Lymphocyte-mediated cytotoxicity is essential to control viral infections, limit lymphocyte expansion and activation, and survey for malignant cells. Humans with defects in lymphocyte cytotoxicity have reduced perforin function resulting in uncontrolled lymphocyte expansion, leading to excessive histiocyte activation and a hemophagocytic disorder. Dog breeds such as Bernese mountain dogs (BMD) have a high incidence of reactive and malignant diseases affecting histiocytes. This study addressed the hypothesis that changes in the perforin gene contribute to the development of hemophagocytic histiocytic sarcoma (HHS) in BMD. Canine perforin DNA was amplified and sequenced through multiple PCR assays from healthy and diseased dogs, and the gene structure determined by rapid amplification of cDNA ends. The coding component of the gene consists of 1679 bp, with two exons of 536 bp and 1143 bp separated by an intron of 865 bp. Gene configuration and location differ from that in other species although the coding sequence is highly conserved. Three silent single nucleotide polymorphisms (SNP) were identified. Analysis of their distribution indicated a consistent genotype among 6 middle-aged to older BMD without histiocytic diseases. Among samples from 10 dogs with HHS and 10 without histiocytic diseases SNP occurred with variable frequency. It was concluded that changes in the amino acid sequence of perforin were not associated with HHS but that a constellation of SNP may characterize BMD without histiocytic disease. Investigation of more dogs is required to confirm a specific genotype. Future studies should focus on the potential contribution of reduced perforin expression and/or function to HHS in dogs.  相似文献

The chemical constituents of 171 specimens of canine urinary calculi have been analysed by infrared spectroscopy. The analysis revealed that 46 per cent of the total contained struvite as the major component; 26 per cent contained cystine; 16 per cent were found to be composed of whewellite and, or, weddellite; 8 per cent of urates; 3 per cent of calcium phosphate (apatite and, or, brushite) and 1 per cent of silica. A series of selected samples was also studied by scanning electron microscopy and energy dispersive X-ray analysis (EDAX). This examination revealed structures similar to human stones. Potassium and complex urates containing two or more cations were commonly found in many areas of ammonium urate and sodium urate calculi. Moreover, EDAX analysis detected a calcium enriched urate in several zones of urate calculi.  相似文献

Most disorders of the equine cervical oesophagus present with a similar range of clinical signs, resulting from acute, chronic or recurrent impediment to the transit of ingesta and saliva. These include profuse frothy nasal discharge and dysphagia with gagging and coughing. Rupture of the cervical oesophagus is characterised by a painful ventral swelling at the affected site with crepitus, cellulitis and potential drainage of necrotic material. Disorders of the thoracic and abdominal segments of the oesophagus are less well recognised and clinical signs are more insidious in onset. Catastrophic damage to the thoracic oesophagus may occur without previous signs of obstruction. Pyrexia, tachycardia, tachypnoea and vague colic signs may be the first manifestations of perforation of the thoracic oesophagus, resulting from a progressive septic pleural effusion. Rupture of the thoracic oesophagus should be a differential in any horse presenting with an unexplained pleural effusion and ruled out by careful endoscopic examination.  相似文献

A six‐year‐old mixed‐breed male dog weighing 7.0 kg was presented with chronic vomiting and regurgitation. Endoscopic examination revealed prominent oesophageal dilation in the thoracic region, multiple small greyish‐white nodules over the oesophageal lumen and cauliflower‐like masses in the caudal oesophagus. Histopathological studies revealed a characteristic pattern of coexisting elements of infiltrating adenocarcinoma and squamous cell carcinoma. Immunohistochemical staining with anti‐cytokeratin AE1 + AE3 was positive in both types of neoplastic cells. Neoplastic glandular cells stained positively for cytokeratin 8 while neoplastic squamous cells stained positively for cytokeratin 5/6. On the basis of these findings, the dog was diagnosed with oesophageal adenosquamous carcinoma. The case history and findings suggest that the malignancy might have developed from Barrett's oesophagus following irritation of the oesophageal mucosa due to chronic vomiting and regurgitation.  相似文献

A 2‐month‐old filly was evaluated for severe colic. Ultrasound, abdominocentesis and physical examination findings prompted an abdominal exploratory surgery. Perforation of the stomach was discovered during the surgery. The filly was humanely subjected to euthanasia under anaesthesia and post mortem examination was performed. In addition to gastric and duodenal ulceration, a thickened, black area of the proximal oesophagus was discovered. Histopathology of the lesion revealed primary malignant melanoma. Although rare, primary melanoma can occur in noncutaneous locations.  相似文献

The effects of stretch and force‐feeding on some physiological properties of the isolated oesophagus of geese was examined. Elasticity was measured at various points, pressure‐volume relations determined and histological sections examined.

It was found that the total oesophageal volume was increased markedly by force‐feeding, particularly in the area of the crop, although the elasticity was unchanged. Enlargement was due to hypertrophy. Pressure‐volume relations indicate that a very large oesophageal volume can be attained at pressures up to 40 mmHg. It is concluded that force‐feeding does not damage the physiological characteristics of the oesophagus, and that efficient filling may be obtained at relatively low pressures.  相似文献

Idiopathic muscular hypertrophy of the oesophagus (IMHO) is reported in equids as an incidental post mortem finding, infrequently associated with clinical signs of oesophageal dysfunction or additional oesophageal abnormalities. Clinicopathological and post mortem findings are presented from a 15‐year‐old gelding that developed a spontaneous perforation of the distal, intrathoracic oesophagus, in association with IMHO. The majority of oesophageal perforations documented in the equine literature have been related to external or iatrogenic trauma resulting in rupture of the cervical portion of the oesophagus. In this case, the early presenting features included pyrexia and inappetance, and progressed over a 4 day period to include signs of tachypnoea, tachycardia and endotoxaemia due to the development of a fatal septic pleuritis and pleural effusion. Oesophageal obstruction was not a presenting feature. Diagnostic investigation in cases of unexplained septic pleural effusion should include oesophageal endoscopic evaluation to rule out perforation of the intrathoracic oesophagus. IMHO may represent a clinically significant, primary disease entity with the potential to lead to oesophageal perforation.  相似文献

The oesophagus is a muscular tube comprised of cervical and thoracic regions. Several studies have clarified the histological structure of the oesophagus. However, its histoarchitecture in relation to variable dietary habits of each species is still unclear. In the current study, 21 pigeons, cattle egrets and ducks, n = 7, each was used. Macroscopically, the oesophagus of cattle egrets either the cervical or thoracic parts was the longest among the pigeons and ducks. Histologically, the oesophagus comprised of four distinct tunicae: mucosa, propria submucosa, musculosa and adventitia or serosa. A great structural variation in these layers among the three investigated species was recorded. In the cervical oesophagus of pigeons, the superficial squamous cells showed perinuclear halo zone, the propria submucosa was characteristically lacked any gland. Moreover, its musculosa was very thick. On the other hand, the intraepithelial glands were characteristically distributed along the whole length of the cattle egret’s oesophagus. Interestingly, the cervical esophagus of the ducks showed submucosal associated lymphatic tissue; diffuse and nodular Ultrastructurally, the oesophageal glands showed secretory granules of variable electron densities, electron - lucent in the pigeons and ducks and electron - dense in the cattle egrets.  相似文献

The gastrointestinal tract of the snake has various distinctions from mammals, birds and other reptiles. Five gopher snakes (Pituophis catenifer) were studied in relation to the gross anatomical measurements of visceral organs relative to body length as well as the histology of the oesophagus and the stomach. The oesophagus closely resembles the empty stomach upon gross examination. A small palpable thickening was observed at the junction of the oesophagus with the stomach. In both the oesophagus and the stomach, there were cellular and structural differences observed in the tunica mucosa which can be linked to the feeding habits and natural biology of the snake when compared to those of mammals. Both oesophagus and stomach were lined by simple columnar to pseudostratified columnar epithelium. There were no glands in the wall of the oesophagus. Scattered ciliated triangular cells (brush cells) were present among the columnar epithelial cells in the distal portion of the oesophagus. The stomach can be divided into three portions (proximal, middle and distal). The stomach has a small non-glandular portion with low folds. After this small non-glandular portion, glands started to appear and gradually increase in quantity. The largest quantity of glands appeared in the middle portion of the stomach with more branching folds resulting in a decrease in the lumen diameter. The tunica muscularis increased in thickness at the oesophageal and the pyloric-duodenal junctions. Positive statistical correlations were established in thickness of the tunica muscularis between proximal and distal portions of the oesophagus and the stomach.  相似文献
