The objective of this work was to study the relationship between female size (weight) and variables of egg and larval stage of Persian sturgeon Acipenser persicus . In this study, 19 female breeders were captured in Caspian Sea and fertilized by routine methods. Positive significant correlations ( P <0.05) were established between female weight and ovulated eggs per female, time of second mitosis division and volume of yolk-sac at hatching. There was not significant correlation ( r =0.33, P =0.161) between female weight and egg diameter. Female weight was not affected weight of larvae at hatching time ( r =0.37, P =0.119), as well as larval length ( r =−0.14, P =0.558) and larval weight at the end of the experiment (48 hours after first feeding) ( r =0.16, P =0.491). Mortality rate during yolk-sac absorption was higher with increased female weight but their correlation was not significant ( r =0.40, P =0.076). During the first feeding stage, mortality rate was 13.39% and there was no significant correlation between mortality rate in this period and female weight ( r =−0.12, P =0.613). Conclusively, as a result female size influenced fecundity, time of second mitosis division and yolk-sac volume at hatching time without affecting mortality rate during yolk-sac absorption and first feeding stage in Persian sturgeon. Thus, smaller female broods do not cause more mortality than larger ones in larval production and they can be used in reproduction procedure.  相似文献

A 3 × 4 factorial design was used to evaluate the dietary protein requirement and to determine the optimum dietary protein to energy (P/DE) ratio in sub‐yearling Persian sturgeon, Acipenser persicus, reared in the indoor system. Twelve experimental diets (40P16, 40P17, 40P18, 40P19, 45P19, 45P17, 45P18, 45P19, 50P16, 50P17, 50P18 and 50P19) were formulated and prepared to contain three protein levels (40%, 45% and 50%) and four digestible energy levels (16, 17, 18 and 19 kJ g?1 diet) at each protein level. Fish averaging 103.3 ± 3.5 (mean ± SD) were fed one of the experimental diets for 14 weeks. At the end of the experimental period, there were significant energy effects (P < 0.05) on weight gain (WG) and specific growth rate (SGR). Weight gain and SGR tended to decrease, although non‐significantly, with increase in dietary protein levels. Furthermore, there were significant protein and energy interaction effects on WG, SGR, hepatosomatic index and protein efficiency ratio. However, there were no significant dietary protein, energy or their interaction effects on feed efficiency for fish fed all diets. Weight gain and SGR of fish fed 40P19 were significantly higher than those of fish fed 40P16, 45P16, 45P17, 50P16 and 50P17 diets (P < 0.05). There were no significant differences in WG and SGR among fish fed 40P17, 40P18, 40P19, 45P18, 45P19, 50P18 and 50P19 diets. These results may indicate that the optimum dietary protein requirement and the P/DE ratio could be 40% protein and 22.0 mg protein kJ?1 (40P18), respectively, in Persian sturgeon, based on growth performance and feed utilization.  相似文献

In this study, metabolic responses of Persian sturgeon fingerlings to hyperosmotic condition were investigated by NMR‐based metabolomics. Persian sturgeon fingerlings (n = 2010, mean total weight: 3.2 ± 0.6 g; mean total length: 8.5 ± 1.5 cm) were held in freshwater (FW) for 96 hr and then acclimated in brackish water (BW) (12 g/L) for 24 hr. Blood samples were taken before and after salinity acclimation. The major metabolite changes corresponding to salinity acclimation were related to amino acids, osmolytes and energy metabolites. The plasma glucose levels increased significantly after 24 hr acclimation in BW (p < .05). Other energetic metabolites (acetate, acetoacetate, beta‐hydroxybutyrate, phosphocreatine, creatine, glycerol) showed no significant changes after 24 hr salinity challenge (p > .05). The osmolytes (taurine, trimethylamine‐N‐oxide, choline, N,N‐dimethylglycine) and amino acids (aspartate, glutamate, glutamine, leucine, phenylalanine, β‐alanine, histidine, threonine, cysteine) declined significantly after 24 hr acclimation in BW (p < .05). However, the levels of glycine elevated after 24 hr acclimation in BW (p < .05). Other amino acids including aspargine, isoluecine, Tyrosine, Tryptophan, Valine, and Lysine showed no significant changes after salinity challenge (p > .05). The considerable decreases in almost all plasma amino acids and all osmolytes showed specific importance of these metabolites in osmotic adaptation of fish in BW. Also, it seems that Persian sturgeon fingerlings rely more on oxidation of glucose to meet energetic requirements of osmoregulation than oxidation of lipids, as we observed no detectable changes in metabolites associated with lipid oxidation during osmotic acclimation in BW. Our results may help to understand osmoregulation in a chondrostei fish species from a metabolic approach.  相似文献

In this study, cryopreservation feasibility of Persian sturgeon (Acipenser persicus) and the effect of different doses of 2‐hydroxypropyl‐beta‐cyclodextrin on thawed spermatozoa quality (motility duration and motility percentage) were investigated. For freezing, semen of seven male individuals was pooled in equal volumes and diluted with 4°C [Tris‐HCl (100 mM), pH = 8, DMSO 10%] extenders containing 0, 5, 10, 15 mM of HβCD in a ratio of 1:1(semen/extenders). Then semen was filled into 0.5‐mL straws, and was frozen with vapour of liquid nitrogen at 4‐cm above surface of liquid nitrogen. After 3 min, straws were plunged in to liquid nitrogen. Thawing was performed at 40°C water baths for 15 s. Motility duration of the 10 mM HβCD treated spermatozoa at days 14 (228.98 ± 16.39) and 56 (199.66 ±21.78) were longer than other treatments. In day 56, the motility percentage in treatment with 10 mM was significantly higher (16.14 ± 2.54) (P < 0.05) compared with 5 mM treatment (8.75 ± 2.47) (P < 0.05). Therefore, it is recommended that 10 mM of HβCD can be used as an additive cryoprotectant for increasing cryopreserved spermatozoa quality in this species.  相似文献

In this study, the efficiency of a novel droplet vitrification technique along with different doses of fish antifreeze protein (AFP) type III on Persian sturgeon thawed spermatozoa quality (motility duration and motility percentage) was investigated. Semen of seven male individuals was pooled in equal volumes and diluted with 4°C Tris‐Hcl (100 mM), pH = 8 extenders containing 0, 5, 10, 15 μM of AFP type III in a ratio of 1:1 (semen/extenders). Treated semen was dropped into liquid nitrogen. Solidified droplets were stored for 2, 60 and 120 days and thawed by plunging them into a tube containing 5 mL Tris‐Hcl (100 mM), pH=8 with 1% BSA at 37°C. Motility duration in all treatments had no significant difference comparing to fresh sperm (P > 0.05), but their motility percentage was significantly lower. Treatment with 10 μM of AFP had significantly higher motility percentage (16.11 ± 0.5%) comparing to other treatments (P < 0.05). There was no significant difference between 0, 5, 15 μM of antifreeze protein treatments (P > 0.05), suggesting that antifreeze protein effectiveness are highly dose dependent, and dose of 10 μM is appropriate in Persian sturgeon spermatozoa droplet vitrification. Besides, the present technique obtained higher quality of spermatozoa comparing to its analogue techniques.  相似文献

The effect of fish protein hydrolysate (FPH) on the growth, feed utilization, digestive enzymes, gut microflora and resistance to bacterial challenge in Persian sturgeon (Acipenser persicus) larvae during a 45‐day feeding trial has been investigated. Artificial diets containing different levels of tuna visceral protein hydrolysate (PH0, PH10, PH25 and PH50) and live foods (Artemia: 5 days, Daphnia: 40 days) were fed to Persian sturgeon larvae. Larvae fed on the PH10 and PH25 had growth, feed utilization, protein content and trypsin activity significantly better than PH0 and PH50 groups (< 0.05). Condition factors, survival rate and pepsin activity were not significantly different (> 0.05). Brush border membrane enzymes to cytosolic enzyme ratio revealed that maturation had been occurred in 41 days post hatch (dph) at moderate FPH levels. In 54 dph larvae, this ratio was not affected by the level of FPH in the diet. No significant differences were observed in the total number of gut microflora or in the resistance to Aeromonas hydrophila (> 0.05), whereas lactic acid bacteria was significantly higher in PH50 (< 0.05). The results of this study show that application of moderate levels of FPH in Persian sturgeon diets could enhance larvae performance due to the early maturation in intestinal.  相似文献

This experiment was conducted to illustrate the effect of dietary taurine levels on Persian sturgeon, Acipenser persicus (mean weight of ~35 g) fed plant‐based diet. A basal diet containing low level of fishmeal (19%) was formulated containing 0.05% endogenous taurine (control diet). To this diet were added five levels of exogenous taurine: 0.25 (0.25T), 0.5 (0.5T), 0.8 (0.8T), 1.2 (1.2T) and 1.6% (1.6T). These diets were fed to the experimental fish for 42 days. Results showed that growth performance decreased and food conversion ratio increased as dietary taurine increased, although no significant difference was observed between the control and 0.25T treatments. Increased dietary taurine led to increased carcass moisture and decreased carcass lipid percentages. Feed intake was significantly and negatively correlated with dietary taurine. Carcass taurine content significantly increased with dietary taurine elevation up to 1.2% and then decreased. Hepatic taurine content showed a significant increase in 0.25T treatment and then decreased along with dietary taurine elevation and reached the control values at 1.6% taurine level. Dietary taurine elevation led to more hepatic damages compared to the control. In conclusion, although within the range reported for other studied fish, the tested taurine levels in this study might be supra‐optimal causing adverse effects in Persian sturgeon and further study with lower taurine levels is encouraged.  相似文献

The physiological ionoregulatory, metabolic and immune responses of Persian sturgeon, Acipenser persicus, to acute stress were investigated. Water levels were lowered to the fish dorsal scutes, and fish were blood‐sampled before stress (pre‐stress), and 0, 6, 24 and 72 h after stress. Results showed that serum cortisol rapidly increased after stress, returning to initial levels at 24 h. Serum glucose significantly increased at 6 h, declining to the pre‐stress levels at 72 h. Serum triglyceride and cholesterol showed significant decreases at 0 h, then increasing to higher than the initial levels at 72 h. Serum T3 and T4 significantly decreased at 0 h and recovered at 72 h. Serum chloride levels showed no significant changes while serum calcium showed a significant increase at 0 h and a further increase until 72 h poststress. Serum total protein and alternative complement activity showed a significant initial decrease and recovery at 24 h with further increase at 72 h. Serum lysozyme activity increased significantly at 24 and 72 h after stress. Serum total immunoglobulin significantly increased at 0 h and peaked at 24 h. This is the first work showing thyroid hormone and immunological responses in Persian sturgeon subjected to stress, and the results show that this species follows a classical hormonal and energetic stress response, although, in the low range among the sturgeons. Although transient and moderate, confinement stress can induce significant changes in the innate immune response.  相似文献

The effects of five different light regimes on growth performance and behaviour of Persian Sturgeon larvae were examined. Larvae were reared under five different photoperiods (24L, 12L:12D, 16L:8D, 8L:16L and 24D), from hatching (25.8 ± 8.1 mg; 12 ± 0.6 mm) to 28 days post‐hatch (dph). Except the last week, there was no significant difference in growth parameters among the experimental groups. Growth performance significantly improved in larvae reared under a continuous light regime. The highest final length (34.5 ± 1.7 mm) was measured in 24L and the lowest one (29.5 ± 0.4 mm) in the constant darkness. Relative to other experimental groups, the continuous darkness had a retarding impact on the yolk sac absorption and swarming behaviour. Among the whole body compositions, the lowest body moisture content was measured in the continuous light group. Persian Sturgeon pre‐larvae and larvae were not completely dependent on light regimes (in 1–21 days), whereas on the basis of measured parameters (total length, wet and dry weight and digestive fullness index) a continuous light regime played a decisive role on growth performance beyond 21 dph.  相似文献

A minimally invasive surgical technique (MIST) for the removal of ovulated eggs from Persian sturgeon Acipenser persicus was tested on broodstocks caught from the wild to determine whether it affected fecundity or hatching rates compared with the traditional stripping method of killing and removing the eggs (commonly used in hatchery). Morphological parameters of females, germinal vesicle (GV) position, weight of obtained egg, number of eggs/gram, fertilization rate, and percentage of hatching during incubation were not significantly different between the MIST and traditional stripping methods. Obtained ova were 4.8 ± 0.4 kg female−1 in the MIST and 4.6 ± 0.5 kg female−1 in traditional stripped groups, respectively; the fertilization rate was 83.4 ± 11.2% and 80.0 ± 7.2% in groups, respectively. The results of this study showed that the minimally invasive surgical technique approach is efficient, practical, and less stressful to Persian sturgeon broodstocks during artificial propagation than other reported egg collection procedures.  相似文献

The aims of this study was to assess the effect of two lactic acid bacteria (LAB), Lactobacillus curvatus and Leuconostoc mesenteroides, originally isolated from gastrointestinal (GI) tract of beluga (Huso huso) and Persian sturgeon (Acipenser persicus), respectively, on growth, survival and digestive enzyme (amylase, lipase and protease) activities and the population level of LAB in the GI tract. The treatments included 10 different groups; control, separate supplements of Lcurvatus and Leumesenteroides at three different counts [2 × 109, 5 × 109 and 9 × 109 colony forming units (CFU) per gram food] and three combinations of the two LAB (2 × 109 + 2 × 109, 5 × 109 + 5 × 109 and 9 × 109 + 9 × 109 CFU per gram food). The bacteria used in this study were added in lyophilized form to chopped Chironomidae. In the beluga study, highest specific growth rate, survival and improved intestinal enzyme activities were noted in the rearing group fed 9 × 109 L. curvatus per gram food. In Persian sturgeon, the inclusion level of 2 × 109 Leu. mesenteroides had similar positive effect. The ability of LAB to colonize the digestive tract seems to involve host specificity, and our bacteriological results are relevant to initiate future probiotic studies in sturgeons and future directions will be discussed.  相似文献

The specific activity of alkaline protease, trypsin and pepsin‐like enzymes was measured in yolk sac stage Acipenser persicus larvae and over a 1‐month feeding experiment using live Artemia nauplii (ND), formulated feed (FD) or mixed food (MD). Artemia nauplii group larvae showed significantly higher growth and survival during the first 15 days while FD larvae showed the lowest growth and survival. At day 30, MD larvae exhibited significantly higher growth than the ND group. Alkaline protease activity showed the lowest activity on day 15; the highest activity was observed in the MD group larvae. Pepsin‐like activity showed a drastic increase from day 1 to 5 in all treatments, but remained stable throughout the next 25 days, with the lowest and the highest activity in the FD treatment on day 10 and in the MD treatment on day 30 post‐feeding respectively. Trypsin‐like activity in group ND remained almost the same from day 5 to 30, whereas in groups MD and FD, it decreased significantly from day 10 to 30. The contribution of the naupliar proteases was moderate but effective. Additionally, better performance in Artemia fed sturgeon larvae may also be due to the structure and digestibility of proteins and the food intake stimulation by the nauplii.  相似文献

This study aimed to evaluate the effects of enriched Artemia by fish and soybean oils supplemented with vitamin E on growth performance, lipid peroxidation, lipase activity and fatty acid composition of Persian sturgeon (Acipenser persicus) larvae. For this purpose, five experimental diets including non‐enriched Artemia (control diet), Artemia enriched with soybean oil supplemented with 15% and 30% vitamin E (S15 and S30 diets) and fish oil supplemented with 15% and 30% vitamin E (F15 and F30 diets) were used. The larvae were fed to apparent satiation four times per day for 22 days. The results indicated that fish fed enriched Artemia had no significant differences compared with those fed non‐enriched Artemia in terms of growth and survival, but increase in vitamin E levels from 15 to 30% improved growth performance of larvae. Vitamin E content in fish fed S15 and S30 diets was significantly higher. Fish fed non‐enriched Artemia had significantly higher thiobarbituric acid and lower lipase activity. The highest HUFA and n‐3/n‐6 ratio were observed in fish fed F15 and F30 diets. Our results demonstrated that fish oil can completely replace with soybean oil in larval diets. Therefore, using S30 diet is recommended for feeding of Persian sturgeon larvae.  相似文献

Persian sturgeon eggs were fertilized with different levels of salinities (0.5, 2, 4, 6, 8, 10, 12?ppt), and then each group was incubated in the same salinity until hatch. The fertility (%), hatching rate as well as larvae cumulative mortality rate and abnormality (%) were measured. Our Results revealed that Persian surgeon eggs could be fertilized in the different salinity concentrations but not more than 4?ppt. Moreover, hatching rate decreased with increase in salinities more than 2 and 4?ppt, respectively, and no larvae hatched in 6?ppt salinity. According to these results, the salinity tolerance threshold for Persian sturgeon larvae hatching in brackish water is less than 4?ppt.  相似文献

Three experiments were performed to develop protocols for cryopreservation of Persian sturgeon Acipenser persicus, sperm. In the first experiment, sperm from six males was individually split in three subsamples and cryopreserved using Modified Tsvetkova's extender (mT) supplemented with dimethyl sulfoxide (DMSO), methanol (MeOH), glycerol (Gly) and ethylene glycol (EG) at concentration of 5%, 10%, 15% and 20%. In the second set of experiments, the effects of six equilibration times (0, 5, 10, 20, 40 and 60 min) and dilution ratios (volume sperm: volume extender 1:0.5, 1:1, 1:2, 1:3, 1:5 and 1:10) and the additive advantage of bovine serum albumin (BSA; 0, 2.5, 5 and 10 mg mL?1) and ascorbic acid (0, 2.5, 5 and 10 U mL?1), on the post‐thaw survival of sperm (triplicate set of six fish) were evaluated. Then, sperm was diluted in 1:1 mT extender with 10 mg mL?1 BSA with selected cryoprotectants (15% MeOH and 10% DMSO) for 5 min. After a month of storage in liquid nitrogen, post‐thawed sperm motility; fertilization and hatching rate and viability of derived larvae were measured (Exp.3). Evaluation of cryoprotectants efficiency showed that MeOH 15% and DMSO 10% were suitable for cryopreservation of Persian sturgeon sperm. Gly and EG resulted in very low post‐thaw motility rates even at lowest concentration. No significant difference was observed among the four different equilibration times (0, 5, 10, 20 min) (P > 0.05) although higher equilibration times than 20 min resulted low post‐thaw motility (P < 0.05). The motility of frozen–thawed sperm did not significantly change when dilution ratio was increased from 1:0.5 to 1:3 (P > 0.05). However, higher dilution ratios (1:5 and 1:10) reduced the percentage of motile sperm. Supplementation of the cryoprotectant solution with 10 mg mL?1 BSA significantly improved post‐thaw motility (P < 0.05), but ascorbic acid did not improve post‐thaw motility (P > 0.05). The results of experiment 3 showed that the highest fertilization (30.2 ± 5.75) and hatching rates (28.2 ± 5.25) were observed when samples were frozen with 15% MeOH (P > 0.05). Our study indicates that the use of mT extender consisting of 10 mg mL?1 BSA in 15% MeOH diluted with sperm at 1:1 ratio for 5 min can be recommended cryopreservation method for Persian sturgeon sperm.  相似文献

Artificial rearing of fry has been introduced as an alternative to supply fry and juvenile sturgeon in order to restock the depleted sturgeon population in the Caspian Sea. The aim of this study is to assess the feed selection characteristics of sturgeon (Acipenser persicus) fry during the period in which they stay in earthen ponds before being released into the river. Sturgeon fry weighing around 150 mg were introduced into five almost similar‐sized earthen ponds. The ponds were enriched with fertilizer to supply natural food for the fries. A weekly sample of six specimens was collected from each pond during the 5 weeks of the experiment. All collected fish were weighed and then dissected for inspection of the stomach and the intestine content. The overall stomach content measured from 150 fish showed that around 74% of the stomach content was composed of chironomids and the rest was daphnia and cyclops. At the beginning of the study (first 2 weeks), sturgeon fry feed more on daphnia and cyclops; this trend, however, reversed in the last 2 weeks of the study. In conclusion, sturgeon fry feed on three prey groups—chironomids, daphnia and cyclops—during their lifespan in earthen pond. Feeding on chironomids larvae increased with sturgeon fry size.  相似文献

To compare the reproductive performance of spring and autumn forms of Persian sturgeon males, Acipenser persicus, the relative abundance of spermiating and non‐spermiating males, milt quality, sex steroids and cortisol were investigated after pituitary preparation (PP) treatment and at the time of spermiation. Our results showed that higher numbers of spring males are spermiated after PP treatment than autumn individuals. The autumn spermiating males had higher and lower values of sperm density and duration of motility than spring males respectively. The milt volume was higher in spring males than in autumn individuals. The percentage of motility was statistically equal in both the groups examined. In both forms of brooders, the serum levels of testosterone (T), 11‐ketotestosterone (11‐KT), progesterone (P), 17α, 20β, 21‐trihydroxy‐4‐pregnen‐3‐one (20βS) and cortisol (C) increased after PP treatment, although such increases did not occur in non‐spermiating males. Our results showed the probable involvement of 20βs, P, T, 11‐KT and C in the final maturation of Persian sturgeon. In addition, it was demonstrated that the spring form of Persian sturgeon shows better reproductive performance than the autumn form in terms of a response to artificial induction and milt quality.  相似文献

The motility and fertilizing ability of the Persian sturgeon, Acipenser persicus, spermatozoa were investigated. Optimum ionic content (Na+, K+, Ca2+ and Mg2+) and pH of activation solution as well as the optimum dilution rate were determined. The results show optimum motility characteristics of spermatozoa in buffered solutions containing 25, 0.2, 3 and 10 mM L?1 Na+, K+, Ca2+ and Mg2+, respectively, at dilution rate 1:50 and pH 8.0. To test the fertilizing ability of sperm, two buffered saline solutions were used as activation solution of sperm motility. The present study indicated (1) spermatozoa motility is one of key factors that influence on fertilizing ability of sperm, (2) a high fertilizing ability of sperm is obtained after dilution in saline solutions rather than in freshwater and (3) a maximum fertilization rate occurs in buffered saline solution containing 0.2 mM L?1 K+. There is also a good correlation between biochemical characteristics of seminal plasma and fertilizing ability of sperm.  相似文献

Four one‐week growth trials were conducted on green sturgeon fry to determine the effect of feeding rate on their growth performance at 18 °C when they were fed a salmonid soft moist feeds containing 445–457 g kg?1 of crude protein and 201–207 g kg?1 of lipid. The fry used in Trials I‐IV were 5–8 weeks after their initiation of exogenous feeding. Their average initial body weights were 1.63 ± 0.01, 2.63 ± 0.03, 5.08 ± 0.08 and 7.49 ± 0.05 g, respectively. Six feeding rates used were as follows: 2.5–15.0% body weight per day (% BW day?1) with a 2.5% increment in Trial I; 1.25–7.50% BW day?1 with a 1.25% increment in Trial II; and 2.0–7.0% BW day?1 with a 1.0% increment in Trials III and IV. Four replicates with 50 fry per tank in Trials I‐III and 30 fry per tank in Trial IV were assigned randomly to each feeding rates. The final body weight, specific growth rate, feed efficiency, protein retention, and whole‐body moisture, lipid, and energy contents were significantly (< 0.05) affected by the feeding rates. The optimum feeding rates determined by the broken‐line model were 7.1, 5.7 and 5.3% BW day?1 for Trials I, II and IV, when the fry were 5, 6 and 8 weeks after their initiation of exogenous feeding, respectively.  相似文献
