Previously, we isolated mixed polyphenolic fractions on a toyopearl matrix (TP-2 to TP-6) from grape cell cultures that were highly potent catalytic inhibitors in a human DNA topoisomerase II assay for cancer chemoprevention. The objectives of this study were to evaluate the potency of, and potential interactions between, individual fractions and some of the purified bioactive polyphenols that comprise these fractions on human DNA topoisomerase II catalytic activity. Treatments that combined anthocyanin-rich fractions (TP-2; 0.5 or 2.0 microg of dried material/mL), fractions containing catechins, procyanidin dimers, and flavanones (TP-4; 0.25 microg of dried material/mL), and/or fractions enriched with procyanidin oligomers and polymers (TP-6; 0.15 or 0.5 microg of dried material/mL) showed additive effects toward catalytic inhibition of the enzyme. Epicatechin gallate (IC50 = 0.029 microM), myricetin (0.39 microM), procyanidin B2 (PB2, 4.5 microM), and resveratrol (65.7 microM), constituents of the most bioactive mixed fraction from grape cell culture (TP-4), each individually provided potent catalytic inhibition of topoisomerase II. In addition, potentiating interactions between the PB2 and the other polyphenolic constituents mentioned above and between myricetin and resveratrol were clearly demonstrated. A synergistic interaction between myricetin and resveratrol was also confirmed with isobolographic analysis at a molar ratio of 1:70.  相似文献

为了探讨葡萄籽提取物对无淀粉火腿肠保质期的影响,将葡萄籽提取物配成质量分数为0.05%、0.10%、0.15%和0.20%的溶液,添加于火腿肠中,于0~4℃贮藏,测定细菌总数、pH值等的变化。结果表明:在贮藏过程中,添加葡萄籽提取物火腿肠的感官品质明显好于对照样;处理样的细菌总数、pH值、挥发性盐基氮(TVB-N值)、硫代巴比妥酸(TBA值)和黄色值(b*)明显低于对照组,亮度(L*)和红色值(a*)明显高于对照组;葡萄籽提取物的浓度越高,对细菌总数、pH值、L*的影响越明显,但对TVB-N值、TBA值、a*和b*的影响没有明显规律;从生产成本和使用效果综合考虑,建议生产中使用质量分数为0.20%的葡萄籽提取物。  相似文献

Topoisomerases are targets of several anticancer agents because their inhibition impedes the processes of cell proliferation and differentiation in carcinogenesis. With very limited information available on the inhibitory activities of peptides derived from dietary proteins, the objectives of this study were to employ co-immunoprecipitation to identify inhibitory peptides in soy protein hydrolysates in a single step and to investigate their molecular interactions with topoisomerase II. For this, soy protein isolates were subjected to simulated gastrointestinal digestion with pepsin and pancreatin, and the human topoisomerase II inhibitory peptides were co-immunoprecipitated and identified on a CapLC- Micromass Q-TOF Ultima API system. The inhibitory activity of these peptides from soy isolates toward topoisomerase II was confirmed using three synthetic peptides, FEITPEKNPQ, IETWNPNNKP,and VFDGEL, which have IC 50 values of 2.4, 4.0, and 7.9 mM, respectively. The molecular interactions of these peptides evaluated by molecular docking revealed interaction energies with the topoisomerase II C-terminal domain (CTD) (-186 to -398 kcal/mol) that were smaller than for the ATPase domain (-169 to -357 kcal/mol) and that correlated well with our experimental IC 50 values ( R (2) = 0.99). In conclusion, three peptides released from in vitro gastrointestinal enzyme digestion of soy proteins inhibited human topoisomerase II activity through binding to the active site of the CTD domain.  相似文献

UV radiation leads to the generation of reactive oxygen species (ROS). These molecules exert a variety of harmful effects by altering key cellular functions and may result in cell death. Several studies have demonstrated that human skin can be protected against UV radiation by using plant-derived antioxidants. Here we evaluated the in vitro capacity of several antioxidant polyphenolic fractions from grape, which differ in their degree of polymerization and percentage of galloylation, to protect HaCaT human keratinocytes against UV-induced oxidative damage. These fractions inhibited both basal and UVB- or UVA-induced intracellular ROS generation in this cell line. Consequently, the same fractions inhibited p38 and JNK1/2 activation induced by UVB or UVA radiation. The highest protective effect was for fractions rich in procyanidin oligomers and gallate esters. These encouraging in vitro results support further research and should be taken into consideration into the clinical pharmacology of plant-derived polyphenolic extracts as novel agents for skin photoprotection.  相似文献

研究了红砂土中加入稻草秸秆和猪粪后土壤中微生物生物量碳、水溶性有机碳、溶解性酚等活性有机质组分的动态变化。有机肥处理一周后 ,微生物生物量碳、水溶性有机碳、溶解性酚等都达到最高值。随着培养时间的推移 ,不同处理的土壤中微生物生物量碳和水溶性有机碳含量都逐渐下降 ,而溶解性酚含量在整个培养过程中基本保持不变。相关分析表明 ,土壤中微生物生物量碳与水溶性有机碳和溶解性酚的含量都呈极显著正相关 ,而水溶性有机碳和溶解性酚的含量不相关。  相似文献

Grapes are rich sources of potentially bioactive polyphenols. However, the phenolic content is variable depending on grape variety, and may be modified during vinification. In this study, we examined the chemical composition and biological activity of phenolic extracts prepared from several red wine grape varieties and their fermented byproduct of winemaking (pomace) on some of the virulence properties of Streptococcus mutans a well-known dental pathogen. Grape phenolic extracts were obtained from Vitis vinifera varieties Cabernet Franc and Pinot Noir and Vitis interspecific hybrid varieties Baco Noir and Noiret. The anthocyanins and flavan-3-ols content were highly variable depending on grape variety and type of extract (whole fruit vs fermented pomace). Nevertheless, all grape phenolic extracts remarkably inhibited glucosyltransferases B and C (70-85% inhibition) at concentrations as low as 62.5 microg/mL (P < 0.01). Furthermore, the glycolytic pH-drop by S. mutans cells was inhibited by the grape extracts without affecting the bacterial viability; an effect that can be attributed to partial inhibition of F-ATPase activity (30-65% inhibition at 125 microg/mL; P < 0.01). The biological activity of fermented pomace was either as effective as or significantly better than whole fruit grape extracts. The results showed that grape phenolic extracts, especially from pomace, are highly effective against specific virulence traits of S. mutans despite major differences in their phenolic content.  相似文献

探讨大田条件下,补镁对缺镁葡萄叶片活性氧含量、保护酶活性和产量的影响,为葡萄科学施肥提供理论依据。采用裂区试验,主区设4个土施硫酸镁梯度,分别为0、112. 5、150、187. 5 kg/hm~2,副区设3个叶面喷施硫酸镁浓度,分别是0%、0. 2%、0. 4%。结果发现:葡萄缺镁时,叶片超氧阴离子产生速率加快,过氧化氢含量增加,超氧化物歧化酶活性升高,但过氧化物酶和过氧化氢酶活性显著降低,叶片脯氨酸和丙二醛含量升高,生物膜受到伤害。通过土施和叶面喷施的方式适量补镁,葡萄叶片活性氧产生速率降低,同时保护酶过氧化物酶和过氧化氢酶活性增加,膜脂过氧化产物丙二醛含量降低,生物膜的受伤程度得到缓解,叶片衰老速率降低,葡萄产量提高,但当施入量过大对生物膜受伤程度的缓解效果降低,葡萄产量降低。土施150 kg/hm~2同时叶面喷施0. 4%硫酸镁肥时,叶片超氧阴离子产生速率低,过氧化氢含量少,膜质过氧化产物丙二醛含量低,葡萄产量高,是该土壤环境下的最佳补镁措施。  相似文献

Three different solvent extracts (methanol, ethyl acetate, and n-hexane) of longan ( Dimocarpus longan Lour.) flowers were assayed with three different antioxidant capacity methods, namely, the DPPH free radical scavenging effect, the oxygen radical absorbance capacity (ORAC) assay, and the inhibition of Cu(2+)-induced oxidation of human low-density lipoprotein (LDL). It was revealed that the methanol extract has the best antioxidative activity, followed by ethyl acetate and n-hexane extracts. The methanol extract was separated by liquid-liquid partition into n-hexane, ethyl acetate, n-butanol, and water fractions. The ethyl acetate fraction was found to have the highest activity of delaying LDL oxidation. After silica gel column chromatography, the fraction having a superior activity was identified as containing two major compounds, (-)-epicatechin and proanthocyanidin A2.  相似文献

The effect of dietary fiber in the form of apple cell walls and pectin extracts on natural antioxidants was examined. Cell walls (CW), isolated from apples ( Malus domestica Borkh. cv. "Pacific Rose"), were incubated with ascorbic acid (AA) or quercetin in N-2-hydroxyethylpiperazine- N'-2-ethanesulfonic acid (HEPES) buffer (pH 6.5) at 37 degrees C for 2 h. The resulting supernatants were characterized by a ferric reducing antioxidant power (FRAP) assay and cyclic voltammetry (CV). The experiments were repeated with pectin isolated from the apple cell walls and commercial pectins and showed that polysaccharide preparations stabilized AA effectively but offered little protection against quercetin oxidation. The water-soluble components from cell walls appeared to be responsible for the observed effects of cell-wall polysaccharide preparations on antioxidant activity.  相似文献

A range of radiolabeled anthocyanins, proanthocyanidins, and other flavonoids were accumulated by cell suspension cultures of two plant species, ohelo (Vaccinium pahalae) and grape (a Vitis hybrid, Bailey Alicant A), after providing uniformly labeled [(14)C]sucrose to the medium. Approximately 15% of administered label was recovered in a series of flavonoid-rich fractions varying in composition. Anthocyanins, and monomers to oligomers of proanthocyanidins, were labeled effectively and characterized from both species. Most of the proanthocyanidin oligomers were based on the flavan-3-ols (+)-catechin and (-)-epicatechin. Cyanidin and peonidin glycosides were the dominant forms of anthocyanins in both species. Whereas the predominant form of flavonoids identified from ohelo cell cultures was proanthocyanidins, grape cell cultures produced mostly anthocyanins. The labeled phytochemicals were produced for use in subsequent in vivo animal feeding studies to gauge their bioavailability and accumulation in target organs.  相似文献

【目的】在大同盆地苏打盐化土上,研究不同有机物料对春玉米产量、土壤有机碳及活性碳组分的影响,明确土壤有机碳及活性碳组分与主要盐碱指标的相关关系,为苏打盐化土改良及有机物料资源化利用提供理论支撑。【方法】2016-2017年在山西省北部怀仁县开展田间定位试验,设对照(CK)、风化煤、生物炭、牛粪和秸秆5个处理,各处理有机物料施用量按照每年9000 kg/hm^2等有机碳投入量折算,收获时对春玉米进行测产。2017年春玉米收获后,采集土壤样品测定土壤有机碳总量(SOC)和水溶性有机碳(WSOC)、易氧化有机碳(EOC)、轻组有机碳(LFOC)含量,分析土壤活性碳组分占有机碳的比例、土壤有机碳及活性碳组分与盐碱指标之间的关系。【结果】与CK相比,生物炭和秸秆处理春玉米产量无明显差异,而风化煤和牛粪处理春玉米产量则分别显著提高30.2%和30.3%。添加有机物料促进了0-20 cm土层SOC累积,其中以风化煤和牛粪处理效果最佳,较CK分别提高47.6%和36.1%。在有机碳组分方面,风化煤和牛粪处理提高WSOC、EOC含量的效果显著高于生物炭、秸秆处理;风化煤、牛粪和秸秆处理的LFOC含量显著高于生物炭处理。四类有机物料处理的WSOC占总有机碳的比例差异不显著,牛粪处理的占比显著高于CK。EOC占总有机碳的比例以牛粪处理最高,风化煤次之,且二者均显著高于CK处理;LFOC占总有机碳的比例则表现为秸秆、牛粪>风化煤、生物炭> CK。此外,添加有机物料能有效降低0-20 cm土层土壤pH、电导率(EC)和碱化度(ESP),其中以风化煤和牛粪处理降幅最大。相关分析表明,土壤SOC与pH、EC和ESP呈显著负相关。【结论】通过有机物料改良效果比较,发现牛粪和风化煤处理能促进苏打盐化土有机碳累积,提高可溶性、易氧化态及轻组有机碳组分在总有机碳中的占比,降低土壤pH、EC和ESP,明显提高春玉米产量。因此,风化煤和牛粪是山西北部苏打盐化土良好的改良剂。  相似文献

Commercially prepared grape (Vitis vinifera), bilberry (Vaccinium myrtillus L.), and chokeberry (Aronia meloncarpa E.) anthocyanin-rich extracts (AREs) were investigated for their potential chemopreventive activity against colon cancer. The growth of colon-cancer-derived HT-29 and nontumorigenic colonic NCM460 cells exposed to semipurified AREs (10-75 microg of monomeric anthocyanin/mL) was monitored for up to 72 h using a sulforhodamine B assay. All extracts inhibited the growth of HT-29 cells, with chokeberry ARE being the most potent inhibitor. HT-29 cell growth was inhibited approximately 50% after 48 h of exposure to 25 microg/mL chokeberry ARE. Most importantly, the growth of NCM460 cells was not inhibited at lower concentrations of all three AREs, illustrating greater growth inhibition of colon cancer, as compared to nontumorigenic colon cells. Extracts were semipurified and characterized by high-pressure liquid chromatography, spectrophotometry, and colorimetry. Grape anthocyanins were the glucosylated derivatives of five different anthocyanidin molecules, with or without p-coumaric acid acylation. Bilberry contained five different anthocyanidins glycosylated with galactose, glucose, and arabinose. Chokeberry anthocyanins were cyanidin derivatives, monoglycosylated mostly with galactose and arabinose. The varying compositions and degrees of growth inhibition suggest that the anthocyanin chemical structure may play an important role in the growth inhibitory activity of commercially available AREs.  相似文献

Stilbenes are phenolic molecules that have antifungal effects in the plant and antioxidant and anti-cancer effects when consumed in the human diet. Glycosylation of stilbenes increases their solubility and may make them more easily absorbed by the intestine. We have found an activity in extracts of cultured cells of Vitis vinifera (cv. Gamay Freaux) that glucosylates the stilbene resveratrol to form piceid. The Km for UDP-Glucose was 1.2 mM, and the Km for resveratrol was 0.06 mM, values similar to those of other phenolic glucosyltransferases. We investigated the resveratrol glucosylating activity of the enzyme extracted from cells grown under different light treatments (dark, visible light, light + ultraviolet (UVC) radiation) and found the activity to be unaffected or slightly reduced. In contrast, UVC light strongly stimulated extractable quercetin glucosyltransferase activity. These results, combined with analysis of phenolic compounds extracted from the differently treated cells, suggest that the resveratrol glucosyltransferase is distinct from the glucosyltransferase(s) active on other phenolics.  相似文献

Biologically active fractions of soil organic matter are important in understanding decomposition potential of organic materials, nutrient cycling dynamics, and biophysical manipulation of soil structure. We evaluated the quantitative relationships among potential C and net N mineralization, soil microbial biomass C (SMBC), and soil organic C (SOC) under four contrasting climatic conditions. Mean SOC values were 28±11 mg g−1 (n=24) in a frigid–dry region (Alberta/British Columbia), 25±5 mg g−1 (n=12) in a frigid–wet region (Maine), 11±4 mg g−1 (n=117) in a thermic–dry region (Texas), and 12±5 mg g−1 (n=131) in a thermic–wet region (Georgia). Higher mean annual temperature resulted in consistently greater basal soil respiration (1.7 vs 0.8 mg CO2–C g−1 SOC d−1 in the thermic compared with the frigid regions, P<0.001), greater net N mineralization (2.8 vs 1.3 mg inorganic N g−1 SOC 24 d−1, P<0.001), and greater SMBC (53 vs 21 mg SMBC g−1 SOC, P<0.001). Specific respiratory activity of SMBC was, however, consistently lower in the thermic than in the frigid regions (29 vs 34 mg CO2–C g−1 SMBC d−1, P<0.01). Higher mean annual precipitation resulted in consistently lower basal soil respiration (1.1 vs 1.3 mg CO2–C g−1 SOC d−1 in the wet compared with the dry regions, P<0.01) and lower SMBC (31 vs 43 mg SMBC g−1 SOC, P<0.001), but had inconsistent effects on net N mineralization that depended upon temperature regime. Specific respiratory activity of SMBC was consistently greater in the wet than the dry regions (≈33 vs 29 mg CO2–C g−1 SMBC d−1, P<0.01). Although the thermic regions were not able to retain as high a level of SOC as the frigid regions, due likely to high annual decomposition rates, biologically active soil fractions were as high per mass of soil and even 2–3-times greater per unit of SOC in the thermic compared with the frigid regions. These results suggest that macroclimate has a large impact on the portion of soil organic matter that is potentially active, but a relatively small impact on the specific respiratory activity of SMBC.  相似文献

为了探讨不同秸秆还田模式对土壤有机碳(total organic carbon,TOC)及活性碳组分的影响,设置了秸秆不还田(CK)、秸秆直接还田(CS)、秸秆转化为食用菌基质,出蘑后菌渣还田(CMS)和秸秆过腹还田(CGS)4种还田模式。通过田间小区试验,研究了不同秸秆还田模式下,土壤有机碳及活性组分的变化规律。结果表明不同秸秆还田模式均提高了土壤有机碳含量,但不同还田模式下土壤有机碳含量差异不显著(P0.05),和CK相比,CS、CMS和CGS处理下,土壤有机碳质量分数分别增加9.0%、23.9%和26.7%。不同秸秆还田模式也提高了土壤活性碳组分含量。在不同秸秆还田模式下,土壤溶解性有机碳(dissolved organic carbon,DOC)含量表现为CSCMSCGSCK,且不同处理间差异显著(P0.01)。和CK相比,CS、CMS和CGS处理下,土壤DOC质量分数分别增加64.6%、29.4%和8.9%。土壤微生物量碳(microbial biomass carbon,MBC)含量表现为CMSCGSCSCK,且差异显著(P0.05)。和CK相比,CS、CMS和CGS处理下,土壤MBC质量分数分别增加28.9%、84.7%和59.3%。土壤易氧化态碳(easily oxidizable carbon,EOC)含量表现为CMSCSCGSCK,且差异显著(P0.01)。和CK相比,CS、CMS和CGS处理下,土壤EOC质量分数分别增加24.1%、55.7%、和9.3%。不同秸秆还田模式显著影响土壤活性碳组分在总有机碳中占的比例,改变土壤有机碳质量。在不同秸秆还田模式下,DOC/TOC表现为CSCMSCKCGS、MBC/TOC表现为CMSCGSCSCK、EOC/TOC表现为CMSCSCKCGS,且不同处理间均差异显著(P0.01)。从提高土壤质量角度,推荐秸秆-菌渣还田模式,在该模式下,土壤MBC/TOC和EOC/TOC均最大,土壤碳素有效性高、易于被微生物利用,有利于作物生长。从提高土壤固碳角度,推荐秸秆过腹还田模式,在该模式下,土壤DOC/TOC最小,且土壤有机碳含量最高,有利于碳的固定和保存。该研究结果可为秸秆合理高效利用、改善农业土壤碳库质量提供参考。  相似文献

The aim of this study was to determine the impact of two different treatments of early defoliation performed before blooming on: grape yield, chemical parameters, polyphenols content, and antioxidant activity of grape and red wine cv. Cabernet Sauvignon from the vineyard located in Ilok, the eastern continental region of Croatia. Two different treatments of leaf removal (LR) were performed: removal of 3 leafs (T1) and 6 leafs (T2) before blooming, together with control (no leaf removal) (T3) during two years (2013 and 2014). Crop yield and average cluster weights per vine were determined. Density, pH and titratable acidity were measured in must, while the total phenols, total anthocyanins and antioxidant activity were measured in the extract of grape skin and produced wine. The analysis of individual anthocyanins in wine was performed by HPLC method. T2 treatment significantly lowered the crop yield and the average cluster weights, and increased total phenols, total anthocyanins, antioxidant activity and most abundant individual anthocyanins in wine. Defoliation did not affect the other chemical parameters in must, grape skin extract and wine. Vintage year is statistically the most significant source of variability for density of must, antioxidant activity in grape skin extract, as well as pH and titratable acidity in wine. This study has showed that the early leaf removal treatment in eastern continental part of Croatia could be used for the production of smaller quantity of high quality Cabernet Sauvignon red wine abundant with anthocyanins.  相似文献

为明确秸秆配施膨润土对旱作农田0~40 cm土层土壤有机碳(SOC)、活性有机碳组分及其分配比例和碳库管理指数的影响,于2019—2021年在内蒙古清水河县一间房试验基地连续3年进行田间定位试验。分别设置不施膨润土和秸秆(CK)、单施秸秆(T1)、单施膨润土(T2)和秸秆配施膨润土(T3)4个处理。结果表明:土壤SOC及活性碳组分在年际间表现为先降低后升高[除颗粒有机碳(POC)含量]的变化趋势,不同土层间表现为10~20 cm > 0~10 cm > 20~40 cm;各处理均提高了0~40 cm土层土壤有机碳及其活性组分含量,秸秆配施膨润土效果优于单施。与CK相比,在0~40 cm土层,T3处理土壤SOC、可溶性有机碳(DOC)、易氧化有机碳(LOC)和POC 3年平均提高了7.16%~9.63%、12.35%~18.05%、15.55%~41.97%和100.73%~127.90%,同时提高了不同土层DOC/SOC、LOC/SOC和POC/SOC以及土壤碳库管理指数;土壤SOC及各组分之间均呈显著正相关关系,以LOC相关系数最大,表明LOC可较好地反映出SOC的变化情况。可见,在旱作区采取秸秆配施膨润土措施能够显著提高土壤SOC及其活性组分含量以及碳库管理指数,对土壤肥力提升、质量改善具有重要意义。  相似文献

Grape byproducts were subjected to an extraction process under various different experimental conditions (namely, solvent type, temperature, solvent-to-solid ratio, time contact, and raw material) in order to study the effect of these conditions on the yield of phenolic compounds and the corresponding antiradical activity of extracts. Although the order of decreasing capacity to extract soluble materials was ethanol > methanol > water, methanol was the most selective for extracting phenolic compounds. Temperature and solvent-to-solid ratio were found to have a critical role in extraction efficiency; values of 50 degrees C (between 25 and 50 degrees C) and 1:1 (between 1:1 and 5:1) maximized the antiradical activity of phenolic extracts. In addition, extracts from grape samples previously subjected to distillation reached higher antiradical values in comparison to those coming directly from pressing; in both cases, seed extracts showed better results than those of stem when ethanol or water was employed, whereas the opposite occurred in the case of methanol. These differences were attributed to the different phenolic compositions of the considered fractions.  相似文献

Non-destructive instrumental neutron activation analysis was examined. Using long-lived isotopes, La, Sm, Ce, Sc, Fe, Co, Cs, Th, Cr, Hf, and Tb, and using short-lived isotopes, Al, V, Na, and Mn in various particle size Tractions of clay, silt, fine sand, and coarse sind, and the oxalic acid extracts of the soil were determined. Relative standard deviations (CV) were in the range of 1.7 to 12%. The reliability of the analysis was also examined. Recovery percentages ere in the range of 87 to 120%. The analytical results of geochemical reference samples of JG-1 and JB-1 showed general agreement with the other worker's results.

Distribution patterns of those elements among the six fractions of highly weathered and ively unweathered soils were briefly discussed in relation to weathering processes.  相似文献
