The objective of this field study was to evaluate in an endemically porcine reproductive and respiratory syndrome (PRRS) virus-infected farm the reproductive performance of sows after their vaccination with a PRRS attenuated vaccine. In a farrow-to-finish pig farm with history of endemic PRRS virus infection, a total of 200 gilts and sows were used. They were divided in 2 groups of 100 animals. The first group was used as untreated controls, while the animals of the second group were vaccinated against PRRS virus using the attenuated Porcilis PRRS vaccine (Intervet International, The Netherlands) based on European strain. All health and reproductive parameters were recorded from the time of vaccination up to next weaning. No adverse systemic or local reactions or side effects relative to vaccination were noted. Compared to controls, vaccinated sows showed significantly improved farrowing rate (89% versus 78%) and a tendency for fewer returns to oestrus, particularly those at irregular intervals. Fewer sows farrowed prematurely and showed post-partum dysgalactia syndrome, but more live pigs were born and weaned in each litter after vaccination. It was concluded that vaccination of sows with Porcilis PRRS attenuated vaccine in farms with endemic PRRSV infection has beneficial effects on their health and fertility.  相似文献

The purposes of this study were to describe the clinical signs observed in PRRS positive herds during a porcine reproductive and respiratory syndrome (PRRS) outbreak in Ontario and to determine associations between these clinical signs and herd demographics and PRRS control strategies. All PRRS polymerase chain reaction-(PCR)-positive submissions to a diagnostic laboratory between September 1, 2004 and August 31, 2007 were identified (n = 1864). After meeting eligibility requirements and agreeing to voluntary study participation, producers from 455 of these submissions were surveyed for information on clinical signs observed in their herds, herd demographics, and PRRS control strategies used in their herds at the time that the PCR-positive samples were taken. Larger herd size was associated with an increased risk of reporting abortion, weakborn piglets, off-feed sows, and sow mortality in sow herds, and with an increased risk of reporting mortality in finishing herds. When disease control strategies were examined, use of a commercial PRRS vaccine in sows and gilts was associated with a decreased risk of reporting weakborn pigs and high pre-weaning mortality, while the use of serum inoculation in breeding animals was associated with an increased risk of reporting off-feed sows and sow mortality. Providing biofeedback of stillborn/mummified piglets, placenta or feces to gilts was associated with an increased risk of reporting respiratory disease and mortality in finishing pigs while all-in/all-out flow in farrowing rooms was associated with an increased risk of reporting sow mortality and weakborn piglets.  相似文献

The objectives of this study were to determine if coinfection of segregated early weaned (SEW) pigs with porcine circovirus type 2 (PCV2) and porcine parvovirus (PPV) induces an increase in the incidence of post-weaning multisystemic wasting syndrome (PMWS) compared to singular PCV2 infection, and to determine if vaccination against PPV protects pigs against PMWS associated with PCV2/PPV coinfection in SEW pigs. Seventy, 3-week-old, SEW pigs were randomly assigned to one of the five groups. Pigs in group 1 (n = 14) served as the negative controls, group 2 pigs (n = 14) were inoculated with PCV2, group 3 pigs (n = 12) were inoculated with PPV, groups 4 (n = 16) and 5 (n = 14) pigs were inoculated with both PCV2 and PPV. Pigs in groups 1-3 and 5 were vaccinated with two doses of a killed parvovirus-leptospira-erysipelothrix (PLE) vaccine prior to inoculation. The PCV2/PPV-coinfected pigs (groups 4 and 5) had significantly (P < 0.05) higher and more persistent fevers than the singular PCV2-infected pigs. One pig in each of the coinfected groups developed clinical disease (fever, respiratory disease, jaundice, weight loss) consistent with PMWS. Lymphoid depletion was significantly (P < 0.05) more severe in the dually-infected pigs at 42 days post-inoculation (DPI). Vaccinated, coinfected pigs (group 5) remained viremic significantly (P < 0.05) longer and had higher copy numbers of genomic PCV2 DNA in sera at 28, 35, and 42 DPI compared to the unvaccinated coinfected pigs (group 4). PPV-viremia was detected only in the unvaccinated group 4 pigs. PLE-vaccination prevented PPV-viremia but did not prevent clinical PMWS or reduce the severity of lymphoid depletion in PCV2/PPV-coinfected pigs. Evidence of increased incidence of clinical PMWS due to vaccination was not observed in this model.  相似文献

The objectives of the present study were to compare Mycoplasma hyopneumoniae (Mh) colonization and serologic status on Mh vaccinated and non-vaccinated sows and to assess the effect of sow vaccination on colonization and serologic status of their piglets at weaning as well as presence of enzootic pneumonia (EP) lung lesions at slaughter. Fifty sows (25 vaccinated and 25 unvaccinated) as well as five of their piglets were included in the study. Blood samples and nasal swabs from sows at 7 weeks pre-farrowing and 1 week post-farrowing and from piglets at 3-4 weeks of age were taken. Nasal swabs and sera were tested by a nested polymerase chain reaction (nPCR) to detect Mh DNA and by an enzyme-linked immunosorbent assay (ELISA) test to detect antibodies to the pathogen, respectively. Finally, at 23 weeks of age, pigs were sent to the slaughter where the extension of EP-compatible gross lesions was assessed. Vaccination with two doses of Mh vaccine resulted in a significantly higher (p<0.05) percentage of seropositive sows than in the non-vaccinated group at 1 week post-farrowing. On the contrary, no statistical significant differences were found in the number of nasal nPCR positive sows among different treatments (p>0.05). At 3-4 weeks of age, a significantly higher percentage (p<0.001) of seropositive piglets came from vaccinated than from non-vaccinated sows. Although the number of Mh infected piglets coming from non-vaccinated sows was higher than the one from vaccinated sows, the difference was not statistically significant (p>0.05). Overall, piglets from vaccinated sows had a significant lower (p<0.05) mean of EP-compatible lung lesions (1.83+/-2.8) than piglets from non-vaccinated sows (3.02+/-3.6). Under the conditions described in this study, sow vaccination did not affect sow or piglet colonization but increased the percentage of seropositive sows and piglets at weaning and reduced significantly the mean EP-compatible lung lesion scoring at slaughter.  相似文献

The purpose of this study was to investigate the safety and efficacy of a commercial European porcine reproductive and respiratory syndrome virus (PRRSV)‐inactivated vaccine after 18‐month use in gilts/sows at a farm with high seroprevalence. In a farrow‐to‐finish farm with 1100 sows, all sows and gilts were systematically vaccinated with the PRRS‐inactivated PROGRESSIS® vaccine for a period of 18 months. Farm's reproductive and litter characteristics were longitudinally recorded for this period and historically compared with those of the year prior to vaccination. Serology, employing immunoperoxidase monolayer assay, had confirmed a high prevalence of PRRS‐specific antibodies in most age groups within the farm prior to vaccination. Seroprevalence during the experiment ranged between 0% and 100% in weaners and growers, but remained at stable high levels (>93%) in finishing pigs and gilts throughout all 2‐year period of serology measurements. No local or systemic vaccine side effects were noted throughout the trial period. Vaccinations had resulted over time in a significant improvement of sow reproductive performance (e.g. reduction of premature farrowings, abortions and increase of farrowing rate) and litter characteristics (e.g. increase of the number of live born and weaned pigs and decrease of stillborn, mummified, weak and splay‐legged piglets). It has also been observed that the higher the degree of immunization of a sow, the better the improvement of her reproductive parameters. Sows after vaccination have shown improved characteristics compared to homoparous sows prior to the application of vaccinations in the farm.  相似文献

OBJECTIVE: To document shedding of porcine reproductive and respiratory syndrome (PRRS) virus in mammary gland secretions of experimentally inoculated sows, to evaluate effects of vaccination during gestation on virus shedding during the subsequent lactation, and to evaluate shedding of PRRS virus in milk of sows in commercial herds. ANIMALS: 6 sows seronegative for PRRS virus were used for experiment 1, and 2 sows were retained for experiment 2. For experiment 3, 202 sows in commercial herds were used. PROCEDURE: In experiment 1, 2 sows were inoculated with PRRS virus, 2 sows were vaccinated with modified-live PRRS virus vaccine, and 2 sows served as control pigs. Mammary gland secretions were assayed for PRRS virus. In experiment 2, pregnant vaccinated sows from experiment 1 were vaccinated with another modified-live PRRS virus vaccine. Mammary gland secretions were assayed in the same manner as for experiment 1. For experiment 3, milk collected from 202 sows in commercial herds was assayed for PRRS virus. RESULTS: In experiment 1, PRRS virus was detected in mammary gland secretions of both vaccinated and 1 of 2 virus-inoculated sows. In experiment 2, virus was not detected in samples from either vaccinated sow. In experiment 3, all samples yielded negative results. CONCLUSIONS AND CLINICAL RELEVANCE: Na?ve sows inoculated late in gestation shed PRRS virus in mammary secretions. Previous vaccination appeared to prevent shedding during the subsequent lactation. Results for samples obtained from sows in commercial herds suggested that virus shedding in mammary gland secretions of such sows is uncommon.  相似文献

In order to control growing problems associated with porcine circovirus type 2 (PCV2) several vaccines for piglets or sows were introduced recently. An objective of the study was to compare an efficacy of three different vaccination protocols in the herd with acute postweaning multisystemic wasting syndrome (PMWS) outbreak affecting 3-month-old pigs. All of three applied protocols, namely vaccination of sows, piglets or sows and piglets with Circovac proved to be efficacious in controlling of PMWS. All production parameters significantly improved after vaccination. Obtained values were as good as before the outbreak or even better as in case of average daily weight gains. However, decreased mortality before weaning was recorded only after vaccination of sows while in groups where piglets were vaccinated significantly lower mortality in fourth month of life was observed. The impact of different protocols on different parameters suggests that they could be adopted in herds with different porcine circovirus associated problems.  相似文献

The trial involved eight large white sows obtained from a closed experimental specific pathogen free herd. Four sows (two each for an experimental vaccine and for Nobi-Vac AR) were vaccinated twice (eight weeks and two weeks before parturition) with 2 ml of vaccine administered intramuscularly. Two unvaccinated sows were used as an infected control group and two unvaccinated sows served as an uninfected control group. Forty-six piglets (28 from vaccinated sows and 18 from unvaccinated sows) were challenged by intranasal instillation of Bordetella bronchiseptica at two days of age and Pasteurella multocida type D, dermonecrotic toxin at seven days of age. Among the infected control group some piglets died and there were clinical signs of pneumonia and severe turbinate atrophy. In the vaccinated groups the results showed that immunisation of the pregnant sows had provided a good level of antibodies, which were transmitted to their offspring. There was a significant reduction in the clinical signs and no lesions were observed in the group vaccinated with the experimental vaccine and only moderate atrophy of the turbinates in the Nobi-Vac AR group. B bronchiseptica and P multocida were never recovered from the lungs of the vaccinated groups and in the nasal cavities their frequency declined with age.  相似文献

In a Haemophilus pleuropneumonia problem herd with piglet production and fattening, sows and weaned pigs were vaccinated with a Danish vaccine (Pleurinord). Due to consequent vaccination of the sows the health of the piglets could be improved decisively, resulting in an increased number of raised piglets per sow and year from 16.8 to 20.3. In the vaccinated fattening pigs compared to the unvaccinated ones the following effects were observed: markedly reduced expenses for medication and a markedly reduced frequency of characteristic lesions in the respiratory tract; the improvement of the daily weight gains and the reduction of losses remained behind the expectations. Decisive causes for this were respiratory and enteric diseases unrelated to Haemophilus, which were favoured by serious mismanagement and inadequacies in the feeding regimen and barn climate. On the example of the vaccinated herd it is shown how important the analysis of a multifactorial disease situation is, in order to be able to objectify better or at all the influence of a vaccination program under field conditions. Within a concept of prevention and control of Haemophilus pleuropneumonia the vaccination is a helpful part.  相似文献

Little is known about the safety and efficacy of extra-label use of the modified live porcine reproductive-and-respiratory syndrome (PRRS) virus vaccine in gestating sows. Our purpose was to determine the impact of vaccination on reproductive performance in 54 herds in Ontario, Manitoba (Canada) and the mid-western United States that were PRRS-positive, PRRS-negative, or concurrently affected by an outbreak of PRRS when initially vaccinated. Majority-vaccinated herds vaccinated ≥50% but <100% of sows at one time, and limit-vaccinated herds vaccinated <50% of sows at one time. Most majority-vaccinated herds did not vaccinate sows in late gestation, and none vaccinated during the initial PRRS outbreak. Numbers of pigs born alive and weaned decreased when pregnant sows were vaccinated. The effect of vaccination on productivity in the gestation following vaccination depended on the vaccination protocol.  相似文献

The use of a vaccine against classical swine fever virus (CSFV) during an outbreak of CSF should lead to a reduction in the horizontal or vertical transmission of CSFV. The reduction of vertical, i.e. transplacental, transmission of a moderate-virulent strain of CSFV from the sow to its offspring was studied in sows vaccinated once or twice with a CSFV E2 subunit vaccine. Two groups of nine sows were vaccinated with one PD95 dose of the E2 subunit vaccine, approximately four weeks before insemination. A third group of nine inseminated sows served as controls. One group of nine sows were vaccinated again at two weeks after insemination. At ten weeks after the primary vaccination, approximately six weeks after insemination, all 27 sows were challenged intranasally with 10(5) TCID50 of a moderate-virulent strain of CSFV, the Van Zoelen strain. The sows were euthanized at five weeks after challenge, and samples from the sows and fetuses were collected for detection of CSFV. All 27 sows were in gestation at the time of slaughter, CSFV was detected in the fetuses of all unvaccinated sows but it was not detected in any of the samples collected from fetuses of the double-vaccinated sows. Virus was however recovered from the fetuses of one out of nine sows vaccinated once. All the sows, except four double-vaccinated sows, developed CSFV Erns antibodies. Transplacental transmission of CSFV was reduced significantly (p <0.001) in all vaccinated sows. When the results from the experiment were extrapolated to a herd level, it could be concluded that, with 95% certainty, approximately 11% (single vaccination) or 0% (double vaccination), confidence intervals of 0.01-0.44 and 0.0-0.30 respectively, of the pregnant sows would still not be protected against vertical transmission of moderate-virulent CSFV. We conclude that vaccination with the CSFV E2 subunit vaccine can reduce the transmission of moderate-virulent strain of CSFV from the sow to its offspring significantly.  相似文献

Potential risk factors for clinical signs of post-weaning multisystemic wasting syndrome (PMWS) and porcine dermatitis and nephropathy syndrome (PDNS) in pigs in the Netherlands were investigated in a matched case-control study using a questionnaire (personal interview). Eighty-two pig farmers were questioned about management, hygiene, husbandry systems, disease history, and preventive health care. In this study, 30 pig herds with (cases) and 30 pig herds without (controls) characteristic clinical signs of PMWS were compared. For PDNS, 11 pig herds with (cases) and II pig herds without (controls) characteristic clinical signs of PDNS were compared. Univariate analysis (P < 0.10) showed that the following occurred relatively more often in the PMWS case herds than in the control herds: 1) clinical signs of PDNS, porcine reproductive and respiratory syndrome (PRRS), porcine parvovirus (PPV) infections, meningitis, coccidiosis, and pre-weaning diarrhoea observed by the farmer; 2) vaccination against PRRS and mycoplasma; 3) non-optimal climatic conditions in the nursery rooms, a large variation in weaning age, a high occurrence of cross-fostering of piglets, a large number of sows with lactation problems, poor colostrum intake by piglets; and 4) (historical) use of breeding stock (including semen for artificial insemination) of Anglo-Saxon origin. In the final multivariate statistical model, one variable remained significantly associated with PMWS case herds, namely, the presence of clinical signs of PRRS (and/or the associated use of vaccination against PRRS). It should be noted that in almost all cases animals were vaccinated against PRRS because of clinical signs of PRRS that appeared a few months after the first occurrence of clinical signs of PMWS. This excludes PRRS vaccination as a primary factor in causing PMWS. Analysis of the PDNS case-control data showed comparable results with those of the PMWS study. In the final statistical model, the presence of clinical signs of PRRS (and/or the associated use of vaccination against PRRS) was significantly associated with PDNS case herds.  相似文献

Several Aujeszky's disease virus (ADV) vaccination protocols of sows were evaluated with regard to the passive protection conferred on piglets in a recently built commercial farm. Three different groups of sows were vaccinated using a Bartha K-61 strain. One group received an inactivated vaccine during pregnancy and the other two groups received attenuated vaccines, either during pregnancy (day 65) or on the seventh day of lactation. At farrowing, sows vaccinated during lactation had lower seroneutralization titres than those vaccinated during pregnancy either with inactivated or attenuated vaccines. Accordingly, their piglets were the ones with lower levels of maternally transferred neutralizing antibodies. At 4 weeks of age, five piglets born of each group of sows were challenged intranasally with a neurotropic strain of ADV. Piglets born of sows vaccinated during pregnancy with inactivated and attenuated vaccines gained 1.50 kg bodyweight and 2.50 kg bodyweight during 7 days, respectively, and did not show clinical signs, while piglets from sows vaccinated during the previous lactation lost 0.60 kg and presented moderate to severe clinical signs of ADV. Vaccination of sows during pregnancy provided more protection against ADV for piglets than sow vaccination before mating. Piglets born from sows vaccinated with attenuated or inactivated vaccines did not present remarkable differences on protection.  相似文献

In a large herd of pigs where the parenteral immunisation of pregnant sows by polyvalent vaccine against enteral coliinfections of new-born piglets was performed successfully for several years, an increased occurrence of diseases and mortality of sucking piglets was observed. After screening tests in the herd, six strains of E. coli, differing from the strains contained in the commercial vaccine, were isolated. The obtained strains were employed for preparation of peroral vaccine for sows by means of the modified method after Kohler. In two cycles of sows (48 animals in experimental group and 60 animals in control group), this method was compared with the effectiveness of the commonly used commercial polyvalent parenteral vaccine against enteral E. coli infections of new-born piglets (manufactured by Bioveta, Ivanovice in Haná). In experimental sow groups vaccinated perorally by the new vaccine, the number of live-born piglets increased by 0.475 piglet per litter, the number of reared piglets up to the age of 28 days by 0.904 piglet per litter and the mortality till the age of 28 days decreased by 6.1% as compared with the control vaccinated by commercial vaccine. The results were not statistically significant. The advantages and disadvantages of both vaccination methods are discussed.  相似文献

Porcine reproductive and respiratory syndrome (PRRS) virus infection causes reproductive failures including return to oestrus, abortion, mummified foetuses, stillborn, and weak-born piglets. The objective of the present study was to investigate reproductive performance of sows in PRRS-virus-seropositive herds with and without PRRS modified live virus (PRRS-MLV) vaccination. The study was conducted in 20 PRRS-virus-seropositive commercial swine herds in Thailand. The data included 211,009 mating and 180,935 farrowing records. The analysed variables included farrowing rate (FR), return rate (RR), abortion rate (AR), total number of piglets born per litter (TB), number of piglets born alive per litter (BA), percentage of stillborn (SB), percentage of mummified foetuses (MM), and number of piglets weaned per litter (WP). The results revealed that FR in non-vaccinated sows was lower than that in vaccinated sows (85.0 vs 89.7 %, respectively, P?<?0.001), and RR in non-vaccinated sows was higher than that in vaccinated sows (6.9 vs 3.7 %, respectively, P?<?0.001). AR did not differ significantly between non-vaccinated and vaccinated sows (1.6 and 2.0 %, respectively, P?=?0.964). TB (11.2 and 11.5, respectively, P?<?0.001), BA (10.0 and 10.6, respectively, P?<?0.001), and WP (9.2 and 9.6, respectively, P?<?0.001) in non-vaccinated sows were lower than those in vaccinated sows. SB (6.9 and 5.1 %, respectively, P?<?0.001) and MM (3.2 and 2.2 %, respectively, P?<?0.001) in PRRS-MLV-vaccinated sows were higher than those in non-vaccinated sows. The improvement in sow reproductive performance in PRRS-MLV-vaccinated herds was most pronounced in gilts and primiparous sows.  相似文献

In order to minimize the effects of porcine reproductive and respiratory syndrome (PRRS) on stillbirth, mummification, and neonatal mortality in swine herds, many producers have vaccinated their herds using a modified-live virus vaccine. The purpose of this study was to determine the association of the PRRS modified-live vaccine and reproductive performance by stage of gestation when the vaccine was administered. A total of 47 swine herds from Ontario and Manitoba, Canada, and from the mid-western USA were included in the study. Participating farms had vaccinated all of their sows at one point in time when they used the vaccine for the first time. The reproductive performance of sows that farrowed in the year prior to use of the vaccine was compared to that of sows vaccinated in each of five stages of gestation and in the gestation that followed the initial use of the vaccine. Sows vaccinated at any time during gestation had a reduced number of pigs born alive, a reduced number of pigs weaned per litter, and increased number of stillborn pigs and an increased number of mummified pigs compared to the sows that farrowed prior to use of the vaccine. The largest association was seen in sows that were vaccinated in the last four weeks of gestation. The largest losses were observed in those herds that were vaccinated concurrently with the initial PRRS herd outbreak. These results suggest that the modified-live vaccine should only be administered to non-gestating sows.  相似文献

The efficacy of a new vaccine against atrophic rhinitis in pigs was tested in the Netherlands and Denmark. The vaccine contained protein dO (a truncated Pasteurella multocida toxin which is immunogenic and non-toxic), inactivated Bordetella bronchiseptica whole cells, and an adjuvant. The sows were either vaccinated intramuscularly with 2 ml of the vaccine at six to eight and two to four weeks before expected farrowing or left unvaccinated as controls. All the piglets were challenged intranasally with B bronchiseptica when three to seven days old and with P multocida three to four days later. Pigs born to the vaccinated sows performed significantly better than pigs born to the control sows when judged on growth, average daily weight gain and snout scores. The challenge organisms were reisolated more frequently from the control pigs than from the pigs in the vaccinated group. The vaccinated sows and their progeny developed high titres of antibodies against B bronchiseptica and P multocida toxin.  相似文献

The possible effect on pig protection after vaccination pregnant sows and their piglets against FMD at various age was examined using the SN test. Three experiments were conducted with three sow in each (8-9 piglets each). In the first experiment sows were not vaccinated but their piglets were vaccinated on 10th, 20th and 60th day of age. In second experiment sows were vaccinated at the end of rest period and in the middle of pregnancy. Pigs from one sow were vaccinated on 10th, from second sow on 20th and from the third sow on the 60th day of age. Pigs which were vaccinated on the 10th and 20th day of age were revaccinated on the 60th day of age. In third experiment sows were vaccinated at the end of the rest period and in the middle of pregnancy. Their piglets were not vaccinated. The conclusion may be drawn that if the epidemiological situation requires, systemic vaccination of all pigs then, on large farms (where pig production is planned) all sows should be vaccinated at the end of the rest period (first vaccination) and on the 55th-60th day of pregnancy. The next vaccination should be done in the middle of the next pregnancy. A vaccination program of piglets, according to our results, should depend on the vaccination program used for their dams.  相似文献

No local or systemic vaccine side effects were noted throughout the trial period. Vaccinations had resulted over time in a significant improvement of sow reproductive performance (e.g. reduction of premature farrowings, abortions and increase of farrowing rate) and litter characteristics (e.g. increase of the number of live born and weaned pigs and decrease of stillborn, mummified, weak and splay-legged piglets). It has also been observed that the higher the degree of immunization of a sow, the better the improvement of her reproductive parameters. Sows after vaccination have shown improved characteristics compared to homoparous sows prior to the application of vaccinations in the farm.  相似文献
