Objective To investigate motor responses to stimulation during the transition from ‘deep’ (burst suppression) to ‘light’ isoflurane anaesthesia in pigs. Study design Prospective, randomized observational study. Animals Five castrated male and five female Norwegian landrace pigs, weighing 19–29 kg. Materials and methods Anaesthesia was induced with isoflurane and the inspired concentration gradually increased until a burst suppression electroencephalogram (EEG) was recorded. End‐tidal isoflurane concentration ( F ) was then allowed to equilibrate for 30 minutes after which the eyelashes, cornea, nasal septum, anus, interdigital skin fold, periople, tail and claw were stimulated. The motor response to stimulation at each location was graded from 0 to 5. End‐tidal isoflurane concentration was decreased 0.3% and the areas re‐stimulated; this was repeated three times in each pig. A linear regression analysis using response as dependent and anaesthetic level as independent variable was performed for each stimulus in each pig. Using Student's t‐statistic a 95% confidence interval for the mean slope of each stimulus was constructed. Results No pig responded to eyelash brushing. The mean slopes for the other stimuli indicated increasing responses with decreasing F . Responses to periople pinching and tail and claw clamping showed significant increases. No stimuli consistently increased the magnitude of response in all pigs, and the appearance and absence of a response was inconsistent between pigs. Motor responses occurred in at least one pig during isoflurane burst suppression anaesthesia to all stimuli except eyelash brushing. Conclusions All the stimuli investigated may elicit movement responses during burst suppression anaesthesia with isoflurane except eyelash brushing. No consistent response pattern between pigs was observed with decreasing isoflurane concentration. Of the stimuli evaluated, clamping the tail or claw and pinching the periople appear the most reliable indicators of anaesthetic depth. Clinical relevance The absence or presence of single reflexes does not accurately reflect the degree of isoflurane‐induced cortical depression in individual pigs. 相似文献

The effects of epidural administration of 250 μg/kg xylazine on EEG responses to surgical stimulation of 5 different intensities were evaluated during isoflurane anaesthesia for an experimental orthopaedic procedure in dogs. The dogs were assigned randomly to one of 2 treatment groups receiving either xylazine (n = 4) or equal volumes of sterile water (n = 4) (control group) epidurally. Intense surgical stimulation during removal of a bone graft from the dorsoiliac spine of the ileum was associated with a significantly (P = 0.0339) higher increase in EEG alpha/delta ratio after epidural administration of sterile water than after epidural injection of 250 μg/kg of xylazine. In addition, the preincision baseline values for 80% spectral edge frequency were significantly (P = 0.0339) lower in the xylazine group compared to control dogs. Our results suggest that epidural administration of 250 μg/kg of xylazine during orthopaedic procedures in dogs exerts antinociceptive effects which may be in part mediated by a supraspinal effect of xylazine. 相似文献

Objective To evaluate electroencephalographic indices, invasive arterial blood pressure and pulse rate as indicators of nociception during isoflurane anaesthesia in pigs. Animals Ten Norwegian Landrace pigs, five castrated males and five females, weighing between 19 and 29 kg. Materials and methods The pigs were anaesthetized with isoflurane and prepared for the measurement of heart rate (HR), mean arterial pressure (MAP) and the electroencephalogram (EEG). The inspired isoflurane concentration (Fi ′ISO) was then adjusted until the suppression ratio (SR) exceeded 20%, after which the corresponding end‐expired concentration (Fe ′ISO) was maintained for 30 minutes. The effects of noxious stimuli, consisting of pinching (nasal septum, interdigital web, anus, periople) and clamping (tail, claw) on spectral edge frequency 95%, median frequency, alpha/delta ratio, beta/delta ratio, theta/delta ratio, total power, suppression ratio, MAP and HR were recorded. Fe ′ISO was then decreased by 0.3% and maintained for 30 minutes after which noxious stimulation was re‐applied. This was repeated twice with Fe ′ISO reductions of 0.3% being applied each time. Changes in measured variables before and after noxious stimulation were then compared at each of four levels of anaesthesia. Results Noxious stimulation caused significant MAP increases at the lowest two levels for Fe ′ISO. There were no significant changes in the other variables examined. Conclusion Of the variables examined, mean arterial blood pressure is the most sensitive indicator of nociception in isoflurane‐anaesthetized pigs. Clinical relevance When evaluating nociception under isoflurane anaesthesia in pigs mean arterial blood pressure should be monitored when surgery is performed. 相似文献

OBJECTIVE: To study pulmonary gas exchange and cardiovascular responses to sedation achieved with romifidine and butorphanol (RB) alone, or combined with acepromazine, and during subsequent tiletamine-zolazepam anaesthesia in horses. ANIMALS: Six (four males and two females) healthy Standardbred trotters aged 3-12 years; mass 423-520 kg. STUDY DESIGN: Randomized, cross-over, experimental study. MATERIALS AND METHODS: Horses were anaesthetized on two occasions (with a minimum interval of 1 week) with intravenous (IV) tiletamine-zolazepam (Z; 1.4 mg kg(-1)) after pre-anaesthetic medication with IV romifidine (R; 0.1 mg kg(-1)) and butorphanol (B; 25 microg kg(-1) IV). At the first trial, horses were randomly allocated to receive (protocol ARBZ) or not to receive (protocol RBZ) acepromazine (A; 35 microg kg(-1)) intramuscularly (IM) 35 minutes before induction of anaesthesia. Each horse was placed in left lateral recumbency and, after tracheal intubation, allowed to breathe room air spontaneously. Respiratory and haemodynamic variables and ventilation-perfusion (; multiple inert gas elimination technique) ratios were determined in the conscious horse, after sedation and during anaesthesia. One- and two-way repeated-measures anova were used to identify within- and between-technique differences, respectively. RESULTS: During sedation with RB, arterial oxygen tension (PaO(2)) decreased compared to baseline and increased mismatch was evident; there was no O(2) diffusion limitation or increase in intrapulmonary shunt fraction identified. With ARB, PaO(2) and remained unaffected. During anaesthesia, intrapulmonary shunt occurred to the same extent in both protocols, and mismatching increased. This was less in the ARBZ group. Arterial O(2) tension decreased in both protocols, but was lower at 25 and 35 minutes of anaesthesia in RBZ than in ARBZ. During sedation, heart rate (HR) and cardiac output (Qt) were lower while arterial-mixed venous oxygen content differences and haemoglobin concentrations were higher in RBZ compared with ARBZ. Total systemic vascular resistance, mean systemic, and mean pulmonary arterial pressures were higher during anaesthesia with RBZ compared to ARBZ. CONCLUSIONS AND CLINICAL RELEVANCE: Acepromazine added to RB generally improved haemodynamic variables and arterial oxygenation during sedation and anaesthesia. Arterial oxygenation was impaired as a result of increased shunt and mismatch during anaesthesia, although acepromazine treatment reduced disturbances and falls in PaO(2) to some extent. Haemodynamic variables were closer to baseline during sedation and anaesthesia when horses received acepromazine. Acepromazine may confer advantages in healthy normovolaemic horses. 相似文献

Anemon I is a new monitoring system that can be used to evaluate autonomic nervous system reactivity in real time by showing a simple, easily interpretated quantitative index (0–200), the Anemon Index (AI) ( Junke et al. 2000 ). This study used the AI to evaluate the quality of analgesia during sevoflurane and fentanyl anaesthesia in pigs. Six healthy pigs, weighing 24.76 ± 3.40 kg, were induced to anaesthesia with 5% sevoflurane (SEVO) in 5 L minute?1 oxygen. After endotracheal intubation SEVO was given at 1 MAC (2.66%) in 3 L minute?1 oxygen. Fentanyl was infused IV at 50 µg kg?1 hour?1 for the first 30 minutes of anaesthesia, discontinued for 30 minutes, and then infused at 100 µg kg?1 hour?1 for another 30 minutes. Three mechanical noxious stimuli (needle prick, pin‐prick and pressure on the abdomen) were applied for 15 seconds at 30, 60 and 90 minutes. The AI, ECG, invasive mean arterial blood pressure (MAP), heart rate (HR), SpO2 by pulse oximetry, tidal volume, Fe′sevo , Fe ′CO2 and respiratory rate were recorded before induction (baseline), after induction, after intubation and extubation, and before and during noxious stimulation at 30, 60 and 90 minutes. Recovery times were recorded. Statistically significant differences were determined by anova . Spearman rank‐correlation was used to evaluate the relationship between AI and hemodynamic variables. A p‐value of < 0.05 was considered significant. A significant (p < 0.01) decrease in AI was recorded after anaesthetic induction, from 82.3 ± 21.1 to 52.7 ± 20.3. After intubation, AI increased slightly, but not significantly, to 71.7 ± 27.1. A significant (p < 0.05) increase of AI occurred after extubation. Nociceptive stimuli did not have any measurable effect either on AI or on recorded cardiovascular variables. There was no movement, respiratory changes, or any other visible response to noxious stimulation. The AI did not change significantly with the different doses of fentanyl. Respiratory depression and apnoea were seen in all animals during the fentanyl infusion; therefore, pigs received intermittent positive pressure ventilation. Anaesthesia with sevoflurane and fentanyl resulted in a significant (p < 0.001) decrease in MAP. Heart rate did not change significantly. There was no correlation between AI and cardiovascular variables (HR and MAP). Endotracheal intubation caused an increase and extubation a greater significant increase in the AI. This suggests that intubation and extubation may represent stressful events during general anaesthesia, although further studies are needed to validate the use of the AI in pigs. Sevoflurane anesthetic induction may not prevent the sympathetic stimulus caused by endotracheal intubation in pigs, as indicated by the increased AI values. 相似文献

Assessment of a complement fixation test to detect Mycoplasma hyopneumoniae infection in pigs 总被引:2,自引:0,他引:2

SUMMARY Serums from pigs slaughtered at abattoirs were tested for evidence of Mycoplasma hyopneumoniae infection using a complement fixation (CF) test which avoids the procomplementary effect of pig serum. To establish a diagnosis of enzootic pneumonia, the lungs from all sampled pigs were examined for pathological and histological changes consistent with the disease and cultures were made for mycoplasmas and bacteria. The study was carried out at Parkville and Bendigo 160 km apart at different times and all serums were tested at both laboratories. The results agreed closely. Thirty-six of 97 pigs at Parkville and 46 of 99 at Bendigo had enzootic pneumonia. About 80% were positive in the CF test. Sixteen per cent of porkers and 36% of baconers gave false negative reactors, that is, a negative test though lesions were present. About 18% to 36% gave false positive reactions but the level in the porkers in the Bendigo group was significantly higher (p<0.02). Possible explanations include, for the false negatives, loss of reactivity caused by circulating antigen and for the false positives, cross reacting antibody produced by another infection or failure to appreciate that lesions of EP were present in lungs because either they were not identified as such or they were not detected. The validity of any serological test for this disease cannot be established while there is a possibility that the present methods used for diagnosis, gross and microscopic examination and recovery of M. hyopneumoniae, fail to detect some infected animals. Other criteria may have to be adopted. 相似文献

The cardiovascular effects of non-abdominal and abdominal surgery during isoflurane anaesthesia (A-group) or isoflurane anaesthesia supplemented with either epidural ropivacaine (AR-group; 0.75 % solution, 0.2 ml/kg) or morphine (AM-group; 0.1 mg/kg diluted in saline to 0.2 ml/kg) were evaluated in 28 healthy pigs with a mean body weight of 30.3 kg SD +/- 4.1 during surgical devascularisation of the liver. Anaesthesia was induced with the intramuscular injection of midazolam (0.3 mg/kg) and ketamine (10 mg/kg). Anaesthesia was deepened with intravenous propofol to enable tracheal intubation and maintained with isoflurane on a circle rebreathing circuit. The vaporiser was set at 2.5% for the A-group and 1.5% for the AR- and AM-groups. Differences between treatment groups were not statistically significant (P > 0.05) for any of the variables. Differences between AM- and AR-groups were marginally significant heart rate (HR) (P = 0.06) and mean arterial blood pressure (MAP) (P = 0.08). Within treatment groups, differences for the A-group were statistically significant (P < 0.05) between non-abdominal and abdominal surgery for HR, systolic blood pressure, diastolic blood pressure (DIA) and MAP. Within the AM-group differences were statistically significant (P < 0.05) for DIA and MAE and within the AR group differences for all variables were not statistically significant (P > 0.05). It was concluded that in isoflurane-anaesthetised pigs, the epidural administration of ropivacaine decreased heart rate and improved arterial blood pressure during surgery. 相似文献

Romifidine as a premedicant to propofol induction and infusion anaesthesia in the dog 总被引:3,自引:0,他引:3
The effects of premedication with four different intravenous doses of romifidine (20, 40, 80 and 120 (μg/kg body weight) and a saline placebo were compared in a group of 20 adult beagles of both sexes, undergoing anaesthesia with propofol for a clinical dental procedure. Anaesthesia was induced 10 minutes after premedication and maintained by intravenous infusion of propofol for a period of 30 minutes. Romifidine had a marked synergistic effect with propofol and reduced the required induction and infusion doses by more than 60 per cent for a standard level of anaesthesia; the synergistic effect was dose related. Following premedication, propofol produced no significant alteration of respiratory rate, heart rate or rectal temperature. Anaesthesia was found to be more stable following romifidine premedication at all doses studied. The quality of induction was unaltered by the dose of the romifidine. Recovery from anaesthesia was smooth and of a similar quality in all cases. There were no differences in the recovery times between the unpremedicated group and the dogs premedicated with any dose of romifidine studied. There were no adverse effects noted following this anaesthetic regimen. The marked dose-related synergism with propofol induction and infusion anaesthesia is relevant should romifidine be used in the dog in clinical veterinary practice. 相似文献

The objective of the study was to investigate the hypothesis that perioperative lymphocytopenia was due to apoptosis of these
cells induced by either halothane or epidural anaesthesia in dogs. The relationship between apoptosis induction and plasma
concentrations of the stress hormone cortisol and the cytokines TNF-alpha and IL-10 was examined as well. The study was performed
on 22 healthy mongrel dogs, equal numbers from both genders, weighing 18.3 ± 2.9 kg, and aged between 3–5 years. Dogs were
divided in three groups. Eight of the animals were anaesthetized with halothane, another eight received epidural anaesthesia
using lidocaine, and six served as controls. Venous blood samples were obtained immediately before (0 minute) anaesthesia,
during deep anaesthesia (120 minute), and on the next day (24 hour) in order to determine the following parameters: the total
lymphocyte counts, the percentage of apoptotic peripheral blood mononuclear cells (PBMC) by flow cytometry, plasma concentrations
of the cytokines tumor necrosis factor – alpha (TNF-alpha) and interleukin – 10 (IL-10) by enzyme-linked immunosorbent assay
(ЕLISA), and plasma cortisol levels by radioimmune assay. Both halothane and epidural anaesthesia in dogs induces apoptosis
of PBMC with slight decrease in total lymphocyte counts. These immunomodulatory effects were transient and faded till the
24th hour. Concerning the mechanism of inducing lymphocyte apoptosis by general or epidural anaesthesia, it seemed that neither
cortisol, nor the tested cytokines TNF-alpha and IL-10 were implicated in this process. Further investigations are necessary
to confirm this assumption. 相似文献

OBJECTIVE: Evaluation of the LMA-ProSeal for positive pressure ventilation (PPV) in the pig. STUDY DESIGN: Prospective observational study. ANIMALS: Twelve German country pigs, weighing 25-62 kg. METHOD: Lungs of pigs were mechanically ventilated under general anaesthesia using the LMA-ProSeal. The ease of insertion, number of attempts and total time until placement of the LMA-ProSeal and gastric tube were recorded. Bronchoscopy was performed to determine the position of the LMA-ProSeal and to detect signs of aspiration. Ventilation variables and the leak airway pressure (P(leak)) were measured. An arterial blood gas sample was taken to determine the adequacy of ventilation. RESULTS: The airway was secured in all pigs within 39 +/- 19 seconds (27-51). Different sizes of LMA-ProSeal were used; up to 30 kg: size 3, up to 43 kg: size 4; and above 43 kg: size 5. In all but one animal the P-LMA and gastric tube were inserted at the first attempt. In nine animals gastric fluid was drained through the gastric tube. There was no evidence of aspiration in any animal. The mean [+/-SD (95%CI)]P(leak) was 28.8 +/- 7.5 cm H(2)O (24.06-33.60) and normal ventilation was achieved in all animals. CONCLUSIONS: The results of this study indicate that the airway of pigs weighing 25-62 kg can be secured safely and reliably with the sizes 3, 4 and 5 LMA-ProSeal. CLINICAL RELEVANCE: Endotracheal intubation in pigs can be difficult so there is a risk of hypoxemia in the apnoeic animal. With the LMA-ProSeal the airway can be secured rapidly, safely and reliably. Use of the Standard-LMA under PPV can be associated with gas leakage into the stomach and the subsequent risk of gastric distension and regurgitation. Both the ability to drain the stomach and the high P(leak) of the LMA-ProSeal could contribute to improved protection against aspiration under PPV. 相似文献

ObjectiveTo evaluate the influence of premedication with tramadol on xylazine–ketamine anaesthesia in young pigs.Study designProspective, randomized, blinded cross-over study.AnimalsTen young Niger hybrid pigs: mean weight 6.1 ± 0.6 kg.MethodsPigs were anaesthetized twice. Xylazine (2.5 mg kg?1), ketamine (25 mg kg?1) and atropine (0.04 mg kg?1) were administered by intramuscular (IM) injection, 5 minutes after either tramadol (5 mg kg?1)) (treatment XKT) or saline (treatment XKS). Time to loss of righting reflex (TLRR), duration of antinociception, duration of recumbency (DR) and recovery times (RCT) were recorded. Quality of induction of anaesthesia including ease of endotracheal intubation was assessed using a subjective ordinal rating score of 1 (worst) to 4 (best). Heart, pulse and respiratory rates, arterial oxygen saturations and rectal temperatures were determined over 60 minutes. Antinociception was assessed by the pigs’ response to artery forceps applied at the interdigital space. Data were compared with Student's t-test, Mann–Whitney's test or analysis of variance (anova) for repeated measures as appropriate and are presented as mean ± standard deviation.ResultsThe quality of anaesthetic induction was significantly better and duration of antinociception significantly longer (p < 0.05) in treatment XKT (3.1 ± 0.7 score; 43.7 ± 15.5 minutes) than in treatment XKS (2.8 ± 0.6 score; 32.0 ± 13.3 minutes). TLRR, DR and RCT did not differ significantly (p > 0.05) between treatment XKT (2.1 ± 0.8, 65.8 ± 17.0 and 13.2 ± 6.7 minutes) and treatment XKS (2.1 ± 1.3, 58.0 ± 14.8 and 10.3 ± 5.6 minutes). Physiological measurements did not differ between the treatments. Conclusion and clinical relevanceTramadol improved the quality of anaesthetic induction and increased the duration of antinociception in xylazine–ketamine anaesthetized young pigs without increasing duration of anaesthesia, nor causing additional depression of the physiological parameters measured. 相似文献

The objective of the study was to examine the relationship between the ‘depth’ of anaesthesia - as determined by clinical signs - and the bispectral index (BIS). Electroencephalograms (EEG)s were recorded in 8 female and 8 castrated male, healthy Norwegian landrace pigs undergoing isoflurane anaesthesia, from which the bispectral index (BIS) was calculated. Isoflurane was delivered in pure oxygen at end-tidal concentrations of 1.6, 1.9, 2.2 and 2.5%, in randomised order, for 30 min after which the EEG was recorded over a 5 min period. Anaesthetic depth was evaluated on a visual analogue scale (VAS) by an experienced anaesthetist. The 95% confidence interval for the mean correlation coefficient between BIS and VAS was calculated to be -0.52–0.30. Confidence intervals (95%) for the mean change in the BIS obtained during the conscious state and that obtained during anaesthesia at different isoflurane concentrations was also calculated. There was a significant decrease in the BIS recorded during consciousness and after 1.6% isoflurane anaesthesia, and between readings after inhalation of 2.2% and 2.5% isoflurane. This indicates that BIS does not accurately reflect ‘depth’ at surgical levels of isoflurane anaesthesia in the pig, and is of no use for this purpose. 相似文献

Objective To compare the distribution of new methylene blue (NMB) solution injected into either the subperiosteal space or under the epidural fat at the first and second lumbar vertebral interspace in cattle. Study design Prospective experimental study. Sample population Nine nonpregnant cows. Methods Needles were advanced towards the first interlumbar epidural space using a dorsal mid‐line approach. Upon confirmation of successful penetration of the ligamentum flavum, group 1 received 5 mL of 0.12% NMB in 0.9% saline. In group 2, the needle was slowly advanced a further 7–10 mm to ensure injection under the epidural fat; the same dose was injected. The extent of cranial and caudal migration of dye, as indicated by staining of epidural fat and the dura mater was measured. Results In group 1 (n = 4), NMB solution migrated only between the periosteum and the epidural fat. There was unilateral distribution in two cows. In contrast, there was no difference between left‐ and right‐sided distribution in group 2 (n = 5), and the NMB solution migrated only between epidural fat and the dura mater. Although the dura mater was not penetrated in any animal of group 2, the dorsal roots were slightly stained and the overlying dura mater was heavily stained in these animals. Conclusions and clinical relevance Variation in distribution of local anaesthetic solution can probably be overcome by deeper needle penetration of the epidural fat. 相似文献

Cryopreservation is a modern technique which assists in the preservation of genetic material from oocytes and embryos for a long time. However, elevated vulnerability to cryopreservation due to the large accumulation of intracellular lipids within oocytes or embryos avoids success of this method. These lipids remain the main crucial factor limiting survival rates of oocytes and embryos after thawing. Lipid ingathering in the oocyte cytoplasm augments lipid peroxidation (LPO) and oxidative stress increases the apoptosis process, declines the viability after thawing, declines cytoskeleton actin filament injuries, lowers the blastocyst rates and reduces cryotolerance in the early stages of embryo development. There have been several attempts to reduce the ingathering of intracellular lipids in oocytes or embryos during the cryopreservation process, in that way enhancing the competence of cryopreserved oocytes or embryos and increasing their viability. One of the most applied agents for chemical delipidation is forskolin. Forskolin exhibited a possible part in improving the oocytes cryopreservation through stimulating cyclic adenosine monophosphate (cAMP) production. The main purpose of cAMP modulation is to provide energy to sustain the mammalian oocytes´ meiotic arrest. The purpose of the existing article is to assess and offer more evidence concerning the forskolin utilization as a modulator of cAMP during the cryopreservation of oocytes and its influence on meiosis completion and the reorganization of cytoplasm, which are prerequisites for the development of oocytes in addition to the contribution to fertilization and subsequently, the development of embryos. 相似文献

The use of porcine small intestinal submucosa as a biomaterial for perineal herniorrhaphy in the dog 总被引:1,自引:0,他引:1
OBJECTIVES: To develop an in vivo perineal hernia model, to develop a technique for using small intestinal submucosa (SIS) in perineal hernia repair, to further elucidate the biological behavior of SIS, and to compare SIS herniorrhaphy with the internal obturator muscle transposition (IOT) technique. STUDY DESIGN: Prospective evaluation comparing SIS herniorrhaphy with IOT. ANIMALS: Twelve adult castrated male, large-breed dogs. METHODS: All dogs had bilateral pelvic diaphragm defects created by complete excision of the levator ani muscle. Each dog had one side repaired using SIS and the other by IOT. Pain and inflammation were subjectively scored. Dogs were killed 2 weeks (n = 4), 12 weeks (n = 4), or 16 weeks (n = 4) after surgery. Each pelvic diaphragm was biomechanically tested to failure. The pelvic diaphragms from 2 normal dogs (n = 4 sides) were also biomechanically tested. Failure site, maximum pressure, displacement at failure, and initial linear stiffness values were determined. Histologic assessment was performed. Statistical analysis was performed with significance set at P <.05 RESULTS: No significant postoperative complications were noted. There were no significant differences in maximum pressure to failure, displacement, or stiffness when comparing normal, SIS, and IOT at any time point. The SIS group had significantly less displacement (P =.004) at 2 weeks than at weeks 12 or 16. For all herniorrhaphy techniques, the failure site was central (n = 22) or at the suture line (n = 2). At 2 weeks, histologic evaluation of tissues from the IOT group showed inflammation, mineralization, and necrosis, which were not present in tissues from the SIS group. Histologic examination at 12 and 16 weeks showed no microscopic differences in cell population or tissue characteristics between the IOT and SIS groups. CONCLUSIONS: SIS herniorrhaphy was successfully performed in this in vivo model of perineal hernia in the dog. CLINICAL RELEVANCE: This study suggests that SIS can be used as a primary means of repair, as augmentation when the internal obturator muscle is thin and friable, or as a salvage procedure in cases of recurrence in dogs with perineal hernia. 相似文献

Measurement of hepatic oxygen extraction was performed on six healthy Greyhound dogs over a two hour period. The Greyhounds were anaesthetised and a right subcostal surgical incision performed. Ultrasonic flow transducers were used to measure flow rate in the hepatic artery and the portal vein. The blood oxygen tensions in arterial blood and in the portal and hepatic veins were also measured. Hepatic oxygen extraction remained stable throughout the study, despite a steady decline in arterial blood pressure. The methodology described in this study provides a direct measure of oxygen uptake by the liver in the dog and could readily be used to investigate hepatic uptake of drugs. 相似文献

ObjectiveTo investigate the relationship between urine specific gravity (USG) and the risk of arterial hypotension during general anaesthesia (GA) in healthy dogs premedicated with dexmedetomidine and methadone.Study designProspective clinical cohort study.AnimalsA total of 75 healthy client-owned dogs undergoing GA for elective tibial plateau levelling osteotomy.MethodsAfter placing an intravenous catheter, dogs were premedicated with dexmedetomidine (5 μg kg–1) and methadone (0.3 mg kg–1) intravenously. After induction of GA with alfaxalone to effect, the bladder was expressed and USG measured. An arterial catheter was placed, and residual blood was used to measure packed cell volume (PCV) and total protein (TP). GA was maintained with isoflurane vaporised in oxygen and a femoral and sciatic nerve block were performed. Arterial blood pressure < 60 mmHg was defined as hypotension and recorded by the anaesthetist. Treatment for hypotension was performed in a stepwise manner following a flow chart. Frequency of hypotension, treatment and response to treatment were recorded. Logistic regression modelling was used to assess the association between USG, TP and PCV and incidence of perioperative hypotension; p < 0.05.ResultsData from 14 dogs were excluded. Of the 61 dogs, 16 (26%) were hypotensive during GA, 15 dogs needed treatment of which 12 were responsive to a decrease in inhalant vaporiser setting. The logistic regression model was not statistically significant (p = 0.8). There was no significant association between USG (p = 0.6), TP (p = 0.4), PCV (p = 0.8) and arterial hypotension during GA.Conclusions and clinical relevanceIn healthy dogs premedicated with dexmedetomidine and methadone and maintained under GA with isoflurane and a femoral and sciatic nerve block, there was no relationship between the specific gravity of urine collected after premedication and intraoperative arterial hypotension. 相似文献
