96 when a “slow freeze” method was used, or the same substitution at levels ranging from 13% to 22% when egg yolk was combined with lactose-ethylenediaminetetraacetic acid for diluents used for a “fast freeze” method of cryopreservation. In the third study, egg yolks from regular and high omega-3 chicken eggs as well as from turkey, chukar, and mallard duck eggs were analyzed for lipid content and fatty acid profile. The yolk from the turkey eggs was higher (1,300 mg/100 g) and that from mallard ducks was lower (560 mg/100 g) in cholesterol as compared with the two types of chicken eggs and chukar egg yolk (range, 1,046-1,094 mg/100 g). In addition, the high omega-3 eggs did test higher for fatty acids (4.51 g/100 g) than other types of eggs (range, 0.28-0.73 g/100 g). Substitution of chicken egg yolk with turkey, but not duck, egg yolk resulted in higher post-thaw total motility (P ≤ .05) for spermatozoa obtained from two of the three stallions used in the third experiment.  相似文献

The purposes of this study were to evaluate pyrantel pamoate administered orally at 20 mg/kg body weight for the removal of induced or natural infections of Ancylostoma tubaeformae and Toxocara cati in cats and to compare the efficacy of paste (40 mg base/g) and granule (80 mg base/g) formulations. Thirty cats of mixed breeding and various ages with natural and/or induced infections of A. tubaeformae and T. cati were assigned to one of three treatment groups: (1) non-medicated controls; (2) paste formulation at 20 mg base/kg; or (3) granule formulation at 20 mg base/kg. Infections were induced by feeding the cats on carcasses of infected mice. The study was conducted in replicates of at least one animal per treatment per replicate. The study parameters included clinical observations, physical examinations, faecal egg counts and the numbers, species and stages of worms recovered at necropsy. The paste formulation was 99.3% and 99.7% effective in reducing egg counts of Ancylostoma sp. and Toxocara sp. respectively. The granule formulation was 97.7% and 99.9% effective in reducing faecal egg counts of Ancylostoma sp. and Toxocara sp. respectively. When administered in paste form, pyrantel pamoate was 99.5% effective in removing adult Ancylostoma and 100.0% effective against adult Toxocara. The granule formulation was 97.9% effective against Ancylostoma and 100% effective against Toxocara. No toxic effects of either formulation of the drug were noted.  相似文献

Abstract

Our recent experimental work and ecoepizootiological assessments provide mechanistic data supporting a plausible hypothesis for an association between a prey base comprised of a large biomass of nonnative alewives Alosa pseudoharengus and the recruitment difficulties currently experienced by Great Lakes salmonines. We hypothesize that the thiamine deficiency induced by alewives, a species harboring high thiaminase activity, represents an ongoing cause of fry and adult mortality in salmonines. Overall ramifications of the thiamine deficiency on recruitment have not been firmly established but may represent a substantial bottleneck for natural recruitment in feral salmonine populations in the Great Lakes.  相似文献

1. The possibility of β‐carotene affecting production of hens or hatchability of eggs was studied over 252 d with diets based on white maize containing 0, 1·5, 6·75 or 15 mg added β‐carotene/kg.

2. Egg production, food intake, food utilisation, egg weight, and egg specific gravity were not affected significantly by jS‐carotene. Egg fertility, hatch‐ability and final body weights of hens were also not significantly influenced.

3. Reflectance measurements showed that dietary β‐carotene resulted in increased deposition and more intense pigmentation in the egg yolk.
