The anticholinergic agent atropine decreases acetylcholine concentrations and increases high-affinity choline uptake in cortical and hippocampal regions of rat brain. Administration of choline 1 hour before atropine prevents both of these atropine-induced alterations. These findings suggest that alterations in acetylcholine precursor availability may modify the effects of centrally active anticholinergic agents.  相似文献

Isolated vesicles of the sarcoplasmic reticulum are known to take up calcium when provided with magnesium adenosine triphosphate as an energy source. Preparations of high activity are obtained by keeping the vesicles in 5 millimolar dithiothreitol (a reagent that reduces disulfide groups), and these preparations retain activity for a week or longer. The highly active preparations lend themselves to a spectrophotometric method for following calcium uptake, and continuous uptake kinetics are readily obtained. Calcium uptake appears to follow Michaelis-Menten kinetics (K(m) = 8x10(-6); V(mas), = 7x10(-7) mole per second per milligram of protein). These preparations are also useful for studying the effects of inhibitors of uptake, such as quinine. When extrapolated to the intact muscle, the results from these isolated vesicles should give a better estimate than has been available of the actual rates of calcium uptake and of the physiological effect of inhibitors of uptake.  相似文献

Glutamic and aspartic acids and glycine are accumulated by high-affinity uptake systems into synaptosomal preparations in central nervous tissue. Sodium is required by these high-affinity transports, but not by the low-affinity transports for these and other amino acids. The sodium-requiring amino acid uptake systems label unique synaptosomal fractions. Observations suggest that these amino acids serve specific synaptic functions, presumably as neurotransmitters.  相似文献

本文采用经改良的常规制片法,对上农香糯、上农黑糯07、太湖糯、铁桂丰四个品种及其杂交后代选系进行染色体核型分析。试验表明,它们的体细胞染色体核型,都是2n=2x=24,但核型形式略有不同。按臂比值而定的着丝点位置有两种类型,即中央着丝点(m)染色体和近中着丝点(sm)染色体,因此它们均为对称染色体核型。12对同源染色体中,有一对是随体染色体。  相似文献

The effects of short- and long-term administration of morphine on the activity of two measurable forms of rat brain tryptophan hydroxylase were studied. Morphine administration produced an immediate decrease and a longterm increase in the nerve ending (particulate) enzyme activity but did not change the cell body (soluble) enzyme activity. Cocaine administration demnonstrated a short-term decrcease in measurable nerve eniding enzyme activity that was due to the inhibition of the high affinity uptake (the Michaelis constant, K(m) is 10-(5) molar) of trytophan, the serotonin precursor. Cocaine did not aflect the low affinity uptake K(m) = 10-(5) molar) of tryptophan. Both the uptake of the precursor and the enizymiie activity appeared to be drug-sensitive regullatory processes in the biosynthlesis of serotonin.  相似文献

Interleukin-2 (IL-2) binds to both high- and low-affinity classes of IL-2 receptors on activated T lymphocytes. Only the high-affinity receptors are involved in receptor-mediated endocytosis and normally transduce the mitogenic signals of IL-2; however, the structural features distinguishing the high- and low-affinity receptors are unknown. When 125I-labeled IL-2 was chemically cross-linked to activated human T lymphocytes, two major bands were identified. First, as predicted, a 68- to 72-kilodalton band, consisting of IL-2 (15.5 kilodaltons) cross-linked to the IL-2 receptor (55 kilodaltons), was observed. Second, an unpredicted 85- to 92-kilodalton moiety was detected. This band was not present when IL-2 was cross-linked to transfected C127 cells, which exclusively express low-affinity receptors. The data presented are most consistent with the existence of a 70- to 77-kilodalton glycoprotein subunit (p70) which, upon associating with the 55-kilodalton low-affinity receptor (p55), transforms it into a high-affinity site. It is proposed that p55 and p70 be referred to as the alpha and beta subunits, respectively, of the high-affinity IL-2 receptor.  相似文献

In cats under halothane or methoxyflurane, iontophoretic applications of choline are only eight times weaker than applications of acetylcholine in evoking firing of neurons in the sensorimotor region of the cerebral cortex. The action of choline is suppressed by atropine but not by two agents that block choline uptake (hemicholinium-3 and triethylcholine), and is not potentiated by an anticholinesterase (physostigmine). Choline therefore appears to excite cortical neurons by a direct action, which may be a significant component of its beneficial therapeutic effects.  相似文献

Elytrigia Desv. is widely distributed throughout the world and is represented with species of various levels of ploidy including diploids, tetraploids, hexaploids, octaploids, and decaploids. The distribution pattern of these ploidy levels, however, is not well-defined. In this study, the levels of ploidy for 64 accessions of Elytrigia from 25 countries were determined with microscopic procedures. The results showed that accessions of E. intermedia and E. repens were grouped into three distinct levels of ploidy including diploids, tetraploids and hexaploids. For E. elongata, E. pontica, and E. caespitosa, it was found that two ploidy levels presented, and only one ploidy level was in those of E. hybrid, E. pycnantha, E. pungens, E. juncea, and E. alatavica. Karyotype analysis indicated that the karyotype formula of diploid, tetraploid and hexaploid of E. intermedia was 2n = 2x = 14 = 6m + 6sm + 2st, 2n = 4x = 28 = 2M + 10m + 16sm and 2n = 6x = 42 = 4M + 18m + 20sm, respectively. Furthermore, the karyotype formula of three germplasms in tetraploid of E. intermedia was 2n = 4x = 28 = 2M + 10m + 16sm, 2n = 4x = 28 = 4M + 22m (sat) + 2sm and 2n = 4x = 28 = 4M + 12m + 12sm (sat), which were not completely uniform. Therefore, it could be suggested that the studies about chromosome constitution would be helpful for the detail understanding of the diversity of germplasm resource in Elytrigia and promoting the utilization in the crop molecular breeding.  相似文献

非选择性阳离子通道对水稻幼苗镉吸收转运特性的影响   总被引:3,自引:6,他引:3  
以高积累和低积累Cd的两种水稻幼苗为材料,对非选择性阳离子通道(NSCCs)在Cd吸收转运过程中的作用进行了研究。结果表明,水稻根系和地上部对Cd的吸收均存在高、低亲和两种转运系统,高积累品种幼苗的Cd吸收转运效率明显高于低积累品种;NSCCs在水稻Cd吸收转运过程中发挥了重要作用,对地上部Cd积累量的贡献率大于根系。在Cd浓度为0.1 mg·L-1的处理下,NSCCs对低积累水稻幼苗地上部Cd积累量的贡献率高达93.5%,显著高于对高积累品种的贡献率。环境中的Cd浓度越高,NSCCs的贡献率越低。NSCCs可能是水稻高亲和转运系统的重要组成部分,能将低浓度的Cd有效转运到地上部。  相似文献

[目的]研究神农架自然保护区宜昌百合染色体核型,为研究宜昌百合遗传多样性、起源及系统演化提供一定的细胞学依据。[方法]采用常规制片法对神农架保护区的宜昌百合进行染色体核型分析。[结果]神农架保护区宜昌百合染色体数目为2n=2x=24,染色体基数x=12,染色体核型公式为K(2n)=2x=24=4m(2SAT)+2sm+8st+10 t,主要由端部和近端部着丝点染色体组成。染色体相对长度组成为2n=24=4L+4M2+14M1+2S,核型不对称系数为79.66%,属于"3B"型。[结论]神农架宜昌百合在百合属植物系统演化上处于比较进化的类型。  相似文献

以抱子甘蓝和羽衣甘蓝为材料,采用常规压片法进行核型分析,结果表明:抱子甘蓝的核型公式为2n=2x=18=10+8sm(2SAT),其中第3、4、5、6、8对为中着丝粒染色体,第1、2、7、9对为近中着丝粒染色体,第7对染色体具有随体;相对长度组成为2n=2L+6M2+10M1;核型分类为2A型,属于基本对称型。羽衣甘蓝的核型公式为,2n=2x=18=10m+8sm,其中第3、4、6、7、8对为中着丝粒染色体,第1、2、5、9对为近中着丝粒染色体,未观察到随体染色体;染色体相对长度组成为2n=8M2+10M1;核型分类为2A型,属于基本对称型。  相似文献

An empirical model describing the diffusion kinetics of oxygen in silicate minerals under hydrothermal conditions has been established for temperatures between 773 and 1073 Kelvin at 100 megapascals of water pressure. The equation, log D = alpha + (beta/T) + [(gamma + (delta/T))Z], where D is the diffusion coefficient, alpha, beta, gamma, and delta are constants, T is the Kelvin temperature, and Z is the total ionic porosity, may be used to predict diffusion coefficients, in most cases to within the reported experimental reproducibility of a factor of 2. For oxygen diffusion, alpha = -2, beta = -3.4 x 10(4)K, gamma = -0.13, and delta = 6.4 x 10(2)K, for D in square centimeters per second. Limited data for the diffusion of argon in silicates suggest that the model describes this system as well.  相似文献

The Voyager 2 encounter with the Neptune system included radio science investigations of the masses and densities of Neptune and Triton, the low-order gravitational harmonics of Neptune, the vertical structures of the atmospheres and ionospheres of Neptune and Triton, the composition of the atmosphere of Neptune, and characteristics of ring material. Demanding experimental requirements were met successfully, and study of the large store of collected data has begun. The initial search of the data revealed no detectable effects of ring material with optical depth tau [unknown] 0.01. Preliminary representative results include the following: 1.0243 x 10(26) and 2.141 x 10(22) kilograms for the masses of Neptune and Triton; 1640 and 2054 kilograms per cubic meter for their respective densities; 1355 +/- 7 kilometers, provisionally, for the radius of Triton; and J(2) = 3411 +/- 10(x 10(-6)) and J(4) = -26(+12)(-20)(x10(-6)) for Neptune's gravity field (J>(2) and J(4) are harmonic coefficients of the gravity field). The equatorial and polar radii of Neptune are 24,764 +/- 20 and 24,340 +/- 30 kllometers, respectively, at the 10(5)-pascal (1 bar) pressure level. Neptune's atmosphere was probed to a pressure level of about 5 x 10(5) pascals, and effects of a methane cloud region and probable ammonia absorption below the cloud are evident in the data. Results for the mixing ratios of helium and ammonia are still being investigated; the methane abundance below the clouds is at least 1 percent by volume. Derived temperature-pressure profiles to 1.2 x 10(5) pascals and 78 kelvins (K) show a lapse rate corresponding to "frozen" equilibrium of the para- and ortho-hydrogen states. Neptune's ionosphere exhibits an extended topside at a temperature of 950 +/- 160 K if H(+) is the dominant ion, and narrow ionization layers of the type previously seen at the other three giant planets. Triton has a dense ionosphere with a peak electron concentration of 46 x 10(9) per cubic meter at an altitude of 340 kilometers measured during occultation egress. Its topside plasma temperature is about 80 +/- 16 K if N(2)(+) is the principal ion. The tenuous neutral atmosphere of Triton produced distinct signatures in the occultation data; however, the accuracy of the measurements is limited by uncertainties in the frequency of the spacecraft reference oscillator. Preliminary values for the surface pressure of 1.6 +/- 0.3 pascals and an equivalent isothermal temperature of 48 +/- 5 K are suggested, on the assumption that molecular nitrogen dominates the atmosphere. The radio data may be showing the effects of a thermal inversion near the surface; this and other evidence imply that the Triton atmosphere is controlled by vapor-pressure equilibrium with surface ices, at a temperature of 38 K and a methane mixing ratio of about 10(-4).  相似文献

A 2×2×2 factorial design was adopted to study the effects of temperature,moisture and choline chloride on vitamin A stability in premix.The results indicated that temperature,moisture and choline chloride damaged vitamin A significantly.The regression equations of vitamin A disappearance rate and storage time were as follows:in room temperature(18 3)℃,y=14.368Ln(x) 4.1425,R~2=-0.978;in high temperature(4℃),y=22.24Ln(x) 13.27,R~2=0.9918;in low moisture(2%-3%),y=10.408Ln(x) 9.5418, R~2=-0.9322;in high moisture(8%-9%),y=26.199Ln(x) 7.8741,R~2=-0.9949;in the condition of choline chloride free,y=9.5125Ln(x) 8.9869,R~2=0.9826;supplemented with choline chloride,y=27.094Ln(x) 8.4276,R~2=0.9984.Temperature had highly significant interaction with moisure and choline chloride on destruction of vitamin A,respectively from the periods of two months storage. However,from the period of the first month storage,the interaction of moisture and choline chloride,as well as the interaction of temperature,moisture and choline destroyed vitamin A remarkably.  相似文献

Acetylcholine is transported along insect chemoreceptor axons at a rate of 12 to 13 centimeters per day after peripheral uptake of choline. Colchicine, vinblastine sulfate, and cytochalasin B all block transport, but transport continues in axons separated surgically from the cell body. These data from an insect are in accord with many studies on vertebrates which have implicated intracellular microtubules in the transport mechanism. The peripheral uptake of choline and its acetylation and transport to nerve terminals in the brain are consistent withthe suggestion that acetylcholine is an antennal sensory transmitter in insects.  相似文献
