The efficacy of an ivermectin jetting fluid against the sheep body louse, Bovicola (Damalinia) ovis, in long-woolled sheep was investigated in three field trials. In all trials, louse populations in hand-jetted sheep were reduced to nondetectable numbers within 2 weeks of treatment. Bv comparison, shorn and unshorn control sheep maintained detectable infestations for the duration of the trials. 相似文献

Groups of sheep infested with strains of Bovicola (Damalinia) ovis were obtained from flocks either with a history of failure to control lice with synthetic pyrethroid (SP) pour-on insecticides, or from farms where SP compounds were not used. The sheep were treated according to the manufacturer's recommendations with registered "off-shears" SP formulations. All treatments were applied under ideal conditions with doses calculated on an individual body weight basis and applied to the dorsal mid-line from the base of the neck to the butt of the tail. Treated sheep were kept in pens and maintained in separate groups. The pour-on SP treatments significantly reduced the lice population but failed to eliminate the infestation in 7 of 13 experiments in sheep carrying strains of lice with resistance factors of greater than 4 to at least one of the SP compounds. Failures occurred with all three of the SP pour-ons currently registered for lice control in NSW and with both water-based and organic solvent-based formulations. 相似文献

AIM: To investigate the efficacy of pour-on anthelmintics against field strains of parasitic nematodes in young cattle on five farms in New Zealand. METHODS: Faecal nematode egg count (FEC) reduction (FECR) tests were carried out on five calf-rearing farms using pour-on formulations of levamisole, ivermectin, eprinomectin, and the simultaneous administration of levamisole and ivermec- tin. Faecal samples were collected per rectum before treatment and about 7, 14, 21 and 28 days after treatment, for FEC and faecal nematode larval culture. RESULTS: Resistance (i.e. <95% reduction in FEC) of Cooperia oncophora to ivermectin and eprinomectin was identified on all five farms. There was limited evidence of possible emerging resistance in Ostertagia spp to ivermectin but not eprinomectin, in short-tailed larvae of Cooperia spp to ivermectin and eprinomec- tin, and in Trichostrongylus spp to ivermectin, eprinomectin and levamisole used separately. Levamisole was effective against C. oncophora, but had variable efficacy against Ostertagia spp in the calves in this study. Simultaneous treatment with levamisole and ivermectin pour-on formulations were effective against all genera on all farms. CONCLUSIONS: To effectively manage roundworm parasites in their calves farmers need to be aware of the resistance status of the parasites on their farms. Levamisole is likely to be an effective anthelmintic on most farms at times of the year when the impact of Ostertagia spp is not high. Simultaneous administration of levamisole and ivermectin pour-on anthelmintics to cattle is likely to control both ML-resistant C. oncophora and stages of Ostertagia spp that are not controlled by levamisole alone. 相似文献

Abstract AIM: To describe the seasonal pattern of development of third-stage infective larvae (L3) from eggs of Teladorsagia (=Ostertagia) circumcincta, Trichostrongylus colubriformis and Haemonchus contortus on pasture in the North Island of New Zealand. METHODS: Sheep faeces containing known numbers of eggs of all three nematode species were deposited on, or buried in, pasture plots at three sites, viz coastal Manawatu, Upper Hutt Valley, and East Cape hill country. Development was measured by recovering L3 from faeces, herbage and soil 28–31 days after deposition on 13–18 occasions, between January 2005 and July 2006. Analysis of the number of larvae recovered used a mixed model including number of eggs deposited, weight of faeces recovered (an assumed indicator of earthworm activity), site, contamination date, and position of deposited faeces, i.e. on the surface or buried. RESULTS: There was a significant effect of contamination date on development of all three species, with maximum numbers ofL3 developing between late spring (November) and early autumn (March), and minimum numbers in June and July. There were large differences between species, with H. contortus exhibiting a long period (April to October) where development was close to zero, whereas T. circumcincta developed to some extent all year round. Development of T. colubriformis was intermediate between the other two species. Burying faeces containing nematode eggs increased the number of L3 recovered compared with surface deposition (p≤0.001), although there were a small number of exceptions involving only T. colubriformis. The weight of faeces recovered at harvest, which was assumed to be an indication of earthworm activity, was correlated with the number of L3 recovered for all species (p<0.001). In a separate analysis, earthworms were assumed tohave been active if <5 g faeces remained at harvest. Where this occurred, the number of L3 of T. colubriformis and T.circumcincta recovered was reduced by 56% and 58%, respectively (p<0.001). CONCLUSIONS: A marked seasonal pattern of development was observed for all three species, with the most larvae developing in spring-early autumn and the least in winter. This seasonal pattern was most pronounced in H. contortus and least obvious in T. circumcincta. Burying faeces containing eggs generally resulted in more L3 being recovered, whilst the apparent activity of earthworms resulted in fewer larvae being recovered. 相似文献

AIM: To investigate the anthelmintic properties of New Zealand native flax (Phormium tenax) for cattle, using a faecal egg count reduction test (FECRT). METHODS: Twenty-six heifer calves with high (>300 epg) faecal nematode egg counts (FEC) were paired into two groups and fed either chopped flax leaves ad libitum, or barley straw at an equivalent dry matter (DM) rate, from Day 0–7; all were supplemented with 1.5 kg/head/day cereal feed supplement. On Days 8–10, all heifers were fed a common diet of 3.5 kg DM/ head/day barley straw and 1.5 kg/head/day cereal feed supplement. Heifers were weighed and faecal samples were collected on Days —6, 0, 7, and 10; samples were analysed in triplicate for FEC. The nutritive value of the flax and straw was analysed. RESULTS: Both the flax and straw had low dry matter digestibility (DMD) and protein content. Although the flax-fed claves grew more than the controls, this may have been related to gut fill. Total daily faecal egg output was similar for flax and straw groups on Day 7 (14.7 vs 15.0 x 106 eggs/day, respectively) and Day 10 (14.9 vs 15.1 x 106 eggs/day, respectively). There was no difference in the change in FEC with time between the calves fed flax or straw diets. CONCLUSION: Consumption of flax leaves did not reduce FEC in calves with a mixed nematode infection. 相似文献

The synthetic pyrethroid deltamethrin (DM) containing a trace of [(14)C]-DM was formulated with non-oxidised sterol and wax ester fractions (F1) of wool grease and as the commercial preparation 'Clout-S'. These were applied as a 'backline' strip to sheep immediately after shearing and the concentration of [(14)C]-DM at meridians adjacent to the application strip and at 1/4 and 3/4 of the dorsal-ventral distance was determined. The F1 formulation resulted in significantly greater lateral spread of DM with less remaining at the application site (66+/-8% of dose) 98 days after treatment compared to 'Clout-S' (94+/-3% dose). Autoradiographic examination of treated wool demonstrated that there was more DM in the lower half of the wool staple when formulated in F1 compared to 'Clout-S'. Greater mortality occurred when sheep biting lice Bovicola (Damalinia) ovis were exposed in vitro to wool containing DM from F1 compared to 'Clout-S' treated sheep. In field trials there was increased efficacy against synthetic pyrethroid resistant B. ovis with F1 formulation than with 'Clout-S'. The study has demonstrated that synthetic pyrethroid availability, and therefore efficacy, can be significantly increased when the insecticide is formulated in a 'carrier' with the physicochemical characteristics of wool grease. 相似文献

Increasing reports of failure of synthetic pyrethroids for the control of Bovicola ovis on sheep lead to a preliminary investigation into the insecticide resistance status of this pest during 1994. Eighteen strains of lice from North and South Island flocks experiencing failure of synthetic pyrethroid treatments to control lice were exposed to cypermethrin (high-cis) in a treated surface (contact) bioassay. Lice from a group of hermit sheep were chosen as a reference susceptible strain against which resistance of field strains was measured and resistance factors (RFs) calculated. Low to moderate resistance to cypermethrin (high-cis) was demonstrated, with RFs ranging from 1.0 to 12.4. 相似文献

Abstract AIM: To identify weather variables associated with the development of eggs of Teladorsagia (=Ostertagia) circumcincta, Trichostrongylus colubriformis and Haemonchus contortus to third-stage infective larvae (L3) under a range of climatic conditions on pasture in the North Island of New Zealand. METHODS: Sheep faeces containing known numbers of eggs of all three nematode species were deposited on, or buried in, pasture plots at three sites, viz coastal Manawatu, Upper Hutt Valley, and East Cape hill country. Development was measured by recovering L3 from faeces, herbage and soil 28–31 days after deposition, on 13–18 occasions between January 2005 until July 2006. Weather data were recorded at each site, and the association between weather variables and number of L3 recovered was analysed using subsets regression to select best-fitting models from several candidate variables, after adjustment for efficiency of recovery of L3. A multiple linear regression model was then developed for each species, to select weather variables that had both significant and substantive effects on the number of L3 recovered. RESULTS: For all species, mean daily temperature was the bestpredictor of the number of L3 recovered (p=0.001). For T.circumcincta the final model included mean daily temperature and soil temperature (R2=51%), and for T. colubriformis the model only included mean daily temperature (R2=55%). For development of H. contortus, mean daily temperature was the most significant variable, but moisture in the form of rainfall entropy, i.e. the temporal distribution of rainfall, overthe first 14 days was also significant in the final model (R2=34%). CONCLUSIONS: Temperature was the most important determinant of developmental success of free-living nematodes on pasture at the study sites, and probably also for other parts ofNew Zealand with similar climates. Moisture was not significant in the development of T. circumcincta or T.colubriformis, implying that under the generally moist temperate climate in New Zealand, moisture is seldom limiting for development of these species. Haemonchus contortusappeared to be sensitive to moisture availability and precipitation in the first 14 days after deposition of faeces. The results of this study will be further developed as part of species-specific climate-driven models of parasite development in New Zealand. 相似文献

Data were obtained from a population consisting of 160 Merino ewes, as well as 64 male and 75 female progeny of these ewes at the 2-tooth age. The age and sex groups were maintained in separate flocks. The population has been divergently selected from the same base since 1986, either for (high or H line) or against (low or L line) maternal multiple rearing ability. All animals were inspected for sheep lice (Bovicola ovis) during September/October 2002 after a wool growth period of at least 4 months. When expressed relative to mean L-line performance, the advantage in lambs weaned per ewe during the lambing season amounted to approximately 110% in the H line (1.01 vs 0.48; P < 0.01). The proportion of animals on which 1 or more lice were observed were markedly lower in 2-tooth replacement ewes than in replacement rams and mature breeding ewes (0.053 vs 0.625 and 0.531, respectively; P < 0.01). One or more lice were observed on a lower proportion of H-line ewes than on those of the L line (0.413 vs 0.571, respectively; P < 0.10), while a similar tendency was observed in 2-tooth ewe hoggets (0.033 vs 0.143, respectively; P = 0.16). When the mean number of lice on individuals on which 1 or more lice were observed was compared between lines, the average number of lice observed on H-line young rams was lower than in the L line (4.36 +/- 0.85 vs 9.71 +/- 1.84 lice, respectively; P < 0.01). A similar tendency was observed in mature breeding ewes (2.15 +/- 0.38 vs 3.22 +/- 0.42 lice, respectively; P < 0.20). The more highly reproductive H-line ewes were not more susceptible to infestation with B. ovis than ewes of the L line. In fact, available evidence suggest that H-line animals could be more tolerant of B. ovis than those of the L line. 相似文献

The application of polymer matrix ear tags impregnated with 8.5% w/w cypermethrin to 6 wethers following shearing reduced lice to non-detectable levels on 4 of them at 29 weeks after tagging. At the conclusion of the study at 45 weeks the mean count of lice on tagged wethers was 3 per sheep compared to 158 on untreated wethers. In a second experiment, 6 non-infected sheep were treated at shearing with cypermethrin tags, 6 with 25 g/l cypermethrin backline formulation, 6 with tags plus backline and 6 were left untreated. Each group was exposed to 6 sheep with moderate to heavy infestations of lice. Compared to controls, all treatments delayed infestation, but cypermethrin tags gave no longer protection than backline treatment. All sheep were infested by 17 weeks after commencement of the study. At 45 weeks mean counts of lice were 38, 27, 20 and 74 respectively for sheep treated with tags, backline formulation, backline plus tags and untreated. Possible reasons for the better effect observed from applying tags to infested sheep than to sheep which were not infected at application but which were subsequently exposed to infested sheep are discussed. 相似文献

Objective To compare the wettability and efficacy of diazinon dip wash made with and without the addition of zinc sulphate.
Design Field experiments using a shower and a plunge dip complemented by in-vitro wettability experiments.
Procedure A flock of infested sheep was divided into groups and treated in a shower dip with clear or cloudy dam water plus up to 1.5% zinc sulphate. Another infested line of sheep was treated using a plunge dip with nil or 1% zinc sulphate. In both experiments, wetting was assessed after dipping and louse counts were conducted for 9 months after treatment. Five in-vitro experiments compared the wettability of dip wash containing diazinon with up to 1.5% zinc sulphate added.
Results In the shower dipping experiment, live lice were found at 1 month after dipping in the cloudy water groups with 0.75%, 1.0% and 1.5% zinc sulphate and at 2 months in the 0.75% zinc sulphate group. No lice were found at subsequent inspections or at any time in the groups that were plunge dipped. Zinc sulphate decreased the amount of dip wash retained by wool staples in all in-vitro experiments (P < 0.05).
Conclusion Zinc sulphate should be considered as a risk factor that could cause failure to eradicate a lice infestation. The risk can be overcome by ensuring that all sheep are saturated at dipping and that the dip wash, and any holding tanks, are agitated throughout the dipping event. 相似文献

Gastrointestinal nematodes are becoming increasingly resistant to the commercial products used to control them. The cost of routine vermifuge applications on herds and the problem of residues in animal products and the environment have prompted research on the anthelmintic activity of plant extracts. This work examines the anthelmintic action of neem and the homeopathic product Fator Vermes in sheep kept in a pasture for 18 months. Forty sheep of the Morada Nova breed were divided into four treatments and the control, according to the EPG. During the experiment, each animal received 100 g/day of shredded corn and did not receive protein supplementation. In treatment 1 (control), the animals received only shredded corn. Treatment 2 received 1.6 g/(animal day) of the homeopathic product mixed with the shredded corn, and treatments 3, 4 and 5 received, respectively, 12.5, 25.0 and 37.5 g/(animal day) of dried Azadirachta indica leaves mixed with the shredded corn. The neem was administered for alternating 15-day periods and the homeopathic product daily for 18 months. There were 39 fortnightly fecal collections made to count the EPG, and fecal cultures were performed monthly. The following genera, in percentage, were identified: Haemonchus: 65.58+/-3.27, Trichostrongylus: 15.92+/-7.38 and Oesophagostomum: 18.50+/-6.22. The treatments evaluated were not effective in controlling gastrointestinal nematodes (P>0.05), whose mean log(10) counts (EPG +1) and standard errors for treatments 1-5 were respectively 3.55+/-0.28; 3.48+/-0.31; 3.90+/-0.29; 2.78+/-0.29 and 3.48+/-0.30. A significant effect (P<0.0001) was observed of the periods of the year when the 39 collections occurred. Because of the diet deficient in raw protein, the sheep had higher average EPG counts, for all the treatments, at the end of the dry season, and the opposite occurred in the middle of the rainy season. 相似文献

Abstract AIM: To investigate the cause of classical swine fever (CSF) virus-seropositive animals in a nucleus pig-breeding herd in New Zealand, where porcine circovirus-associated disease had been diagnosed. CASE HISTORY AND CLINICAL FINDINGS: An exotic disease investigation was undertaken to exclude CSF and porcine reproductive and respiratory syndrome (PRRS) on a nucleus pig-breeding herd comprising approximately 300 breeding sows, 1,000 weaners, and 650 grower pigs. The herd was experiencing poor reproductive performance in sows, and breeding records showed a declining farrowing rate attributable to a single manager. The growing pigs (10–15 weeks old) were experiencing respiratory disease and wasting, and the mortality rate by pen varied between 9 and 20%. Post-mortem changes in affected grower pigs were consistent with circovirus-associated diseases. DIAGNOSTIC TESTING: Serological screening using an IDEXX-ELISA gave negative results for PRRS virus antibodies, but two grower pigs and one sow tested positive for CSF virus antibodies. These three seropositive animals remained positive to CSF virus, using three commercial ELISA test kits, over 27 weeks. A newly developed virus neutralisation test (VNT), using a New Zealand isolate of border disease (BD) virus, demonstrated that the seropositive pig sera had higher antibody titres to BD virus than to bovine viral diarrhoea (BVD) virus and CSF virus. PCR performed on tonsil, kidney, ileum and spleen gave negative results for CSF virus, and histopathology on lymph nodes, intestine, lung, kidney, liver and brain showed no evidence of the disease. Virus isolation performed on a number of samples was negative. CLINICAL RELEVANCE: The seropositive samples for CSF virus found in this investigation were likely to be a cross reaction to a pestivirus other than CSF virus. The finding of a possible endemic pestivirus capable of being transmitted between sheep and pigs on this farm may explain findings from previous serological survey work in New Zealand, and supports experience elsewhere, where BD virus was found to be the predominant ruminant pestivirus infecting pigs. The results show that pestivirus cross reactivity can result in unexpectedly high titres, and that testing with a full set of (local) pestiviruses is necessary to reach the correct conclusion. The investigation has direct relevance where pig herds with a low seroprevalence are encountered during surveillance for CSF. 相似文献

A study was undertaken on weaned 4–5 month old farmed red deer to test the efficacy of moxidectin and abamectin anthelmintics, given by three different routes of administration, compared with an untreated control. Faecal samples were collected on days 0, 7 and 14 for a faecal egg count reduction test (FECRT), blood samples were collected on days 0, 0.5, 1, 2, 3, 5, 7, 10 and 14 for pharmacokinetics, and the deer were killed on days 14 or 15 for total nematode count. The control group averaged 1264 adult Ostertagia-type nematode parasite species and treatment efficacy was 77.4% for moxidectin injection, 26% for oral moxidectin and 27.6% for pour-on moxidectin, while the treatment efficacy was 72.4% for abamectin injection, 70.1% for oral abamectin (Hi-Mineral) and 34.1% for pour-on abamectin. Both moxidectin and abamectin injections were significantly more efficacious than their equivalent pour-ons. There was a significant difference in efficacy between oral abamectin (Hi-Mineral) and oral moxidectin (P < 0.01).The control group averaged 2956 adult lungworm (Dictyocaulus eckerti) and 50 Oesophagostomum venulosum in the large intestine and treatment efficacy against these nematodes was 100% for all treatments. There were negligible numbers of other gastro-intestinal nematodes. At slaughter, there was a significant correlation (P = 0.02) between FEC and Ostertagia-type nematodes in the untreated controls. Relatively few eggs were found in faeces from treated animals at 7 and 14 days post-treatment despite significant worm burdens in all six treatment groups, suggesting egg-laying suppression in resistant nematodes, and all three different FECRT calculations tended to overestimate the efficacy of the treatments compared with actual nematode counts.Peak plasma concentrations (Cmax) for both actives were measured 12 h after treatment for injection and oral and at 5 days for pour-on. Cmax (ng/ml) for moxidectin injection, oral and pour-on were 71.8, 8.3 and 0.4, respectively, and for abamectin injection, oral and pour-on were 62.1, 30.3 and 10.0, respectively. Area under the curve (AUC) estimates for moxidectin injection, oral and pour-on were 106.6, 12.9 and 6.1, respectively, and for abamectin injection, oral and pour-on were 162.7, 57.5 and 74.3, respectively. The results demonstrate that significant anthelmintic resistance to moxidectin and abamectin is present on this deer farm. However, the injection was the most effective route of administration in young deer for both anthelmintics, although <80% efficacious. We conclude that the FECRT is unreliable in deer when anthelmintic resistance is present. 相似文献

Having the ability to control litter size is important for sheep farmers and breeders worldwide. However, making genetic gain in key livestock traits like reproductive performance needs typically a lot of time, and both the fecundity and fertility traits have a great economic importance. Attention has therefore turned to better understanding the genes that control reproductive performance. Of these genes, research has focussed on the growth differentiation growth factor 9 (GDF9) gene (GDF9). In this study, a PCR-single strand conformation polymorphism (PCR-SSCP) approach was used to investigate variation in this gene in separate groups of purebred Finnish Landrace sheep, Finnish Landrace × Texel-cross sheep and composite sheep of undefined breed background, but based on New Zealand Romney-type genetics. Three GDF9 variants (named A, B and C) were found, and upon DNA sequencing, the nucleotide substitutions c.978A>G, c.994G>A and c.1111G>A were revealed. The frequency of variant A (containing nucleotides c.978A, c.994G and c.1111G) in the Finnish Landrace, Finnish Landrace × Texel-cross and composite sheep was 0.86, 0.78 and 0.76, respectively. In these three sheep groups, the frequency of B (defined by the presence of nucleotides c.978G and c.994A) was 0.01, 0.03 and 0.23 and for C (containing c.1111A) was 0.13, 0.18 and 0.01, respectively. An animal model was used to estimate the additive effect of fertility data for Finnish Landrace × Texel-cross sheep and revealed an association between litter size and the c.1111G>A variation (p = .036), but this was not observed for the Finnish Landrace sheep (p = .27) or the composite sheep (p = .17). When all the sheep were analysed together, the presence of c.1111A was associated (p < .05) with increased litter size, when compared to ewes that had c.1111G. Litter size did not differ between sheep with and without c.994A in all three groups of sheep investigated. This study suggests that c.1111A could be a useful genetic marker for improving fecundity in New Zealand sheep breeds and that it could be introgressed into other breeds, but analysis of more sheep will be required to confirm the associations that have been observed here. 相似文献

Two single nucleotide polymorphisms in the promoter of the ovine myostatin gene (MSTN) and their effect on growth and carcass muscle traits in New Zealand Romney sheep 下载免费PDF全文

Myostatin is a negative regulator of muscle growth and development in mammals, and variation in ovine myostatin gene (MSTN) has been demonstrated to be associated with variation in the muscularity of sheep. Polymerase chain reaction–single‐stranded conformational polymorphism (PCR–SSCP) was used to look for single nucleotide polymorphisms (SNPs) in a 304‐bp amplicon from the promoter region of ovine MSTN. Sequence analyses revealed two previously identified SNPs (c.?2449G/C and c. ?2379T/C) that resulted in three haplotypes (H1 (c.[?2449G; ?2379C]), H2 (c.[?2449C; ?2379C]) and H3 (c.[?2449G; ?2379T]). The effect of these SNPs on growth and carcass traits was investigated in 357 NZ Romney lambs. General linear mixed‐effect models revealed that sheep with the genotype c.?2449GC had a higher loin meat yield (p = 0.032) and proportion loin yield (p = 0.028), than those with the genotype c.?2449GG. The genotype c.?2379CC was associated with an increase in three weight traits: birthweight (p = 0.003), tailing weight (p = 0.009) and weaning weight (p = 0.028), when compared with the genotype c.?2379TC, but it was not found to have an association with growth rate. This suggests that c.?2379T/C has an effect that originates at, or before birth. Haplotype H3 was associated with a decrease in birthweight (p = 0.002), tailing weight (p = 0.003) and weaning weight (p = 0.011). Haplotype H2 was associated with increased loin yield (p = 0.012) and proportion loin yield (p = 0.002). The SNPs may have value as genetic markers for improved Romney breeding. 相似文献
