The ovaries of 59 pluriparous cows of unknown reproductive history were palpated, scanned and dissected on the day of slaughter to compare the accuracy of rectal palpation and transvaginal ultrasonography with a 5 MHz linear array for the detection of corpora lutea and follicles. The rectal palpation was first carried out to judge the presence of follicles of more than 5 mm diameter, and corpora lutea which were classified as young (days 1 to 4), mid-cycle (days 5 to 16) or old (days 17 to 21) according to morphological criteria. The cows were then examined for follicles and corpora lutea by ultrasonography and the corpora lutea were again classified directly as young, mid-cycle or old according to their appearance. The cows were then slaughtered, their ovaries dissected, and the follicles over 5 mm in diameter were counted and the corpora lutea were classified in the above mentioned age categories. For the detection of a mid-cycle corpus luteum the sensitivity and predictive value of rectal palpation were, respectively, 83.3 per cent and 73.2 per cent and for ultrasonography the sensitivity and predictive value were 80.6 per cent and 85.3 per cent, respectively. However, both techniques were inaccurate for the detection of young and old corpora lutea. For detecting follicles ultrasonography was a significantly better method than rectal palpation. Ultrasonography detected 95 per cent of follicles larger than 10 mm whereas rectal palpation detected only 71 per cent of these follicles. Both techniques failed with follicles 5 to 10 mm in diameter; only 21.5 per cent were detected by rectal palpation and 34.3 per cent by ultrasonography.  相似文献

In this study diagnostic certainty of ultrasonography and rectal palpation concerning the detection of follicles and C.I. was compared by evaluation of the findings obtained with ultrasonography in waterbath and dissection of the ovaries after slaughter. Clinical examinations were performed on a total of 30 cows (transrectally and ultrasonographically, 5.0 mhz, linear) in slaughterhouse. In the laboratory ovaries were evaluated after slaughter both macroscopically and by ultrasonography in waterbath. Diagnostic reliabilities of these methods were compared. No difference between the methods was determined concerning the longitudinal measurements of corpora lutea (19.96 +/- 4.83 mm, 20.41 +/- 5.41 mm, 21.45 +/- 5.26 mm by ultrasonography, waterbath and macroscopy respectively). By means of determining the correct identification of corpora lutea, the error rate was 24.1% and 17.2% for rectal palpation and ultrasonography respectively. The comparison of rectal palpation and macroscopy showed that three small corpora lutea and two corpora lutea with small cavity were determined wrongly as small follicles and two corpora lutea were determined whereas they were not present actually. With ultrasonography four small C.I. could not be detected and one C.I. with cavity was wrongly determined as follicle. It was noticed that follicles bigger than 10 mm (F2 = 10-15 mm, F3 = 16-20 mm) could be determined more accurately by means of ultrasonography than by rectal palpation (with ultrasonography: F2 = 90.48%, F3 = 100.0%; with rectal palpation, F2 = 61.9%, F3 = 200.0%). The correlation of the findings of rectal palpation or ultrasonography and blood progesterone levels was 86.2% and 89.7% respectively. Using rectal palpation and laparoscopy, the relationship of ovarian contents to plasma progesterone concentration during the oestrous cycle, early pregnancy and post partum periods in the swamp buffalo (Bubalus bubalis) was studied. During the oestrous cycle, four stages in the lifespan of the corpus luteum were seen laparoscopically. The mean (+/- sd) concentrations of progesterone in plasma in cows with and without a corpus luteum on their ovaries were 1.49 +/- 0.78 ng/ml (n = 31) and 0.14 +/- 0.09 ng/ml (n = 14), respectively. Plasma progesterone levels reflected age-dependent changes occurring in the cyclic corpus luteum. The accuracy of diagnosing ovarian contents was 82 and 91 per cent for rectal palpation and plasma progesterone levels respectively. Approximately 29 per cent follicles (larger than 10 mm) were incorrectly diagnosed as corpora lutea by rectal palpation.  相似文献

Fifteen Holstein cows were treated with two doses of 25 mg of a prostaglandin F2α (PGF2α as dinoprost tromethamine) administered intramuscularly 11 days apart. The cows were then divided into three groups and inseminated either at 72, 80 or 72 and 96 hours after the second dose of PGF2α. Thirteen cows ovulated after the second prostaglandin treatment. Eight cows were diagnosed pregnant by rectal palpation 42 days after insemination but only five calved. PGF2α induced luteolysis in cows with active corpora lutea as evidenced by the dramatic decreases in the plasma progesterone concentrations after treatment. In contrast, following PGF2α administration to cows in follicular or late luteal phase the concentrations of plasma progesterone either increased gradually or remained low for several days before increasing to maximal levels. The ovulatory rate after the two doses of PGF2α11 days apart was high. However, the pregnancy rate after this treatment was influenced by other factors including abnormal ovarian function.  相似文献

The reliability of clinical ovarian findings was assessed as an indicator of luteal function in primiparous dairy cows. The postpartum period of 103 cows following their first parturition was studied by thrice weekly rectal palpation of ovaries and whole milk progesterone assay from 1 week after parturition to the first insemination. The relationship between milk progesterone levels and 1101 ovarian findings was compared during the follicular phases, short luteal phases and during the early, mid and late thirds of normal luteal phases. The compatibility between elevated progesterone and palpable corpus luteum was 71%, and between low progesterone and lack of corpus luteum 77%. In 10% of all rectal examinations the finding was unspecified; i.e. the clinician could not differentiate between luteal and follicular activity. During the acyclic period prior to the initiation of luteal function, the proportion of false corpus luteum findings was 11%. The corpora lutea of the short oestrous cycles were more difficult to palpate than those of normal cycles. During early dioestrus the corpus luteum was significantly more difficult to palpate than during the rest of dioestrus. The percentage of unspecified findings was highest during early dioestrus.The paper discusses the reliability of rectal examination as a method of diagnosing cyclicity and of evaluating the responsiveness of a cow to prostaglandin treatment.  相似文献

The objective of this study was to determine whether induced luteolysis of one of the two corpora lutea in twin pregnancies would provoke spontaneous twin reduction. In Experiment 1, 12 post‐partum cows with two corpora lutea in the same ovary were assigned to (three cows per group): Group I, Group II, Group III or Group IV receiving into one of the corpora lutea puncture with no treatment, 0.5 mg dinoprost, 1.5 mg dinoprost and 2.5 mg dinoprost, respectively. One of the two corpora lutea showed clear signs of luteolysis on Day 2 and was practically non‐detectable on Day 7 after treatment in the three cows of the Group IV. In Experiment 2, 11 cows carrying live twins with two corpora lutea on Day 28 of gestation, eight bilateral and three unilateral, received 2.5 mg dinoprost into one of the corpora lutea. Corpus luteum reduction and embryo reduction after treatment were registered in 10 and 9 cows, respectively. In bilateral twin pregnancies, four cows suffering embryo reduction remained pregnant. In unilateral twin pregnancies, membrane detachment resulted in the death of both cotwins. In conclusion, although observations were based on few animals, there seems to be a mechanism that operates locally to transfer ovarian progesterone to the uterus, and also a quantitative relationship between the amount of progesterone secreted and support of conceptuses, resulting in death of one twin embryonic vesicle when one corpus luteum regresses.  相似文献

The objective of this study was to determine if corpora lutea anticipated to have short lifespans were more responsive to the luteolytic action of prostaglandin F2 alpha (PGF2 alpha) than corpora lutea anticipated to have normal lifespans. Sixteen cows were allotted randomly to a hysterectomized-control (HC) or hysterectomized-progestogen (norgestomet) implant (HN) group. To verify that progestogen treatment of postpartum cows prior to induction of ovulation with gonadotropin-releasing hormone (GnRH) results in an increased number of cows exhibiting normal-length luteal phases, 21 additional cows were allotted randomly to a uterine intact-control (IC) or a uterine intact-progestogen implant (IN) group. Cows allotted to the HN and IN groups received norgestomet ear implants for 9 d beginning 17 to 21 d postcalving. All cows were injected (i.m.) with 100 micrograms GnRH 28 to 32 d postcalving (48 h after implant removal in the HN and IN groups) to induce ovulation. Two or 3 d after GnRH injection (d 0), cows in the HC (n = 8) and HN (n = 8) groups were hysterectomized to remove the major endogenous source of PGF2 alpha, and on d 7 cows were injected (i.m.) with 10 mg PGF2 alpha to assess luteal sensitivity. The proportion of corpora lutea having normal lifespans was greater (P less than .1) for the IN than for the IC group. In HC and HN groups, concentration of progesterone (P) increased similarly from d 0 to 6. Injection of PGF2 alpha in HC and HN groups on d 7 decreased (P less than .01) concentration of P approximately 50% by 6 h after injection (similar for both groups). Complete luteolysis was induced by PGF2 alpha in none of eight and two of eight cows in the HC and HN groups, respectively. In remaining cows (HC and HN groups) concentration of P increased (P less than .01; similar for HC and HN groups) beginning 24 h after PGF2 alpha and remained elevated through d 30 to 34 (end of experimental-period). In summary, corpora lutea anticipated to be short-lived were not more responsive to PGF2 alpha than corpora lutea anticipated to have normal lifespans.  相似文献

Ovarian function was assessed from milk progesterone profiles in 236 cows that were presented for clinical examination after parturition and in 227 unexamined herdmates. By 30 days after calving, 81 per cent of the cows had ovulated and by 42 days 92 per cent had resumed cycling. On average milk samples were taken for 155 days, and during this period 67 luteal cysts or cystic corpora lutea and 24 follicular cysts were recorded, and 88 cows were anoestrus for more than 30 days. Luteal cysts, follicular cysts and prolonged anoestrus all occurred twice as frequently in the cows which were clinically examined as in those which were not examined. Progesterone profiles indicated that at the time of clinical examination 72.4 per cent of the cows examined were cycling normally, 11.3 per cent were anoestrus, 6.7 per cent had follicular cysts, 8.0 per cent had luteal cysts and 1.6 per cent were pregnant. Rectal palpation correctly identified ovarian function in only 63.5 per cent of examinations, with 29.8 per cent of cows that were cycling normally, 58.1 per cent that were anoestrus, 70.3 per cent with follicular cysts and 25 per cent with luteal cysts being incorrectly diagnosed. Progesterone profiles confirmed normal oestrous cyclicity in 81.9 per cent of these cows diagnosed as cycling normally by the clinician. The clinical treatments administered included prostaglandin, gonadotrophin-releasing hormone (GnRH), progesterone (PRID) and the physical expression of ovarian structures. Cows diagnosed as cycling normally were left untreated. We examined the response to exogenous prostaglandin F2α in cattle with or without palpable structures believed to be ovarian follicles. All animals had ovarian structures diagnosed by palpation as corpora lutea. The cows were placed into two groups: those with follicles which were estimated by the palpators to be ≤13 mm diameter (n=60); and cows with no palpable follicles or with follicles <13 mm diameter (n=133). Comparisons of proportion in estrus within five days, days to estrus, and milk progesterone levels failed to show significant differences between the groups.  相似文献

A total of 137 cycling zebu cows, each receiving a single dose of prostaglandin PGF(2alpha)were used in an oestrus synchronization programme on three different farms. Of the cows on the three farms, 60.6 and 90.5% showed overt oestrus and luteolysis, respectively. Pregnancy rate to fixed time inseminations following single injection of PGF(2alpha)was 61.4% for farm 1, significantly higher than the values of 45.7 and 46.9% for farms 2 and 3, respectively. The pregnancy rates to second service of rebred cows were 53.3, 50.0 and 50.0% for the three farms, respectively, with no significant differences between each.Fertility classification of the cows based on progesterone (P(4)) concentration showed that 6.6% of cows on the three farms were incorrectly diagnosed as having corpora lutea; 2.9% of them had incomplete luteolysis and 5.1% may have lost their embryos between days 21 and 45 post-insemination. The pregnancy rate was 10% higher in the rainy season than in the dry season. Cows with body condition scores of 3 and 4 had a higher overall pregnancy rates than those with a body condition score of 2. The findings of this study further confirm the luteolytic efficacy of prostaglandin in inducing oestrus in zebu cattle and indicate that the nutritional status of the cows must be satisfactory before embarking on oestrus synchronization programmes.  相似文献

The PMSG-superovulatory response was determined by ovarian dissection in 20 cows and compared with estimates made by laparoscopy laparotomy and rectal palpation at day 7 of the induced estrus. The corpora lutea count made transrectally seriously underestimated real corpora lutea numbers when more than nine corpora lutea were present. Rectal palpation failed to correctly identify the number of follicles ≥ 10 mm when more than four follicles were present. After ovarian dissection, laparotomy was the most constant and accurate method to obtain the number of ovulations, followed by laparoscopy. The excessive weight and the large size of the ovaries associated with the presence of large unovulated luteinized follicles were often responsible for the erroneous estimates of the ovulation rate by rectal palpation.  相似文献

Fourteen anovulatory postpartum (38.0 +/- 1.9 d) beef cows that ovulated after an injection of 250 micrograms gonadotropin releasing hormone (GnRH) in saline were used to examine the influence of indomethacin on luteal function. Beginning the d after GnRH, 6 cows were given intrauterine infusions of indomethacin for 14 d and the other eight cows received vehicle. After GnRH treatment, concentrations of progesterone in serum were elevated longer (P less than .01) for indometacin-treated cows than for vehicle-treated cows. At the same time prostaglandin metabolite (PGFM) concentrations were lower (P less than .01) in indomethacin-treated cows than in vehicle-treated cows. In summary, indomethacin suppressed PGFM concentrations and enhanced function of corpora lutea induced in postpartum suckled beef cows.  相似文献

Ovarian function in 91 dairy cows with cystic ovarian disease was assessed by rectal palpation and by plasma hormone analysis before and after treatment. Plasma analysis showed that 84% of the cysts were correctly classified clinically and only these cows are considered further. Luteinised cysts occurred in 59 cows whereas only 18 had non-luteinised cysts. The mean plasma concentrations of luteinising hormone (LH), follicular stimulating hormone (FSH), progesterone, oestradiol and testosterone were not significantly different when compared with values at relevant stages of the oestrous cycle in normal cows. Success of treatment with progesterone, a synthetic prostaglandin, human gonadotrophin (HCG), or gonadotrophin releasing hormone (GnRH) was not dependent upon prior hormone concentrations, except for the prostaglandin which required active luteal tissue. LH and FSH concentrations in cows with luteinised cysts were not significantly different before and after successful treatment with GnRH or progesterone. Normal luteal function was not always established after treatment of non-luteinised cysts with GnRH.  相似文献

A study was undertaken in a California dairy to estimate the risk of transmission of bovine leukemia virus associated with rectal palpation of cows. The probability of seroconversion within 3 months after rectal palpation was examined as a function of the prevalence of infection in cows at the time of palpation. It was hypothesized that the probability of seroconversion within 3 months after palpation would be higher for cows in a group with high prevalence of infection than for cows in a group with low prevalence. The probability of seroconversion after rectal palpation was equal to 0.034, and results of logistic regression, using data from 1,116 noninfected and 1,047 infected cows, did not indicate any significant association between probability of seroconversion after rectal palpation and prevalence of infection (P = 0.189). Results of this study suggest that risk of bovine leukemia virus transmission by rectal palpation typically used in reproductive tract examination of cows either does not occur or is uncommon.  相似文献

AIMS: To determine the level of agreement between transrectal ultrasonography, manual palpation and measurement of progesterone in the detection of corpora lutea in post-partum cows. To evaluate the reproductive performance of cows not detected in oestrus prior to mating, but detected with a corpus luteum at the start of mating. METHODS: Cows from seven herds which were not detected in oestrus were examined between 5 and 7 days before the herd's planned start of mating. Ovaries were examined by palpation and ultrasonography in 160 cows and plasma progesterone concentrations were also measured in 103 cows. Insemination dates and pregnancy data were recorded in 159 cows not detected in oestrus but having a corpus luteum and 1405 cycling herd mates. RESULTS: The level of agreement between ultrasonography and measurement of progesterone (kappa = 0.74) was higher than with manual palpation (kappa = 0.67) although it was significant for both techniques (p <0.001). Cows which were not detected in oestrus, but detected with a corpus luteum had a longer interval to first service (12 v. 10 days, p <0.05) and conception (19 v. 15 days, p = 0.01) than cycling herd mates and had a higher empty rate (9.7% v. 3.8%, p <0.001). CONCLUSIONS: These data indicate that ultrasonography may provide a better diagnostic tool for examining cows not detected in oestrus than manual palpation or measurement of progesterone concentration. Cows not detected in oestrus but having a corpus luteum were shown to have significantly poorer reproductive performance than cycling herd mates.  相似文献

Estimates were made of the superovulation response in donor cows using ovarian size, the number of corpora lutea (palpated per rectum) and blood progesterone levels. Neither the estimated number of corpora lutea nor ovarian size gave a satisfactory prediction of superovulation response. There was a large discrepancy between the number of corpora lutea present on the ovaries of nine superovulated cows (166) and the estimated number (106). All the corpora lutea on superovulated ovaries were smaller than normal (1.95 g). Blood progesterone levels at the time of embryo recovery were correlated with both the number of corpora lutea on the superovulated ovaries (r = 0.9, P less than 0.001) and the weight of the corpora lutea (r = 0.95, P less than 0.001). There were 67 embryos and ova recovered from the nine donors, representing 40 per cent of the actual and 63 per cent of the palpated corpora lutea. However, neither ovarian size, the number of corpora lutea nor blood progesterone levels were correlated with embryo production.  相似文献

Two methods for a scheduled post partum examination were compared on a commercial dairy farm. In Group 1, all cows (n = 601) were examined by rectal palpation between day 20 and 26 post partum for signs of endometritis (vaginal discharge, enlarged uterus). In Group 2, all cows (n = 652) were examined by external inspection for vaginal discharge as a sign for endometritis. In both groups all cows with endometritis were treated twice with prostaglandin F2 alpha (25 mg of dinoprost) in a 14-day interval. The proportion of cows with signs of endometritis was 33.3% and 17.2% in Group 1 and Group 2, respectively (p < 0.05). In Group 1, the conception rate (39.0% vs 49.3%) and the proportion of cows pregnant (60.5% vs 72.6%) were lower for cows with endometritis than for cows without endometritis. In Group 2, no significant differences were found in reproductive performance for cows with and without signs of endometritis. No significant differences in reproductive performance were found for cows with endometritis between the two groups. Also, for cows without endometritis no significant differences in reproductive performance were found between the groups. It is concluded that the post partum examination by rectal palpation was more sensitive in finding cows with endometritis. However, with regard to the reproductive performance the more sensitive method was not more effective than the method based on systematic external inspection.  相似文献

In 160 cows with ovarian cysts as determined by rectal palpation, differentiation was made of follicular cyst, luteal cyst, and cystic corpus luteum on the basis of milk progesterone concentrations estimated by an enzyme immunoassay before and at 10 days after cows were treated with gonadotropin-releasing hormone. Cows having a progesterone concentration in skim milk less than 1.0 ng/ml were considered to have follicular cysts and those with concentrations of 1.0 ng/ml or higher were regarded as the cases of luteal cyst or cystic corpus luteum. Luteal cyst was characterized by progesterone values remaining high in the cows for 10 days after treatment, and cystic corpus luteum was characterized by a decrease in progesterone concentration after cows were treated. By the rectal palpation procedure it was impossible to differentiate luteal cyst and cystic corpus luteum from follicular cyst. The frequencies of follicular cyst, luteal cyst, and cystic corpus luteum were 65%, 19%, and 16%, respectively. Of 104 cows with follicular cysts as defined by milk progesterone assay result, 73 (70%) responded to the treatment with gonadotropin-releasing hormone, the milk progesterone concentration increasing from 0.7 +/- 0.2 ng/ml (mean +/- SD) to 1.8 +/- 1.1 ng/ml. The accuracy of rectal palpation 10 days after treatment for judgment of luteinization of follicular cyst confirmed by milk progesterone analysis was only 30% (48 cows of 160).  相似文献

OBJECTIVE: To evaluate the sensitivity and specificity of single manual examination of ovarian surface structures and of ovarian size as tests for the detection of corpora lutea in non-cycling cows DESIGN: Non cycling cows were dichotomously classified for the presence of palpable corpora lutea and for ovarian size. The performance characteristics of the palpable corpus luteum test and the ovarian size test were estimated using Bayesian analysis. Previously published information on prevalence and test performance was combined with observed data using the Gibbs Sampler to derive posterior distributions for prevalence and test parameters. RESULTS: Prior distributions for the prevalence of corpora lutea in cows not detected on heat before mating within seasonal herds were centred on 25%, and for the corpus luteum test sensitivity were centred on 70%. No prior assumptions for any other test parameter were made. From a total of 650 cows, 144 were found to have at least one corpus luteum, and 156 were found to have two small ovaries. The posterior estimate obtained for prevalence was 0.30 (95% Cl 0.22 to 0.43), for corpus luteum test sensitivity was 0.71 (95% Cl 0.49 to 0.93), and for corpus luteum test specificity was 0.98 (95% Cl 0.49 to 1.00). For the ovarian size test, the posterior estimate for size test sensitivity was 0.98 (95% Cl 0.95 to 1.00) and for size test specificity was 0.34 (95% CI 0.28 to 0.43). Sensitivity analysis indicated that corpus luteum test sensitivity ranged from 0.60 to 0.70 in most herds. CONCLUSIONS: Errors associated with pre-mating heat detection are likely to result in between 15 to 30% of cycling cows not detected in oestrous before mating, resulting in the inclusion of cycling cows within the population of non-cycling cows. This mixed population of cows is then subjected to manual examination by veterinarians in order to assign cows to treatment groups. The corpus luteum test has modest sensitivity and high specificity and the size test has high sensitivity and low specificity. Therefore the use of a single examination of the ovaries of cows not detected on heat before the mating period, in order to classify them as anovulatory anoestrous cows with or without a corpus luteum, is not sufficient to accurately classify them and thus to recommend treatment.  相似文献

The aim of this study was to investigate the effect of applying a progesterone‐based oestrous synchronization protocol at 51–57 days postpartum in high‐producing dairy cows. The data analysed were derived from 1345 lactating cows. Cows between 51 and 57 days postpartum were assigned to the groups: control, PRID (receiving a progesterone‐releasing intravaginal device for 9 days, and prostaglandin F 24 h before PRID removal) or GnRH–PRID (the same as the PRID group plus GnRH at PRID insertion). Oestrus was detected by using pedometers and confirmed by examination of the genital tract at AI. Oestrous and conception rates before days 71–77 postpartum, pregnancy loss in early pregnant cows or the cumulative conception rate registered on day 120 postpartum were considered as the dependent variables in four consecutive logistic regression analyses. Based on the odds ratios, the oestrous rate increased by a factor of 1.73 in cows showing oestrus before treatment for each unit increase in the number of previous oestruses; decreased by a factor of 0.44 in the control group with respect to the treatment groups; and by a factor of 0.61 in cows without luteal structures at treatment with respect to cows with corpora lutea. The conception rates of cows inseminated before days 71–77 postpartum remained similar across the groups, whereas the likelihood of pregnancy loss for cows becoming pregnant during this period was 0.11 times lower in the PRID group than in the control. Based on the odds ratio, the likelihood of a higher cumulative conception rate on day 120 postpartum: increased in cows showing oestrus before treatment by a factor of 1.41 for each unit increase in the number of previous oestruses, was reduced 0.56‐fold in control cows compared with treated cows, and was also reduced by a factor of 0.98 for each kilogram of milk production increase recorded at treatment. In conclusion, although oestrous synchronization programmes performed in this study did not improve fertility, cows treated with progesterone could be inseminated earlier than untreated cows, such that the treatments increased the cumulative pregnancy rates determined on day 120 postpartum. In addition, fewer pregnancy losses were observed in early pregnant cows in the PRID group than the GnRH–PRID group.  相似文献
