Young feral goats harbouring 3, 7 and 36 day old experimental infections of strains of gastrointestinal nematodes, known to be susceptible to anthelmintics in sheep, were dosed orally with commercially available anthelmintics at the manufacturers' recommended rates for sheep. The drugs and overall removals of Ostertagia circumcincta and Trichostrongylus spp. (T. colubriformis and T. vitrinus) respectively were: morantel citrate 69% and 68%; levamisole hydrochloride 55% and >99%; fenbendazole 98.5% and >99%; oxfendazole >99% and >99%. Results for Haemonchus contortus, while not inconsistent with high efficacy for all four drugs, must be interpreted with caution, in view of the erratic establishment of this worm in the controls.  相似文献

Observations on a beef cattle farm in Flanders led to the suspicion of resistance to levamisole in a strain of Ostertagia ostertagi. After treating a group of six animals with levamisole (5 mg kg-1 L.W., i.m.) the reduction in the number of trichostrongylid eggs per gram of faeces varied between 0 and 66.6%, whereas a similar group treated with fenbendazole (7.5 mg kg-1 L.W., p.o.) showed a reduction in worm burdens of 100%. Coproculture showed that the remaining eggs in the first treatment group were all Ostertagia sp. The suspected field strain was compared with a reference strain of O. ostertagi by means of the in vitro larval paralysis test. This test showed LC95 values of 9.12 micrograms ml-1 and of 99.04 micrograms ml-1 for the reference and the field strain respectively, which indicates a resistance factor for the latter of 10.9. These results were not unequivocally confirmed by the post mortem findings on a tracer calf necropsied 4 days after treatment with levamisole.  相似文献

Faecal egg count reduction tests were used to identify a strain of Ostertagia circumcincta/trifurcata complex in goats which was resistant to: ivermectin (at 0.2 mg/kg and 0.4 mg/kg orally), oxfendazole (at 5 mg/kg orally), levamisole (at 12 mg/kg orally) and fenbendazole (at 5 mg/kg orally) combined with levamisole (at 9.4 mg/kg orally). The percentage reductions achieved in these faecal egg count reduction tests were respectively 27%, 83%, 82%, 79% and 82%. Moxidectin (at 0.2 mg/kg by subcutaneous injection), fenbendazole (at 10 mg/kg orally) combined with levamisole (at 18.8 mg/kg orally), ivermectin (at 0.4 mg/kg orally) combined with oxfendazole (at 10 mg/kg orally) and ivermectin (at 0.4 mg/kg orally) combined with levamisole (at 12 mg/kg orally) were effective in removing these nematodes in goats as determined by faecal egg count reduction tests. These drenches achieved reductions of 100%, 100%, 98% and 100% respectively.  相似文献

A high level of host resistance to Trichostrongylus colubriformis was established in a group of Saanen goats infected weekly with 10,000 infective larvae for 10 weeks, treated with an anthelmintic and then challenged twice with 50,000 larvae, with a further anthelmintic treatment 28 days after the first challenge. A second group of goats exposed only to the two challenge infections developed worm burdens intermediate between, and significantly different from, the first group and a third control group given only the second challenge infection. Nematode fecundity (eggs per female) and male:female ratios were also significantly decreased in the first two groups. Faecal egg counts and worm counts were highly correlated and this relationship was dominated by the influence of the number of eggs/female nematode. The goats were approximately 9 months old at the start of the experiment.  相似文献

Field strains of Trichostrongylus colubriformis and Ostertagia circumcincta, designated PF4 and PF5 respectively, were recovered from a farm on which the sole use of levamisole over a preceding 12 year period led to the development of anthelmintic resistance. The results of field observations and preliminary critical trials in both Merino and crossbred sheep showed that both species have varying degrees of resistance to three major anthelmintics; levamisole, morantel tartrate and thiabendazole. Mean worm count reductions for adult T colubriformis (PF4) for therapeutic doses of morantel tartrate, thiabendazole and levamisole in crossbreds were 45.7 per cent, 97.3 per cent and zero respectively, and for Merinos 80.7 per cent, 88.3 per cent and 92.0 per cent respectively. Against O circumcincta the corresponding reductions for crossbreds were 51.4 percent, 95.4 per cent and 20.3 per cent and for Merinos, 52.5 per cent, 73.1 per cent and 29.8 per cent. There was no statistically significant difference in the responses of both parasite species to either levamisole or morantel. This result suggests that resistance to the two chemically unrelated drugs may be co-inherited.  相似文献

AIM: To assess the in vivo anthelmintic activity of condensed tannins (CT) in the forage species Dorycnium rectum and Medicago sativa, and in an extract from grape (Vitus vinifera) seeds (GSE), against two species of parasite, Teladorsagia (Ostertagia) circumcincta and Trichostrongylus colubriformis, at different stages of their life cycle, in sheep that were parasite-na?ve or previously exposed to nematodes. METHODS: In Trial 1, a factorial treatment structure was used to compare faecal nematode egg counts (FEC) and worm burdens in 40 weaned Romney lambs fed either the CT-containing forage D. rectum (12% dry matter; DM) or M. sativa (lucerne; 0.2% DM). Twenty na?ve and 20 previously-exposed lambs were drenched free of parasites then reinfected with known species and numbers of parasites, and housed in pens indoors on a diet of lucerne pellets and chaffed hay. Groups of lambs (n=5 lambs per group) were fed one of the forages over one of two time periods within the parasite's life cycle. Six to nine days after the last feeding of fresh forages, faecal samples were collected for FEC, and all lambs were slaughtered and worm counts conducted. In Trial 2, 12 Suffolk x Romney lambs were surgically implanted with an abomasal cannula and then housed indoors in metabolism crates. After infection with parasites, six lambs were infused continuously over a 14-day period with a commercially available CT GSE (96% DM, made up to 34 g/L in water); the remaining lambs were infused with water. During infusion, samples were collected for egg hatch and larval development assays. After infusion, samples were collected for FEC, and all lambs were slaughtered and worm counts conducted. RESULTS: In Trial 1, there was a significant (p<0.001) difference in burdens of O. circumcincta between na?ve lambs and those previously exposed to parasites, but no other differences were recorded. In Trial 2, lambs infused with GSE had significantly (p<0.05) fewer T. colubriformis at slaughter and significantly (p<0.001) fewer eggs hatched in the egg hatch assay (EHA) than for lambs infused with water. Overall, the differences attributable to GSE were small in magnitude, being an 11% drop in egg hatch, and an 18% drop in numbers of adult T. colubriformis after 14 days of continuous infusion. No other differences were recorded. CONCLUSION: The results indicate that the in vivo anthelmintic activity of these CT sources is, at best, modest and is unlikely to be of any practical value. Further, these data emphasise that in vitro activity is an unreliable indicator of in vivo efficacy for CT-containing forages and extracts.  相似文献

Sheep were allowed to graze pasture that had been seeded with benzimidazole-resistant Haemonchus contortus and Ostertagia circumcincta in order to acquire a burden of arrested larvae. Following housing, sheep were dosed orally with either oxfendazole at a dose rate of 4.7 mg/kg (to confirm the benzimidazole-resistant status of the species of nematode), levamisole at a dose rate of 7.5 mg/kg, or an oxfendazole/levamisole mixture at a dose rate of 4.6 mg/kg oxfendazole and 8.1 mg/kg levamisole. The efficacies of the treatments were assessed by estimation of the arrested larval burden in the abomasum of each sheep, either at 10 or 11 days (oxfendazole and oxfendazole/levamisole mixture), or 12 or 13 days (levamisole), after treatment. Compared to the untreated controls, the protection afforded by a single dose of either levamisole or the oxfendazole/levamisole mixture was >99% against the arrested stages of both Haemonchus contortus and Ostertagia circumcincta. Treatment with oxfendazole confirmed the benzimidazole-resistance status of the two species.  相似文献

The efficacy of a mixture of albendazole sulphoxide and levamisole, 3.6 and 8.25 mg/kg respectively, at single and double dose rates, was compared with the recommended dose rate of each anthelmintic alone. The comparison was conducted on groups of 6 to 14-week-old lambs on 22 farms, 16 of which had evidence of multiple resistance to benzimidazole and levamisole. A single dose of the mixture reduced mean egg counts by 95% on half the farms with multiple resistance and on all the remaining farms. Consequently, the mixture should be included in an assessment of effective anthelmintics on farms to determine its value for nematode control. A double dose rate of mixture was effective on all but 4 farms. Reductions caused by the mixture were due to the additive actions of the drugs on 18 of 22 farms; synergistic action was noted on only 3 farms. It was concluded that the mixture of albendazole sulphoxide and levamisole offered many farmers an effective anthelmintic for use in preventive control programs. Recommendations for such programs include annual rotation of effective anthelmintics as a means of delaying selection for drug resistance.  相似文献

The immediate as well as the persistent anthelmintic efficacies of topically applied eprinomectin were evaluated in goats against induced infections with Teladorsagia circumcincta (2800 L3) and Trichostrongylus colubriformis (6000 L3). Twenty-three culled dairy goats were allocated to the following groups: control animals (group 1), animals treated 21 days prior to nematode infection (group 2), animals treated 7 days prior to nematode infection (group 3) and animals treated 21 days after nematode infection (group 4). Eprinomectin was applied at twice the cattle dose rate (1.0 mg/kg BW). According to the groups, necropsies were undertaken 28 days after nematode infection (groups 1-3) or 14 days after the anthelmintic treatment (group 4). Worm counts were determined for abomasum and small intestine. The curative anthelmintic efficacy of eprinomectin at 1.0 mg/kg BW on existing worm burdens was 100% against T. circumcincta and T. colubriformis. Quite similar worm burdens reductions were observed when eprinomectin was administered 7 days before infection whereas they were only 52.4 and 17.8% for T. circumcincta and T. colubriformis, respectively, for an administration of the drug 21 days prior to the nematode infection.  相似文献

Parasitic nematodes of ruminants can be expected to experience temperatures in excess of 40 degrees C in faeces on pasture and, perhaps, in the host. L3 Ostertagia (Teladorsagia) circumcincta survived for at least 90 min at 45 degrees C in vitro in water, but the larvae were inactivated rapidly by only slightly higher temperatures. The glycolytic enzymes hexokinase and pyruvate kinase were inactivated in a similar temperature range, whereas malate dehydrogenase maintained its activity at temperatures in excess of 50 degrees C. These data imply that the loss of glycolytic activity might explain the loss of larval motility at temperatures between 45 degrees C and 50 degrees C.  相似文献

The use of tanniferous plants or tannins represents one alternative approach to the control of gastrointestinal parasites in ruminants but most data have been obtained in sheep. The current study was therefore performed in goats with two objectives: firstly, to investigate the effects of condensed tannins (CT) on adult populations of Trichostrongylus colubriformis and Teladorsagia circumcincta; secondly, to examine their effects on the establishment of infective larvae of these two species. In experiment 1, two groups of kids were infected with 6 000 L3 of T. colubriformis and 6 000 L3 of T. circumcincta. After 7 weeks, quebracho extracts were administered per os for 8 days to one group. A comparable group which did not receive tannins was included as the control. The kids were slaughtered on week 11. Parasitological and pathophysiological parameters were measured weekly. Worm counts were assessed and mast cells, globule leukocytes and eosinophils were counted in the abomasal and intestinal mucosae. Tannin administration was associated with a decrease in egg excretion, and a decrease in female fecundity, but with no changes in worm numbers. These changes were associated with an increased number of intestinal mast cells. In experiment 2, 24 goats were used according to a 2 x 2 factorial design, depending on infection and tannin administration. Two groups were either infected with 6 000 L3 of T. colubriformis or T. circumcincta. Within each group, the goats were either drenched or undrenched with tannin extracts. Pathophysiological parameters were measured weekly. Twelve days after the cessation of tannin administration, the goats were slaughtered. Worm counts and female worm fecundity were determined. Tannin consumption was associated with a significant reduction (P < 0.01) of Trichostrongylus populations and a close to significant reduction for Teladorsagia. No effect on fecundity was observed. Our results (1) confirm the consequences of condensed tannins on nematodes in goats as in sheep and (2) indicate divergent effects depending on the parasitic stage exposed to the condensed tannins.  相似文献

This paper describes the nematode control strategy adopted by a Scottish hill sheep farmer. It provides an example of the limited use of anthelmintic drugs, targeted towards control of the periparturient rise in faecal nematode egg output in ewes and exploitation of grazing management, resulting in limited exposure of naive sheep to infective larvae on pasture. Resistance to benzimidazole, imidazothiazole and macrocyclic lactone anthelmintics, and to a combination of a macrocyclic lactone and imidazothiazole anthelmintic drugs was diagnosed. The targeted use of a persistent anthelmintic drug to control a periparturient rise in faecal nematode egg output in ewes may be unnecessary and selects strongly for resistance when the reservoir of anthelmintic-susceptible nematodes in refugia is small at the time of treatment. However, the use of a persistent anthelmintic drug in a selective proportion of ewes can be important and probably does not select strongly for resistance when the reservoir of anthelmintic-susceptible nematodes in refugia is large at the time of treatment. The former circumstances might arise on many Scottish hill sheep farms, whereas the latter may occur on upland and lowground farms, depending on previous grazing management, anthelmintic use and winter weather conditions. These factors must be taken into account when preparing sustainable health plans for nematode parasite control in individual sheep flocks.  相似文献

Control of a benzimidazole-, levamisole- and ivermectin-resistant population of Teladorsagia circumcincta was attempted using moxidectin administered orally at the manufacturer's recommended dose rate of 200 microg/kg bodyweight. Ewes were dosed after lambing with the aim of controlling their periparturient rise in faecal egg output and lambs were dosed at six-week intervals throughout the summer. This regimen failed to suppress the establishment of significant numbers of infective helminth larvae on the pasture, resulting in unsatisfactory lamb production. Oral dosing with moxidectin was effective in removing adult female burdens of ivermectin-resistant T. circumcincta, but the effect of the drug did not persist against the resistant helminth population.  相似文献

AIM: To investigate the occurrence of resistance to macrocyclic lactone (ML) anthelmintics by Ostertagia circumcincta in lambs on a sheep and cattle property in the North Island of New Zealand. METHODS: Thirty lambs were randomly allocated to one of five equal-sized groups, consisting of an untreated control and four treatment groups. The treatments, which were administered at the manufacturer's recommended dose rates, included oral moxidectin, oral abamectin (both at 0.2 mg/kg), an albendazole-levamisole combination, and an albendazole-levamisole-ivermectin combination. Post mortem worm counts were undertaken 7 days after treatment to determine the efficacy of each anthelmintic. RESULTS: The albendazole-levamisole and albendazole-levamisole-ivermectin combinations both reduced O. circumcincta burdens to zero whereas for moxidectin and abamectin efficacies of only 72% and 29%, respectively, were recorded. CONCLUSIONS: These results clearly demonstrated the occurrence of resistance to MLs by O. circumcincta. Although this is not the first occasion where resistance to this anthelmintic family has been detected in this parasite in sheep in New Zealand, it is the first instance that resistance to either moxidectin or abamectin has been reported.  相似文献

RESULTS: The albendazole-levamisole and albendazole-levam-isole-ivermectin combinations both reduced O. circumcincta burdens to zero whereas for moxidectin and abamectin efficacies of only 72% and 29%, respectively, were recorded.

CONCLUSIONS: These results clearly demonstrated the occurrence of resistance to MLs by O. circumcincta.
