Sheep from local farms with and without previous exposure to pigs were tested for their skin and airway responses to a commercial Ascaris suum antigen. There was an immediate reaction to intradermal injection of the antigen in 90% of 101 sheep. A bronchial provocation test by aerosol of the same antigen was undertaken on 43 of the sheep with a positive skin reaction. About 70% of sheep showed an immediate airway response to the antigen as an aerosol, reflected as a significant increase in airway resistance and/or decrease of dynamic lung compliance. The mean peak airway resistance and mean lowest dynamic lung compliance were 165% above and 61% below their baselines, respectively. No significant changes were recorded when the same animals were given an aerosol of phosphate buffered saline. Similarly, no correlation was found between the degree of skin reaction and the magnitude of bronchoconstriction (p>0.05). The sheep with previous exposure to pigs showed no significant differences in airway responses to antigen challenge, although they showed significantly greater skin reactions than those without exposure to pigs. These results indicate that the majority of Romney sheep in the Manawatu have a natural skin and airway sensitivity to A. suum antigen and may therefore be used as an animal model to study human airway hypersensitivity. The origin of this sensitivity has yet to be determined.  相似文献

To introduce antigen to the respiratory mucosa, killed Actinobacillus pleuropneumoniae with quil A as adjuvant was administered to pigs as an aerosol. Immunisation by this aerosol induced a marked IgA response in the bronchoalveolar and nasal fluids, and in the serum. Following challenge with live bacteria two weeks after the last exposure to the aerosol, the immunised pigs were protected from the severe pleuropneumonia which developed in non-immunised pigs. The immunised pigs had lower antibody titres in the mucosal fluids and serum after exposure to the challenge. The immune response after experimental infection of non-immunised animals was a weak IgA antibody response in the bronchoalveolar and nasal fluids, whereas the systemic immune response after challenge included both IgA and IgG antibodies.  相似文献

The influence of A. suum infection on the haematology, liver-related serum enzymes, blood urea and live weight gain in Mukota and Large White (LW) weaner pigs was compared. Six pigs of each genotype were infected with a single dose of 4000 A. suum eggs per pig and another six were not. The pigs were kept for 100 days. Blood was collected daily for the first 7 days and also after 100 days. In the infected pigs, there was an increase (p<0.05) in alanine aminotransferase (ALT) activity in the LW but not in the Mukota pigs. Although the alkaline phosphatase (ALP) activity rose (p<0.05) in both infected and non-infected LW pigs from day 1 to day 3, the activity in the non-infected LW pigs then decreased, while that of the infected LW pigs remained elevated. The infected LW pigs had higher (p<0.05) levels of ALT, ALP and aspartate aminotransferase than their non-infected counterparts. Non-infected LW pigs tended to have higher (p<0.05) haematological parameters, daily weight gain and urea concentrations than infected LW pigs, but these differences were not significant. These preliminary findings suggest that more A. suum larvae reached the livers in the LW than in the Mukota pigs and that the latter may be more resistant to A. suum infection.  相似文献

Eighty-six clinically normal adult dogs from southern Florida, USA, with no history of dermatitis, were intradermally skin tested with Greer flea antigen 1:1000 w/v to determine the prevalence of positive immediate intradermal reactivity. This study describes the test group of animals and reports the prevalence of sensitivity to the Greer whole-body flea antigen in normal dogs living in a flea-rich environment. Similar to previous research, the highest prevalence of reactivity to flea antigen occurred at 3–4 years of age. The results indicate that 24% of clinically normal dogs from a flea-rich environment exhibited positive immediate skin test reactivity. These dogs had no clinical signs of flea allergic dermatitis (FAD) in spite of ongoing flea exposure. A 2-year follow-up telephone call to the owners of these flea antigen intradermal skin test (IDST)-positive dogs indicated that only two of 21 dogs had developed clinical signs of FAD.  相似文献

OBJECTIVE: To describe the spectrum of nonspecific airway reactivity in a group of clinically normal foals. ANIMALS: 12 clinically normal mixed-breed foals, 48 to 92 days old, without history of clinical lung disease. PROCEDURE: Nonspecific airway reactivity was determined by measuring the extent of changes in dynamic compliance during nebulization of incrementally increasing concentrations of histamine aerosol. Degree of airway reactivity was expressed as the dose of histamine that evoked a decrease in dynamic compliance (Cdyn) to 65% of the after saline nebulization value (PC65Cdyn) or increase in pulmonary resistance (R(L)) to 135% of baseline (PC135R(L)). RESULTS: In all foals, it was possible to induce a decrease in Cdyn in dose-dependent manner to < or = 65% of baseline. Response of foals in terms of R(L) was more erratic, and, in 1 foal, R(L) decreased after histamine exposure. Mean+/-SD PC65Cdyn was 5.43+/-1.74 (range, 0.77 to 19.56) mg/ml, and mean PC135R(L) was 3.34+/-1.52 (range, -0.749 to 17.35) mg/ml. Body weight was not correlated to baseline Cdyn, R(L), PC65Cdyn, or PC135R(L). CONCLUSIONS AND CLINICAL RELEVANCE: Clinically normal foals had a wide range of airway reactivity, which may contribute to variation in clinical responses of foals to otherwise similar stimuli, such as infection, inflammation, and challenge exposure with environmental irritants.  相似文献

Seven cats diagnosed as flea allergic by specific criteria and seven normal control cats were exposed to flea bites in a controlled manner and were given intradermal injections of 1:1000 w/v flea antigen. Subjective evaluation of gross lesions and documentation of histological changes at flea antigen intradermal skin test (IDST) and flea bite sites were performed at 15 min, 24 h and 48 h after IDST or flea exposure. Control cats did not develop an immediate gross reaction to either flea bites or the intradermal injection of flea antigen. All seven flea-allergic cats had an immediate gross reaction at the site of IDST with flea antigen; five of these cats also developed immediate gross reactions to flea bites. Three of seven flea-allergic cats developed a gross 24 h and/or 48 h delayed reaction at the flea antigen IDST sites. These three and one other cat had both an immediate and delayed gross reaction to flea bites. Histological examination of 15 min skin specimens from IDST and flea bite sites of flea-allergic cats were similar with a mild lymphocytic, histiocytic and mastocytic superficial perivascular dermatitis. Histological examination of 24 h and 48 h skin specimens from IDST and flea bite sites of flea-allergic cats showed that they were often indistinguishable. Histological features of IDST and flea bite sites of flea-allergic cats at 24 h consisted of a perivascular to diffuse predominately eosinophilic dermatitis and mural folliculitis with variable epidermal necrosis and ulceration.  相似文献

Experimental infestations of Sarcoptes scabiei (De Geer) were established to study development of cutaneous hypersensitivity in pigs. Forty-eight pigs in six isolation rooms were used in two trials lasting 51 and 65 days, respectively. Treatments of 0 (control), 100 (low dose), and 1000 mites (high dose) per pig were randomly assigned to rooms. Intradermal skin tests with a sterile mite extract were done weekly to assess hypersensitivity. Control pigs never responded to the extract, whereas most infested pigs progressed through phases of (1) no response, (2) delayed hypersensitivity alone, (3) immediate and delayed hypersensitivity together, and (4) immediate hypersensitivity alone. High-dose pigs developed delayed and immediate hypersensitivity sooner than low-dose pigs (P less than 0.005). When related to cumulative mite-days, a measure of exposure to mite antigens, low-dose and high-dose pigs developed delayed responses after experiencing the same level of exposure. Thereafter, immediate responses developed sooner in low-dose pigs, suggesting that immediate hypersensitivity develops at a rate that is independent of rate of antigen exposure.  相似文献

Reasons for performing study: Studies investigating the clinical efficacy of albuterol administered with the same propellant and commercially available delivery devices in horses with recurrent airway obstruction (RAO) are not currently available. Objectives: To determine the efficacy of aerosolised albuterol administered to horses with RAO by means of 2 commercially available, hand‐held delivery devices. Methods: Ten horses with RAO were kept in a dusty environment and fed mouldy hay to induce airway obstruction. Lung mechanics were measured before and after the procedure. ΔPmax was measured 5 min after administration of 180 µg of albuterol from a pressurised metered dose inhaler, using an aerosol delivery device chosen randomly. This process was repeated every 5 min until maximal bronchodilation was achieved. After a 24 h washout period, lung mechanics data were again collected using the other aerosol delivery device. Results: Aerosolised albuterol induced a significant and rapid bronchodilation in the horses using both aerosol delivery devices. No statistically significant difference in pulmonary function was observed in response to albuterol therapy between the 2 devices. The dose required to achieve 50% of maximal bronchodilation was not statistically different between the 2 devices (173.35 ± 78.35 µg with Device 1 and 228.49 ± 144.99 µg with Device 2, P = 0.26). The decrease in lung resistance tended to be more pronounced after albuterol administration with Device 1 (P = 0.066). Conclusions: Aerosolised albuterol is an effective bronchodilator in horses with recurrent airway obstruction. There is no statistically significant difference between the 2 commercially available aerosol delivery devices in terms of efficacy. Potential relevance: Aerosolised albuterol is effectively delivered using currently available devices leading to maximal bronchodilation in horses with RAO at an average dose of 540 µg.  相似文献

Three techniques to reduce the exposure of humans and pigs to aerosols in pig houses have been investigated; filtration and recirculation of air in weaner rooms (FR), weekly vacuum cleaning in a finishing house (VC) and weekly washing with water of weaned piglets (WA). All experiments have been carried out in rooms for 70 (FR), 96 (VC) or 72 (WA) animals. For FR seven replications, for VC two replications and for WA six replications have been carried out.Average reduction in gravimetrical dried aerosol concentrations was 40% (1.58 vs. 2.65 mg/m3) for FR (P<0.0001), but did not result in significant changes in daily feed intake nor daily gain of piglets during the weaning stage. Incidence of respiratory health disorders was very low in both groups.Average reduction in aerosol concentrations was 6% (2.34 vs. 2.50 mg/m3) for VC (P>0.05). Reductions improved from −2% on the day following vacuum cleaning to over 10% from 4 days after vacuum cleaning onwards. No significant differences in feed intake or daily gain of pigs were observed. Pigs in rooms without vacuum cleaning received more veterinary treatments for respiratory health disorders, resulting in less pigs with lung lesions.Average reduction in aerosol concentrations was 10% (4.17 vs. 4.66 mg/m3) for WA (P<0.05) compared to the reference level. For weekly washing of piglets the reduction in aerosol concentration was highest (25%) on the day of washing. No significant improvements in production of pigs have been found.Costs of reducing aerosol concentrations in pighouses are not compensated by improved production of pigs.  相似文献

200 SPF pigs were infected by aerosol with Mycoplasma hyopneumoniae and the development of clinical signs, serological and pathological reactions were studied. Mean time to onset of coughing was 13 days. A mean delay of 9 days was observed from onset of coughing until seroconversion against M. hyopneumoniae as measured by ELISA. At an individual level, the sensitivity for this ELISA was estimated to 98–100% and the specificity to 93–100%. Pasteurella multocida was isolated from the majority of the lungs 4 weeks post inoculation with M. hyopneumoniae and the lung lesions in pigs were significantly larger when P. multocida was present as compared to pigs with M. hyopneumoniae alone. An evaluation of cultivation, immunofluorescence, ELISA and polymerase chain reaction for demonstration of M. hyopneumoniae in lungs showed that all four methods have a high sensitivity in the acute stages of pneumonia. In the later stages the sensitivity of cultivation was superior to the other methods. No differences in specificity were observed between the methods. The antigen-ELISA OD values and the immunofluorescence scores revealed a strong positive correlation. Nasal swabs were additionally used for demonstration of M. hyopneumoniae and the polymerase chain reaction was found superior to the other methods.  相似文献

Direct application of antigens to skin together with an adjuvant, a procedure called transcutaneous immunization (TCI), can induce systemic immune responses in mice, humans, cats and dogs. In previous studies we found that cholera toxin (CT) applied topically on unbroken skin induces systemic antibody and lymphocyte proliferative responses in sheep. The current study examined whether concurrent administration of CT and tetanus toxoid (TT) delivered transcutaneously could induce specific antibody responses to both antigens in sheep. Antibodies to both TT and CT were induced by TCI although antibody titres in serum to TT were higher in sheep receiving TT plus alum by intramuscular injection (n=5) than TT plus CT by TCI (n=5). The ratio of IgG1/IgG2 antibody to TT in serum was near unity, and the route of immunization, TCI versus injection, did not influence this ratio. In contrast, the ratio of IgG1/IgG2 antibody differed significantly between the two antigens, TT and CT, delivered by TCI, with a higher proportion of IgG1 antibody in serum to CT than TT. Antibody to TT was detected in lung washes from TCI and injection groups, with IgG1 predominating over IgG2 in both groups. IgA antibodies to CT and TT were detected in sera of CT and TT-immunized groups respectively but in lung washes IgA antibody to TT was detected only in the injection group. Results show that TCI induced systemic antibody responses to CT and the co-administered antigen TT, whereas no evidence was obtained for mucosal IgA responses following TCI.  相似文献

In a survey of 2500 market weight pigs in a Saskatchewan abattoir, 37% were infected with adult Ascaris suum and in 46% there were milkspot hepatic lesions. A total of 60% of the pigs examined had some evidence of A. suum infection. Most infected pigs contained less than 50 adult ascarids.

In a second abattoir survey of a further 2500 market weight pigs, 44% of the animals had milk-spot hepatic lesions. In approximately 12% of the affected pigs these lesions were very severe and a very similar proportion of pigs' livers were condemned or sent for use as animal feed.

The results of these surveys are discussed with particular reference to the significance of A. suum infection in growing pigs in Saskatchewan.

Caseous lymphadenitis (CLA) infection was established experimentally in a total of 30, 2-tooth merino wethers. In the first experiment a broth culture of the causative organism, Corynebacterum pseudotuberculosis was placed directly on to a freshly shorn area on the right shoulder of 21 sheep after pretreatment with a defatting agent, water or 4 different commercial sheep dips. Within 6–13 days focal skin lesions appeared on 7 sheep and these were followed by abscessation of the right prescapular lymph node. Lung lesions of CLA developed in 6 sheep; 5 of which had skin lesions. In the second experiment C. Pseudotuberculosis broth culture was applied over the right shoulder region of 14 sheep, 24 hours post shearing. The 7 animals had only 1 application of the culture and the remaining 7 had 4 applications on consecutive days. A total of 9 of the 14 sheep developed skin lesions and subsequent CLA in the right prescapular lymph nod but there was no significant difference between the 2 methods of application. As part of the second experiment a broth culture of C. pseudotuberculosis was added to a commercial arsenical sheep dip and 2 hours later sprayed on to the right side of sheep shorn 24 hours or 2 weeks previously. Within 6–13 days all 14 sheep treated in this way developed skin lesions from which C. pseudotuberculiosis was recovered in dense pure growth. Healing time of skin lesions in all affected sheep ranged from 17–72 days post inoculation. A serological test to detect the presence of C. pseudotuberculosis antitoxin was used at regular intervals on all experimental sheep. The earliest positive reaction in an affected sheep was recorded 4 weeks post inoculation. In 23 known affected sheep tested at 35 weeks post inoculation, 10 were positive to the Zaki test. No false positive reactors were recorded. The localisation of focal lung lesions in the interstitial tissue suggests that pulmonary abscessation caused by C. pseudotuberculosis develops via the haematogenous route.  相似文献

During the last 30 years, pig production in Uganda and neighbouring counties has increased markedly. Pigs are mainly kept as a source of income for small-scale farmers; however, the pig production is subject to several constraints, one of them being worm infections. A study was carried out in rural communities in Kabale District in the South Western part of Uganda in September and October 2007 in order to estimate the prevalence of gastrointestinal nematode parasites in pigs based on coprological examination. Fifty-six households were randomly selected and visited. Housing system and deworming history were recorded. Faeces was sampled from rectum of one to five pigs (age, 3–12 months) per household. A total of 106 pigs were examined coprologically of which 91% excreted nematode eggs. The following prevalences of nematode eggs were recorded: strongyles (89%), Ascaris suum (40%), Trichuris suis (17%) and spiruroid eggs (48%). On household level, rearing pigs on slatted floors in pens significantly reduced the faecal egg excretion of strongyle eggs with almost 80% (p = 0.010) and a significant interaction between floor type and anthelmintic treatment was found for spiruroids (p = 0.037). Fifteen T. suis egg positive pigs were selected for post-mortem examination of the gastrointestinal tract. The post-mortem examinations revealed that 93% pigs were infected with Oesophagostomum spp. (worm burden, min–max 10–2,180), 73% with A. suum (1–36), 67% with T. suis (6–58), and 20% with Hyostrongylus rubidus (worms not quantified). In general, nematode infections were widespread and polyparasitism common in pigs in Uganda. However, worm burdens were moderate which may be related to recent deworming or to the practice of rearing pigs on slatted floors in wooden elevated pens.  相似文献

Three experiments were performed to evaluate the immunogenic potency of a soluble fraction ofPasteurella haemolytica against pneumonic pasteurellosis in calves. A soluble antigen was extracted by a 2.5% saline solution fromP.haemolytica. Weaned Holstein bull calves, seronegative for infectious bovine rhinotracheitis virus (IBRV) and the pasteurella antigen, were vaccinated either by repeated subcutaneous (SC) vaccination, or by exposure 3 times to the aerosol ofP.haemolytica antigen. Challenge exposure to aerosol ofP.haemolytica was preceded by infection with IBRV, or in experiments 2 and 3, the virus exposures were combined with a stress treatment. The lung lesions were examined at necropsy 3 to 8 days post infection. In the first experiment, all the vaccinated calves produced specific antibody response to the pasteurella antigen, and none of the calves including controls showed significant lesions in the lung. In the second experiment 2 aerogenically vaccinated calves had no lesions. One of the two SC-vaccinated calves had mild consolidated lesions. Two control calves, one of which died 3 days following the challenge, developed severe fibrinous pneumonia with consolidation of 50% or more of the lung surfaces.P.haemolytica was isolated only from the 2 control animals. In the third experiment, 2 of the 3 control calves developed moderate to severe consolidation, butP.haemolytica was isolated only from one of them. Two of the three aerosol-vaccinated calves also developed significant lesions and one of them yielded the bacteria from the lung. Three SC-vaccinated calves had slight lesions and the organism was not isolated from their lungs. The results did not consistently indicate an immunogenic potential of the soluble antigen againstP.haemolytica-related pneumonia. The effect of stress on the pathogenesis of bovine viral penumonia and correlation between pneumonic lesions and antibacterial resistancein situ are discussed.  相似文献

Two enzyme-linked immunosorbent assays to measure antibody to the cell wall antigen (C-ELISA) and toxin antigen (T-ELISA) of C. pseudotuberculosis were evaluated on serum from 6 separate groups of sheep. For sheep naturally infected with C. pseudotuberculosis the sensitivity and specificity of the C-ELISA was 76% and 73% respectively and for the T-ELISA 67% and 77% respectively. For sheep slaughtered one year after artificial infection the sensitivity of both tests was greater than or equal to 83% and the specificity greater than or equal to 72%. For sheep slaughtered 4 months after artificial infection the specificity of both tests was less than 30% while the mean sensitivity was 85%. The C-ELISA in conjunction with the T-ELISA detected 92% of sheep with lung lesions.  相似文献

An indirect enzyme-linked immunosorbent assay (iELISA) was evaluated for its diagnostic capability in detecting antibodies against Fasciola gigantica infection in cattle, sheep and donkeys sera using crude worm, excretory–secretory and glutathione S-transferase antigens prepared from adult liver fluke. Presence of F. gigantica worms at post-mortem examination of cattle, sheep and donkey’s livers was taken as a gold standard for the evaluation of the assay. The diagnostic sensitivity, specificity and accuracy percentages of iELISA were determined for each antigen. Excretory–secretory antigen gave the best results for the serodiagnosis of F. gigantica infection in cattle, sheep and donkeys using iELISA with diagnostic sensitivity percentages of 93.3%, 94.9% and 93.3%, respectively, while the specificity percentages were 96.7%, 97.2% and 96.3%, respectively, whereas the accuracy percentages were 95%, 96% and 95.7%, respectively. The diagnostic sensitivity percentages of iELISA using crude worm antigen were 96.7%, 100% and 93.3%, respectively, while the specificity percentages were 80%, 83.3% and 85.2%, respectively, whereas the accuracy percentages were 88.3%, 86.7% and 87%, respectively. The diagnostic sensitivity percentages of iELISA using glutathione S-transferase antigen were 66.7%, 71.8% and 60%, respectively, while the specificity percentages were 70%, 77.8% and 77.8%, respectively, whereas the accuracy percentages were 68.3%, 74.7% and 73.9%, respectively. Conclusively, excretory–secretory antigen dependent iELISA can be used as a reliable serodiagnostic test for F. gigantica infection in cattle, sheep and donkeys.  相似文献
