Summary The increasing popularity of organic agriculture and health food products has led to a renewed interest in hulled wheat species such as emmer (Triticum turgidum ssp. dicoccumSchubler). Knowledge on agronomic and quality traits is required for effective and efficient use of germplasm collections in breeding programs. The objective of this study was to estimate agronomic and grain quality traits of emmer wheat cultivated in Italy. A total of 20 emmer accessions consisting of landraces, breeding lines or cultivars selected from landraces and modern cultivars were examined under low input conditions. The study was conducted for three successive years (2002–2004) at one location of Southern Italy (Foggia). The entries were characterized for agronomic and qualitative traits [grain yield (t ha−1), thousand grain weight (g), test weight (kg hl−1), grain protein content (%), HMWG composition, dry gluten content (%), gluten index and yellow index, alveograph indices and Total Organic Matter (TOM) on cooked pasta]. The results showed a large genetic variability for most of the traits measured and, even if most of the accessions showed inferior bread- and pasta-making performance, modern cultivars exhibited improved quality traits with some potential to perform healthy and tasty food.  相似文献

Chloroplast DNA variation in 96 Prunus avium L. cultivars was assessed and compared with the results of a previous study of cpDNA diversity in 23 wild populations of the species. The polymerase chain reaction‐restriction fragment length polymorphism method was used in these studies. Approximately 9% of the chloroplast genome was analyzed, using five universal primer pairs and three restriction enzymes. Ten polymorphic fragments were common to both the wild and sweet cherry; eight polymorphic fragments were found only in the wild cherry. In the cultivars, all mutations were small (5‐30 bp) indels. In the wild populations, a point mutation was also detected in addition to indels. The mutational combinations revealed three haplotypes in the cultivars, which are the main haplotypes in the wild cherry populations. Chloroplast DNA diversity in wild cherry is higher (16 haplotypes) than in sweet cherry cultivars (three haplotypes). The probable wild origin of the sweet cherry cultivars in the maternal line, on the basis of haplotypic similarity, was discussed.  相似文献

Perennial rhizomatous grasses (PRGs) tend to have a high yield combined with a low environmental impact. Cultivation in marginal or poorly cultivated land is recommended in order not to compromise food security and to overcome land use controversies. However, the environmental impacts of using different types of soil are still unclear. We thus assessed the environmental impact of two giant reed (GR) systems cultivated in a fertile soil (FS) and in a marginal soil (MS) through a cradle-to-plant gate LCA. We analyzed energy balance, GHG emissions (including LUC, not including iLUC), and the main impacts on air, water and soil quality. In both systems the annualized soil carbon sequestration was more than twofold the total GHG emitted, equal to −6464 kg CO2eq ha−1 in FS and −5757 kg CO2eq ha−1 in MS. Overall, soil characteristics affected not only GR yield level, but also its environmental impact, which seems to be higher in the MS system both on a hectare and tonne basis. The production of GR biomass in marginal soil could thus lead to higher environmental impacts and a more extensive land requirement.  相似文献

In pot experiments under controlled conditions we investigated the effects of water stress on oilseed rape. Yield and yield components were mainly affected by water shortage occurring from flowering to the end of seed set. The greatest reduction (48%) was observed when only 37% of full water requirement was supplied to the plant during this stage. The number of seeds per plant was the main yield component affected. Some compensation occurred when the water supply was restored. The 1000-seed weight was only affected by a water stress from the stage when the pods were swollen until the seeds colored stage. The results demonstrated a marked reduction in oil concentration when water deficit occurred from anthesis to maturity. There was an inverse relationship between oil and protein concentration. The most marked effect observed in this experiment was on the glucosinolate concentration where increases of up to 60% were observed. These results may explain effects on seed quality of field grown oilseed rape.  相似文献

The extent and distribution of incompatibility between gene pools (Mesoamerican and Andean) and evolutionary classes (wild, landrace and bred) of the common bean (Phaseolus vulgaris) was explored by seeking anomalous values of highly heritable quantitative traits in the progeny of crosses. Clear incompatibility (no progeny or sterile or deformed progeny) was shown by 28 crosses in a 12-parent wild × bred diallel cross, and 37 crosses in a 12-parent landrace × bred diallel cross. Incompatibility was particularly common in the progeny of certain genotypes, but was not consistently associated with the division between gene pools or evolutionary classes. When crosses showing clear incompatibility were eliminated from the data, days to flowering, number of seeds per pod, log (weight per seed) and seed roundness in the F1 generation gave a good fit to an additive-dominance model, confirming that there is no overall tendency to incompatibility between the gene pools. There was a division between the gene pools with regard to log (weight per seed), as expected, but there was no such division, with regard either to the means of the parent lines or the distribution of the statistics V r and W r (which indicate the distribution of dominant alleles between genotypes), for the other quantitative variables. Differences between reciprocal crosses were strikingly widespread, and appeared generally to be due to cytoplasmic effects or cytoplasmic × nuclear interactions rather than maternal effects, indicating that the direction in which a cross is made may have a perceptible effect on the progeny that can be obtained from it. Successful castor (Ricinus communis L.) cropping in Greece depends on the yielding ability and yield stability of the cultivars (hybrids or inbreds) as well as the reliability of production systems. The adaptation and yielding ability of 19 modern castor oil genotypes were studied for 3 years in two sites of Northern Greece. Genotypes combining high seed and oil yield and desirable morphological characteristics were tested for 2 or 3 years, whereas the rest were tested for 1 year only. The growing period in both locations was long enough for ripening the first raceme and a number of secondary racemes depending on the genotypes. The plant height was dependent mainly on the genotypes but also was affected by the site and the year of the experimentation and ranged from 79 to 278 cm. The seed yield varied between 2.5 and 5.0 Mg ha−1, values that are among the highest reported in the literature. The seed yield was higher in the site where plants produced and ripened more secondary racemes. The seed oil content was dependent mainly on the genotype and ranged from 44.5 to 54.2%. The oil yield followed the changes in seed yield. The variation in seed yield between years was low and in most genotypes less than 20%. Results indicate that the castor oil crop was satisfactorily adapted in the area.  相似文献

Summary Six wild barley (Hordeum spontaneum) accessions, from a diverse range of habitats, and two spring-cultivated barleys, were examined for variation in durations of development phases. The durations of the leaf initiation and spikelet initiation phases were longer and spikelet growth phases shorter, in wild than in cultivated barley. Across all wild and cultivated barleys the rate and duration of spikelet initiation were negatively correlated, but neither was related to the number of spikelets per spike. The number of spikelets was positively correlated with the number of leaves and the ratio of the number of spikelets to the number of leaves declined with increasing time to anthesis, indicating that each successive leaf was associated with a diminishing increase in the number of spikelets. The duration of culm elongation and final culm length were shorter in accessions of cultivated barley compared with wild barley. West African cotton production has increased rapidly in recent years. Cotton is being cropped under new ecological conditions by new cotton-producing farmers, but the cropping techniques recommended by developers have essentially remained the same. Methodologies are needed to generate a broad scope of recommendations on cropping techniques to deal with the increasing diversity concerning farmers and cropping conditions.

A conceptual model of a cotton field was developed that approaches a crop field as a biophysical system under the influence of a “technical system” (i.e. the combination of farmers’ practices implemented in the field). The system outputs were restricted to yield and the main yield components. A theoretical model was first designed on the basis of published data and expert knowledge on cotton physiology, local soil–climate conditions and farmers’ practices. It was based on five specific hypotheses on links between technical and biophysical systems. The hypotheses were tested in a local farmers’ network. Thirty “cropping situations” (soil–crop–technique combinations) were selected in farmers’ fields around Katogo village (Mali), a village that had been previously selected for a cotton crop management prototyping program. Homogeneous groups of situations were drawn up on the basis of the dynamics of crop aerial biomass accumulation. They were compared for their management and environment features. The initial conceptual model was then simplified, while taking the measured variability in its components and the sensitivity of the outputs to these components into account. This conceptual model is being evaluated in other villages, where we have partnerships with farmers, in order to develop a version adapted to a broad range of situations.  相似文献

Summary To determine the origin of Ogura male sterile cytoplasm in radish (Raphanus sativus L.), wild and cultivated radishes were crossed. Three types of progeny resulted from the F1 hybrids between the wild radish from Kushikino with Ogura-type mtDNA and the cultivars (Uchiki-Gensuke or Comet). The segregation patterns of the male sterility were compared with those of Ogura cytoplasm. The male sterility induced in the F1 hybrid was maintained by crossing with Uchiki-Gensuke, that maintains Ogura male sterility. In the two types of progeny, in which Comet (a restorer of Ogura cytoplasm) was used as one of the parents, both fertile and sterile plants segregated at the predicted ratio on the assumption that a single dominant fertility restoring gene exists in the restorer. From these results, we concluded that the Ogura cytoplasm is identical to that of the wild radish, and the former originated in a population of Japanese wild radish.  相似文献

Summary Two self-compatible (sc) dihaploids, G254 and B16, and one self-incompatible (si) dihaploid, G609, from Solanum tuberosum L. were intercrossed reciprocally. Segregation ratios sc : si : pc (pseudo-compatible) were determined in all 6 F1's in three successive years and critically tested and discussed. Genotypes at the S-locus could be assigned to the dihaploid parents and the S-allele on the translocation in sc G254 identified as S 1. Using these genotypes all sc and si genotypes were derived which could be expected in the F1's.Incompatibility groups were detected in each F1 from the results of complete diallels involving si plants. The genotype of each group was identified by test crosses. Compatibility groups could be both detected and identified by crossing in each F1 the sc plants as females with the already identified si sibs. In this way a complete series of 6 si testers and corresponding sc genotypes was obtained involving four alleles at the S-locus and S 1 and S x on the translocation.Certative disadvantage of pollen carrying the translocation could be ruled out as a possible cause of unexpected ratios. The hypothesis of an S-bearing translocation as the cause of self-compatibility could account for all results on the assumption that translocation homozygotes are lethal and the S-allele on the translocation is active in the pollen only.The following bachelor students have contributed to the experimental data used in this article: Janny Olsder, J. Barley is the second most widely cultivated cereal on the island of Sardinia. It is grown for green fodder, grain and straw, all of which are used for animal feet. The cultivation of modem varieties is limited and many farmers grow local populations of the six-row landrace ‘S'orgiu sardu’. Twelve local populations (20 strains per population) were evaluated for 13 random amplified polymorphic DNA (RAPD) markers, sisisozyme loci and five morphophenological traits. Results showed that Sardinian barley is characterized by a lax six-row spike and is. in general, a spring type. Isozyme polymorphism was delected for only two esterase loci (Est and Est 4). The genetic variation for all types of murkers, was found located mostly within population (about 84%, for isozymes aud morphaphenological traits and 89%, for RAPDs). This finding appears to confirm the hypothesis that the populations studied belong to the same land race called ‘S'orgiu sardu’ by farmers. A geographical differential ion between southern and northern populations was foand in relation to distinct environmental characteristics. Taking into account all types of marker 209 (90.5%) different genotypes were identified out of 231 individuals. The overall results confirm the high variability that can be found within landrace populations, underlining the value of landraccs as a source of genetic variation.  相似文献

Amplification of the coding region, and upstream and downstream sequences of a low-molecular-weight glutenin subunits (LMW-GS) gene from wild emmer wheat (Triticum turgidum L. var. dicoccoides, 2n = 4x = 28, AABB) accession TD22 was carried out using designed allele-specific PCR (AS-PCR) primers. The complete 1,176 bp sequence of a novel LMW-i type subunit gene at the Glu-A3 locus, named LMW-TD22, is described. Analysis of the nucleotide and deduced amino acid sequences showed that this gene possessed striking characteristics although its molecular structure was generally similar to those of previously reported i-type LMW-GS genes that were isolated from common wheat and related species. The deduced amino acid sequence of LMW-TD22 gene contained 390 amino acid residues with the predicted molecular weight being 43,009.3 Da, which appeared to be the longest gene among the cloned LMW-i type genes from bread wheat and related species. The distinct feature of LMW-TD22 was two long polyglutamine stretches of 12 and 17 glutamines occurring in the repetitive and C-terminal domains as well as a cysteine residue present in the seventh amino acid residue of the signal peptide. These polyglutamine repeats are believed to improve the structure of gluten polymer and increase the strength of dough formed from the polymer. In addition, the putative 44 k subunit encoded by LMW-TD22 was verified by N-terminal microsequencing, gel electrophoresis and matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) analysis. Certain types of post-translational modifications, such as phosphorylation and glycosylation, may be associated with this LMW-i type subunit. A. Wang and Y. 在农杆菌介导的转基因组织培养再生后代中发现了一个无绒有絮的纤维发育突变体,通过自交选择T3代获得其纯合体,命名为CM突变体。尽管CM突变体从转基因后代中发现,但和转基因插入位点无关,推测是在组织培养过程中产生的点突变所致。通过与陆地棉遗传标准系TM-1,海岛棉军海1号,以及新乡小吉无绒有絮(XinFLM),新乡小吉无绒无絮(XinWX),徐州142无绒无絮(XZ142WX),显性光子N1N1,隐性光子n2n2、SL-7-1、MD17及T586等一系列纤维发育突变体分别配制F2组合进行突变基因的遗传及等位性分析,结果表明CM突变体与纤维发育正常的TM-1和军海1号杂交,F1表型均为无绒有絮,F2表现无绒有絮和有绒有絮3∶1分离,说明该突变体与纤维发育正常材料相比,在短绒发育方面存在一个位点的差异,该突变性状由单显性基因控制。等位性测验及分子定位均表明, 控制该突变体短绒的基因与控制N1N1显性光子的N1基因等位。扫描电镜进一步证明该基因突变会造成纤维起始突起延迟。与N1N1突变体相比,CM突变体的衣分比N1N1显著高,而百粒重比N1N1极显著低。推测CM突变体中的突变基因与显性N1基因为复等位基因。  相似文献

A hybrid plant was regenerated from embryo-derived callus after crossing Hordeum vulgare with Psathyrostachys fragilis. A mciotic analysis of pollen mother cells revealed the frequent occurrence of one ring bivalent and twelve univalents. It appeared that eight H. vulgare and six P. fragilis chromosomes were present and an unknown duplicate H. vulgare chromosome had compensated for the loss of one P. fragilis chromosome. Although it remains unclear whether the anomaly was present in the original explain or had occurred during the callus phase, the technique could be used to investigate genome homology between the two species.  相似文献

Genetic diversity was studied in wild forage species subjected to grazing along a rainfall gradient in West Niger, within the Sahel with aim of identifying adapted genotypes, which could be used to reclaim degraded land. Two legumes (Alysicarpus ovalifolius and Zornia glochidiata) and two grasses (Brachiaria xantholeuca and Cenchrus biflorus) were selected to relate phenotypic adaptation to genetic diversity in response to grazing and rainfall. Populations of each species were sampled from both heavily grazed and ungrazed sites along a rainfall gradient, approximating 200 mm yr−1 to 800 mm yr−1 rainfall isohyets. The adaptative phenotypic expressions to aridity and grazing of the populations from each of the species were characterised by morphological measures performed on the plants sampled in the field. These analyses were then compared with the results from genetic analyses using the PCR-based techniques of amplified fragment length polymorphism (AFLP) and random amplified polymorphic DNA (RAPD). Analyses of molecular data using cluster analysis (UPGMA), principal co-ordinates analysis (PCO), Mantel tests and an Analysis of Molecular Variance (AMOVA), revealed genotypic distinction between populations subjected to both differing aridity and grazing. The majority of the total genotypic variation sampled in all species occurred among individuals within a population. The significant morphological differentiation found among populations subjected to varying grazing and aridity stresses, determined through linear regression analyses, did not correlate significantly with the genotypic differentiation, as revealed by Mantel tests. The results suggest that grazing does not cause a loss of genetic diversity in the wild forage species studied, although with increasing aridity the impact of grazing on the genetic diversity of populations may increase.R. Growth trials were conducted outdoors in the UK to determine the yield, yield components and plant architectural differences between determinate and indeterminate Phaseolus vulgaris bean plants. F3 lines derived from crosses between ‘Prelude’, a determinate cultivar and ‘V8’, an indeterminate landrace, were grown together with the parents and ‘Carioca’, an indeterminate landrace from Brazil. Data were recorded on flowering date, number of nodes on main stem at flowering, plant height at maturity, number of pods/plant, number of seeds/pod, number of seeds/plant, 100-seed weight, seed mass/plant, percentages of diseased and healthy seeds/plant. Determinate F3 lines had significantly lower (P < 0.05) seed mass/plant, fewer pods/plant, fewer seeds/pod, fewer seeds/plant, lower harvest index, shorter stems, earlier date of flowering, fewer nodes at flowering and fewer healthy seeds/plant than indeterminate F3 lines. However, determinate genotypes had significantly larger (P < 0.05) pods to accommodate their larger seeds compared with indeterminate lines. A similar situation was found when the yield and yield components of ‘V8’, ‘Prelude’ and ‘Carioca’ were evaluated; the highest seed mass/plant was given by ‘V8’ and the lowest by ‘Prelude’. Furthermore, some indeterminate genotypes with Type IV growth habit yielded significantly higher (P < 0.05) than the high-yielding parent, ‘V8’, indicating a positive influence on seed yield by crossing different genotypes.  相似文献

Screening of 136 faba bean lines forresistance to chocolate spot caused by thefungus Botrytis fabae was conductedin field conditions with artificialinoculation. Detached leaves of thesegenotypes were also assessed for diseaseseverity after inoculation under controlledconditions with a B. fabaesuspension. The BPL710 and Aguadulcegenotypes were inserted as resistant andsusceptible checks, respectively. Diseasesymptoms were scored visually in bothexperiments. Diameter of lesions andproduction of spores were measured in thedetached leaf assay. The disease scoreindex and diameter of lesions were used tocalculate the area under the diseaseprogress curve (AUDPC) to measure thedisease progress. Significant differenceswere detected among genotypes for reactionto the disease in the field. However, nocomplete resistance was observed. Forhighly susceptible and highly resistantgenotypes, the laboratory assay generallyled to the same result as the field test,nevertheless, for more intermediate levelsthe agreement was weak. On detached leaves,diameter of lesions was significantlycorrelated to the AUDPC of disease scores(r = 0.89) and moderately correlated tosporulation (r = 0.52). The correlationbetween field AUDPC and detached leaf AUDPCof disease scores was low (r = 0.26). A lowcorrelation was also observed between fieldAUDPC and lesion size (r = 0.30) andbetween field AUDPC and sporulation (r =0.32). Among the 136 genotypes evaluated inthis study, nine were convincingly andhighly resistant in both tests. FRYM167 andFRYA58 genotypes were the most resistantwith low AUDPC of disease scores, low AUDPCof lesion diameter and low sporeproduction. These genotypes obviously havepartial resistance.  相似文献
