The effects of dietary restriction of vitamin E (Vit E) and selenium (Se) on lymphocyte proliferation, natural killer (NK) cell activity, antibody-dependent cell-mediated cytotoxicity (ADCC), and on burst respiratory response of stimulated granulocytes as measured by chemiluminescence (CL) were studied in pigs. Six male weanling pigs were maintained for 25 d on a torula yeast-based diet containing no measurable amount of alpha-tocopherol and less than .02 mg of Se per kilogram of feed. Six others received the same basal diet supplemented with 33 IU of DL-alpha-tocopheryl acetate and .2 mg of Se per kilogram of feed. All pigs were inoculated with Salmonella typhisuis on d 21 of the feeding period and killed on d 25. Tests to measure cellular immune functions were performed on cells isolated from blood samples taken on d 21 and 25. After 21 d of feeding, lymphocyte blastogenesis responses to phytohemagglutinin, concanavalin A, and pokeweed mitogen in pigs fed the Vit E- and Se-deficient diet were normal compared with the response in pigs fed the supplemented diet. Moreover, the cytotoxic activity of NK cells, the ADCC response, and the CL response of granulocytes were not affected. After 25 d, a marked suppression of lymphocyte response to mitogens occurred in pigs fed the Vit E- and Se-deficient diet when the cells were cultured in the presence of autologous serum. When fetal bovine serum replaced autologous serum in the cultures, no suppression was observed. No effect on NK activity and ADCC was observed, whereas the CL peak response of granulocytes tended to be higher in pigs fed the deficient diet.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Three trials using 80 Yorkshire x Hampshire x Duroc crossbred pigs (avg initial wt, 6.9 kg) were conducted to determine the effects of four dietary vitamin E levels (11, 110, 220 and 550 IU/kg of feed) on the humoral and cell-mediated immune response and performance of 4-wk-old weanling pigs housed at two nursery temperatures (19 or 30 degrees C). Interactive effects of temperature and vitamin E were not observed for any measurements. Serum and liver vitamin E levels increased linearly with increasing level of dietary vitamin E, but performance, cortisol, antibody levels and mitogen-induced stimulation indices were not affected by supplemental levels of vitamin E. Average daily gain and feed intake were higher (P less than .01) for pigs housed at 19 degrees C than for pigs housed at 30 degrees C, but feed:gain ratios, mitogen stimulation index of white blood cells, plasma cortisol levels and antibody titers were not altered. Although supplemental vitamin E above the NRC estimated requirement increased serum and liver vitamin E concentrations, no differences were observed in humoral and cell-mediated immune response, cortisol levels or performance for weanling pigs housed at either 19 or 30 degrees C temperatures.  相似文献

Avidity indices of antibody to hen egg-white lysozyme (HEWL) were measured by chaotropic ion (SCN-) elution enzyme-linked immunosorbent assay (ELISA) in pigs grouped as high, control or low for various immune and innate resistance-related traits. The avidity index was the molar concentration of SCN- required to reduce by 50% the ELISA optical density value for a given serum. The index was independent of the amount of antibody. Eight- to ten-week-old Yorkshire pigs were immunized with HEWL and serum antibody measured by ELISA as one of five traits used to assign them to high, low or control response groups. Serum antibody avidity for HEWL was evaluated on Day 14 and Day 30 after primary (Day 0) and secondary (Day 14) immunization. The effects of response group, gender, litter, serum IgG concentration and anti-HEWL antibody on avidity were determined using a linear model. Antibody avidity indices varied amongst individuals. Mean avidity indices for sera collected on Days 14 and 30 were 0.61 +/- 0.43 and 1.22 +/- 0.56, with maximum indices of 2.64 and 2.86 respectively. Avidity of secondary response antibody was significantly higher (P less than or equal to 0.05). Pigs of the high response group had significantly higher secondary antibody avidity than those of the control (P less than or equal to 0.08) and low groups (P less than or equal to 0.01). Avidity index was positively correlated with antibody to HEWL on Days 14 and 30 but not to preimmunization serum IgG concentration or to other measured traits.  相似文献

Pigs selected for high (H) or low (L) combined antibody and cell-mediated immune response were infected with Mycoplasma hyorhinis. Following the infection, arthritis was more severe in the H pigs, while pleuritis and peritonitis were more severe in the L pigs. Since Mycoplasma infections in pigs often cause just mild signs, indicators of the inflammatory response may aid diagnosis of such infections. In addition, data about the genetic influence on inflammatory response indicators are scanty in the pig. The objectives of the study were therefore: firstly, to determine interferon-alpha (IFNalpha) and haptoglobin in M. hyorhinis infected pigs and, secondly, to investigate if the inflammatory response as determined by these indicators was influenced by genetic selection. There was no consistent increase of IFN-alpha in serum following infection. The serum haptoglobin concentration started to increase 3 days post-infection and there was no difference between the two breeding lines. Hence, M. hyorhinis infection in pigs is reflected in increased serum haptoglobin concentration, but no effect of the magnitude of the inflammatory response on this indicator by selection for high or low immune response was observed.  相似文献

The cellular immune response to an experimental infection by Haemophilus parasuis, the etiological agent of Glässer’s disease in pigs, was characterized studying changes in peripheral blood mononuclear cells (PBMC) in colostrum-deprived pigs. Five groups were studied, four of those were previously immunized with different formulations and the fifth was maintained as non-immunized control. All groups were challenged with 5 × 109 CFU of H. parasuis serotype 5. The non-commercial bacterin conferred a complete protection, while the OMP-vaccine and the exposure to a subletal dose of 105 CFU of H. parasuis protected only partially, and the recombinant Tbp B-vaccine induced no protection. PBMC were analyzed using monoclonal antibodies against porcine CD45+, CD3+, CD4+, CD8α+, CD25+, CD4+ naïve, αIgM+ and SWC3+ cells in single-colour fluorescence, and CD4+/CD8α+ and CD8α+/CD8β+ combinations in two-colour fluorescence. The different groups showed no significant changes in PBMC subsets following vaccination, and only minor changes were encountered after challenge, consisting mainly of significant increases (P < 0.05) in the relative proportions of monocytes and granulocytes (SWC3+) and B cells (αIgM+), as well as a significant reduction in CD3+ cells (P < 0.05). These changes were similar for the five groups compared, except for the significant increase of CD25+ cells, which was only observed for the bacterin-vaccinated group. These results suggest an increase of trafficking of inflammatory cells and the onset of the adaptive antibody response against H. parasuis infection; in addition, the blood cellular response developed by the different groups was not relevant to protection.  相似文献

Immunoglobulin (Ig) function varies by isotype and antibody activity is best mediated by isotypes most able to control the inciting infection. In pigs, a high ratio of IgG1:IgG2 is associated with resistance to disease caused by the extra-cellular bacterium Actinobacillus pleuropneumoniae. This ratio is controlled by type 1/type 2 cytokines in vitro, reflecting cell- (CMI) or antibody-mediated immune (AMI) responses, respectively. Animals were used which had been previously selectively bred for high (HIR) or low (LIR) combined AMI and CMI and had been immunized with hen eggwhite lysozyme (HEWL) in Quil A (days 0 and 14) while Bacillus Calmette Guérin was given on day 9. To test the hypothesis that lines do not differ in IgG isotype expression as antibody to HEWL, the ratio of anti-HEWL associated with IgG1 and IgG2 was determined at days 0, 9, 14 and 21. The ratio of IgG1:IgG2-associated antibody was always <1.0 indicating a type 1 response and differed significantly over time in HIR and LIR animals. After primary and secondary immunizations, the HIR animals' IgG1:IgG2-associated antibody ratio increased and approached 1 while for LIR animals the ratio decreased. Thus anti-HEWL antibody in HIR, but not LIR, approached balance in type 2:type 1 expression. Individual variation in immune response was frequently significant within each immune response group. Thus, proportional production of anti-HEWL antibody associated with IgG isotypes varies by individual and differs over time as a function of genotype in pigs selectively bred for HIR or LIR.  相似文献

Immune function (response to concanavalin A, cytokine production, and lymphocyte profiles) and blood chemistry variables were measured in growing-finishing pigs (Yorkshire/Landrace/Duroc dam × Hampshire sire) fed varying percentages of CLA (0, 0.12, 0.25, 0.50, and 1.0%). Blood was collected at 0, 14, 28, 42, and 56 d on feed (DOF). Total white blood cell (WBC) count increased (P < 0.01) linearly to 42 DOF. No differences (P = 0.53) were observed for WBC across CLA treatment. Nitric oxide was greater (P < 0.01) for the 1.0% CLA treatment compared with all other treatments. Flow cytometry using fluorescent labeled monoclonal antibodies to the CD4, CD8, double-positive CD4/CD8, and CD2 surface markers was used to determine lymphocyte subpopulations. Supplementation of CLA had no effect (P = 0.61) on lymphocyte subpopulation cell distribution. Most blood chemistry variables were within the normal metabolic range for pigs. A decrease was observed over DOF for P (P < 0.01) and K (P < 0.05). Additionally, Na and Cl concentrations increased (P < 0.05) from 14 to 28 DOF and decreased over the remainder of the trial. Electrolyte balance was not different (P = 0.38) across CLA treatments and was likely explained by no differences in feed intake among the CLA treatment groups. Blood lipid variables indicated that total cholesterol (P < 0.001), triglycerides (P < 0.001), high-density lipoproteins (P < 0.001), and low-density lipoproteins (P < 0.01) increased as the amount of CLA in the diet increased, but none of the results from these treatments exceeded the normal range of acceptability. These results suggested that CLA was safe when fed to growing-finishing pigs and had little effect on their immune function and blood chemistry variables.  相似文献

Ten sets of 5 littermate pigs from each of 2 genetic strains were utilized to determine the impact of the dietary concentration of 5 B vitamins (riboflavin, niacin, pantothenic acid, cobalamin, and folacin) on growth from 9 to 28 kg of BW in pigs with high or moderate capacity for lean growth. All pigs (penned individually) were reared via a segregated, early weaning scheme, so that the lean growth potential of each strain could be expressed. The basal diet provided the 5 test vitamins at concentrations of total and estimated bioavailability equivalent to a minimum of 100 and 70%, respectively, of their estimated requirements (NRC, 1998) for 5- to 10-kg pigs. At a BW of 9 +/- 0.9 kg, pigs within each litter were allotted to the basal diet supplemented with sources of the 5 test vitamins equivalent to an additional 0, 100, 200, 300, or 400% (bioavailable) of the NRC requirements. Pigs from the high lean strain consumed less feed (P < 0.05) and gained BW faster (P < 0.02) and more efficiently (P < 0.01) than pigs of the moderate lean strain. In both lean strains, the rate and efficiency of growth were improved (P < 0.01) as dietary B vitamin concentrations were increased. However, the dietary B vitamin concentrations needed to optimize G:F were greater (P < 0.03) in the high (>470% of NRC, 1998) vs. moderate (270%) lean strain. Based on these data, the dietary needs for 1 or more of the 5 B vitamins are greater than current NRC (1998) estimates, particularly in pigs expressing a high rate of lean tissue growth. The greater need for these vitamins is not associated with greater dietary energy intake or body energy accretion rate but is potentially due to shifts in the predominant metabolic pathways.  相似文献

To investigate the influence of baseline enterotypes and dietary starch type on the concentration of short-chain fatty acids (SCFA), numbers of butyrate producing bacteria and the expression of genes related to intestinal barrier and inflammatory response in the colon of finishing pigs, a 60-d in vivo trial was conducted. A 2-wk pre-trial with 102 crossbred (Duroc × [Landrace × Yorkshire]) finishing barrows (90 d old) was conducted to screen enterotypes. Then, a total of 32 pigs (87.40 ± 2.76 kg) with high (HPBR, ≥ 14) and low (LPBR, ≤ 2) Prevotella-to-Bacteroides ratios (PBR) in equal measure were selected and randomly divided into 4 groups with 8 replicates per group and 1 pig per replicate. The trial was designed following a 2 (PBR) × 2 (amylose-to-amylopectin ratio, AMR) factorial arrangement. Pigs with different PBR were fed diets based on corn-soybean meal with high AMR (HAMR, 1.24) or low AMR (LAMR, 0.23), respectively. Results showed that neither PBR nor AMR influenced the growth performance of pigs. HPBR pigs fed HAMR diet had a higher number of colonic Clostridium cluster XIVa and higher gene expression of butyrate kinase compared to the LPBR pigs (P < 0.05). The HPBR pigs fed HAMR diets also had increased colonic concentrations of total SCFA and propionate compared to the LPBR pigs (P < 0.05). Comparing with other pigs, HPBR pigs fed HAMR diets showed a lower (P < 0.05) expression of histone deacetylases (HDAC) gene and higher (P < 0.05) expression of G protein-coupled receptor 43 gene (GPR 43) in the colonic mucosa. The interaction (P < 0.05) of HPBR and HAMR was also found to decrease the gene expression of interleukin (IL)-6, IL-12, IL-1β and tumor necrosis factor-α (TNF-α) in colonic mucosa. These findings show that HAMR diet increased the abundance and activity of butyrate-producing bacteria and the concentration and absorption of SCFA, which may be associated with the decreased gene expression of inflammatory cytokines in the colonic mucosa of pigs with Prevotella-rich enterotype. All these alterations are likely to have a positive effect on the intestinal health of finishing pigs.  相似文献

The susceptibility of a given muscle tissue to lipid oxidation may not only depend on the presence of unsaturated fatty acids and the balance between antioxidants and prooxidants, but also on the composition of the skeletal muscle. In the present study, the effects of dietary supplementation of vitamin E (dl-alpha-tocopheryl acetate) and copper in combination with a high level of monounsaturated fatty acids were examined with regard to the antioxidant concentration and the susceptibility to lipid oxidation of two muscles, longissimus (LD) and psoas major (PM), representing different oxidative capacity. In addition, fatty acid profiles of the backfat and the intramuscular lipids, as well as fresh meat quality traits, were studied. Pigs were allotted to a 3x3 factorial experiment with three levels of dl-alpha-tocopheryl acetate (0, 100, and 200 mg/kg of feed) and three levels of copper (0, 35, and 175 mg/kg of feed) added to a diet containing 6% rapeseed oil. A basal diet (without rapeseed oil) was added to the experimental design, giving a total of 10 dietary treatments. Muscle alpha-tocopherol concentrations increased (P<.001) with increasing dl-alpha-tocopheryl acetate in the feed. The antioxidative status was higher in PM than in LD, when considering the concentration of alpha-tocopherol (P<.001) and the activity of antioxidant enzymes (superoxide dismutase, P<.001; glutathione peroxidase, P = .06). Supplemental copper did not give rise to any deposition of copper in muscle tissue or backfat, but the antioxidant status of PM increased. The susceptibility to lipid oxidation was reduced in LD with increasing dietary dl-alpha-tocopheryl acetate and in PM with increasing dietary copper. Supplemental dl-alpha-tocopherol acetate improved the water-holding capacity of LD (P = .005) and PM (P = .003). The fatty acid composition of the backfat and the triglyceride fraction of the intramuscular fat became more unsaturated with the addition of rapeseed oil to the feed. Higher intakes of monounsaturated fatty acids due to the rapeseed oil were also reflected in the phospholipid fraction of the intramuscular fat, but no influence on the proportion of saturated fatty acids was seen. The susceptibility to lipid oxidation of PM was lower for pigs on the rapeseed oil-based diet than for those on the basal diet. The energy metabolic status of the muscles and the accumulation of calcium by the sarcoplasmic reticulum were not influenced by the dietary treatments, but there were differences between muscle types. The addition of rapeseed oil to the diet reduced the muscular content of glycogen (LD, P = .02; PM, P = .06) and elevated the plasma concentration of free fatty acids (P = .05). Overall, dietary fat, dl-alpha-tocopherol acetate, and copper affected the oxidative status of pig muscles, and the results differed depending on muscle type.  相似文献

Fifty-two Targhee twin-bearing ewes were used in a factorial arrangement of treatments to investigate the role of supplemental vitamin E (vit E); 0 (NE) vs 400 IU of vit E x ewe x (-1)d(-1) (E) and parainfluenza type 3 (PI3) vaccination; none (NP) vs PI3 vaccination (P) in immune function. Parainfluenza type 3 vaccination was used to evoke an immune response. Ewes receiving PI3 were vaccinated at 49 and 21 d before the expected lambing date. Ewes receiving vit E were orally dosed daily, 32 to 0 d before lambing. Blood was collected from ewes at the time of the initial PI3 vaccination and 4 h postpartum. Blood was collected from lambs (n = 104) at 3 d postpartum. Ewe and lamb sera were analyzed for anti-PI3 antibody titers, immunoglobulin G (IgG) titers, and vit E concentrations. Colostrum was collected 4 h postpartum and analyzed for IgG. The model for ewe and lamb analysis included the main effects of vit E and PI3, sex (lambs model only), and their interactions. No interactions were detected (P > 0.20) for any ewe or lamb variables. Serum anti-PI3 titers were greater (P < 0.01) in P ewes and their lambs than NP ewes and their lambs. Serum vit E concentrations were greater (P < 0.01) in E ewes and their lambs than NE ewes and their lambs. Colostral IgG titers and serum anti-PI3 titers did not differ (P > 0.20) between E and NE ewes. Serum IgG titers in E ewes and their lambs did not differ (P > 0.15) from IgG titers in NE ewes and their lambs. Lamb anti-PI3 titers did not differ (P = 0.76) between lambs reared by E and NE ewes. These results indicate that, although supplemental vit E to the ewe increased lamb serum vit E concentration, it had no effect on measures used in this study to assess humoral immunity in the ewe or passive immunity to the lamb.  相似文献

Phosphorylated mannans derived from the yeast cell wall of Saccharomyces cerevisiae may beneficially modulate immune function in the weanling pig, possibly providing an alternative to the use of dietary growth-promoting antibiotics. Therefore, in this study, 32 pigs averaging 19 d of age and 5.7 +/- 0.2 kg initial BW were randomly assigned to 16 pens in an environmentally controlled nursery to determine the effects of dietary supplementation with phosphorylated mannans on growth and immune function. Average daily gain and G:F ratio increased (P < 0.05) when pigs were fed diets supplemented with mannans from d 0 to 14 after weaning and in the overall experiment. Percentage of neutrophils was lower (P < 0.08) and percentage of lymphocytes was higher (P < 0.05) in blood from pigs fed mannans than when pigs were fed the basal diet. Lamina propria macrophages isolated from pigs fed diets containing mannans phagocytosed a greater (P < 0.05) number of sheep red blood cells (2.63 +/- 0.11) than did lamina propria macrophages isolated from pigs fed the basal diet (2.31 +/- 0.11). On d 19 after weaning, pigs fed diets supplemented with mannans tended to have a greater (P < 0.10) percentage of CD14+ lamina propria leukocytes than did pigs fed the basal diet. On d 21 following weaning, the percentage of CD14+MHCII+ leukocytes isolated from lamina propria tissue tended (P < 0.10) to be lower when pigs were fed mannans than when pigs were fed the basal diet. Pigs fed diets containing mannans had a lower (P < 0.05) ratio of CD3+CD4+:CD3+CD8+ T lymphocytes isolated from jejunal lamina propria tissue only on d 21 after weaning compared with pigs fed the basal diet. Supplementation of mannans in the diets of weanling pigs improved gain and efficiency, and intermittently affected selected components of the young pigs' immune function both systemically and enterically.  相似文献
