The structure and histochemical properties of the intestine in bronze corydoras (Corydoras aeneus), a stomach‐containing teleost, are described, with emphasis on goblet cells and mucin types. The proximal intestine displayed a normal structure for teleosts, whereas the distal intestine was wide, translucent, thin‐walled, richly vascularized and constantly filled with air, suggesting an important respiratory role. Goblet cells were common throughout the entire intestine and displayed a variable, but mainly faint metachromatic colour after toluidine blue. They were moderately coloured by alcian blue at both pH 2.5 and 0.2 and displayed no colour after periodic acid followed by Schiff's solution (PAS), but a distinct purple‐brown colour after high iron diamine followed by alcian blue (pH 2.5). Together, these results suggest that the mucin in the intestine goblet cells consists mainly of sulphated proteoglycans. Further, the results from the present lectin and neuraminidase tests suggest that these mucins contain much N‐acetylglucoseamines and some N‐acetylgalactosamines and sialic acid, but seem to lack glucose and mannose. They also contain some galactose‐N‐acetylgalactosamines sequences, normally hidden by sialic acid. The distinct brush border and mucus layer on the epithelial cells in the respiratory intestine may indicate some digestive roles, such as absorption of water, ions and simple carbohydrates. As sulphated proteoglycans are tough and attract much water, this mucus may play important roles in the protection against mechanical and chemical damages and in the defence against micro‐organisms throughout the entire intestine, but in the respiratory intestine it may impede significantly the oxygen uptake. However, as this part of the intestine usually contains no digesta, but is completely filled with air, frequently renewed by dry air from the atmosphere, and the main function of the mucus may be to protect the respiratory epithelium against a destroying and dangerous desiccation.  相似文献

The appearance and histochemical properties of mucus cells in various parts of digestive tract in prenatal larvae of platyfish (Xiphophorus maculatus), a stomachless teleost, are described and compared. Such cells occurred regularly throughout the entire epithelial cell layer in mouth, pharynx, oesophagus and intestine in 6 mm long larvae i.e. approximately 1 week before delivery, but were very scarce in 4 mm long larvae. The mucin in these cells was strongly stained by alcian blue at pH 2.5 and weakly coloured by this dye at pH 1.0. It displayed a clean blue colour after high iron diamine followed by alcian blue (pH 2.5), except in some very few cells which displayed a purple-brown colour after this treatment. The mucus cells displayed various colours between purple-blue and magenta when the alcian blue (pH 2.5) was followed by periodic acid-Schiff (PAS) solution; the magenta-coloured cells were mainly confined to oesophagus. Together these findings suggest that the mucin in the digestive tract of prenatal platyfish consists mainly of non-sulphated, carboxylated glycoconjugates. The observations from the present lectin histochemistry suggest that the amount of galactosamine and glucosamine in this mucin varies much from cell to cell. Also, the various colours after PAS suggest a significant cellular heterogeneity in the mucus cell population. This may reflect different stages in development and maturation or a true and permanent cellular heterogeneity, suggesting that this mucus is able to play a number of important roles even a week before the larvae start to feed and digest.  相似文献

A histochemical study using conventional carbohydrate histochemistry (periodic‐acid staining including diastase controls, alcian blue staining at pH 1 and 2.5) as well as using a battery of 14 fluorescein isothiocyanate (FITC)‐labelled lectins to identify glycoconjugates present in 10 different areas of the skin of a catfish (Arius tenuispinis) was carried out. The lectins used were: mannose‐binding lectins (Con A, LCA and PSA), galactose‐binding lectins (PNA, RCA), N‐acetylgalactosamine‐binding lectins (DBA, SBA, SJA and GSL I), N‐acetylglucosamine‐binding lectins (WGA and WGAs), fucose‐binding lectins (UEA) and lectins which bind to complex carbohydrate configurations (PHA E, PHA L). Conventional glycoconjugate staining (PAS staining, alcian blue at pH 1 and 2.5) showed that the mucous goblet cells contain a considerable amount of glycoconjugates in all locations of the skin, whereas the other unicellular gland type, the club cells, lacked these glycoconjugates. The glycoproteins found in goblet cells are neutral and therefore stain magenta when subjected to PAS staining. Alcian blue staining indicating acid glycoproteins was distinctly positive at pH 1, but gave only a comparable staining at pH 2.5. The mucus of the goblet cells therefore also contains acid glycoproteins rich in sulphate groups. Using FITC‐labelled lectins, the carbohydrate composition of the glycoproteins of goblet cells could be more fully characterized. A distinct staining of the mucus of goblet cells was found with the mannose‐binding lectins LCA and PSA; the galactosamine‐binding lectins DBA, SBA and GLS I; the glucosamine‐binding lectin WGA; and PHA E which stains glycoproteins with complex carbohydrate configurations. No reaction occurred with the fucose‐binding lectin UEA and the sialic acid‐specific lectin SNA. In addition, the galactose‐binding lectins PNA and RCA showed only a weak or completely negative staining of the mucus in the goblet cells. The specificity of the lectin staining could be proved by inhibiting binding of the lectins by competitive inhibition with the corresponding sugars. From these data, we can conclude that the mucus produced by the epidermal goblet cells of A. tenuispinis is rich in mannose, N‐acetylgalactosamine and N‐acetylglucosamine residues.  相似文献

Histopathologic features of hepatic peribiliary cysts were described in a young slaughtered pig. The animal was an apparently healthy 6-month-old pig of mixed breed. Macroscopically, all lobes of the liver contained numerous cysts of varying size containing serous fluid in all lobes. Histopathologically, the cysts were located mainly around the large bile duct and in the connective tissue of the portal tracts. Within serial sections, these cysts were assumed to be solitary or multilocular, but they were separated from the bile duct. The cysts were lined by a single layer of columnar, cuboidal, and flattened epithelial cells. Occasionally, goblet cells were observed. The epithelial cells were stained with periodic acid-Schiff/alcian blue and high-iron diamine/alcian blue, indicating the presence of neutral mucin, sialomucin, and sulfomucin. Grimalius' method revealed the presence of endocrine cells in the lining epithelium. There was no bile pigment in the cysts by the Hall method.  相似文献

The objectives of these preliminary studies were to evaluate the use of hydrogen peroxide (H2O2) for the treatment of selected species of ornamental fishes and its efficacy in treating external bacteria and parasites. In the first part of the study, fish of five species (serpae tetra Hyphessobrycon eques (also known as Serpa tetra H. serpae), tiger barb Puntius tetrazona, blue gourami Trichogaster trichopterus, suckermouth catfish Hypostomus plecostomus, and green swordtail Xiphophorus hellerii) were exposed to H2O2 for 1 h at concentrations between 6 and 34 mg/L or for 24 h at concentrations between 1 and 6 mg/L. The results were species specific: green swordtails tolerated all of the treatments, serpae tetras and tiger barbs were sensitive only to the highest concentration, and mortalities of suckermouth catfish and blue gourami were recorded in every treatment. In the second part of the study, clinically healthy green swordtails and fish infested with external motile rod-shaped bacteria (i.e., Ichthyobodo spp., Trichodina spp., and Gyrodactylus spp.) were treated with several concentrations of H2O2. A single H2O2 treatment of 3.1 mg/L or more for 1 h effectively eliminated external bacteria, concentrations of 6.5 mg/L or more appeared to effectively kill Ichthyobodo spp., and none of the treatments tested was effective against Trichodina spp. or Gyrodactylus spp. These preliminary findings suggest that H2O2 is effective for treating certain external bacterial infections and flagellate infestations in some species of ornamental fish at the dosages tested. Other treatment regimens may need to be tested for effectiveness against Trichodina spp. and Dactylogyrus spp.  相似文献

Mycoplasma hyopneumoniae causes pneumonia in pigs. The effect of infection by this organism on histochemical characteristics of airway mucin within epithelial cells was studied. Seven- to 10-week-old pigs were inoculated intratracheally with M hyopneumoniae or culture broth, and lung tissues were collected from inoculated and control pigs at 2, 4, and 6 weeks after inoculation. Tissue sections were stained with periodic acid-Schiff/Alcian blue, pH 2.5 or high iron diamine/Alcian blue. Histologic features of randomly selected bronchi, bronchioles, and submucosal glands were compared in sections stained with periodic acid-Schiff/Alcian blue. Bronchial goblet cell sulfomucin and sialomucin were quantitated by image analysis of sections stained with high iron diamine/Alcian blue. Bronchi and bronchioles of infected pigs contained proportionately fewer goblet cells with mucin at all stages of infection than age-matched control pigs. Goblet cells in bronchi of infected pigs contained significantly less total mucin and sialomucin, and significantly more sulfomucin than goblet cells of control pigs. Increased sulfated mucin in bronchial goblet cells may reflect altered glycoprotein production or secretion in response to infection with M hyopneumoniae.  相似文献

Two different, computer-based, image analysis methods were employed to complement subjective assessment of patterns of mucin staining (by periodic acid Schiff/alcian blue, aldehyde fuchsin/alcian blue and potassium hydroxide-alcian blue/phenylhydrazine hydrocholoride) in digitised images of sections of major and minor salivary glands from cats and dogs. Image analysis based on red, green and blue (RGB) staining was not suitable for quantitation of histochemical staining of mucin in salivary glands. Image analysis based on hue, saturation and lightness (HSL) allowed quantitative assessment of staining by different stain components and of mixed staining, and enabled comparison of staining of different glands in dogs and cats. Quantitative analysis based on HSL allowed differentiation of differences in staining patterns of major and minor cat and dog salivary glands, and between the species; such differences would have been impossible to distinguish on subjective grounds alone. Quantitative assessment of normal salivary gland histochemistry allows comparison with staining patterns in disease.  相似文献

Mucinous carcinoma of the mammary gland is a rare tumor characterized by excessive mucin production. In human and canine pathology, the diagnosis of mucinous carcinoma is based on the demonstration of an epithelial phenotype of mucus-producing cells and periodic acid-Schiff (PAS)-diastase positivity of the mucin. The histologic and immunohistologic characteristics of feline mucinous mammary carcinoma were examined. Of 656 cases of feline mammary neoplasms and dysplasias, 3.2% were found to be mucin-producing tumors. Cytokeratin 19 (16 cases positive, 4 heterogenous, and 1 negative) and vimentin (15 cases positive, 2 heterogenous, and 4 negative) expression were examined, and the mucin produced was alcian blue positive. PAS-diastase staining was variable (38.1%). Based on these findings, mucinous mammary carcinoma in the cat varies significantly from the human and canine varieties and alcian blue is the prominent stain in the diagnosis of feline mucinous carcinoma.  相似文献

The number of goblet cells containing neutral and acidic mucins, including sulphomucins and sialomucins, was investigated in the small intestine of goat kids fed with lyophilized bovine colostrum in the period of passive immunity acquisition. At 0, 7 and 14 h of life, 15 male newborns received 5% of body weight of lyophilized bovine colostrum (LBC) and 14 male newborns received goat colostrum (GC), both with 55 mg/mL of IgG. Three additional animals were sampled at birth, without colostrum intake. Duodenum, jejunum and ileum samples were collected at 18, 36 and 96 h of life. Histological stains, periodic acid-Schiff, 1% alcian blue pH 2.5 and 1% alcian blue pH 1.0 were used to identify neutral and acidic mucins and acidic sulphated mucins, respectively. The number of goblet cells containing neutral and acidic mucins, including sulphomucins and sialomucins, does not differ in the duodenum (P>0.05). In the jejunum, LBC showed a higher number of goblet cells containing sialomucins compared to GC (P<0.05). The highest number of goblet cells containing acidic and neutral mucins and total number of goblet cells were observed at 96 h (P<0.05). In this segment, vacuoles of colostrum were present at 18 and 36 h mainly in the upper region of the villi, while the goblet cells were located at the bottom. At 96 h, vacuoles of colostrum were not detected, only goblet cells distributed throughout the villi. In the ileum, the number of goblet cells containing sulphomucins was higher (P<0.05) at 96 h than at 18 h. The LBC group showed higher (P<0.05) number of goblet cells containing sulphomucins at 96 h and total number of goblet cells at 36 and 96 h than the 0-h group. The present work revealed that the greater the absorption of colostrum in the goat kids' jejunum epithelium, the smaller the number of goblet cells. Considering this segment, feeding newborns with heterologous colostrum caused alteration in the number of goblet cells containing sialomucin. This condition suggested a reaction of the intestinal epithelium with increasing secretion due to the presence of non-recognized substances from the lyophilized bovine colostrum.  相似文献

Mucin hypersecretion is considered to be one of the most common components of the immune response to gastrointestinal nematode infection. However, investigations have not been conducted in the Cattle–Cooperia oncophora system to verify the findings largely derived from murine models. In this study, we examined the expression of seven mucins and seven enzymes in the mucin biosynthesis pathway involved in O-linked glycosylation in the bovine small intestine including goblet cells enriched using laser capture microdissection during a primary C. oncophora infection. At the mRNA level, MUC2 expression was significantly higher in both lamina propria and goblet cells at 28 days post-infection compared to the naïve control. MUC5B expression at the mRNA level was also higher in lamina propria at 28 dpi. Expression of MUC1, MUC4, MUC5AC, and MUC6 was extremely low or not detectable in goblet cells, columnar epithelial cells, and lamina propria from both naïve control and infected animals. Among the seven enzymes involved in post-translational O-linked glycosylation of mucins, GCNT3, which may represent one of the key rate-limiting steps in mucin biosynthesis, was up-regulated in goblet cells, columnar epithelial cells, lamina propria, and gross small intestine tissue during the course of infection. Western blot analysis revealed that MUC2 glycoprotein was strongly induced by infection in both gross small intestine tissue and its mucosal layer. In contrast, the higher MUC5B protein expression was observed only in the mucosal layer. Immunohistochemistry provided further evidence of the mucin glycoprotein production and localization. Our results provided insight into regulation of mucin biosynthesis in various cell types in the bovine small intestine during gastrointestinal nematode infection and will facilitate our understanding of mucins and their role in immune response against parasitic nematodes.  相似文献

Few studies exist regarding the distribution of intestinal mucins in fetuses of mammalians such as cattle and sheep. In this study, we aimed to describe the changes in the mucin production by ileal epithelium of bovine fetuses during their prenatal development. The goblet cells showed heterogeneity in mucins and the apical cytoplasm of the enterocytes demonstrated Periodic acid Schiff‐positive reaction which declined gradually towards the birth. Moreover, the number of the goblet cells containing acidic and mixed mucins augmented, whereas those containing neutral mucins decreased with advancing gestational age. After sixth month of gestation, with the initiation of the ileal Peyer patches and follicle‐associated epithelium development, a gradual increase in the number of goblet cells containing sulfomucins was also noticed towards the birth. The presence of different mucins in the ileum of bovine fetuses throughout prenatal development might play a role in the protection of the intestinal mucosa against urinary waste products in swallowed amniotic fluid and bile. Furthermore, mucins can also contribute for the formation of meconium in intra‐uterine life and building of strong intestinal barrier with predominating sulfomucins, protecting the intestine against potential pathogens and digestive enzymes after birth.  相似文献

This study was conducted with the aim of determining the morphology, distribution and heterogeneity of mast cells in the tongues of seagull (Larus fuscus), common buzzard (Buteo buteo), goose (Anser anser), white stork (Ciconia ciconia) and Gerze rooster. The study used five samples of tongue material from each of the healthy adult avian species. The samples were fixed in 10% neutral‐buffered formalin (NBF) solution, then, after routine tissue follow‐up, the samples blocked with paraplast. Cross‐sections with 5–6 μm of thickness were stained with the 0.5% toluidine blue and alcian blue/safranin O (AB/SO). In all five avian species, it was found that the mast cells were in different sizes and round, oval or spindle‐shaped based on their place of distribution. Mast cell numbers were determined in stained with toluidine blue, examined ×40 objectives in a 1 mm2 area. It was observed that mast cell density in subepithelial lamina propria and microscopic papilla was higher in the tongues of all species. Mast cell distribution and heterogeneity varied through the tongue, and there were more mast cells in the dorsal side of the tongue than the ventral side. The highest amount of mast cells was found in the tongue of the Gerze rooster among all five species. In the tongue cross‐sections stained with the combined method of alcian blue/safranin O (AB/SO), the mast cells were stained as AB (+), SO (+) and AB/SO (+) (mixed).  相似文献

The morphology of the intestine has been studied in a species of warm water fish, Tilapia spp., a hybrid teleost of notable economic importance. Light and electron microscope results show that the intestine is a relatively undifferentiated muscular tube lined with a simple columnar epithelium interspersed with goblet cells. The proximal region has a greater surface area, manifested by elongated mucosal ridges. The enterocytes are covered apically with uniform microvilli and exhibit the typical ultrastructural features of pinocytosis, namely extensive invaginations of the luminal plasma membrane and massive accumulation of vesicles in the apical cytoplasm. The distal intestine mucosa is thinner and less elaborately folded and consists of columnar cells with shorter and sparser microvilli. Their supranuclear cytoplasm contains abundant clear vacuoles. Numerous endocrine cells can also be seen. Regional cellular ultrastructural features are correlated with digestive functions.  相似文献

Mucins are related to infectious and non-infectious diseases in Veterinary and Human Medicine. MUC1 mucin is a transmembrane glycoprotein expressed on the apical surface of human epithelia while MUC5AC is the predominant secreted mucin expressed in human gastric epithelium and goblet cells of lung and eyes. MUC5AC C-terminus cysteine rich regions and the cytoplasmic tail of MUC1 domains are conserved among several mammalian species. Objective: to compare the expression of MUC1 and MUC5AC mucins in mammalian epithelia. CT33 anti-MUC1 cytoplasmic tail (MUC1CT) polyclonal antibody and 45M1 anti-MUC5AC monoclonal antibody were employed. By immunohistochemistry, MUC1CT was expressed in most tissues while MUC5AC was restricted to gastric surface epithelium and goblet cells from trachea and lung. By western blot, MUC1CT showed a band at approximately 35 kDa in most tissues; MUC5AC revealed bands at >180 kDa in stomach and lung secretions from rat, cat, pig and cow. When rat MUC5AC was immunoprecipitated, a band at about 180 kDa was obtained.  相似文献

1. The effect of vitamin A on the small intestine was examined in vitamin-A-deficient meat-type chickens. 2. Maturation and activity of the small intestinal cells were assayed by detection of proliferating cells with proliferating cells nuclear antigen, goblet cells with Alcian blue, mature cells with alkaline phosphatase and extent of RNA expression with dot blot analysis. 3. Vitamin A deficiency caused hyperproliferation of enterocytes, a decrease in the number of goblet cells, decreased alkaline phosphatase activity and decreased expression of 2 brush-border enzymes. 4. Our findings suggest that the absence of vitamin A interferes with the normal growth rate in chickens because it influences functionality of the small intestine by altering proliferation and maturation of cells in the small intestinal mucosa.  相似文献

The histology and characteristics of mucins secreted by epithelial mucous cells of the digestive tract in yellow catfish, Pelteobagrus fulvidraco were investigated using light microscope and transmission electron microscope. The digestive tract was divided into a pharynx, oesophagus, U-shaped stomach (with a cardiac, fundic and pyloric part) and intestine, composed of anterior intestine, middle intestine and posterior intestine, which consisted of a mucosa (epithelial layer), lamina propria-submucosa, muscularis and serosa. A large number of isolated longitudinal striated muscular bundles were present in the lamina propria-submucosa of pharynx. Goblet cells were observed throughout the digestive tract, except in the stomach. In the cardiac and fundic stomach, a plenty of gastric glands were observed, whereas they were absent in the pyloric part. Numerous mitochondria and endoplasmic reticulum were observed in the columnar epithelial cells of the intestine, especially of the anterior part. The epithelial mucous cells contained neutral or other two mixtures of acid and neutral mucins, the first being the most common. The neutral mucin was the only type of mucins in the stomach, anterior intestine and middle intestine. The results of this study will be helpful for understanding the digestive physiology and diagnosing some gastrointestinal diseases in yellow catfish.  相似文献

通过组织学观察,探讨野生黄鼬消化管的组织结构特点.将8只野生黄鼬经乙醚麻醉后处死,解剖取食管、胃、小肠、大肠,制作石蜡切片,观察其组织结构.结果显示野生黄鼬食管的黏膜为复层扁平上皮,食管腺发达,肌层以骨骼肌为主.胃贲门部有发达的皱襞和贲门腺;胃底腺有大量的主细胞和壁细胞;胃大弯部的腺体以壁细胞为主,仅有少量主细胞;胃幽门部有发达的幽门腺和大量壁细胞.十二指肠黏膜层有小肠腺,内有潘氏细胞存在,黏膜下层含有十二指肠腺;空肠可见孤立淋巴小结、弥散淋巴组织及集合淋巴小结.结肠无皱襞和肠绒毛,大肠腺排列紧密,其中杯状细胞特别多;直肠固有膜内有发达的大肠腺.所以野生黄鼬消化管的特点是胃各部胃腺发达,壁细胞特别多.  相似文献

Morphofunctional characteristics of the large intestine are rarely explored to understand the physiology, behavior and ecology of neotropical primates. In this study, we analyzed the histometric parameters of the large intestine of hybrid marmosets (Callithrix sp.) captured in forest fragments of Viçosa-Brazil, under seasonal interference. These animals were predominantly insectivorous in the rainy season and gummivores in the dry season. Large intestine fragments were collected and processed according to histological methods and stained with toluidine blue for general analysis, periodic acid of Schiff (PAS) and Alcian blue (AB) for goblet cells, Grimelius and Masson Fontana for argyrophil and argentaffin endocrine cells. Several histometric parameters were more expressive in the large intestine of the rainy season marmosets: greater thickness of the parietal layers, greater number of argyrophil and argentaffin endocrine cells, and AB-positive goblet cells, characteristics favor secretomotor functions and reduce the passage time of the fecal bolus, which is consistent with an insectivorous diet. In contrast, parameters such as crypt width, height of the absorptive cells and striated border, and the number of PAS-positive cells were more expressive in the dry season marmosets, reflecting the need for longer passage time for digestion and absorption of food items from tree gum, which are more complex and demand the action of microorganisms present in the large intestine, as well as greater protection against the abrasive action of dietary fibers and against microorganisms. Thus, it can be said that the marmoset's large intestine has morphological adaptations to maximize energy intake from the diet, which alternates under the influence of seasonality.  相似文献

采用光镜和扫描电镜技术 ,对泰山螭霖鱼 (Varicorhinusmacrolepis)的肠道进行了观察。结果表明 :泰山螭霖鱼无胃 ,食管之后是肠道 ,起始端膨大呈球状。肠道由前肠、中肠和后肠组成 ,肠管直径由前肠到后肠逐渐变小。各段肠壁均分为粘膜、肌层和浆膜 3层。粘膜上皮由柱状细胞和杯状细胞组成 ,肌层分内环行和外纵行 2层。粘膜向肠腔内突出形成许多粘膜褶 ,有的呈指状、杵状 ,有的有分支。由前肠到后肠 ,粘膜褶由高变低 ,数量逐渐减少 ;杯状细胞数目由多变少 ;肌层逐渐变薄。扫描电镜下 ,肠道的粘膜褶大体上呈纵向锯齿状 ,并且粘膜褶上还有次级皱褶。柱状上皮细胞表面多呈圆形 ,前肠、中肠柱状上皮细胞轮廓和界限清楚 ,常呈隆起状 ,而后肠上皮细胞表面较平坦。前肠柱状上皮细胞游离面的微绒毛长而密 ,后肠的短而稀疏。前肠的杯状细胞常常有较大的分泌孔 ,周围有分泌物 ,粘膜表面有粗大的分泌颗粒 ;后肠杯状细胞的分泌孔较小 ,粘膜表面有较多细小分泌颗粒。  相似文献
