Foal first diarrhoea is one of the most prominent problems in the early life of horses. Probiotics might have the potency to prevent or at least diminish neonatal diarrhoea. We hypothesised that the treatment of foals with probiotic strains of Lactobacillus rhamnosus and Enterococcus faecium starting early after birth and then daily over 2 weeks would prevent or mitigate foal heat diarrhoea. The influence of this probiotic treatment on diarrhoea incidence and growth and health performance of young foals was investigated. Thirty‐four foals were randomly allocated to two groups. From day 1 to 14 of life, the foals received either placebo (PG, n = 16) or the probiotic treatment (TG, n = 18). Clinical examination was performed, and the faeces consistency score (FCS, 1–5; with diarrhoea defined by ≤3) was recorded once per day in weeks 1 and 2 and once weekly in weeks 3–8 of life (WL). The body height was measured at birth and after two and eight WL. Diarrhoea occurred in the 1st WL in 19% and 61% of PG and TG foals respectively. In the 1st WL, diarrhoea lasted 0.3 ± 0.8 and 1.6 ± 1.4 days in PG and TG foals respectively. In the 2nd WL, diarrhoea occurred in 94% and 84% of PG and TG foals, respectively, and lasted for 3.0 ± 1.5 and 3.7 ± 1.6 days respectively. At least two periods of diarrhoea developed in 33% and 65% of PG and TG foals respectively. The TG foals grew slightly slower than the PG foals. The results indicated that the probiotic treatment of neonatal foals as performed in this study was not suitable to reduce diarrhoea within the first two WL, because contrary to the hypothesis, the TG foals suffered more frequently and for longer periods from diarrhoea than the PG foals.  相似文献

Lactic acid bacteria play an essential role in the food industry in the manufacture of many fermented products (cheese, yogurt, fermented vegetables, etc.). Application of these organisms is now being extended to the area of health improvement, as their probiotic activities become known. Probiotics are defined as viable microorganisms that exert a beneficial effect on the health of the host when they are ingested in sufficient quantity. Lactic acid bacteria and bifidobacteria isolated from the human intestine are the most common probiotics used for human consumption. The development of new probiotics with new beneficial effects is eagerly awaited in the food industry. This review introduces Lactococcus, which are one of the genera of lactic acid bacteria and are mainly isolated from dairy products and fermented vegetables, as new probiotics, focusing especially on Lactococcus lactis H61, which improves skin status in Japanese women with oral intake of heat‐killed or live cells. The deduced mechanisms associated with the beneficial effects of strain H61 are also discussed.  相似文献

The effects of the administration of four Lactobacillus strains as probiotics on the constitution of microbial populations in the intestine of pigs were investigated by using a litter of pigs. The experimental group was fed a diet blended with the freeze‐dried powder containing lactobacilli at a concentration of 0.2% (W/W). After 4 weeks of probiotics administration, the total viable count of anaerobes in the feces was not significantly different between the pigs fed a diet supplemented with probiotics and the pigs given the non‐supplemented diet. However, viable counts of eubacteria were significantly higher (P < 0.01) in the feces of pigs fed a diet supplemented with probiotics than in the pigs given the non‐supplemented diet. Conversely, viable counts of clostridia were significantly lower (P < 0.01) in the feces of pigs fed a diet supplemented with probiotics than the pigs given the non‐supplemented diet. The total volatile fatty acid concentrations in the feces of pigs that were given a diet supplemented with probiotics were significantly lower (P < 0.05) than those given the non‐supplemented diet. Further, the molar proportion of n‐butyric acids in the feces of pigs that were given a diet supplemented with probiotics, was significantly lower (P < 0.05) than those given the non‐supplemented diet.  相似文献

This study investigated the in vitro effect of Lactobacillus strains, a major group of probiotic lactic acid bacteria, on immunoglobulin E (IgE)‐ and antigen‐induced mast cell degranulation and subsequent gene expression. Bone marrow‐derived mast cells (BMMCs) from DBA/2 mice were cultured with heat‐killed Lactobacillus strains for 24 h. Some strains significantly inhibited IgE‐ and antigen‐induced β‐hexosaminidase release from BMMCs. Furthermore, Lactobacillus reuteri NBRC 15892, which exhibited the strongest inhibitory activity, significantly reduced the elevated interleukin (IL)‐4, IL‐13, tumor necrosis factor‐α, and cyclooxygenase‐2 expression levels that was induced by 1–2 h of stimulation with IgE and antigens. The suppressive effect of NBRC 15892 strain on BMMC degranulation was significantly reduced in the presence of a toll‐like receptor (TLR)2‐neutralizing antibody. In addition, downregulation of cell surface FcεRIα expression was observed after 6 h of NBRC 15892 treatment. These results suggest that some Lactobacillus strains inhibited IgE‐mediated mast cell degranulation and subsequent late‐phase reactions involving mast cells via a TLR2‐dependent mechanism with FcεRIα downregulation.  相似文献

Early weaning induces villous atrophy in the small intestine (SI) of piglets. Oral administration of live lactic acid bacteria (LAB) can improve villous shortening. In this study, we evaluated the oral administration of a heat‐killed and dried cell preparation of Enterococcus faecalis (a LAB) strain EC‐12 against villous atrophy in early‐weaned mice (Experiment 1) and pigs (Experiments 2 and 3). Twelve 16‐days‐old mice were divided into two groups in Experiment 1: gavage of EC‐12 (10 mg/kg body weight (BW)/day), or control. On day 21, SI was collected. Eighteen 21‐day‐old pigs were divided into two groups in Experiment 2: gavage of EC‐12 (10 mg/kg BW/day), or control. After 10 days, the villous height of jejunum was measured. Six 21‐day‐old pigs were divided into two groups in Experiment 3: the basal diet supplemented with EC‐12 at 0.05%‐fed group, or the basal diet‐fed group. After 10 days, the villous height of jejunum was measured. The villous heights in SI were significantly higher by EC‐12 administration in all experiments. EC‐12 successfully improved the villous atrophy in the early‐weaned mice and pigs when EC‐12 was administered orally.  相似文献

Probiotics have gained considerable attention with respect to their beneficial effects on livestock performance and health. The most significant effects of probiotics on the gut microbiota and the host animals take place when they are included in diets during particularly stressful periods such as weaning and/or at the beginning of the lactation period. The probiotics Bacillus mesentericus strain TO‐A at 1 × 108 colony forming units (CFU)/g, Clostridium butyricum strain TO‐A at 1 × 108 CFU/g and Enterococcus faecalis strain T‐110 at 1 × 109 CFU/g were used. Litter weight at delivery and ratio of return to estrous improved significantly (17% and 24% improvement, respectively) by probiotic administration to sows (0.2% (w/w)). Furthermore, the feed intake of the probiotics‐administered sows was greater than that of the control sows during the late lactation period. Post‐weaning diarrheal incidence and growth performance was improved by probiotics administration to neonates (0.02% (w/w)), while the combined use of probiotics in sows and their neonates induced the enlargement of villous height and prevented muscle layer thinning in the small intestine of weaning piglets. The administration of probiotics of three species of live bacteria improved the porcine reproductive performance around stressful periods of sows (farrowing) and piglets (weaning). Levels of fecal or intestinal lactobacilli, Escherichia coli and Clostridium perfringens, and the prevalence of clostridial alpha toxin gene and heat‐stable toxin (ST) gene of enterotoxigenic E. coli (ETEC) were monitored in weaned piglets before (day 0) and during (days 7, 14, and 21) the administration of Lactobacillus plantarum strain Lq80. Lactobacilli were enumerated in a culture‐dependent method. The remainders were determined by quantitative real‐time PCR. In this quantitative real‐time PCR method, the detection limit was proved to be as low as 103 cells/g feces or intestinal contents. Number of lactobacilli increased from day 0 to day 7 (P < 0.05), to day 14 (P < 0.05), and to day 21 (P = 0.07) in the Lq80‐administered group. L. plantarum contributed to as low as 10% of the lactobacillal population in the Lq80‐administered group. The number of E. coli and C. perfringens, and the prevalence of alpha toxin gene in feces or intestinal contents of the Lq80‐administered group decreased, at least in the first week of the postweaning period. Oral administration of L. plantarum strain Lq80 can stimulate the growth of indigenous lactobacilli and decrease ST‐producing ETEC and C. perfringens in the intestine of postweaning piglets.  相似文献

随机选择24头出生时间相近的健康杜×长×大外三元仔猪,分为2个组,每组3个重复;每个重复4头。分别在仔猪刚出生、3日龄及5日龄灌服10%灭菌脱脂奶(对照组)或含有屎肠球菌活菌(5×10。~6×10。CFU/mL)的10%脱脂奶重悬液(处理组),每次2mL/头,仔猪25日龄断奶。记录哺乳期仔猪腹泻发生率。断奶当天屠宰仔猪,采集肠道内容物及黏膜样品,检测盲肠内容物主要菌群的数量、各肠段pH值和结肠内容物乳酸含量,用实时定量PCR检测空肠黏膜化学屏障相关基因的表达量。结果显示,屎肠球菌使仔猪盲肠的乳酸菌比率升高8.98%,致病性肠杆菌的数量和比例分别下降28.68%(P〈0.05)和13.01%,仔猪腹泻率也下降了43.21%,但不影响细菌总数和乳酸菌数量,也不影响肠道pH值、结肠乳酸含量以及空肠黏膜MUC2、pBD-1和PMAP-37mRNA的表达量,却显著下调LYSmRNA的表达。结果表明,口服屎肠球菌改善哺乳期仔猪肠道菌群组成,促进肠道健康,同时它对溶菌酶基因表达的抑制作用可能是维持乳猪肠道微生态稳定的重要因素之一。  相似文献

Effects of probiotics on the intestinal microbiota of foals are yet insufficiently studied. The aim of this study was to investigate whether supplementation of Lactobacillus rhamnosus (DSM 7133) and Enterococcus faecium (DSM 7134) influences the bacterial composition of the faecal microbiota of foals. A total of 34 newborn foals were randomly assigned to the placebo group (PG, n = 16) and the treatment group (TG, n = 18). From day 1 to day 14 of life, foals orally received 3 ml of either a probiotic preparation (1.05 × 109 CFU E. faecium and 4.50 × 108 CFU L. rhamnosus) or placebo (carrier) once a day. Faeces were collected directly from the rectum immediately after birth (meconium) and at day 14 and day 56 of life. Samples of 12 foals per group were selected for microbiological analysis. DNA was extracted and used for polymerase chain reaction–denaturing gradient gel electrophoresis (PCR‐DGGE) and quantitative PCR. No DNA or amplicons were obtained from meconium. There were no differences in richness of bands and Shannon index of diversity regarding the Clostridium cluster XIVa between groups. Cluster analysis and principal coordinate analysis of DGGE data showed a clear effect of age. Band‐based similarity of bacterial clusters (Dice coefficient) decreased from day 14 to day 56 of life (p < 0.001) in PG foals only resulting in lower similarity in PG versus TG foals when 2 month old (p < 0.01). Five of thirty re‐amplified bands were identified on species level. Others were assigned either to family (mainly Lachnospiraceae) or genus level (Akkermansia). The bands related to Akkermansia muciniphila or Akkermansia spp. appeared almost in all DGGE profiles. Two‐week supplementation of the probiotic preparation to foals had no significant impact on the composition of the faecal microbiota but it appears to have prevented the reduction of bacterial similarity between 2 and 8 weeks of age observed in not treated foals.  相似文献

Numerous experiments have demonstrated that feeding a lower protein diet decreases protein fermentation in the gastrointestinal tract (GIT) and reduces the incidence of post‐weaning diarrhea (PWD). However, there is a lack of holistic evidence underpinning the relationship between feeding a lower protein diet and PWD in relation to physiological responses and protein fermentation in the GIT. The scope of this article, therefore, will: (i) focus on the impact of dietary protein levels on selected indices of GIT health in weaned pigs without and with experimental infection with an enterotoxigenic strain of Escherichia coli; and (ii) attempt to conduct regression analysis to examine the relationships between dietary‐origin protein intake, nitrogen fermentation indices, fecal consistency and the incidence of PWD. We used datasets generated from a series of four intensive experiments in weaned pigs. The collective results derived from these datasets indicate that restriction of daily protein intake to less than 60 g through feeding a lower protein diet for as little as 7 days after weaning reduced the incidence of PWD commensurate with a reduction in protein fermentation indices.  相似文献

Lipoic acid (LA) is a naturally existing substance which widely distributed in the cellular membranes and cytosol of animal cells. Its intracellular functions include quenching of free radicals and repairing oxidized proteins. The purpose of this study was to evaluate the effects of LA on post‐weaning diarrhoea using a rat model. Sixty weaned rats were fed either a basal diet or a LA‐supplemented diet, or a zinc oxide (ZnO)‐supplemented diet as a positive control. Rats in the LA and ZnO groups had better performance and reduced incidence of diarrhoea (p < 0.05). Both LA and ZnO treatments enhanced intestinal homeostatic and architecture, significantly decreased urinary lactulose to mannitol ratios (p < 0.05) and increased the expression of the intestinal mucosal tight junction proteins occludin (OCLN) and zonula occludens protein‐1 (ZO‐1) (p < 0.05). LA significantly increased the activities of antioxidant enzymes, and reduced glutathione while decreasing the levels of oxidative glutathione and malondialdehyde in the intestinal mucosa (p < 0.05). Furthermore, an in vitro study indicated that supplementation with LA in IEC‐6 intestinal epithelial cells significantly enhanced the expression of OCLN and ZO‐1 under hydrogen peroxide‐induced oxidative stress. Collectively, these results suggest that LA relieves post‐weaning diarrhoea by reducing intestinal permeability and improving antioxidant indices.  相似文献

Heat stress (HS) disrupts redox balance and insulin‐related metabolism. Supplementation with supranutritional amounts of selenium (Se) may enhance glutathione peroxidase (GPX) activity and reduce oxidative stress, but may trigger insulin resistance. Therefore, the aim of this experiment was to investigate the effects of a short‐term high Se supplementation on physiology, oxidative stress and insulin‐related metabolism in heat‐stressed pigs. Twenty‐four gilts were fed either a control (0.20 ppm Se) or a high Se (1.0 ppm Se yeast, HiSe) diet for 2 weeks. Pigs were then housed in thermoneutral (20°C) or HS (35°C) conditions for 8 days. Blood samples were collected to study blood Se and oxidative stress markers. An oral glucose tolerance test (OGTT) was conducted on day 8 of thermal exposure. The HS conditions increased rectal temperature and respiration rate (both p < .001). The HiSe diet increased blood Se by 12% (p < .05) and ameliorated the increase in rectal temperature (p < .05). Heat stress increased oxidative stress as evidenced by a 48% increase in plasma advanced oxidized protein products (AOPPs; p < .05), which may be associated with the reductions in plasma biological antioxidant potential (BAP) and erythrocyte GPX activity (both p < .05). The HiSe diet did not alleviate the reduction in plasma BAP or increase in AOPPs observed during HS, although it tended to increase erythrocyte GPX activity by 13% (p = .068). Without affecting insulin, HS attenuated lipid mobilization, as evidenced by a lower fasting NEFA concentration (p < .05), which was not mitigated by the HiSe diet. The HiSe diet increased insulin AUC, suggesting it potentiated insulin resistance, although this only occurred under TN conditions (p = .066). In summary, HS induced oxidative stress and attenuated lipid mobilization in pigs. The short‐term supranutritional Se supplementation alleviated hyperthermia, but did not protect against oxidative stress in heat‐stressed pigs.  相似文献

This study was aimed at investigating the long‐term effects of provision of liquid milk replacer (MR) and solid starter diet (SD) during lactation on post‐weaning (PW) growth of pigs. In experiment 1, 33 cross‐bred litters were allotted to four treatments: no supplement (CON), MR ad libitum, SD ad libitum and 100 g SD/litter/day from lactation day 4 through weaning at day 21 during late fall. In experiment 2, 40 litters received MR or none in July. PW pigs received commercial diets to marketing. In experiment 1, weaning weight (WW), pre‐weaning average daily gain (ADG) and mortality (2.4%) were not influenced by creep‐feeding MR or SD. ADG was greater (P < 0.05) in the MR group versus CON during days 21–54, but did not differ across the treatments during days 54–162. In experiment 2, ADG during lactation and WW were greater in the MR group versus CON, with mortality lower in the former (5.6 vs. 10.3%). However, PW ADG to day 175 did not differ between the two groups. Results suggest that creep‐feeding MR or SD has no effect on PW growth. However, it remains possible that MR reduces PW mortality during the hot season.  相似文献

The influence of medium‐chain glycerides on performance and gastrointestinal well‐being in weaning piglets was assessed. First, caproic (C6), caprylic (C8) and capric (C10) acid activity against Escherichia coli was screened in vitro. Pig flora of the whole small intestine was used as inoculum. Seven in vitro incubations were done in duplicate at pH = 3 and 5: C10 (15 mM), C8 (12 mM), C6 (15, 12, 10 mM), a non‐incubated‐negative control and incubated negative control. Culture suspensions were plated on E. coli‐selective agar. Controls showed bacterial growth. C6 and C8 showed no growth at both pH‐values, where C10 showed growth at pH = 5. Secondly, an in vivo study was done with 80 weaned piglets over 42 days, housed in pens of eight animals (five pens/treatment), fed a basal diet containing broken rice/soya bean meal/fish meal and supplemented with C6 and C8 in medium‐chain glyceride form (MCT6/8, 0.175%) or antibiotic growth promoter (AGP, 0.020%) (Kasetsart University, Thailand) serving as control. Feed intake, daily gain and feed‐to‐gain ratio did not differ between MCT6/8 and AGP. Per replicate, two random selected piglets were challenged intravenously with E. coli‐lipopolysaccharide (LPS) or saline solution (S) at Days 21 and 28. All challenged animals were sacrificed; blood and digestive tract samples (jejunum/ileum) were collected at Day 35. LPS challenge consistently reduced villus height and crypt depth for MCT6/8 and AGP. However, LPS‐challenged piglets supplemented with MCT6/8 restored villus height, where AGP did not. MCT6/8 piglets had higher serum IgA, more jejunal IgA‐positive plasma cells and goblet cells than AGP. At the ileal level, results were similar, though less pronounced. The present study offers new insight in the benefits of MCT6/8 over AGP in the post‐weaning period. There is in vitro anti‐microbial action of C6 and C8 on E. coli. In vivo, MCT6/8 also has protective effects in the small intestine that may result in growth promotion.  相似文献

From porcine rectal swabs or feces, we isolated lactic acid bacteria and used porcine Peyer's patch cells to select them for inducibility of IgA production as an indicator of probiotic effects. The strain selected as a new probiotic was named ‘Enterococcus faecium NHRD IHARA‘. To verify the probiotic effects of this strain in vivo, 536 piglets at age 25 days were assigned to either the trial group, which administrated the strain, or the control group. An increase in IgA in the feces was observed at age 45 days (P < 0.05 compared with the control group); a significant increase in serum IgA was also observed at the end of the study (P < 0.01) in the trial group. In addition, significant differences between the groups in terms of body weight (P < 0.05) and average daily gain (P < 0.01) were observed. The rate of detection of swine‐pathogenic Escherichia coli gene in the feces tended to be lower in the trial group than in the controls. The novel probiotic strain; E. faecium NHRD IHARA may have beneficial effects on swine growth by inducing IgA production and reducing rates of colonization by pathogens in the body.  相似文献

Porcine edema disease (ED) is caused by Shiga toxin 2e‐producing Escherichia coli (STEC). Post‐weaned piglets often suffer from ED as a result of intestinal infection with STEC, which causes impaired growth performance and high mortality. Antimicrobial therapy is a curative treatment for piglets infected with STEC, but the emergence of antimicrobial‐resistant STEC has become a serious problem for Japanese pig farmers. Therefore, an alternative strategy other than antimicrobial therapy is needed for the prevention or treatment of ED. In this study, we evaluated the effect of oral administration of Bacillus subtilis DB9011 (DB9011) to prevent the experimental infection of STEC in weaning piglets. Eight 21‐day‐old piglets were divided into two groups: STEC challenge with the basal diet, and STEC challenge with DB9011 supplemented diet. The challenge was carried out when the animals were 25, 26 and 27 days old using STEC contained in capsules resistant against gastric digestion. All pigs were euthanized at 36 days of age. DB9011 improved the symptoms of ED and decreased the number of STEC in the ileal digesta and feces. Accordingly, oral administration of DB9011 in weaned piglets prevents ED through the suppression of the growth of STEC in the ileum.  相似文献

For the last several decades, antimicrobial compounds have been used to promote piglet growth at weaning through the prevention of subclinical and clinical disease. There are, however, increasing concerns in relation to the development of antibiotic‐resistant bacterial strains and the potential of these and associated resistance genes to impact on human health. As a consequence, European Union (EU) banned the use of antibiotics as growth promoters in swine and livestock production on 1 January 2006. Furthermore, minerals such as zinc (Zn) and copper (Cu) are not feasible alternatives/replacements to antibiotics because their excretion is a possible threat to the environment. Consequently, there is a need to develop feeding programs to serve as a means for controlling problems associated with the weaning transition without using antimicrobial compounds. This review, therefore, is focused on some of nutritional strategies that are known to improve structure and function of gastrointestinal tract and (or) promote post‐weaning growth with special emphasis on probiotics, prebiotics, organic acids, trace minerals and dietary protein source and level.  相似文献

In 2008, we identified vancomycin‐resistant enterococci (VRE) in Michigan swine, which was the first report of VRE in livestock from North America. Continued sampling in 2009 and 2010 was conducted to determine whether VRE persisted in Michigan. In 2009, swine faecal and feed samples (n = 56), county fair pig barn manure samples (n = 9) and pooled Michigan State Fair pig barn manure samples (n = 18) were screened for VRE. In 2010, swine faecal samples were collected from 26 county fairs (n = 73) and nine commercial swine farms in six states (n = 28). Recovered VRE isolates were molecularly evaluated by polymerase chain reaction, restriction fragment length polymorphism, pulsed‐field gel electrophoresis (PFGE), S1 nuclease digestion and multilocus sequence typing (MLST). Six VRE isolates were identified in 2009 from the State Fair, and another six (8.2%) were recovered from the five county fairs in 2010. All 12 isolates were highly related to the first‐reported VRE from Michigan swine: all were confirmed to be vancomycin‐resistant Enterococcus faecium (VREf) carrying vanA gene on Tn1546 (Type D), were negative for IS1251, hyl and esp gene, carried a 150–160 kb megaplasmid, and have closely similar PFGE patterns with >80% similarity. Classified as ST5, ST6 or ST185 by MLST, all belong to the clonal complex 5, a strain recognized to be circulating among European pigs. This study reveals that VREf are widespread in Michigan swine and persist in the historical absence of the use of agricultural glycopeptides.  相似文献
