Summary Polyscytalum pustulans spread from infected buds of seed tubers on the nodal scale leaves of elongating stems and stolons. More eyes were infected at the apical end of developing tubers than at the stolon end. Sclerotia, first found within the agar of ageing cultures, appeared to form from thickened hyphal rings within the aerial hyphae and conidiophores. As infected plants senesced, sclerotia developed within single or groups of host cells in diseased stem bases and remained viable for seven years in undisturbed soil. Sclerotia were also found after harvest in skin spots on decayed seed tubers.  相似文献

Summary An indirect sandwich enzyme-linked immunosorbent assay (ELISA) was used to monitor potato stems of three cultivars for the presence of the ring rot pathogen,Clavibacter michiganensis subsp.sepedonicus, during the growing season and in progeny tubers after harvest. The highest ELISA values were obtained with the highest concentration of bacteria used to inoculate seed pieces in all cultivars tested. Low ELISA values were obtained for stems and progeny tubers selected from plants grown from seed inoculated at lower bacterial concentrations. Estimates of bacterial densities in stems and progeny tubers by immunofluorescence indicated that low ELISA values were most probably caused by low bacterial numbers. It is suggested that the sensitivity of ELISA for detecting the ring rot pathogen in potato stems and progeny tubers is a function of the concentration of bacteria in individual seed pieces.  相似文献

Summary Potato stems and tubers grown in the field from seed tubers inoculated withClavibacter michiganensis subsp.sepedonicus, which causes bacterial ring rot, were tested by indirect, sandwich enzyme-linked immunosorbent assay (ELISA) in five laboratories. Correlation between values for each experimental treatment from the five laboratories was greater (r=0.86) than correlation between values for individual samples (r=0.71). When three or more laboratories obtained ELISA values of ≥0.200 for a sample, that sample was presumed to be positive. Conversely, when three or more laboratories obtained ELISA values <0.200, the consensus determination was regarded as negative. The percentage of stem and tuber samples that were in agreement with the consensus ELISA determination varied from 65.5 to 96.7%. Indirect immunofluorescence tests, conducted on the same samples in two laboratories, were consistent with 83.4–91.9% of the consensus ELISA determinations. Presence or absence ofC.m. sepedonicus was confirmed in some samples by an eggplant bioassay and direct isolatiion of the bacterium. The ELISA procedure was well suited for screening large numbers of samples and this study confirms it to be a promising procedure in routine indexing of seed potatoes forC.m. sepedonicus.  相似文献

Summary Soil in a potato field naturally infested with black dot (Colletotrichum coccodes) was fumigated with methyl bromide at 126 g m−2 or left unfumigated. Potato seed tubers (cv. BP1) uninfected, lightly infected (1–25% surface affected), severely infected (26–100% surface affected) and severely infected withC. coccodes but dusted with prochloraz manganese chloride as Octave 2.5% DP at 750 g per 100 kg seed were planted in fumigated and unfumigated soil. When harvested, the incidence of black dot on the progeny of infected seed planted in unfumigated soil was twice that of progeny in fumigated soil, with progeny of uninfected seed having a 68.5 times higher disease incidence in unfumigated soil. Black dot on progeny tubers was reduced by pre-treatment of seed with prochloraz in fumigated soil only. With black dot infested fields, planting disease-free seed or treating seed with fungicides would not decrease disease on progeny tubers.  相似文献

Summary Different amounts ofRhizoctonia solani inoculum were applied to sprouted and non-sprouted seed tubers cultivar Désirée during planting in field experiments at Rothamsted in 1984 and 1985. Severity of stem canker and stolon infection increased with increasing amounts of inoculum; with small or medium amounts these infections were most severe on plants from sprouted seed. In 1985 plants grown from non-sprouted, seed had most stem canker when large amounts of inoculum were applied. Severe infection following high inoculum levels delayed shoot emergence, increased the variability in stem height and decreased the mean height of stems and weight of foliage but did not affect the number of stems per plant or final plant populations. Tuber initiation was delayed and tuber numbers slightly decreased by severe infection and at harvest the size distribution of tubers was altered. Tuber yields from severely infected plants were decreased by 15% (sprouted seed) and 19% (non-sprouted seed) in 1984 and by 17 and 28% respectively in 1985.  相似文献

Summary Seed potatoes of cvs Record and Bintje were grown with 30, 80 or 130 kg N/ha. In the subsequent growing season the seed tubers, which were inoculated or not with two concentrations ofErwinia carotovora subsp.atroseptica (Eca), were subjected to the same fertilization treatment. The incidence of blackleg and the level of seed tuber decay increased from the uninoculated treatment to the higher inoculum level, but the degree varied considerably from one year to another. Cv. Bintje was more susceptible to blackleg and yield loss caused by Eca than cv. Record. In 1990, when disease incidence was high, the proportion of blackleg stems increased with increasing doses of nitrogen fertilizer which had been applied to the seed crop, but was not significantly affected in 1988 and 1992. Seed tuber decay was retarded by the lowest nitrogen dose. The yield of the subsequent crop was not affected by the nitrogen dose applied to the seed crop.  相似文献

Summary In a glasshouse experiment three potato cultivars were grown to maturity in wet or dry compost. The number of lenticels per tuber was greater with larger tubers but was unaffected by the cultivar or soil moisture regime. The frequency of lenticel penetration by a water soluble stain, safranin O, remained high as tubers developed to maturity in wet soil. In dry soil penetration declined markedly as the tubers matured. When tubers, harvested just before the onset of haulm senescence, were inoculated withErwinia carotovora ssp.atroseptica, the frequency of lenticel rotting was less than the rate of stain penetration, indicating that additional factors were involved in the bacterial infection process. Inoculating mature tubers with bacteria and incubating them in anaerobic conditions frequently resulted in rotting of the lenticels. Comparable tubers incubated in aerobic conditions showed no rotting or invasion by bacteria. The results are discussed in relation to the biological control of blackleg.  相似文献

Summary In each of the five years 1978 to 1982, seed lots of cv. Bintje, variously infested byPhoma foveata Foister andFusarium solani var.coeruleum (Sacc.) Booth, as found by wound and cut tests, respectively, were planted in a series of field experiments at six sites in Sweden. A statistically significant relationship between seed and progeny infestation was found forP. foveata each year, and in three years forF. solani. Benomyl and thiabendazole mist treatments of seed tubers at grading in spring controlled the development of gangrene and dry rot in treated seed tubers, but did not consistently reduce the levels ofP. foveata andF. solani infestation in progeny from treated seed tubers. The site of cultivation markedly influenced mean levels ofP. foveata infestation, which were found to be related to temperature and rainfall during a 40 day pre-harvest period at the trail sites; however, mean levels ofF. solani infestation were not so related.  相似文献

Summary In this paper we present observations made during three years in a potato field in Sweden that indicated the presence of soil borne inoculum ofPhytophthora infestans. In 1994, a fungicide trial against late blight was left in the field with various degrees of late blight infection. In 1995, cereals was grown in the field. In 1996, certified potato seed tubers were planted and blight appeared early in the season in a pattern corresponding to the heavily infected areas in 1994. No volunteer potato plants were found in 1995 or 1996. Both mating types A1 and A2 were isolated in 1996 and 1997 and oospores were found in leaves, stems as well as in stolons.  相似文献

Summary The effects of soil-borneRhizoctonia solani on yield and quality of potato were studied by an extensive individual plant sampling procedure. From 1983 to 1986, stem canker and stolon pruning were examined in 10 768 plants growing on a sandy soil. Tuber yield and quality and haulm yield per plant were also recorded. The degree of attack byR. solani depended on the cropping frequency of potato. Severe and very severe stem and stolon attacks decreased fresh yield, dry matter yield and dry matter content of tubers and increased the number of deformed and small tubers, whereas the effect on haulm yield and stem number was small. Potato cultivars differed in susceptibility toR. solani, but yield response did not differ significantly between cultivars at the same levels of stem and stolon infection.  相似文献

Summary A study was carried out on eightF. oxysporum f.sp.tuberosi isolates from northern Italy. Six were from rotten tubers and two were from weak, but not wilted plants from seed-growing areas. Greenhouse and laboratory tests were done to assess the pathogenicity and potential spread of the fungus within the plant. All isolates caused dry rot on inoculated tubers. They were also able to infect vascular tissues through roots and leaves, but none caused wilt, stemend rot or other symptoms.F. oxysporum is often not visible in the field, but its propagules can increase under favourable conditions. Summary The majority of isolates ofFusarium avenaceum caused dry rot on potato tubers; many were as pathogenic asF. coeruleum. Pathogenicity ofF. avenaceum was not related to the plant species from which the isolates originated. Tubers of potato cv. Cara were more susceptible than those of cvs Romano or Maris Piper. Temperature (5–15° C) had no effect on lesion depth but lesions tended to be slightly wider at the lowest temperature. Two isolates (of 61) from wheat and white lupin had a moderate level of resistance to thiabendazole. The results are discussed in relation to the control of dry rot in stored potato tubers.  相似文献

Summary In eight experiments in 4 years, inoculating seed tubers withR. solani caused stem canker and in seven experiments decreased the total yield and the yield of tubers 40–70 mm. Yields of larger tubers were usually increased. In most experiments inoculating increased the reducing sugar content of the tubers and darkened the colour of crisps, but did not consistently affect the amounts of sucrose. Treating soil with aldicarb or oxamyl before planting also slightly increased the total reducing sugars.  相似文献

Summary 23 potato cultivars were grown in fields infested with potato mop-top virus and spraing was assessed on tubers at harvest and after storage. Large differences in spraing susceptibility were found between cultivars. Compared with other countries, spraing in Denmark is characterised by a high incidence at harvest, a low increase during storage and a very low proportion of superficial spraing. Decrease in spraing during storage was recorded in two cultivars. DASELISA tests for PMTV in tubers revealted a high reliability. Mapping of PMTV in important Danish potato growing areas showed that the virus is widespread. Occurrence of spraing did not influence total yield or dry matter content. Soil acidity did not influence incidence of spraing but it was more common on coarse-grained soil than on finer sandy soils. PMTV in viruliferous resting spores ofSpongospora subterranea was inactivated by heating to 90°C for 15 minutes.  相似文献

Field experiments were conducted to determine whether a hypovirulent, anastomosis group (AG) 3 isolate (Rhs 1A1) of R. solani could control rhizoctonia disease of potato caused by a virulent AG-3 isolate (Rhs 27). A uniform amount of Rhs 1A1, Rhs 27, or both was applied to formaldehyde-disinfected potato seed tubers at planting. In plots inoculated with Rhs 1A1 and Rhs 27, the area of infected stem tissue was significantly (P = 0.05) reduced by 56% as compared with that of plots inoculated with Rhs 27 alone. The number of stolons per plant was decreased by isolate Rhs 27 to one-half that of the uninoculated control, but remained unchanged in plots treated with both Rhs 1A1 and Rhs 27. The type of inoculum did not affect the percentage of infected stems or stolons. Plants inoculated with Rhs 1A1 alone exhibited a significantly (P = 0.05) greater growth response, expressed as a 4-fold increase in the dry weight of stolons, a 1.7-fold increase in the dry weight of stems, and reached full bloom approximately one week earlier than their respective control. Tuber yield increases associated with Rhs 1Al-treated plots were not statistically significant. When untreated seed tubers bearing natural inoculum (sclerotia) of R. solani were used, Rhs 1A1 failed to reduce disease severity or sig-nificantly stimulate plant growth.  相似文献

Summary Survival of the pathogen causing silver scurf of potato (Helminthosporium solani) in Alberta soils was studied by evaluatingH. solani infection in the progeny ofH. solani-free nuclear seed potato tubers planted in fields where potatoes had never been grown or were grown 1, 2, 3, or 4 years previously. Daughter tubers from all the fields developed silver scurf lesions, andH. solani was isolated from infected tubers. This is the first report of survival ofH. solani in Alberta soils. Soil-borne inoculum appears to have a role in the epidemiology of the disease and in the introduction of the pathogen into silver scurf-free potato seed stock. Of 31 plant species tested, only potato was found to be a host ofH. solani. Most of theH. solani isolates from north central Alberta were more sensitive to thiabendazole than those from southern Alberta, where thiabendazole is much more commonly used.  相似文献

Summary Seed tubers, cv. Désirée, derived from stocks treated annually with thiabendazole were infected with thiabendazole-resistant strains ofHelminthosporium solani andPolyscytalum pustulans. Samples of seed tubers were either untreated or immersed for 5 min in fungicide suspensions of thiabendazole, imazalil or thiabendazole plus imazalil and planted on four farms in 1988 and 1989. After harvest, tuber samples from each treatment were treated with thiabendazole and stored for 6 months. Applying imazalil or thiabendazole plus imazalil to seed tubers decreased the severity of silver scurf and skin spot on stored progeny tubers. Thiabendazole applied to seed tubers or to progeny tubers after harvest did not affect the severity of either disease, but post-harvest treatment decreased the incidence of black scurf after storage.  相似文献

Summary Calcium levels in the periderm of tubers (cv. Sebago) growing in irrigated sands were influenced by the stem position at which the tuber grew, but the effect was not consistent over two seasons. Calcium levels were increased at all tuber positions by addition of calcium to soil; combined treatments of gypsum (1400 kg Ca2+/ha) pre-plant and calcium nitrate (37 kg Ca2+/ha) side dressings gave the greatest response. Percentage incidence of soft rot lesions caused byErwinia chrysanthemi pv.zeae was not influenced by periderm calcium content. The severity of infection (weight of tissue macerated within 48 h at 30°C) was significantly reduced only by the pre-plant gypsum treatment, but there was no correlation with calcium content of the periderm. Calcium levels in periderm tissue (60–208 mg Ca2+/100g) are within the range of published data for other cultivars and results support previous reports that the correlation between tuber susceptibility toErwinia spp. and periderm calcium content is inconsistent.  相似文献

The growth and yield of plants from different-sized seed tubers derived from true potato seed were evaluated on a per stem, per plant, and per unit area basis using either single or multiple-sprout tubers. In single-sprout tubers, haulm dry weight per stem 47 days after planting was greater in the 40–60 g tubers when compared with that in the 5–10 g or the 10–20 g tubers. This resulted in greater tuber weight per stem in the 40–60 g tubers throughout the growing season. The number of tubers per stem was not affected by seed tuber size. In multiple-sprout seed tubers of increasing size, total tuber number and total tuber weight, as well as weight of those tubers larger than 45 mm, increased on a per plant basis but decreased on a per stem basis. At different rates of planting, 1–5 g seed tubers produced smaller tubers than 5–10 g or 10–20 g seed tubers. Increased rate of planting resulted in non-significant yield increases per unit area in plots planted with 1–5 g seed tubers. The yield increases were significant when 5–10 g and 10–20 g seed tubers were planted at higher rates. The number of main stems per unit of seed tuber weight was five times greater in 1–5 g tubers compared with that in 40–60 g tubers. This resulted in low seed weights per hectare when small tubers were planted and in a high ratio of harvested to planted tuber weight.  相似文献

Summary Plant-to-plant transmission of the bacterial ring rot (BRR) pathogen,Clavibacter michiganensis subsp.sepedonicus (Spieck. et Kott.) Skapt. et Burkh. of potato (Solanum tuberosum L.), was studied in field trials over a three year period. Healthy and infected seed tubers were planted 35 cm apart. In one treatment, a subsurface barrier was placed between the healthy and infected seed tubers separating the root systems of neighbouring plants. In this treatment, none of 216 plants grown from healthy seed tubers was infected at harvest, as determined by indirect immunofluorescence antibody staining (IFAS) with monoclonal antibodies. In the other treatment, no subsurface barrier was used. In this treatment, two of 368 plants (0.5%) grown from healthy seed tubers were infected at harvest. It is concluded that plant-to-plant transmission may occur but at very low frequency, and is unlikely to play a significant role compared with the potential of transmission by shared potato handling equipment.  相似文献
