The thermal property of bovine intramuscular adipose tissue (IAT) was evaluated using differential scanning calorimetry (DSC) and compared with the melting point temperature (MP) of the fat extract of IAT, which was measured using the slip point method. The beef samples were classified according to the beef marbling score (BMS). Beef with a high BMS contained less protein than that with middle or low BMS. Beef with a high BMS contained significantly more fat than that with a low BMS (< 0.05). The endothermic point temperature (EP) of IAT, measured by DSC, was significantly higher than the MP of IAT fat (< 0.05). The EP showed no significant difference among the three marbling grade groups. Although the MP was correlated with the monounsaturated fatty acids (MUFA) content of IAT (R2 = 0.505), there was no correlation between the EP and the MUFA (R2 = 0.040). However, the EP of IAT treated with collagenase was relatively highly correlated with the MP (R2 = 0.655). Thus, these results suggested that DSC analysis would give us the practical thermal information regarding the melt‐in the‐mouth of beef such as the gelatinization of collagen, along with the melting of fat in IAT.  相似文献

Effects of intramuscular fat deposition on the chemical composition, tenderness, and free amino acids (FAA) concentration of beef were studied using various classified carcasses of 21 Japanese Black (Wagyu) steers. The Beef Marbling Standard (BMS) No., judged on the market in 1998, and fat content, ranged from 2 to 10 and 4.8 to 39.0% in the M. longissimus thoracis, respectively. Moisture content was negatively correlated with the fat content (r =?0.98, P < 0.01). In contrast, protein content was relatively constant up to approximately 23% fat, corresponding to BMS No.4, and decreased as the fat content increased. Cooking loss was also constant up to approximately 28% fat, corresponding to BMS No.4, and markedly decreased with fat content increase. The beef in these cases were, however, classified into BMS No. around 9 and 11, respectively, according to the BMS model in 1988. A negative correlation was found between the shear‐force value and the fat content (r =?0.83, P < 0.05). Most FAA concentrations on the wet weight of meat were correlated negatively with the fat content, except glutamine, and this negative relationship was still observed when the concentrations were recalculated on the basis of protein. These results indicate that higher marbling Wagyu beef (above approx. 23% fat) would have an extremely lower content of protein, which would partly explain the lower cooking loss and FAA with fat increase. In addition, some other reasons not relating to protein content (e.g. high fat content preventing the breakdown of protein to FAA) seem to explain the negative correlation between fat and FAA.  相似文献

The percentage of MUFA to total fatty acids of beef differs among intermuscular, intramuscular, and subcutaneous fat even within an individual cow. Our objective was to investigate the variation of the percentage of MUFA by geometric and sectional change of marbling flecks in rib eye. Longissimus muscles of 8 Japanese Black steers from a common sire and a common maternal grand sire were used. Three slices (1 from rib roast and 2 from sirloin) from each animal were selected for analysis. Five marbling flecks from each slice were randomly taken to obtain the percentage of MUFA using gas chromatography. High-quality digital images of all slices were taken with a mirror-type camera. The area and location of each marbling fleck were calculated by image analysis. The marbling flecks were categorized by area [small <0.4 cm(2), medium 0.4 to 2.0 cm(2), large >2.0 cm(2)], by location (dorsal and ventral), and by slice section through the LM (front, middle, and back). The effects of classification according to the area, location, and slice section were statistically significant (P < 0.05) for the percentage of MUFA. Least squares means of the percentage of MUFA for marbling flecks of sizes small, medium, and large were 56.8, 58.4, and 60.2%, respectively, indicating that larger marbling flecks had greater MUFA (P < 0.05). Those of dorsal, ventral, front, middle, and back were 59.1, 57.8, 55.4, 59.9, and 60.1%, respectively. The percentages of MUFA of the marbling flecks located in the dorsal part were greater than those in the ventral part (P < 0.05). The percentages of MUFA from middle and back were greater than those from front (P < 0.01). We suggest that the area, location, and slice section of marbling would be the determining factors for the percentage of MUFA of marbling.  相似文献

Twenty 10‐month‐old Japanese Black steers were used to evaluate the effects of clay on fat necrosis and carcass characteristics. Ten steers (Clay group) were fed the clay (50 g/day) during 10–30 months of age. The other 10 steers (Control group) were not fed it. There was no significant difference in body weight or average daily gain between the two groups (P > 0.05). The occurrence of fat necrotic mass in the Clay group (30%) was lower (P < 0.05) than that in the Control group (90%) at slaughter. The size of necrotic masses in the Clay group was smaller (P < 0.05) than that in the Control group. There was no significant difference in the marbling score, beef color, Longissimus muscle area or subcutaneous fat thickness between the two groups. These results suggest that the clay prevented the occurrence of fat necrosis and did not affect the carcass characteristics in Japanese Black steers.  相似文献

Crude fat content of longissimus (ribeye) muscle of beef cattle was predicted from a ratio of fat area (RFA) to area of ribeye muscle calculated from computer image analysis (CIA). Cross sections of 64 ribeyes taken from the 6-7th rib from cattle at experiment station A and cross sections of 94 ribeyes taken from the 6-7th rib from cattle at Experiment Station B were used in this study. Slices (1 to 1.5 cm in thickness) of just the Longissimus dorsi were homogenized and sampled for chemical estimation of crude fat content using petroleum ether. Crude fat content as determined from chemical analysis was used as the true estimate of fat content. A CCD (charge-coupled device) camera was used as the input device at Experiment Station A, and a single-lens reflex camera was used at Experiment Station B to photograph ribeyes for CIA. The contour comparison method, which assigns a threshold value for each marbling particle, was used to obtain accurate binarization in this study. Minimum and maximum of chemical measurements of crude fat were 2.1 and 39.8%, and for CIA calculation of the RFA were 6.1 and 56.8%, respectively. This range covered almost the complete range of the beef marbling standard used in carcass grading in Japan. The equation for the regression of the crude fat content (Y) on RFA (X) calculated from CIA for all of the data was Y = .793X-3.04 with r2 = .96. Regression equations for prediction of crude fat percentage from RFA taking into consideration the effect of experiment station were Y = .741X-2.22 with r2 = .91 for Experiment Station A, and Y = .782X-2.54 with r2 = .91 for Experiment Station B. Analysis of covariance showed that the effects of experiment stations on intercepts and slopes were not significant (P > .10). The ranges of differences between actual and predicted crude fat content from the prediction equation that was calculated without consideration of the effect of station were -6.4 to 4.0%. CIA of cross sections of the ribeye muscle seems to have potential for prediction of crude fat content.  相似文献

The objective of this study was to estimate variance components related to imprinting for carcass traits and physiochemical characteristics in Japanese Black cattle. The carcass records obtained from 4,220 Japanese Black feedlot cattle included carcass weight (CW), rib eye area (REA), rib thickness, subcutaneous fat thickness, and beef marbling score (BMS), and the physiochemical characteristics were fat, moisture, glycogen per proportion of moisture content, oleic acid, and monounsaturated fatty acids (MUFA). To detect gametic effects, an imprinting model was fitted. High additive heritabilities were estimated for all traits (from 0.516 for glycogen to 0.853 for fat) and were reduced in Mendelian heritability. The range of the differences was from 0.002 (CW) to 0.331 (fat and moisture), and the reductions were due to their imprinting variances. The ratio of the imprinting variance to the total additive genetic variance for REA (0.374), BMS (0.291), fat (0.387), moisture (0.388), and MUFA (0.337) were large (p < 0.05). These imprinting variances were due to the maternal contribution and suggested the existence of maternally expressed genomic imprinting effects on the traits in Japanese Black cattle. Therefore, maternal gametic effects should be considered in breeding programs for Japanese Black cattle.  相似文献

To understand the molecular mechanisms that regulate intramuscular fat deposition (marbling), cDNA clones expressed in adipose tissues of Korean cattle were identified and characterized. One clone had a total length of 1262 nucleotides coding for 314 amino acids. It was identified as one encoding bovine homolog of human CGI-105 mRNA. CGI-105 is a member of fumarylacetoacetate hydrolase family. Comparison of the deduced amino acid sequences of bovine CGI-105 with those of human revealed more than 89% identity. High levels of CGI-105 mRNA expression were detected in muscle, heart, and kidney tissues among various bovine tissues. Carcass traits, including backfat thickness, rib eye area, yield index, marbling score, and quality grade were analyzed in steer of Korean cattle. A CCAAT/enhancer binding protein alpha (C/EBPα) is one of adipocyte differentiation factors that may affect deposition of fat in muscle. mRNA levels of CGI-105 and C/EBPα genes were determined in the loin muscle tissues of steers. Correlation between carcass traits and mRNA levels of the genes was estimated by Pearson's correlation coefficient. The mRNA levels of C/EBPα showed strong positive correlation (r = 0.83, p < 0.01) with marbling scores. The results of the present study indicate that the manipulation of the expression of the C/EBPα gene may contribute to the development of a method for enhancing intramuscular fat deposition in beef.  相似文献

The deposition of fat in muscle, recognized by the consumer as marbling, is an important meat quality trait. The objective of the study was to provide additional insights into the quantitative extent of marbling by means of computer image analysis. Fifty-one F(2) generation German Holstein and Charolais crossbreed cattle, 18 mo of age, were used to determine relationships among marbling traits, adipocyte size, and the amount of adipose tissue in different depots. Differences were recorded among the size of i.m. adipocytes in different groups of marbling flecks, divided according to the location in the muscle cross-section and to the size of the marbling flecks. The results showed positive correlation between i.m. adipocyte size and the weight of s.c. fat, intestinal fat, omental fat, and perirenal fat (r = 0.50, 0.61, 0.70, and 0.63, respectively, P < 0.001). The i.m. adipocyte size was correlated with i.m. fat content, number of marbling flecks, proportion of marbling fleck area, and total length of marbling flecks (r = 0.71, 0.44, 0.62, and 0.55, respectively, P < 0.01). The number of marbling flecks was also correlated with i.m. fat content, proportion of marbling fleck area, and total length of marbling flecks (r = 0.58, 0.62, and 0.91, P < 0.01, respectively). The ventral marbling flecks had a 5-fold larger fleck area, 4-fold more adipocytes, and larger adipocytes (P < 0.001). Larger marbling flecks contained larger adipocytes (P < 0.001). Moreover, compared with the small marbling flecks, there was a 48-fold larger fleck area and 26-fold more adipocytes in the large marbling flecks. The results indicate that i.m. fat deposition increases concurrently with the other fat depots but is still independent. Furthermore, the i.m. fat is preferentially deposited in the ventral area of LM. Although the i.m. adipocyte size has an important effect on the traits of marbling flecks, cell number plays a greater role in i.m. fat deposition than cell size.  相似文献

本研究旨在对肉用西门塔尔牛与杂交和牛两试验群体进行肉质性状比较,以期为肉品质性状遗传机制剖析及优质肉牛培育提供理论指导。收集肉用西门塔尔牛与杂交和牛两试验群体肉质性状表型数据,包括眼肌面积、剪切力、系水力、pH、肉色、脂肪颜色和大理石花纹等指标,进行相关性及差异性分析,从而综合评价两群体的肉品质。结果显示,两群体的脂肪颜色、系水力及剪切力呈现两两正相关;肉色与脂肪颜色、pH呈正相关,且肉色与pH呈显著相关(P < 0.05),而系水力与pH、肉色和大理石花纹呈负相关。在两试验群体间,和牛杂交群体的大理石花纹、眼肌面积、pH性状表现为极显著优于肉用西门塔尔牛(P < 0.01)。从性别比较来看,肉用西门塔尔牛母牛在大理石花纹、pH、剪切力等性状上极显著或显著优于公牛(P < 0.01;P < 0.05);而和牛杂交群体母牛的肉色和剪切力性状表现为显著或极显著高于公牛(P < 0.05;P < 0.01)。总体而言,和牛杂交群体的肉质性能优于肉用西门塔尔牛;群体内比较表明,肉用西门塔尔牛群体母牛肉质性能优于公牛,而和牛杂交群体母牛肉质性能低于公牛。  相似文献

Marbling, defined by the amount and distribution of intramuscular fat, is an economically important trait of beef cattle in Japan. The c2‐11#2 expressed sequence tag (EST) has been previously shown to possess expression difference in musculus longissimus muscle between low‐marbled and high‐marbled steer groups, and to be located within genomic region of a quantitative trait locus for marbling. Thus, the ribosomal protein L27a (RPL27A) gene containing the c2‐11#2 EST sequence was considered as a positional candidate for the gene responsible for marbling. In the present study, a single nucleotide polymorphism (SNP) in the promoter region of the RPL27A, referred to as g.3109537C>T, was detected between the 2 steer groups. The SNP was associated with the predicted breeding value for beef marbling standard number by the analyses using Japanese Black beef cattle population. The effect of genotypes of the SNP on the predicted breeding value for subcutaneous fat thickness was not statistically significant. These findings suggest that the RPL27A SNP may be useful for effective marker‐assisted selection to increase the levels of marbling in Japanese Black beef cattle.  相似文献

The objective of this study was to develop mathematical equations for describing the change in marbling in Japanese Black steers using longitudinal measurements. Serial ultrasound measurements were taken at 14, 16, 20, and 26 months of age and analyzed using an image analysis software. The longitudinal marbling measurements from the ultrasound images and carcasses were fitted into a nonlinear logistic curve. Data used for the analysis consisted of 749 steers that converged in nonlinear curve fitting and showed reasonable estimated parameters of the logistic curves. The average predicted mature beef marbling score (BMS) and maturation rate were 6.26 and 0.353, respectively, and the average maturity levels at 24 months of age were 83.9%. The heritability estimates for the predicted maturity traits were moderate, indicating that these traits may have potential for genetic improvement. There was a negative relationship between the expected progeny differences between carcass BMS and maturity traits, suggesting that genetic improvement by carcass BMS may lead to the selection of bulls with late maturity for marbling. The results indicate that ultrasound and model building for marbling can be useful tools to correctly select candidate bulls with high marbling in the early fattening period.  相似文献

Ectopic fats have been recognized as a new risk factor for metabolic syndrome. In obese humans, ectopic fat accumulations are affected by body fat distribution. Intramuscular adipose tissue is categorized as one of the ectopic fats. Japanese black cattle (Wagyu) are characterized by the ability to accumulate high amounts of intramuscular adipose tissue. In Japan, the marbling level is indicated by the beef marbling standard number (BMS No.), which reflects the intramuscular fat content of longissimus muscle. We hypothesized that the intramuscular fat accumulation is affected by the body fat distribution in Wagyu cattle. In this study, we showed that the BMS No. was not correlated with the subcutaneous and visceral adipocyte diameter. In contrast, the BMS No. was positively correlated with intramuscular adipocyte diameter. These results indicate that the intramuscular adipocyte diameter of Wagyu is hypertrophied with an increase in the intramuscular fat accumulation. In addition, we showed that the BMS No. was positively correlated with the subcutaneous fat percentage. In contrast, the BMS No. was negatively correlated with the visceral fat percentage. These results indicate that highly marbled Wagyu cattle have a higher percentage of subcutaneous fat and a lower percentage of visceral fat.  相似文献

The bovine growth hormone gene (bGH) possesses three haplotypes, A, B and C, that differ by amino acid mutations at positions 127 and 172 in the fifth exon: (leucine 127, threonine 172), (valine 127, threonine 172) and (valine 127, methionine 172) respectively. The correlation between meat quality or carcass weight and these haplotypes was investigated in Japanese black cattle. Altogether, 940 bGH haplotypes were compared with respect to six carcass traits: carcass weight, longissimus muscle area, rib thickness, subcutaneous fat thickness, beef marbling score and beef colour. The frequency of the B haplotype was higher (0.421) than that of A (0.269) and C (0.311). High carcass weight and low beef marbling were associated with haplotype A (p < 0.05 and p < 0.01 respectively), whereas beef marbling was increased by haplotype C (p < 0.05). Estimated regression coefficient of the A haplotype substitution effect for carcass weight and beef marbling score were 5.55 (13.1% of the phenotypic SD) and -0.31 (17.0%) respectively. That of the C haplotype for beef marbling score was 0.20 (11.0%). The other traits showed no relationship to the haplotypes examined. The results of this investigation suggest that information pertaining to bGH polymorphisms in Japanese black cattle could be used to improve the selection of meat traits.  相似文献

Monosaccharides such as glucose contribute to the development of meat flavor upon heating via the Maillard reaction; therefore, monosaccharide content is related to beef palatability. Here, we analyzed the effects of genetic and environmental factors on the content of glycogen, one of the precursors of monosaccharides, in the muscles of 958 fattened Japanese Black cattle from Yamagata Prefecture. Analysis of variance showed that muscle glycogen content was affected by the farm and postmortem periods, but not by sex, slaughter age, slaughter month or number of days detained at the slaughter yard. Additionally, consumption of digestible brown rice feed elevated muscle glycogen levels. Glycogen heritability was estimated to be 0.34, and genetic correlations between glycogen and carcass weight (CW) or beef marbling standard (BMS) were weak. The predicted breeding values varied among paternal lines. These results demonstrated that genetic factors might improve muscle glycogen content and therefore beef palatability, but do not influence CW or BMS.  相似文献

To assess the effects of slaughter weight and sex on APGS (Animal Products Grading Service) quality and APGS yield grade of Korean Hanwoo (n = 20,881) cattle, data were collected from cow, bull, and steer carcasses during a 1-yr period. Factors used to determine quality grade (marbling, meat color, fat color, texture, and overall maturity score) and yield grade (cold carcass weight, adjusted fat thickness, and longissimus muscle area) by the Korean grading system were recorded. Both yield and quality grades were improved (P < 0.01) with heavier slaughter weight, but there was no difference in yield grade for Hanwoo cattle classes heavier than 551 kg (P > 0.01). Longissimus muscle area, adjusted fat thickness, and marbling score increased (P < 0.01) with carcass weight. Bull carcasses showed higher yield but lower quality than those of cows or steers (P < 0.01). The quality grade of steer carcasses was higher (P < 0.01) than that of cow carcasses due to higher marbling scores, lower maturity scores, and heavier carcass weights. Hanwoo carcasses with larger longissimus muscle areas in relation to their carcass weight had lower APGS quality grades. The APGS quality grades were different between yield grade A and B carcasses (P < 0.01), but quality grade was not improved by increased fat thickness beyond the point of yield grade B. Adjusted fat thickness and marbling score showed significant (P < 0.01) differences among all yield grade classes, and this resulted in increased quality grade as yield grade decreased. Adjusted fat thickness showed the strongest correlation (r = -0.63) with yield grade, whereas marbling score had the strongest correlation (r = 0.81) with quality grade. Results showed a negative effect of castration on yield but a positive effect on quality. Also, data showed that Hanwoo carcasses with heavier weights had higher quality grades than those of lighter weight.  相似文献

Vitamin A (VA) restriction in beef cattle improves meat marbling; however, the underlying molecular mechanisms remain incompletely understood. We performed microarray analysis to clarify the effect of VA restriction on Longissimus thoracis gene expressions in Japanese Black steers. Six Japanese Black steers 13–14 months of age were divided into two groups: S group (n = 3), which received VA supplementation, and R group (n = 3), in which dietary VA intake was restricted. Steers were fattened for 7 months, following which tissue samples were obtained. Extracted RNA samples were analyzed by Affymetrix Genechip Bovine Genome Array. Lists of genes highly expressed in the R and S groups were obtained. The lists were functionally interpreted using functional annotation software, DAVID. In the R and S groups, 48 and 40 genes were significantly highly expressed, respectively. The gene list of the R group included CD36, LPL, GPAM, DGAT2, and SCD and additional genes annotated ‘PPAR signaling pathway,’ ‘lipid biosynthesis’ and ‘mitochondrion,’ whereas that of the S group included COL1A2, FN1 and DCN and additional genes annotated ‘extracellular matrix.’ Changes in the expression of these genes are possibly involved in marbling improvement in beef cattle by VA restriction.  相似文献

To estimate the direct and correlated responses in meatiness and meat quality, simulated selection was applied using one‐trait selection and index selection including muscle fibre traits. In a total of 2024 pigs of German Landrace, Large White, Leicoma, and Schwerfurter breeds, carcass composition, meat quality, and fibre characteristics of the M. longissimus lumborum were analysed and genetic parameters were calculated by using REML variance component estimation. Coefficients of heritability ranged from low to moderate (growth traits: h2 = 0.22–0.32; meat quality traits: h2 = 0.17–0.27; muscle fibre traits: h2 = 0.12–0.20). The total fibre number and the frequency of white fibres correlated positively with live weight (rg = 0.44 and 0.44, respectively) and with loin muscle area (rg = 0.38 and 0.19, respectively) while the relationships to pH value and drip loss were not as close (rg = ?0.29–0.19). Selection indices were constructed from live weight and muscle area, from pH value and drip loss, and from muscle structure traits. As expected, simulated selection for live weight or loin muscle area lead to direct positive effects but these were associated with negative effects on meat quality traits (selection intensities: SI = 0.1; 0.5). Using selection for an index from loin area and muscle structure (loin muscle area + total fibre number – frequency of white fibres – frequency of giant fibres) the adverse effect on meat quality was minimized (responses in pH value: ?0.05; ?0.01) or was changed towards desired direction (responses in drip loss: ?0.65%; ?0.26%). The results show that fibre characteristics of pig muscle can be used as selection criteria for simultaneous improvement of carcass composition and meat quality in pigs by including indices developed from performance and fibre traits.  相似文献

From a calibration trial involving computer tomography (CT) scanning and dissection of 45 lambs, a prediction equation was derived to estimate total internal fat weight in Scottish Blackface lambs from measurements taken on cross‐sectional CT images. Using data from two cross‐sectional images (at the hip and loin) internal fat can be predicted with relatively high accuracy (adjusted R2 = 62.2%, r = 0.79). The derived equation was then used to predict internal fat weights in a further 427 Scottish Blackface lambs from a separate trial. Phenotypic correlations were calculated between predicted internal fat weight and weights of total carcass fat, muscle and bone, predicted using previously derived equations. When considering absolute tissue weights, adjusted for fixed effects, internal fat showed the strongest positive correlation with carcass fat (0.58), followed by muscle (0.36), and then by bone (0.32). When tissue weights were adjusted for fixed effects and total carcass weight (so considering tissue weights relative to size), internal fat showed a lower correlation with carcass fat weight (0.36) and negative correlations with muscle (?0.35) and bone (?0.19). These results provide the basis for more complex studies of relationships (phenotypic and genetic) between internal fat in hill lambs and economically important traits, such as carcass composition and survival of lambs, and tissue levels in different depots in hill ewes.  相似文献

Genomic imprinting should be considered in animal breeding systems to avoid lead in bias in genetic parameter estimation. The objective of this study was to clarify the effects of pedigree information on imprinting variances for carcass traits and fatty acid composition in Japanese Black cattle. Carcass records [carcass weight, rib eye area, rib thickness (RT), subcutaneous fat thickness and beef marbling score (BMS)] and fatty acid composition were obtained for 11,855 Japanese Black feedlot cattle. To estimate and compare the imprinting variances for the traits, two imprinting models with different pedigree information [the sire–dam gametic relationship matrix (Model 1) and the sire–maternal grandsire (MGS) numerator relationship matrix (Model 2)] were fitted. The ratio of the imprinting variance to the total additive genetic variance for RT (6.33%) and BMS (19.00%) was significant in Model 1, but only that for BMS (21.09%) was significant in Model 2. This study revealed that fitting the sire–MGS model could be useful in estimating imprinting variance under certain conditions, such as when restricted pedigree information is available. Furthermore, the present result suggested that the maternal gametic effects on BMS should be included in breeding programmes for Japanese Black cattle to avoid selection bias caused by imprinting effects.  相似文献

Genetic selection in favor of muscle growth at the expense of fat should affect characteristics of muscles, and therefore beef quality. This study was conducted with two extreme groups of six animals selected among 64 Charolais young bulls ranked according to their genetic potential for muscle growth. Muscle characteristics were assessed in Rectus abdominis (RA, slow oxidative) and Semitendinosus (ST, fast glycolytic) muscles. Intramuscular fat content and proportions of myosin heavy chains I (slow) and IIA (fast oxido‐glycolytic) and certain indicators of oxidative metabolism (activities of citrate synthase (CS), isocitrate dehydrogenase and cytochrome‐c oxidase (COX); expression of H‐fatty acid binding protein (FABP)) were higher in RA than in ST muscle. Genetic selection for muscle growth reduced intramuscular fat content and the activities of some oxidative metabolism indicators (namely CS, COX only). The positive correlation between muscle triacylglycerol content and A‐FABP messenger RNA level (a marker of adipocyte differentiation) (r = 0.53, P < 0.05) suggests that A‐FABP may be a good marker of the ability of bovines to deposit intramuscular fat. In conclusion, the metabolic muscle characteristics which respond to the selection process in favor of muscle growth clearly differ from the muscle characteristics which allow muscle types to be differentiated.  相似文献
