应用光镜和透射电镜观察,恒河猴肾上腺皮质可分为球状带、束状带和网状带。其特点是:(1)被膜较发达,有时可伸入球状带将其细胞分隔为团块。(2)球状带有时呈指状指入束状带和网状带,其内可见嗜铬细胞;球状带细胞可分为明、暗2种类型,他们排列成团球状、滤泡状、索状和不规则形。(3)网状带较发达,有的可分为内、外2个区;有时网状带呈刺状伸入束状带;有时网状带细胞可见于被膜下或髓质内。(4)有时部分被膜连同皮质一起呈指状伸入髓质内。(5)皮质细胞均含丰富的溶酶体、线粒体和内质网,但不同部位的细胞所含的脂滴在数量、大小、形态和电子密度等方面存在差异。  相似文献

以大鼠为试验对象,研究冷应激情况下肾上腺组织结构的变化。结果表明:应激后大鼠肾上腺皮质束状带和网状带宽度显著增高.且分界明显。应激后束状带和髓质细胞数目显著高于常温对照组。球状带细胞数目增多.出现分裂情况;束状带细胞有脱颗粒现象,使细胞呈空泡状;网状带细胞胞质内脂质颗粒少,细胞小;髓质细胞体积增大,胞核大且密集,胞浆染色深。  相似文献

There is little published information describing the differences in adrenal structure between strains of a single species despite quite well known functional differences. In this paper we report morphological differences in the adrenal glands between three strains of laboratory rat; Dark Agouti (DA), Sprague-Dawley (SD) and Wistar (W). Significant differences in adrenal weights (as a percentage of body weight) were not detected between any of the groups. While there were no significant differences in the volume of medullary or zonae glomerulosa or fasciculata tissue, the volume fraction for the zona reticularis was significantly smaller in DAs compared to the other strains. The functional significance of these differences is unknown. However, it is suggested that the reduced volume of zona reticularis tissue may contribute to a reduced capacity for glucocorticoid synthesis or storage.  相似文献

The adrenal glands of 56 dogs ranging in age from birth to 13.6 years were examined for a determination of normal zonal and cellular morphology patterns throughout development, maturity and aging. Routine light microscopic techniques as well as lipid and cholesterol staining were used. The zona arcuata is described as developing between the fibrous and cellular layers of the capsule forming the cortical blastema layer, a zona biproliferativa. It is proposed that both centrifugal and centripetal migration of cortical parenchyma can result. The formation of accessory nodules is described as compensatory hyperplasia. Histomorphometric analysis revealed growth in the zona fasciculata to be by hyperplasia and the zonal volume percentages to be arcuata, 27%; fasciculata, 50%; reticularis, 23%.  相似文献

A 6-month-old female beagle dog, assigned to the low-dose group in a toxicity study, was evaluated for compound toxicity, and spontaneous hyperadrenocorticism was suspected. The animal had an externally apparent distended abdomen on clinical examination upon arrival. Pre-dose clinical pathology showed slightly higher erythroid parameters and stress leukogram on hematology; plasma biochemistry showed higher total protein, gamma-glutamyl transferase, total cholesterol, and triglyceride levels than the reference data. On necropsy, a prominent increase in adipose tissues of the subcutis and abdomen and increased weight of the adrenal gland and liver were observed. Histopathology revealed diffuse hyperplasia of adrenocortical cells in the zona fasciculata and reticularis, cortical atrophy of the thymus, and abundant glycogen accumulation in the hepatocytes. These findings were incidental and not test-substance-related. Electron microscopy of the adrenocortical cells in the zona fasciculata revealed decreased typical translucent lipid droplets, increased electron-dense lipid droplets, and abundant smooth endoplasmic reticulum and lysosomes. Additionally, increased numbers of various sizes and forms of mitochondria with tubular, vesicular, or lamellar cristae compared to that of normal animals were observed. These ultrastructural characteristics of the adrenocortical cells suggested hyperfunction. The pre-dose plasma cortisol levels were slightly higher than those of other females assigned to the toxicity study, while plasma adrenocorticotropic hormone levels were within the normal range. These findings indicate that hyperadrenocorticism is a possible cause of the systemic changes in this case.  相似文献

Adrenal glands of adult and young male mink were examined for a period of twelve months from July, 1970 and the seasonal variations in the relative widths of the cortex and medulla were investigated using histometric procedures. The medulla to cortex ratio in adult mink was noted to be increased during the months immediately following the breeding season although no consistent change was noted in the total size of the adrenal gland. Histochemical analysis using periodic acid Schiff reagent (PAS) for carbohydrates, and Oil red-0-stain for lipids revealed focal accumulations of pigment cells in the zona reticularis during the post breeding season. The presence of strands of PAS positive cells in the outer zones of adrenal cortex just before the peak of the breeding season indicates that these cells may be in the process of migration and infiltration from the zona glomerulosa to the zona reticularis where they become encased by a thin layer of connective tissue. The aggregation of PAS and Oil red-0 positive cells in the zona reticularis, previously assumed to be a response to hormone treatment, may be a normal feature associated with the onset of testicular quiescence in sexually mature mink.  相似文献

The studies reported here were undertaken to better characterize the role of the adrenal cortex in the increased susceptibility to stress observed in porcine stress syndrome. Since adrenal hypofunction was suggested during early studies as a factor in events leading to porcine stress syndrome it was decided to produce such a state by the exogenous administration of the glucocorticoids prednisolone and fluoroprednisolone.

A total of 34 pigs (weighing 90 to 100 kg) received a series of graded doses (given intramuscularly) of one or the other of the foregoing compounds and were necropsied 48, 72, and 120 hours after the last treatment. The major adrenocortical changes produced by these treatments were atrophy, an accentuation of degenerative changes which could be observed in milder form in many of the 30 control pigs examined, a marked increase in stainable lipid 120 hours after the last fluoroprednisolone treatment and, ultrastructurally, an increase in lysosome-like bodies and cellular debris (“myelin bodies”).

Adrenocortical atrophy is not a feature of porcine stress syndrome nor were the localized depositions of stainable lipid within the zona reticularis, described in porcine stress syndrome, found in the glucocorticoid treated pigs. Consequently, the findings reported here bear a somewhat limited resemblance to those observed in pigs affected by porcine stress syndrome and cast some doubt on whether adrenal hypofunction plays a role in the pathogenesis of the latter condition.

ACTH is the primary regulator of adrenal function during acute stress. However, during chronic inflammatory stress additional factors play a major role in the regulation of adrenal secretion. Many cytokines circulate in the blood and are synthesized and released from adrenal tissue. Furthermore, these peptides modify adrenal function. Recently, interleukin-4 (IL-4) was demonstrated to be released from a human adrenal tumor cell line. Therefore, we hypothesized that normal bovine adrenocortical cells could express IL-4 and that this cytokine may modify adrenal function. We determined that IL-4 and IL-4 receptors (IL-4R) are expressed in the bovine adrenal cortex whereas the expression of IL-4 and IL-4R in the adrenal medulla was not apparent. Exposure of dispersed bovine adrenocortical cells isolated from the zona fasciculate to IL-4 did not modify basal release of cortisol. However, the ACTH-stimulated release of cortisol from the bovine adrenal cells was augmented by IL-4. IL-4 exposure had no affect on adrenal androgen release from bovine zona reticularis cells, but IL-4 inhibited the ACTH-stimulated release of adrenal androgens from these cells. The effects of IL-4 on ACTH-stimulated cortisol and adrenal androgen release were dependent upon the IL-4 incubation interval and the IL-4 concentration. Because communication between the immune and endocrine systems is important in inflammatory conditions, IL-4 may play a role in coordinating the adrenal response to inflammatory stress.  相似文献

The lesions in the adrenal glands of seven dogs with hyperadrenocorticism that had been treated with trilostane were studied histologically. The glands of the six dogs with pituitary-dependent hyperadrenocorticism had moderate to severe cortical hyperplasia that was either diffuse or nodular. The lesions were more pronounced in the zona fasciculata than in the zona reticularis, and the zona glomerulosa was normal. In the dog with a functional adrenal tumour the non-tumour bearing adrenal gland showed mild nodular hyperplasia. Five of the seven dogs had variable degrees of adrenal necrosis, which was severe in two of them. The terminal deoxynucleotidyl transferase-mediated DUTP nick-end labelling (TUNEL) reaction specified areas of cell death as apoptosis in three of the dogs, and was positive in one of the dogs without visible areas of cell death. There were variable degrees of cortical haemorrhage in three of the dogs. In some of the dogs the lesions were severe enough to lead to hypoadrenocorticism.  相似文献

Cell cultures of embryonic calf kidney which had been infected with bovid herpes virus 2 were examined for cytological and histochemical changes. The morphological changes recorded from cells damaged by virus infection included the formation of gigant syncytial cells and intranuclear inclusions of Cowdry Type A. The cytological changes in the infected cells were accompanied by variation in enzyme activity. Recordable were rise in lactate dehydrogenase and alkaline phosphatase as well as decline in succinate dehydrogenase and acid phosphate activity. These phenomena were found to have resulted from impediment of cell metabolism by virus action.  相似文献

Body weights of finishing pigs can be variable within a finishing barn near the time of slaughter; therefore, it is common to market pigs over a period of time. This allows lighter pigs more time to gain BW and approach a desired end point. Use of immunological castration late in life to control boar taint, as an alternative to physical castration early in life, increases cutting yields of finishing male pigs compared with physical castrates. Because of common marketing strategies, it is important for advantages in cutting yields to span a broad spectrum of slaughter ages and BW. The primary objectives in this study were to evaluate carcass cutting yields, pork quality, belly quality, and bacon processing characteristics of immunologically castrated (IC) male pigs fed a moderate level of distillers dried grains with solubles and slaughtered at either 4 wk (early slaughter group) or 6 wk (late slaughter group) post-second injection. A total of 156 male pigs (physical castrates or IC males) were selected from a population of 1,200 finishing pigs. Data were analyzed with the MIXED procedure of SAS as a split-split plot design. Body weights of IC males were 3.60 kg heavier (P = 0.03) than physical castrates when slaughtered at 4 wk post-second injection and 7.52 kg heavier (P < 0.0001) than physical castrates when slaughtered at 6 wk post-second injection. Because of a lack of interaction (P > 0.05) between sex and time of slaughter post-second injection, some response variables were pooled. Hot carcass weights were not different (P = 0.57) between physical castrates (91.98 kg) and IC males (92.52 kg). There was a 2.77 percentage unit decrease (P < 0.001) in dressing percentage of IC males (71.78%) compared with physical castrates (74.55%). Lean cutting yields of IC males were 2.62 percentage units greater (P < 0.0001) than physical castrates and carcass cutting yields were 2.27 percentage units greater (P < 0.0001) for IC males when compared with physical castrates. There were no differences between IC males and physical castrates for shear force (P = 0.09), ultimate pH (P = 0.57), objective color (P ≥ 0.31), subjective color score (P = 0.64), or drip loss (P = 0.30). Bellies from IC males were thinner (P = 0.01) and had narrower belly flops (P < 0.0001) than bellies from physical castrates. There were no differences (P = 0.74) in cured belly cooked yield between IC males and physical castrates. Overall, immunological castration improved cutting yields, did not affect pork quality, made fresh bellies thinner, and did not affect cured belly characteristics when pigs were fed a moderate level of distillers dried grains with solubles during the finishing phase of production.  相似文献

The structure of the adrenal gland was studied in 11 bottlenose dolphins ( Tursiops truncatus ), and five striped dolphins ( Stenella coeruleoalba ). These species are legally protected in Croatia. All examined animals died of natural causes and were found stranded along eastern Adriatic coast. In both species the adrenal gland consists of a cortex and a medulla; the cortex is divided into three zones. Whereas in the bottlenose dolphin, there is a zona arcuata which contains columnar cells arranged in the form of arches; in the striped dolphin this zone is replaced by zona glomerulosa containing rounded clusters of polygonal cells. In both species, the zona fasciculata consists of radially oriented cords of polygonal cells, whereas in zona reticularis cells are arranged in branching and anastomosing cords. The adrenal medulla in both species contains dark, epinephrine-secreting cells and light norepinephrine-secreting cells. Epinephrine-secreting cells are localized in the outer part of the medulla, whereas norepinephrine-secreting cells are found in the inner part, arranged in clusters and surrounded by septa of thin connective tissue. The gland is surrounded by a thick connective-tissue capsule, from where thick trabeculae extend towards the interior. In the bottlenose dolphin, group of cells resembling both medullar and cortical cells can be seen within the capsule; whereas only groups of cells resembling cortical cells are found within the capsule of the striped dolphin. In the bottlenose dolphin invagination of the adrenal cortex into the medulla is obvious as well as medullary protrusions extending through cortex to the connective tissue capsule.  相似文献

Seventy-two crossbred (Large White X Landrace) pigs were used in a 3 X 7 factorial experiment to investigate the response of two strains of boars (strains A and B) and of castrated male pigs (strain B) to seven levels of intake of a single diet (ranging from 5.3 Mcal digestible energy [DE]/d to ad libitum) between 45 and 90 kg live weight. All aspects of growth performance and body composition were affected to different degrees by both strain and sex. At all levels of energy intake strain A boars grew faster, had a lower feed to gain ratio and contained less fat and more water in the empty body than strain B boars, which in turn exhibited faster live weight gain and more efficient and leaner growth than castrated males. The magnitude of the differences in growth performance between strain A and strain B boars and castrates increased with increased energy intake above 7.88 Mcal DE/d, and these differences were associated with concomitant strain differences in their respective capacity for protein growth and in the relationship between energy intake and protein deposition. For strain A boars the rate of protein deposition increased linearly from 92 to 188 g/d with increased energy intake from 5.3 Mcal DE/d to ad libitum. For strain B boars and castrates the rate of protein deposition increased linearly with increased energy intake up to 7.88 Mcal DE/d, but thereafter it remained constant at 128 and 85 g/d, respectively. For castrates protein deposition was depressed (P less than .01) when the diet was offered ad libitum. Strain A boars had a higher energy requirement for maintenance (3.55 Mcal DE/d) than strain B boars (2.77 Mcal DE/d) or castrates (2.60 Mcal DE/d). Strain A boars also contained less protein and more water in the fat free empty body than the other two pig types.  相似文献

With 14 figures and 3 tables SUMMARY: Each adrenal gland consisted of cortex and medulla that developed from different embryological origins and presented different cellular organization. One hundred male or female camel embryos or fetuses with crown vertebral rump lengths (CVRL) that ranged from 0.8 to 117 cm were examined. The adrenal cortex, which is derived from intermediate mesoderm, was first observed in the 0.8-cm CVRL camel embryo. The adrenal cortex initially was combined with the gonad as a thickened region of proliferating cells derived from splanchnic intermediate mesoderm. Adrenocortical tissue was first separated from the gonadal tissue in the 2-cm CVRL camel fetus and was observed as a separate dorso-medial mass of cells. At 2.5-cm CVRL, the adrenocortical tissue was surrounded by a capsule of undifferentiated mesenchymal cells, except at its proximal pole, where an invagination was located through which chromaffinoblast cells entered the cortex. The chromaffinoblast cells migrated from the neural crest to form the medulla of the developing adrenal gland. In the 3.5-cm CVRL camel fetus, the adrenocortical cells differentiated into two layers: the inner fetal cortex and the outer definitive cortex. As development proceeded, the fetal cortex degenerated and the definitive cortex formed the zona glomerulosa and zona fasciculata. The zona reticularis did not form until the end of gestation. During prenatal life, the adrenal medulla was much thicker than the cortex.  相似文献

The effects of sex, genotype, and adipose depot on lipogenic enzyme activity have been investigated in Holstein and Pirenaican bulls and heifers, taking into account differences in adipocyte size. Fifteen Pirenaican bulls and 15 heifers and 15 Holstein bulls and 13 heifers were fattened until slaughter (12 to 13 mo old and 450 to 500 kg of body weight). During the fattening period, animals had ad libitum access to commercial concentrates and straw. The 10th rib was dissected to determine the fat content. Adipocyte size and activities of the following lipogenic enzymes were determined: glycerol 3-phosphate dehydrogenase, fatty acid synthase, nicotinamide adenine dinucleotide phosphate (NADP)-malate dehydrogenase, glucose 6-phosphate dehydrogenase, and NADP-isocitrate dehydrogenase, in the omental, perirenal, subcutaneous, and intermuscular adipose depots, respectively. Because adipocyte mean cell volume varied with sex, breed, and depot, regression analyses of log(e) activity per cell and log(e) cell volume were used to compare activities per unit volume. Sex, breed and depot had no effect (P > 0.05) on the gradients of regressions, which did not differ significantly from 1. Thus, activity per unit volume did not vary with cell size. Consequently, sex, breed, and depot effects on the regression analyses were equivalent to effects on activity per unit volume. Females had greater amounts of fat in the 10th rib (P < 0.001), larger adipocytes (P < 0.001) and, in general, greater (P < 0.05) lipogenic activity per cell, even when adjusted for cell size, than males. These findings suggest that differences in adiposity between sexes are mainly due to females having a greater capacity for lipid synthesis, and hence, hypertrophy, than males. When adjusted for differences in carcass weight, Holsteins had larger adipocytes than Pirenaicans. The abdominal depots, omental and perirenal, had a greater adipocyte size (P < 0.001) and, in general, greater lipogenic enzyme activities per cell (P < 0.05) than the subcutaneous and intermuscular carcass depots. However, when activity per cell was adjusted for cell size, subcutaneous depots had greater fatty acid synthae, glucose 6-phosphate dehydrogenase, and NADP-malate dehydrogenase activities than omental and perirenal, indicating that other factors such as nutrient supply may restrict hypertrophy of carcass adipocytes.  相似文献

Lagomorpha are often used as animal models in reproductive experiments. The aim of the present study was to examine the glycoconjugate modifications occurring mainly in the zona pellucida during oocyte growth in the rabbit and hare, using a battery of lectins combined with sialidase digestion and chemical treatments. This histochemical approach made it possible to identify sulpho- and asulpho-carbohydrates in the terminal and/or subterminal position linked to sialic acid residues. The lectins that stained the zona pellucida of both species most effectively were SBA, PNA and WGA, indicating the presence of beta-D-N-acetylgalactosamine, beta-D-galactosamine and N-acetylglucosamine residues. The differences in the glucidic residue content and in their spatial distribution that depended on the species and stage of follicle development were also detected.  相似文献

Histochemical activities of several enzymes were investigated in the olfactory epithelium (OE) and vomeronasal organ (VNO) of the golden hamster. Activities of adenosine triphosphatase, lactate dehydrogenase and succinate dehydrogenase were intense in the OE, and the sensory (VSE) and respiratory epithelium (VRE) of the VNO. The activity of acid phosphatase was intense in both the OE and the VSE, while that of non-specific esterase was intense in the VSE alone. The activity of alkaline phosphatase was detectable only in the VRE. Activities of monoamine oxidase and acetylcholine esterase were negative in all of the OE, VSE and VRE. These similarities and differences in the histochemical distribution of enzymes between OE and VSE may reflect the common olfactory function and/or functional specialization in these epithelia. On the other hand, the VRE was considerably different from the OE and VSE in the enzymatic distribution. This may reflect the non-olfactory function of this epithelium.  相似文献

Fibre characteristics and enzyme activities were determined for the gluteus, semitendinosus, vastus lateralis and triceps brachii muscles of 55 Standardbred trotters of different ages. Four fibre types (I, IIA, IIB, IIC) were demonstrated by histochemical staining of myofibrillar adenosine triphosphatase after preincubation at different pH values. Type II fibres predominated in all the muscles and the type IIA/IIB ratio was higher in horses over 5 years than in younger horses, except in the vastus in which the IIA/IIB ratio did not change with age. The vastus had the highest proportion of type IIA fibres and the semitendinosus the highest proportion of type IIB fibres. Histochemical demonstration of NADH dehydrogenase disclosed that almost 100 per cent of the type IIA and many of the type I and IIB fibres were medium-stained; the remaining type I fibres were darkly stained and the type IIB fibres lightly stained. In older horses more fibres were stained for NADH dehydrogenase. The activity of triosephosphate dehydrogenase decreased that that of 3-hydroxy-acyl-coA dehydrogenase and citrate synthase increased in all the muscles except the vastus with increasing age. The greatest increase in oxidative capacity occurred in the gluteus and triceps. Training, rather than age, was regarded as the factor inducing these changes. The results emphasise that histochemical data are only semiquantitative, and there are apparent discrepancies in the intensities of histochemical staining and the biochemical evaluation of various enzymes.  相似文献

1. M. iliotibialis (MIT) and M. pectoralis (MP) of the BUT Big 6 and Kelly BBB turkey were characterised with respect to physical properties, mitochondrial function, metabolic state, morphology and meat quality. 2. Mitochondrial enzyme activity and respiration rates in MP declined with increasing age while glycolytic enzyme activity remained nearly constant. 3. There were no major differences between BUT Big 6 and Kelly BBB with respect to histological, histochemical or biochemical variables. In spite of the greater adult weight of BUT Big 6, body proportion was equal in both strains. 4. In agreement with the histochemical findings MIT showed higher oxidative capacities, while glycolytic enzyme activity was higher in MP. 5. Pyruvate was the best substrate for oxidative phosphorylation in MIT, but not in MP. Pyruvate dehydrogenase (PDH) activity was up to 15 times less in MP and blood lactate was correlated with intramuscular pH. 6. Turkey breast muscle was restricted in its ability to oxidise pyruvate, especially in those animals that tended to develop intramuscular acidosis post mortem. 7. It is concluded that the in vivo metabolic environment results in acidosis and impaired meat quality, at least in turkey M. pectoralis.  相似文献

The objective of this experiment was to determine if increasing lysine in the diets of immunologically castrated (IC) male pigs would affect further processed product characteristics when compared with physical castrates or entire males. Raw materials for this experiment were derived from a previous experiment evaluating carcass characteristics. Physical castrates, IC males, and entire males were assigned to 1 of 4 diet programs with increasing lysine in a step-down lysine inclusion program that culminated with the following concentrations in the late finishing diet: physical castrate with low lysine (0.7%), IC with low lysine (0.7%), IC with low/medium lysine (0.8%), IC with medium/high lysine (0.9%), IC with high lysine (1.0%), and entire with high lysine (1.0%). Bellies were injected with a cure solution to a target of 110% of original green weight, and weighed again to determine brine uptake. Hams were injected with same cure solution to a target of 130% of green weight. Cure solution was formulated for a finished product inclusion of 1.5% salt, 0.34% phosphate, 0.05% sodium erythorbate, 0.11% sugar, and 0.014% sodium nitrate. Physical castrates had thicker (3.77 cm) bellies (P<0.05) than all treatment groups, except IC males fed low/medium lysine (3.73 cm). Entire males (2.85 cm) had the thinnest (P<0.05) bellies of all treatment groups. There were no differences (P>0.05) in percentage brine uptake for cured bellies among IC males regardless of dietary lysine (range 9.93 to 10.67%). Cooked yield of cured bellies was not different (P>0.05) among physical castrates or IC males regardless of lysine inclusion. Cooked yield of cured bellies from entire males (95.12%) was less (P<0.05) than cooked yield for any other treatment group. Pumped weight differences of cured hams among treatment groups were similar to green weight differences, and there were no differences (P>0.05) among any treatment groups for pump uptake percentage. There were also no differences in cook loss percentages among any treatment group. Therefore, differences in cooked yield are a reflection of initial green weight. There were no differences (P>0.05) for protein fat-free values among any treatment groups. Therefore, it can be concluded, in this population of pigs, there were no differences in further processed product characteristics among physical castrates and IC males.  相似文献
